1203 Modern Seismic Design Approach for Buildings with Hysteretic Dampers

Authors: Vanessa A. Segovia, Sonia E. Ruiz

Abstract:

The use of energy dissipation systems for seismic applications has increased worldwide, thus it is necessary to develop practical and modern criteria for their optimal design. Here, a direct displacement-based seismic design approach for frame buildings with hysteretic energy dissipation systems (HEDS) is applied. The building is constituted by two individual structural systems consisting of: 1) a main elastic structural frame designed for service loads; and 2) a secondary system, corresponding to the HEDS, that controls the effects of lateral loads. The procedure implies to control two design parameters: a) the stiffness ratio (α=Kframe/Ktotal system), and b) the strength ratio (γ=Vdamper/Vtotal system). The proposed damage-controlled approach contributes to the design of a more sustainable and resilient building because the structural damage is concentrated on the HEDS. The reduction of the design displacement spectrum is done by means of a damping factor (recently published) for elastic structural systems with HEDS, located in Mexico City. Two limit states are verified: serviceability and near collapse. Instead of the traditional trial-error approach, a procedure that allows the designer to establish the preliminary sizes of the structural elements of both systems is proposed. The design methodology is applied to an 8-story steel building with buckling restrained braces, located in soft soil of Mexico City. With the aim of choosing the optimal design parameters, a parametric study is developed considering different values of હ and ઻. The simplified methodology is for preliminary sizing, design, and evaluation of the effectiveness of HEDS, and it constitutes a modern and practical tool that enables the structural designer to select the best design parameters.

1202 Diagnosis of Multivariate Process via Nonlinear Kernel Method Combined with Qualitative Representation of Fault Patterns

Authors: Hyun-Woo Cho

Abstract:

The fault detection and diagnosis of complicated production processes is one of essential tasks needed to run the process safely with good final product quality. Unexpected events occurred in the process may have a serious impact on the process. In this work, triangular representation of process measurement data obtained in an on-line basis is evaluated using simulation process. The effect of using linear and nonlinear reduced spaces is also tested. Their diagnosis performance was demonstrated using multivariate fault data. It has shown that the nonlinear technique based diagnosis method produced more reliable results and outperforms linear method. The use of appropriate reduced space yielded better diagnosis performance. The presented diagnosis framework is different from existing ones in that it attempts to extract the fault pattern in the reduced space, not in the original process variable space. The use of reduced model space helps to mitigate the sensitivity of the fault pattern to noise.

1201 Effect of Magnetic Field on the Biological Clock through the Radical Pair Mechanism

Authors: Chathurika D. Abeyrathne, Malka N. Halgamuge, Peter M. Farrell

Abstract:

There is an ongoing controversy in the literature related to the biological effects of weak, low frequency electromagnetic fields. The physical arguments and interpretation of the experimental evidence are inconsistent, where some physical arguments and experimental demonstrations tend to reject the likelihood of any effect of the fields at extremely low level. The problem arises of explaining, how the low-energy influences of weak magnetic fields can compete with the thermal and electrical noise of cells at normal temperature using the theoretical studies. The magnetoreception in animals involve radical pair mechanism. The same mechanism has been shown to be involved in the circadian rhythm synchronization in mammals. These reactions can be influenced by the weak magnetic fields. Hence, it is postulated the biological clock can be affected by weak magnetic fields and these disruptions to the rhythm can cause adverse biological effects. In this paper, likelihood of altering the biological clock via the radical pair mechanism is analyzed to simplify these studies of controversy.

Authors: Liang Zhao, Feng Wang, Bo Deng, Bo Yang

Abstract:

Software testability is proposed to address the problem of increasing cost of test and the quality of software. Testability measure provides a quantified way to denote the testability of software. Since 1990s, many testability measure models are proposed to address the problem. By discussing the contradiction between domain testability and domain range ratio (DRR), a new testability measure, semantic fault distance, is proposed. Its validity is discussed.

1199 Image Adaptive Watermarking with Visual Model in Orthogonal Polynomials based Transformation Domain

Authors: Krishnamoorthi R., Sheba Kezia Malarchelvi P. D.

