430 An Information Theoretic Approach to Rescoring Peptides Produced by De Novo Peptide Sequencing
Authors: John R. Rose, James P. Cleveland, Alvin Fox
Abstract:
Tandem mass spectrometry (MS/MS) is the engine driving high-throughput protein identification. Protein mixtures possibly representing thousands of proteins from multiple species are treated with proteolytic enzymes, cutting the proteins into smaller peptides that are then analyzed generating MS/MS spectra. The task of determining the identity of the peptide from its spectrum is currently the weak point in the process. Current approaches to de novo sequencing are able to compute candidate peptides efficiently. The problem lies in the limitations of current scoring functions. In this paper we introduce the concept of proteome signature. By examining proteins and compiling proteome signatures (amino acid usage) it is possible to characterize likely combinations of amino acids and better distinguish between candidate peptides. Our results strongly support the hypothesis that a scoring function that considers amino acid usage patterns is better able to distinguish between candidate peptides. This in turn leads to higher accuracy in peptide prediction. Keywords: Tandem mass spectrometry, proteomics, scoring, peptide, de novo, mutual information

Authors: Bing Song, Kexi Han, Xuewei Lv
Abstract:
Titania slag is a high quality raw material containing titanium in the subsequent process of titanium pigment. The effects of cooling approaches of granulating, water cooling, and air cooling on chemical, phases, and acidolysis of Panzhihua titania slag were investigated. Compared to the original slag which was prepared by the conventional processing route, the results show that the titania slag undergoes oxidation of Ti3+during different cooling ways. The Ti2O3 content is 17.50% in the original slag, but it is 16.55% and 16.84% in water cooled and air-cooled slag, respectively. Especially, the Ti2O3 content in granulated slag is decreased about 27.6%. The content of Fe2O3 in granulated slag is approximately 2.86% also obviously higher than water (<0.5%) or air-cooled slag (<0.5%). Rutile in cooled titania slag was formed because of the oxidation of Ti3+. The rutile phase without a noticeable change in water cooled and air-cooled slag after the titania slag was cooled, but increased significantly in the granulated slag. The rate of sulfuric acid acidolysis of cooled slag is less than the original slag. The rate of acidolysis is 90.61% and 92.46% to the water-cooled slag and air-cooled slag, respectively. However, the rate of acidolysis of the granulated slag is less than that of industry slag about 20%, only 74.72%.

Abstract:
Potassium borates, which are widely used in welding and metal refining industry, as a lubricating oil additive, cement additive, fiberglass additive and insulation compound, are one of the important groups of borate minerals. In this study the production of a potassium borate mineral via hydrothermal method is aimed. The potassium source of potassium nitrate (KNO3) was used along with a sodium source of sodium hydroxide (NaOH) and boron source of boric acid (H3BO3). The constant parameters of reaction temperature and reaction time were determined as 80°C and 1 h, respectively. The molar ratios of 1:1:3 (as KNO3:NaOH:H3BO3), 1:1:4, 1:1:5, 1:1:6 and 1:1:7 were used. Following the synthesis the identifications of the produced products were conducted by X-Ray Diffraction (XRD), Fourier Transform Infrared Spectroscopy (FT-IR) and Raman Spectroscopy. The results of the experiments and analysis showed in the ratio of 1:1:6, the Santite mineral with powder diffraction file number (pdf no.) of 01-072-1688, which is known as potassium pentaborate (KB5O8·4H2O) was synthesized as best. Keywords: Hydrothermal synthesis, potassium borate, potassium nitrate, santite.

