1169 Insight into Structure and Functions of of Acyl CoA Binding Protein of Leishmania major

Abstract:
Acyl-CoA binding protein (ACBP) is a housekeeping protein which functions as an intracellular carrier of acyl-CoA esters. Given the fact that the amastigote stage (blood stage) of Leishmania depends largely on fatty acids as the energy source, of which a large part is derived from its host, these proteins might have an important role in its survival. In Leishmania major, genome sequencing suggests the presence of six ACBPs, whose function remains largely unknown. For functional and structural characterization, one of the ACBP genes was cloned, and the protein was expressed and purified heterologously. Acyl-CoA ester binding and stoichiometry were analyzed by isothermal titration calorimetry and Dynamic light scattering. Our results shed light on high affinity of ACBP towards longer acyl-CoA esters, such as myristoyl-CoA to arachidonoyl-CoA with single binding site. To understand the binding mechanism & dynamics, Nuclear magnetic resonance assignments of this protein are being done. The protein's crystal structure was determined at 1.5Å resolution and revealed a classical topology for ACBP, containing four alpha-helical bundles. In the binding pocket, the loop between the first and the second helix (16 – 26AA) is four residues longer from other extensively studied ACBPs (PfACBP) and it curls upwards towards the pantothenate moiety of CoA to provide a large tunnel space for long acyl chain insertion.Keywords: acyl-coa binding protein (ACBP), acyl-coa esters, crystal structure, isothermal titration, calorimetry, Leishmania

Abstract:
The purpose of this research was to develop and evaluate mucoadhesive films for buccal administration of itraconazole using film-forming and mucoashesive polymers. Buccal films of chitosan bearing Itraconazole were prepared by solvent casting technique. The films have been evaluated in terms of film weight, thickness, density, surface pH, FTIR, X-ray diffraction analysis, bioadhesion, swelling properties, and in vitro drug release studies. It was found that film formulations of 2 cm2 size having weight in the range of 204 ± 0.76 to 223 ± 2.09 mg and film thickness were in the range of 0.44 ± 0.11 to 0.57 ± 0.19 mm. Density of the films was found to be 0.102 to 0.126 g/ml. Drug content was found to be uniform in the range of 8.23 ± 0.07 to 8.73 ± 0.09 mg/cm2 for formulation A1 to A4. Maximum bioadhesion force was recorded for HPMC buccal films (A2) i.e. 0.57 ± 0.47 as compared to other films. In vitro residence time was in range of 1.7 ± 0.12 to 7.65 ± 0.15 h. The drug release studies show that formulations follow non-fickian diffusion. These mucoadhesive formulations could offer many advantages in comparison to traditional treatments.Keywords: biovariability, buccal patches, itraconazole, Mucoadhesion

Abstract:
Up to now only five different equations of motion can be derived from velocity time graph without needing to know the normal and frictional forces acting at the point of contact. In this paper we obtained all possible requisite conditions to be considering an equation as an equation of motion. After that we classified equations of motion by considering two equations as fundamental kinematical equations of motion and other three as additional kinematical equations of motion. After deriving these five equations of motion, we examine the easiest way of solving a wide variety of useful numerical problems. Abstract:
Morphological evaluation of Indian languages is one of the burgeoning fields in the area of Natural Language Processing (NLP). The evaluation of a language is an eminent task in the era of information retrieval and text mining. The extraction and classification of knowledge from text can be exploited for sentiment analysis and morphological evaluation. This study coalesce morphological evaluation and sentiment analysis for the task of classification of farmer suicide cases reported in Punjab state of India. The pre-processing of Punjabi text involves morphological evaluation and normalization of Punjabi word tokens followed by the training of proposed model using deep learning classification on Punjabi language text extracted from online Punjabi news reports. The class-wise accuracies of sentiment prediction for four negatively oriented classes of farmer suicide cases are 93.85%, 88.53%, 83.3%, and 95.45% respectively. The overall accuracy of sentiment classification obtained using proposed framework on 275 Punjabi text documents is found to be 90.29%.Keywords: deep neural network, farmer suicides, morphological processing, punjabi text, sentiment analysis