Abstract:

In this paper, an image adaptive, invisible digital watermarking algorithm with Orthogonal Polynomials based Transformation (OPT) is proposed, for copyright protection of digital images. The proposed algorithm utilizes a visual model to determine the watermarking strength necessary to invisibly embed the watermark in the mid frequency AC coefficients of the cover image, chosen with a secret key. The visual model is designed to generate a Just Noticeable Distortion mask (JND) by analyzing the low level image characteristics such as textures, edges and luminance of the cover image in the orthogonal polynomials based transformation domain. Since the secret key is required for both embedding and extraction of watermark, it is not possible for an unauthorized user to extract the embedded watermark. The proposed scheme is robust to common image processing distortions like filtering, JPEG compression and additive noise. Experimental results show that the quality of OPT domain watermarked images is better than its DCT counterpart.

Authors: Manash Pratim Sarma, Kandarpa Kumar Sarma

Abstract:

A set of Artificial Neural Network (ANN) based methods for the design of an effective system of speech recognition of numerals of Assamese language captured under varied recording conditions and moods is presented here. The work is related to the formulation of several ANN models configured to use Linear Predictive Code (LPC), Principal Component Analysis (PCA) and other features to tackle mood and gender variations uttering numbers as part of an Automatic Speech Recognition (ASR) system in Assamese. The ANN models are designed using a combination of Self Organizing Map (SOM) and Multi Layer Perceptron (MLP) constituting a Learning Vector Quantization (LVQ) block trained in a cooperative environment to handle male and female speech samples of numerals of Assamese- a language spoken by a sizable population in the North-Eastern part of India. The work provides a comparative evaluation of several such combinations while subjected to handle speech samples with gender based differences captured by a microphone in four different conditions viz. noiseless, noise mixed, stressed and stress-free.

Abstract:

Recent research in neural networks science and neuroscience for modeling complex time series data and statistical learning has focused mostly on learning from high input space and signals. Local linear models are a strong choice for modeling local nonlinearity in data series. Locally weighted projection regression is a flexible and powerful algorithm for nonlinear approximation in high dimensional signal spaces. In this paper, different learning scenario of one and two dimensional data series with different distributions are investigated for simulation and further noise is inputted to data distribution for making different disordered distribution in time series data and for evaluation of algorithm in locality prediction of nonlinearity. Then, the performance of this algorithm is simulated and also when the distribution of data is high or when the number of data is less the sensitivity of this approach to data distribution and influence of important parameter of local validity in this algorithm with different data distribution is explained.

In recent years, the use of vector variance as a measure of multivariate variability has received much attention in wide range of statistics. This paper deals with a more economic measure of multivariate variability, defined as vector variance minus all duplication elements. For high dimensional data, this will increase the computational efficiency almost 50 % compared to the original vector variance. 1192 Detection and Correction of Ectopic Beats for HRV Analysis Applying Discrete Wavelet Transforms

Authors: Desmond B. Keenan

Abstract:

The clinical usefulness of heart rate variability is limited to the range of Holter monitoring software available. These software algorithms require a normal sinus rhythm to accurately acquire heart rate variability (HRV) measures in the frequency domain. Premature ventricular contractions (PVC) or more commonly referred to as ectopic beats, frequent in heart failure, hinder this analysis and introduce ambiguity. This investigation demonstrates an algorithm to automatically detect ectopic beats by analyzing discrete wavelet transform coefficients. Two techniques for filtering and replacing the ectopic beats from the RR signal are compared. One technique applies wavelet hard thresholding techniques and another applies linear interpolation to replace ectopic cycles. The results demonstrate through simulation, and signals acquired from a 24hr ambulatory recorder, that these techniques can accurately detect PVC-s and remove the noise and leakage effects produced by ectopic cycles retaining smooth spectra with the minimum of error.

1191 Dimension Reduction of Microarray Data Based on Local Principal Component

Authors: Ali Anaissi, Paul J. Kennedy, Madhu Goyal

Abstract:

Analysis and visualization of microarraydata is veryassistantfor biologists and clinicians in the field of diagnosis and treatment of patients. It allows Clinicians to better understand the structure of microarray and facilitates understanding gene expression in cells. However, microarray dataset is a complex data set and has thousands of features and a very small number of observations. This very high dimensional data set often contains some noise, non-useful information and a small number of relevant features for disease or genotype. This paper proposes a non-linear dimensionality reduction algorithm Local Principal Component (LPC) which aims to maps high dimensional data to a lower dimensional space. The reduced data represents the most important variables underlying the original data. Experimental results and comparisons are presented to show the quality of the proposed algorithm. Moreover, experiments also show how this algorithm reduces high dimensional data whilst preserving the neighbourhoods of the points in the low dimensional space as in the high dimensional space.

