248 Antimicrobial Properties of Copper in Gram-Negative and Gram-Positive Bacteria

Abstract:

For centuries humans have used the antimicrobial properties of copper to their advantage. Yet, after all these years the underlying mechanisms of copper mediated cell death in various microbes remain unclear. We had explored the hypothesis that copper mediated increased levels of lipid peroxidation in the membrane fatty acids is responsible for increased killing in Escherichia coli. In this study we show that in both gram positive (Staphylococcus aureus) and gram negative (Pseudomonas aeruginosa) bacteria there is a strong correlation between copper mediated cell death and increased levels of lipid peroxidation. Interestingly, the non-spore forming gram positive bacteria as well as gram negative bacteria show similar patterns of cell death, increased levels of lipid peroxidation, as well as genomic DNA degradation, however there is some difference in loss in membrane integrity upon exposure to copper alloy surface.

247 Integrated Cultivation Technique for Microbial Lipid Production by Photosynthetic Microalgae and Locally Oleaginous Yeast

Abstract:

The objective of this research is to study of microbial lipid production by locally photosynthetic microalgae and oleaginous yeast via integrated cultivation technique using CO2 emissions from yeast fermentation. A maximum specific growth rate of Chlorella sp. KKU-S2 of 0.284 (1/d) was obtained under an integrated cultivation and a maximum lipid yield of 1.339g/L was found after cultivation for 5 days, while 0.969g/L of lipid yield was obtained after day 6 of cultivation time by using CO2 from air. A high value of volumetric lipid production rate (QP, 0.223 g/L/d), specific product yield (YP/X, 0.194), volumetric cell mass production rate (QX, 1.153 g/L/d) were found by using ambient air CO2 coupled with CO2 emissions from yeast fermentation. Overall lipid yield of 8.33 g/L was obtained (1.339 g/L of Chlorella sp. KKU-S2 and 7.06g/L of T. maleeae Y30) while low lipid yield of 0.969g/L was found using non-integrated cultivation technique. To our knowledge this is the unique report about the lipid production from locally microalgae Chlorella sp. KKU-S2 and yeast T. maleeae Y30 in an integrated technique to improve the biomass and lipid yield by using CO2 emissions from yeast fermentation.

246 Bacteriological Screening and Antibiotic – Heavy Metal Resistance Profile of the Bacteria Isolated from Some Amphibian and Reptile Species of the Biga Stream in Turkey

Abstract:

In this article, the antibiogram and heavy metal resistance profile of the bacteria isolated from total 34 studied animals (Pelophylax ridibundus = 12; Mauremys rivulata = 14; Natrix natrix = 8) captured around the Biga Stream, are described. There was no database information on antibiogram and heavy metal resistance profile of bacteria from these area’s amphibians and reptiles. A total of 200 bacteria were successfully isolated from cloaca and oral samples of the aquatic amphibians and reptiles as well as from the water sample. According to Jaccard’s similarity index, the degree of similarity in the bacterial flora was quite high among the amphibian and reptile species under examination, whereas it was different from the bacterial diversity in the water sample. The most frequent isolates were A. hydrophila (31.5%), B. pseudomallei (8.5%), and C. freundii (7%). The total numbers of bacteria obtained were as follows: 45 in P. ridibundus, 45 in N. natrix 30 in M. rivulata, and 80 in the water sample. The result showed that cefmetazole was the most effective antibiotic to control the bacteria isolated in this study and that approximately 93.33% of the bacterial isolates were sensitive to this antibiotic. The multiple antibiotic resistances (MAR) index indicated that P. ridibundus (0.95) > N. natrix (0.89) > M. rivulata (0.39). Furthermore, all the tested heavy metals (Pb+2, Cu+2, Cr+3, and Mn+2) inhibit the growth of the bacterial isolates at different rates. Therefore, it indicated that the water source of the animals was contaminated with both antibiotic residues and heavy metals.

245 Production of Glucose from the Hydrolysis of Cassava Residue using Bacteria Isolates from Thai Higher Termites

Abstract:

The possibility of using cassava residue containing 49.66% starch, 21.47% cellulose, 12.97% hemicellulose, and 21.86% lignin as a raw material to produce glucose using enzymatic hydrolysis was investigated. In the experiment, each reactor contained the cassava residue, bacteria cells, and production medium. The effects of particles size (40 mesh and 60 mesh) and strains of bacteria (A002 and M015) isolated from Thai higher termites, Microcerotermes sp., on the glucose concentration at 37°C were focused. High performance liquid chromatography (HPLC) with a refractive index detector was used to determine the quantity of glucose. The maximum glucose concentration obtained at 37°C using strain A002 and 60 mesh of the cassava residue was 1.51 g/L at 10 h.