Authors: N. Sanmanee, K. Panishkan, K. Obsuwan, S. Dharmvanij
Abstract:
The increments of aromatic structures are widely used to monitor the degree of humification. Compost derived from mix manures mixed with agricultural wastes was studied. The compost collected at day 0, 7, 14, 21, 28, 35, 49, 77, 91, 105, and 119 was divided into 3 stages, initial stage at day 0, thermophilic stage during day 1-48, and mature stage during day 49-119. The change of highest absorptions at wavelength range between 210-235 nm during day 0- 49 implied that small molecules such as nitrates and carboxylic occurred faster than the aromatic molecules that were found at wavelength around 280 nm. The ratio of electron-transfer band at wavelength 253 nm by the benzonoid band at wavelength 230 nm (E253/E230) also gradually increased during the fermenting period indicating the presence of O-containing functional groups. This was in agreement with the shift change from aliphatic to aromatic structures as shown by the relationship with C/N and H/C ratios (r = - 0.631 and -0.717, p< 0.05) since both were decreasing. Although the amounts of humic acid (HA) were not different much during the humification process, the UV spectral deconvolution showed better qualitative characteristics to help in determining the compost quality. From this study, the compost should be used at day 49 and should not be kept longer than 3 months otherwise the quality of HA would decline regardless of the amounts of HA that might be rising. This implied that other processes, such as mineralization had an influence on the humification process changing HA-s structure and its qualities.

Abstract:
Lightweight concrete is being used in the construction industry as a building material in its own right. Ultra-lightweight concrete can be applied as a filler and support material for the manufacturing of composite building materials. This paper is about the development of a stable and reproducible ultra-lightweight concrete with the inclusion of poly-lactic acid (PLA) beads and assessing the feasibility of PLA as a lightweight aggregate that will deliver advantages such as a more eco-friendly concrete and a non-petroleum polymer aggregate. In total, sixty-three samples were prepared and the effectiveness of mineral admixture, curing conditions, water-cement ratio, PLA ratio, EPS ratio and perlite ratio on compressive strength of perlite concrete are studied. The results show that PLA particles are sensitive to alkali environment of cement paste and considerably shrank and lost their strength. A higher compressive strength and a lower density was observed when expanded polystyrene (EPS) particles replaced PLA beads. In addition, a set of equations is proposed to estimate the water-cement ratio, cement content and compressive strength of perlite concrete. Keywords: Perlite concrete, poly-lactic acid, expanded polystyrene, concrete.

Abstract:
Traditionally, Yemini Sidr honey has been reported to cure liver problems, stomach ulcers, and respiratory disorders. In this experiment, we evaluated Yemeni Sidr honey for its ability to protect inflammations caused by acetic acid and formalin -induced writhing, carrageenan and histamine-induced paw oedema in experimental rat model. Hyperpyrexia, membrane stabilizing activity, and phytochemical screening of the honey was also examined. Yemini Sidr Honey at (100, 200 and 500 mg/kg) exhibited a concentration dependant inhibition of acetic acid induced and formalin induced writhing, paw oedema induced by carrageenan & histamine, and hyperpyrexia induced by brewer's yeast, it also inhibited membrane stabilizing activity. Phytochemical screenings of the honey reveal the presence of flavonoids, steroid, alkaloids, saponins and tannins. This study suggested that Yemeni Sidr honey possess very strong antiinflammatory, analgesic and antipyretic effects and these effects would be a result of the phytochemicals present. Keywords: Anti-inflammatory, Analgesic, Carrageenan, Aceticacid, Histamine, Yemini Sidr Honey

Abstract:
Date palm (Phoenix dactylifera L.) seeds are waste streams which are considered a major problem to the food industry. They contain potentially useful protein (10-15% of the whole date-s weight). Global production, industrialisation and utilisation of dates are increasing steadily. The worldwide production of date palm fruit has increased from 1.8 million tons in 1961 to 6.9 million tons in 2005, thus from the global production of dates are almost 800.000 tonnes of date palm seeds are not currently used [1]. The current study was carried out to convert the date palm seeds into useful protein powder. Compositional analysis showed that the seeds were rich in protein and fat 5.64 and 8.14% respectively. We used several laboratory scale methods to extract proteins from seed to produce a high protein powder. These methods included simple acid or alkali extraction, with or without ultrafiltration and phenol trichloroacetic acid with acetone precipitation (Ph/TCA method). The highest protein content powder (68%) was obtained by Ph/TCA method with yield of material (44%) whereas; the use of just alkali extraction gave the lowest protein content of 8%, and a yield of 32%.