1189 Heart Rate Variability Analysis for Early Stage Prediction of Sudden Cardiac Death

Authors: Reeta Devi, Hitender Kumar Tyagi, Dinesh Kumar

Abstract:

In present scenario, cardiovascular problems are growing challenge for researchers and physiologists. As heart disease have no geographic, gender or socioeconomic specific reasons; detecting cardiac irregularities at early stage followed by quick and correct treatment is very important. Electrocardiogram is the finest tool for continuous monitoring of heart activity. Heart rate variability (HRV) is used to measure naturally occurring oscillations between consecutive cardiac cycles. Analysis of this variability is carried out using time domain, frequency domain and non-linear parameters. This paper presents HRV analysis of the online dataset for normal sinus rhythm (taken as healthy subject) and sudden cardiac death (SCD subject) using all three methods computing values for parameters like standard deviation of node to node intervals (SDNN), square root of mean of the sequences of difference between adjacent RR intervals (RMSSD), mean of R to R intervals (mean RR) in time domain, very low-frequency (VLF), low-frequency (LF), high frequency (HF) and ratio of low to high frequency (LF/HF ratio) in frequency domain and Poincare plot for non linear analysis. To differentiate HRV of healthy subject from subject died with SCD, k –nearest neighbor (k-NN) classifier has been used because of its high accuracy. Results show highly reduced values for all stated parameters for SCD subjects as compared to healthy ones. As the dataset used for SCD patients is recording of their ECG signal one hour prior to their death, it is therefore, verified with an accuracy of 95% that proposed algorithm can identify mortality risk of a patient one hour before its death. The identification of a patient’s mortality risk at such an early stage may prevent him/her meeting sudden death if in-time and right treatment is given by the doctor.

Authors: D. S. C. Soares, D. G. Costa, J. T. S., A. K. S. Abud, T. P. Nunes, A. M. Oliveira Júnior

Abstract:

Mathematical models of drying are used for the purpose of understanding the drying process in order to determine important parameters for design and operation of the dryer. The jackfruit is a fruit with high consumption in the Northeast and perishability. It is necessary to apply techniques to improve their conservation for longer in order to diffuse it by regions with low consumption. This study aimed to analyze several mathematical models (Page, Lewis, and Midilli) to indicate one that best fits the conditions of convective drying process using performance indicators associated with each model: accuracy (Af) and noise factors (Bf), mean square error (RMSE) and standard error of prediction (% SEP). Jackfruit drying was carried out in convective type tray dryer at a temperature of 50°C for 9 hours. It is observed that the model Midili was more accurate with Af: 1.39, Bf: 1.33, RMSE: 0.01%, and SEP: 5.34. However, the use of the Model Midilli is not appropriate for purposes of control process due to need four tuning parameters. With the performance indicators used in this paper, the Page model showed similar results with only two parameters. It is concluded that the best correlation between the experimental and estimated data is given by the Page’s model.

1187 DCBOR: A Density Clustering Based on Outlier Removal

Authors: A. M. Fahim, G. Saake, A. M. Salem, F. A. Torkey, M. A. Ramadan

Abstract:

Data clustering is an important data exploration technique with many applications in data mining. We present an enhanced version of the well known single link clustering algorithm. We will refer to this algorithm as DCBOR. The proposed algorithm alleviates the chain effect by removing the outliers from the given dataset. So this algorithm provides outlier detection and data clustering simultaneously. This algorithm does not need to update the distance matrix, since the algorithm depends on merging the most k-nearest objects in one step and the cluster continues grow as long as possible under specified condition. So the algorithm consists of two phases; at the first phase, it removes the outliers from the input dataset. At the second phase, it performs the clustering process. This algorithm discovers clusters of different shapes, sizes, densities and requires only one input parameter; this parameter represents a threshold for outlier points. The value of the input parameter is ranging from 0 to 1. The algorithm supports the user in determining an appropriate value for it. We have tested this algorithm on different datasets contain outlier and connecting clusters by chain of density points, and the algorithm discovers the correct clusters. The results of our experiments demonstrate the effectiveness and the efficiency of DCBOR.

