3200 Mechanisms of Ginger Bioactive Compounds Extract Using Soxhlet and Accelerated Water Extraction
Authors: M. N. Azian, A. N. Ilia Anisa, Y. Iwai
Abstract:
The mechanism for extraction bioactive compounds from plant matrix is essential for optimizing the extraction process. As a benchmark technique, a soxhlet extraction has been utilized for discussing the mechanism and compared with an accelerated water extraction. The trends of both techniques show that the process involves extraction and degradation. The highest yields of 6-, 8-, 10-gingerols and 6-shogaol in soxhlet extraction were 13.948, 7.12, 10.312 and 2.306 mg/g, respectively. The optimum 6-, 8-, 10-gingerols and 6-shogaol extracted by the accelerated water extraction at 140oC were 68.97±3.95 mg/g at 3min, 18.98±3.04 mg/g at 5min, 5.167±2.35 mg/g at 3min and 14.57±6.27 mg/g at 3min, respectively. The effect of temperature at 3mins shows that the concentration of 6-shogaol increased rapidly as decreasing the recovery of 6-gingerol.

Abstract:
Team distillation assisted by microwave extraction (SDAM) considered as accelerated technique extraction is a combination of microwave heating and steam distillation, performed at atmospheric pressure. SDAM has been compared with the same technique coupled with the cryogrinding of seeds (SDAM -CG). Isolation and concentration of volatile compounds are performed by a single stage for the extraction of essential oil from Cuminum cyminum seeds. The essential oils extracted by these two methods for 5 min were quantitatively (yield) and qualitatively (aromatic profile) no similar. These methods yield an essential oil with higher amounts of more valuable oxygenated compounds, and allow substantial savings of costs, in terms of time, energy and plant material. SDAM and SDAM-CG is a green technology and appears as a good alternative for the extraction of essential oils from aromatic plants. Keywords: Steam distillation, microwave extraction, Cuminum cyminum, chromatography, mass spectrometry

Abstract:
Subcritical water extraction was investigated as a novel and alternative technology in the food and pharmaceutical industry for the separation of Mannitol from olive leaves and its results was compared with those of Soxhlet extraction. The effects of temperature, pressure, and flow rate of water and also momentum and mass transfer dimensionless variables such as Reynolds and Peclet Numbers on extraction yield and equilibrium partition coefficient were investigated. The 30-110 bars, 60-150°C, and flow rates of 0.2-2 mL/min were the water operating conditions. The results revealed that the highest Mannitol yield was obtained at 100°C and 50 bars. However, extraction of Mannitol was not influenced by the variations of flow rate. The mathematical modeling of experimental measurements was also investigated and the model is capable of predicting the experimental measurements very well. In addition, the results indicated higher extraction yield for the subcritical water extraction in contrast to Soxhlet method. Keywords: Extraction, Mannitol, Modeling, Olive leaves, Soxhlet extraction, Subcritical water.

Abstract:
An impetus towards green consumerism and implementation of sustainable techniques, consumption of natural products and utilization of environment friendly techniques have gained accelerated acceptance. Butein, a natural colorant, has many medicinal properties apart from its use in dyeing industries. Extraction of butein from the flowers of flame of forest was carried out using ultrasonication bath. Solid loading (2-6 g), extraction time (30-50 min), volume of solvent (30-50 mL) and types of solvent (methanol, ethanol and water) have been studied to maximize the yield of butein using the Taguchi method. The highest yield of butein 4.67% (w/w) was obtained using 4 g of plant material, 40 min of extraction time and 30 mL volume of methanol as a solvent. The present method provided a greater reduction in extraction time compared to the conventional method of extraction. Hence, the outcome of the present investigation could further be utilized to develop the method at a higher scale.

Authors: R. Kandiah, K. Miyamoto
Abstract:
In Sri Lanka, the Jaffna area is a high drought affected area and depends mainly on groundwater aquifers for water needs. Water for daily activities is extracted from wells. As households manually extract water from the wells, it is not drawn from mid evening to early morning. The water inflow at night provides the maximum water level that decreases during the daytime due to extraction. The storage volume of water in wells is limited or at its lowest level during the dry season. This study analyzes the domestic water budget during the dry season in the Jaffna area. In order to evaluate the water inflow rate into wells, storage volume and extraction volume from wells over time, water pressure is measured at the bottom of three wells, which are located in coastal area denoted as well A, in nonspecific area denoted as well B, and agricultural area denoted as well C. The water quality at the wells A, B, and C, are mostly fresh, modest fresh, and saline respectively. From the monitoring, we can find that the daily inflow amount of water into the wells and daily water extraction depend on each other, that is, higher extraction yields higher inflow. And, in the dry season, the daily inflow volume and the daily extraction volume of each well are almost in balance.