Abstract:
The use of probiotics engineered to express specific enzymes has been the subject of considerable attention in poultry industry because of increased nutrient availability and reduced cost of enzyme supplementation. Phytase enzyme is commonly added to poultry feed to improve digestibility and availability of phosphorus from plant sources. To construct a probiotic with potential of phytate degradation, phytase gene (appA) from E. coli was cloned and transformed into two probiotic bacteria Lactobacillus salivarius and Lactococcus lactis. L. salivarous showed plasmid instability, unable to express the gene. The expression of appA gene in L. lactis was analyzed by detecting specific RNA and zymography assay. Phytase enzyme was isolated from cellular extracts of recombinant L. lactis, showing a 46 kDa band upon the SDS-PAGE analysis. Zymogram also confirmed the phytase activity of the 46 kDa band corresponding to the enzyme. An enzyme activity of 4.9U/ml was obtained in cell extracts of L. lactis. The growth of native and recombinant L. lactis was similar in the presence of two concentrations of ox bile. Keywords: Lactobacillus salivarus, Lactococcus lactis, recombinant, phytase, poultry.

Abstract:
In this paper, we used a high-quality anodization technique to make a colored wind chime with a nano-tube structure anodic film, which controls the length-to-diameter ratio of an aluminum rod and controls the oxide film structure on the surface of the aluminum rod by an anodizing method. The research experiment used hard anodization to grow a controllable thickness of anodic film on an aluminum alloy surface. The hard anodization film has high hardness, high insulation, high-temperature resistance, good corrosion resistance, colors, and mass production properties that can be further applied to transportation, electronic products, biomedical fields, or energy industry applications. This study also provides in-depth research and a detailed discussion of the related process of aluminum alloy surface hard anodizing, including pre-anodization, anodization, and post-anodization. The experiment parameters of anodization include using a mixed acid solution of sulfuric acid and oxalic acid as an anodization electrolyte and controlling the temperature, time, current density, and final voltage to obtain the anodic film. In the results of the experiments, the properties of the anodic film, including thickness, hardness, insulation, and corrosion characteristics, the microstructure of the anode film were measured, and the hard anodization efficiency was calculated. Thereby it can obtain different transmission speeds of sound in the aluminum rod. And, different audio sounds can present on the aluminum rod. Another feature of the present experiment result is the use of the anodizing method and dyeing method, laser engraving patterning and electrophoresis method to make good-quality colored aluminum wind chimes.

Abstract:
Performing culture and characterization of mycobacteria in low resource settings like Nigeria is a very difficult task to undertake because of the very few and limited laboratories carrying out such an experiment; this is a largely due to stringent and laborious nature of the tests. Hence, a rapid, simple and accurate test for characterization is needed. The “SD BIOLINE TB Ag MPT 64 Rapid ®” is a simple and rapid immunochromatographic test used in differentiating Mycobacteria into Mycobacterium tuberculosis (NTM). The 100 sputa were obtained from patients suspected to be infected with tuberculosis and presented themselves to hospitals for check-up and treatment were involved in the study. The samples were cultured in a class III Biosafety cabinet and level III biosafety practices were followed. Forty isolates were obtained from the cultured sputa, and there were identified as Acid-fast bacilli (AFB) using Zeihl-Neelsen acid-fast stain. All the isolates (AFB positive) were then subjected to the SD BIOLINE Analyses. A total of 31 (77.5%) were characterized as MTBC, while nine (22.5%) were NTM. The total turnaround time for the rapid assay was just 30 minutes as compared to a few days of phenotypic and genotypic method. It was simple, rapid and reliable test to differentiate MTBC from NTM.

