231 Antibacterial and Antifungal Activity of Essential Oil of Eucalyptus camendulensis on a Few Bacteria and Fungi

Abstract:

Red River Gum (Eucalyptus camaldulensis) is a tree of the genus Eucalyptus widely distributed in Algeria and in the world. The value of its aromatic secondary metabolites offers new perspectives in the pharmaceutical industry. This strategy can contribute to the sustainable development of our country. Preliminary tests performed on the essential oil of Eucalyptus camendulensis showed that this oil has antibacterial activity vis-à-vis the bacterial strains (Enterococcus feacalis, Enterobacter cloaceai, Proteus microsilis, Escherichia coli, Klebsiella pneumonia, and Pseudomonas aeruginosa) and antifungic (Fusarium sporotrichioide and Fusarium graminearum). The culture medium used was nutrient broth Muller Hinton. The interaction between the bacteria and the essential oil is expressed by a zone of inhibition with diameters of MIC indirectly expression of. And we used the PDA medium to determine the fungal activity. The extraction of the aromatic fraction (essentially oilhydrolat) of the fresh aerian part of the Eucalyptus camendulensis was performed by hydrodistillation. The average essential oil yield is 0.99%. The antimicrobial and fungal study of the essential oil and hydrosol showed a high inhibitory effect on the growth of pathogens.

230 Negative Impact of Bacteria Legionella Pneumophila in Hot Water Distribution Systems on Human Health

Abstract:

Safe drinking water is one of the biggest issues facing the planet this century. The primary aim of this paper is to present our research focused on theoretical and experimental analysis of potable water and in-building water distribution systems from the point of view of microbiological risk on the basis of confrontation between the theoretical analysis and synthesis of gathered information in conditions of the Slovak Republic. The presence of the bacteria Legionella in water systems, especially in hot water distribution system, represents in terms of health protection of inhabitants the crucial problem which cannot be overlooked. Legionella pneumophila discovery, its classification and its influence on installations inside buildings are relatively new. There are a lot of guidelines and regulations developed in many individual countries for the design, operation and maintenance for tap water systems to avoid the growth of bacteria Legionella pneumophila, but in Slovakia we don-t have any. The goal of this paper is to show the necessity of prevention and regulations for installations inside buildings verified by simulation methods.

229 Exploratory Tests of Crude Bacteriocins from Autochthonous Lactic Acid Bacteria against Food-Borne Pathogens and Spoilage Bacteria

Abstract:

The aim of the present work was to test in vitro inhibition of food pathogens and spoilage bacteria by crude bacteriocins from autochthonous lactic acid bacteria. Thirty autochthonous lactic acid bacteria isolated previously, belonging to the genera: Lactobacillus, Carnobacterium, Lactococcus, Vagococcus, Streptococcus, and Pediococcus, have been screened by an agar spot test and a well diffusion assay against Gram-positive and Gram-negative harmful bacteria: Bacillus cereus, Bacillus subtilis ATCC 6633, Escherichia coli ATCC 8739, Salmonella typhimurium ATCC 14028, Staphylococcus aureus ATCC 6538, and Pseudomonas aeruginosa under conditions means to reduce lactic acid and hydrogen peroxide effect to select bacteria with high bacteriocinogenic potential. Furthermore, crude bacteriocins semiquantification and heat sensitivity to different temperatures (80, 95, 110°C, and 121°C) were performed. Another exploratory test concerning the response of St. aureus ATCC 6538 to the presence of crude bacteriocins was realized. It has been observed by the agar spot test that fifteen candidates were active toward Gram-positive targets strains. The secondary screening demonstrated an antagonistic activity oriented only against St. aureus ATCC 6538, leading to the selection of five isolates: Lm14, Lm21, Lm23, Lm24, and Lm25 with a larger inhibition zone compared to the others. The ANOVA statistical analysis reveals a small variation of repeatability: Lm21: 0.56%, Lm23: 0%, Lm25: 1.67%, Lm14: 1.88%, Lm24: 2.14%. Conversely, slight variation was reported in terms of inhibition diameters: 9.58± 0.40, 9.83± 0.46 and 10.16± 0.24 8.5 ± 0.40 10 mm for, Lm21, Lm23, Lm25, Lm14and Lm24, indicating that the observed potential showed a heterogeneous distribution (BMS = 0.383, WMS = 0.117). The repeatability coefficient calculated displayed 7.35%. As for the bacteriocins semiquantification, the five samples exhibited production amounts about 4.16 for Lm21, Lm23, Lm25 and 2.08 AU/ml for Lm14, Lm24. Concerning the sensitivity the crude bacteriocins were fully insensitive to heat inactivation, until 121°C, they preserved the same inhibition diameter. As to, kinetic of growth , the µmax showed reductions in pathogens load for Lm21, Lm23, Lm25, Lm14, Lm24 of about 42.92%, 84.12%, 88.55%, 54.95%, 29.97% in the second trails. Inversely, this pathogen growth after five hours displayed differences of 79.45%, 12.64%, 11.82%, 87.88%, 85.66% in the second trails, compared to the control. This study showed potential inhibition to the growth of this food pathogen, suggesting the possibility to improve the hygienic food quality.

