39 The Induction of Antioxidant Enzyme Activities in Cabbage Seedlings by Heavy Metal Stress

Abstract:

Cabbage seedlings grown in vitro were exposed to excess levels of heavy metals, including Cd, Mo, and Zn. High metal levels affected plant growth at cotyledonary stage. Seedlings under Cd, Mo, and Zn treatments could not produce root hairs and true leaves. Under stress conditions, seedlings accumulated a higher amount of anthocyanins in their cotyledons than those in the control. The pigments isolated from Cd and Zn stressed seedling cotyledons appeared as pink, while under Mo stress, was dark pink or purple. Moreover, excess Mo stress increased antioxidant enzyme activities of APX, CAT, SOD. These results suggest that, under excess Mo stress, the induced antioxidant enzyme activity of cabbage seedlings may function as a protective mechanism to shield the plants from toxicity and exacerbated growth.

38 Effect of Shallow Groundwater Table on the Moisture Depletion Pattern in Crop Root Zone

Abstract:

Different techniques for estimating seasonal water use from soil profile water depletion frequently do not account for flux below the root zone. Shallow water table contribution to supply crop water use may be important in arid and semi-arid regions. Development of predictive root uptake models, under influence of shallow water table makes it possible for planners to incorporate interaction between water table and root zone into design of irrigation projects. A model for obtaining soil moisture depletion from root zone and water movement below it is discussed with the objective to determine impact of shallow water table on seasonal moisture depletion patterns under water table depth variation, up to the bottom of root zone. The role of different boundary conditions has also been considered. Three crops: Wheat (Triticum aestivum), Corn (Zea mays) and Potato (Solanum tuberosum), common in arid & semi-arid regions, are chosen for the study. Using experimentally obtained soil moisture depletion values for potential soil moisture conditions, moisture depletion patterns using a non linear root uptake model have been obtained for different water table depths. Comparative analysis of the moisture depletion patterns under these conditions show a wide difference in percent depletion from different layers of root zone particularly top and bottom layers with middle layers showing insignificant variation in moisture depletion values. Moisture depletion in top layer, when the water table rises to root zone increases by 19.7%, 22.9% & 28.2%, whereas decrease in bottom layer is 68.8%, 61.6% & 64.9% in case of wheat, corn & potato respectively. The paper also discusses the causes and consequences of increase in moisture depletion from top layers and exceptionally high reduction in bottom layer, and the possible remedies for the same. The numerical model developed for the study can be used to help formulating irrigation strategies for areas where shallow groundwater of questionable quality is an option for crop production.

37 Transcritical CO2 Heat Pump Simulation Model and Validation for Simultaneous Cooling and Heating

Abstract:

In the present study, a steady-state simulation model has been developed to evaluate the system performance of a transcritical carbon dioxide heat pump system for simultaneous water cooling and heating. Both the evaporator (including both two-phase and superheated zone) and gas cooler models consider the highly variable heat transfer characteristics of CO2 and pressure drop. The numerical simulation model of transcritical CO2 heat pump has been validated by test data obtained from experiments on the heat pump prototype. Comparison between the test results and the model prediction for system COP variation with compressor discharge pressure shows a modest agreement with a maximum deviation of 15% and the trends are fairly similar. Comparison for other operating parameters also shows fairly similar deviation between the test results and the model prediction. Finally, the simulation results are presented to study the effects of operating parameters such as, temperature of heat exchanger fluid at the inlet, discharge pressure, compressor speed on system performance of CO2 heat pump, suitable in a dairy plant where simultaneous cooling at 4oC and heating at 73oC are required. Results show that good heat transfer properties of CO2 for both two-phase and supercritical region and efficient compression process contribute a lot for high system COPs.

36 Privacy Concerns and Law Enforcement Data Collection to Tackle Domestic and Sexual Violence

Abstract:

It has been observed that violent or coercive behaviour has been apparent from initial conversations on dating apps like Tinder. Child pornography, stalking, and coercive control are some criminal offences from dating apps, including women murdered after finding partners through Tinder. Police databases and predictive policing are novel approaches taken to prevent crime before harm is done. This research will investigate how police databases can be used in a privacy-preserving way to characterise users in terms of their potential for violent crime. Using the COPS database of NSW Police, we will explore how the past criminal record can be interpreted to yield a category of potential danger for each dating app user. It is up to the judgement of each subscriber on what degree of the potential danger they are prepared to enter into. Sentiment analysis is an area where research into natural language processing has made great progress over the last decade. This research will investigate how sentiment analysis can be used to interpret interchanges between dating app users to detect manipulative or coercive sentiments. These can be used to alert law enforcement if continued for a defined number of communications. One of the potential problems of this approach is the potential prejudice a categorisation can cause. Another drawback is the possibility of misinterpreting communications and involving law enforcement without reason. The approach will be thoroughly tested with cross-checks by human readers who verify both the level of danger predicted by the interpretation of the criminal record and the sentiment detected from personal messages. Even if only a few violent crimes can be prevented, the approach will have a tangible value for real people.

