13990 Molecular Analysis of Somaclonal Variation in Tissue Culture Derived Bananas Using MSAP and SSR Marker
Authors: Emma K. Sales, Nilda G. Butardo
Abstract:
The project was undertaken to determine the effects of modified tissue culture protocols e.g. age of culture and hormone levels (2,4-D) in generating somaclonal variation. Moreover, the utility of molecular markers (SSR and MSAP) in sorting off types/somaclones were investigated. Results show that somaclonal variation is in effect due to prolonged subculture and high 2,4-D concentration. The resultant variation was observed to be due to high level of methylation events specifically cytosine methylation either at the internal or external cytosine and was identified by methylation sensitive amplification polymorphism (MSAP). Simple sequence repeats (SSR) on the other hand, was able to associate a marker to a trait of interest. These therefore, show that molecular markers can be an important tool in sorting out variation/mutants at an early stage. Keywords: methylation, MSAP, somaclones, SSR, subculture, 2, 4-D

Authors: W. Scott Jackson
Abstract:
This paper explains the declension of Puritan clerical power following the Great Migration up until when Massachusetts lost its charter in 1684. Historian Perry Miller argued that an overall declension in Puritan culture occurred during this period. However, that notion has been dispelled. There is a resurging field exploring declension in areas outside of Miller’s scope of Puritan culture. I determine that colonial New England existed as a functional theocracy by using Pierre Bourdieu’s theory of symbolic capital to explain clerical power through symbolic and religious misdirection and conversion. I explore civil and economic power struggles in colonial New England during the decades following the Great Migration to establish that Puritan culture did not largely decline. Instead, it was the Puritan clergy’s power that waned during this period. Keywords: Bourdieu, Historical Sociology, Symbolic Capital, Puritan

Authors: Melissa Valle
Abstract:
This paper draws upon the studies of visual culture and intersectionality to illuminate how visuality can naturalize social hierarchies. Through the use of iconography, it decodes the denotative, connotative and ideological meanings of symbols of ritualistic events in the context of the Colombian Atlantic Coast. An examination of such exceptional moments, i.e. of the spectacle, brings into focus how such performances are imbued with meaning by both the on-looker and the performer. Through an analysis of preexisting visuals (e.g., advertisements, social media) and visual materials produced by the researcher for the purpose of photo-elicitation interviews, this paper provides a contextual analysis of the ways in which three representations, popular during Colombian Atlantic coastal festivals (Negrita Puloy, Las Palenqueras, and El Son de Negro), have been historically, culturally and politically constituted. This work reveals that the visualizations are born out of and reproduce typifications systems heavily based upon race, gender, class, and ethnicity. Understanding the ways these categories are mutually constituted through the cultural practice of visual representation is essential to a more comprehensive understanding of the role such representation plays in the reproduction of social difference. Keywords: Colombia, festivals, intersectionality, visual culture

Authors: Alapa Peters Odugbo
Abstract:
The remarkable but fascinatingly intricate book 'The Lives of Jessie Sampter', by Sarah Imhoff, which describes Jessie Sampter's three different lives as a queer, a disabled person, and a Zionist, served as the main inspiration for this work. Her second chapter of Imhoff, which covers disability in-depth, inspired the focus of my study. This paper critically explores how culture, religion, and medicine contribute to and sustain discriminatory practices against people with disabilities in Nigeria. These practices include continued and often unchallenged stigmatization, unequal treatment, and denial of basic social services, employment prospects, and fundamental human rights. The paper makes crucial recommendations to help combat and eliminate these practices and negative perceptions toward people with disabilities in Nigeria, as well as to safeguard and promote their interests and rights. Keywords: disability, culture, religion, medicine