Abstract:
Rambutan is a tropical fruit which peel possesses antioxidant properties. This work was conducted to optimize extraction conditions of phenolic compounds from rambutan peel. Response surface methodology (RSM) was adopted to optimize subcritical water extraction (SWE) on temperature, extraction time and percent solvent mixture. The results demonstrated that the optimum conditions for SWE were as follows: temperature 160°C, extraction time 20min. and concentration of 50% ethanol. Comparison of the phenolic compounds from the rambutan peels in maceration 6h, soxhlet 4h, and SWE 20min., it indicated that total phenolic content (using Folin-Ciocalteu-s phenol reagent) was 26.42, 70.29, and 172.47mg of tannic acid equivalent (TAE) per g dry rambutan peel, respectively. The comparative study concluded that SWE was a promising technique for phenolic compounds extraction from rambutan peel, due to much more two times of conventional techniques and shorter extraction times.

Abstract:
The traditional method for essential oil extraction from agarwood (Aquilaria Crassna) is to soak it in water and follow with hydrodistillation. The effect of various agarwood pretreatments: ethanol, acid, alkaline, enzymes, and ultrasound, and the effect of subcritical water extraction(SWE) was studied to compare with the traditional method. The major compositions of agarwood oil from hydrodistillation were aroma compounds as follow: aristol-9-en-8- one (21.53%), selina-3, 7(11)-diene (12.96%), τ-himachalene (9.28%), β-guaiene (5.79%), hexadecanoic acid (4.90%) and guaia- 3,9-diene (4.21%). Whereas agarwood oil from pretreatments with ethanol and ultrasound, and SWE got fatty acid compounds. Extraction of agarwood oil using these pretreatments could improve the agarwood oil yields up to 2 times that of the traditional method. The components of the pretreated sample with diluted acid (H2SO4) at pH 4 gave quite similar results as the traditional method. Therefore, the enhancement of essential oil from agarwood depends on requirement of type of extracted oil that involved extraction methods.

The aim of the current research was to determine best method for extraction of phenolic compounds from horseradish roots showing high antiradical activity. Three genotypes (No. 105; No. 106 and variety ‘Turku’) of horseradish roots were extracted with eight different solvents: n-hexane, ethyl acetate, diethyl ether, 2-propanol, acetone, ethanol (95%), ethanol / water / acetic acid (80/20/1 v/v/v) and ethanol / water (80/20 by volume) using two extraction methods (conventional and Soxhlet). As the best solvents ethanol and ethanol / water solutions can be chosen. Although in Soxhlet extracts TPC was higher, scavenging activity of DPPH˙ radicals did not increase. It can be concluded that using Soxhlet extraction method more compounds that are not effective antioxidants.

Isolation and concentration of volatile compounds are performed by a single stage. (ASDAM) has been compared with (ASDAM) with cryogrinding of seeds (CG) and a conventional technique, hydrodistillation assisted by microwave (HDAM), hydro-distillation (HD) for the extraction of essential oil from aromatic herb as caraway and cumin seeds. The essential oils extracted by (ASDAM) for 1 min were quantitatively (yield) and qualitatively (aromatic profile) no similar to those obtained by ASDAM-CG (1 min) and HD (for 3 h). The accelerated microwave extraction with cryogrinding inhibits numerous enzymatic reactions as hydrolysis of oils. Microwave radiations constitute the adequate mean for the extraction operations from the yields and high content in major component majority point view, and allow to minimise considerably the energy consumption, but especially heating time too, which is one of essential parameters of artifacts formation. The ASDAM and ASDAM-CG are green techniques and yields an essential oil with higher amounts of more valuable oxygenated compounds comparable to the biosynthesis compounds, and allows substantial savings of costs, in terms of time, energy and plant material. Keywords: Microwave, steam distillation, caraway, cumin, cryogrinding, GC-MS, GCxGC-MS.