35 Textile Dyeing with Natural Dye from Sappan Tree (Caesalpinia sappan Linn.) Extract

Abstract:

Natural dye extracted from Caesalpinia sappan Linn. was applied to a cotton fabric and silk yarn by dyeing process. The dyestuff component of Caesalpinia sappan Linn. was extracted using water and ethanol. Analytical studies such as UV–VIS spectrophotometry and gravimetric analysis were performed on the extracts. Brazilein, the major dyestuff component of Caesalpinia sappan Linn. was confirmed in both aqueous and ethanolic extracts by UV–VIS spectrum. The color of each dyed material was investigated in terms of the CIELAB (L*, a* and b*) and K/S values. Cotton fabric dyed without mordant had a shade of reddish-brown, while those post-mordanted with aluminum potassium sulfate, ferrous sulfate and copper sulfate produced a variety of wine red to dark purple color shades. Cotton fabric and silk yarn dyeing was studied using aluminum potassium sulfate as a mordant. The observed color strength was enhanced with increase in mordant concentration.

34 Bioremediation of Sewage Sludge Contaminated with Fluorene Using a Lipopeptide Biosurfactant

Abstract:

The disposal and the treatment of sewage sludge is an expensive and environmentally complex problem. In this work, a lipopeptide biosurfactant extracted from corn steep liquor was used as ecofriendly and cost-competitive alternative for the mobilization and bioremediation of fluorene in sewage sludge. Results have demonstrated that this biosurfactant has the capability to mobilize fluorene to the aqueous phase, reducing the amount of fluorene in the sewage sludge from 484.4 mg/Kg up to 413.7 mg/Kg and 196.0 mg/Kg after 1 and 27 days respectively. Furthermore, once the fluorene was extracted the lipopeptide biosurfactant contained in the aqueous phase allowed the biodegradation, up to 40.5% of the initial concentration of this polycyclic aromatic hydrocarbon.

33 Influence of Probiotics on Dairy Cows Diet

Abstract:

The main goal of this paper was evaluate the effect of diets containing different levels of probiotic on performance and milk composition of lactating cows. Eight Holstein cows were distributed in two 4x4 Latin square. The diets were based on corn silage, concentrate and the treatment (0, 3, 6 or 9 grams of probiotic/animal/day). It was evaluated the dry matter intake of nutrients, milk yield and composition. The use of probiotics did not affect the nutrient intake (p>0.05) neither the daily milk production or corrected to 4% fat (p>0.05). However, it was observed that there was a significant fall in milk composition with higher levels of probiotics supplementation. These results emphasize the need of further studies with different experimental designs or improve the number of Latin square with longer periods of adaptation.

32 Furniko Flour: An Emblematic Traditional Food of Greek Pontic Cuisine

Abstract:

Although the gastronomy of the Greeks of Pontus is well-known, it has not received the same level of scientific scrutiny as another Greek local cuisine, that of Crete. As a result, we planned to concentrate our research on Greek Pontic cuisine to shed light on its distinct recipes, food products, and, ultimately, its characteristics. The Greeks of Pontus have one of Greece's most distinguished local cuisines, having lived for a long time in the northern part (Black Sea Region) of modern Turkey and now widely inhabiting northern Greece. Despite its simplicity, their cuisine contains several mouthwatering delicacies. Even though they have been in Greece for a century, their gastronomic culture remains an important part of their collective identity. As a first step toward understanding Greek Pontic cuisine, furniko flour, one of its most well-known traditional products, was investigated. For this project, we targeted residents of Western Macedonia, a province in northern Greece with a large population of descendants of Pontus Greeks who are primarily engaged in agricultural activities. In this quest, we approached a descendant of Pontus Greeks who is involved in the production of furniko flour and agreed to show us the entire process as we participated in it. Furniko flour is made from heirloom non-hybrid corn. When the moisture content of the seeds is low enough to make them suitable for roasting, they are harvested by hand. Harvesting by hand entails removing the cob from the plant and separating the husks. The harvested cobs are then roasted in a traditional wood oven for 24 hours. After that, the these are collected and stored in sacks. The next step is to extract the seeds by rubbing the cobs together. Ideally, the seeds should be ground in a traditional stone hand mill. The outcome of this process is aromatic, dark golden furniko flour, which is used to make havitz. Along with the furniko flour preparation, we also documented the havitz cooking process (a porridge-like corn flour dish). One of the most delectable dishes in Greek Pontic cuisine, this savory delicacy is simple to prepare. Because of the ingredients of furniko flour, havitz is a highly nutritious dish, according to the research participant. Furthermore, he claims that preparing havitz is a wonderful way to bring families together, share stories, and revisit happy memories. Finally, as an initial effort to highlight elements of Pontic Greek cuisine, this study illustrates the traditional preparation of furniko flour and its use in various traditional recipes. Our next objective would be to evaluate the nutritional value of furniko flour by analyzing its chemical components.

