29 Evaluation of Broad Leaf Weed Herbicides on Weed Control and Productivity of Wheat (Triticum Aestivum L.)

Abstract:

-- A field experiment was conducted at Holetta research center and farmers fields during 2017 and 2018 to determine the effects of haulauxifen-methyl + florasulam (QULEX 200 WG) on broadleaf weeds in wheat. The design was a Randomized Complete Block with three replications. The treatments were included haulauxifen-Methyl + florasulam @ 25gm, 50gm and 75gm ha-1, (King-D) 2, 4-D dimethyl amine @1.0 L ha-1, 2, 4-Dichlorophenoxy acetic acid @1.0 L ha-1 rate (standard check), farmers practice twice hand weeding (25-30 and 55-60) days after sowing and weedy check. Herbicides were applied with knapsack sprayer with a spray volume of 200 L ha-1. The wheat variety “Denda” was sown at 20 cm spacing. The recommended rate of fertilizer was applied. Weed density and biomass were recorded at (25-30 and 55-60) days after sowing. The results revealed that post emergence application of haulauxifen-methyl + florasulam @50gm ha-1 had a significant (P<0.05) effect on Guizotia scabra, Polygonum nepalense, Plantago lanceolata, Galinsoga parviflora, Sonchus spp., Galium spurium, Amaranthus hybridus, Raphanus raphanistrum and Medicago polymorpha population. The magnitude ranged from two to four folds when comparing with weed densities recorded in the unweeded plot. The grain yield harvested from the untreated check plot was significantly lower than the rest treatments. The grain yield was improved by 17.3% over the standard check with better performance.

28 Characterization of Some Bread Wheat Genotypes for Drought Tolerance Using Molecular Markers

Abstract:

Drought is the most important factor that limiting the production and productivity of wheat in the world. The yield of wheat, which is one of the most important crop in the world, reduced depend on drought. Researches to minimize effects of drought are one of the most important about breeding of drought resistant varieties. In recent years, benefiting from the drought resistance wild species and rapid advances in molecular biology studies, researches about drought have been accelerated and number of studies were made on molecular plant breeding which included the molecular mechanisms related to drought resistance. The aim of the present study was characterization of some bread wheat lines for drought tolerance which commonly cultivated in different location of Turkey. In this study, registered 9 bread wheat varieties which on the physiological tests about drought tolerance and 10 bread wheat line has been developed by Transitional Zone Agricultural Research Institute were used. SSR, STS, RAPD and SNP markers that associated with drought tolerance were used. The polymorphisms of the markers were determined by screening of two control varieties. For these purpose 40 molecular markers were used and 12 markers of them were polymorphic among the drought tolerance and the drought sensitive varieties. Control varieties were screened using polymorphic markers. All the DNAs on the genotypes will be searched for the presence of QTLs mapped to different chromosomes. Result of the research, the studied genotypes will be grouped according to drought tolerance and will be detected drought tolerance varieties by molecular markers. In addition, the results will be compared also with physiological tests. The drought tolerant wheat genotypes may be used in breeding studies related to drought stress.

27 Phosphorus Uptake of Triticale (Triticosecale Wittmack) Genotypes at Different Growth Stages

Abstract:

Triticale (Triticosecale Wittmack) is a man-made crop developed by crossing wheat (Triticum L.) and rye (Secale cereale L.). Triticale has until now been used mostly for animal feed; however, it can be consumed by humans in the form of biscuits, cookies, and unleavened bread. Moreover, one of the reasons for the development of triticale is that it is more efficient in nutrient deficient soil than wheat cultivars. After nitrogen fertilizer, phosphorus (P) is the most used fertilizer for crop production because P fixation occurs highly when it is applied the soil. The aim of the present study was to evaluate P uptake of winter triticale genotypes under different P fertilizer rates in different growth stages. The experiment was conducted in Eskisehir, Central Anatolia, Turkey. Treatments consisted of five triticale lines and one triticale cultivars (Samursortu) with four rates of P fertilization (0, 30, 60 and 120 kg P2O5 ha⁻¹). Phosphorus uptake of triticale genotypes in tillering, heading, as well as grain and straw at harvest stage and yield of grain and straw were determined. The results showed that a P rate of 60 kg/ha and the TCL-25 genotype produced the highest yields of straw and grain at harvest. Phosphorus uptake was the highest in tillering stage, and it decreased towards to harvest time. Phosphorus uptake of all growth stage increased as P rates raised and the application of 120 kg/ha P₂O₅ had the highest P uptake. Phosphorus uptake of genotypes was found differently. The regression analyses indicated that P uptake at tillering stage was the most effective on grain yield. These results will provide useful information to triticale growers about suitable phosphorus fertilization for both forage and food usage.

26 Diversification of Sweet Potato Blends and Utilization for Malnutrition and Poverty Alleviation

Abstract:

Value addition to agricultural produce is of possible potential in reducing poverty, improving food security and malnutrition, therefore the need to develop small and micro-enterprises of sweet potato production. The study was carried out in Nigeria to determine the acceptability of blends sweet potato (Ipomea batatas) and commodities yellow maize (Zea mays), millet (Pennisetum glaucum), soybean (Glycine max), bambara groundnut (Vigna subterranean), guinea corn (Sorghum vulgare), wheat (Triticum aestivum), and roselle (Hibiscus sabdariffa) through sensory evaluation. Sweet potato (Ipomea batatas) roots were processed using two methods. The first method involved the use of a fabricated gas powered cabinet dryer to dry sulphited chips and the second method was the use of traditional sun drying method without the addition of the chemical. The blends were also assessed in terms of functional, chemical and color properties. Most acceptable blends include BAW (80:20 of sweet potato/wheat), BBC (80:20 of sweet potato/guinea corn), AAB (60:40 of sweet potato/guinea corn), YTE (100% soybean), TYG (100% sweet potato), KTN (100% wheat flour), XGP (80:20 of sweet potato/soybean), XAX (60:40 of sweet potato/wheat), LSS (100% Roselle), CHK (100% Guinea corn), and ABC (60:40% of sweet potato/ yellow maize). In addition, chemical analysis carried out revealed that sweet potato has high percentage of vitamins A and C, potassium (K), manganese (Mn), calcium (Ca), magnesium (Mg) and iron (Fe) and fibre content. There is also an increase of vitamin A and Iron in the blended products.