Abstract:
Yogurt is a coagulated milk product obtained from the lactic acid fermentation by the action of Lactobacillus bulgaricus and Streptococcus thermophilus. The additions of fruits into milk may enhance the taste and the therapeutical values of milk products. However fruits also may change the fermentation behaviour. In this present study, the changes in physicochemical, the peptide concentration, total phenolics content and the antioxidant potential of yogurt upon the addition of Hylocereus polyrhizus and Hylocereus undatus (white and red dragon fruit) were investigated. Fruits enriched yogurt (10%, 20%, 30% w/w) were prepared and the pH, TTA, syneresis measurement, peptide concentration, total phenolics content and DPPH antioxidant inhibition percentage were determined. Milk fermentation rate was enhanced in red dragon fruit yogurt for all doses (-0.3606 - -0.4126 pH/h) while only white dragon fruit yogurt with 20% and 30% (w/w) composition showed increment in fermentation rate (-0.3471 - -0.3609 pH/h) compared to plain yogurt (-0.3369pH/h). All dragon fruit enriched yogurts generally showed lower pH readings (pH 3.95 - 4.03) compared to plain yogurt (pH 4.05). Both fruit yogurts showed a higher lactic acid percentage (1.14-1.23%) compared to plain yogurt (1.08%). Significantly higher syneresis percentage (57.19 - 70.32%) compared to plain yogurt (52.93%) were seen in all fruit enriched yogurts. The antioxidant activity of plain yogurt (19.16%) was enhanced by the presence of white and red dragon fruit (24.97- 45.74%). All fruit enriched yogurt showed an increment in total phenolic content (36.44 - 64.43mg/ml) compared to plain yogurt (20.25mg/ml). However, the addition of white and red dragon fruit did not enhance the proteolysis of milk during fermentation. Therefore, it could be concluded that the addition of white and red dragon fruit into yogurt enhanced the milk fermentation rate, lactic acid content, syneresis percentage, antioxidant activity, and total phenolics content in yogurt. Keywords: Antioxidant activity, Hylocereus polyrhizus, Hylocereus undatus, yogurt

Authors: Mochamad Chalid, Imam Prabowo
Abstract:
Today, the pollution due to non-degradable material such as plastics, has led to studies about the development of environmental-friendly material. Because of biodegradability obtained from natural sources, polylactid acid (PLA) and ijuk fiber are interesting to modify into a composite. This material is also expected to reduce the impact of environmental pollution. Surface modification of ijuk fiber through alkalinization with 0.25 M NaOH solution for 30 minutes was aimed to enhance its compatibility to PLA, in order to improve properties of the composite such as the mechanical properties. Alkalinization of the ijuk fibers annihilates some surface components such as lignin, wax and hemicelloluse, so the pore on the surface clearly appeared, decreasing of the density and diameter of the ijuk fibers. The change of the ijuk fiber properties leads to increase the mechanical properties of PLA composites reinforced the ijuk fibers through strengthening of the mechanical interlocking with the PLA matrix. An addition to enhance the distribution of the fibers in the PLA matrix, the stirring during DCM solvent evaporation from the mixture of the ijuk fibers and the dissolved-PLA can reduce amount of the trapped-voids and fibers pull-out phenomena, which can decrease the mechanical properties of the composite.

