1541 Integration of Microarray Data into a Genome-Scale Metabolic Model to Study Flux Distribution after Gene Knockout

Abstract:
Prediction of perturbations after genetic manipulation (especially gene knockout) is one of the important challenges in systems biology. In this paper, a new algorithm is introduced that integrates microarray data into the metabolic model. The algorithm was used to study the change in the cell phenotype after knockout of Gss gene in Escherichia coli BW25113. Algorithm implementation indicated that gene deletion resulted in more activation of the metabolic network. Growth yield was more and less regulating gene were identified for mutant in comparison with the wild-type strain.Keywords: Metabolic network, gene knockout, flux balance analysis, microarray data, integration.

Abstract:
The cycles of the steam-injection gas-turbine systems are studied. The analyses of the parametric effects and the optimal operating conditions for the steam-injection gas-turbine (STIG) system and the regenerative steam-injection gas-turbine (RSTIG) system are investigated to ensure the maximum performance. Using the analytic model, the performance parameters of the system such as thermal efficiency, fuel consumption and specific power, and also the optimal operating conditions are evaluated in terms of pressure ratio, steam injection ratio, ambient temperature and turbine inlet temperature (TIT). It is shown that the computational results are presented to have a notable enhancement of thermal efficiency and specific power.

Abstract:
In this investigation, synchrotron X-ray imaging is used to study water transport inside polymer electrolyte membrane fuel cells. Two measurement techniques are used, namely in-situ radiography and quasi-in-situ tomography combining together in order to reveal the relationship between the structures of the microporous layers (MPLs) and the gas diffusion layers (GDLs), the operation temperature and the water flow. The developed cell is equipped with a thick GDL and a high back pressure MPL. It is found that these modifications strongly influence the overall water transport in the whole adjacent GDM.Keywords: Polymer electrolyte membrane fuel cell, microporous layer, water transport, radiography, tomography.

Abstract:
The method of moments combined with the method of symmetrical components is used for the analysis of interstitial hyperthermia applicators. The basis and testing functions are both piecewise sinusoids, qualifying our technique as a Galerkin one. The dielectric coatings are modeled by equivalent volume polarization currents, which are simply related to the conduction current distribution, avoiding in that way the introduction of additional unknowns or numerical integrations. The results of our method for a four dipole circular array, are in agreement with those already published in literature for a same hyperthermia configuration. Apart from being accurate, our approach is more general, more computationally efficient and takes into account the coupling between the antennas.Keywords: Hyperthermia, integral equations, insulated antennas, method of symmetrical components.

Abstract:
In this article, biomechanical aspects of hen-s eggshell as a natural ceramic structure are studied. The images, taken by a scanning electron microscope (SEM), are used to investigate the microscopic aspects of the egg. It is observed that eggshell has a three-layered microstructure with different morphological and structural characteristics. Studies on the eggshell membrane (ESM) as a prosperous tissue suggest that it is placed to prevent the penetration of microorganisms into the egg. Finally, numerical models of the egg are presented to study the stress distribution and its deformation under different loading conditions. The effects of two different types of loading (hydrostatic and point loadings) on two different shell models (with constant and variable thicknesses) are investigated in detail.

Abstract:
The main parameter of the orifice hole diameter was designed according to the range of throttle diameter ratio which gave the required discharge coefficient. The discharge coefficient is determined by difference diameter ratios. The value of discharge coefficient is 0.958 occurred at throttle diameter ratio 0.5. The throttle hole diameter is 80 mm. The flow analysis is done numerically using ANSYS 17.0, computational fluid dynamics. The flow velocity was analyzed in the upstream and downstream of the orifice meter. The downstream velocity of non-standard orifice meter is 2.5% greater than that of standard orifice meter. The differential pressure is 515.379 Pa in standard orifice.