25 Susceptibility Assessment and Genetic Diversity of Iranian and CIMMYT Wheat Genotypes to Common Root Rot Disease Bipolaris sorokiniana

Abstract:

Wheat, Triticum aestivum L. is one of the most important and strategic crops in the human diet. Several diseases threaten this particular crop. Common root rot disease of wheat by a fungal agent, Bipolaris sorokiniana is one of the important diseases, causing considerable losses worldwide. Resistant sources are the only feasible and effective method of control for managing diseases. In this study, the response of 33 domestic and exotic wheat genotypes, including cultivars and promising lines were screened to B. sorokiniana at greenhouse and field conditions, based on five scoring scale indexes of 0 to 100 severity percentage. The screening was continued on resistant wheat genotypes and repeated several times to confirm the greenhouse and field results. Statistical and cluster analysis of data was performed using SAS and SPSS software, respectively. The results showed that, the response of wheat genotypes to the disease in the greenhouse and field conditions was highly significant. The highest rate of common root rot disease infection, B. sorokiniana in the greenhouse and field, was of CVS. Karkheh and Beck Cross-Roshan with 60.83% and 59.16% disease severity respectively, and the lowest one were in cv. Alvand with 18.33%, followed by cv. Baharan with 19.16% disease severity, with a highly significant difference respectively. The remaining wheat genotypes were located in between these two highest and lowest infected groups to B. sorokiniana significantly. There was a high correlation coefficient between the related statistical groups and cluster analysis.

24 Effect of Plant Growth Regulator on Vegetative Growth and Yield Components of Winter Wheat under Different Levels of Irrigation

Abstract:

Field experiment were carried out to investigate the effect of the plant growth regulator on vegetative growth and yield components of reduced height isogenic lines of the wheat (Triticum aestivum L.) cultivar Mercia. The Field experiment compared the growth regulator response of seven isogenic lines of Mercia. Growth regulators reduced plant height significantly in all lines. Growth regulator decreased total dry matter and grain yield with greatest reduction generally for the control and Rht8 lines. Rht1 was the least affected. There were few significant effects of growth regulator on gas exchange and chlorophyll fluorescence but the trend was for greater values with growth regulator. In this field experiment, a rate of 2.0 l ha-1 applied just before the third node detectable stage under non water stressed and water stressed conditions gave slight increases in yield of up to 14% except for line Rht10 which increased significantly in non-stressed conditions. In the second glasshouse experiment, a rate of 2.5 l ha-1 applied at the start of stem elongation under 30% FC and 100% FC gave reductions in yield up to 16% for the growth regulator and 55% under water stress. In the field experiment, rates of 2.5 and 3.0 l ha-1 applied at the start of stem elongation gave reductions in yield up to 20% mainly through individual seed weight. In the final glasshouse experiment, rates of 2.5 and 3.0 l ha-1 applied at 6 leaves unfolded and 1st node detectable both reduced grain yield.

23 Carbon Dioxide (CO₂) and Methane (CH₄) Fluxes from Irrigated Wheat in a Subtropical Floodplain Soil Increased by Reduced Tillage, Residue Retention, and Nitrogen Application Rate

Abstract:

Quantifying carbon (C) sequestration in soils is necessary to help better understand the effect of agricultural practices on the C cycle. The estimated contribution of agricultural carbon dioxide (CO₂) and methane (CH₄) to global warming potential (GWP) has a wide range. The underlying causes of this huge uncertainty are the difficulties to predict the regional CO₂ and CH₄ loss due to the lack of experimental evidence on CO₂ and CH₄ emissions and associated drivers. The CH₄ and CO₂ emissions were measured in irrigated wheat in subtropical floodplain soils which have been under two soil disturbance levels (strip vs. conventional tillage; ST vs. CT being both with 30% residue retention) and three N fertilizer rates (60, 100, and 140% of the recommended N fertilizer dose, RD) in annual wheat (Triticum aestivum)-mungbean (Vigna radiata)-rice (Oryza sativa L) for seven consecutive years. The highest CH₄ and CO₂ emission peak was observed on day 3 after urea application in both tillages except CO₂ flux in CT. Nitrogen fertilizer application rate significantly influenced mean and cumulative CH₄ and CO₂ fluxes. The CH₄ and CO₂ fluxes decreased in an optimum dose of N fertilizer except for ST for CH₄. The CO₂ emission significantly showed higher emission at minimum (60% of RD) fertilizer application at both tillages. Soil microbial biomass carbon (MBC), organic carbon (SOC), Particulate organic carbon (POC), permanganate oxidisable carbon (POXC), basal respiration (BR) were significantly higher in ST which were negative and significantly correlated with CO₂. However, POC and POXC were positively and significantly correlated with CH₄ emission.

22 Energy Budgeting, Carbon and Water Footprints Under Conventional and Conservation Tillage Practices of Rice-Wheat Double Cropping System

Abstract:

Amid the present environmental crises, developing environment-resilient and cost-effective conservation agriculture strategies to feed the world's ever-growing population is pertinent. Therefore, a field study was conducted to test the hypothesis that residue retention under no-till (NTR) would enhance energy productivity (EP) and energy use efficiency (EUE) while offsetting the carbon footprints (CF), water footprints (WF) and greenhouse gases emissions (GHGs) in rice (Oryza sativa L.)-wheat (Triticum aestivum L.) double cropping system. Two tillage systems viz., conventional tillage (CT) and conservation tillage (no-till; NT), with or without residue retention, were combined into four treatments as CT0 (puddled rice, conventional wheat - residue); CTR (puddled rice, conventional wheat + residue); NT0 (direct rice seeding, zero-tilled wheat - residue); NTR (direct rice seeding, zero-tilled wheat + residue) were evaluated. Overall, results showed that the NT system had 34.2% lower energy consumption, 1.2 times more EP than CT system. Moreover, NTR had 19.8% higher EUE than CT0. The overall system grain yield ranged from 7.8 to 9.3 Mg ha−1 under NT0 and CTR, respectively. The NTR had 56.6% and 17.9% lesser CF and WF, respectively, than CT0. The net GHGs emissions (CO2-eq kg ha−1) under CT0 were the highest, while NTR had the lowest emissions. The NTR enhanced carbon sequestration in soil that can offset half of the system's CO2 emissions. The findings of this study might help develop a suitable strategy for resource/energy conservation and higher productivity while offsetting GHGs emissions in the Indo-Gangetic Plains.