31 Comparative Analysis between Corn and Ramon (Brosimum alicastrum) Starches to Be Used as Sustainable Bio-Based Plastics

Polymers from renewable resources have attracted an increasing amount of attention over the last two decades, predominantly due to two major reasons: firstly environmental concerns, and secondly the realization that our petroleum resources are finite. Finding new uses for agricultural commodities is also an important area of research. Therefore, it is crucial to get new sources of natural materials that can be used in different applications. Ramon tree (Brosimum alicastrum) is a tropical plant that grows freely in Yucatan countryside. This paper focuses on the seeds recollection, processing and starch extraction and characterization in order to find out about its suitability as biomaterial. Results demonstrated that it has a high content of qualities to be used not only as comestible but also as an important component in polymeric blends.

30 Sustainable Production of Oyster Mushroom (Pleurotus ostreatus) in Chiapas, Mexico

Abstract:

Pleurotus ostreatus is a common edible mushroom with a number of properties that can help to solve the nutritional and economical problems of people in Chiapas, Mexico. The objective of this project was to produce the mushroom under a sustainable management in which only regional products were allowed as a way to promote the cultivation and consumption of Pleurotus ostreatus; 5 different substrates were tested as well as 2 sanitation methods. The obtained results showed that the highest yields were obtained using corn husk and a thermal sanitation method. Pests and diseases were not a problem during the project but they appeared more in the substrates sanitized with calcium hydroxide.

29 Model-Driven and Data-Driven Approaches for Crop Yield Prediction: Analysis and Comparison

Abstract:

Crop yield prediction is a paramount issue in agriculture. The main idea of this paper is to find out efficient way to predict the yield of corn based meteorological records. The prediction models used in this paper can be classified into model-driven approaches and data-driven approaches, according to the different modeling methodologies. The model-driven approaches are based on crop mechanistic modeling. They describe crop growth in interaction with their environment as dynamical systems. But the calibration process of the dynamic system comes up with much difficulty, because it turns out to be a multidimensional non-convex optimization problem. An original contribution of this paper is to propose a statistical methodology, Multi-Scenarios Parameters Estimation (MSPE), for the parametrization of potentially complex mechanistic models from a new type of datasets (climatic data, final yield in many situations). It is tested with CORNFLO, a crop model for maize growth. On the other hand, the data-driven approach for yield prediction is free of the complex biophysical process. But it has some strict requirements about the dataset. A second contribution of the paper is the comparison of these model-driven methods with classical data-driven methods. For this purpose, we consider two classes of regression methods, methods derived from linear regression (Ridge and Lasso Regression, Principal Components Regression or Partial Least Squares Regression) and machine learning methods (Random Forest, k-Nearest Neighbor, Artificial Neural Network and SVM regression). The dataset consists of 720 records of corn yield at county scale provided by the United States Department of Agriculture (USDA) and the associated climatic data. A 5-folds cross-validation process and two accuracy metrics: root mean square error of prediction(RMSEP), mean absolute error of prediction(MAEP) were used to evaluate the crop prediction capacity. The results show that among the data-driven approaches, Random Forest is the most robust and generally achieves the best prediction error (MAEP 4.27%). It also outperforms our model-driven approach (MAEP 6.11%). However, the method to calibrate the mechanistic model from dataset easy to access offers several side-perspectives. The mechanistic model can potentially help to underline the stresses suffered by the crop or to identify the biological parameters of interest for breeding purposes. For this reason, an interesting perspective is to combine these two types of approaches.

28 Probiotic Properties of Lactic Acid Bacteria Isolated from Fermented Food

Abstract:

The objectives of this study were to isolate LAB from various sources, dietary supplement, Thai traditional fermented food, and freshwater fish and to characterize their potential as probiotic cultures. Out of 1,558 isolates, 730 were identified as LAB based on isolation on MRS agar supplemented with a bromocresol purple indicator&CaCO3 and Gram-positive, catalase- and oxidase-negative characteristics. Eight isolates showed the potential probiotic properties including tolerance to acid, bile salt & heat, proteolytic, amylolytic & lipolytic activities and oxalate-degrading capability. They all showed the antimicrobial activity against some Gram-negative and Gram-positive pathogenic bacteria. Based on 16S rDNA sequence analysis, they were identified as Enterococcus faecalis BT2 & MG30, Leconostoc mesenteroides SW64 and Pediococcus pentosaceous BD33, CF32, NP6, PS34 & SW5. The health beneficial effects and food safety will be further investigated and developed as a probiotic or protective culture used in Nile tilapia belly flap meat fermentation.