228 Identification Common Microbes Observed on Polyester Tufting

Abstract:

Tufting carpet is a very suitable substrate for growing microorganism such as pathogenic microbes, due to the direct touch with human body, long washing periods and laying on the floor; in fact there are 3 major problems: To risk human health, Prepare bad odors and Destruction of the products.. In the presented research, for investigation of presence most common microbes on polyester tufting, first goods laid in a public place (in the corridor fair) for 30 days and the existence of some microbes were investigate on it with two methods of enrichment in nutrient environments such as thioglycolate and noutrunt brath, and shake the dust off the polyester tufting onto cultivation mediums such as blood agar and noutrunt agar. After the microorganism colonics are grown, the colonies were separated and six microbial tests such as cataloes and sitrat were carried out in five phases on the colonics for identifying the varieties of bacteria. As a result of tests, 5 type of bacteria, such as Escherichia coli, staphylococcus saprophytic as were identified. Each of the mentioned bacteria can be seriously harmful for the heath of human.

227 Fermentation of Germinated Native Black Rice Milk Mixture by Probiotic Lactic Acid Bacteria

Abstract:

This research aimed to demonstrate probiotic germinated native black rice juice fermentation by lactic acid bacteria (Lactobacillus casei TISTR 390). Germinated native black rice juice was inoculated with a 24-h old lactic culture and incubated at 30 °C for 72 hours. Changes in pH, acidity, total soluble solid, and viable cell counts during fermentation under controlled conditions at 0-h, 24-h, 48-h, and 72-h fermentations were evaluated. The study found out that the change in pH and total soluble solid of probiotic germinated black rice juice significantly (p ≤ 0.05) decreased at 72-h fermentation (5.67±0.12 to 2.86±0.04 and 7.00±0.00 to 6.40±0.00 ºbrix at 0-h and 72-h fermentations, respectively). On the other hand, the amount of titratable acidity expressed as lactic acid and the viable cell count significantly (p≤0.05) increased at 72-h fermentation (0.11±0.06 to 0.43±0.06 (% lactic acid) and 3.60 x 106 to 2.75 x 108 CFU/ml at 0-h and 72-h fermentations, respectively). Interestingly, the amount of γ-Amino Butyric Acid (GABA) had a significant difference (p≤0.05) twice as high as that of the control group (0.25±0.01 and 0.13±0.01 mg/100g, respectively). In addition, the free radical scavenging capacity assayed by DPPH method also showed that the IC50 values were significantly (p≤0.05) higher than the control (147.71±0.96 and 202.55±1.24 mg/ml, respectively). After 4 weeks of cold storage at 4 °C, the viable cell counts of lactic acid bacteria reduced to 1.37 x 106 CFU/ml. In conclusion, fermented germinated native black rice juice could be served as a healthy beverage for vegans and people who are allergic to cow milk products.

226 Communities of Ammonia-oxidizing Archaea and Bacteria in Enriched Nitrifying Activated Sludge

Abstract:

In this study, communities of ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB) in nitrifying activated sludge (NAS) prepared by enriching sludge from a municipal wastewater treatment plant in three continuous-flow reactors receiving an inorganic medium containing different ammonium concentrations of 2, 10, and 30 mM NH4 +-N (NAS2, NAS10, and NAS30, respectively) were investigated using molecular analysis. Results suggested that almost all AOA clones from NAS2, NAS10, and NAS30 fell into the same AOA cluster and AOA communities in NAS2 and NAS10 were more diverse than those of NAS30. In contrast to AOA, AOB communities obviously shifted from the seed sludge to enriched NASs and in each enriched NAS, communities of AOB varied particularly. The seed sludge contained members of N. communis cluster and N. oligotropha cluster. After it was enriched under various ammonium loads, members of N. communis cluster disappeared from all enriched NASs. AOB with high affinity to ammonia presented in NAS 2, AOB with low affinity to ammonia presented in NAS 30, and both types of AOB survived in NAS 10. These demonstrated that ammonium load significantly influenced AOB communities, but not AOA communities in enriched NASs.