Authors: Prachi Choudhary, Jai Gopal Sharma
Abstract:
Microbial cultures determine the type of organism, its abundance in the tested sample, or both. It is one of the primary diagnostic methods of microbiology. It is used to determine the cause of infectious disease by letting the agent multiply in a predetermined medium. Different bacteria produce colonies that may be very distinct from the bacterial species that produced them. To culture any pathogen or microorganism, we should first know about the types of media used in microbiology for culturing. Sometimes sub culturing is also done in various microorganisms if some mixed growth is seen in culture. Nearly 3 types of culture media based on consistency – solid, semi-solid, and liquid (broth) media; are further explained in the report. Then, The Five I's approach is a method for locating, growing, observing, and characterizing microorganisms, including inoculation and incubation. Isolation, inspection, and identification. For identification of bacteria, we have to culture the sample like urine, sputum, blood, etc., on suitable media; there are different methods of culturing the bacteria or microbe like pour plate method, streak plate method, swabbing by needle, pipetting, inoculation by loop, spreading by spreader, etc. After this, we see the bacterial growth after incubation of 24 hours, then according to the growth of bacteria antibiotics susceptibility test is conducted; this is done for sensitive antibiotics or resistance to that bacteria, and also for knowing the name of bacteria. Various methods like the dilution method, disk diffusion method, E test, etc., do antibiotics susceptibility tests. After that, various medicines are provided to the patients according to antibiotic sensitivity and resistance. Keywords: inoculation, incubation, isolation, antibiotics suspectibility test, characterizing

Authors: Arif Sikander, Imran Saeed
Abstract:
The behavioral pattern and performance of airline pilots are influenced by the level of stress, their ethical decision-making ability and above all their leadership style as part of the Crew Management process. Cultural differences of pilots, especially while working in ex-country airlines, could influence the stress perception. Culture also influences ethical decision making. Leadership style is also a variable dimension, and pilots need to adapt to the cultural settings while flying with the local pilots as part of their team. Studies have found that age, education, gender, and management experience are statistically significant factors in ethical maturity. However, in the decades to come, more studies are required to validate the results over and over again; thereby, providing support for the validity of the Moral Development Theory. Leadership style plays a vital role in ethical decision making. This study is grounded in the Moral Development theory and seeks to analyze the styles of leadership of airline pilots related to ethical decision making and also the influence of the culture on their stress perception. The sample for the study included commercial pilots from a National Airline. It is expected that these results should provide useful input to the literature in the context of developing appropriate Talent Management strategies. The authors intend to extend this study (carried out in one country) to major national carriers (many countries) to be able to develop a ultimate framework on Talent Management which should serve as a benchmark for any international airline as most of them (e.g., Emirates, Etihad, Cathay Pacific, China Southern, etc.) are dependent on the supply of this scarce resource from outside countries. Keywords: ethics, leadership, pilot, stress

Authors: Hannah Jennica P. Ello, Regina Via G. Garcia
Abstract:
Films are famous expressions of art in the country. As an expression of art, it serves as a medium in which a culture is reflected. This paper studied how films reflected the Filipino culture. In this study, social semiotics was used to analyze the semiotic resources identified in the film. The films studied were 'Feng Shui', 'Sukob', and 'The Healing', which were three of the highest grossing horror films of Chito S. Roño. The objectives of the paper were (1) to identify the semiotic resources in the film, (2) to extract their meanings, and (3) to determine how these resources were perceived in the Filipino culture. The semiotic resources identified in each film are organized into three categories: color, practices and supernatural occurrences. Each semiotic resource is analyzed through the four dimensions of social semiotics, genre, style, modality, and discourse. For color, some of the semiotic resources identified are red, white and blue; for practices, Hagiolatry, and Mariolatry, faith healing and the belief in superstitions; and for supernatural occurrences, haunting ghosts, doppelganger attacks and returning from the dead were identified. The practices that are prominent in the films are Hagiolatry and Mariolatry, belief in feng shui and belief in faith healers and albularyos. The belief of these practices shows that Filipinos have a dual faith; belief in religion and a belief in superstitions. In short, Filipinos highly practice folk Catholicism and because of this, a mixture of different cultures can be seen, as having molded the Filipino culture to what it is today. Keywords: culture, film, semiotics, social semiotics

