866 Identification Characterization and Production of Phytase from Endophytic Fungi

Abstract:
Phytases are acid phosphatase enzymes, which efficiently cleave phosphate moieties from phytic acid, thereby generating myo-inositol and inorganic phosphate. Thirty four isolates of endophytic fungi to produce of phytases were isolated from leaf, stem and root fragments of soybean. Screening of 34 isolates of endophytic fungi identified the phytases produced by Rhizoctonia sp. and Fusarium verticillioides . The phytase production were the best induced by phytic acid and rice bran compared the others inducer in submerged fermentation medium used. The phytase produced by both Rhizoctonia sp. and F. verticillioides have pH optimum at 4.0 and 5.0 respectively. The characterization of phytase from Fusarium verticillioides showed that temperature optimum was 500C and stability until 600C, the pH optimum 5.0 and pH stability was 2.5 – 6.0, and substrate specificity were rice bran>soybean meal>corn> coconut cake, respectively.Keywords: endophytic fungus, phytase, soybean, Rhizoctoniasp., Fusarium verticillioides,

Abstract:
This paper represents the results of experimental work to investigate the suitability of a waste material (WM) for soft soil stabilisation. In addition, the effect of particle size distribution (PSD) of the waste material on its performance as a soil stabiliser was investigated. The WM used in this study is produced from the incineration processes in domestic energy power plant and it is available in two different grades of fineness (coarse waste material (CWM) and fine waste material (FWM)). An intermediate plasticity silty clayey soil with medium organic matter content has been used in this study. The suitability of the CWM and FWM to improve the physical and engineering properties of the selected soil was evaluated dependant on the results obtained from the consistency limits, compaction characteristics (optimum moisture content (OMC) and maximum dry density (MDD)); along with the unconfined compressive strength test (UCS). Different percentages of CWM were added to the soft soil (3, 6, 9, 12 and 15%) to produce various admixtures. Then the UCS test was carried out on specimens under different curing periods (zero, 7, 14, and 28 days) to find the optimum percentage of CWM. The optimum and other two percentages (either side of the optimum content) were used for FWM to evaluate the effect of the fineness of the WM on UCS of the stabilised soil. Results indicated that both types of the WM used in this study improved the physical properties of the soft soil where the index of plasticity (IP) was decreased significantly. IP was decreased from 21 to 13.64 and 13.10 with 12% of CWM and 15% of FWM respectively. The results of the unconfined compressive strength test indicated that 12% of CWM was the optimum and this percentage developed the UCS value from 202kPa to 500kPa for 28 days cured samples, which is equal, approximately 2.5 times the UCS value for untreated soil. Moreover, this percentage provided 1.4 times the value of UCS for stabilized soil-CWA by using FWM which recorded just under 700kPa after 28 days curing.

This paper presents numerical flow and slope stability simulations in three typical sections of earth dams built in tropical regions, two homogeneous with different slope inclinations, and the other one heterogeneous with impermeable core. The geotechnical material parameters used in this work were obtained from a lab testing of physical characterization, compaction, consolidation, variable load permeability and saturated triaxial type CD for compacted soil samples with standard proctor energy at optimum moisture content (23%), optimum moisture content + 2% and optimum moisture content +5%. The objective is to analyze the general behavior of earth dams built in rainy regions where optimum moisture is exceeded. The factor of safety is satisfactory for the three sections compacted in all moisture content during the stages of operation and end of construction. On The other hand, the rapid drawdown condition is the critical phase for homogeneus dams configuration, the factor of safety obtained were unsatisfactory. In general, the heterogeneous dam behavior is more efficient due to the fact that the slopes are made up of gravel, which favors the dissipation of pore pressures during the rapid drawdown. For the critical phase, the slopes should have lower inclinations of the upstream and downstream slopes to guarantee stability, although it increases the costs.