1186 Optimal Power Allocation for the Proposed Asymmetric Turbo Code for 3G Systems

Authors: K. Ramasamy, B. Balamuralithara, Mohammad Umar Siddiqi

Abstract:

We proposed a new class of asymmetric turbo encoder for 3G systems that performs well in both “water fall" and “error floor" regions in [7]. In this paper, a modified (optimal) power allocation scheme for the different bits of new class of asymmetric turbo encoder has been investigated to enhance the performance. The simulation results and performance bound for proposed asymmetric turbo code with modified Unequal Power Allocation (UPA) scheme for the frame length, N=400, code rate, r=1/3 with Log-MAP decoder over Additive White Gaussian Noise (AWGN) channel are obtained and compared with the system with typical UPA and without UPA. The performance tests are extended over AWGN channel for different frame size to verify the possibility of implementation of the modified UPA scheme for the proposed asymmetric turbo code. From the performance results, it is observed that the proposed asymmetric turbo code with modified UPA performs better than the system without UPA and with typical UPA and it provides a coding gain of 0.4 to 0.52dB.

This research aims at identifying and mapping mangroves types along Karachi coast ranging from 24.790 to 24.850 in latitude and 66.910 to 66.970 in longitude using hyperspectral remote sensing data and techniques. Image acquired during February, 2012 through Hyperion sensor have been used for this research. Image pre processing includes geometric and radiometric correction followed by Minimum Noise Fraction (MNF) and Pixel Purity Index (PPI). The output of MNF and PPI has been analyzed by visualizing it in n-dimensions for end member extraction. Well distributed clusters on the n-dimensional scatter plot have been selected with the region of interest (ROI) tool as end members. These end members have been used as an input for classification techniques applied to identify and map mangroves species including Spectral Angle Mapper (SAM), Spectral Feature Fitting (SFF) and Spectral Information Diversion (SID). Only two types of mangroves namely Avicennia Marina (White Mangroves) and Avicennia germinans (Black Mangroves) have been observed throughout the study area.

1184 A Wavelet Based Object Watermarking System for Image and Video

Authors: Abdessamad Essaouabi, Ibnelhaj Elhassane

Abstract:

Efficient storage, transmission and use of video information are key requirements in many multimedia applications currently being addressed by MPEG-4. To fulfill these requirements, a new approach for representing video information which relies on an object-based representation, has been adopted. Therefore, objectbased watermarking schemes are needed for copyright protection. This paper proposes a novel blind object watermarking scheme for images and video using the in place lifting shape adaptive-discrete wavelet transform (SA-DWT). In order to make the watermark robust and transparent, the watermark is embedded in the average of wavelet blocks using the visual model based on the human visual system. Wavelet coefficients n least significant bits (LSBs) are adjusted in concert with the average. Simulation results shows that the proposed watermarking scheme is perceptually invisible and robust against many attacks such as lossy image/video compression (e.g. JPEG, JPEG2000 and MPEG-4), scaling, adding noise, filtering, etc.

1183 Super Resolution Blind Reconstruction of Low Resolution Images using Wavelets based Fusion

Authors: Liyakathunisa, V. K. Ananthashayana

Abstract:

Crucial information barely visible to the human eye is often embedded in a series of low resolution images taken of the same scene. Super resolution reconstruction is the process of combining several low resolution images into a single higher resolution image. The ideal algorithm should be fast, and should add sharpness and details, both at edges and in regions without adding artifacts. In this paper we propose a super resolution blind reconstruction technique for linearly degraded images. In our proposed technique the algorithm is divided into three parts an image registration, wavelets based fusion and an image restoration. In this paper three low resolution images are considered which may sub pixels shifted, rotated, blurred or noisy, the sub pixel shifted images are registered using affine transformation model; A wavelet based fusion is performed and the noise is removed using soft thresolding. Our proposed technique reduces blocking artifacts and also smoothens the edges and it is also able to restore high frequency details in an image. Our technique is efficient and computationally fast having clear perspective of real time implementation.

1182 Optimal Control Strategies for Speed Control of Permanent-Magnet Synchronous Motor Drives

Authors: Roozbeh Molavi, Davood A. Khaburi

Abstract:

The permanent magnet synchronous motor (PMSM) is very useful in many applications. Vector control of PMSM is popular kind of its control. In this paper, at first an optimal vector control for PMSM is designed and then results are compared with conventional vector control. Then, it is assumed that the measurements are noisy and linear quadratic Gaussian (LQG) methodology is used to filter the noises. The results of noisy optimal vector control and filtered optimal vector control are compared to each other. Nonlinearity of PMSM and existence of inverter in its control circuit caused that the system is nonlinear and time-variant. With deriving average model, the system is changed to nonlinear time-invariant and then the nonlinear system is converted to linear system by linearization of model around average values. This model is used to optimize vector control then two optimal vector controls are compared to each other. Simulation results show that the performance and robustness to noise of the control system has been highly improved.

