71 Chitin Degradation in Pseudomonas fluorescens
Authors: Azhar Alhasawi, Vasu D. Appanna
Abstract:
Chitin, the second most abundant bio-polymer in nature after cellulose, composed of β (1→4) linked N-acetylglucosamine (GlcNAc), is a major structural component in the cell walls of fungi and the shells of crustaceans. Chitin and its derivatives are gaining importance of economic value due to its biological activity and its industrial and biomedical applications. There are several methods to hydrolyze chitin to NAG, but they are typically expensive and environmentally unfriendly. Chitinase which catalyzes the breakdown of chitin to NAG has received much attention owing to its various applications in biotechnology. The presented research examines the ability of the versatile soil microbe, Pseudomonas fluorescens grown in chitin medium to produce chitinase and a variety of value-added products under abiotic stress. We have found that with high pH, Pseudomonas fluorescens enable to metabolize chitin more than with neutral pH and the overexpression of chitinase was also increased. P-dimethylaminobenzaldehyde (DMAB) assay for NAG production will be monitored and a combination of sodium dodecyl polyacrylamide gels will be used to monitor the proteomic and metabolomic changes as a result of the abiotic stress. The bioreactor of chitinase will also be utilized. Keywords: Pseudomonas fluorescens, chitin, DMAB, chitinase

Authors: I. Alhajri, G. Zahedi, R. Alazmi, A. Akbari
Abstract:
In this paper, an Artificial Neural Network (ANN) was developed to predict Asphaltene Precipitation (AP) during the injection of carbon dioxide into crude oil reservoirs. In this study, the experimental data from six different oil fields were collected. Seventy percent of the data was used to develop the ANN model, and different ANN architectures were examined. A network with the Trainlm training algorithm was found to be the best network to estimate the AP. To check the validity of the proposed model, the model was used to predict the AP for the thirty percent of the data that was unevaluated. The Mean Square Error (MSE) of the prediction was 0.0018, which confirms the excellent prediction capability of the proposed model. In the second part of this study, the ANN model predictions were compared with modified Hirschberg model predictions. The ANN was found to provide more accurate estimates compared to the modified Hirschberg model. Finally, the proposed model was employed to examine the effect of different operating parameters during gas injection on the AP. It was found that the AP is mostly sensitive to the reservoir temperature. Furthermore, the carbon dioxide concentration in liquid phase increases the AP.Keywords: artificial neural network, asphaltene, CO2 injection, Hirschberg model, oil reservoirs

Authors: O. M. Lemine
Abstract:
The effect of milling parameters on the structural and magnetic properties of nickel powder was investigated. The samples were characterized by X-ray powder diffraction and vibrating sample magnetometer (VSM). The results did not reveal any phase change of nickel during the milling. The average crystallite size decreases with a prolongation of milling times, whereas the lattice parameters increase. The hysteresis loop reveals the intrinsic magnetic behaviour. It was observed an increase in the magnetization which can be correlated to the volume expansion showed by XRD results. Keywords: nickel powders, nanocrystallines, XRD, VSM

Abstract:
Bio-polyols are important components in polyurethane industries. The preliminary studies into the synthesis of bio-polyol products (epoxy-polyol and epoxyl-polyurethanes) from Jatropha curcas were investigated. The reactions were followed by both infrared and nuclear magnetic resonance. Physico-chemical characterisation of the samples for iodine value (IV), acid value (AV), saponification value (SV) and hydroxyl value (HV) were carried out. Thermal transitions of the products were studied by heating 5 mg of the sample from 20ºC to 800ºC and then cooling down to -500ºC on a differential scanning calorimeter (DSC). The preparation of epoxylpolyol and polyurethane from Jatropha curcas oil was smooth and efficient. Results of film and solubility properties revealed that coatings of Jatropha curcas epoxy-polyurethanes performed better with increased loading of toluylene 2, 4-diisocyanate (TDI) up to 2 wt% while their solvent resistance decreased beyond a TDI loading of 1.2 wt%. DSC analysis shows the epoxy-polyurethane to be less stable compared to the epoxy-polyol.Keywords: synthesis, epoxy-polyol, epoxy-polyurethane, jatropha curcas oil

