475 Computational Model for Predicting Effective siRNA Sequences Using Whole Stacking Energy (% G) for Gene Silencing

Authors: Reena Murali, David Peter S.

Abstract:

The small interfering RNA (siRNA) alters the regulatory role of mRNA during gene expression by translational inhibition. Recent studies show that upregulation of mRNA because serious diseases like cancer. So designing effective siRNA with good knockdown effects plays an important role in gene silencing. Various siRNA design tools had been developed earlier. In this work, we are trying to analyze the existing good scoring second generation siRNA predicting tools and to optimize the efficiency of siRNA prediction by designing a computational model using Artificial Neural Network and whole stacking energy (%G), which may help in gene silencing and drug design in cancer therapy. Our model is trained and tested against a large data set of siRNA sequences. Validation of our results is done by finding correlation coefficient of experimental versus observed inhibition efficacy of siRNA. We achieved a correlation coefficient of 0.727 in our previous computational model and we could improve the correlation coefficient up to 0.753 when the threshold of whole tacking energy is greater than or equal to -32.5 kcal/mol.

473 Using Adaptive Pole Placement Control Strategy for Active Steering Safety System

Authors: Hadi Adibi-Asl, Alireza Doosthosseini, Amir Taghavipour

Abstract:

This paper studies the design of an adaptive control strategy to tune an active steering system for better drivability and maneuverability. In the first step, adaptive control strategy is applied to estimate the uncertain parameters on-line (e.g. cornering stiffness), then the estimated parameters are fed into the pole placement controller to generate corrective feedback gain to improve the steering system dynamic’s characteristics. The simulations are evaluated for three types of road conditions (dry, wet, and icy), and the performance of the adaptive pole placement control (APPC) are compared with pole placement control (PPC) and a passive system. The results show that the APPC strategy significantly improves the yaw rate and side slip angle of a bicycle plant model.

468 Use of Curcumin in Radiochemotherapy Induced Oral Mucositis Patients: A Control Trial Study

Authors: Shivayogi Charantimath

Abstract:

Radiotherapy and chemotherapy are effective for treating malignancies but are associated with side effects like oral mucositis. Chlorhexidine gluconate is one of the most commonly used mouthwash in prevention of signs and symptoms of mucositis. Evidence shows that chlorhexidine gluconate has side effects in terms of colonization of bacteria, bad breadth and less healing properties. Thus, it is essential to find a suitable alternative therapy which is more effective with minimal side effects. Curcumin, an extract of turmeric is gradually being studied for its wide-ranging therapeutic properties such as antioxidant, analgesic, anti-inflammatory, antitumor, antimicrobial, antiseptic, chemo sensitizing and radio sensitizing properties. The present study was conducted to evaluate the efficacy and safety of topical curcumin gel on radio-chemotherapy induced oral mucositis in cancer patients. The aim of the study is to evaluate the efficacy and safety of curcumin gel in the management of oral mucositis in cancer patients undergoing radio chemotherapy and compare with chlorhexidine. The study was conducted in K.L.E. Society’s Belgaum cancer hospital. 40 oral cancer patients undergoing the radiochemotheraphy with oral mucositis was selected and randomly divided into two groups of 20 each. The study group A [20 patients] was advised Cure next gel for 2 weeks. The control group B [20 patients] was advised chlorhexidine gel for 2 weeks. The NRS, Oral Mucositis Assessment scale and WHO mucositis scale were used to determine the grading. The results obtained were calculated by using SPSS 20 software. The comparison of grading was done by applying Mann-Whitney U test and intergroup comparison was calculated by Wilcoxon matched pairs test. The NRS scores observed from baseline to 1st and 2nd week follow up in both the group showed significant difference. The percentage of change in erythema in respect to group A was 63.3% for first week and for second week, changes were 100.0% with p = 0.0003. The changes in Group A in respect to erythema was 34.6% for 1st week and 57.7% in second week. The intergroup comparison was significant with p value of 0.0048 and 0.0006 in relation to group A and group B respectively. The size of the ulcer score was measured which showed 35.5% [P=0.0010] of change in Group A for 1st and 2nd week showed totally reduction i.e. 103.4% [P=0.0001]. Authors: D. I. Ekine, C. C. Akpanibah

Abstract:

This paper analyses the productivity of median scale battery cage and deep litter chicken egg producers in Rivers State, Nigeria. 90 battery cage and 90 deep litter farmers giving a total of 180 farmers were sampled through a multistage sampling procedure. Mean productivity was higher for the battery cage than the deep litter farmers at 2.65 and 2.33 respectively. Productivity of battery cage farmers were positively influenced by age, extension contacts, experience and feed quantity while the productivity of deep litter farmers was positively influenced by age, extension contacts, household size, experience and labour. The major constraints identified by both categories are high cost of feed, high price of day-old chick, inadequate finance, lack of credit and high cost of drug/vaccination. Furthermore, the work recommends that government should assist chicken egg farmers through subsidies of input resources and put policies to make financial institutions give out loans at low interest rate to the farmers. The farmers should abide by the recommended number of birds per unit area while stocking.