225 Study on Nitrite Accumulation Characteristics and Nitrifying Population Dynamics at Different Growth Environments

Abstract:

Novel nitrogen removal technologies via nitrite pathway attract increasing interest in recent years. In this study, batch experiments were performed to investigate nitrite accumulation characteristics and shifts in nitrifying community structure at different growth environments including ammonia concentration, pH and alkalinity. It was found that nitrite accumulation ratios were maintained at around 95% at studied conditions, and the optimum pH and Alk/N (ratio between alkalinity and nitrogen) for ammonium oxidization were 8.5 and 8.33, respectively. Fluorescence in situ hybridization analysis of nitrifying bacteria showed that high free ammonia (from influent ammonium or caused by high pH) significantly altered the structure of nitrifying community, leading to abundance of ammonia-oxidizing bacteria (AOB), especially Nitrososmonas, and inhibition of nitrite-oxidizing bacteria (NOB). The results suggest that free ammonia plays more important role than other studied conditions on nitrite accumulation.

224 Antibacterial Activity of Some Medicinal Plant Extracts

Abstract:

Medicinal plants are now gaining attractiveness in treatment of bacterial infections and food preservation. The objective of this study was to assess antibacterial activity of some medicinal plants on pathogenic bacteria. Screening of antibacterial activity of aqueous and methanol extracts of some plants: Jojoba, Ginger, Sage, Thyme and Clove against Bacillus cereus, Salmonella typhimurium, Staphylococcus aureus, Clostridium perfringens and Escherichia coli were investigated. Antibacterial activity was performed by agar diffusion and disc diffusion method. Jatropha, Jojoba, Clove and Ginger extracts showed notable bacterial activity in the first screening step then selected to be tested against Bacillus cereus (Gram+), Staphylococcus aureus (Gram+) and Salmonella typhimurium (Gram−) and their effect was compared using antibiotics as control. Screening results showed potential antibacterial activity of the tested plant extracts against the screened bacterial strains. It was found that methanol extracts exhibited higher antibacterial activity than aqueous extracts. Methanol extract of Jatropha showed the highest inhibition zone against Staphylococcus aureus (Gram+) with 24.00 mm diameter, compared to the other plant extracts followed by clove. Meanwhile, the inhibition zones of methanol extracts of Jojoba and Ginger were the same (12mm).The Gram-positive bacteria were found to be more sensitive to aqueous and methanol extracts than Gram-negative bacteria.

223 Extracellular Protein Secreted by Bacillus subtilis ATCC21332 in the Presence of Streptomycin Sulfate

Abstract:

The extracellular proteins secreted by bacteria may be increased in stressful surroundings, such as in the presence of antibiotics. It appears that many antibiotics, when used at low concentrations, have in common the ability to activate or repress gene transcription, which is distinct from their inhibitory effect. There have been comparatively few studies on the potential of antibiotics as a specific chemical signal that can trigger a variety of biological functions. Therefore, this study was carried out to determine the effect of Streptomycin Sulfate in regulating extracellular proteins secreted by Bacillus subtilis ATCC21332. Results of Microdilution assay showed that the Minimum Inhibition Concentration (MIC) of Streptomycin Sulfate on B. subtilis ATCC21332 was 2.5 mg/ml. The bacteria cells were then exposed to Streptomycin Sulfate at concentration of 0.01 MIC before being further incubated for 48h to 72 h. The extracellular proteins secreted were then isolated and analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). Proteins profile revealed that three additional bands with approximate sizes of 30 kDa, 22 kDa and 23 kDa were appeared for the treated bacteria with Streptomycin Sulfate. Thus, B. subtilis ATCC21332 in stressful condition with the presence of Streptomycin Sulfate at low concentration could induce the extracellular proteins secretion.

222 Aureobasidium pullulans Used as a Biological Control Agent under Field Conditions Affects the Microbial Quality of Winter Wheat Grain

Abstract:

The biological activity of A. pullulans isolates against species of the genus Fusarium, bacteria of the genus Azotobacter and pseudomonads colonizing wheat kernels was evaluated. A field experiment was carried out in 2009-2011, in north-eastern Poland. Winter wheat (cv. Bogatka) plants were sprayed with a cell suspension of A. pullulans at a density of 106 - 108 per cm3 water at the stem elongation stage and the heading stage. Untreated plants served as control. The abundance of epiphytic yeasts, bacteria of the genus Azotobacter, pseudomonads and Fusarium pathogens on wheat grain was estimated at harvest and after six months’ storage. The average size of yeast communities was significantly greater on wheat kernels treated with a cell suspension of A. pullulans, compared with control samples. In 2010-2011, biological control reduced the abundance of some species of the genus Fusarium.