1181 Hybrid Markov Game Controller Design Algorithms for Nonlinear Systems

Authors: R. Sharma, M. Gopal

Abstract:

Markov games can be effectively used to design controllers for nonlinear systems. The paper presents two novel controller design algorithms by incorporating ideas from gametheory literature that address safety and consistency issues of the 'learned' control strategy. A more widely used approach for controller design is the H∞ optimal control, which suffers from high computational demand and at times, may be infeasible. We generate an optimal control policy for the agent (controller) via a simple Linear Program enabling the controller to learn about the unknown environment. The controller is facing an unknown environment and in our formulation this environment corresponds to the behavior rules of the noise modeled as the opponent. Proposed approaches aim to achieve 'safe-consistent' and 'safe-universally consistent' controller behavior by hybridizing 'min-max', 'fictitious play' and 'cautious fictitious play' approaches drawn from game theory. We empirically evaluate the approaches on a simulated Inverted Pendulum swing-up task and compare its performance against standard Q learning.

1179 Removing Ocular Artifacts from EEG Signals using Adaptive Filtering and ARMAX Modeling

Authors: Parisa Shooshtari, Gelareh Mohamadi, Behnam Molaee Ardekani, Mohammad Bagher Shamsollahi

Abstract:

EEG signal is one of the oldest measures of brain activity that has been used vastly for clinical diagnoses and biomedical researches. However, EEG signals are highly contaminated with various artifacts, both from the subject and from equipment interferences. Among these various kinds of artifacts, ocular noise is the most important one. Since many applications such as BCI require online and real-time processing of EEG signal, it is ideal if the removal of artifacts is performed in an online fashion. Recently, some methods for online ocular artifact removing have been proposed. One of these methods is ARMAX modeling of EEG signal. This method assumes that the recorded EEG signal is a combination of EOG artifacts and the background EEG. Then the background EEG is estimated via estimation of ARMAX parameters. The other recently proposed method is based on adaptive filtering. This method uses EOG signal as the reference input and subtracts EOG artifacts from recorded EEG signals. In this paper we investigate the efficiency of each method for removing of EOG artifacts. A comparison is made between these two methods. Our undertaken conclusion from this comparison is that adaptive filtering method has better results compared with the results achieved by ARMAX modeling.

1178 Best Combination of Design Parameters for Buildings with Buckling-Restrained Braces

Authors: Ángel de J. López-Pérez, Sonia E. Ruiz, Vanessa A. Segovia

Abstract:

Buildings vulnerability due to seismic activity has been highly studied since the middle of last century. As a solution to the structural and non-structural damage caused by intense ground motions, several seismic energy dissipating devices, such as buckling-restrained braces (BRB), have been proposed. BRB have shown to be effective in concentrating a large portion of the energy transmitted to the structure by the seismic ground motion. A design approach for buildings with BRB elements, which is based on a seismic Displacement-Based formulation, has recently been proposed by the coauthors in this paper. It is a practical and easy design method which simplifies the work of structural engineers. The method is used here for the design of the structure-BRB damper system. The objective of the present study is to extend and apply a methodology to find the best combination of design parameters on multiple-degree-of-freedom (MDOF) structural frame – BRB systems, taking into account simultaneously: 1) initial costs and 2) an adequate engineering demand parameter. The design parameters considered here are: the stiffness ratio (α = Kframe/Ktotal), and the strength ratio (γ = Vdamper/Vtotal); where K represents structural stiffness and V structural strength; and the subscripts "frame", "damper" and "total" represent: the structure without dampers, the BRB dampers and the total frame-damper system, respectively. The selection of the best combination of design parameters α and γ is based on an initial costs analysis and on the structural dynamic response of the structural frame-damper system. The methodology is applied to a 12-story 5-bay steel building with BRB, which is located on the intermediate soil of Mexico City. It is found the best combination of design parameters α and γ for the building with BRB under study.

Authors: Kaushalendra K. Khadanga, Lee Hee Hyol

Abstract:

Passenger comfort has been paramount in the design of suspension systems of high speed cars. To analyze the effect of vibration on vehicle ride quality, a vertical model of a six degree of freedom railway passenger vehicle, with front and rear suspension, is built. It includes car body flexible effects and vertical rigid modes. A second order linear shaping filter is constructed to model Gaussian white noise into random rail excitation. The temporal correlation between the front and rear wheels is given by a second order Pade approximation. The complete track and the vehicle model are then designed. An active secondary suspension system based on a Linear Quadratic Gaussian (LQG) optimal control method is designed. The results show that the LQG control method reduces the vertical acceleration, pitching acceleration and vertical bending vibration of the car body as compared to the passive system.