Authors: Elif Doğru, Bülent Kızılduman, Huriye İcil
Abstract:
Throughout history, Cyprus has been involved in various commercial and cultural relationships with different civilizations, owing to its strategic location. Particularly during the Late Bronze Age, Cyprus emerged as a significant region engaged in interactions with the Mycenaeans and other Mediterranean civilizations. Presently, findings from archaeological excavations provide valuable insights into Cyprus' cultural history and its connections with other civilizations. Painted Mycenaean ceramics discovered during the excavations at Kaleburnu-Kral Tepesi (Galinaporni-Vasili), dated to the Late Bronze Age in Cyprus, are considered significant archaeological findings that carry traces of the art and culture of that era, reflecting the island's commercial and cultural connections. Considering these findings, there is a need for archaeometric studies to aid in the understanding of the commercial and cultural ties at Kaleburnu-Kral Tepesi. In line with this need, analytical studies have been initiated concerning the provenance and production techniques of the Mycenaean ceramics discovered in the excavations at Kaleburnu-Kral Tepesi, dated to the Late Bronze Age. In the context of origin analysis studies, it is advocated that understanding the techniques and materials used for the figures and designs applied on Mycenaean ceramics would significantly contribute to a better comprehension of historical contexts. Hence, the adopted approach involves not only the analysis of the ceramic raw material but also the characterization of the pigments on the ceramics as a whole. In light of this, in addition to the studies aimed at determining the provenance and production techniques of the Mycenaean ceramic bodies, the characterization of the pigments used in the decorations of the relevant ceramics has been included in the research scope. Accordingly, this study aims to characterize the pigments used in the decorations of Mycenaean ceramics discovered at Kaleburnu-Kral Tepesi, dated to the Late Bronze Age. The X-Ray diffraction (XRD), Fourier Transform Infrared Spectroscopy (FTIR), and Scanning Electron Microscopy with Energy Dispersive X-ray Spectroscopy (SEM-EDX) methods have been employed to determine the surface morphology and chemical properties of the Mycenaean pigments. The characterization has been conducted through the combination of multiple analytical methods. The characterization of the pigments of Mycenaean ceramics aims to enhance the scientific perspective adopted for understanding the contributions of Mycenaean ceramics found in Cyprus to the island's culture, by providing scientific data on the types and origins of pigments used during the Late Bronze Age.Keywords: mycenaean, ceramic, provenance, pigment

Authors: Roman Kusaiko
Abstract:
For decades, remaking was one of the film industry’s main practices but has become vivid in recent years. The latest geopolitical developments, though, are becoming a new challenge for filmmakers regarding cultural landscapes and contextual differences. Deglobalization may also affect transnational remaking practices. Thus, these upcoming challenges can be addressed through the analysis of contemporary academic thought, primarily from adaptation and film studies and their understanding of the issues of transmediality and how it affects film remaking. However, the analysis would be insufficient without conducting case studies. This paper is part of broader research about transnational remaking practices and their cultural and contextual specifics. This paper aims to understand whether shifting medium affects remaking as a critical category and present case studies of the popular Hong Kong motion picture Infernal Affairs and its transition into the Japanese remake Double Face. Consequently, the analysis of their contextual distinctions will lead to the correct categorization of the transnational remakes allowing scholars and filmmakers to better understand the existing remaking practices and whether they affect the final result. Keywords: cinema, context, culture, films, remaking, transmediality

Authors: Siddhi Pedamkar, Reenu Singh
Abstract:
This report is based on how a culture transforms from one era to another era in commercial space. This transformation is observed in commercial as well as residential spaces. The spaces have specific color concepts, surface detailing furniture, and function-specific layouts. But the cultural impact is very rarely seen in commercial spaces, mostly because the interior is divine by function to a large extent. Information was collected from books and research papers. A quantitative survey was conducted to understand people's perceptions about the impact of culture on design entities and how culture dictates the different types of space and their character. The survey also highlights the impact of types of interior lighting, colour schemes, and furniture types on the interior environment. The questionnaire survey helped in framing design parameters for contemporary interior design. The design parameters are used to propose design options for new-age furniture that can be used in co-working spaces. For the new and contemporary working spaces, new age design furniture, interior elements such as visual partition, semi-visual partition, lighting, and layout can be transformed by cultural changes in the working style of people and organization. Keywords: commercial space, culture, environment, furniture, interior