221 Influence of Yeast Strains on Microbiological Stability of Wheat Bread

Abstract:

Problem of food preservation is extremely important for mankind. Viscous damage ("illness") of bread results from development of Bacillus spp. bacteria. High temperature resistant spores of this microorganism are steady against 120°C) and remain in bread during pastries, potentially causing spoilage of the final product. Scientists are interested in further characterization of bread spoiling Bacillus spp. species. Our aim was to find weather yeast Saccharomyces cerevisiae strains that are able to produce natural antimicrobial killer factor can preserve bread illness. By diffusion method, we showed yeast antagonistic activity against spore-forming bacteria. Experimental technological parameters were the same as for bakers' yeasts production on the industrial scale. Risograph test during dough fermentation demonstrated gas production. The major finding of the study was a clear indication of the presence of killer yeast strain antagonistic activity against rope in bread causing bacteria. After demonstrating antagonistic effect of S. cerevisiae on bacteria using solid nutrient medium, we tested baked bread under provocative conditions. We also measured formation of carbon dioxide in the dough, dough-making duration and quality of the final products, when using different strains of S. cerevisiae. It is determined that the use of yeast S. cerevisiae RCAM 01730 killer strain inhibits appearance of rope in bread. Thus, natural yeast antimicrobial killer toxin, produced by some S. cerevisiae strains is an anti-rope in bread protector.

220 The Efficiency of Irrigation System and Nitrogen Fixation for inoculated Soybeans by using N15 Tracer Techniques

Abstract:

Repeated additions of the unfertilized bacteria led to increase the activity of Nitrogen-fixing bacteria in the root zone with drip irrigation system compared to traditional manual vaccination to increase the proportion of Nitrogen from 29% to 64%, and the efficiency of adding Nitrogen fertilizer did not exceed 9.5% while dropped to 4%, due to the amount of fertilizer added was not exceed 20kg N/h, and the second was the existence of a large amount of available Nitrogen in the soil by fixation, while the efficiency of irrigation system between 2.08 to 2.26 kg/m3.

219 Comparison between Antibacterial Effects of Ethanolic and Isopropyl: Hexan (7:3) Extracts of Zingiber officinale Rose

Abstract:

In this investigation, the antibacterial effects of ethanolic and 7:3 isopropyl –hexane mixture extracts of Zingiber officinale were evaluated against three Gram positive bacteria, B. cereus, S.epidermidis, S. aureus and three Gram negative bacteria, E. coli, K.pneumonia and P.areuginosa. Utilizing paper disk diffusion and well methods in-vitro, MIC and MBC were determined by macrodilution. The results showed that ethanolic rhizome extract of ginger had significantly active than Isopropyl –hexan extract. Further work needs to be done in these extracts including fractionation to isolate active constituents and subsequent pharmacological evaluation.

218 Influential Effect of Self-Healing Treatment on Water Absorption and Electrical Resistance of Normal and Light Weight Aggregate Concretes

Abstract:

Interest in using bacteria in cement materials due to its positive influences has been increased. Cement materials such as mortar and concrete basically suffer from higher porosity and water absorption compared to other building materials such as steel materials. Because of the negative side-effects of certain chemical techniques, biological methods have been proposed as a desired and environmentally friendly strategy for reducing concrete porosity and diminishing water absorption. This paper presents the results of an experimental investigation carried out to evaluate the influence of Sporosarcina pasteurii bacteria on the behaviour of two types of concretes (light weight aggregate concrete and normal weight concrete). The resistance of specimens to water penetration by testing water absorption and evaluating the electrical resistance of those concretes was examined and compared. As a conclusion, 20% increase in electrical resistance and 10% reduction in water absorption of lightweight aggregate concrete (LWAC) and for normal concrete the results show 7% decrease in water absorption and almost 10% increase in electrical resistance.

217 Sterility Examination and Comparative Analyses of Inhibitory Effect of Honey on Some Gram Negative and Gram Positive Food Borne Pathogens in South West Nigeria

Abstract:

Food borne illnesses have been reported to be a global health challenge. Annual incidences of food–related diseases involve 76 million cases, of which only 14 million can be traced to known pathogens. Poor hygienic practices have contributed greatly to this. It has been reported that in the year 2000 about 2.1 million people died from diarrheal diseases, hence, there is a need to ensure food safety at all level. This study focused on the sterility examination and inhibitory effect of honey samples on selected gram negative and gram positive food borne pathogen from South West Nigeria. The laboratory examinations revealed the presence of some bacterial and fungal contaminations of honey samples and that inhibitory activity of the honey sample was more pronounced on the gram negative bacteria than the gram positive bacterial isolates. Antibiotic sensitivity test conducted on the different bacterial isolates also showed that honey was able to inhibit the proliferation of the tested bacteria than the employed antibiotics.