Abstract:
The plant Eclipta alba Hassk. (Family: Compositae) contains coumestans (wedelolactone and demethyl wedelolactone) used in liver disorders. The objective of the present investigation was to develop a formulation of these coumestans in combination with the soya phosphatidylcholine (PC), in order to overcome the limitation of absorption and to investigate the protective effect of coumestans–phosphatidylcholine complex (C-PC) on carbon tetrachloride induced acute liver damage in rats. Methanolic extract (ME) of the whole plant of Eclipta alba was fractionated with water and then with ehylacetate. Coumestans were characterized in the ethylacetate fraction of methanolic extract (EFME). The C-PC was prepared by dissolving EFME and PC in 1:1 ratio in dichloromethane and heating at 60°C for 2 h. The C-PC was characterized by DSC and FTIR spectroscopy. In vitro drug release from EFME and C-PC through egg membrane was measured using UV-Visible spectrophotometer. The hepatoprotective activity of C-PC (equivalent to 5.35 and 10.7 mg/kg body weight of EFME), ME 250 mg/kg and EFME 5.35 mg/kg was evaluated by measuring various enzymes level. C-PC significantly provided better protection to the liver by restoring the enzyme levels of SGPT, SGOT, ALP and total billirubin with respect to carbon tetrachloride (CCl4) treated group (P < 0.001). Histopathological studies were also performed. The C-PC provided better protection to rat liver than ME and EFME at similar doses as well as shown significant regeneration of hepatocytes, central vein, intact cytoplasm, and nucleus.Keywords: hepatotoxicity, wedelolactone, soya phosphatidylcholine, eclipta alba

Abstract:
Transcranial direct current stimulation (tDCS) has shown promise as a novel approach to enhance cognitive performance and provide therapeutic benefits for various brain disorders. However, the exact underlying brain mechanisms are not fully understood. We conducted a study to examine the brain's functional changes when subjected to simultaneous tDCS and music (Indian classical raga). During the study, participants in the experimental group underwent a 20-minute session of tDCS at two mA while listening to music (raga) for a duration of seven days. In contrast, the control group received a sham stimulation for two minutes at two mA over the same seven-day period. The objective was to examine whether repetitive tDCS could lead to the formation of additional functional connections between the medial prefrontal cortex (the stimulated area) and the auditory cortex in comparison to a sham stimulation group. In this study, 26 participants (5 female) underwent pre- and post-intervention scans, where changes were compared after one week of either tDCS or sham stimulation in conjunction with music. The study revealed significant effects of tDCS on functional connectivity between the stimulated area and the auditory cortex. The combination of tDCS applied over the mPFC and music resulted in newly formed connections. Based on our findings, it can be inferred that applying anodal tDCS over the mPFC enhances functional connectivity between the stimulated area and the auditory cortex when compared to the effects observed with sham stimulation.Keywords: fNIRS, tDCS, neuroplasticity, music

Abstract:
Many materials are known to mankind that is widely used for synthesis of corrosion resistant hydrophobic coatings. In the current work, novel hydrophobic composite was synthesized by mixing polytetrafluoroethylene (PTFE) and 20 weight% ceria particles followed by sintering. This composite had same hydrophobic behavior as PTFE. Moreover, composite showed better scratch resistance than virgin PTFE. Pits of plasma sprayed Ni₃Al coating were exploited to hold PTFE composite on the substrate as Superni-75 alloy surface through sintering process. Plasma sprayed surface showed good adhesion with the composite coating during scratch test. Potentiodynamic corrosion test showed 100 fold decreases in corrosion rate of coated sample this may be attributed to inert and hydrophobic nature of PTFE and ceria.Keywords: polytetrafluoroethylene, PTFE, ceria, coating, corrosion