Abstract:

Today, biogenic magnetite nanoparticles among magnetic nanoparticles have unique attracted attention because of their magnetic characteristics and potential applications in various fields such as therapeutic and diagnostic. A well known example of these biogenic nanoparticles is magnetosomes of magnetotactic bacteria. In this research, we used two different types of technique for the isolation and purification of magnetosome nanoparticles from the isolated magnetotactic bacterial cells, heat-alkaline treatment and sonication. Also we evaluated pyrogen content and sterility of synthesized the isolated individual magnetosome by the Limulus Amoebocyte Lysate test and direct impedimetric method respectively.

242 Identification of Anaerobic Microorganisms for Converting Kitchen Waste to Biogas

Abstract:

Anaerobic digestion process is one of the alternative methods to convert organic waste into methane gas which is a fuel and energy source. Activities of various kinds of microorganisms are the main factor for anaerobic digestion which produces methane gas. Therefore, in this study a modified Anaerobic Baffled Reactor (ABR) with working volume of 50 liters was designed to identify the microorganisms through biogas production. The mixture of 75% kitchen waste and 25% sewage sludge was used as substrate. Observations on microorganisms in the ABR showed that there exists a small amount of protozoa (5%) and fungi (2%) in the system, but almost 93% of the microorganism population consists of bacteria. It is definitely clear that bacteria are responsible for anaerobic biodegradation of kitchen waste. Results show that in the acidification zone of the ABR (front compartments of reactor) fast growing bacteria capable of growth at high substrate levels and reduced pH was dominant. A shift to slower growing scavenging bacteria that grow better at higher pH was occurring towards the end of the reactor. Due to the ability of activity in acetate environment the percentages of Methanococcus, Methanosarcina and Methanotrix were higher than other kinds of methane former in the system.

241 Manipulation of Probiotics Fermentation of Yogurt by Cinnamon and Licorice: Effects on Yogurt Formation and Inhibition of Helicobacter Pylori Growth in vitro

Abstract:

Probiotic bacteria especially Lactobacillus spp. and Bifidobacterium exert suppressive effect on Helicobacter pylori. Cinnamon and licorice have been traditionally used for the treatment of gastric ulcer. The objectives of this study were to determine the effects of herbs on yogurt fermentation, the level of probiotic bacteria in yogurt during 28 days storage and the effect of herbal yogurt on the growth of H. pylori in vitro. Cinnamon or licorice was mixed with milk and the mixture was fermented with probiotic bacteria to form herbal-yogurt. Changes of pH and total titratable acids were monitored and the viability of probiotic bacteria was evaluated during and after refrigerated storage. The in vitro inhibition of H. pylori growth was determined using agar diffusion and minimum inhibitory concentration (MIC) method. The presence of herbs did not affect the probiotic population during storage. There were no significant differences in pH and TTA between herbal-yogurts and plain-yogurt during fermentation and storage. Water extract of cinnamon-yogurt showed the highest inhibition effect (13.5mm) on H. pylori growth in comparison with licorice-yogurt (11.2mm). The present findings indicate cinnamon and licorice has bioactive components to decrease the growth of H. pylori.

240 The Dynamics of Microorganisms in Dried Yogurt Storages at Different Temperatures

Abstract:

Yoghurt is a fermented milk product. The process of making yogurt involves fermenting milk with live and active bacterial cultures by adding bacteria directly to the dairy product. It is usually made with a culture of Lactobacillus sp. (L. acidophilus or L. bulgaricus) and Streptococcus thermophilus. Many people like to eat it plain or flavored and it's also use as ingredient in many dishes. Yogurt is rich in nutrients including the microorganism which have important role in balancing the digestion and absorption of the boy.Consumers will benefit from lactic acid bacteria more or less depending on the amount of bacteria that lives in yogurt while eating. When purchasing yogurt, consumers should always check the label for live cultures. Yoghurt must keep in refrigerator at 4°C for up to ten days. After this amount of time, the cultures often become weak. This research studied freezing dry yogurt storage by monitoring on the survival of microorganisms when stored at different temperatures. At 300C, representative room temperature of country in equator zone, number of lactic acid bacteria reduced 4 log cycles in 10 week. At 400C, representative temperature in summer of country in equator zone, number of lactic acid bacteria also dropped 4 log cycle in 10 week, similar as storage at 300C. But drying yogurt storage at 400C couldn’t reformed to be good character yogurt as good as storage at 400C only 4 week storage too. After 1 month, it couldn’t bring back the yogurt form. So if it is inevitable to keep yogurt powder at a temperature of 40°C, yoghurt is maintained only up to 4 weeks.