21 Perspectives and Challenges Functional Bread with Yeast Extract to Improve Human Diet

Abstract:

In the last decades, the urban population has been characterized by sedentary lifestyles, low physical activity, and "fast food". These changes in diet and physical nonactivity have been associated with an increase in chronic diseases. Bread is one of the most popular wheat products consumed worldwide. Spelt wheat has shown potential in various food applications, including bread, pasta, breakfast cereal, and other products of altered nutritional characteristics compared to conventional wheat products. It has very high protein content and even 30 to 60% higher concentration of mineral elements Fe, Zn, Cu, Mg and P compared to Triticum Aestivum. Spelt wheat is growing without the use of pesticides in harsh ecological conditions and it is an old cultivar. So it can be used for organic and health-safe food. Changes in the formulation of bread with the aim of improving its nutritional and functional properties usually lead to changes in the dough's properties, which are related to the quality of the finished product. The aim of this paper is to research the impact of adding yeast extract to bread on sensory characteristics and consumer acceptance of a new product as a key factor for the successful marketing of a distinct product. The sensory analysis of bread with 5% yeast extract is as follows: the technological quality is very good (3.8), and the color of the product is excellent (4.85). Based on data, consumers' survey declared that they liked the taste of bread with 5% yeast extract (74%), consumers marked the product as likable (70%), and 75% of the total number of respondents would buy this new product. This paper is promoting a type of bread with 5% yeast extract (Z score 0.80) to improve diet and a product intended for consumers conscious about their health and diet.

19 Management Effects on Different Sustainable Agricultural with Diverse Topography

Abstract:

Crop yields are influenced by many factors, including natural ones, such as soil and environmental characteristics of the agricultural land, as well as manmade ones, such as management applications. One of the factors that frequently affect crop yields in undulating Midwest landscapes is topography, which controls the movement of water and nutrients necessary for plant life. The main objective of this study is to examine how field topography influences performance of different management practices in undulated terrain of southwest Michigan. A total of 26 agricultural fields, ranging in size from 1.1 to 7.4 ha, from the Scale-Up at Kellogg Biological Station were included in the study. The two studied factors were crop species with three levels, i.e., corn (Zea mays L.) soybean (Glycine max L.), and wheat (Triticum aestivum L.), and management practice with three levels, i.e., conventional, low input, and organic managements. They were compared under three contrasting topographical settings, namely, summit (includes summits and shoulders), slope (includes backslopes), and depression (includes footslope and toeslope). Yield data of years 2007 through 2012 was processed, cleaned, and filtered, average yield then was calculated for each field, topographic setting, and year. Topography parameters, including terrain, slope, curvature, flow direction and wetness index were computed under ArcGIS environment for each topographic class of each field to seek their effects on yield. Results showed that topographical depressions produced greatest yields in most studied fields, while managements with chemical inputs, both low input and conventional, resulted in higher yields than the organic management.

18 Bio Energy from Metabolic Activity of Bacteria in Plant and Soil Using Novel Microbial Fuel Cells

Abstract:

Microbial fuel cells (MFCs) are an emerging and promising method for achieving sustainable energy since they can remove contaminated organic matter and simultaneously generate electricity. Our approach was driven in three different ways like Bacterial fuel cell, Soil Microbial fuel cell (Soil MFC) and Plant Microbial fuel cell (Plant MFC). Bacterial MFC: Sulphate reducing bacteria (SRB) were isolated and identified as the efficient electricigens which is able to produce ±2.5V (689mW/m2) and it has sustainable activity for 120 days. Experimental data with different MFC revealed that high electricity production harvested continuously for 90 days 1.45V (381mW/m2), 1.98V (456mW/m2) respectively. Biofilm formation was confirmed on the surface of the anode by high content screening (HCS) and scanning electron Microscopic analysis (SEM). Soil MFC: Soil MFC was constructed with low cost and standard Mudwatt soil MFC was purchased from keegotech (USA). Vermicompost soil (V1) produce high energy (± 3.5V for ± 400 days) compared to Agricultural soil (A1) (± 2V for ± 150 days). Biofilm formation was confirmed by HCS and SEM analysis. This finding provides a method for extracting energy from organic matter, but also suggests a strategy for promoting the bioremediation of organic contaminants in subsurface environments. Our Soil MFC were able to run successfully a 3.5V fan and three LED continuously for 150 days. Plant MFC: Amaranthus candatus (P1) and Triticum aestivium (P2) were used in Plant MFC to confirm the electricity production from plant associated microbes, four uniform size of Plant MFC were constructed and checked for energy production. P2 produce high energy (± 3.2V for 40 days) with harvesting interval of two times and P1 produces moderate energy without harvesting interval (±1.5V for 24 days). P2 is able run 3.5V fan continuously for 10days whereas P1 needs optimization of growth conditions to produce high energy.

17 Amino Acid Responses of Wheat Cultivars under Glasshouse Drought Accurately Predict Yield-Based Drought Tolerance in the Field

Abstract:

Water limits crop productivity, so selecting for minimal yield-gap in drier environments is critical to mitigate against climate change and land-use pressures. To date, no markers measured in glasshouses have been reported to predict field-based drought tolerance. In the field, the best measure of drought tolerance is yield-gap; but this requires multisite trials that are an order of magnitude more resource intensive and can be impacted by weather variation. We investigated the responses of relative water content (RWC), stomatal conductance (gs), chlorophyll content and metabolites in flag leaves of commercial wheat (Triticum aestivum L.) cultivars to three drought treatments in the glasshouse and field environments. We observed strong genetic associations between glasshouse-based RWC, metabolites and Yield gap-based Drought Tolerance (YDT): the ratio of yield in water-limited versus well-watered conditions across 24 field environments spanning sites and seasons. Critically, RWC response to glasshouse drought was strongly associated with both YDT (r2 = 0.85, p < 8E-6) and RWC under field drought (r2 = 0.77, p < 0.05). Multiple regression analyses revealed that 98% of genetic YDT variance was explained by drought responses of four metabolites: serine, asparagine, methionine and lysine (R2 = 0.98; p < 0.01). Fitted coefficients suggested that, for given levels of serine and asparagine, stronger methionine and lysine accumulation was associated with higher YDT. Collectively, our results demonstrate that high-throughput, targeted metabolic phenotyping of glasshouse-grown plants may be an effective tool for the selection of wheat cultivars with high YDT in the field.