Abstract:
Laboratory trial results of mixing crumb rubber produced from discarded tires with 60/70 pen grade Kuwaiti bitumen are presented on this paper. PG grading and multiple stress creep recovery tests were conducted on Kuwaiti bitumen blended with 15% and 18% crumb rubber at temperatures ranging from 40 to 70 °C. The results from elastic recovery and non-recoverable creep presented optimum performance at 18% rubber content. The optimum rubberized-bitumen mix was next transformed into a pelletized form (PelletPave®), and was used as a partial replacement to the conventional bitumen in the manufacture of continuously graded hot mix asphalts at a number of binder contents. The trialed PelletPave® contents were at 2.5%, 3.0%, and 3.5% by mass of asphalt mix. In this investigation, it was not possible to utilize the results of standard Marshall method of mix design (i.e. volumetric, stability and flow tests) and subsequently additional assessment of mix compactability was carried out using gyratory compactor in order to determine the optimum PelletPave® and total binder contents.

Abstract:
The advantage of using non-linear passive damping system in vibration control of two adjacent structures is investigated under their base excitation. The base excitation is El Centro earthquake record acceleration. The damping system is considered as an optimum and effective non-linear viscous damper that is connected between two adjacent structures. A MATLAB program is developed to produce the stiffness and damping matrices and to determine a time history analysis of the dynamic motion of the system. One structure is assumed to be flexible while the other has a rule as laterally supporting structure with rigid frames. The response of the structure has been calculated and the non-linear damping coefficient is determined using optimum LQR algorithm in an optimum vibration control system. The non-linear parameter of damping system is estimated and it has shown a significant advantage of application of this system device for vibration control of two adjacent tall building.

Abstract:
A combination of photosynthetic bacteria along with anaerobic acidogenic bacteria is an ideal option for efficient hydrogen production. In the present study, the optimum concentration of substrates for the growth of Rhodobacter sphaeroides was found by response surface methodology. The optimum combination of three individual fatty acids was determined by Box Behnken design. Increase of volatile fatty acid concentration decreased the growth. Combination of sodium acetate and sodium propionate was most significant for the growth of the organism. The results showed that a maximum biomass concentration of 0.916 g/l was obtained when the concentrations of acetate, propionate and butyrate were 0.73g/l,0.99g/l and 0.799g/l, respectively. The growth was studied under an optimum concentration of volatile fatty acids and at a light intensity of 3000 lux, initial pH of 7 and a temperature of 35°C.The maximum biomass concentration of 0.92g/l was obtained which verified the practicability of this optimization.Keywords: Biohydrogen, Response Surface Methodology, Rhodobacter sphaeroides, Volatile fatty acid

Abstract:
This paper aims at determining the soil characteristics that influence the irrigation process of green landscapes and deciding on the optimum amount of water needed for irrigation. The laboratory experiments were conducted using the constant head methodology to determine the soil infiltration rates. The steady state infiltration rate was reached after 10 minutes of infiltration at a rate of 200 mm/hr. The effects of different water heads on infiltration rates were also investigated, and the head of 11 cm was found to be the optimum head for the test. The experimental results showed consistent infiltration results for the range between 11 cm and 15 cm. The study also involved finding the initial moisture content, which ranged between 5% and 25%, and finding the organic content, which occupied 1% to 2% of the soil. These results will be later utilized, using the water balance approach, to estimate the optimum amount of water needed for irrigation for changing weather conditions.

Abstract:
In this study, statistical optimization design was used to study the optimum disinfection parameters using defatted crude Moringa oleifera seed extracts against Escherichia coli (E. coli) bacterial cells. The classical one-factor-at-a-time (OFAT) and response surface methodology (RSM) was used. The possible optimum range of dosage, contact time and mixing rate from the OFAT study were 25mg/l to 200mg/l, 30minutes to 240 minutes and 100rpm to 160rpm respectively. Analysis of variance (ANOVA) of the statistical optimization using faced centered central composite design showed that dosage, contact time and mixing rate were highly significant. The optimum disinfection range was 125mg/l, at contact time of 30 minutes with mixing rate of 120 rpm.