27 Characterization of Novel Atrazine-Degrading Klebsiella sp. isolated from Thai Agricultural Soil

Abstract:

Atrazine, a herbicide widely used in sugarcane and corn production, is a frequently detected groundwater contaminant. An atrazine-degrading bacterium, strain KB02, was obtained from long-term atrazine-treated sugarcane field soils in Kanchanaburi province of Thailand. Strain KB02 had a rod-to-coccus morphological cycle during growth. Sequence analysis of the PCR product indicated that the 16S rRNA gene in strain KB02 was ranging from 97-98% identical to the same region in Klebsiella sp. Based on biochemical, physiological analysis and 16S rDNA sequence analysis of one representative isolate, strain KB02, the isolates belong to the genus Klebsiella in the family Enterobacteriaceae. Interestingly that the various primers for atzA, B and C failed to amplify genomic DNA of strain KB02. Whereas the expected PCR product of atzA, B and C were obtained from the reference strain, Arthrobacter sp. strain KU001.

26 Effect of Restaurant Fat on Milk Yield and Composition of Dairy Cows Limit-Fed Concentrate Diet with Free Access to Forage

Abstract:

Ten lactating multiparous Holstein cows were used in a cross-over design with two dietary treatments and 28-d periods (with 14 d as an adaptation) to study the effect of restaurant fat on milk production and composition. Each cow was offered 14.7 kg DM /d of the basal concentrate diet based on barley and corn (crude protein = 17.7%, neutral detergent fiber = 23.5%, and acid detergent fiber = 5.8% of dry matter) with free access to alfalfa. Dietary treatments were arranged as supplying each cow with 0 (CONTROL) or 150 g/day (RF) of restaurant fat. Supplemental RF did not significantly (P > 0.25) affect milk yield, composition, and composition yields, except for milk fat contents. Milk fat contents were depressed (P < 0.05) with supplemental RF. Our results indicate that RF could depress milk fat without affecting milk yield and that the depression in milk fat in response to RF precedes the depression in milk yield.

25 Energy Evaluation and Utilization of Cassava Peel for Lactating Dairy Cows

Abstract:

The experiment was then conducted to investigate the effect of cassava peel addition in the concentrate on the performance of lactating dairy cows. Twenty four Holstein Friesian crossbred (>87.5% Holstein Friesian) lactating dairy cows in mid lactation; averaging 12.2+2.1 kg of milk, 119+45 days in milk, 44.1+6.2 months old and 449+33 kg live weight, were stratified for milk yield, days in milk, age, stage of lactation and body weight, and then randomly allocated to three treatment groups. The first, second and third groups were fed concentrates containing the respective cassava peel, 0, 20 and 40%. All cows were fed ad libitum corn silage and freely access to clean water. Dry matter intake, 4%FCM, milk composition and body weight change were affected (P<0.05) by the third treatments (40%). The present study indicated that 20% cassava peel can be used in the concentrate for lactating dairy cows.

24 Mathematical Modeling for Continuous Reactive Extrusion of Poly Lactic Acid formation by Ring Opening Polymerization Considering Metal/Organic Catalyst and Alternative Energies

Abstract:

PLA emerged as a promising polymer because of its property as a compostable, biodegradable thermoplastic made from renewable sources. PLA can be polymerized from monomers (Lactide or Lactic acid) obtained by fermentation processes from renewable sources such as corn starch or sugarcane. For PLA synthesis, ring opening polymerization (ROP) of Lactide monomer is one of the preferred methods. In the literature, the technique mainly developed for ROP of PLA is based on metal/bimetallic catalyst (Sn, Zn and Al) or other organic catalysts in suitable solvent. However, the PLA synthesized using such catalysts may contain trace elements of the catalyst which may cause toxicity. This work estimated the usefulness and drawbacks of using different catalysts as well as effect of alternative energies and future aspects for PLA production.