16 Bioefficacy of Diclosulam for Controlling Weeds in Soybean [Glycine Max (L.) Merrill] and Its Carry Over Effect on Succeeding Wheat (Triticum Aestivum) Crop

Abstract:

The experiment was conducted at Agricultural Research Station, Agriculture University, Kota, Rajasthan, India during kharif and rabi 2020-21 and 2021-22 to study the biofficacy of diclosulam and its residual effect on succeeding wheat crop. The treatments comprised of Diclosulam 84 % WDG viz. 6.25, 12.50, 25.00 and 37.50 g/ha as pre emergence (PE), Pendimethalin 30% EC 3.33 l/ha, Sulfentrazon 48% SC 750 g/ha, hand weeding at 30 and 45 DAS and weedy check, were evaluated in randomized block design in three replications. The experimental soil was clay in texture and non-calcareous. Experimental field was mainly dominated by grasses-Echinochloa colonum, E.crusgalli,Cynodon dactylon, Sedges-Cyperus rotundus and broad leaved weeds Celosia argentea and Digera arvensis.The result revealed that application of Diclosulam 84 % WDG 25 g/ha PE was found effective in controlling mostly weed species and registered higher weed control efficiency 81.2, 74.3, 69.6 per cent at 30, 45 days after sowing and at harvest. Diclosulam 84 % WDG (6.25-25.0 g/ha) was found selective to the soybean crop as no any phytotoxicity symptoms were observed. Among the herbicidal treatments, Diclosulam 84 % WDG 25 g/ha registered maximum and significantly higher soybean seed yield (1889 and 1431 kg/ha during kharif 2020 and 2021, respectively and was at par with Sulfentrazone 48% SC 750 g/ha and over weedy check( 1027 and 667 kg/ha).The wheat crop growth, yield attributes and seed yield were not influenced due to carry over effect of the Diclosulam 84 % WDG( 6.25-25.0 g/ha) and no any phytotoxicity symptoms were observed. Henceforth, the Diclosulam 84 % WDG 25.0 g/ha as pre emergence may be used in the soybean for effective weed control without carry over effect on succeeding wheat crop.

15 Influence of Digestate Fertilization on Soil Microbial Activity, Greenhouse Gas Emissions and Yield

Abstract:

Agricultural wastes contribute significantly to global climate change through greenhouse gas emissions if not adequately recycled and sustainably managed. A recurring agricultural waste is livestock wastes that have consistently served as feedstock for biogas systems. The objective of this study was to access the influence of digestate fertilization on soil microbial activity and greenhouse gas emissions in agricultural fields. Wheat (Triticum spp. L.) was fertilized with different types of animal wastes digestates (organic fertilizers) and mineral nitrogen (inorganic fertilizer) for three years. The 170 kg N ha⁻¹ presented in digestates were split fertilized at an application rate of 90 and 80 kg N ha⁻¹. The soil microorganism activity could be predicted significantly using the dehydrogenase activity and soil microbial biomass carbon. By combining the two different monitoring approaches, the different methods applied in this study were sensitive to enzymatic activities and organic carbon in the living component of the soil organic matter. The emissions of greenhouse gasses (carbon dioxide (CO₂), methane (CH₄), and nitrous oxide (N₂O) were monitored directly by a static chamber system. The soil and environmental variables were measured to determine their influence on greenhouse gas emissions. Emission peaks was observed in N₂O and CO₂ after the first application of fertilizers with the emissions flattening out over the cultivating season while CH₄ emission was negligible with no apparent patterns observed. Microbial biomass carbon and dehydrogenase activity were affected by the fertilized organic digestates. A significant difference was recorded between the control and the digestate treated soils for the microbial biomass carbon and dehydrogenase. Results also showed individual and cumulative emissions of CO₂, CH₄ and N₂O from the digestates were relatively low suggesting the digestate fertilization can be an efficient method for improving soil quality and reducing greenhouse gases from agricultural sources in temperate climate conditions.

14 Seed Priming Winter Wheat (Triticum aestivum L.) for Germination and Emergence

Abstract:

In order to evaluate the effect of the different sources of salt on germination and early growth of five wheat cultivars (Katea, Bezostaja, Koksal-2000, Golia, Pehlivan) an experiment was conducted at the seed laboratory of the Uludag University, Agricultural Faculty, Department of Field Crops in Bursa/Turkey. Seeds were applied in five different resources media (KCl % 2, KCl %4, KNO₃ %0,5, KH₂PO₄ %0,5, PEG %10) and distilled water as the control). The seed was fully immersed in priming media at a temperature of 24ᵒC for durations of 12 and 24hours. Six different agronomic characters (seed germination, stem length, stem weight, radicle length, fresh weight, dry weight) were measured in 7th days and 14th days. Maximum seed germination percentage of seven days are Pehlivan was observed when the seeds were applied by KH₂PO₄ and Katea by distilled water as a control. The most stem length and stem weight were obtained for seeds were applied by KH₂PO₄ %0,5 with Katea and Bezostja immersed in priming media at 12h intervals beginning 7d after planting. Seeds were applied KH₂PO₄ %0,5 media produced maximum radicle length by Koksal and dry weight by Katea. The freshest weight obtains in Katea by KNO₃ %0,5 immersed in priming media at 24h. The most germination percent, dry weight, stem length of fourteen days was observed in Katea which subjected to KH₂PO₄ %0,5 solution. The most radicle length was observed Katea and Koksal in media of KH₂PO₄ %0,5. The most stem length was obtained for seeds were applied by KH₂PO₄ %0,5 and KNO₃ with Katea and Bezostaja. When the applied chemicals and all days examined KH₂PO₄ %0,5 treatment in fourteen days and immersed for the duration of 24 hours had better effects than other medias, seven days treatments and 12hours immersed. As a result of this research, the best response of media for the wheat germination can be said that the KH₂PO₄ %0,5 immersed in priming media at 24h intervals beginning 14 days after planting.

13 The Source of Fibre and Roxazyme® G2 Interacted to Influence the Length of Villi in the Ileal Epithelium of Growing Pigs Fed Fibrous Maize-Soybean Diets

Abstract:

The effects of dietary fibre source on the histomorphology of the ileal epithelium were examined in growing pigs fed high fibre (242-250 g total dietary fibre kg-1 dry matter) diets fortified with Roxazyme® G2. The control was a standard, low fibre (141 g total dietary fibre kg-1 dry matter) diet formulated from dehulled soybean (Glycine max), maize (Zea Mays) meal and hominy chop. Five fibrous diets were evaluated in which fibre was increased by partial substitution of the grains in the control diet with maize cobs, soybean hulls, barley (Hordeum vulgare L) brewer’s grains, Lucerne (Medicago sativa) hay or wheat (Triticum aestivum) bran. Each diet was duplicated and 220 mg Roxazyme® G2 kg-1 dry mater was added to one of the mixtures. Seventy-two intact Large White X Landrace male pigs of weight 32 ± 5.6 kg pigs were randomly allocated to the diets in a complete randomised design with a 2 (fibre source) X (enzyme) factorial arrangement of treatments. The pigs were fed ad libitum for 10 weeks. Ileal tissue samples were taken at slaughter, at a point 50cm above the ileal-caecal valve. Villi length and area, and crypt depth were measured by computerised image analyses. The villi length: crypt ratio was calculated. The diet and the supplemental enzyme cocktail did not affect (p>0.05) any of the measured parameters. Significant (p=0.016) diet X enzyme interaction was observed for villi length whereby the enzyme reduced the villi length of pigs on the soy-hulls, standard and wheat bran diets, with an opposite effect on pigs on the maize cob, brewer’s grain, Lucerne diets. The results suggested fibre-source dependent changes in the morphology of the ileal epithelium of pigs fed high fibre, maize-soybean diets fortified with Roxazyme® G2.