Authors: L. Kloetzer, M. Poştaru, A. I. Galaction, D. Caşcaval
Abstract:
Gentamicin is an aminoglycoside antibiotic industrially obtained by biosynthesis of Micromonospora purpurea or echinospora, the product being a complex mixture of components with very similar structures. Among them, three exhibit the most important biological activity: gentamicins C1, C1a, C2, and C2a. The separation of gentamicin from the fermentation broths at industrial scale is rather difficult and it does not allow the fractionation of the complex mixture of gentamicins in order to increase the therapeutic activity of the product. The aim of our experiments is to analyze the possibility to selectively separate the less active gentamicin, namely gentamicin C1, from the biosynthetic mixture by reactive extraction with di-(2-ethylhexyl) phosphoric acid (D2EHPA) dissolved in dichloromethane, followed selective re-extraction of the most active gentamicins C1a, C2, and C2a. The experiments on the reactive extraction of gentamicins indicated the possibility to separate selectively the gentamicin C1 from the mixture obtained by biosynthesis. The extraction selectivity is positively influenced by increasing the pH-value of an aqueous solution and by using a D2EHPA concentration in organic phase closer to the value needed for an equimolecular ratio between the extractant and this gentamicin. For quantifying the selectivity of separation, the selectivity factor, calculated as the ratio between the degree of reactive extraction of gentamicin C1 and the overall extraction degree of gentamicins were used. The possibility to remove the gentamicin C1 at an extractant concentration of 10 g l-1 and pH = 8 is presented. In these conditions, it was obtained the maximum value of the selectivity factor of 2.14, which corresponds to the modification of the gentamicin C1 concentration from 31.92% in the biosynthetic mixture to 72% in the extract. The re-extraction of gentamicins C1, C1a, C2, and C2a with sulfuric acid from the extract previously obtained by reactive extraction (mixture A – extract obtained by non-selective reactive extraction; mixture B – extract obtained by selective reactive extraction) allows for separating selectively the most active gentamicins C1a, C2, and C2a. For recovering only the active gentamicins C1a, C2, and C2a, the re-extraction must be carried out at very low acid concentrations, far below those corresponding to the stoichiometry of its chemical reactions with these gentamicins. Therefore, the mixture resulted by re-extraction contained 92.6% gentamicins C1a, C2, and C2a. By bringing together the aqueous solutions obtained by reactive extraction and re-extraction, the overall content of the active gentamicins in the final product becomes 89%, their loss reaching 0.3% related to the initial biosynthetic product. Keywords: di-(2-ethylhexyl) phosphoric acid, gentamicin, reactive extraction, selectivity factor

Authors: Raghul Vignesh Kumar, M. Vadivel
Abstract:
With the recent years many industries and companies have turned their attention in realizing how going 'green' can benefit public relations, lower cost, and reduce global emissions from industrial manufacturing. Green Computing has become an originative way on how technology and ecology converge together. It is a growing import subject that creates an urgent need to train next generation computer scientists or practitioners to think ‘green’. However, green computing has not yet been well taught in computer science or computer engineering courses as a subject. In this modern IT world it’s impossible for an organization or common man to work without hardware like servers, desktop, IT devices, smartphones etc. But it is also important to consider the harmful impact of those devices and steps to achieve energy saving and environmental protection. This paper presents the magnitude of green computing and steps to be followed to go green. Keywords: green computing, carbon-dioxide, greenhouse gas, energy saving, environmental protection agency

Authors: Muhammad Naeem, Abdulrahman A. Al-Rabiah, Wasif Mughees
Abstract:
Technical mixture of C4 containing 1-butene and n-butane are very close to each other with regard to their boiling points i.e. -6.3°C for 1-butene and -1°C for n-butane. Extractive distillation process is used for the separation of 1-butene from the existing mixture of C4. The solvent is the essential of extractive distillation, and an appropriate solvent plays an important role in the process economy of extractive distillation. Aspen Plus has been applied for the separation of these hydrocarbons as a simulator. Moreover, NRTL activity coefficient model was used in the simulation. This model indicated that the material balances in this separation process were accurate for several solvent flow rates. Mixture of acetonitrile and water used as a solvent and 99% pure 1-butene was separated. This simulation proposed the ratio of the feed to solvent as 1: 7.9 and 15 plates for the solvent recovery column. Previously feed to solvent ratio was more than this and the number of proposed plates were 30, which shows that the separation process can be economized. Keywords: extractive distillation, 1-butene, aspen plus, ACN solvent

