1682 A Heat-Inducible Transgene Expression System for Gene Therapy
Authors: Masaki Yamaguchi, Akira Ito, Noriaki Okamoto, Yoshinori Kawabe, Masamichi Kamihira
Abstract:
Heat-inducible gene expression vectors are useful for hyperthermia-induced cancer gene therapy, because the combination of hyperthermia and gene therapy can considerably improve the therapeutic effects. In the present study, we developed an enhanced heat-inducible transgene expression system in which a heat-shock protein (HSP) promoter and tetracycline-responsive transactivator were combined. When the transactivator plasmid containing the tetracycline-responsive transactivator gene was co-transfected with the reporter gene expression plasmid, a high level of heat-induced gene expression was observed compared with that using the HSP promoter without the transactivator. In vitro evaluation of the therapeutic effect using HeLa cells showed that heat-induced therapeutic gene expression caused cell death in a high percentage of these cells, indicating that this strategy is promising for cancer gene therapy. Keywords: Inducible gene expression, Gene therapy, Hyperthermia, Heat shock protein, Tetracycline transactivator.

Authors: L. Parisi
Abstract:
In this research work, neural networks were applied to classify two types of hip joint implants based on the relative hip joint implant side speed and three components of each ground reaction force. The condition of walking gait at normal velocity was used and carried out with each of the two hip joint implants assessed. Ground reaction forces’ kinetic temporal changes were considered in the first approach followed but discarded in the second one. Ground reaction force components were obtained from eighteen patients under such gait condition, half of which had a hip implant type I-II, whilst the other half had the hip implant, defined as type III by Orthoload®. After pre-processing raw gait kinetic data and selecting the time frames needed for the analysis, the ground reaction force components were used to train a MLP neural network, which learnt to distinguish the two hip joint implants in the abovementioned condition. Further to training, unknown hip implant side and ground reaction force components were presented to the neural networks, which assigned those features into the right class with a reasonably high accuracy for the hip implant type I-II and the type III. The results suggest that neural networks could be successfully applied in the performance assessment of hip joint implants.

Abstract:
The present paper is an experimental investigation of roughness effects on nucleate pool boiling of refrigerant R113 on horizontal circular copper surfaces. The copper samples were treated by different sand paper grit sizes to achieve different surface roughness. The average surface roughness of the four samples was 0.901, 0.735, 0.65, and 0.09, respectively. The experiments were performed in the heat flux range of 8 to 200kW/m2. The heat transfer coefficient was calculated by measuring wall superheat of the samples and the input heat flux. The results show significant improvement of heat transfer coefficient as the surface roughness is increased. It is found that the heat transfer coefficient of the sample with Ra=0.901 is 3.4, 10.5, and 38.5% higher in comparison with surfaces with Ra of 0.735, 0.65, and 0.09 at heat flux of 170 kW/m2. Moreover, the results are compared with literature data and the well known Cooper correlation. Keywords: Nucleate Boiling, Pool Boiling, R113, SurfaceRoughness

Abstract:
In a nuclear reactor, an array of fuel rods containing stacked uranium dioxide pellets clad with zircalloy is the heat source for a thermodynamic cycle of energy conversion from heat to electricity. After fuel is used in a nuclear reactor, the assemblies are stored underwater in a spent nuclear fuel pool at the nuclear power plant while heat generation and radioactive decay rates decrease before it is placed in packages for dry storage or transportation. A computational model of a Boiling Water Reactor spent fuel assembly is modeled using FLUENT, the computational fluid dynamics package. Heat transfer simulations were performed on the two-dimensional 9x9 spent fuel assembly to predict the maximum cladding temperature for different input to the FLUENT model. Uncertainty quantification is used to predict the heat transfer and the maximum temperature profile inside the assembly. Keywords: Spent nuclear fuel, conduction, heat transfer, uncertainty quantification.

Abstract:
This study presents the numerical simulation of three-dimensional incompressible steady and laminar fluid flow and conjugate heat transfer of a trapezoidal microchannel heat sink using water as a cooling fluid in a silicon substrate. Navier-Stokes equations with conjugate energy equation are discretized by finite-volume method. We perform numerical computations for a range of 50 ≦ Re ≦ 600, 0.05W ≦ P ≦ 0.8W, 20W/cm2 ≦q"≦ 40W/cm2. The present study demonstrates the numerical optimization of a trapezoidal microchannel heat sink design using the response surface methodology (RSM) and the genetic algorithm method (GA). The results show that the average Nusselt number increases with an increase in the Reynolds number or pumping power, and the thermal resistance decreases as the pumping power increases. The thermal resistance of a trapezoidal microchannel is minimized for a constant heat flux and constant pumping power.