12 Development of a New Method for the Evaluation of Heat Tolerant Wheat Genotypes for Genetic Studies and Wheat Breeding

Abstract:

Heat is one of the major abiotic stresses limiting wheat production worldwide. To identify heat tolerant genotypes, a newly designed system involving a large plastic box holding many layers of filter papers positioned vertically with wheat seeds sown in between for the ease of screening large number of wheat geno types was developed and used to study heat tolerance. A collection of 499 wheat geno types were screened under heat stress (35ºC) and non-stress (25ºC) conditions using the new method. Compared with those under non-stress conditions, a substantial and very significant reduction in seedling length (SL) under heat stress was observed with an average reduction of 11.7 cm (P<0.01). A damage index (DI) of each geno type based on SL under the two temperatures was calculated and used to rank the genotypes. Three hexaploid geno types of Triticum aestivum [Perenjori (DI= -0.09), Pakistan W 20B (-0.18) and SST16 (-0.28)], all growing better at 35ºC than at 25ºC were identified as extremely heat tolerant (EHT). Two hexaploid genotypes of T. aestivum [Synthetic wheat (0.93) and Stiletto (0.92)] and two tetraploid genotypes of T. turgidum ssp dicoccoides [G3211 (0.98) and G3100 (0.93)] were identified as extremely heat susceptible (EHS). Another 14 geno types were classified as heat tolerant (HT) and 478 as heat susceptible (HS). Extremely heat tolerant and heat susceptible geno types were used to develop re combinant inbreeding line populations for genetic studies. Four major QTLs, HTI4D, HTI3B.1, HTI3B.2 and HTI3A located on wheat chromosomes 4D, 3B (x2) and 3A, explaining up to 34.67 %, 28.93 %, 13.46% % and 11.34% phenotypic variation, respectively, were detected. The four QTLs together accounted for 88.40% of the total phenotypic variation. Random wheat geno types possessing the four heat tolerant alleles performed significantly better under the heat condition than those lacking the heat tolerant alleles indicating the importance of the four QTLs in conferring heat tolerance in wheat. Molecular markers are being developed for marker assisted breeding of heat tolerant wheat.

11 Trehalose Application Increased Membrane Stability and Cell Viability to Affect Growth of Wheat Genotypes under Heat Stress

Abstract:

Heat stress is one of the major environmental factors drastically reducing wheat production. Crop heat tolerance can be enhanced by preconditioning of plants by exogenous application of osmoprotectants. Presently, the effect of trehalose pretreatment (at 1 mM, and 1.5 nM) under heat stress of 35±2˚C (moderate) and 40±2˚ (severe) for four and eight hour was conducted in wheat (Tricticum aestivum L.) genotypes viz. HD2967, PBW 175, PBW 343, PBW 621, and PBW 590. Heat stress affects wide spectrum of physiological processes within plants that are irreversibly damaged by stress. Membrane thermal stability (MTS) and cell viability was significantly decreased under heat stress for eight hours. Pretreatment with trehalose improved MTS and cell viability under stress and this effect was more promotory with higher concentration. Thermal stability of photosynthetic apparatus differed markedly between genotypes and Hill reaction activity was recorded more in PBW621 followed by C306 as compared with others. In all genotypes photolysis of water showed decline with increase in temperature stress. Trehalose pretreatment helped in sustaining Hill reaction activity probably by stabilizing the photosynthetic apparatus against heat-induced photo inhibition. Both plant growth and development were affected by temperature in both shoot and root under heat stress. The reduction was compensated partially by trehalose (1.5 mM) application. Adaption to heat stress is associated with the metabolic adjustment which led to accumulation of soluble sugars including non-reducing and reducing for their role in adaptive mechanism. Higher acid invertase activity in shoot of tolerant genotypes appeared to be a characteristic for stress tolerance. As sucrose synthase play central role in sink strength and in studied wheat genotype was positively related to dry matter accumulation. The duration of heat stress for eight hours had more severe effect on these parameters and trehalose application at 1.5 mM ameliorated it to certain extent.

10 Grain Yield, Morpho-Physiological Parameters and Growth Indices of Wheat (Triticum Aestivum L.) Varieties Exposed to High Temperature under Late Sown Condition

Abstract:

A field experiment was carried out in Factorial Randomized Block Design (FRBD) with three replications at Instructional Farm Krushigadh, Junagadh Agricultural University, Junagadh, India to assess the biochemical parameters of wheat in order to assess the thermotolerance. Nine different wheat varieties GW 433, GW 431, HI 1571, GW 432, RAJ 3765, HD 2864, HI 1563, HD 3091 and PBW 670 sown in timely and late sown conditions (i.e., 22 Nov and 6 Dec 2012) were analysed. All the varieties differed significantly with respect to grain yield morpho-physiological parameters and growth indices for time of sowing, varieties and varieties x time of sowing interactions. The observations on morpho-physiological parameters viz., germination percentage, canopy temperature depression and growth indices viz., leaf area index (LAI), leaf area ratio (LAR) were recorded. Almost all the morpho-physiological parameters, growth indices and grain yield studied were affected adversely by late sowing, registering reduction in their magnitude. Germination percentage was reduced under late sown condition but variety PBW 670 was the best. Varieties GW 432 performed better with respect to canopy temperature depression while sown late. Under late sown condition, variety GW 431 recorded higher LAI while HI 1563 had maximum LAR. Considering yield performance, HD 2864 was best under timely sown condition, while GW 433 was best under late sown condition. Varieties HI 1571, GW 433 and GW 431 could be labelled as thermo-tolerant because there was least reduction in grain yield under late sown condition (1.75 %, 7.90 % and13.8 % respectively). Considering correlation coefficient, grain yield showed very strong significant positive association with germination percentage. Leaf area ratio was strongly and significantly correlated with grain yield but in negative direction. Canopy temperature depression and leaf area index also had positive correlation with grain yield but were non-significant.