Authors: H. B. Rekha, Usha N. Murthy, Prashanth, Ashoka
Abstract:
Dyes are manufactured to have high chemical resistance because they are normally species, very difficult to degrade (reactive dyes). It damages flora and fauna. Furthermore, coloured components are highly hazardous. So removal of dyes becomes a challenge for both textile industry and water treatment facility. Dyeing wastewater is usually treated by conventional methods such as biological oxidation and adsorption but nowadays them becoming in-adequate because of large variability of composition of waste water. In the present investigation, mild steel electrodes of varying surface area were used for treatment of synthetic textile dye. It appears that electro-chemical coagulation could be very effective in removing coloured from wastewater; it could also be used to remove other parameters like chlorides, COD, and solids to some extent. In the present study, coloured removal up to 99% was obtained for surface area of mild steel electrode of 80 cm2 and 96% of surface area of mild steel electrode of 50 cm2. The findings from this study could be used to improve the design of electro-chemical treatment systems and modify existing systems to improve efficiency. Keywords: electrochemical coagulation, mild steel, colour, environmental engineering

Authors: Zita I. Šereš, Ljubica P. Dokić, Dragana M. Šoronja Simović, Cecilia Hodur, Zsuzsanna Laszlo, Ivana Nikolić, Nikola Maravić
Abstract:
In this work the concentration of steep-water from corn starch industry is monitored using ultrafiltration membrane. The aim was to examine the conditions of ultrafiltration of steep-water by applying the membrane of 2.5nm. The parameters that vary during the course of ultrafiltration, were the transmembrane pressure, flow rate, while the permeate flux and the dry matter content of permeate and retentive were the dependent parameter constantly monitored during the process. Experiments of ultrafiltration are conducted on the samples of steep-water, which were obtained from the starch wet milling plant Jabuka, Pancevo. The procedure of ultrafiltration on a single-channel 250mm length, with inner diameter of 6.8mm and outer diameter of 10mm membrane were carried on. The membrane is made of a-Al2O3 with TiO2 layer obtained from GEA (Germany). The experiments are carried out at a flow rate ranging from 100 to 200lh-1 and transmembrane pressure of 1-3 bars. During the experiments of steep-water ultrafiltration, the change of permeate flux, dry matter content of permeate and retentive, as well as the absorbance changes of the permeate and retentive were monitored. The experimental results showed that the maximum flux reaches about 40lm-2h-1. For responses obtained after experiments, a polynomial model of the second degree is established to evaluate and quantify the influence of the variables. The quadratic equitation fits with the experimental values, where the coefficient of determination for flux is 0.96. The dry matter content of the retentive is increased for about 6%, while the dry matter content of permeate was reduced for about 35-40%, respectively. During steep-water ultrafiltration in permeate stays 40% less dry matter compared to the feed. Keywords: ultrafiltration, steep-water, starch industry, ceramic membrane