23 Aflatoxins Aggravate the Incidence of Salmonellosis Outbreak in Fattening Calves: A Case Study

Abstract:

Fever, bloody diarrhea and high mortality rate were the main clinical finding in a group of fattening calves. Analysis of corn silage revealed presence of aflatoxins at level of 370 ppb. This level of aflatoxins in the feed of cattle is somewhat low to be the main cause of reported signs. Leukocytopenia, anemia, decreased lymphocytic activity and lowered phagocytic index are the main hematological and immunological alterations in diseased calves. Bacteriological investigation revealed isolation of pathogenic Salmonella typhimurium from the rectal swabs of diseased calves. Our results suggested that, long duration of exposure to aflatoxins even at small concentrations may considered as predisposing factor for the incidence of natural infectious outbreaks as salmonellosis due to its immunosuppressive effect. We can conclude that the veterinarians and owners must be given an attention to the relation between aflatoxicosis and salmonellosis under field condition. We are recommended that the treatment program during similar outbreaks must be including anti-aflatoxins preparations beside the antimicrobial therapy.

22 Identification Characterization and Production of Phytase from Endophytic Fungi

Abstract:

Phytases are acid phosphatase enzymes, which efficiently cleave phosphate moieties from phytic acid, thereby generating myo-inositol and inorganic phosphate. Thirty four isolates of endophytic fungi to produce of phytases were isolated from leaf, stem and root fragments of soybean. Screening of 34 isolates of endophytic fungi identified the phytases produced by Rhizoctonia sp. and Fusarium verticillioides . The phytase production were the best induced by phytic acid and rice bran compared the others inducer in submerged fermentation medium used. The phytase produced by both Rhizoctonia sp. and F. verticillioides have pH optimum at 4.0 and 5.0 respectively. The characterization of phytase from Fusarium verticillioides showed that temperature optimum was 500C and stability until 600C, the pH optimum 5.0 and pH stability was 2.5 – 6.0, and substrate specificity were rice bran>soybean meal>corn> coconut cake, respectively.

21 Determination of Sea Transport Route for Staple Food Distribution to Achieve Food Security in the Eastern Indonesia

Abstract:

Effectiveness and efficiency of food distribution is necessary to maintain food security in a region. Food supply varies among regions depending on their production capacity; therefore, it is necessary to regulate food distribution. Sea transportation could play a great role in the food distribution system. To play this role and to support transportation needs in the Eastern Indonesia, sea transportation shall be supported by fleet which is adequate and reliable, both in terms of load and worthiness. This research uses Linear Programming (LP) method to analyze food distribution pattern in order to determine the optimal distribution system. In this research, transshipment points have been selected for regions in one province. Comparison between result of modeling and existing shipping route reveals that from 369 existing routes, 54 routes are used for transporting rice, corn, green bean, peanut, soybean, sweet potato, and cassava.

20 Utilization of Mustard Leaves (Brassica juncea) Powder for the Development of Cereal Based Extruded Snacks

Abstract:

Mustard leaves are rich in folates, vitamin A, K and B-complex. Mustard greens are low in calories and fats and rich in dietary fiber. They are rich in potassium, manganese, iron, copper, calcium, magnesium and low in sodium. It is very rich in antioxidants and Phytonutrients. For the optimization of process variables (moisture content and mustard leave powder), the experiments were conducted according to central composite Face Centered Composite design of RSM. The mustard leaves powder was replaced with composite flour (a combination of rice, chickpea and corn in the ratio of 70:15:15). The extrudate was extruded in a twin screw extruder at a barrel temperature of 120°C. The independent variables were mustard leaves powder (2-10 %) and moisture content (12-20 %). Responses analyzed were bulk density, water solubility index, water absorption index, lateral expansion, antioxidant activity, total phenolic content, and overall acceptability. The optimum conditions obtained were 7.19 g mustard leaves powder in 100g premix having 16.8% moisture content (w.b).

19 The Residual Effects of Different Doses of Atrazine+Alachlor and Foramsulfuron on the Growth and Physiology of Rapeseed (Brassica napus L.)

Abstract:

A pot experiment was carried out under controlled conditions to evaluate the residual effects of different doses of atrazine+alachlor and foramsulfuron used in corn fields on the growth and physiology of rapeseed (Brassica napus L.). A split-plot experiment in CRD with 4 replications was used. The main plots consisted of herbicide type (atrazine+alachlor mixture and foramsulfuron) and the sub-plots were different residual doses of the herbicides (0, 1%, 5%, 10%, 20%, 40%, 50% and 100%). 7 cm diameter pots were filled with a virgin soil and seeds of rapeseed cv. Hayola were planted in them. The pots were kept under controlled conditions for 8 weeks after germination. At harvest, the growth parameters and the chlorophyll contents of the leaves were determined. The results showed that the growth of rapeseed plants was completely prevented at the highest residual doses of the herbicides (50 and 100 %). The growth parameters of rapeseed plants were affected by all doses of both types of the herbicide as compared to the controls. The residual effects of atrazine+alachlor mixture in reducing the growth parameters of rapeseed were more pronounced as compared to the residual effects of foramsulfuron alone.