216 Effect of Azespirilium Bacteria in Reducing Nitrogen Fertilizers (Urea) and the Interaction of it with Stereptomyces Sp due the Biological Control on the Wheat (Triticum Asstivum) Sustinibelation Culture

Abstract:

An experiment was conducted in October 2008 due the ability replacement plant associate biofertilizers by chemical fertilizers and the qualifying rate of chemical N fertilizers at the moment of using this biofertilizers and the interaction of this biofertilizer on each other. This field experiment has been done in Persepolis (Throne of Jamshid) and arrange by using factorial with the basis of randomized complete block design, in three replication Azespirilium SP bacteria has been admixed with consistence 108 cfu/g and inoculated with seeds of wheat, The streptomyces SP has been used in amount of 550 gr/ha and concatenated on clay and for the qualifying range of chemical fertilizer 4 level of N chemical fertilizer from the source of urea (N0=0, N1=60, N2=120, N3=180) has been used in this experiment. The results indicated there were Significant differences between levels of Nitrogen fertilizer in the entire characteristic which has been measured in this experiment. The admixed Azespirilium SP showed significant differences between their levels in the characteristics such as No. of fertile ear, No. of grain per ear, grain yield, grain protein percentage, leaf area index and the agronomic fertilizer use efficiency. Due the interaction streptomyses with Azespirilium SP bacteria this actinomycet didn-t show any statistically significant differences between it levels.

215 Mechanisms Involved In Organic Solvent Resistance in Gram-Negative Bacteria

Abstract:

The high world interest given to the researches concerning the study of moderately halophilic solvent-tolerant bacteria isolated from marine polluted environments is due to their high biotechnological potential, and also to the perspective of their application in different remediation technologies. Using enrichment procedures, I isolated two moderately halophilic Gram-negative bacterial strains from seawater sample, which are tolerant to organic solvents. Cell tolerance, adhesion and cells viability of Aeromonas salmonicida IBBCt2 and Pseudomonas aeruginosa IBBCt3 in the presence of organic solvents depends not only on its physicochemical properties and its concentration, but also on the specific response of the cells, and the cellular response is not the same for these bacterial strains. n-hexane, n-heptane, propylbenzene, with log POW between 3.69 and 4.39, were less toxic for Aeromonas salmonicida IBBCt2 and Pseudomonas aeruginosa IBBCt3, compared with toluene, styrene, xylene isomers and ethylbenzene, with log POW between 2.64 and 3.17. The results indicated that Aeromonas salmonicida IBBCt2 is more susceptible to organic solvents than Pseudomonas aeruginosa IBBCt3. The mechanisms underlying solvent tolerance (e.g., the existance of the efflux pumps) in Aeromonas salmonicida IBBCt2 and Pseudomonas aeruginosa IBBCt3 it was also studied.

214 Nitrification Efficiency and Community Structure of Municipal Activated Sewage Sludge

Abstract:

Nitrification is essential to biological processes designed to remove ammonia and/or total nitrogen. It removes excess nitrogenous compound in wastewater which could be very toxic to the aquatic fauna or cause serious imbalance of such aquatic ecosystem. Efficient nitrification is linked to an in-depth knowledge of the structure and dynamics of the nitrifying community structure within the wastewater treatment systems. In this study, molecular technique was employed for characterizing the microbial structure of activated sludge [ammonia oxidizing bacteria (AOB) and nitrite oxidizing bacteria (NOB)] in a municipal wastewater treatment with intention of linking it to the plant efficiency. PCR based phylogenetic analysis was also carried out. The average operating and environmental parameters as well as specific nitrification rate of plant was investigated during the study. During the investigation the average temperature was 23±1.5oC. Other operational parameters such as mixed liquor suspended solids and chemical oxygen demand inversely correlated with ammonia removal. The dissolved oxygen level in the plant was constantly lower than the optimum (between 0.24 and 1.267 mg/l) during this study. The plant was treating wastewater with influent ammonia concentration of 31.69 and 24.47 mg/L. The influent flow rates (ML/Day) was 96.81 during period. The dominant nitrifiers include: Nitrosomonas spp. Nitrobacter spp. and Nitrospira spp. The AOB had correlation with nitrification efficiency and temperature. This study shows that the specific ammonia oxidizing rate and the specific nitrate formation rates can serve as good indicator of the plant overall nitrification performance.

213 The Antibacterial Efficacy of Gold Nanoparticles Derived from Gomphrena celosioides and Prunus amygdalus (Almond) Leaves on Selected Bacterial Pathogens

Abstract:

Gold nanoparticles (AuNPs) have gained increasing interest in recent times. This is greatly due to their special features, which include unusual optical and electronic properties, high stability and biological compatibility, controllable morphology and size dispersion, and easy surface functionalization. In typical synthesis, AuNPs were produced by reduction of gold salt AuCl4 in an appropriate solvent. A stabilizing agent was added to prevent the particles from aggregating. The antibacterial activity of different sizes of gold nanoparticles was investigated against Staphylococcus aureus, Salmonella typhi and Pseudomonas pneumonia using the disk diffusion method in a Müeller–Hinton Agar. The Au-NPs were effective against all bacteria tested. That the Au-NPs were successfully synthesized in suspension and were used to study the antibacterial activity of the two medicinal plants against some bacterial pathogens suggests that Au-NPs can be employed as an effective bacteria inhibitor and may be an effective tool in medical field. The study clearly showed that the Au-NPs exhibiting inhibition towards the tested pathogenic bacteria in vitro could have the same effects in vivo and thus may be useful in the medical field if well researched into.