Authors: Tareg M. Elsunaki, Suleiman A. Arafa, Mohamed A. Abd-Alla
Abstract:
New interesting thermal stable polyesters containing 1-methyl-4-piperidone moiety in the main chain have been synthesized. These polyesters were synthesized by interfacial polycondensation technique of 3,5-bis(4-hydroxybenzylidene)-1-methyl-4-piperidone (I) and 3,5-bis(4-hydroxy-3-methoxy benzyli-dene)-1-methyl-4-piperidone (II) with terphthaloyl, isophthaloyl, 4,4'-diphenic, adipoyl and sebacoyl dichlorides. The yield and the values of the reduced viscosity of the produced polyesters were found to be affected by the type of an organic phase. In order to characterize these polymers, the necessary model compounds (A), (B) were prepared from (I), (II) respectively and benzoyl chloride. The structure of monomers (I), (II), model compounds and resulting polyesters were confirmed by IR, elemental analysis and 1HNMR spectroscopy. The various characteristic of the resulting polymers including solubility, thermal properties, viscosity and X-ray analysis were also studied. Keywords: synthesis, characterization, new polyesters, chemistry
Procedia PDF Downloads 46360 Effect of Ramp Rate on the Preparation of Activated Carbon from Saudi Date Tree Fronds (Agro Waste) by Physical Activation Method
Authors: Muhammad Shoaib, Hassan M Al-Swaidan
Abstract:
Saudi Arabia is the major date producer in the world. In order to maximize the production from date tree, pruning of the date trees is required annually. Large amount of this agriculture waste material (palm tree fronds) is available in Saudi Arabia and considered as an ideal source as a precursor for production of activated carbon (AC). The single step procedure for the preparation of micro porous activated carbon (AC) from Saudi date tree fronds using mixture of gases (N2 and CO2) is carried out at carbonization/activation temperature at 850°C and at different ramp rates of 10, 20 and 30 degree per minute. Alloy 330 horizontal reactor is used for tube furnace. Flow rate of nitrogen and carbon dioxide gases are kept at 150 ml/min and 50 ml/min respectively during the preparation. Characterization results reveal that the BET surface area, pore volume, and average pore diameter of the resulting activated carbon generally decreases with the increase in ramp rate. The activated carbon prepared at a ramp rate of 10 degrees/minute attains larger surface area and can offer higher potential to produce activated carbon of greater adsorption capacity from agriculture wastes such as date fronds. The BET surface areas of the activated carbons prepared at a ramp rate of 10, 20 and 30 degree/minute after 30 minutes activation time are 1094, 1020 and 515 m2/g, respectively. Scanning electron microscopy (SEM) for surface morphology, and FTIR for functional groups was carried out that also verified the same trend. Moreover, by increasing the ramp rate from 10 and 20 degrees/min the yield remains same, i.e. 18%, whereas at a ramp rate of 30 degrees/min the yield increases from 18 to 20%. Thus, it is feasible to produce high-quality micro porous activated carbon from date frond agro waste using N2 carbonization followed by physical activation with CO2 and N2 mixture. This micro porous activated carbon can be used as adsorbent of heavy metals from wastewater, NOx SOx emission adsorption from ambient air and electricity generation plants, purification of gases, sewage treatment and many other applications. Keywords: activated carbon, date tree fronds, agricultural waste, applied chemistry

Authors: Chih Yuan Chen, Chien Chon Chen, Jin-Shyong Lin
Abstract:
The microstructure characterization of tempered nanocrystalline bainitic steel is investigated in the present study. It is found that two types of plastic relaxation, dislocation debris and nanotwin, occurs in the displacive transformation due to relatively low transformation temperature and high carbon content. Because most carbon atoms trap in the dislocation, high dislocation density can be sustained during the tempering process. More carbides only can be found in the high tempered temperature due to intense recovery progression. Keywords: nanostructure bainitic steel, tempered, TEM, nano-twin, dislocation debris, accommodation

Authors: Zue Chin Chang, Shih-Chang Liang
Abstract:
RF magnetron sputtering is used on the ceramic targets, each of which contains zinc oxide (ZnO), zinc oxide doped with aluminum (AZO) and zinc oxide doped with gallium (GZO). The electric conduction mechanism of the AZO and GZO films came mainly from the Al and Ga, the oxygen vacancies, Zn interstitial atoms, and Al and/or Ga interstitial atoms. AZO and GZO films achieved higher conduction than did ZnO film, it being ion vacant and nonstoichiometric. The XRD analysis showed a preferred orientation along the (002) plane for ZnO, AZO, and GZO films. Keywords: ZnO, AZO, GZO, doped, sputtering