Aluminium and its alloys play have excellent corrosion resistant properties, ease of fabrication and high specific strength to weight ratio. In this investigation an attempt has been made to study the effect of different post weld heat treatment methods on the mechanical and metallurgical properties of TIG welded joints of the commercial aluminium alloy. Three different methods of post weld heat treatments are, solution heat treatment, artificial aged and combination of solution heat treatment and artificial aging are given to TIG welded aluminium joints. The proliferation of wireless sensor networks and Internet of Things (IoT) devices in the quickly changing digital landscape has highlighted the urgent need for strong security solutions that can handle these systems’ limited resources. A key solution to this problem is the emergence of ultralightweight security protocols, which provide strong security features while respecting the strict computational, energy, and memory constraints imposed on these kinds of devices. This in-depth analysis explores the field of ultralightweight security protocols, offering a thorough examination of their evolution, salient features, and the particular security issues they resolve. We carefully examine and contrast different protocols, pointing out their advantages and disadvantages as well as the compromises between resource limitations and security resilience. We also study these protocols’ application domains, including the Internet of Things, RFID systems, and wireless sensor networks, to name a few. In addition, the review highlights recent developments and advancements in the field, pointing out new trends and possible avenues for future research. Abstract:
This paper proposed a generalized class of estimators, an exponential class of estimators based on the adaption of Sharma and Singh (2015) and Solanki and Singh (2013), and a simple difference estimator for estimating unknown population mean in the case of Poisson distributed population in simple random sampling without replacement. The expressions for mean square errors of the proposed classes of estimators are derived from the first order of approximation. It is shown that the adapted version of Solanki and Singh (2013), the exponential class of estimator, is always more efficient than the usual estimator, ratio, product, exponential ratio, and exponential product type estimators and equally efficient to simple difference estimator. Moreover, the adapted version of Sharma and Singh's (2015) estimator is always more efficient than all the estimators available in the literature. In addition, theoretical findings are supported by an empirical study to show the superiority of the constructed estimators over others with an application to earthquake data of Turkey.Keywords: auxiliary attribute, point bi-serial, mean square error, simple random sampling, Poisson distribution

Decrement in vigilance task performance echoes impediment in effortful attention; here attention fluctuated in the realm of external and internal milieu of a person. To examine this fluctuation across time period, we employed two experiments of vigilance task with variation in thought probing rate, which was embedded in the task. The thought probe varies in terms of <2 minute per thought probe and <4 minute per thought probe during vigilance task. A 2x4 repeated measure factorial design was used. 15 individuals participated in this study with an age range of 20-26 years. It was found that thought probing rate has a negative trend with vigilance task performance whereas the subjective measures of mind-wandering have a positive relation with thought probe rate.Keywords: criterion response, mental status, mind-wandering, thought probe, vigilance

Abstract:
In the current investigation plasma spray technique was used for depositing hydroxyapatite (HA) and HA – silicon oxide (SiO2) coatings on 316L SS substrate. In HA-SiO2 coating, 20 wt% SiO2 was mixed with HA. The feedstock and coatings were characterized by X-ray diffraction (XRD) and scanning electron microscopy (SEM)/energy-dispersive X-ray spectroscopy (EDX) analyses. The corrosion resistance of the uncoated, HA coated and HA + 20 wt% SiO2 coated 316L SS was investigated by electrochemical corrosion testing in simulated human body fluid (Ringer’s solution). The influence of SiO2 (20 wt%) on corrosion resistance was determined. After the corrosion testing, the samples were analyzed by XRD and SEM/EDX analyses. The addition of SiO2 reduces the crystallinity of the coating. The corrosion resistance of the 316L SS was found to increase after the deposition of the HA + 20 wt% SiO2 and HA coatings.Keywords: HA, SiO2, corrosion, Ringer’s solution, 316L SS

Abstract:
A major problem related to the use of rotavator is wear of rotavator blades due to abrasion by soil hard particles, as it seriously affects tillage quality and agricultural production economy. The objective of this study was to increase the wear resistance by covering the rotavator blades with two different hard facing electrodes. These blades are generally produced from low carbon or low alloy steel. During the field work i.e. preparing land for the cultivation these blades are subjected to severe wear conditions. Comparative wear tests on a regular rotavator blade and two kinds of hardfacing with electrodes were conducted in the field. These two different hardfacing electrodes, which are designated HARD ALLOY-400 and HARD ALLOY-650, were used for hardfacing. The wear rate in the field tests was found to be significantly different statistically. When the cost is taken into consideration; HARD ALLOY-650 and HARD ALLOY-400 have been found to be the best hardfacing electrodes.Keywords: hardfacing, rotavator blades, hard alloy-400, abrasive wear