239 Incidence of Pathogenic Bacteria in Cakes and Tarts Displayed for Sale in Tripoli, Libya

Abstract:

This study was conducted to investigate the incidence of pathogenic bacteria: Salmonella, Shigella, Escherichia coli O157 and Staphylococcus aureus in cakes and tarts collected from thirtyfive confectionery producing and selling premises located within Tripoli city, Libya. The results revealed an incidence of S. aureus with 94.4 and 48.0 %, E. coli O157 with 14.7 and 4.0 % and Salmonella sp. with 5.9 and 8.0 % in cakes and tarts samples respectively; while Shigella was not detected in all samples. In order to determine the source of these pathogenic bacteria, cotton swabs were taken from the hands of workers on the production line, the surfaces of preparation tables and cream whipping instruments. The results showed that the cotton swabs obtained from the hands of workers contained S. aureus and Salmonella sp. with an incidence of 42.9 and 2.9 %, the cotton swabs obtained from the surfaces of preparation tables 22.9 and 2.9 % and the cotton swabs obtained from the cream whipping instruments 14.3 and 0.0 % respectively; while E. coli O157 and Shigella sp. were not detected in all swabs. Additionally, other bacteria were isolated from the hands of workers and the Surfaces of producing equipments included: Aeromonas sp., Pseudomonas sp., E. coli, Klebsiella sp., Enterobacter sp., Citrobacter sp., Proteus sp., Serratia sp. and Acinetobacter sp. These results indicate that some of the cakes and tarts might pose threat to consumer's health. Meanwhile, occurrences of pathogenic bacteria on the hands of those who are working in production line and the surfaces of equipments reflect poor hygienic practices at most confectionery premises examined in this study. Thus, firm and continuous surveillance of these premises is needed to insure the consumer's health and safety.

238 Zno Nanocomposites: Control of Enviromental Effects for Preservation of old Manuscripts

Abstract:

We investigate the ZnO role in the inherent protection of old manuscripts to protect them against environmental damaging effect of ultraviolet radiation, pollutant gasses, mold and bacteria. In this study a cellulosic nanocomposite of ZnO were used as protective coating on the surface of paper fibers. This layered nanocomposite can act as a consolidate materials too. Furthermore, to determine how well paper works screen objects from the damaging effects, two accelerated aging mechanisms due to light and heat are discussed. Results show good stability of papers with nanocomposite coating. Also, a good light stability was shown in the colored paper that treated with this nanocomposite. Furthermore, to demonstrate the degree of antifungal and antibacterial properties of coated papers, papers was treated with four common molds and bacteria and the good preventive effects of coated paper against molds and bacteria are described.

237 Coordinated Design of PSS and STATCOM for Power System Stability Improvement Using Bacteria Foraging Algorithm

Abstract:

This paper presents the coordinated controller design of static synchronous compensator (STATCOM) and power system stabilizers (PSSs) for power system stability improvement. Coordinated design problem of STATCOM-based controller with multiple PSSs is formulated as an optimization problem and optimal controller parameters are obtained using bacteria foraging optimization algorithm. By minimizing the proposed objective function, in which the speed deviations between generators are involved; stability performance of the system is improved. The nonlinear simulation results show that coordinated design of STATCOM-based controller and PSSs improve greatly the system damping oscillations and consequently stability improvement.

236 Effect of Different Lactic Acid Bacteria on Phytic Acid Content and Quality of whole Wheat Toast Bread

Abstract:

Nowadays, consumption of whole flours and flours with high extraction rate is recommended, because of their high amount of fibers, vitamins and minerals. Despite nutritional benefits of whole flour, concentration of some undesirable components such as phytic acid is higher than white flour. In this study, effect of several lactic acid bacteria sourdough on Toast bread is investigated. Sourdough from lactic acid bacteria (Lb. plantarum, Lb. reuteri) with different dough yield (250 and 300) is made and incubated at 30°C for 20 hour, then added to dough in the ratio of 10, 20 and 30% replacement. Breads that supplemented with Lb. plantarum sourdough had lower phytic acid. Higher replacement of sourdough and higher DY cause higher decrease in phytic acid content. Sourdough from Lb. plantarum, DY = 300 and 30% replacement cause the highest decrease in phytic acid content (49.63 mg/100g). As indicated by panelists, Lb. reuteri sourdough can present the greatest effect on overall quality score of the breads. DY reduction cause a decrease in bread quality score. Sensory score of Toast bread is 81.71 in the samples that treated with Lb. reuteri sourdough with DY = 250 and 20% replacement.