Abstract:
We introduce an effective approach for automatic offline au- thentication of handwritten samples where the forgeries are skillfully done, i.e., the true and forgery sample appearances are almost alike. Subtle details of temporal information used in online verification are not available offline and are also hard to recover robustly. Thus the spatial dynamic information like the pen-tip pressure characteristics are considered, emphasizing on the extraction of low density pixels. The points result from the ballistic rhythm of a genuine signature which a forgery, however skillful that may be, always lacks. Ten effective features, including these low density points and den- sity ratio, are proposed to make the distinction between a true and a forgery sample. Abstract:
One of the issues that arises with microscale lab-on-a-chip technology is that the laminar flow within the microchannels limits the mixing of fluids. To combat this, micromixers have been introduced as a means to try and incorporate turbulence into the flow to better aid the mixing process. This study presents an electroosmotic micromixer that balances vortex generation and degeneration with the inlet flow velocity to greatly increase the mixing efficiency. A comprehensive parametric study was performed to evaluate the role of the relevant parameters on the mixing efficiency. It was observed that the suggested micromixer is perfectly suited for biological applications due to its low pressure drop (below 10 Pa) and low shear rate. The proposed micromixer with optimized working parameters is able to attain a mixing efficiency of 95% in a span of 0.5 seconds using a frequency of 10 Hz, a voltage of 0.7 V, and an inlet velocity of 0.366 mm/s.

Abstract:
Biomass is renewable and sustainable. As an energy source, it will not release extra carbon dioxide into the atmosphere. Hence, tremendous efforts have been made to develop technologies capable of transforming biomass into suitable forms of bio-fuel. One of the viable technologies is gasifying biomass in supercritical water (SCW), a green medium for reactions. While previous studies overwhelmingly selected glucose as a model compound for biomass, the present study adopted fructose for the sake of comparison. The gasification of fructose in SCW was investigated experimentally to evaluate the applicability of supercritical water processes to biomass gasification. Experiments were conducted with an autoclave reactor. Gaseous product mainly consists of H2, CO, CO2, CH4 and C2H6. The effect of two major operating parameters, the reaction temperature (673-873 K) and the dosage of oxidizing agent (0-0.5 stoichiometric oxygen), on the product gas composition, yield and heating value was also examined, with the reaction pressure fixed at 25 MPa.

Abstract:
The sand production problem has led researchers into making various attempts to understand the phenomenon. The generally accepted concept is that the occurrence of sanding is due to the in-situ stress conditions and the induced changes in stress that results in the failure of the reservoir sandstone during hydrocarbon production from wellbores. By using a hypothetical cased (perforated) well, an approach to the problem is presented here by using Finite Element numerical modelling techniques. In addition to the examination of the erosion problem, the influence of certain key parameters is studied in order to ascertain their effect on the failure and subsequent erosion process. The major variables investigated include: drawdown, perforation depth, and the erosion criterion. Also included is the determination of the optimal mud pressure for given operational and reservoir conditions. The improved understanding between parameters enables the choice of optimal values to minimize sanding during oil production.

Abstract:
This paper describes in details a commercial green building that has been designed and constructed in Marsa Matrouh, Egypt. The balance between homebuilding and the sustainable environment has been taken into consideration in the design and construction of this building. The building consists of one floor with 3 m height and 2810 m2 area while the envelope area is 1400 m2. The building construction fulfills the natural ventilation requirements. The glass curtain walls are about 50% of the building and the windows area is 300 m2. 6 mm greenish gray tinted temper glass as outer board lite, 6 mm safety glass as inner board lite and 16 mm thick dehydrated air spaces are used in the building. Visible light with 50% transmission, 0.26 solar factor, 0.67 shading coefficient and 1.3 W/m2.K thermal insulation U-value are implemented to realize the performance requirements. Optimum electrical distribution for lighting system, air conditions and other electrical loads has been carried out. Power and quantity of each type of the lighting system lamps and the energy consumption of the lighting system are investigated. The design of the air conditions system is based on summer and winter outdoor conditions. Ventilated, air conditioned spaces and fresh air rates are determined. Variable Refrigerant Flow (VRF) is the air conditioning system used in this building. The VRF outdoor units are located on the roof of the building and connected to indoor units through refrigerant piping. Indoor units are distributed in all building zones through ducts and air outlets to ensure efficient air distribution. The green building energy consumption is evaluated monthly all over one year and compared with the consumed energy in the non-green conditions using the Hourly Analysis Program (HAP) model. The comparison results show that the total energy consumed per year in the green building is about 1,103,221 kWh while the non-green energy consumption is about 1,692,057 kWh. In other words, the green building total annual energy cost is reduced from 136,581 $ to 89,051 $. This means that, the energy saving and consequently the money-saving of this green construction is about 35%. In addition, 13 points are awarded by applying one of the most popular worldwide green energy certification programs (Leadership in Energy and Environmental Design “LEED”) as a rating system for the green construction. It is concluded that this green building ensures sustainability, saves energy and offers an optimum energy performance with minimum cost.