Abstract:
Chalcopyrite (CuFeS2) is the most common primary mineral used for the commercial production of copper. The low dissolution efficiency of chalcopyrite in sulfate media has prevented an efficient industrial leaching of this mineral in sulfate media. Ferric ions, bacteria, oxygen and other oxidants have been used as oxidizing agents in the leaching of chalcopyrite in sulfate and chloride media under atmospheric or pressure leaching conditions. Two leaching methods were studied to evaluate chalcopyrite (CuFeS2) dissolution in acid media. First, the conventional oxidative acid leaching method was carried out using sulfuric acid (H2SO4) and potassium dichromate (K2Cr2O7) as oxidant at atmospheric pressure. Second, microwave-assisted acid leaching was performed using the microwave accelerated reaction system (MARS) for same reaction media. Parameters affecting the copper extraction such as leaching time, leaching temperature, concentration of H2SO4 and concentration of K2Cr2O7 were investigated. The results of conventional acid leaching experiments were compared to the microwave leaching method. It was found that the copper extraction obtained under high temperature and high concentrations of oxidant with microwave leaching is higher than those obtained conventionally. 81% copper extraction was obtained by the conventional oxidative acid leaching method in 180 min, with the concentration of 0.3 mol/L K2Cr2O7 in 0.5M H2SO4 at 50 ºC, while 93.5% copper extraction was obtained in 60 min with microwave leaching method under same conditions. Keywords: Extraction, copper, microwave-assisted leaching, chalcopyrite, potassium dichromate.

Ground kernels of raw and heat treated kernels, each with and without Viscozyme treatment were used. Microscopic images of the kernels were taken to analyse the visible effects of each treatment on the cotyledon cell structure of the kernels. Heat pretreated kernels before both extraction processes produced enhanced oil extraction yields than the control, with steam explosion the most efficient. In hexane extraction, applying a combination of steam explosion and Viscozyme treatments to the kernels before the extraction gave the maximum oil extractable in 1 hour; while for aqueous extraction, raw kernels treated with Viscozyme gave the highest oil extraction yield. Remarkable cotyledon cell disruption was evident in kernels treated with Viscozyme; whereas steam explosion and conventional heat treated kernels had similar effects.

Authors: P. Parthiban, J. Karthikeyan
Abstract:
This paper deals with the performance of semi-light weight concrete, prepared by using wood ash pellets as coarse aggregates which were improved by partial replacement of cement with alccofine. Alccofine is a mineral admixture which contains high glass content obtained through the process of controlled granulation. This is finer than cement which carries its own pozzolanic property. Therefore, cement could be replaced by alccofine as 0%, 5%, 10%, 15%, 20%, 25%, 30%, 35%, 40%, 45%, 50%, 55%, 60%, 65%, and 70% to enhance the strength and durability properties of concrete. High range water reducing admixtures (HRWA) were used in these mixes which were dosed up to 1.5% weight of the total cementitious content (alccofine & cement). It also develops the weaker transition zone into more impermeable layer. Specimens were subjected in both the accelerated curing method as well as conventional curing method. Experimental results were compared and reported, in that the maximum compressive strength of 32.6 MPa was achieved on 28th day with 30% replacement level in a density of 2200 kg/m3 to a conventional curing, while in the accelerated curing, maximum compressive strength was achieved at 40% replacement level. Rapid chloride penetration test (RCPT) output results for the conventional curing method at 0% and 70% give 3296.7 and 545.6 coulombs.

Authors: Ratchadaporn Oonsivilai, Jutarat Manatwiyangkool, Anant Oonsivilai
Abstract:
Theoptimal extraction condition of dried Phaseolus vulgaris powderwas studied. The three independent variables are raw material concentration, shaking and centrifugaltime. The dependent variables are both yield percentage of crude extract and alphaamylase enzyme inhibition activity. The experimental design was based on box-behnkendesign. Highest yield percentage of crude extract could get from extraction condition at concentration of 1, 0,1, concentration of 0.15 M ,extraction time for 2hour, and separationtime for60 min. Moreover, the crude extract with highest alpha-amylase enzyme inhibition activityoccurred by extraction condition at concentration of 0.10 M, extraction time for 2 min, and separation time for 45 minKeywords: Extraction time, Optimal condition, Alpha-amylase enzymeinhibition activity