Abstract:
The aim of the present investigation was to study the effect of ultrasonic vibration on the cladding of the AISI 308 on the mild steel plates using the shielded metal arc welding (SMAW). Ultrasonic vibrations were applied to molten austenitic stainless steel during the welding process. Due to acoustically induced cavitations and streaming there is a complete mixture of the clad metal and the base metal. It was revealed that cladding of AISI 308 over mild steel along with ultrasonic vibrations result in uniform and finer grain structures. The effect of the vibration on the dilution, mechanical properties and metallographic studies were also studied. It was found that the welding done using the ultrasonic vibration has the less dilution and CVN value for the vibrated sample was also high.Keywords: surfacing, ultrasonic vibrations, mechanical properties, shielded metal arc welding

Abstract:
In the modern agricultural landscape, precision farming has emerged as a pivotal strategy for enhancing crop yield and optimizing resource utilization. This paper introduces an innovative Crop Recommendation System (CRS) that leverages the Internet of Things (IoT) technology and the Message Queuing Telemetry Transport (MQTT) protocol to collect critical environmental and soil data via sensors deployed across agricultural fields. The system is designed to address the challenges of real-time data acquisition, efficient data transmission, and dynamic crop recommendation through the application of advanced Artificial Intelligence (AI) and Machine Learning (ML) models. The CRS architecture encompasses a network of sensors that continuously monitor environmental parameters such as temperature, humidity, soil moisture, and nutrient levels. This sensor data is then transmitted to a central MQTT server, ensuring reliable and low-latency communication even in bandwidth-constrained scenarios typical of rural agricultural settings. Upon reaching the server, the data is processed and analyzed by AI/ML models trained to correlate specific environmental conditions with optimal crop choices and cultivation practices. These models consider historical crop performance data, current agricultural research, and real-time field conditions to generate tailored crop recommendations. Mobile phones have become a vital part of everyone’s life. Mobile phone and mobile phone towers emit RF-EMR (Radiofrequency Electromagnetic Radiation), which becomes a cause of concern to the general public. The study was designed to evaluate the effect of 3G (RF-EMR) on the reproductive system of male Wistar rats. Adult male Wistar rats were used for the study. Animals were divided into two groups, RF-exposed, and sham-exposed (control). RF-exposed rats were exposed to radio frequency radiation (2100 MHz) for 2 hours/day for 45 days. Emitted power density and specific absorption rate (SAR) values were measured during exposure. At the end of the exposure, testis and epididymis were excised out, and their weights were recorded. Sperm cell count, morphology, viability, and reactive oxygen species (ROS) levels were checked. Lipid peroxidation and sperm mitochondrial activity were measured. Histopathology of testis and ultrastructure analysis of sperm were also checked. Result showed a decrease in organ weight and sperm count with alteration in the sperm morphology in exposed group rats. A significant decrease in sperm viability, membrane integrity, and mitochondrial activity was found. Also, an increase in lipid peroxidation and ROS level were found in exposed group animals as compared to control. It may be concluded that exposure to radiofrequency radiation emits from mobile phones leads to oxidative stress-mediated changes in reproductive parameters.Keywords: electromagnetic radiation, oxidative stress, reactive oxygen species, sperm