18 Isolation and Probiotic Characterization of Arsenic-Resistant Lactic Acid Bacteria for Uptaking Arsenic

Abstract:

The growing health hazardous impact of arsenic (As) contamination in environment is the impetus of the present investigation. Application of lactic acid bacteria (LAB) for the removal of toxic and heavy metals from water has been reported. This study was performed in order to isolate and characterize the Asresistant LAB from mud and sludge samples for using as efficient As uptaking probiotic. Isolation of As-resistant LAB colonies was performed by spread plate technique using bromocresol purple impregnated-MRS (BP-MRS) agar media provided with As @ 50 μg/ml. Isolated LAB were employed for probiotic characterization process, acid and bile tolerance, lactic acid production, antibacterial activity and antibiotic tolerance assays. After As-resistant and removal characterizations, the LAB were identified using 16S rDNA sequencing. A total of 103 isolates were identified as As-resistant strains of LAB. The survival of 6 strains (As99-1, As100-2, As101-3, As102-4, As105-7, and As112-9) was found after passing through the sequential probiotic characterizations. Resistant pattern pronounced hollow zones at As concentration >2000 μg/ml in As99-1, As100-2, and As101-3 LAB strains, whereas it was found at ~1000 μg/ml in rest 3 strains. Among 6 strains, the As uptake efficiency of As102-4 (0.006 μg/h/mg wet weight of cell) was higher (17 – 209%) compared to remaining LAB. 16S rDNA sequencing data of 3 (As99- 1, As100-2, and As101-3) and 3 (As102-4, As105-7, and As112-9) LAB strains clearly showed 97 to 99% (340 bp) homology to Pediococcus dextrinicus and Pediococcus acidilactici, respectively. Though, there was no correlation between the metal resistant and removal efficiency of LAB examined but identified elevated As removing LAB would probably be a potential As uptaking probiotic agent. Since present experiment concerned with only As removal from pure water, As removal and removal mechanism in natural condition of intestinal milieu should be assessed in future studies.

17 Development of Efficient Fungal Biomass-Degrading Enzyme Mixtures for Saccharification of Local Lignocellulosic Feedstock

Abstract:

Conversion of lignocellulosic biomass is the basis process for production of fuels, chemicals and materials in the sustainable biorefinery industry. Saccharification of lignocellulosic biomass is an essential step which produces sugars for further conversion to target value-added products e.g. bio-ethanol, bio-plastic, g-valerolactone (GVL), 5-hydroxymethylfuroic acid (HMF), levulinic acid, etc. The goal of this work was to develop an efficient enzyme for conversion of biomass to reducing sugar based on crude fungal enzyme from Chaetomium globosum BCC5776 produced by submerged fermentation and evaluate its activity comparing to a commercial Acremonium cellulase. Five local biomasses in Thailand: rice straw, sugarcane bagasse, corncobs, corn stovers, and palm empty fruit bunches were pretreated and hydrolyzed with varying enzyme loadings. Saccharification of the biomass led to different reducing sugar levels from 115 mg/g to 720 mg/g from different types of biomass using cellulase dosage of 9 FPU/g. The reducing sugar will be further employed as sugar feedstock for production of ethanol or commodity chemicals. This work demonstrated the use of promising enzyme candidate for conversion of local lignocellulosic biomass in biorefinery industry.

16 Preservation of Coconut Toddy Sediments as a Leavening Agent for Bakery Products

Abstract:

Toddy sediment (TS) was cultured in a PDA medium to determine initial yeast load, and also it was undergone sun, shade, solar, dehumidified cold air (DCA) and hot air oven (at 400, 500 and 60oC) drying with a view to preserve viability of yeast. Thereafter, this study was conducted according to two factor factorial design in order to determine best preservation method. Therein the dried TS from the best drying method was taken and divided into two portions. One portion was mixed with 3: 7 ratio of TS: rice flour and the mixture was divided in to two again. While one portion was kept under in house condition the other was in a refrigerator. Same procedure was followed to the rest portion of TS too but it was at the same ratio of corn flour. All treatments were vacuum packed in triple laminate pouches and the best preservation method was determined in terms of leavening index (LI). The TS obtained from the best preservation method was used to make foods (bread and hopper) and organoleptic properties of it were evaluated against same of ordinary foods using sensory panel with a five point hedonic scale. Results revealed that yeast load or fresh TS was 58×106 CFU/g. The best drying method in preserving viability of yeast was DCA because LI of this treatment (96%) is higher than that of other three treatments. Organoleptic properties of foods prepared from best preservation method are as same as ordinary foods according to Duo trio test.