Abstract:
This paper presents the DC voltage control design of D-STATCOM when the D-STATCOM is used for load voltage regulation. Although, the DC voltage can be controlled by active current of the D-STATCOM, reactive current still affects the DC voltage. To eliminate this effect, the control strategy with elimination effect of the reactive current is proposed and the results of the control with and without the elimination the effect of the reactive current are compared. For obtaining the proportional and integral gains of the PI controllers, the symmetrical optimum and genetic algorithms methods are applied. The stability margin of these methods are obtained and discussed in detail. In addition, the performance of the DC voltage control based on symmetrical optimum and genetic algorithms methods are compared. Effectiveness of the controllers designed was verified through computer simulation performed by using Power System Tool Block (PSB) in SIMULINK/MATLAB. The simulation results demonstrated that the DC voltage control proposed is effective in regulating DC voltage when the DSTATCOM is used for load voltage regulation.

Abstract:
The synergistic impact and optimization of gas flow rate, concentration of CO2, and light intensity on CO2 biofixation rate were investigated using wastewater as a medium to cultivate Chlorella vulgaris under different conditions (gas flow rate 1-8 L/min), CO2 concentration (0.03-7%), and light intensity (150-400 µmol/m2.s)). Response Surface Methodology and Box-Behnken experimental Design were applied to find optimum values for gas flow rate, CO2 concentration, and light intensity. The optimum values of the three independent variables (gas flow rate, concentration of CO2, and light intensity) and desirability were 7.5 L/min, 3.5%, and 400 µmol/m2.s, and 0.904, respectively. The highest amount of biomass produced and CO2 biofixation rate at optimum conditions were 5.7 g/L, 1.23 gL-1d-1, respectively. The synergistic effect between gas flow rate and concentration of CO2, and between gas flow rate and light intensity was significant on the three responses, while the effect between CO2 concentration and light intensity was less significant on CO2 biofixation rate. The results of this study could be highly helpful when using microalgae for CO2 biofixation in wastewater treatment.

In this research paper were investigated the main regularities of a radical bromination reaction of decalin. There had been studied the temperature effect, durations of reaction, frequency rate of process, a ratio of initial components, type and number of the initiator on decalin bromination degree. There were specified optimum conditions of synthesis of a perbromodecalin by the method of a decalin bromination. There are developed the technological flowchart of receiving a perbromodecalin and the mass balance of process on the first and the subsequent loadings of components. The results of research of antibacterial and antifungal activity of synthesized bromoderivatives have been represented.

Abstract:
Wireless charging of electric vehicles is becoming more and more attractive as large amount of power can now be transferred to a reasonable distance using magnetic resonance coupling method. However, proper tuning of the compensation network is required to achieve maximum power transmission. Due to the variation of coil distance from the nominal value as a result of change in tire condition, change in weight or uneven road condition, the tuning of the compensation network has become challenging. In this paper, a tuning method has been described to determine the optimum values of the compensation network in order to maximize the average output power. The simulation results show that 5.2% increase in average output power is obtained for 10% variation in coupling coefficient using the optimum values without the need of additional space and electro-mechanical components. The proposed method is applicable to both static and dynamic charging of electric vehicles.

Speech enhancement is the process of eliminating noise and increasing the quality of a speech signal, which is contaminated with other kinds of distortions. This paper is on developing an optimum cascaded system for speech enhancement. This aim is attained without diminishing any relevant speech information and without much computational and time complexity. LMS algorithm, Spectral Subtraction and Kalman filter have been deployed as the main de-noising algorithms in this work. Biodiesel, as an alternative renewable fuel, has been receiving increasing attention due to the limited supply of fossil fuels and the increasing need for energy. Microalgae are promising source for lipids, which can be converted to biodiesel. The biodiesel production from microalgae lipids using lipase catalyzed reaction in supercritical CO2 medium has several advantages over conventional production processes. However, identifying the optimum microalgae lipid extraction and transesterification conditions is still a challenge. In this study, the quality of biodiesel produced from lipids extracted from Scenedesmus sp. and their enzymatic transesterification using supercritical carbon dioxide have been investigated. At the optimum conditions, the highest biodiesel production yield was found to be 82%. The fuel properties of the produced biodiesel, without any separation step, at optimum reaction condition, were determined and compared to ASTM standards. The properties were found to comply with the limits, and showed a low glycerol content, without any separation step.Keywords: Biodiesel, fuel standards, lipase, microalgae, Supercritical CO2.