The existing interdependent relationship between attention and consciousness has been put to debate since long. To testify the nature, dual-task paradigm has been used to simultaneously manipulate awareness and attention. With central discrimination task which is attentional demanding, participants also perform simple discrimination task in the periphery in near absence of attention. Individual-based analysis of performance accuracy in single and dual condition showed and above chance level performance i.e. more than 80%. In order to widen the understanding of extent of discrimination carried in near absence of attention, natural image and its geometric equivalent shape were presented in the periphery; synthetic objects accounted to lower level of performance than natural objects in dual condition. The gaze plot and heatmap indicate that peripheral performance do not necessarily involve saccade every time, verifying the discrimination in the periphery was in near absence of attention. The present research focused to improve the machinability of Hastelloy C-276 at different machining speeds such as 31, 55, and 79 m/min. The use of CO2 gas and Minimum quantity lubrication (MQL) was applied as coolant and lubrication purposes to enhance the machinability of the superalloy. The output in the form of surface roughness (S.R) and heat generation was monitored under dry, MQL, and MQL-CO2-cooled conditions. The Design of the Experiment was prepared using MINITAB software utilizing Taguchi L-27 orthogonal arrays followed by ANOVA analysis for finding the impact of input variables on output responses. At different speeds and lubrication conditions, different behavioral patterns for Surface Roughness and the temperature was observed. ANOVA analysis depicted that the cooling environment impacted the S.R. majorly (50%) followed by cutting speed (29.84%), feed rate (5.09%), and least through depth of cut (4.95%). On the other side, the temperature was greatly influenced by cutting speed (69.12%), Cryo-MQL (8.09%), feed rate (7.59%), and depth of cut (6.20%). Abstract:
Thermo-mechanical treatment (TMT) of rebars is a critical process to impart sufficient strength and ductility to rebar. TMT rebars are produced by the Tempcore process, involves an 'in-line' heat treatment in which hot rolled bar (temperature is around 1080°C) is passed through water boxes where it is quenched under high pressure water jets (temperature is around 25°C). The quenching rate dictates composite structure consisting (four non-homogenously distributed phases of rebar microstructure) pearlite-ferrite, bainite, and tempered martensite (from core to rim). The ferrite and pearlite phases present at core induce ductility to rebar while martensitic rim induces appropriate strength. The TMT process is difficult to model as it brings multitude of complex physics such as heat transfer, highly turbulent fluid flow, multicomponent and multiphase flow present in the control volume. Additionally the presence of film boiling regime (above Leidenfrost point) due to steam formation adds complexity to domain. A coupled heat transfer and fluid flow model based on computational fluid dynamics (CFD) has been developed at product technology division of Tata Steel, India which efficiently predicts temperature profile and percentage martensite rim thickness of rebar during quenching process. The model has been validated with 16 mm rolling of New Bar mill (NBM) plant of Tata Steel Limited, India. Abstract:
In this paper, a steganographic scheme for digital images using Integer Wavelet Transform (IWT) is proposed. The cover image is decomposed into wavelet sub bands using IWT. Each of the subband is divided into blocks of equal size and secret data is embedded into the largest and smallest pixel values of each block of the subband. Visual quality of stego images is acceptable as PSNR between cover image and stego is above 40 dB, imperceptibility is maintained. Experimental results show better tradeoff between capacity and visual perceptivity compared to the existing algorithms. Maximum possible error analysis is evaluated for each of the wavelet subbands of an image. Abstract:
Friction Stir Welding Process emerged as promising solid-state welding and eliminates various welding defects like cracks and porosity in joining of dissimilar aluminum alloys. In the present research, Friction Stir Welding (FSW) is carried out on dissimilar aluminum alloys 2000 series and 6000 series this combination of alloys are highly used in automobile and aerospace industry due to their good strength to weight ratio, mechanical, and corrosion properties. The joints characterized by applying various destructive and non-destructive tests. Three critical welding parameters were considered i.e. Tool Rotation speed, Transverse speed, and Tool Geometry. The effective range of tool rotation speed from 1200-1800 rpm and transverse speed from 60-240 mm/min and tool geometry was studied. The two-different difficult to weld alloys were successfully welded. All the samples showed different microstructure with different set of welding parameters. The fundamental problem of subspace coding is to explore the maximum possible cardinality Aq(n, d, k) of a set of k-dimensional subspaces of an n-dimensional vector space over Fq such that the subspace distance satisfies ds(W1, W2) ≥ d for any two distinct subspaces W1, W2 in this set. In this paper, we construct a new class of constant dimension codes (CDCs) by generalizing the coset construction and combining it with CDCs derived from parallel linkage construction and coset construction with an aim to improve the new lower bounds of Aq(n, d, k). Abstract:
In the present investigation, 75Cr3C2-25NiCr coating was deposited on T91 boiler tube steel substrate by high velocity oxy-fuel (HVOF) process to enhance high-temperature corrosion resistance. High-temperature performance of bare, as well as HVOF-coated steel specimens was evaluated for 1500 h under cyclic conditions in the platen superheater zone coal-fired boiler, where the temperature was around 900 °C. Experiments were carried out for 15 cycles each of 100 h duration followed by 1 h cooling at ambient temperature. The performance of the bare and coated specimens was assessed via metal thickness loss corresponding to the corrosion scale formation and the depth of internal corrosion attack. 75Cr3C2-25NiCr coating deposited on T91 steel imparted better hot corrosion resistance than the uncoated steel. Inferior resistance of bare T91 steel is attributed to the formation of pores and loosely bounded oxide scale rich in Fe2O3.Keywords: 75Cr3C2-25NiCr, HVOF process, boiler steel, coal fired boilers