460 Gold-Mediated Modification of Apoferritin Surface with Targeting Antibodies

Authors: Simona Dostalova, Pavel Kopel, Marketa Vaculovicova, Vojtech Adam, Rene Kizek

Abstract:

To ensure targeting of apoferritin nanocarrier with encapsulated doxorubicin drug, we used a peptide linker based on a protein G with N-terminus affinity towards Fc region of antibodies. To connect the peptide to the surface of apoferritin, the C-terminus of peptide was made of cysteine with affinity to gold. The surface of apoferritin with encapsulated doxorubicin (APODOX) was coated either with gold nanoparticles (APODOX-Nano) or gold(III) chloride hydrate reduced with sodium borohydride (APODOX-HAu). The reduction with sodium borohydride caused a loss of doxorubicin fluorescent properties and probably accompanied with the loss of its biological activity. Fluorescent properties of APODOX-Nano were similar to the unmodified APODOX; therefore it was more suited for the intended use. To evaluate the specificity of apoferritin modified with antibodies, ELISA-like method was used with the surface of microtitration plate wells coated by the antigen (goat anti-human IgG antibodies). To these wells, the nanocarrier was applied. APODOX without the modification showed 5× lower affinity to the antigen than APODOX-Nano modified gold and targeting antibodies (human IgG antibodies).

457 Multi-criteria Optimization of Square Beam using Linear Weighted Average Model

Authors: Ali Farhaninejad, Rizal Zahari, Ehsan Rasooliyazdi

Abstract:

Increasing energy absorption is a significant parameter in vehicle design. Absorbing more energy results in decreasing occupant damage. Limitation of the deflection in a side impact results in decreased energy absorption (SEA) and increased peak load (PL). Hence a high crash force jeopardizes passenger safety and vehicle integrity. The aims of this paper are to determine suitable dimensions and material of a square beam subjected to side impact, in order to maximize SEA and minimize PL. To achieve this novel goal, the geometric parameters of a square beam are optimized using the response surface method (RSM).multi-objective optimization is performed, and the optimum design for different response features is obtained.

Abstract:

In our modern society electricity is vital to our health, safety, comfort and well-being. While our daily use of electricity is often taken for granted, public concern has arisen about potential adverse health effects from electric and magnetic – electromagnetic – fields (EMFs) produced by our use of electricity. This paper aims to compare between the measured magnetic field values and the simulated models for the indoor medium to low voltage (MV/LV) distribution substations. To calculate the magnetic flux density in the substations, interactive software SUBCALC is used which is based on closed form solution of the Biot-Savart law with 3D conductor model. The comparison between the measured values and the simulated models was acceptable. However there were some discrepancies, as expected, may be due to the current variation during measurements.

451 Effect of Concrete Nonlinear Parameters on the Seismic Response of Concrete Gravity Dams

Authors: Z. Heirany, M. Ghaemian

Abstract:

Behavior of dams against the seismic loads has been studied by many researchers. Most of them proposed new numerical methods to investigate the dam safety. In this paper, to study the effect of nonlinear parameters of concrete in gravity dams, a twodimensional approach was used including the finite element method, staggered method and smeared crack approach. Effective parameters in the models are physical properties of concrete such as modulus of elasticity, tensile strength and specific fracture energy. Two different models were used in foundation (mass-less and massed) in order to determine the seismic response of concrete gravity dams. Results show that when the nonlinear analysis includes the dam- foundation interaction, the foundation-s mass, flexibility and radiation damping are important in gravity dam-s response.

With high speed vessels getting ever more sophisti-cated, travelling at higher and higher speeds and operating in With high speed vessels getting ever more sophisticated, travelling at higher and higher speeds and operating in areas of high maritime traffic density, training becomes of the highest priority to ensure that safety levels are maintained, and risks are adequately mitigated. Training onboard the actual craft on the actual route still remains the most effective way for crews to gain experience. However, operational experience and incidents during the last 10 years demonstrate the need for supplementary training whether in the area of simulation or man to man, man/ machine interaction. Training and familiarisation of the crew is the most important aspect in preventing incidents. The use of simulator, computer and web based training systems in conjunction with onboard training focusing on critical situations will improve the man machine interaction and thereby reduce the risk of accidents. Today, both ship simulator and bridge teamwork courses are now becoming the norm in order to improve further emergency response and crisis management skills. One of the main causes of accidents is the human factor. An efficient way to reduce human errors is to provide high-quality training to the personnel and to select the navigators carefully.areas of high maritime traffic density, training becomes of the highest priority to ensure that safety levels are maintained, and risks are adequately mitigated. 446 The Analysis of TRACE/PARCS in the Simulation of Ultimate Response Guideline for Lungmen ABWR

Authors: J. R. Wang, W.Y. Li, H.T. Lin, B.H. Lee, C. Shih, S.W. Chen

Abstract:

In this research, the TRACE/PARCS model of  Lungmen ABWR has been developed for verification of ultimate  response guideline (URG) efficiency. This ultimate measure was  named as DIVing plan, abbreviated from system depressurization,  water injection and containment venting. The simulation initial  condition is 100% rated power/100% rated core flow. This research  focuses on the estimation of the time when the fuel might be damaged  with no water injection by using TRACE/PARCS first. Then, the  effect of the reactor core isolation system (RCIC), control  depressurization and ac-independent water addition system (ACIWA),  which can provide the injection with 950 gpm are also estimated for  the station blackout (SBO) transient.