Abstract:
In recent years, medical science has improved chemotherapy, radiation therapy and adjuvant therapy and has developed newer targeted therapies as well as refining surgical techniques for removing cancer. However, the chances of surviving the disease depend greatly on the type and location of the cancer and the extent of the disease at the start of treatment. Moreover, mainstream forms of cancer treatment have side effects which range from the unpleasant to the fatal. Therefore, the continuation of progress in anti-cancer therapy may depend on placing emphasis on other existing but less thoroughly investigated therapeutic approaches such as Sonodynamic Therapy (SDT). SDT is based on the local activation of a so called 'sonosensitizer', a molecule able to be excited by ultrasound, the radical production as a consequence of its relaxation processes and cell death due to different mechanisms induced by radical production. The present work deals with synthesis, characterization and preliminary in vitro test of Single Walled Carbon Nanotubes (SWCNT) decorated with porphyrins and biological vectors. The SWCNT’s surface was modified exploiting 1, 3-dipolar cycloaddition or Dies Alder reactions. For this purpose, different porphyrins scaffolds were ad-hoc synthesized using also non-conventional techniques. To increase cellular specificity of porphyrin-conjugated SWCNTs and to improve their ability to be suspended in aqueous solution, the modified nano-tubes were grafted with suitable glutamine or hyaluronic acid derivatives. These nano-sized sonosensitizers were characterized by several methodologies and tested in vitro on different cancer cell lines. Keywords: sonodynamic therapy, porphyrins synthesis and modification, SWNCT grafting, hyaluronic acid, anti-cancer treatment

Authors: Ali A. Mutair
Abstract:
The concentration levels of iron (Fe), copper (Cu), zinc (Zn), cadmium (Cd) and lead (Pb) in different cigarette brands commonly produced and sold in Yemen were determined. Convenient sample treatment for cigarette tobacco of freshly opened packs was achieved by a sample preparation method based on dry digestion, and the concentrations of the analysed metals were measured by Flame Atomic Absorption Spectrometry (FAAS). The mean values obtained for Fe, Zn, Cu, Cd, and Pb in different Yemeni cigarette tobacco were 311, 52.2, 10.11, 1.71 and 4.06 µg/g dry weight, respectively. There is no more significant difference among cigarette brands tested. It was found that Fe was at the highest concentration, followed by Zn, Cu, Pb and Cd. The average relative standard deviation (RSD) ranged from 1.77% to 19.34%. The accuracy and precision of the results were checked by blank and recovery tests. The results show that Yemeni cigarettes contain heavy metal concentration levels that are similar to those in foreign cigarette brands reported by other studies in the worldwide. Keywords: iron, zinc, copper, lead, cadmium, tobacco, Yemeni cigarette brands, atomic absorption spectrometry

Abstract:
Cesiumiodide with Na doping (CsI(Na)) solution or melt is easily forming three- dimension dendrites on the free surface. The defects or bobbles form inside the CsI(Na) during the solution or melt solidification. The defects or bobbles can further effect the x-ray path in the CsI(Na) crystal and decrease the scintillation characteristics of CsI(Na). In order to enhance the CsI(Na) scintillated property we made single crystal of CsI(Na) column in the anodic aluminum oxide (AAO) template by hydraulic pressure injection method. It is interesting that when CsI(Na) melt is confined in the small AAO channels, the column grow as stable single column without any dendrites. The high aspect ratio (100~10000) of AAO and nano to sub-micron channel structure which is a suitable template for single of crystal CsI(Na) formation. In this work, a new low-cost approach to fabricate scintillator crystals using anodic aluminum oxide (AAO) rather than Si is reported, which can produce scintillator crystals with a wide range of controllable size to optimize their performance in X-ray detection. Keywords: cesiumiodide, AAO, scintillator, crystal, X-ray