Authors: Asif Hasan
Abstract:
Among the reported suitable mariculture species in Bangladesh, seabass and mullet are the two most popular candidates due to their high market values. Several field studies conducted on the culture of seabass in Bangladesh, it still remains a challenge to commercially grow this species due to its exclusive carnivorous nature. In contrast, the grey mullet (M. cephalus) is a fast-growing, omnivorous euryhaline fish that has shown excellent growth in many areas including South Asia. Choice of a sustainable aquaculture technique must consider the productivity and yield as well as their environmental suitability. This study was designed to elucidate the ecologically suitable culture technique of M. cephalus in brakishwater ponds by comparing the biotic and abiotic components of pond ecosystem. In addition to growth parameters (yield, ADG, SGR, weight gain, FCR), Physicochemical parameters (Temperature, DO, pH, salinity, TDS, transparency, ammonia, and Chlorophyll-a concentration) and biological community composition (phytoplankton, zooplankton and benthic macroinvertebrates) were investigated from ponds under Semi-intensive, Improve extensive and Traditional culture system. While temperature were similar in the three culture types, ponds under improve-extensive showed better environmental conditions with significantly higher mean DO and transparency, and lower TDS and Chlorophyll-a. The abundance of zooplankton, phytoplankton and benthic macroinvertebrates were apparently higher in semi-intensive ponds. The Analysis of Similarity (ANOSIM) suggested moderate difference in the planktonic community composition. While the fish growth parameters of M. cephalus and total yield did not differ significantly between three systems, M. cephalus yield (kg/decimal) was apparently higher in semi-intensive pond due to high stocking density and intensive feeding. The results suggested that the difference between the three systems were due to more efficient utilization of nutrients in improve extensive ponds which affected fish growth through trophic cascades. This study suggested that different culture system of M. cephalus is an alternative and more beneficial method owing to its ecological and economic benefits in brackishwater ponds. Keywords: Mugil cephalus, pond ecosystem, mariculture, fisheries management

Abstract:
An efficient protocol for simultaneous production of forskolin and rosmarinic acid in in vitro callus derived from the leaves of Coleus forskohlii Briq. has been developed. MS media was used for the establishment of cultures and NAA + 6-BA (1.0 ppm) was found best for callus growth. The callus was further subjected to treatment with various elicitor/precursors viz. chitosan, thidiazuron and methyl jasmonate to observe their effect on production of biomass and accumulation of secondary metabolites. The content of forskolin and rosmarinic acid were estimated by HPTLC, in comparison to natural explant which showed 2 fold and 10 fold rise in forskolin and rosmarinic acid content, respectively. Methy1 jasmonate 50 µM was found best for production of forskolin, whereas thidiazuron showed best results in the yield of rosmarinic acid, separately in static culture. However, combined treatment in suspension culture showed moderated effect for increase in secondary metabolites but the biomass increased significantly as compared to static culture. Keywords: plant tissue culture, secondary metabolites, coleus, forskolin, rosmarinic acid, HPTLC

Abstract:
Crop improvement through genetic engineering depends on effective and reproducible plant regeneration systems. Immature embryos are the most widely used explant source for in vitro regeneration in barley (Hordeum vulgare L.). However, immature embryos require the continuous growth of donor plants and the suitable stage for their culture is also certainly limited. On the other hand, mature embryos can be procured and stored easily; they can be studied throughout the year. In this study, an effective callus induction and plant regeneration were aimed to develop from mature embryos of different barley genotypes. The effect of medium (MS1 and MS2), auxin type (2,4-D, dicamba, picloram and 2,4,5-T) and concentrations (2, 4, 6 mg/l) on callus formation and effect of cytokinin type (TDZ, BAP) and concentrations (0.2, 0.5, 1.0 mg/l) on green plant regeneration were evaluated in mature embryo culture of barley. Callus and shoot formation was successful for all genotypes. By depending on genotype, MS1 is the best medium, 4 mg/l dicamba is the best growth regulator in the callus induction and MS1 is the best medium, 1 mg/l BAP is the best growth regulator in the shoot formation were determined. Keywords: barley, callus, embryo culture, mature embryo