We present a new model based on viscoelasticity for the Non-Newtonian fluids.We use a matrix formulated algorithm to approximate solutions of a class of partial integro-differential equations with the given initial and boundary conditions. Some numerical results are presented to simplify application of operational matrix formulation and reduce the computational cost. Convergence analysis, error estimation and numerical stability of the method are also investigated. Abstract:
The potential use of liposomes as drug carriers by i.v. injection is limited by their low stability in blood stream. Firstly, phospholipid exchange and transfer to lipoproteins, mainly HDL destabilizes and disintegrates liposomes with subsequent loss of content. To avoid the pain associated with injection and to obtain better patient compliance studies concerning various dosage forms, have been developed. Conventional liposomes (unilamellar and multilamellar) have certain drawbacks like low entrapment efficiency, stability and release of drug after single breach in external membrane, have led to the new type of liposomal systems. The challenge has been successfully met in the form of Double Liposomes (DL). DL is a recently developed type of liposome, consisting of smaller liposomes enveloped in lipid bilayers. The outer lipid layer of DL can protect inner liposomes against various enzymes, therefore DL was thought to be more effective than ordinary liposomes. This concept was also supported by in vitro release characteristics i.e. DL formation inhibited the release of drugs encapsulated in inner liposomes. DL consists of several small liposomes encapsulated in large liposomes, i.e., multivesicular vesicles (MVV), therefore, DL should be discriminated from ordinary classification of multilamellar vesicles (MLV), large unilamellar vesicles (LUV), small unilamellar vesicles (SUV). However, for these liposomes, the volume of inner phase is small and loading volume of water-soluble drugs is low. In the present study, the potential of phosphatidylethanolamine (PE) lipid anchored double liposomes (DL) to incorporate two drugs in a single system is exploited as a tool to augment the H. pylori eradication rate. Preparation of DL involves two steps, first formation of primary (inner) liposomes by thin film hydration method containing one drug, then addition of suspension of inner liposomes on thin film of lipid containing the other drug. The success of formation of DL was characterized by optical and transmission electron microscopy. Quantitation of DL-bacterial interaction was evaluated in terms of percent growth inhibition (%GI) on reference strain of H. pylori ATCC 26695. To confirm specific binding efficacy of DL to H. pylori PE surface receptor we performed an agglutination assay. Agglutination in DL treated H. pylori suspension suggested selectivity of DL towards the PE surface receptor of H. pylori. Monotherapy is generally not recommended for treatment of a H. pylori infection due to the danger of development of resistance and unacceptably low eradication rates. Therefore, combination therapy with amoxicillin trihydrate (AMOX) as anti-H. pylori agent and ranitidine bismuth citrate (RBC) as antisecretory agent were selected for the study with an expectation that this dual-drug delivery approach will exert acceptable anti-H. pylori activity.Keywords: Helicobacter pylorI, amoxicillin trihydrate, Ranitidine Bismuth citrate, phosphatidylethanolamine, multi vesicular systems