Abstract:
Prolonged immobilization leads to significant weakness and atrophy of the skeletal muscle and can also impair the recovery of muscle strength following injury. Therefore, it is important to minimize the period under immobilization and accelerate the return to normal activity. This study examined the effects of heat treatment and rest-inserted exercise on the muscle activity of the lower limb during knee flexion/extension. Twelve healthy subjects were assigned to 4 groups that included: (1) heat treatment + rest-inserted exercise; (2) heat + continuous exercise; (3) no heat + rest-inserted exercise; and (4) no heat + continuous exercise. Heat treatment was applied for 15 mins prior to exercise. Continuous exercise groups performed knee flexion/extension at 0.5 Hz for 300 cycles without rest whereas rest-inserted exercise groups performed the same exercise but with 2 mins rest inserted every 60 cycles of continuous exercise. Changes in the rectus femoris and hamstring muscle activities were assessed at 0, 1, and 2 weeks of treatment by measuring the electromyography signals of isokinetic maximum voluntary contraction. Significant increases in both the rectus femoris and hamstring muscles were observed after 2 weeks of treatment only when both heat treatment and rest-inserted exercise were performed. These results suggest that combination of various treatment techniques, such as heat treatment and rest-inserted exercise, may expedite the recovery of muscle strength following immobilization. Keywords: Electromyography, Heat Treatment, Muscle, Rest-Inserted Exercise.

Experimental investigation of heat transfer and friction factor characteristics of circular tube fitted with 300 right-left helical screw inserts with 100 mm spacer of different twist ratio has been presented for laminar and turbulent flow.. The experimental data obtained were compared with those obtained from plain tube published data. The heat transfer coefficient enhancement for 300 RL inserts with 100 mm spacer is quite comparable with for 300 R-L inserts. Performance evaluation analysis has been made and found that the performance ratio increases with increasing Reynolds number and decreasing twist ration with the maximum for the twist ratio 2.93. Abstract:
Heat transfer enhancement objects like extended surfaces, fins etc. are chosen for their thermal performance as well as for other design parameters depending on various applications. The present paper is on experimental study to investigate the heat transfer enhancement through wire mesh fin arrays equipped with horizontal base plate. The data used in performance analysis were obtained experimentally for the material (mild steel) for different heat inputs such as 40, 60, 80, 100 and 120 watt, by varying wire mesh diameter, fin height and spacing between two fin arrays. Using the Taguchi experimental design method, optimum design parameters and their levels were investigated. Average heat transfer coefficient was considered as a performance characteristic parameter. An L9 (33) orthogonal array was selected as an experimental plan. Optimum results were found by experimenting. It is observed that the wire mesh diameter and fin height have a higher impact on heat transfer coefficient as compared to spacing between two fin arrays. Keywords: Heat transfer enhancement, finned surface, wire mesh diameter, natural convection.

Abstract:
Heat source addition to the axisymmetric supersonic inlet may improve the performance parameters, which will increase the inlet efficiency. In this investigation the heat has been added to the flow field at some distance ahead of an axisymmetric inlet by adding an imaginary thermal source upstream of cowl lip. The effect of heat addition on the drag coefficient, mass flow rate and the overall efficiency of the inlet have been investigated. The results show that heat addition causes flow separation, hence to prevent this phenomena, roughness has been added on the spike surface. However, heat addition reduces the drag coefficient and the inlet mass flow rate considerably. Furthermore, the effects of position, size, and shape on the inlet performance were studied. It is found that the thermal source deflects the flow streamlines. By improper location of the thermal source, the optimum condition has been obtained. For the optimum condition, the drag coefficient is considerably reduced and the inlet mass flow rate and its efficiency have been increased slightly. The optimum shape of the heat source is obtained too. Keywords: Drag coefficient, heat source, performanceparameters, supersonic inlet.

Abstract:
Solar panels that use photovoltaic (PV) cells are popular for converting solar radiation into electricity. One of the major problems impacting the performance of PV panels is the overheating caused by excessive solar radiation and high ambient temperatures, which degrades the efficiency of the PV panels remarkably. To overcome this issue, an aluminum heat sink was used to dissipate unwanted heat from PV cells. The dimensions of the heat sink were determined considering the optimal fin spacing that fulfils hot climatic conditions. In this study, the effects of cooling on the efficiency and power output of a PV panel were studied experimentally. Two PV modules were used: one without and one with a heat sink. The experiments ran for 11 hours from 6:00 a.m. to 5:30 p.m. where temperature readings in the rear and front of both PV modules were recorded at an interval of 15 minutes using sensors and an Arduino microprocessor. Results are recorded for both panels simultaneously for analysis, temperate comparison, and for power and efficiency calculations. A maximum increase in the solar to electrical conversion efficiency of 35% and almost 55% in the power output were achieved with the use of a heat sink, while temperatures at the front and back of the panel were reduced by 9% and 11%, respectively. Keywords: Photovoltaic cell, natural convection, heat sink, efficiency.