Authors: Krisna Damayanti
Abstract:
Negative issue about the relationship between auditors and clients often heard. It arises in view of the rise of a variety of phenomena resulting from the audit practice of greed and do not appreciate the independence of the audit profession and professional code of ethics. It is a logical consequence of the practice of capitalism in accounting. The purpose of this paper would like to uncover the existing auditing practices in Indonesia, especially Java, which is associated with a strong influence of Javanese culture with reluctant/"shy", politely, "legowo", "ngemong" friendly, "not mentholo", "tepo seliro", "ngajeni", "acquiescent". The method used by interpretive approach that emphasizes the role of language, interpret and understand and see social reality as something other than a label, name or concept. Auditing practices in each country has a culture that will affect the standard set by those regulatory standards although there has been an adaptation of IAS. In Indonesia the majority of parties dominated by Javanesse racial regulators, so Java culture is embedded in every audit practices thus conditions in Java requires auditors to behave like that, sometimes interfere with standard Java code of conduct that must be executed by an auditor. Auditors who live in Java have the characters of Javanese culture that is hard to avoid in the audit practice. However, in practice, the auditor still are relevant in their profession. Keywords: auditors, java, character, profession, code of ethics, client

Abstract:
The Research Goal: With the growing demand for regenerative medicine, an effective mass cell culture process is required. In a repetitive subculture process for proliferating cells, preparing single cell suspension which does not contain any cell aggregates is highly required because cell aggregates often raise various undesirable phenomena, e.g., apoptosis and decrease of cell proliferation. Since cell aggregates often occur in cell suspension during conventional subculture processes, this study proposes a single cell sorter driven by a resonance vibration of a cell culture substrate. The Method and the Result: The single cell sorter is simply composed of a cell culture substrate and a glass pipe vertically placed against the cell culture substrate with a certain gap corresponding to a cell diameter. The cell culture substrate is made of biocompatible stainless steel with a piezoelectric ceramic disk glued to the bottom side. Applying AC voltage to the piezoelectric ceramic disk, an out-of-plane resonance vibration with a single nodal circle of the cell culture substrate can be excited at 5.5 kHz. By doing so, acoustic radiation force is emitted, and then cell suspension containing only single cells is pumped into the pipe and collected. This single cell sorter is effective to collect single cells selectively in spite of its quite simple structure. We collected C2C12 myoblast cell suspension by the single cell sorter with the vibration amplitude of 12 µmp-p and evaluated the ratio of single cells in number against the entire cells in the suspension. Additionally, we cultured the collected cells for 72 hrs and measured the number of cells after the cultivation in order to evaluate their proliferation. As a control sample, we also collected cell suspension by conventional pipetting, and evaluated the ratio of single cells and the number of cells after the 72-hour cultivation. The ratio of single cells in the cell suspension collected by the single cell sorter was 98.2%. This ratio was 9.6% higher than that collected by conventional pipetting (statistically significant). Moreover, the number of cells cultured for 72 hrs after the collection by the single cell sorter yielded statistically more cells than that collected by pipetting, resulting in a 13.6% increase in proliferated cells. These results suggest that the cell suspension collected by the single cell sorter driven by the resonance vibration hardly contains cell aggregates whose diameter is larger than the gap between the cell culture substrate and the pipe. Consequently, the cell suspension collected by the single cell sorter maintains high cell proliferation. Conclusions: In this study, we developed a single cell sorter capable of sorting and pumping single cells by a resonance vibration of a cell culture substrate. The experimental results show the single cell sorter collects single cell suspension which hardly contains cell aggregates. Furthermore, the collected cells show higher proliferation than that of cells collected by conventional pipetting. This means the resonance vibration of the cell culture substrate can benefit us with the increase in efficiency of mass cell culture process for clinical applications. Keywords: acoustic radiation force, cell proliferation, regenerative medicine, resonance vibration, single cell sorter