The prosopis seed for this work were obtained from Enugu State in the South East part of Nigeria. The seeds were prepared by separating the endosperm from the seed coat and cotyledon. Three methods were used to separate them, which are acidic method, roasting method and boiling method. 20g of seed were treated with different concentrations (25, 40, 55, 70, and 85% w/w) at 100°C and constant time (30 minutes), under continuous stirring with magnetic stirrer. Also 20g of seed were treated with sulphuric acid of concentrations 40% w/w at 100°C with different time (10, 15, 20, 25, 30 minutes), under continuous stirring with magnetic stirrer. Finally, 20g of seed were treated with sulphuric acid of concentrations 40% w/w at different temperature (20°C, 40°C, 60°C, 80°C, and 100°C) with constant time (30 minutes), under continuous stirring with magnetic stirrer. The whole endosperm extracted was adhesive. The physical properties of the adhesive were determined (appearance, odour, taste, solubility, pH, size, and binding strength). The percentage of the adhesive yield makes the commercialization of the seed in Nigeria possible and profitable. Abstract:
Sickle cell anemia is a recessive genetic disease caused by the presence in the red blood cell, of abnormal hemoglobin called hemoglobin S. It results from the replacement in the beta chain of the acid glutamic acid by valin at position 6. Topics may be homozygous (SS) or heterozygous (AS) most often asymptomatic. Other mutations result in compound heterozygous: - Synthesis of hemoglobin C mutation in the sixth leucin codon (heterozygous SC); - ß-thalassemia (heterozygous S-ß thalassemia). SS homozygous, heterozygous SC and S- ß -thalassemia are grouped under the major sickle cell syndromes. To make a laboratory diagnosis of hemoglobinopathies in a portion of the population in region of Batna, our study was conducted on 115 patients with suspected sickle cell anemia, all cases have benefited from hematological tests as blood count (count RBC, calculated erythrocyte indices, MCV and MCHC, measuring the hemoglobin concentration) and a biochemical test in this case electrophoresis CAPILLARYS HEMOGLOBIN (E). The results showed: 27 cases of sickle cell anemia were found on 115 suspected cases, 73,03% homozygous sickle cell disease and 59,25% sickle cell trait. About 750ml Pawpaw pulp was measured into 2 measuring cylinders A & B of capacity 1 litre heated to 400C, cooled to 200C. 30mls pectinase was added into cylinder A, while 30mls distilled water was added into cylinder B. Enzyme treated sample (A) was allowed to digest for 5hours after which it was heated to 900C for 15 minutes to inactivate the enzyme. The heated sample was cooled and with the aid of a mucillin cloth the pulp was filtered to obtain the clarified pawpaw juice. The juice was filled into 100ml plastic bottles, pasteurized at 950C for 45 minutes, cooled and stored at room temperature. The sample treated with 30mls distilled water also underwent the same process. Freshly pasteurized sample was analyzed for specific gravity, titratable acidity, pH, sugars and ascorbic acid. The remaining sample was then stored for 2 weeks and the above analyses repeated. There were differences in the results of the freshly pasteurized samples and stored sample in pH and ascorbic acid levels, also sample treated with pectinase yielded higher volumes of juice than that treated with distilled water.

Abstract:
The objective of this research is to study the physical and mechanical properties and durability of self-compacting mortars prepared by substituting a part of cement up to a percentage of 30% pozzolan according to different Blaine specific surface area (SSB1=7000 cm2/g and SSB=9000 cm2/g)). Order to evaluate durability, mortars were subjected to chemical attacks in various aggressive environments, a solution of a mixture of nitric acid and ammonium nitrate (HNO3 + NH4NO3) and a magnesium sulfate salt solution (MgSO4)) with a concentration of 10%, for a period of one month. This study is complemented by a comparative study of the durability of mortars elaborated with sulphate resistant cement (SRC). The results show that these mortars develop long-term, mechanical and chemical resistance better than mortars based Portland cement with 5% gypsum (CEM 1) and SRC. We found that the mass losses are lowest in mortars elaborated with pozzolanic cement (30% substitution with SSB2) in both of chemical attack solutions (3.28% in the solution acid and 1.16% in the salt solution) and the compressive strength gains of 14.68% and 8.5% respectively in the two media. This is due to the action of pozzolan which fixes portlandite to form hydrated calcium silicate (CSH) from the hydration of tricalcic silicate (C3S).

