Authors: Shweta Kumari, Parmjit S. Panesar, Manab B. Bera
Abstract:
The hydrolysis of lactose using β-galactosidase is one of the most promising biotechnological applications, which has wide range of potential applications in food processing industries. However, due to intracellular location of the yeast enzyme, and expensive extraction methods, the industrial applications of enzymatic hydrolysis processes are being hampered. The use of permeabilization technique can help to overcome the problems associated with enzyme extraction and purification of yeast cells and to develop the economically viable process for the utilization of whole cell biocatalysts in food industries. In the present investigation, standardization of permeabilization process of novel yeast isolate was carried out using a statistical model approach known as Response Surface Methodology (RSM) to achieve maximal b-galactosidase activity. The optimum operating conditions for permeabilization process for optimal β-galactosidase activity obtained by RSM were 1:1 ratio of toluene (25%, v/v) and ethanol (50%, v/v), 25.0 oC temperature and treatment time of 12 min, which displayed enzyme activity of 1.71 IU /mg DW.

Abstract:
A. niger XP isolated from Vietnam produces very low amount of acidic phytase with optimal pH at 2.5 and 5.5. The phytase production of this strain was successfully improved through gene cloning and expression. A 1.4 - kb DNA fragment containing the coding region of the phyA gene was amplified by PCR and inserted into the expression vector pPICZαA with a signal peptide α- factor, under the control of AOX1 promoter. The recombined plasmid was transformed into the host strain P. pastoris KM71H and X33 by electroporation. Both host strains could efficiently express and secret phytase. The multicopy strains were screened for over expression of phytase. All the selected multicopy strains of P. pastoris X33 were examined for phytase activity, the maximum phytase yield of 1329 IU/ml was obtained after 4 days of incubation in medium BMM. The recombinant protein with MW of 97.4 KW showed to be the only one protein secreted in the culture broth. Multicopy transformant P. pastoris X33 supposed to be potential candidate for producing the commercial preparation of phytase.

Abstract:
The fault current levels through the electric devices have a significant impact on failure probability. New fault current results in exceeding the rated capacity of circuit breaker and switching equipments and changes operation characteristic of overcurrent relay. In order to solve these problems, SFCL (Superconducting Fault Current Limiter) has rising as one of new alternatives so as to improve these problems. A fault current reduction differs depending on installed location. Therefore, a location of SFCL is very important. Also, SFCL decreases the fault current, and it prevents surrounding protective devices to be exposed to fault current, it then will bring a change of reliability. In this paper, we propose method which determines the optimal location when SFCL is installed in power system. In addition, the reliability about the power system which SFCL was installed is evaluated. The efficiency and effectiveness of this method are also shown by numerical examples and the reliability indices are evaluated in this study at each load points. Authors: Dominik Böhm, Konrad Czerski
Abstract:
A customized two-stage model has been developed to simulate, analyse, and visualize distillation of actinides as a useful alternative low-pressure separation method in the nuclear recycling cases. Under the most optimal conditions of idealized thermodynamic equilibrium stages and under total reflux of distillate the investigated cases of chloride systems for the separation of such actinides are (A) UCl4-CsCl-PuCl3 and (B) ThCl4-NaCl-PuCl3. Simulatively, uranium tetrachloride in case A is successfully separated by distillation into a six-stage distillation column, and thorium tetrachloride from case B into an eight-stage distillation column. For this, a permissible mole fraction value of 1E-06 has been assumed for the residual impurification degree. With further separation effort of eleven to seventeen required separation stages, the monochlorides of plutonium trichloride from both systems A and B are simulatively shown to be separated as high pure distillation products.

Authors: Ritsuko Kawasaki (Aiba), Takeshi Hiromatsu
Abstract:
Management is required to understand all information security risks within an organization, and to make decisions on which information security risks should be treated in what level by allocating how much amount of cost. However, such decision-making is not usually easy, because various measures for risk treatment must be selected with the suitable application levels. In addition, some measures may have objectives conflicting with each other. It also makes the selection difficult. Moreover, risks generally have trends and it also should be considered in risk treatment. Therefore, this paper provides the extension of the model proposed in the previous study. The original model supports the selection of measures by applying a combination of weighted average method and goal programming method for multi-objective analysis to find an optimal solution. The extended model includes the notion of weights to the risks, and the larger weight means the priority of the risk.