235 Depyritization of US Coal Using Iron-Oxidizing Bacteria: Batch Stirred Reactor Study

Abstract:

Microbial depyritization of coal using chemoautotrophic bacteria is gaining acceptance as an efficient and eco-friendly technique. The process uses the metabolic activity of chemoautotrophic bacteria in removing sulfur and pyrite from the coal. The aim of the present study was to investigate the potential of Acidithiobacillus ferrooxidans in removing the pyritic sulfur and iron from high iron and sulfur containing US coal. The experiment was undertaken in 8L bench scale stirred tank reactor having 1% (w/v) pulp density of coal. The reactor was operated at 35ºC and aerobic conditions were maintained by sparging the air into the reactor. It was found that at the end of bio-depyritization process, about 90% of pyrite and 67% of pyritic sulfur was removed from the coal. The results indicate that the bio-depyritization process is an efficient process in treating the high pyrite and sulfur containing coal.

234 Effect of Supplemental Irrigation, Nitrogen Chemical Fertilizer, and Inoculation with Rhizobium Bacteria on Grain Yield and Its Components of Chickpea (Cicer arietinum L.) Under Rainfed Conditions

Abstract:

In order to study the effects of supplemental irrigation, different levels of nitrogen chemical fertilizer and inoculation with rhizobium bacteria on the grain yield of chickpea, an experiment was carried out using split plot arrangement in randomize complete block design with three replication in agricultural researches station of Zanjan, Iran during 2009-2010 cropping season. The factors of experiment consisted of irritation (without irrigation (I1), irrigation at flowering stage (I2), irrigation at flowering and grain filling stages (I3) and full irrigation (I4)) and different levels of nitrogen fertilizer (without using of nitrogen fertilizer (N0), 75 kg.ha-1 (N75), 150 kg.ha-1 (N150) and inoculation with rhizobium bacteria (N4). The results of the analysis of variance showed that the effects of irrigation, nitrogen fertilizer levels and bacterial inoculation, were significant affect on number of pods per plant, number grains per plant, grain weight, grain yield, biological yield and harvest index at 1% probability level. Also Results showed that the grain yield in full irrigation treatment and inoculated with rhizobium bacteria was significantly higher than the other treatments.

233 A Bacterial Foraging Optimization Algorithm Applied to the Synthesis of Polyacrylamide Hydrogels

Abstract:

The Bacterial Foraging Optimization (BFO) algorithm is inspired by the behavior of bacteria such as Escherichia coli or Myxococcus xanthus when searching for food, more precisely the chemotaxis behavior. Bacteria perceive chemical gradients in the environment, such as nutrients, and also other individual bacteria, and move toward or in the opposite direction to those signals. The application example considered as a case study consists in establishing the dependency between the reaction yield of hydrogels based on polyacrylamide and the working conditions such as time, temperature, monomer, initiator, crosslinking agent and inclusion polymer concentrations, as well as type of the polymer added. This process is modeled with a neural network which is included in an optimization procedure based on BFO. An experimental study of BFO parameters is performed. The results show that the algorithm is quite robust and can obtain good results for diverse combinations of parameter values.

232 Microorganisms Isolated from Surgical Wounds Infection and Treatment with Different Natural Products and Medications

Abstract:

Surgical site infections (SSIs) are the most common nosocomial infection in surgical patients resulting in significant increases in postoperative morbidity and mortality. The commonly causative bacteria developed resistance to virtually all antibiotics available. The aim of this study was to isolation and identification the most common bacteria that cause SSIs in Medical Research Institute, and to compare their sensitivity to selected group of antibiotics and natural products (garlic, oregano, olive, and Nigella sativa oils). The isolated pathogens collected from infected surgical wounds were identified, and their sensitivities to the antibiotics commonly available for clinical use, and also to the different concentrations of the used natural products were investigated. The results indicate to the potential therapeutic effect of the tested natural products in treatment of surgical wound infections.