Authors: M. E. Ali Mohsin, Agus Arsad, Othman Y. Alothman
Abstract:
This study explains the influence of secondary filler on the dispersion of carbon nanotube (CNT) reinforced high density polyethylene (HDPE) nanocomposites (CNT/HDPE). In order to understand the mixed-fillers system, Montmorillonite (MMT) was added to CNT/HDPE nanocomposites. It was followed by investigating their effect on the thermal, mechanical and morphological properties of the aforesaid nanocomposite. Incorporation of 3 wt% each of MMT into CNT/HDPE nanocomposite resulted to the increased values for the tensile and flexural strength, as compared to the pure HDPE matrix. The thermal analysis result showed improved thermal stability of the formulated nanocomposites. Transmission electron microscopy (TEM) images revealed that larger aggregates of CNTs were disappeared upon addition of these two components leading to the enhancement of thermo-mechanical properties for such composites. Keywords: secondary filler, montmorillonite, carbon nanotube, nanocomposite
Procedia PDF Downloads 36849 Achievement of High L-Cysteine Yield from Enzymatic Conversion Using Eutectic Mixtures of the Substrate ATC
Authors: Deokyeong Choe, Sung Hun Youn, Younggon Kim, Chul Soo Shin
Abstract:
L-Cysteine, a sulfur-containing amino acid, has been often used in the pharmaceutical, cosmetic, food, and feed additive industries. This amino acid has been usually produced by acid-hydrolysis of human hair and poultry feathers. There are many problems, such as avoidance for use of animal hair, low yields, and formation of harmful waste material. As an alternative, the enzymatic conversion of D, L-2-amino-Δ2-thiazoline-4-carboxylic acid (ATC) to L-cysteine has been developed as an environmental-friendly method. However, the substrate solubility was too low to be used in industry. In this study, high concentrations of eutectic substrate solutions were prepared to solve the problem. Eutectic melting occurred at 39°C after mixing ATC and malonic acid at a molar ratio of 1:1. The characteristics of eutectic mixtures were analyzed by FE-SEM, EDS mapping, and XPS. However, since sorbitol, MnSO4, and NaOH should be added as supplements to the substrate mixture for the activation and stabilization of the enzyme, strategies for sequential addition of total five compounds, ATC, malonic acid, sorbitol, MnSO4, and NaOH were established. As a result, eutectic substrate mixtures of 670 mM ATC were successfully formulated. After 6 h of enzymatic reaction, 550 mM L-cysteine was made. Keywords: D, L-2-amino-Δ2-thiazoline-4-carboxylicacid, enzymatic conversion, eutectic solution, l-cysteine
Procedia PDF Downloads 42948 High Dissolution of ATC by pH Control and Its Enzymatic Conversion to L-Cysteine
Authors: Deokyeong Choe, Sung Hun Youn, Younggon Kim, Chul Soo Shin
Abstract:
L-Cysteine is extensively used as a supplement of pharmaceuticals, cosmetics, food and feed additives. It has obtained industrially by hydrolysis of human hair and poultry feathers. However, there are some problems such as the restriction of using materials from animals and the intractable waste pollution. The enzymatic conversion has been regarded as an environmental-friendly method. Currently, the biggest bottle-neck of enzymatic conversion is the low yield of L-cysteine due to the low substrate solubility. In this study, the method of enhancing the solubility of the substrate D,L-2-amino-Δ2-thiazoline-4-carboxylicacid (ATC) was developed and the enzymatic reaction at high concentration levels was performed. A large amount of substrate in aqueous solutions was dissolved by pH control using salts. As the pH of the solution increased, the solubility of ATC increased. It was thought that a shift of ATC from acid form (-COOH) to dissociated carboxylic group (-COO-) would improve its hydrophilicity leading to solubility increase. The highest solubility of ATC was 610 mM at pH 10.5, whereas the maximum reaction rate was obtained at pH 8.3. As a result, a high L-cysteine yield of 250 mM was achieved at pH 9.1, which was obtained from a combination of optimum pH conditions for ATC solubility and enzymatic conversion. This yield corresponds to approximately 18 times of that in previous reports. Keywords: D, L-2-amino-Δ2-thiazoline-4-carboxylicacid, enzymatic conversion, high-substrate solubilization, L-Cysteine

Abstract:
Carbon dioxide removal from natural gas is an important process because the existence of carbon dioxide in natural gas contributes to pipeline corrosion, reduces the heating value, and takes up volume in the pipeline. In this study, bacterial cellulose was chosen for the CO2/CH4 gas separation membrane due to its unique structure and prominent properties. Additionally, it can simply be obtained by culturing the bacteria so called “Acetobacter xylinum” through fermentation of coconut juice. Bacterial cellulose membranes with and without silver ions were prepared and studied for the separation performance of CO2 and CH4.Keywords: bacterial cellulose, CO2, CH4 separation, membrane, nata de coco