212 Effect of Real Wastewater on Biotransformation of 17α-ethynylestradiol by Ammonia-Oxidizing Bacteria in Nitrifying Activated Sludge

Abstract:

17α-ethynylestradiol (EE2) is a synthetic estrogen used as a key ingredient in an oral contraceptives pill. EE2 is an endocrine disrupting compound, high in estrogenic potency. Although EE2 exhibits low degree of biodegradability with common microorganisms in wastewater treatment plants (WWTPs), this compound can be biotransformed by ammonia-oxidizing bacteria (AOB) via a co-metabolism mechanism in WWTPs. This study aimed to investigate the effect of real wastewater on biotransformation of EE2 by AOB. A preliminary experiment on the effect of nitrite and pH levels on abiotic transformation of EE2 suggested that the abiotic transformation occurred at only pH <6.8. Biotransformation of EE2 under the presence of municipal or industrial wastewater demonstrated that different types of wastewater affect EE2 biotransformation differently. Organic matters in wastewater were believed to deteriorate EE2 biotransformation via the competition effect. At a lower initial ammonium concentration, EE2 biotransformation can be retarded and the extent of the deterioration was COD-concentration dependent. However, when an initial ammonium concentration was elevated, thisphenomena disappeared. This is because when increasing the amount of the primary substrate, more AMO enzymes can be produced resulting in unlimited transformation of all compounds in the tests reducing the competitive effect of organic matters on EE2.

211 Effect of Plant Growth Promoting Bacteria Inoculation, Addition of Biochar, and Mineral N Fertilization on Mycorrhizal Colonization

Abstract:

Strong anthropogenic impact has uncontrolled consequences on the nature of the soil. Hence, up-to-date sustainable methods of soil state improvement are essential. Investigators provide the evidence that biochar can positively effects physical, chemical, and biological soil properties and the abundance of mycorrhizal fungi which are in the focus of this study. The main aim of the present investigation is to demonstrate the effect of two types of plant growth promoting bacteria (PGPB) inoculums along with the beech wood biochar and mineral N additives on mycorrhizal colonization. Experiment has been set up in laboratory conditions with containers filled with arable soil from the protection zone of the main water source “Brezova nad Svitavou”. Lactuca sativa (lettuce) has been selected as a model plant. Based on the obtained data, it can be concluded that mycorrhizal colonization increased as the result of combined influence of biochar and PGPB inoculums amendment. In addition, correlation analyses showed that the numbers of main groups of cultivated bacteria were dependent on the degree of mycorrhizal colonization.

210 PCR based Detection of Food Borne Pathogens

Abstract:

Many high-risk pathogens that cause disease in humans are transmitted through various food items. Food-borne disease constitutes a major public health problem. Assessment of the quality and safety of foods is important in human health. Rapid and easy detection of pathogenic organisms will facilitate precautionary measures to maintain healthy food. The Polymerase Chain Reaction (PCR) is a handy tool for rapid detection of low numbers of bacteria. We have designed gene specific primers for most common food borne pathogens such as Staphylococci, Salmonella and E.coli. Bacteria were isolated from food samples of various food outlets and identified using gene specific PCRs. We identified Staphylococci, Salmonella and E.coli O157 using gene specific primers by rapid and direct PCR technique in various food samples. This study helps us in getting a complete picture of the various pathogens that threaten to cause and spread food borne diseases and it would also enable establishment of a routine procedure and methodology for rapid identification of food borne bacteria using the rapid technique of direct PCR. This study will also enable us to judge the efficiency of present food safety steps taken by food manufacturers and exporters.

209 Characterization of the Microbial Induced Carbonate Precipitation Technique as a Biological Cementing Agent for Sand Deposits

Abstract:

The population increase in Egypt is urging for horizontal land development which became a demand to allow the benefit of different natural resources and expand from the narrow Nile valley. However, this development is facing challenges preventing land development and agriculture development. Desertification and moving sand dunes in the west sector of Egypt are considered the major obstacle that is blocking the ideal land use and development. In the proposed research, the sandy soil is treated biologically using Bacillus pasteurii bacteria as these bacteria have the ability to bond the sand partials to change its state of loose sand to cemented sand, which reduces the moving ability of the sand dunes. The procedure of implementing the Microbial Induced Carbonate Precipitation Technique (MICP) technique is examined, and the different factors affecting on this process such as the medium of bacteria sample preparation, the optical density (OD600), the reactant concentration, injection rates and intervals are highlighted. Based on the findings of the MICP treatment for sandy soil, conclusions and future recommendations are reached.