231 Promotion of Growth and Modulation of As- Induced Stress Ethylene in Maize by As- Tolerant ACC Deaminase Producing Bacteria

Abstract:

One of the major pollutants in the environment is arsenic (As). Due to the toxic effects of As to all organisms, its remediation is necessary. Conventional technologies used in the remediation of As contaminated soils are expensive and may even compromise the structure of the soil. An attractive alternative is phytoremediation, which is the use of plants which can take up the contaminant in their tissues. Plant growth promoting bacteria (PGPB) has been known to enhance growth of plants through several mechanisms such as phytohormone production, phosphate solubilization, siderophore production and 1-aminocyclopropane-1- carboxylate (ACC) deaminase production, which is an essential trait that aids plants especially under stress conditions such as As stress. Twenty one bacteria were isolated from As-contaminated soils in the vicinity of the Janghang Smelter in Chungnam Province, South Korea. These exhibited high tolerance to either arsenite (As III) or arsenate (As V) or both. Most of these isolates possess several plant growth promoting traits which can be potentially exploited to increase phytoremediation efficiency. Among the identified isolates is Pseudomonas sp. JS1215, which produces ACC deaminase, indole acetic acid (IAA), and siderophore. It also has the ability to solubilize phosphate. Inoculation of JS1215 significantly enhanced root and shoot length and biomass accumulation of maize under normal conditions. In the presence of As, particularly in lower As level, inoculation of JS1215 slightly increased root length and biomass. Ethylene increased with increasing As concentration, but was reduced by JS1215 inoculation. JS1215 can be a potential bioinoculant for increasing phytoremediation efficiency.

230 Prevalence and Antimicrobial Susceptibility Patterns of Enteric Bacteria Isolated from Water and Fish in Lake Victoria Basin of Western Kenya

Abstract:

A cross sectional study design and standard microbiological procedures were used to determine the prevalence and antimicrobial susceptibility patterns of Escherichia coli, Salmonella enterica serovar typhimurium and Vibrio cholerae O1 isolated from water and two fish species Rastrineobola argentea and Oreochromis niloticus collected from fish landing beaches and markets in the Lake Victoria Basin of western Kenya. Out of 162 samples analyzed, 133 (82.1%) were contaminated, with S. typhimurium as the most prevalent (49.6%), followed by E. coli (46.6%), and lastly V. cholerae (2.8%). All the bacteria isolates were sensitive to ciprofloxacin. E. coli isolates were resistant to ampicillin, tetracycline, cotrimoxazole, chloramphenical and gentamicin while S. typhimurium isolates exhibited resistance to ampicillin, tetracycline, and cotrimoxazole. The V. cholerae O1 isolates were resistant to tetracycline and ampicillin. The high prevalence of drug resistant enteric bacteria in water and fish from the study region needs public health intervention from the local government.

229 Heavy Metals and Antibiotic Resistant Bacteria in Green Turtles are Indicators of Environmental Pollution

Abstract:

Freshly laid eggs from green turtles, Chelonia mydas, were randomly collected from Ras Al-Hadd Reserve, Oman. Eggshells taken from eggs and sand collected from the body chamber were analyzed for eight heavy metals (Al, Br, Cd, Co, Cu, Fe, S, and Zn) using inductively coupled plasma mass spectrometry (ICP). Heavy metal concentrations varied significantly (P<0.05) between nest sand and eggshells. Zn values were significantly higher than the other heavy metals. A total of 60 heterotrophic bacteria belong to eight genera were isolated from fresh egg contents (albumen and yolk). Resistance of the isolates to Ak = amikacin, Ak = amikacin, Amp= ampicillin, Gm= gentamycin, Cn = chloramphenicol, Min = minocycline, N = Neomycin, S= streptomycin, Smx = sulphamethoxazole, Tmp = trimethoprim, Tob = tobramycin was tested. More than 40% of the isolates were multiple resistant to 2-10 antibiotics. Most of the resistant strains were also resistant to Zn. The value of these findings may indicate that the origin of pollution is of human contaminated effluents.