Abstract:
Steam reforming is industrially important as it is incorporated in several major chemical processes including the production of ammonia, methanol, hydrogen and ox alcohols. Due to the strongly endothermic nature of the process, a large amount of heat is supplied by fuel burning (commonly natural gas) in the furnace chamber. Reaction conversions, tube catalyst life, energy consumption and CO2 emission represent the principal factors affecting the performance of this unit and are directly influenced by the high operating temperatures and pressures. This study presents a simulation of the performance of the reforming of methane in a primary reformer, through a developed empirical relation which enables to investigate the effects of operating parameters such as the pressure, temperature, steam to carbon ratio on the production of hydrogen, as well as the fraction of non-converted methane. It appears from this analysis that the exit temperature Te, the operating pressure as well the steam to carbon ratio has an important effect on the reforming of methane. Keywords: reforming, methane, performance, hydrogen, parameters

Authors: Ahmad I. Ayesh
Abstract:
This work reports on the fabrication of tin nanoclusters by sputtering and inert-gas condensation inside an ultra-high vacuum compatible system. This technique allows to fine tune the size and yield of nanoclusters by controlling the nanocluster source parameters. The produced nanoclusters are deposited on SiO2/Si substrate with pre-formed electrical electrodes to produce a nanocluster device. Those devices can be potentially used for gas sensor applications. Keywords: tin, nanoclusters, inert-gas condensation, nanotechnology

Abstract:
Whey acidic proteins (WAP) domain-containing proteins in crustacean are involved in innate immune response against microbial invasion. In the present study, a novel double WAP domain (DWD)-containing protein gene 3 was identified from Chinese mitten crab Eriocheir sinensis (designated EsDWD3) by expressed sequence tag (EST) analysis and PCR techniques. The full-length cDNA of EsDWD3 was of 1223 bp, consisting of a 5′-terminal untranslated region (UTR) of 74 bp, a 3′ UTR of 727 bp with a polyadenylation signal sequence AATAAA and a polyA tail, and an open reading frame (ORF) of 423 bp. The ORF encoded a polypeptide of 140 amino acids with a signal peptide of 22 amino acids. The deduced protein sequence EsDWD3 showed 96.4 % amino acid similar to other reported EsDWD1 from E. sinensis, and phylogenetic tree analysis revealed that EsDWD3 had closer relationships with the reported two double WAP domain-containing proteins of E. sinensis species. Keywords: Chinese mitten crab, Eriocheir sinensis, cloning, double WAP domain-containing protein

Authors: Fengmei Li, Li Xu, Guoliang Xia
Abstract:
Lysozyme is a catalytic enzyme that performs bacterial cell lysis by cleaving the β-1,4-glycosidic bond between N-acetylmuramic acid and N-acetylglucosamine of peptidoglycan in cell walls. In the present study, an invertebrate-type (i-type) lysozyme gene was cloned from Chinese mitten crab Eriocheir sinensis (designated as EsLysozyme) based on PCR-based rapid amplification of cDNA ends (RACE) technology. The full-length cDNA of EsLysozyme was of 831 bp. SMART and SIGNALP 3.0 program analysis revealed that EsLysozyme contained a signal peptide and a destabilase domain. The five amino acid residues (Tyr63, Trp64, Tyr91, His110, Pro114) and the conserved motif GSLSCG(P/Y)FQI and CL(E/L/R/H)C(I/M)C in i-type lysozymes were also found in EsLysozyme. The high similarity of EsLysozyme with L. vannamei lysozymes and phylogenetic analysis suggested that EsLysozyme should be a new member of i-type lysozyme family. Keywords: i-type lysozyme, Eriocheir sinensis, cloning, characterization

Abstract:
Liquid-Liquid Equilibrium (LLE) data are measured for the ternary mixtures of water + 1-butanol + butyl acetate and quaternary mixtures of water + 1-butanol + butyl acetate + glycerol at atmospheric pressure at 313.15 K. In addition, isothermal Vapor–Liquid–Liquid Equilibrium (VLLE) data are determined experimentally at 333.15 K. The region of heterogeneity is found to increase as the hydrophilic agent (glycerol) is introduced into the aqueous mixtures. The experimental data are correlated with the NRTL model. The predicted results from the solution model with the model parameters determined from the constituent binaries are also compared with the experimental values. Keywords: LLE, VLLE, hydrophilic agent, NRTL