15 Effect of Bacillus subtilis Pb6 on Growth and Gut Microflora in Clostridium perfringens Challenged Broilers

Abstract:

The objective of current study was to investigate the effect of Bacillus subtilis PB6 (CloSTAT) as a probiotic in broilers. The corn-soybean based diet was divided into four treatment groups; T1 (basal diet with no probiotic and no Clostridium perfringens); T2 (basal diet challenged with C. perfringens without probiotic); T3 (basal diet challenged with C. perfringens having 0.05% probiotic); T4 (basal diet challenged with C. perfringens having 0.1% probiotic). Every treatment group had four replicates with 24 birds each. Body weight and feed intake were measured on weekly basis, while ileal bacterial count was recorded on day-28 following Clostridium perfringens challenge. The 0.1% probiotic treatment showed 7.2% increase in average feed intake (P=0.05) and 8% increase in body weight compared to T2. In 0.1% treatment body weight was 5% higher than T3 (P=0.02). It was also observed that 0.1% treatment had improved feed conversion ratio (1.77) on 6th week. No effect of treatment was observed on mortality and ileal bacterial count. The current study indicated that 0.1% use of probiotic had positive response in C. perfringens challenged broilers.

14 Supplementation of Saccharomyces Cerevisiae or Lactobacillus Acidophilus in Goats Diets

Abstract:

This experiment was performed with the purpose of investigating effect of additional blend of probiotics Saccharomyces cerevisiae and Lactobacillus acidophilus on plasma fatty acid profiles particularly conjugated linoleic acid (CLA) in growing goats fed corn silage, and selected the optimal levels of the probiotics for further study. Twenty-four growing crossbred (Thai native x Anglo-Nubian) goats that weighed (14.2 ± 2.3) kg, aged about 6 months, were purchased and allocated to 4 treatments according to Randomized Complete Block Design (RCBD) with 6 goats in each treatment. The blocks were made by weight into heavy, medium, and light goats and each of the treatments contained two goats from each of the blocks. In the mean time, ruminal average pH unaffected, but the NH3-N and also plasma urea nitrogen (p<0.05), total volatile fatty acid (p>0.05) were raised, but propionic proportion (p<0.05) and butyric proportion (p>0.05) were reduced in concurrent with raise of acetic proportion and resultantly C2:C3 ratio (p>0.05). On plasma fatty acid profiles, total saturated fatty acids (p>0.05) was increased, and contrasted with decrease of C15:0 (p<0.01), C16:0 (p>0.05), and C18-C22 polyunsaturated fatty acids (p<0.05 or p<0.01). In addition, the experiment proved that the supplemented probiotics was in force for heightening CLA (p<0.01); for raising desirable fatty acids (p<0.05); for reducing ratio of PUFA: SFA (p>0.05) and for raising ratio of n6:n3 (p<0.05).

13 Oxidative and Hormonal Disruptions Underlie Bisphenol A - Induced Testicular Toxicity in Male Rabbits

Abstract:

The presence of endocrine-disrupting compounds, such as bisphenol A (BPA), in the environment can cause serious health problems. However, there are controversial opinions. This study investigated the reproductive, metabolic, oxidative and immunologic-disrupting effects of bisphenol A in male rabbits. Rabbits were divided into five groups. The first four rabbit groups were administered oral BPA (1, 10, 50, or 100 mg/kg/day) for ten weeks. The fifth group was administered corn oil as the vehicle. BPA significantly decreased serum testosterone, estradiol and the free androgen index (FAI) and significantly increased sex hormone binding globulin (SHBG) compared with the placebo group. The higher doses of BPA showed a significant decrease in follicular stimulating hormone (FSH) and luteinizing hormone (LH). A significant increase in blood glucose levels was identified in the BPA groups. The non-significant difference in insulin levels is a novel finding. The cumulative testicular toxicity of BPA was clearly demonstrated by the dose-dependent decrease in absolute testes weight, primary measures of semen quality and a significant increase in testicular malonaldehyde (MDA). Moreover, BPA significantly decreased total antioxidant capacity (TAC) and significantly increased immunoglobulin G (IgG) at the highest concentration. Our results suggest that BPA, especially at higher doses, is associated with many adverse effects on metabolism, oxidative stress, immunity, sperm quality and markers of androgenic action.