208 Effects of Ophiocordyceps dipterigena BCC 2073 β-Glucan as a Prebiotic on the in vitro Growth of Probiotic and Pathogenic Bacteria

Abstract:

The  β-glucan produced by Ophiocordyceps dipterigena BCC 2073 is a (1, 3)-β-D-glucan with highly branching O-6-linkedside chains that is resistant to acid hydrolysis (by hydrochloric acid and porcine pancreatic alpha-amylase). This β-glucan can be utilized as a prebiotic due to its advantageous structural and biological properties. The effects of using this β-glucan as the sole carbon source for the in vitro growth of two probiotic bacteria (L. acidophilus BCC 13938 and B. animalis ATCC 25527) were investigated. Compared with the effect of using 1% glucose or fructo-oligosaccharide (FOS) as the sole carbon source, using 1% β-glucan for this purpose showed that this prebiotic supported and stimulated the growth of both types of probiotic bacteria and induced them to produce the highest levels of metabolites during their growth. The highest levels of lactic and acetic acid, 10.04 g·L-1 and 2.82 g·L-1, respectively, were observed at 2 h of cultivation using glucose as the sole carbon source. Furthermore, the fermentation broth obtained using 1% β-glucan as the sole carbon source had greater antibacterial activity against selected pathogenic bacteria (B. subtilis TISTR 008, E. coli TISTR 780, and S. typhimurium TISTR 292) than did the broths prepared using glucose or FOS as the sole carbon source. The fermentation broth obtained by growing L. acidophilus BCC 13938 in the presence of β-glucan inhibited the growth of B. subtilis TISTR 008 by more than 70% and inhibited the growth of both S. typhimurium TISTR 292 and E. coli TISTR 780 by more than 90%. In conclusion, O. dipterigena BCC 2073 is a potential source of a β-glucan prebiotic that could be used for commercial production in the near future.

207 Investigation on Toxicity of Manufactured Nanoparticles to Bioluminescence Bacteria Vibrio fischeri

Abstract:

Acute toxicity of nano SiO2, ZnO, MCM-41 (Meso pore silica), Cu, Multi Wall Carbon Nano Tube (MWCNT), Single Wall Carbon Nano Tube (SWCNT) , Fe (Coated) to bacteria Vibrio fischeri using a homemade luminometer , was evaluated. The values of the nominal effective concentrations (EC), causing 20% and 50% inhibition of biouminescence, using two mathematical models at two times of 5 and 30 minutes were calculated. Luminometer was designed with Photomultiplier (PMT) detector. Luminol chemiluminescence reaction was carried out for the calibration graph. In the linear calibration range, the correlation coefficients and coefficient of Variation (CV) were 0.988 and 3.21% respectively which demonstrate the accuracy and reproducibility of the instrument that are suitable. The important part of this research depends on how to optimize the best condition for maximum bioluminescence. The culture of Vibrio fischeri with optimal conditions in liquid media, were stirring at 120 rpm at a temperature of 150C to 180C and were incubated for 24 to 72 hours while solid medium was held at 180C and for 48 hours. Suspension of nanoparticles ZnO, after 30 min contact time to bacteria Vibrio fischeri, showed the highest toxicity while SiO2 nanoparticles showed the lowest toxicity. After 5 min exposure time, the toxicity of ZnO was the strongest and MCM-41 was the weakest toxicant component.

206 Product Development and Derivatives Exploration by using Photosynthetic Bacteria

Abstract:

Lycopene, which can be extracted from plants and is very popular for fruit intake, is restricted for healthy food development due to its high price. On the other hand, it will get great safety concerns, especially in the food or cosmetic application, if the raw material of lycopene is produced by chemical synthesis. In this project, we provide a key technology to bridge the limitation as mentioned above. Based on the abundant bioresources of BCRC (Bioresource Collection and Research Center, Taiwan), a promising lycopene output will be anticipated by the introduction of fermentation technology along with industry-related core energy. Our results showed that addition of tween 80(0.2%) and span 20 produced higher amount of lycopene. And piperidine, when was added at 48hr to the cultivation medium, could promote lycopene excretion effectively also.