Their mean age, weight and height were 20.7±2.4 vs. 20.3±0.61 years, 66.5±12.1 vs. 68.57±12.2 kg and 166.7±7.8 vs. 164.28 ±7.59 cm. for group I vs. group II. Data were collected using the Biodex Isokinetic system. The participants were tested twice; before and after a 6-week period during which group I performed a core stability training program. Results: The 2x2 Mixed Design ANOVA revealed that there were no significant differences (p>0.025) in the trunk flexors’/extensors’ peak torque ratio between the pre-test and post-test conditions for either group. Moreover, there were no significant differences (p>0.025) in the trunk flexion/extension ratios between both groups at either condition. However, the 2x2 Mixed Design MANOVA revealed significant increases (p<0.025) in the trunk flexors’ and extensors’ peak torques in the post-test condition compared with the pre-test in group I with no significant differences (p>0.025) in group II. Moreover, there was a significant increase (p<0.025) in the trunk flexors’ peak torque only in group I compared with group II in the post-test condition with no significant differences in the other conditions. Interpretation/Conclusion: The improvement in muscle performance indicated by the increase in the trunk flexors’ and extensors’ peak torques in the experimental group recommends including core stability training in the exercise programs that aim to improve muscle performance.

1175 Appraisal of Relativistic Effects on GNSS Receiver Positioning

Authors: I. Yakubu, Y. Y. Ziggah, E. A. Gyamera

Abstract:

The Global Navigation Satellite System (GNSS) started with the launch of the United State Department of Defense Global Positioning System (GPS). GNSS systems has grown over the years to include: GLONASS (Russia); Galileo (European Union); BeiDou (China). Any GNSS architecture consists of three major segments: Space, Control and User Segments. Errors such as; multipath, ionospheric and tropospheric effects, satellite clocks, receiver noise and orbit errors (relativity effect) have significant effects on GNSS positioning. To obtain centimeter level accuracy, the impacts of the relative motion of the satellites and earth need to be taken into account. This paper discusses the relevance of the theory of relativity as a source of error for GNSS receivers for position fix based on available relevant literature. Review of relevant literature reveals that due to relativity; Time dilation, Gravitational frequency shift and Sagnac effect cause significant influence on the use of GNSS receivers for positioning by an error range of ± 2.5 m based on pseudo-range computation.

1174 A Novel Multiplex Real-Time PCR Assay Using TaqMan MGB Probes for Rapid Detection of Trisomy 21

Abstract:

Cytogenetic analysis still remains the gold standard method for prenatal diagnosis of trisomy 21 (Down syndrome, DS). Nevertheless, the conventional cytogenetic analysis needs live cultured cells and is too time-consuming for clinical application. In contrast, molecular methods such as FISH, QF-PCR, MLPA and quantitative Real-time PCR are rapid assays with results available in 24h. In the present study, we have successfully used a novel MGB TaqMan probe-based real time PCR assay for rapid diagnosis of trisomy 21 status in Down syndrome samples. We have also compared the results of this molecular method with corresponding results obtained by the cytogenetic analysis. Blood samples obtained from DS patients (n=25) and normal controls (n=20) were tested by quantitative Real-time PCR in parallel to standard G-banding analysis. Genomic DNA was extracted from peripheral blood lymphocytes. A high precision TaqMan probe quantitative Real-time PCR assay was developed to determine the gene dosage of DSCAM (target gene on 21q22.2) relative to PMP22 (reference gene on 17p11.2). The DSCAM/PMP22 ratio was calculated according to the formula; ratio=2 -ΔΔCT. The quantitative Real-time PCR was able to distinguish between trisomy 21 samples and normal controls with the gene ratios of 1.49±0.13 and 1.03±0.04 respectively (p value <0.001). These results represent the presence of 3 copies of target gene in DS samples Vs 2 copies in normal controls. The results of quantitative Real-time PCR were in complete agreement with results of cytogenetic analysis. This study confirms previous reports regarding successful implementation of quantitative Real-time PCR for detection of trisomy 21. However, the assay has been improved by using MGB probes and more accurate data analysis. This assay, in particular, when performed in combination with another molecular assay such as QF-PCR or MLPA, can be used as a reliable technique for rapid prenatal diagnosis of trisomy 21.