228 Assessment of Photodynamic Therapy for Staphylococcus Aureus Infected Wounds using Diffuse Reflectance Spectrometry

Abstract:

In this paper we evaluated the efficacy of photodynamic treatment of infected wounds on pig animal model by diffuse reflectance spectrometry. The study was conducted on fifteen wounds contaminated with Staphylococcus aureus bacteria that were incubated for 30 min with methylene blue solution (c = 3.3 x 10-3 M) and exposed to laser radiations (λ = 670 nm, P = 15 mW) for 15 min. The efficiency of photodynamic inactivation of bacteria was evaluated by microbiological exams and diffuse reflectance spectrometry. The results of the microbiological exams showed that the bacterial concentration has decreased from 6.93±0.138 logCFU/ml to 3.12±0.108 logCFU/ml. The spectral examination showed that the diffuse reflectance of wounds contaminated with Staphylococcus aureus has decreased from 5.06±0.036 % to 3.36±0.025 %. In conclusion, photodynamic therapy is an effective method for the treatment of infected wounds and there is a correlation between the CFU count and diffuse reflectance.

227 Investigation on the Effectiveness of Zinc Sulphate and Biofertilizer on Mustard Plant

Abstract:

The present work was conducted to find out the effect of biofertilizer formulated with four species of bacteria (two species of Azotobacter and two species of Lysobacter) and zinc sulphate. Field experiments with mustard plant were conducted to study the effectiveness of soil application of zinc sulphate and biofertilizer at 0, 10, 20, 30, 40, 50 days after sowing. Plant height and condition of plant was found to be increased significantly using a mixture of biofertilizer and zinc sulphate than other treatments after 40 days sowing. Three treatments were also used in this field experiment such as bacteria only, zinc sulphate only and mixture of biofertilizer and zinc sulphate. The treatment using a mixture of zinc sulphate and biofertilizer had the best yield (4688.008 kg/ha) within 50 days of sowing and performed better than other treatments. Field experiment using zinc sulphate only was second best yield (3380.75Kg/ha) and biofertilizer only treatment gave (2639.04kg/ha).

226 Analysis and Remediation of Fecal Coliform Bacteria Pollution in Selected Surface Water Bodies of Enugu State of Nigeria

Abstract:

The assessment of surface waters in Enugu metropolis for fecal coliform bacteria was undertaken. Enugu urban was divided into three areas (A1, A2 and A3), and fecal coliform bacteria analysed in the surface waters found in these areas for four years (2005-2008). The plate count method was used for the analyses. Data generated were subjected to statistical tests involving; Normality test, Homogeneity of variance test, correlation test, and tolerance limit test. The influence of seasonality and pollution trends were investigated using time series plots. Results from the tolerance limit test at 95% coverage with 95% confidence, and with respect to EU maximum permissible concentration show that the three areas suffer from fecal coliform pollution. To this end, remediation procedure involving the use of saw-dust extracts from three woods namely; Chlorophora-Excelsa (C-Excelsa),Khayan-Senegalensis,(CSenegalensis) and Erythrophylum-Ivorensis (E-Ivorensis) in controlling the coliforms was studied. Results show that mixture of the acetone extracts of the woods show the most effective antibacterial inhibitory activities (26.00mm zone of inhibition) against E-coli. Methanol extract mixture of the three woods gave best inhibitory activity (26.00mm zone of inhibition) against S-areus, and 25.00mm zones of inhibition against E-Aerogenes. The aqueous extracts mixture gave acceptable zones of inhibitions against the three bacteria organisms.

225 Study the Biological Activities of Tribulus Terrestris Extracts

Abstract:

In this study the extracts of the Iraqi herb Tribulus terrestris (Al-Hassage or Al-Kutub) was done by using of polar and non polar solvents, then the biological activity of these extractants was studied in three fields, First, the antibacterial activity (in vitro) on gram positive bacteria (Staphylococcus aureus), and gram negative bacteria (E. coli, Proteus vulgaris, Pseudomonas aerugiuosa, and Klebsiella), all extracts showed considerable activity against all bacteria. Second, the effect of extracts on free serum testosterone level in male mice (in vivo), the alcoholic, and acetonitrilic extracts showed significant (P < 0.05) increase in free serum testosterone level, and we found that the extracts contained compounds with less genotoxic effects in mice germ cells. 3rd, was to study the effect of methanolic extract of T. terrestris in diabetes management.