Abstract:
This paper describes the research carried out to develop a process to increase the NaCl percentage of crude salt which is obtained from the conventional solar evaporation process. In this study refined salt was produced from crude solar salt by a chemico-physical method which consists of coagulation, precipitation and filtration. Initially crude salt crystals were crushed and dissolved in water. Optimum amounts of calcium hydroxide, sodium carbonate and Polyaluminiumchloride (PAC) were added to the solution respectively.
Refined NaCl solution was separated out by a filtration process. The solution was tested for Total Suspended Solids, SO42-, Mg2+, Ca2+. With optimum dosage of reagents, the results showed that a level of 99.60% NaCl could be achieved. Further this paper discusses the economic viability of the proposed process. A 83% profit margin can be achieved by this process and it is an increase of 112.3% compared to the traditional process.

Abstract:
This work presents a new algorithm based on a combination of fuzzy (FUZ), Dynamic Programming (DP), and Genetic Algorithm (GA) approach for capacitor allocation in distribution feeders. The problem formulation considers two distinct objectives related to total cost of power loss and total cost of capacitors including the purchase and installation costs. The novel formulation is a multi-objective and non-differentiable optimization problem. The proposed method of this article uses fuzzy reasoning for sitting of capacitors in radial distribution feeders, DP for sizing and finally GA for finding the optimum shape of membership functions which are used in fuzzy reasoning stage. The proposed method has been implemented in a software package and its effectiveness has been verified through a 9-bus radial distribution feeder for the sake of conclusions supports. A comparison has been done among the proposed method of this paper and similar methods in other research works that shows the effectiveness of the proposed method of this paper for solving optimum capacitor planning problem.

Abstract:
A suitable choice of flotation parameters and reagents have a strong effect on the effectiveness of flotation process. The objective of this paper is to give an overview of the flotation of chalcopyrite with the different conditions and dispersants. Flotation parameters such as grinding time, pH, type, and dosage of dispersant were investigated. In order to understand the interaction of some dispersants, sodium silicate, sodium hexametaphosphate and sodium polyphosphate were used. The optimum results were obtained at a pH of 11.5 and a grinding time of 10 minutes. A copper concentrate was produced assaying 29.85% CuFeS2 and 65.97% flotation recovery under optimum rougher flotation conditions with sodium silicate.

This paper presents an extraction of maximum energy from Solar Photovoltaic Array (SPVA) under partial shaded conditions by optimum selection of array size using Particle Swarm Optimization (PSO) technique. In this paper a detailed study on the output reduction of different SPVA configurations under partial shaded conditions have been carried out. A generalized MATLAB M-code based software model has been used for any required array size, configuration, shading patterns and number of bypass diodes. Comparative study has been carried out on different configurations by testing several shading scenarios. While the number of shading patterns and the rate of change are very low for stationary SPVA but these may be quite large for SPVA mounted on a mobile platforms. This paper presents the suitability of PSO technique to select optimum configuration for mobile arrays by calculating the global peak (GP) of different configurations and to transfer maximum power to the load.

Hard coatings are widely used in cutting and forming tool industries. Titanium Nitride (TiN) possesses good hardness, strength, and corrosion resistance. The coating properties are influenced by many process parameters. The coatings were deposited on steel substrate by changing the process parameters such as substrate temperature, nitrogen flow rate and target power in a D.C planer magnetron sputtering. The structure of coatings were analysed using XRD. The hardness of coatings was found using Micro hardness tester. The dilute acid pretreatment and enzymatic saccharification of lignocellulosic substrate, cogon grass (Imperata cylindrical, L.) was optimized prior ethanol fermentation using simultaneous saccharification and fermentation (SSF) method. The optimum pretreatment conditions, temperature, sulfuric acid concentration, and reaction time were evaluated by determining the maximum sugar yield at constant enzyme loading. Cogon grass, at 10% w/v substrate loading, has optimum pretreatment conditions of 126°C, 0.6% v/v H2SO4, and 20min reaction time. These pretreatment conditions were used to optimize enzymatic saccharification using different enzyme combinations. The maximum saccharification yield of 36.68mg/mL (71.29% reducing sugar) was obtained using 25FPU/g-cellulose cellulase complex combined with 1.1% w/w of cellobiase, ß-glucosidase, and 0.225% w/w of hemicellulase complex, after 96 hours of saccharification. Using the optimum pretreatment and saccharification conditions, SSF of treated substrates was done at 37°C for 120 hours using industrial yeast strain HBY3, Saccharomyces cerevisiae. The ethanol yield for cogon grass at 4% w/w loading was 9.11g/L with 5.74mg/mL total residual sugar.Keywords: Acid pretreatment, bioethanol, biomass, cogon grass, fermentation, lignocellylose, SSF.