Abstract:
Conversion of lignocellulosic biomass is the basis process for production of fuels, chemicals and materials in the sustainable biorefinery industry. Saccharification of lignocellulosic biomass is an essential step which produces sugars for further conversion to target value-added products e.g. bio-ethanol, bio-plastic, g-valerolactone (GVL), 5-hydroxymethylfuroic acid (HMF), levulinic acid, etc. The goal of this work was to develop an efficient enzyme for conversion of biomass to reducing sugar based on crude fungal enzyme from Chaetomium globosum BCC5776 produced by submerged fermentation and evaluate its activity comparing to a commercial Acremonium cellulase. Five local biomasses in Thailand: rice straw, sugarcane bagasse, corncobs, corn stovers, and palm empty fruit bunches were pretreated and hydrolyzed with varying enzyme loadings. Saccharification of the biomass led to different reducing sugar levels from 115 mg/g to 720 mg/g from different types of biomass using cellulase dosage of 9 FPU/g. The reducing sugar will be further employed as sugar feedstock for production of ethanol or commodity chemicals. This work demonstrated the use of promising enzyme candidate for conversion of local lignocellulosic biomass in biorefinery industry.

Analytical instrumentation protocols for the unequivocal detection of biomarkers in suitable geological matrices are critical for future unmanned explorations, including the forthcoming ESA ExoMars mission to search for life on Mars scheduled for 2018 and Raman spectroscopy is currently a part of the Pasteur instrumentation suite of this mission. Here, Raman spectroscopy using 785nm excitation was evaluated for determining various concentrations of beta-carotene in admixture with polyaromatic hydrocarbons and usnic acid have been investigated by Raman microspectrometry to determine the lowest levels detectable in simulation of their potential identification remotely in geobiological conditions in Martian scenarios. Information from this study will be important for the development of a miniaturized Raman instrument for targetting Martian sites where the biosignatures of relict or extant life could remain in the geological record.

Authors: Bianca Schweiger, Lucile Bahnweg, Barbara Palm, Ute Steinfeld
Abstract:
The occurrence and removal of trace organic contaminants in the aquatic environment has become a focus of environmental concern. For the selective removal of carbamazepine from loaded waters molecularly imprinted polymers (MIPs) were synthesized with carbamazepine as template. Parameters varied were the type of monomer, crosslinker, and porogen, the ratio of starting materials, and the synthesis temperature. Best results were obtained with a template to crosslinker ratio of 1:20, toluene as porogen, and methacrylic acid (MAA) as monomer. MIPs were then capable to recover carbamazepine by 93% from a 10-5 M landfill leachate solution containing also caffeine and salicylic acid. By comparison, carbamazepine recoveries of 75% were achieved using a nonimprinted polymer (NIP) synthesized under the same conditions, but without template. In landfill leachate containing solutions carbamazepine was adsorbed by 93-96% compared with an uptake of 73% by activated carbon. The best solvent for desorption was acetonitrile, with which the amount of solvent necessary and dilution with water was tested. Selected MIPs were tested for their reusability and showed good results for at least five cycles. Adsorption isotherms were prepared with carbamazepine solutions in the concentration range of 0.01 M to 5*10-6 M. The heterogeneity index showed a more homogenous binding site distribution. Keywords: Carbamazepine, landfill leachate, removal, reuse

Authors: Mai Thanh Truc, Masao Yoshida
Abstract:
Global temperature had increased by about 0.5oC over the past century, increasing temperature leads to a loss or a decrease of soil organic matter (SOM). Whereas soil organic matter in many tropical soils is less stable than that of temperate soils, and it will be easily affected by climate change. Therefore, conservation of soil organic matter is urgent issue nowadays. This paper presents the effect of different doses (5%, 15%) of Ca-type zeolite in conjunction with organic manure, applied to soil samples from Philippines, Paraguay and Japan, on the decomposition resistance of soil organic matter under high temperature. Results showed that a remain or slightly increase the C/N ratio of soil. There are an increase in percent of humic acid (PQ) that extracted with Na4P2O7. A decrease of percent of free humus (fH) after incubation was determined. A larger the relative color intensity (RF) value and a lower the color coefficient (6logK) value following increasing zeolite rates leading to a higher degrees of humification. The increase in the aromatic condensation of humic acid (HA) after incubation, as indicates by the decrease of H/C and O/C ratios of HA. This finding indicates that the use of zeolite could be beneficial with respect to SOM conservation under global warming condition. Keywords: Global warming, Humic substances, Soil organicmatter, Zeolite.