224 Toxicity Study of Two Different Synthesized Silver Nanoparticles on Bacteria Vibrio Fischeri

Abstract:

A comparative evaluation of acute toxicity of synthesized nano silvers using two different procedures (biological and chemical reduction methods) and silver ions on bacteria Vibrio fischeri was investigated. The bacterial light inhibition test as a toxicological endpoint was used by applying of a homemade luminometer. To compare the toxicity effects as a quantitative parameter, a nominal effective concentrations (EC) of chemicals and a susceptibility constant (Z-value) of bacteria, after 5 min and 30 min exposure times, were calculated. After 5 and 30 min contact times, the EC50 values of two silver nanoparticles and the EC20 values were about similar. It demonstrates that toxicity of silvers was independent of their procedure. The EC values of nanoparticles were larger than those of the silver ions. The susceptibilities(Z- Values) of V.fischeri (L/mg) to the silver ions were greater than those of the nano silvers. According to the EC and Z values, the toxicity of silvers decreased in the following order: Silver ions >> silver nanoparticles from chemical reduction method ~ silver nanoparticles from biological method.

223 Identification of the Antimicrobial Effect of Liquorice Extracts on Gram-Positive Bacteria: Determination of Minimum Inhibitory Concentration and Mechanism of Action Using a luxABCDE Reporter Strain

Abstract:

Natural preservatives have been used as alternatives to traditional chemical preservatives; however, a limited number have been commercially developed and many remain to be investigated as sources of safer and effective antimicrobials. In this study, we have been investigating the antimicrobial activity of an extract of Glycyrrhiza glabra (liquorice) that was provided as a waste material from the production of liquorice flavourings for the food industry, and to investigate if this retained the expected antimicrobial activity so it could be used as a natural preservative. Antibacterial activity of liquorice extract was screened for evidence of growth inhibition against eight species of Gram-negative and Gram-positive bacteria, including Listeria monocytogenes, Listeria innocua, Staphylococcus aureus, Enterococcus faecalis and Bacillus subtilis. The Gram-negative bacteria tested include Pseudomonas aeruginosa, Escherichia coli and Salmonella typhimurium but none of these were affected by the extract. In contrast, for all of the Gram-positive bacteria tested, growth was inhibited as monitored using optical density. However parallel studies using viable count indicated that the cells were not killed meaning that the extract was bacteriostatic rather than bacteriocidal. The Minimum Inhibitory Concentration [MIC] and Minimum Bactericidal Concentration [MBC] of the extract was also determined and a concentration of 50 µg ml-1 was found to have a strong bacteriostatic effect on Gram-positive bacteria. Microscopic analysis indicated that there were changes in cell shape suggesting the cell wall was affected. In addition, the use of a reporter strain of Listeria transformed with the bioluminescence genes luxABCDE indicated that cell energy levels were reduced when treated with either 12.5 or 50 µg ml-1 of the extract, with the reduction in light output being proportional to the concentration of the extract used. Together these results suggest that the extract is inhibiting the growth of Gram-positive bacteria only by damaging the cell wall and/or membrane.

222 Analysis of Sulphur-Oxidizing Bacteria Attack on Concrete Based On Waste Materials

Abstract:

Concrete durability as an important engineering property of concrete, determining the service life of concrete structures very significantly, can be threatened and even lost due to the interactions of concrete with external environment. Bio-corrosion process caused by presence and activities of microorganisms producing sulphuric acid is a special type of sulphate deterioration of concrete materials. The effects of sulphur-oxidizing bacteria Acidithiobacillus thiooxidans on various concrete samples, based on silica fume and zeolite, were investigated in laboratory during 180 days. A laboratory study was conducted to compare the performance of concrete samples in terms of the concrete deterioration influenced by the leaching of calcium and silicon compounds from the cement matrix. The changes in the elemental concentrations of calcium and silicon in both solid samples and liquid leachates were measured by using X – ray fluorescence method. Experimental studies confirmed the silica fume based concrete samples were found out to have the best performance in terms of both silicon and calcium ions leaching.

221 Antibacterial and Antifungal Activity of Essential Oil of Eucalyptus camendulensis on a Few Bacteria and Fungi

Abstract:

Red River Gum (Eucalyptus camaldulensis) is a tree of the genus Eucalyptus widely distributed in Algeria and in the world. The value of its aromatic secondary metabolites offers new perspectives in the pharmaceutical industry. This strategy can contribute to the sustainable development of our country. Preliminary tests performed on the essential oil of Eucalyptus camendulensis showed that this oil has antibacterial activity vis-à-vis the bacterial strains (Enterococcus feacalis, Enterobacter cloaceai, Proteus microsilis, Escherichia coli, Klebsiella pneumonia, and Pseudomonas aeruginosa) and antifungic (Fusarium sporotrichioide and Fusarium graminearum). The culture medium used was nutrient broth Muller Hinton. The interaction between the bacteria and the essential oil is expressed by a zone of inhibition with diameters of MIC indirectly expression of. And we used the PDA medium to determine the fungal activity. The extraction of the aromatic fraction (essentially oilhydrolat) of the fresh aerian part of the Eucalyptus camendulensis was performed by hydrodistillation. The average essential oil yield is 0.99%. The antimicrobial and fungal study of the essential oil and hydrosol showed a high inhibitory effect on the growth of pathogens.