Abstract:
The main objective of this paper is to evaluate the use of plastic waste as a low cost asphalt binder modifier. For this purpose Marshall mix design procedure was used. Marshall mix design procedure seeks to select the Optimum Binder Content (OBC) to be added to a specific aggregate blend resulting in a mixture that satisfies the desired properties of strength and durability. In order to evaluate the plastic waste modified (PWM) asphalt mixtures, the OBC for the conventional asphalt mix was first identified, and then different percentages of crushed plastic waste by weight of the identified OBC were tested. Marshall test results for the modified asphalt mixtures were analyzed to find the optimum PWM content. Finally, the static indirect tensile strength (IDT) was determined for all mixtures using the splitting tensile test. It was found that PWM content of 7.43% by weight of OBC is recommended as the optimum PWM content needed for enhancing the performance of asphalt mixtures. It enhanced stability by 42.56%, flow by 89.91% and strength by 13.54%. This would lead to a more durable pavement by improving the pavement resistance to fatigue cracking and rutting.Keywords: Binder content modifier, Marshall test, plastic waste, polyethylene terephthalate.

Abstract:
The content of omega-3 in soybean oil is important in the development of infants and is an alternative for the omega-3 in fish oils. The investigation of extraction of soybean oil is needed to obtain the bioactive compound in the extract. Supercritical carbon dioxide extraction is modern and green technology to extract herbs and plants to obtain high quality extract due to high diffusivity and solubility of the solvent. The aim of this study was to obtain the optimum condition of soybean oil extraction by modified supercritical carbon dioxide. The soybean oil was extracted by using modified supercritical carbon dioxide (SC-CO2) under the temperatures of 40, 60, 80 °C, pressures of 150, 250, 350 Bar, and constant flow-rate of 10 g/min as the parameters of extraction processes. An experimental design was performed in order to optimize three important parameters of SC-CO2 extraction which are pressure (X1), temperature (X2) to achieve optimum yields of soybean oil. Box Behnken Design was applied for experimental design. From the optimization process, the optimum condition of extraction of soybean oil was obtained at pressure 338 Bar and temperature 80 °C with oil yield of 2.713 g. Effect of pressure is significant on the extraction of soybean oil by modified supercritical carbon dioxide. Increasing of pressure will increase the oil yield of soybean oil.

Abstract:
Optimal routing in communication networks is a major issue to be solved. In this paper, the application of Tabu Search (TS) in the optimum routing problem where the aim is to minimize the computational time and improvement of quality of the solution in the communication have been addressed. The goal is to minimize the average delays in the communication. The effectiveness of Tabu Search method is shown by the results of simulation to solve the shortest path problem. Abstract:
Xanthan gum is a microbial polysaccharide of great commercial significance. The purpose of this study was to select the optimum fermentation time for xanthan gum production by Xanthomonas campestris (NRRL-B-1459) using 10% sugar beet molasses as a carbon source. The pre-heating of sugar beet molasses and the supplementation of the medium were investigated in order to improve xanthan gum production. Maximum xanthan gum production in fermentation media (9.02 g/l) was observed after 4 days shaking incubation at 25°C and 240 rpm agitation speed. A solution of 10% sucrose was used as a control medium. Results indicated that the optimum period for xanthan gum production in this condition was 4 days.Keywords: Biomass, Molasses, Xanthan gum, Xanthomonascampestris