Authors: M. Mohammadi
Abstract:
In order to evaluate the effect of phosphate and Zn bio-fertilizers on the yield, phytic acid (PA), Zn concentration and PA/Zn molar ratio in bean, a field experiment was carried out for two years. The treatments included two cultivars of bean (Talash and Sadri), four levels of P (P0, P1: 100 kg ha-1 triple super phosphate (TSP), P2: 50 kg ha-1 TSP + phosphate bio-fertilizer, P3: phosphate bio-fertilizer), three levels of Zn (Zn0, Zn1: 50 kg ha-1 ZnSO4, Zn2: Zn bio-fertilizer). Phosphate bio-fertilizer consisted of inoculum of mycorrhizal fungus and Azotobacter and Zn bio-fertilizer consisted of Pseudomonas bacteria. The results revealed that there was significant difference between yield and Zn concentration between years. The effect of cultivar was significant on studied parameters. The lowest content of PA and PA/Zn were obtained from Talash. P treatment caused to significant difference on parameters in which P2 caused to increase yield, P and Zn concentration, and decrease PA and PA/Zn by 21.8%, 38.2%, 33.4%, 17.4% and 38.6% respectively. Zn treatment caused to significant difference on studied parameters. The maximum number of parameters were obtained from Zn1 and Zn2. The higher Zn concentration led to lower content of PA and PA/Zn. Using of P and Zn bio–fertilizers were caused to increasing nutrient uptake, improving growth condition and reducing PA and PA/Zn molar ratio.

Authors: R. Guliyev
Abstract:
The enhanced need for food items is receiving more importance due to a gradual increase in the world population and, in this scenario, fertilizers play a very important role in agriculture. In this study, the production of the normal superphosphate was investigated with a continuous chamber method by adding potassium chloride to a mixture of Hibin apatite and Kingisepp phosphorite. In the experiments, the following parameters were selected: The concentration of sulfuric acid (54–66% (w/w)), the stoichiometric norm of sulfuric acid (100, 107, 110, 114% (w/w)), the ratio of apatite/phosphorite in the mixture of phosphate (95/5, 90/10, 85/15, 80/20, 75/25, 70/30, 65/35,60/40, 55/45, 50/50 (w/w)), potassium chloride/the mixture of phosphate (1/50, 2/50, 3/50,4/50, 5/50 (w/w)), and the reaction time (2–8 min). It was observed that by adding potassium chloride to a low-grade phosphorite and using it to substitute a fraction of high-grade apatite in the normal superphosphate production not only resulted in a high-quality product but also eliminated the waiting period for the maturation of superphosphate in the storage. The objective of this study was to produce a normal superphosphate fertilizer by using a continuous chamber method in order to accelerate the production process and to reduce the environmental pollution caused by fluoride gases by eliminating the maturation time in the storage.

Authors: N. A. Zambri, A. Mohamed, H. Shareef, M. Z. C. Wanik
Abstract:
This paper presents the modeling and simulation of a hybrid proton exchange membrane fuel cell (PEMFC) with an energy storage system for use in a stand-alone distributed generation (DG) system. The simulation model consists of fuel cell DG, lead-acid battery, maximum power point tracking and power conditioning unit which is modeled in the MATLAB/Simulink platform. Poor loadfollowing characteristics and slow response to rapid load changes are some of the weaknesses of PEMFC because of the gas processing reaction and the fuel cell dynamics. To address the load-tracking issues in PEMFC, a hybrid PEMFC and battery storage system is considered and modelled. The model utilizes PEMFC as the main energy source whereas the battery functions as energy storage to compensate for the limitations of PEMFC.Simulation results are given to show the overall system performance under light and heavyloading conditions.