205 An Advanced Approach Based on Artificial Neural Networks to Identify Environmental Bacteria

Abstract:

Environmental micro-organisms include a large number of taxa and some species that are generally considered nonpathogenic, but can represent a risk in certain conditions, especially for elderly people and immunocompromised individuals. Chemotaxonomic identification techniques are powerful tools for environmental micro-organisms, and cellular fatty acid methyl esters (FAME) content is a powerful fingerprinting identification technique. A system based on an unsupervised artificial neural network (ANN) was set up using the fatty acid profiles of standard bacterial strains, obtained by gas-chromatography, used as learning data. We analysed 45 certified strains belonging to Acinetobacter, Aeromonas, Alcaligenes, Aquaspirillum, Arthrobacter, Bacillus, Brevundimonas, Enterobacter, Flavobacterium, Micrococcus, Pseudomonas, Serratia, Shewanella and Vibrio genera. A set of 79 bacteria isolated from a drinking water line (AMGA, the major water supply system in Genoa) were used as an example for identification compared to standard MIDI method. The resulting ANN output map was found to be a very powerful tool to identify these fresh isolates.

204 Characterization and Optimization of Culture Conditions for Sulphur Oxidizing Bacteria after Isolation from Rhizospheric Mustard Soil, Decomposing Sites and Pit House

Abstract:

Sulphur oxidizing bacteria (SOB) have marked their significant role in perspectives of maintaining healthy environment as researchers from all over the world tested and apply these in waste water treatment plants, bioleaching of heavy metals, deterioration of bridge structures, concrete and for bioremediation purposes, etc. Also, these SOB are well adapted in all kinds of environment ranging from normal soil, water habitats to extreme natural sources like geothermal areas, volcanic eruptions, black shale and acid rock drainage (ARD). SOB have been isolated from low pH environment of anthropogenic origin like acid mine drainage (AMD) and bioleaching heaps, hence these can work efficiently in different environmental conditions. Besides having many applications in field of environment science, they may be proven to be very beneficial in area of agriculture as sulphur is the fourth major macronutrients required for the growth of plants. More amount of sulphur is needed by pulses and oilseed crops with respect to the cereal grains. Due to continuous use of land for overproduction of more demanding sulphur utilizing crops and without application of sulphur fertilizers, its concentration is decreasing day by day, and thus, sulphur deficiency is becoming a great problem as it affects the crop productivity and quality. Sulphur is generally found in soils in many forms which are unavailable for plants (cannot be use by plants) like elemental sulphur, thiosulphate which can be taken up by bacteria and converted into simpler forms usable by plants by undergoing a series of transformations. So, keeping the importance of sulphur in view for various soil types, oilseed crops and role of microorganisms in making them available to plants, we made an effort to isolate, optimize, and characterize SOB. Three potential strains of bacteria were isolated, namely SSF7, SSA21, and SSS6, showing sulphate production of concentration, i.e. 2.268, 3.102, and 2.785 mM, respectively. Also, these were optimized for various culture conditions like carbon, nitrogen source, pH, temperature, and incubation time, and characterization was also done.

203 Microbial Evaluation of Geophagic and Cosmetic Clays from Southern and Western Nigeria: Potential Natural Nanomaterials

Abstract:

Geophagic and cosmetic clays are among potential nanomaterial which occur naturally and are of various forms. The use of these nanoclays is a common practice in both rural and urban areas mostly due to tradition and medicinal reasons. These naturally occurring materials can be valuable sources of nanomaterial by serving as nanocomposites. The need to ascertain the safety of these materials is the motivation for this research. Physical Characterization based on the hue value and microbiological qualities of the nanoclays were carried out. The Microbial analysis of the clay samples showed considerable contamination with both bacteria and fungi with fungal contaminants taking the lead. This observation may not be unlikely due to the ability of fungi species to survive harsher growth conditions than bacteria. ‘Atike pupa’ showed no bacterial growth. The clay with the largest bacterial count was Calabash chalk (Igbanke), while that with the highest fungal count was ‘Eko grey’. The most commonly isolated bacteria in this study were Clostridium spp. and Corynebacterium spp. while fungi included Aspergillus spp. These results are an indication of the need to subject these clay materials to treatments such as heating before consumption or topical usage thereby ascertaining their safety.

202 Characterization of Lactose Consumption during the Biogas Production from Acid Whey by FT-IR Spectroscopy

Abstract:

The consumption of lactose in acid cheese whey anaerobic fermentation process under fed-batch conditions was studied. During fermentation for 100 hours the biogas production (CO2 and CH4) was analyzed online. Among the standard analyses FT-IR spectroscopy was used to follow the consumption of lactose by bacteria. The absorption bands at 990, 894 and 787 cm-1 in the 2nd derivative spectra were shown to be characteristic for lactose and were used to follow the lactose conversion. It was shown that acid cheese whey lactose was converted by bacteria in first 7 hours. In the spectra of 17, 18 and 95 hour fermentation samples lactose was not identified and these results correlated with the HPLC data.