220 Negative Impact of Bacteria Legionella Pneumophila in Hot Water Distribution Systems on Human Health

Abstract:

Safe drinking water is one of the biggest issues facing the planet this century. The primary aim of this paper is to present our research focused on theoretical and experimental analysis of potable water and in-building water distribution systems from the point of view of microbiological risk on the basis of confrontation between the theoretical analysis and synthesis of gathered information in conditions of the Slovak Republic. The presence of the bacteria Legionella in water systems, especially in hot water distribution system, represents in terms of health protection of inhabitants the crucial problem which cannot be overlooked. Legionella pneumophila discovery, its classification and its influence on installations inside buildings are relatively new. There are a lot of guidelines and regulations developed in many individual countries for the design, operation and maintenance for tap water systems to avoid the growth of bacteria Legionella pneumophila, but in Slovakia we don-t have any. The goal of this paper is to show the necessity of prevention and regulations for installations inside buildings verified by simulation methods.

219 Exploratory Tests of Crude Bacteriocins from Autochthonous Lactic Acid Bacteria against Food-Borne Pathogens and Spoilage Bacteria

Abstract:

The aim of the present work was to test in vitro inhibition of food pathogens and spoilage bacteria by crude bacteriocins from autochthonous lactic acid bacteria. Thirty autochthonous lactic acid bacteria isolated previously, belonging to the genera: Lactobacillus, Carnobacterium, Lactococcus, Vagococcus, Streptococcus, and Pediococcus, have been screened by an agar spot test and a well diffusion assay against Gram-positive and Gram-negative harmful bacteria: Bacillus cereus, Bacillus subtilis ATCC 6633, Escherichia coli ATCC 8739, Salmonella typhimurium ATCC 14028, Staphylococcus aureus ATCC 6538, and Pseudomonas aeruginosa under conditions means to reduce lactic acid and hydrogen peroxide effect to select bacteria with high bacteriocinogenic potential. Furthermore, crude bacteriocins semiquantification and heat sensitivity to different temperatures (80, 95, 110°C, and 121°C) were performed. Another exploratory test concerning the response of St. aureus ATCC 6538 to the presence of crude bacteriocins was realized. It has been observed by the agar spot test that fifteen candidates were active toward Gram-positive targets strains. The secondary screening demonstrated an antagonistic activity oriented only against St. aureus ATCC 6538, leading to the selection of five isolates: Lm14, Lm21, Lm23, Lm24, and Lm25 with a larger inhibition zone compared to the others. The ANOVA statistical analysis reveals a small variation of repeatability: Lm21: 0.56%, Lm23: 0%, Lm25: 1.67%, Lm14: 1.88%, Lm24: 2.14%. Conversely, slight variation was reported in terms of inhibition diameters: 9.58± 0.40, 9.83± 0.46 and 10.16± 0.24 8.5 ± 0.40 10 mm for, Lm21, Lm23, Lm25, Lm14and Lm24, indicating that the observed potential showed a heterogeneous distribution (BMS = 0.383, WMS = 0.117). The repeatability coefficient calculated displayed 7.35%. As for the bacteriocins semiquantification, the five samples exhibited production amounts about 4.16 for Lm21, Lm23, Lm25 and 2.08 AU/ml for Lm14, Lm24. Concerning the sensitivity the crude bacteriocins were fully insensitive to heat inactivation, until 121°C, they preserved the same inhibition diameter. As to, kinetic of growth , the µmax showed reductions in pathogens load for Lm21, Lm23, Lm25, Lm14, Lm24 of about 42.92%, 84.12%, 88.55%, 54.95%, 29.97% in the second trails. Inversely, this pathogen growth after five hours displayed differences of 79.45%, 12.64%, 11.82%, 87.88%, 85.66% in the second trails, compared to the control. This study showed potential inhibition to the growth of this food pathogen, suggesting the possibility to improve the hygienic food quality.