12 Co-Administration Effects of Conjugated Linoleic Acid and L-Carnitine on Weight Gain and Biochemical Profile in Diet Induced Obese Rats

Abstract:

Obesity as a global health challenge motivates pharmaceutical industries to produce anti-obesity drugs. However, effectiveness of these agents is remained unclear. Because of popularity of dietary supplements, the aim of this study was tp investigate the effects of Conjugated Linoleic Acid (CLA) and L-carnitine (LC) on serum glucose, triglyceride, cholesterol and weight changes in diet induced obese rats. 48 male Wistar rats were randomly divided into two groups: Normal fat diet (n=8), and High fat diet (HFD) (n=32). After eight weeks, the second group which was maintained on HFD until the end of study, was subdivided into four categories: a) 500 mg Corn Oil (as control group), b) 500 mg CLA, c) 200 mg LC, d) 500 mg CLA+ 200 mg LC.All doses are planned per kg body weights, which were administered by oral gavage for four weeks. Body weights were measured and recorded weekly by means of a digital scale. At the end of the study, blood samples were collected for biochemical markers measurement. SPSS Version 16 was used for statistical analysis. At the end of 8th week, a significant difference in weight was observed between HFD and NFD group. After 12 weeks, LC significantly reduced weight gain by 4.2%. Trend of weight gain in CLA and CLA+LC groups was insignificantly decelerated. CLA+LC reduced triglyceride level significantly, but just CLA had significant influence on total cholesterol and insignificant decreasing effect on FBS. Our results showed that an obesogenic diet in a relative short time led to obesity and dyslipidemia which can be modified by LC and CLA to some extent.

11 Supplementation of Annatto (Bixa orellana)-Derived δ-Tocotrienol Produced High Number of Morula through Increased Expression of 3-Phosphoinositide- Dependent Protein Kinase-1 (PDK1) in Mice

Abstract:

Several embryonic cellular mechanism including cell cycle, growth and apoptosis are regulated by phosphatidylinositol-3- kinase (PI3K)/Akt signaling pathway. The goal of present study is to determine the effects of annatto (Bixa orellana)-derived δ-tocotrienol (δ-TCT) on the regulations of PI3K/Akt genes in murine morula. Twenty four 6-8 week old (23-25g) female balb/c mice were randomly divided into four groups (G1-G4; n=6). Those groups were subjected to the following treatments for 7 consecutive days: G1 (control) received tocopherol stripped corn oil, G2 was given 60 mg/kg/day of δ-TCT mixture (contains 90% delta & 10% gamma isomers), G3 was given 60 mg/kg/day of pure δ-TCT (>98% purity) and G4 received 60 mg/kg/day α-TOC. On Day 8, females were superovulated with 5 IU Pregnant Mare’s Serum Gonadotropin (PMSG) for 48 hours followed with 5 IU human Chorionic Gonadotropin (hCG) before mated with males at the ratio of 1:1. Females were sacrificed by cervical dislocation for embryo collection 48 hours post-coitum. About fifty morulas from each group were used in the gene expression analyses using Affymetrix QuantiGene Plex 2.0 Assay. Present data showed a significant increase (p<0.05) in the average number (mean + SEM) of morula produced in G2 (27.32 + 0.23), G3 (25.42 + 0.21) and G4 (27.21 + 0.34) compared to control group (G1 – 14.61 + 0.25). This is parallel with the high expression of PDK1 gene with increase of 2.75-fold (G2), 3.07-fold (G3) and 3.59-fold (G4) compared to G1. From the present data, it can be concluded that supplementation with δ-TCT(s) and α-TOC induced high expression of PDK1 in G2-G4 which enhanced the PI3K/Akt signaling activity, resulting in the increased number of morula.

10 Influence of Environment-Friendly Organic Wastes on the Properties of Sandy Soil under Growing Zea mays L. in Arid Regions

Abstract:

Environment-friendly organic wastes of Brewers' spent grain, a byproduct of the brewing process, have recently used as soil amendment to improve soil fertility and plant production. In this work, treatments of 1% (T1) and 2% (T2) of spent grains, 1% (C1) and 2% (C2) of compost and mix of both sources (C1T1) were used and compared to the control for growing Zea mays L. on sandy soil under arid Mediterranean climate. Soils were previously incubated at 65% saturation capacity for a month. The most relevant soil physical and chemical parameters were analysed. Water holding capacity and soil organic matter (OM) increased significantly along the treatments with the highest values in T2. Soil pH decreased along the treatments and the lowest pH was in C1T1. Bicarbonate decreased by 69% in C1T1 comparing to control. Total nitrogen (TN) and available P varied significantly among all treatments and T2, C1T1 and C2 treatments increased 25, 17 and 11 folds in TN and 1.2, 0.6 and 0.3 folds in P, respectively related to control. Available K showed the highest values in C1T1. Soil micronutrients increased significantly along all treatments with the highest values in T2. After corn germination, significant variation was observed in the velocity of germination coefficients (VGC) among all treatments in the order of C1T1>T2>T1>C2>C1>control. The highest records of final germination and germination index were in C1T1 and T2. The spent grains may compensate deficiencies of macro and micronutrients in newly reclaimed sandy soils without adverse effects to sustain crop production with a rider that excessive or continuous use need to be circumvented.