Abstract:
Direct fermentation of 226 white rose tapioca stem to ethanol by Fusarium oxysporum was studied in a batch reactor. Fermentation of ethanol can be achieved by sequential pretreatment using dilute acid and dilute alkali solutions using 100 mesh tapioca stem particles. The quantitative effects of substrate concentration, pH and temperature on ethanol concentration were optimized using a full factorial central composite design experiment. The optimum process conditions were then obtained using response surface methodology. The quadratic model indicated that substrate concentration of 33g/l, pH 5.52 and a temperature of 30.13oC were found to be optimum for maximum ethanol concentration of 8.64g/l. The predicted optimum process conditions obtained using response surface methodology was verified through confirmatory experiments. Leudeking-piret model was used to study the product formation kinetics for the production of ethanol and the model parameters were evaluated using experimental data.Keywords: Fusarium oxysporum, Lignocellulosic biomass, Product formation kinetics, Statistical experimental design

Abstract:
This paper presents performance of two robust gradient-based heuristic optimization procedures based on 3n enumeration and tunneling approach to seek global optimum of constrained integer problems. Both these procedures consist of two distinct phases for locating the global optimum of integer problems with a linear or non-linear objective function subject to linear or non-linear constraints. In both procedures, in the first phase, a local minimum of the function is found using the gradient approach coupled with hemstitching moves when a constraint is violated in order to return the search to the feasible region. In the second phase, in one optimization procedure, the second sub-procedure examines 3n integer combinations on the boundary and within hypercube volume encompassing the result neighboring the result from the first phase and in the second optimization procedure a tunneling function is constructed at the local minimum of the first phase so as to find another point on the other side of the barrier where the function value is approximately the same. In the next cycle, the search for the global optimum commences in both optimization procedures again using this new-found point as the starting vector. The search continues and repeated for various step sizes along the function gradient as well as that along the vector normal to the violated constraints until no improvement in optimum value is found. The results from both these proposed optimization methods are presented and compared with one provided by popular MS Excel solver that is provided within MS Office suite and other published results.

Abstract:
Heat source addition to the axisymmetric supersonic inlet may improve the performance parameters, which will increase the inlet efficiency. In this investigation the heat has been added to the flow field at some distance ahead of an axisymmetric inlet by adding an imaginary thermal source upstream of cowl lip. The effect of heat addition on the drag coefficient, mass flow rate and the overall efficiency of the inlet have been investigated. The results show that heat addition causes flow separation, hence to prevent this phenomena, roughness has been added on the spike surface. However, heat addition reduces the drag coefficient and the inlet mass flow rate considerably. Furthermore, the effects of position, size, and shape on the inlet performance were studied. It is found that the thermal source deflects the flow streamlines. By improper location of the thermal source, the optimum condition has been obtained. For the optimum condition, the drag coefficient is considerably reduced and the inlet mass flow rate and its efficiency have been increased slightly. In this paper, an automatic control system design based on Integral Squared Error (ISE) parameter optimization technique has been implemented on longitudinal flight dynamics of an UAV. It has been aimed to minimize the error function between the reference signal and the output of the plant. In the following parts, objective function has been defined with respect to error dynamics. An unconstrained optimization problem has been solved analytically by using necessary and sufficient conditions of optimality, optimum PID parameters have been obtained and implemented in control system dynamics.Keywords: Optimum Design, KKT Conditions, UAV, Longitudinal Flight Dynamics, ISE Parameter Optimization.

Abstract:
In this paper we are to find the optimum multiwavelet for compression of electrocardiogram (ECG) signals. At present, it is not well known which multiwavelet is the best choice for optimum compression of ECG. In this work, we examine different multiwavelets on 24 sets of ECG data with entirely different characteristics, selected from MITBIH database. For assessing the functionality of the different multiwavelets in compressing ECG signals, in addition to known factors such as Compression Ratio (CR), Percent Root Difference (PRD), Distortion (D), Root Mean Square Error (RMSE) in compression literature, we also employed the Cross Correlation (CC) criterion for studying the morphological relations between the reconstructed and the original ECG signal and Signal to reconstruction Noise Ratio (SNR). The simulation results show that the cardbal2 by the means of identity (Id) prefiltering method to be the best effective transformation.Keywords: ECG compression, Multiwavelet, Prefiltering.