Abstract:
Methods for measuring or estimating ground shape by a laser range finder and a vision sensor (Exteroceptive sensors) have critical weaknesses in terms that these methods need a prior database built to distinguish acquired data as unique surface conditions for driving. Also, ground information by Exteroceptive sensors does not reflect the deflection of ground surface caused by the movement of UGVs. Therefore, this paper proposes a method of recognizing exact and precise ground shape using an Inertial Measurement Unit (IMU) as a proprioceptive sensor. In this paper, firstly this method recognizes the attitude of a robot in real-time using IMU and compensates attitude data of a robot with angle errors through analysis of vehicle dynamics. It is found that there exists an optimum value for depth of dimples in which the largest wall heat flux can be achieved. Also, the results show a critical value for the ratio of wall thermal conductivity to the one of fluid in which the dependence of wall heat flux to this ratio almost vanishes. Abstract:
This paper reports on the effects of heat treatment on 3CR12 and AISI 316 stainless steel grades. Heat treatment was conducted on the steel grades and cooled using two different media; air and water in order to study the effect of each medium on the evolving properties of the samples. The heat treated samples were characterized through the evolving microstructure and hardness. It was found that there was a significant grain size reduction in both the heat treated stainless steel specimens compared to the parent materials. The finer grain sizes were achieved as a result of impediment to growth of one phase by the other. The Vickers microhardness values of the heat treated samples were higher compared to the parent materials due to the fact that each of the steel grades had a proportion of martensitic structures in their microstructures thereby improving the integrity of the material.

Abstract:
This study evaluated to facilitate separation of ABS plastics from other waste plastics by froth flotation after surface hydrophilization of ABS with heat treatment. The mild heat treatment at 100oC for 60s could selectively increase the hydrophilicity of the ABS plastics surface (i.e., ABS contact angle decreased from 79o to 65.8o) among other plastics mixture. The SEM and XPS results of plastic samples sufficiently supported the increase in hydrophilic functional groups and decrease contact angle on ABS surface, after heat treatment. As a result of the froth flotation (at mixing speed 150 rpm and airflow rate 0.3 L/min) after heat treatment, about 85% of ABS was selectively separated from other heavy plastics with 100% of purity. The effect of optimum treatment condition and detailed mechanism onto separation efficiency in the froth floatation was also investigated. Abstract:
The hydrodynamic and thermal lattice Boltzmann methods are applied to investigate the turbulent convective heat transfer in the wavy channel flows. In this study, the turbulent phenomena are modeling by large-eddy simulations with the Smagorinsky model. As a benchmark, the laminar and turbulent backward-facing step flows are simulated first. The results give good agreement with other numerical and experimental data. For wavy channel flows, the distribution of Nusselt number and the skin-friction coefficients are calculated to evaluate the heat transfer effect and the drag force. It indicates that the vortices at the trough would affect the magnitude of drag and weaken the heat convection effects on the wavy surface. In turbulent cases, if the amplitude of the wavy boundary is large enough, the secondary vortices would be generated at troughs and contribute to the heat convection. Finally, the effects of different Re on the turbulent transport phenomena are discussed.

Authors: Alireza Mortezaei, Kimia Mortezaei
Abstract:
Recordings from recent earthquakes have provided evidence that ground motions in the near field of a rupturing fault differ from ordinary ground motions, as they can contain a large energy, or “directivity" pulse. This pulse can cause considerable damage during an earthquake, especially to structures with natural periods close to those of the pulse. Failures of modern engineered structures observed within the near-fault region in recent earthquakes have revealed the vulnerability of existing RC buildings against pulse-type ground motions. This may be due to the fact that these modern structures had been designed primarily using the design spectra of available standards, which have been developed using stochastic processes with relatively long duration that characterizes more distant ground motions. Many recently designed and constructed buildings may therefore require strengthening in order to perform well when subjected to near-fault ground motions. Fiber Reinforced Polymers are considered to be a viable alternative, due to their relatively easy and quick installation, low life cycle costs and zero maintenance requirements. The objective of this paper is to investigate the adequacy of Artificial Neural Networks (ANN) to determine the three dimensional dynamic response of FRP strengthened RC buildings under the near-fault ground motions. For this purpose, one ANN model is proposed to estimate the base shear force, base bending moments and roof displacement of buildings in two directions. A training set of 168 and a validation set of 21 buildings are produced from FEA analysis results of the dynamic response of RC buildings under the near-fault earthquakes. It is demonstrated that the neural network based approach is highly successful in determining the response.

Maxwell-Garnetts (MG) and Brinkman models have been utilized for calculating the effective thermal conductivity and dynamic viscosity of nanofluid, respectively. Influences of Rayleigh number (Ra = 103−106), nanoparticle volume concentration (f = 0-0.04) and Hartmann number (Ha = 0-90) on the flow and heat transfer characteristics have been examined. The results indicate that the Hartmann number influences the heat transfer at Ra = 106 more than other Raleigh numbers, as the least effect is observed at Ra = 103. Although the TEG hot-side temperature and thus the electric power output can be increased by inserting an elliptical pin-fin heat sink into the chimney tunnel to increase the heat transfer area, the pin fin heat sink would cause extra pumping power at the same time. The main purpose of this study is to analyze the effects of geometrical parameters on the electric power output and chimney pressure drop characteristics. The effects of different operating conditions, including various inlet velocities (Vin= 1, 3, 5 m/s), inlet temperatures (Tgas = 450, 550, 650K) and different fin height (0 to 150 mm) are discussed in detail. The predicted numerical data for the power vs. current (P-I) curve are in good agreement (within 11%) with the experimental data.