It helps to provide early warning for a special event in a process, and finding its assignable cause can be obtained. This work presents a hybrid diagnostic schmes for batch processes. Nonlinear representation of raw process data is combined with classification tree techniques. The nonlinear kernel-based dimension reduction is executed for nonlinear classification decision boundaries for fault classes. In order to enhance diagnosis performance for batch processes, filtering of the data is performed to get rid of the irrelevant information of the process data. Abstract:
Fiber reinforced concrete is important material for load bearing structural elements. Usually fibers are homogeneously distributed in a concrete body having arbitrary spatial orientations. At the same time, in many situations, fiber concrete with oriented fibers is more optimal. Is obvious, that is possible to create constructions with oriented short fibers in them, in different ways. Present research is devoted to one of such approaches- fiber reinforced concrete prisms having dimensions 100mm ×100mm ×400mmwith layers of non-homogeneously distributed fibers inside them were fabricated.
Simultaneously prisms with homogeneously dispersed fibers were produced for reference as well. Prisms were tested under four point bending conditions. During the tests vertical deflection at the center of every prism and crack opening were measured (using linear displacements transducers in real timescale). Prediction results were discussed.

Abstract:
Network coding has recently attracted attention as an efficient technique in multicast/broadcast services. The problem of finding the optimal network coding mechanism maximizing the bandwidth efficiency is hard to solve and hard to approximate. Lots of network coding-based schemes have been suggested in the literature to improve the bandwidth efficiency, especially network coding-based automatic repeat request (NCARQ) schemes. However, existing schemes have several limitations which cause the performance degradation in resource limited systems. To improve the performance in resource limited systems, we propose NCARQ with overlapping selection (OS-NCARQ) scheme. The advantages of OS-NCARQ scheme over the traditional ARQ scheme and existing NCARQ schemes are shown through the analysis and simulations.

This paper presents current understanding of morphological traits in pineapple roots, highlighting studies using incubation periods and various solid MS media treated with different sucrose concentrations and pH, which directly assess in vitro environmental factors. Rooting parameters had different optimal sucrose concentrations and incubation periods. All shoots failed to root in medium supplemented with sucrose at 5 g/L and no roots formed within the first 45 days in medium enriched with sucrose at 10 g/L. After 75 days, all shoots rooted in medium enriched with 10 and 20 g/L sucrose. Moreover, MS medium supplied with 20 g/L sucrose resulted in the longest and the highest number of roots with 27.3 mm and 4.7, respectively. Root function, such as capacity for P and N uptake, declined rapidly with root length. Abstract:
Electric cars with their fast innovation cycles and their disruptive character offer a high degree of freedom regarding innovative design for remanufacturing. Remanufacturing increases not only the resource but also the economic efficiency by a prolonged product life time. The reduced power train wear of electric cars combined with high manufacturing costs for batteries allow new business models and even second life applications. Modular and intermountable designed battery packs enable the replacement of defective or outdated battery cells, allow additional cost savings and a prolongation of life time. This paper discusses opportunities for future remanufacturing value chains of electric cars and their battery components and how to address their potentials with elaborate designs. Based on a brief overview of implemented remanufacturing structures in different industries, opportunities of transferability are evaluated. In addition to an analysis of current and upcoming challenges, promising perspectives for a sustainable electric car circular economy enabled by design for remanufacturing are deduced. Two mathematical models describe the feasibility of pursuing a circular economy of lithium ion batteries and evaluate remanufacturing in terms of sustainability and economic efficiency. Taking into consideration not only labor and material cost but also capital costs for equipment and factory facilities to support the remanufacturing process, cost benefit analysis prognosticate that a remanufacturing battery can be produced more cost-efficiently. The ecological benefits were calculated on a broad database from different research projects which focus on the recycling, the second use and the assembly of lithium ion batteries. The results of this calculations show a significant improvement by remanufacturing in all relevant factors especially in the consumption of resources and greenhouse warming potential. Exemplarily suitable design guidelines for future remanufacturing lithium ion batteries, which consider modularity, interfaces and disassembly, are used to illustrate the findings. For one guideline, potential cost improvements were calculated and upcoming challenges are pointed out.

Authors: Piyali Chaudhuri, Peter T. Martin, Aleksandar Z. Stevanovic, Chongkai Zhu
Abstract:
Loop detectors report traffic characteristics in real time. They are at the core of traffic control process. Intuitively, one would expect that as density of detection increases, so would the quality of estimates derived from detector data. However, as detector deployment increases, the associated operating and maintenance cost increases. Thus, traffic agencies often need to decide where to add new detectors and which detectors should continue receiving maintenance, given their resource constraints. This paper evaluates the effect of detector spacing on freeway travel time estimation. A freeway section (Interstate-15) in Salt Lake City metropolitan region is examined. The research reveals that travel time accuracy does not necessarily deteriorate with increased detector spacing. Rather, the actual location of detectors has far greater influence on the quality of travel time estimates. The study presents an innovative computational approach that delivers optimal detector locations through a process that relies on Genetic Algorithm formulation. Keywords: Detector, Freeway, Genetic algorithm, Travel timeestimate.