Authors: Goksen Arik, Mihriban Korukluoglu
Abstract:
Lactic acid bacteria (LAB) and some yeast species are common microorganisms found in dairy products and most of them are responsible for the fermentation of foods. Such cultures are isolated and used as a starter culture in the food industry because of providing standardisation of the final product during the food processing. Choice of starter culture is the most important step for the production of fermented food. Isolated LAB and yeast cultures which have the ability to create a biofilm layer can be preferred as a starter in the food industry. The biofilm formation could be beneficial to extend the period of usage time of microorganisms as a starter. On the other hand, it is an undesirable property in pathogens, since biofilm structure allows a microorganism become more resistant to stress conditions such as antibiotic presence. It is thought that the resistance mechanism could be turned into an advantage by promoting the effective microorganisms which are used in the food industry as starter culture and also which have potential to stimulate the gastrointestinal system. Development of the biofilm layer is observed in some LAB and yeast strains. The resistance could make LAB and yeast strains dominant microflora in the human gastrointestinal system; thus, competition against pathogen microorganisms can be provided more easily. Based on this circumstance, in the study, 10 LAB and 10 yeast strains were isolated from various dairy products, such as cheese, yoghurt, kefir, and cream. Samples were obtained from farmer markets and bazaars in Bursa, Turkey. As a part of this research, all isolated strains were identified and their ability of biofilm formation was detected with two different methods and compared with each other. The first goal of this research was to determine whether isolates have the potential for biofilm production, and the second was to compare the validity of two different methods, which are known as “Tube method” and “96-well plate-based method”. This study may offer an insight into developing a point of view about biofilm formation and its beneficial properties in LAB and yeast cultures used as a starter in the food industry.

Authors: P. Pinyaphong, S. Phutrakul
Abstract:
Palm oil could be converted to cocoa butter equivalent by lipase-catalyzed interesterification. The objective of this research was to investigate the structure modification of palm oil to cocoa butter equivalent using Carica papaya lipase –catalyzed interesterification. The study showed that the compositions of cocoa butter equivalent were affected by acyl donor sources, substrate ratio, initial water of enzyme, reaction time, reaction temperature and the amount of enzyme. Among three acyl donors tested (methyl stearate, ethyl stearate and stearic acid), methyl stearate appeared to be the best acyl donor for incorporation to palm oil structure. The best reaction conditions for cocoa butter equivalent production were : substrate ratio (palm oil : methyl stearate, mol/mol) at 1 : 4, water activity of enzyme at 0.11, reaction time at 4 h, reaction temperature at 45 ° C and 18% by weight of the enzyme. The chemical and physical properties of cocoa butter equivalent were 9.75 ± 0.41% free fatty acid, 44.89 ± 0.84 iodine number, 193.19 ± 0.78 sponification value and melting point at 37-39 °C.

Authors: Mekhaldi Abdelkader, Bouznad Ahcen, Djibaoui Rachid, Hamoum Hakim
Abstract:
This study presents an attempt to evaluate the antioxidant potential and antimicrobial activity of methanolic extract, and essential oils prepared from the leaves of sage (Salvia officinalis L.). The content of polyphenol in the methanolic extracts from the leaves of Salvia officinalis was determined spectrophotometrically, calculated as gallic acid and catechin equivalent. The essential oils and methanol extract were also subjected to screenings for the evaluation of their antioxidant activities using 2, 2-diphenyl-1- picrylhydrazyl (DPPH) test. While the plant essential oils showed only weak antioxidant activities, its methanol extract was considerably active in DPPH (IC50 = 37.29 μg/ml) test. Appreciable total polyphenol content (31.25 mg/g) was also detected for the plant methanol extract as gallic acid equivalent in the Folin–Ciocalteu test. The plant was also screened for its antimicrobial activity and good to moderate inhibitions were recorded for its essential oils, and methanol extracts against most of the tested microorganisms. The present investigation revealed that this plant had rich source of antioxidant properties. It is for this reason that sage has found increasing application in food formulations.