9 The Effects of Fungicide and Genetics on Fungal Diseases on Wheat in Nebraska With Emphasis on Stem Rust

Abstract:

Wheat (Triticum aestivum L.) production continues to be challenged by foliar fungal diseases although significant improvement has been made to manage the diseases through developing resistant varieties and the fungicide use to ensure sufficient wheat is produced to meet the growing population’s need. Significant crop losses have been recorded in the history of grain production and yield losses due to fungal diseases, and the trend continues to threat food security in the world and particularly in the less developed countries. The impact of individual fungal diseases on grain yield has been studied extensively to determine crop losses. However, there is limited research available to find out the combined effects of fungal diseases on grain yield and the ways to effectively manage the diseases. Therefore, the objectives of this research were to study the effect of fungal pathogens on grain yield of pre-released winter wheat genotypes in fungicide treated and untreated plots, and to determine whether S7b gene was present in ‘Gage’ wheat as previously hypothesized. Sixty winter wheat genotypes in fungicide treated and untreated plots were studied across four environments. There was a significant effect of fungicide on grain yield consistently across four environments in three years. Fungicide treated wheat lines demonstrated (4,496 kg/ ha-1) grain yield compared to (3,147 kg/ ha-1) grain yield in untreated wheat lines indicating 43% increased grain yield due to severity of foliar fungal diseases. Furthermore, fungicide application also caused an increase in protein concentration from 153 (g kg-1) to 164 (g kg-1) in treated plots in along with test weight from 73 to 77 (kg hL-1) respectively. Gage wheat variety and ISr7b-Ra were crossed to determine presence of Sr7b in Gage. The F2 and F2:3 segregating families were screened and evaluated for stem rust resistance. The segregation of families fell within 15:1 ratio for two separate resistance genes suggesting that Sr7b segregates independently from an unknown resistance gene in Gage that needs to be characterized for its use in the future wheat breeding program to develop resistant wheat varieties.

8 Biological Control of Karnal Bunt by Pseudomonas fluorescens

Abstract:

Pseudomonas species possess a variety of promising properties of antifungal and growth promoting activities in the wheat plant. In the present study, Pseudomonas fluorescens MTCC-9768 is tested against plant pathogenic fungus Tilletia indica, causing Karnal bunt, a quarantine disease of wheat (Triticum aestivum) affecting kernels of wheat. It is one of the 1/A1 harmful diseases of wheat worldwide under EU legislation. This disease develops in the growth phase by the spreading of microscopically small spores of the fungus (teliospores) being dispersed by the wind. The present chemical fungicidal treatments were reported to reduce teliospores germination, but its effect is questionable since T. indica can survive up to four years in the soil. The fungal growth inhibition tests were performed using Dual Culture Technique, and the results showed inhibition by 82.5%. The interaction of antagonist bacteria-fungus causes changes in the morphology of hyphae, which was observed using Lactophenol cotton blue staining and Scanning Electron Microscopy (SEM). The rounded and swollen ends, called ‘theca’ were observed in interacted fungus as compared to control fungus (without bacterial interaction). This bacterium was tested for its antagonistic activity like protease, cellulose, HCN production, Chitinase, etc. The growth promoting activities showed increase production of IAA in bacteria. The bacterial secondary metabolites were extracted in different solvents for testing its growth inhibiting properties. The characterization and purification of the antifungal compound were done by Thin Layer Chromatography, and Rf value was calculated (Rf value = 0.54) and compared to the standard antifungal compound, 2, 4 DAPG (Rf value = 0.54). Further, the in vivo experiments showed a significant decrease in the severity of disease in the wheat plant due to direct injection method and seed treatment. Our results indicate that the extracted and purified compound from the antagonist bacteria, P. fluorescens MTCC-9768 may be used as a potential biocontrol agent against T. indica. This also concludes that the PGPR properties of the bacteria may be utilized by incorporating it into bio-fertilizers.

7 Screening of Wheat Wild Relatives as a Gene Pool for Improved Photosynthesis in Wheat Breeding

Abstract:

The rate of genetic progress in wheat production must be improved to meet global food security targets. However, past selection for domestication traits has reduced the genetic variation in modern wheat cultivars, a fact that could severely limit the future rate of genetic gain. The genetic variation in agronomically important traits for the wild relatives and progenitors of wheat is far greater than that of the current domesticated cultivars, but transferring these traits into modern cultivars is not straightforward. Between the elite cultivars of wheat, photosynthetic capacity is a key trait for which there is limited variation. Early screening of wheat wild relative and progenitors has shown differences in photosynthetic capacity and efficiency not only between wild relative species but marked differences between the accessions of each species. By identifying wild relative accessions with improved photosynthetic traits and characterising the genetic variation responsible, it is possible to incorporate these traits into advanced breeding programmes by wide crossing and introgression programmes. To identify the potential variety of photosynthetic capacity and efficiency available in the secondary and tertiary genepool, a wide scale survey was carried out for over 600 accessions from 80 species including those from the genus Aegilops, Triticum, Thinopyrum, Elymus, and Secale. Genotype data were generated for each accession using a ‘Wheat Wild Relative’ Single Nucleotide Polymorphism (SNP) genotyping array composed of 35,000 SNP markers polymorphic between wild relatives and elite hexaploid wheat. This genotype data was combined with phenotypic measurements such as gas exchange (CO₂, H₂O), chlorophyll fluorescence, growth, morphology, and RuBisCO activity to identify potential breeding material with enhanced photosynthetic capacity and efficiency. The data and associated analysis tools presented here will prove useful to anyone interested in increasing the genetic diversity in hexaploid wheat or the application of complex genotyping data to plant breeding.

6 The Efficacy of Salicylic Acid and Puccinia Triticina Isolates Priming Wheat Plant to Diuraphis Noxia Damage

Abstract:

Russian wheat aphid (Diuraphis noxia, Kurdjumov) is considered an economically important wheat (Triticum aestivum L.) pest worldwide and in South Africa. The RWA damages wheat plants and reduces annual yields by more than 10%. Even though pest management by pesticides and resistance breeding is an attractive option, chemicals can cause harm to the environment. Furthermore, the evolution of resistance-breaking aphid biotypes has out-paced the release of resistant cultivars. An alternative strategy to reduce the impact of aphid damage on plants, such as priming, which sensitizes plants to respond effectively to subsequent attacks, is necessary. In this study, wheat plants at the seedling and flag leaf stages were primed by salicylic acid and isolate representative of two races of the leaf rust pathogen Puccinia triticina Eriks. (Pt), before RWA (South African RWA biotypes 1 and 4) infestation. Randomized complete block design experiments were conducted in the greenhouse to study plant-pest interaction in primed and non-primed plants. Analysis of induced aphid damage indicated salicylic acid differentially primed wheat cultivars for increased resistance to the RWASA biotypes. At the seedling stage, all cultivars were primed for enhanced resistance to RWASA1, while at the flag leaf stage, only PAN 3111, SST 356 and Makalote were primed for increased resistance. The Puccinia triticina efficaciously primed wheat cultivars for excellent resistance to RWASA1 at the seedling and flag leaf stages. However, Pt failed to enhance the four Lesotho cultivars' resistance to RWASA4 at the seedling stage and PAN 3118 at the flag leaf stage. The induced responses at the seedling and flag leaf stages were positively correlated in all the treatments. Primed plants induced high activity of antioxidant enzymes like peroxidase, ascorbate peroxidase and superoxide dismutase. High antioxidant activity indicates activation of resistant responses in primed plants (primed by salicylic acid and Puccina triticina). Isolates of avirulent Pt races can be a worthy priming agent for improved resistance to RWA infestation. Further confirmation of the priming effects needs to be evaluated at the field trials to investigate its application efficiency.