Abstract:
Oil debris signal generated from the inductive oil debris monitor (ODM) is useful information for machine condition monitoring but is often spoiled by background noise. To improve the reliability in machine condition monitoring, the high-fidelity signal has to be recovered from the noisy raw data. Considering that the noise components with large amplitude often have higher frequency than that of the oil debris signal, the integral transform is proposed to enhance the detectability of the oil debris signal. To cancel out the baseline wander resulting from the integral transform, the empirical mode decomposition (EMD) method is employed to identify the trend components. An optimal reconstruction strategy including both de-trending and de-noising is presented to detect the oil debris signal with less distortion. The proposed approach is applied to detect the oil debris signal in the raw data collected from an experimental setup. The result demonstrates that this approach is able to detect the weak oil debris signal with acceptable distortion from noisy raw data. Keywords: Integral transform, empirical mode decomposition, oil debris, signal processing, detection.

Authors: George Yungchih Wang
Abstract:
This paper argues that increased uncertainty, in certain situations, may actually encourage investment. Since earlier studies mostly base their arguments on the assumption of geometric Brownian motion, the study extends the assumption to alternative stochastic processes, such as mixed diffusion-jump, mean-reverting process, and jump amplitude process. A general approach of Monte Carlo simulation is developed to derive optimal investment trigger for the situation that the closed-form solution could not be readily obtained under the assumption of alternative process. The main finding is that the overall effect of uncertainty on investment is interpreted by the probability of investing, and the relationship appears to be an invested U-shaped curve between uncertainty and investment. The implication is that uncertainty does not always discourage investment even under several sources of uncertainty. Furthermore, high-risk projects are not always dominated by low-risk projects because the high-risk projects may have a positive realization effect on encouraging investment. Keywords: real options, geometric Brownian motion, mixeddiffusion-jump process, mean- reverting process, jump amplitudeprocess

Abstract:
In this research, we have developed a new efficient heuristic algorithm for the dynamic facility layout problem with budget constraint (DFLPB). This heuristic algorithm combines two mathematical programming methods such as discrete event simulation and linear integer programming (IP) to obtain a near optimum solution. In the proposed algorithm, the non-linear model of the DFLP has been changed to a pure integer programming (PIP) model. Then, the optimal solution of the PIP model has been used in a simulation model that has been designed in a similar manner as the DFLP for determining the probability of assigning a facility to a location. After a sufficient number of runs, the simulation model obtains near optimum solutions. Finally, to verify the performance of the algorithm, several test problems have been solved. The results show that the proposed algorithm is more efficient in terms of speed and accuracy than other heuristic algorithms presented in previous works found in the literature. Keywords: Budget constraint, Dynamic facility layout problem, Integer programming, Simulation

This paper describes an investigation on designing and enhancing the performance of H-Darrieus Wind Turbine (HDWT) of 10 kW at the medium wind speed. The aerodynamic characteristics of this turbine were investigated by both theoretical and numerical approaches. The optimal design procedure was first proposed to enhance the power coefficient under various effects, such as airfoil type, number of blades, solidity, aspect ratio, and tip speed ratio. As a result, the overall design of the 10 kW HDWT was well achieved, and the power characteristic of this turbine was found by numerical approach. Additionally, the maximum power coefficient predicted is up to 0.41 at the tip speed ratio of 3.7 and wind speed of 8 m/s. Particularly, a generalized correlation of power coefficient with tip speed ratio and wind speed is first proposed. Improved thermal, electrical and exergy models are used to calculate the thermal and electrical parameters, overall energy efficiency, exergy components and exergy efficiency of a typical PV/T air collector. A computer simulation program is also developed. The results of numerical simulation are in good agreement with the experimental measurements noted in the previous literature. Finally, multi-objective optimization has been carried out under given climatic, operating and design parameters. The optimized ranges of inlet air velocity, duct depth and the objective functions in optimal Pareto front have been obtained. Furthermore, non-sensitive solutions from energy or exergy point of view in the results of multi-objective optimization have been shown. Keywords: Solar photovoltaic thermal (PV/T) air collector, Overall energy efficiency, Exergy efficiency, Multi-objectiveoptimization, Sensitivity analysis.

Abstract:
A total of 115 yeast strains isolated from local cassava processing wastes were measured for crude protein content. Among these strains, the strain MSY-2 possessed the highest protein concentration (>3.5 mg protein/mL). By using molecular identification tools, it was identified to be a strain of Pichia kudriavzevii based on similarity of D1/D2 domain of 26S rDNA region. In this study, to optimize the protein production by MSY-2 strain, Response Surface Methodology (RSM) was applied. The tested parameters were the carbon content, nitrogen content, and incubation time. Here, the value of regression coefficient (R2) = 0.7194 could be explained by the model which is high to support the significance of the model. Under the optimal condition, the protein content was produced up to 3.77 g per L of the culture and MSY-2 strain contains 66.8 g protein per 100 g of cell dry weight. These results revealed the plausibility of applying the novel strain of yeast in single-cell protein production. Keywords: Single cell protein, response surface methodology, yeast, cassava processing waste.