5 Impact of ‎Foliar ‎Formulations of Macro and Micro Nutrients on ‎the ‎Tritrophic Association of Wheat Aphid ‎and Entomophagous Insects

In Pakistan, wheat (Triticum aestivum L.) is seriously attacked by the wheat ‎aphid. Naturally, bio control agents play an important role in managing wheat aphid. However, association ‎among pest, natural enemies and host plant is highly affected by food resource ‎concentration and predator/parasitoid factor of any ecosystem. The present ‎study was conducted to estimate the effect of different dose levels of macro ‎and micronutrients on the aphid population and its entomophagous insect ‎on wheat and their tri-trophic association. The experiment was laid out in ‎RCBD with six different combinations of macro and micronutrients and a control treatment. The data was initiated from the second week of ‎the February till the maturity of the crop. Data regarding aphid population and ‎coccinellids counts were collected on weekly basis and subjected to analysis of ‎variance and mean comparison. The data revealed that aphid ‎population was at peak in the last week of March. Coccinellids population ‎increased side by side with aphid population and declined after second week of ‎April. Aphid parasitism was maximum 25% on recommended dose of Double and ‎Flasher and minimum 8.67% on control treatment. Maximum aphid population was observed on first April with 687.2 specimens. However, this maximum population was shown against the application of Double + Flasher treatment. The minimum aphid population was recorded after the application of HiK Gold + Flasher recommended dose on 15th April. The coccinellids population was at peak level at on 8th April and against the treatment double recommended dose of HiK gold + Flasher. Amount of nitrogen, phosphorus and potassium percentage dry leaves ‎components was maximum (2.33, 0.18 and 2.62 % dry leaves. respectively) in ‎plots treated with recommended double dose mixture of Double + Flasher and ‎Hi-K Gold + Flasher while it was minimum (1.43, 0.12 and 1.77 dry leaves ‎respectively) in plots where no nutrients applied. The result revealed that maximum parasitism was at recommended level of micro and macro nutrients application.‎ Maximum micro nutrients zinc, copper, manganese, iron and boron found with values 46.67 ppm, 21.81 ppm, 62.35 ppm, 152.69 ppm and 36.78 respectively. The result also showed that Over application of macro and micro nutrients should be avoided because it do not help in pest control, conversely it may cause stress on plant. The treatment Double and Flasher recommended dose ratio is almost comparable with recommended dose and present studies confirm its usefulness on wheat.

4 Assessment of Bisphenol A and 17 α-Ethinyl Estradiol Bioavailability in Soils Treated with Biosolids

The application of these biosolids to land without the total reclamation and the treated wastewater can transfer these compounds into terrestrial and aquatic environments, giving rise to potential accumulation in plants. The general aim of this study was to evaluate the bioavailability of bisphenol A (BPA), and 17 α-ethynyl estradiol (EE2) in a soil-biosolid system using wheat (Triticum aestivum) plant assays and a predictive extraction method using a solution of hydroxypropyl-β-cyclodextrin (HPCD) to determine if it is a reliable surrogate for this bioassay. Two soils were obtained from the central region of Chile (Lo Prado and Chicauma). Biosolids were obtained from a regional wastewater treatment plant. The soils were amended with biosolids at 90 Mg ha-1. Soils treated with biosolids, spiked with 10 mgkg-1 of the EE2 and 15 mgkg-1 and 30 mgkg-1of BPA were also included. The BPA, and EE2 concentration were determined in biosolids, soils and plant samples through ultrasound assisted extraction, solid phase extraction (SPE) and gas chromatography coupled to mass spectrometry determination (GC/MS). The bioavailable fraction found of each one of soils cultivated with wheat plants was compared with results obtained through a cyclodextrin biosimulator method. The total concentration found in biosolid from a treatment plant was 0.150 ± 0.064 mgkg-1 and 12.8±2.9 mgkg-1 of EE2 and BPA respectively. BPA and EE2 bioavailability is affected by the organic matter content and the physical and chemical properties of the soil. The bioavailability response of both compounds in the two soils varied with the EE2 and BPA concentration. It was observed in the case of EE2, the bioavailability in wheat plant crops contained higher concentrations in the roots than in the shoots. The concentration of EE2 increased with increasing biosolids rate. On the other hand, for BPA, a higher concentration was found in the shoot than the roots of the plants. The predictive capability the HPCD extraction was assessed using a simple linear correlation test, for both compounds in wheat plants. The correlation coefficients for the EE2 obtained from the HPCD extraction with those obtained from the wheat plants were r= 0.99 and p-value ≤ 0.05. On the other hand, in the case of BPA a correlation was not found. Therefore, the methodology was validated with respect to wheat plants bioassays, only in the EE2 case. 3 Development of Allergenic and Melliferous Floral Pollen Spectrum Using Scanning Electron Microscopy

Abstract:

Morphological features of pollen (sculpturing) were useful for identification of different floral taxa. In this study 49 pollen grains, types belonging to 25 families were studied using Scanning Electron Microscope. Shape and sculpturing of pollen ranging from Psilate, scabrate to reticulate, bireticulate and echinolophate. Honey pollen was identified using morphological features, number and arrangement of pore and colpi, size and shape. It presents the first attempt from Pakistan involving extraction of pollen from honey, its identification and taxonomic analysis. Among pollen studied diversity in shape and sculpturing has been observed ranging from Psilate, scabrate to reticulate to bireticulate and echinolophate condition. Pollen has been identified with the help of morphological feature, number and arrangement of pore and colpi, size and shape, reference slides, light microscopic data and previous literature have been consulted for pollen identification. Pollen of closely related species resemble each other therefore pollen identification of airborne and honey pollen is not possible till species level. Survey of flora was carried in parallel to keep the record about the allergenic and melliferous preference of specific sites through surveys and interviews. Their pollination season and geographical distribution were recorded. Two hundred and five including wild and cultivated taxa were identified belonging to sixty-seven families. Major bee attracting wild shrub and trees includes Justicia adhatoda, Acacia nilotica, Ziziphus jujuba, Taraxicum officinalis, Artemisia dubia, Casuarina sp., Ulmus sp., Broussonetia papyrifera, Cupressus sp. or Pinus roxburghii etc. Cultivated crops like Pennisetum typhoides, Nigella sativa, Triticum sativum along with fruit trees of Pyrus, Prunus, Eryobotria, Citrus etc. are popular melliferous floras. Exotic/ introduced species like Eucalyptus or Parthenium hysterophorus, are also frequently visited by bees indicating the significance of those plants in the honey industry. It is concluded that different microscopic analysis techniques give more clear and authentic pictures of and melliferous pollen identification which is well supported by the floral calendar. The diversity of pollen are observed in case of melliferous pollen, and most of the windborne pollen were found less sculptured or psilate expressing the adaptation to the specific mode of pollination. Pollen morphology and sculpturing would serve as a reference for future studies.

2 Effect of Chemical Fertilizer on Plant Growth-Promoting Rhizobacteria in Wheat

Abstract:

The deleterious effect of chemical fertilizer on rhizobacterial diversity has been well documented using 16S rRNA gene amplicon sequencing and predictive metagenomics. Biofertilization is a cost-effective and sustainable alternative; improving strategies depends on isolating beneficial soil microorganisms. Although culturing is widespread in biofertilization, it is unknown whether the composition of cultured isolates closely mirrors native beneficial rhizobacterial populations. This study aimed to determine the relative abundance of culturable plant growth-promoting rhizobacteria (PGPR) isolates within total soil DNA and how potential PGPR populations respond to chemical fertilization in a commercial wheat variety. It was hypothesized that PGPR will be reduced in fertilized relative to unfertilized wheat. Triticum aestivum cv. Cadenza seeds were sown in a nutrient depleted agricultural soil in pots treated with and without nitrogen-phosphorous-potassium (NPK) fertilizer. Rhizosphere and rhizoplane samples were collected at flowering stage (10 weeks) and analyzed by culture-independent (amplicon sequence variance (ASV) analysis of total rhizobacterial DNA) and -dependent (isolation using growth media) techniques. Rhizosphere- and rhizoplane-derived microbiota culture collections were tested for plant growth-promoting traits using functional bioassays. In general, fertilizer addition decreased the proportion of nutrient-solubilizing bacteria (nitrate, phosphate, potassium, iron and, zinc) isolated from rhizocompartments in wheat, whereas salt tolerant bacteria were not affected. A PGPR database was created from isolate 16S rRNA gene sequences and searched against total soil DNA, revealing that 1.52% of total community ASVs were identified as culturable PGPR isolates. Bioassays identified a higher proportion of PGPR in non-fertilized samples (rhizosphere (49%) and rhizoplane (91%)) compared to fertilized samples (rhizosphere (21%) and rhizoplane (19%)) which constituted approximately 1.95% and 1.25% in non-fertilized and fertilized total community DNA, respectively. The analyses of 16S rRNA genes and deduced functional profiles provide an in-depth understanding of the responses of bacterial communities to fertilizer; this study suggests that rhizobacteria, which potentially benefit plants by mobilizing insoluble nutrients in soil, are reduced by chemical fertilizer addition. This knowledge will benefit the development of more targeted biofertilization strategies.

1 Impact of Elevated Temperature on Spot Blotch Development in Wheat and Induction of Resistance by Plant Growth Promoting Rhizobacteria

Abstract:

Plants are constantly interacting with various abiotic and biotic stresses. In changing climate scenario plants are continuously modifying physiological processes to adapt to changing environmental conditions which profoundly affect plant-pathogen interactions. Spot blotch in wheat is a fast-rising disease in the warmer plains of South Asia where the rise in minimum average temperature over most of the year already affecting wheat production. Hence, the study was undertaken to explore the role of elevated temperature in spot blotch disease development and modulation of antioxidative responses by plant growth promoting rhizobacteria (PGPR) for biocontrol of spot blotch at high temperature. Elevated temperature significantly increases the susceptibility of wheat plants to spot blotch causing pathogen Bipolaris sorokiniana. Two PGPR Bacillus safensis (W10) and Ochrobactrum pseudogrignonense (IP8) isolated from wheat (Triticum aestivum L.) and blady grass (Imperata cylindrical L.) rhizophere respectively, showing in vitro antagonistic activity against Bipolaris sorokiniana were tested for growth promotion and induction of resistance against spot blotch in wheat. GC-MS analysis showed that Bacillus safensis (W10) and Ochrobactrum pseudogrignonense (IP8) produced antifungal and antimicrobial compounds in culture. Seed priming with these two bacteria significantly increase growth, modulate antioxidative signaling and induce resistance and eventually reduce disease incidence in wheat plants at optimum as well as elevated temperature which was further confirmed by indirect immunofluorescence assay using polyclonal antibody raised against Bipolaris sorokiniana. Application of the PGPR led to enhancement in activities of plant defense enzymes- phenylalanine ammonia lyase, peroxidase, chitinase and β-1,3 glucanase in infected leaves. Immunolocalization of chitinase and β-1,3 glucanase in PGPR primed and pathogen inoculated leaf tissue was further confirmed by transmission electron microscopy using PAb of chitinase, β-1,3 glucanase and gold labelled conjugates. Activity of ascorbate-glutathione redox cycle related enzymes such as ascorbate peroxidase, superoxide dismutase and glutathione reductase along with antioxidants such as carotenoids, glutathione and ascorbate and osmolytes like proline and glycine betain accumulation were also increased during disease development in PGPR primed plant in comparison to unprimed plants at high temperature. Real-time PCR analysis revealed enhanced expression of defense genes- chalcone synthase and phenyl alanineammonia lyase. Over expression of heat shock proteins like HSP 70, small HSP 26.3 and heat shock factor HsfA3 in PGPR primed plants effectively protect plants against spot blotch infection at elevated temperature as compared with control plants. Our results revealed dynamic biochemical cross talk between elevated temperature and spot blotch disease development and furthermore highlight PGPR mediated array of antioxidative and molecular alterations responsible for induction of resistance against spot blotch disease at elevated temperature which seems to be associated with up-regulation of defense genes, heat shock proteins and heat shock factors, less ROS production, membrane damage, increased expression of redox enzymes and accumulation of osmolytes and antioxidants.