Traditional mechanical control systems in thrust vectoring are efficient in rocket thrust guidance but their costs and their weights are excessive. The fluidic injection in the nozzle divergent constitutes an alternative procedure to achieve the goal. In this paper, we present a 3D analytical model for fluidic injection in a supersonic nozzle integrating an orifice. The fluidic vectoring uses a sonic secondary injection in the divergent. As a result, the flow and interaction between the main and secondary jet has built in order to express the pressure fields from which the forces and thrust vectoring are deduced. A novel method is presented for obtaining the stress field induced by an edge dislocation in a multilayered composite. To demonstrate the applications of the obtained solution, we consider the problem of an interfacial crack in a periodically layered bimaterial medium. The crack is modelled as a continuous distribution of edge dislocations and the Distributed Dislocation Technique (DDT) is utilized to obtain numerical results for the energy release rate (ERR). Abstract:
ANSYS Fluent will be used to simulate Computational Fluid Dynamics (CFD) for an efficient lens and nozzle design which will be explained in this paper. We have designed and characterized an aerodynamic lens and a divergent nozzle for focusing flow that transmits sub 25 nm particles through the aerodynamic lens. The design of the lens and nozzle has been improved using CFD for particle trajectories. We obtained a case for calculating nanoparticles (25 nm) flowing through the aerodynamic lens and divergent nozzle. Nanoparticles are transported by air, which is pumped into the aerodynamic lens through the nozzle at 1 atmospheric pressure. We have also developed a computational methodology that can determine the exact focus characteristics of aerodynamic lens systems. Particle trajectories were traced using the Lagrange approach. The simulation shows the ability of the aerodynamic lens to focus on 25 nm particles after using a divergent nozzle.

The paper deals with the classical fiber bundle model of equal load sharing, sometimes referred to as the Daniels’ bundle or the democratic bundle. Daniels formulated a multidimensional integral and also a recursive formula for evaluation of the strength cumulative distribution function. This paper describes three algorithms for evaluation of the recursive formula and also their implementations with source codes in the Python high-level programming language. A comparison of the algorithms are provided with respect to execution time. This research involved the use of word distributions and morphological knowledge by speakers of Arabic learning English connected different allomorphs in order to realize how the morphology and syntax of English gives meaning through using interactive crossword puzzles (ICP). Fifteen chapters covered with a class of nine learners over an academic year of an intensive English program were reviewed using the ICP. Learners were questioned about how the use of this gaming element enhanced and motivated their learning of English. The findings were positive indicating a successful implementation of ICP both at creational and user levels. Electrospinning is a broadly used technology to obtain polymeric nanofibers ranging from several micrometers down to several hundred nanometers for a wide range of applications. It offers unique capabilities to produce nanofibers with controllable porous structure. With smaller pores and higher surface area than regular fibers, electrospun fibers have been successfully applied in various fields, such as, nanocatalysis, tissue engineering scaffolds, protective clothing, filtration, biomedical, pharmaceutical, optical electronics, healthcare, biotechnology, defense and security, and environmental engineering. In this study, polyurethane nanofibers were obtained under different electrospinning parameters. Abstract:
A high thermal dissipation performance polyethylene terephthalate heat pipe has been fabricated and tested in this research. Polyethylene terephthalate (PET) is used as the container material instead of copper. Copper mesh and methanol are sealed in the middle of two PET films as the wick structure and working fluid. Although the thermal conductivity of PET (0.15-0.24 W/m·K) is much smaller than copper (401 W/m·K), the experiment results reveal that the PET heat pipe can reach a minimum thermal resistance of 0.146 (oC/W) and maximum effective thermal conductivity of 18,310 (W/m·K) with 36.9 vol% at 26 W input power. However, when the input power is larger than 30 W, the laminated PET will debond due to the high vapor pressure of methanol.

A steam power plant is housed with various types of equipments like boiler, turbine, heat exchanger etc. These equipments are mainly connected with piping systems. Such a piping layout design depends mainly on stress analysis and flexibility. It will vary with respect to pipe geometrical properties, pressure, temperature, and supports. The present paper is to analyze the presence and effect of hangers and expansion joints in the piping layout/routing using CAESAR-II software. Main aim of piping stress analysis is to provide adequate flexibility for absorbing thermal expansion, code compliance for stresses and displacement incurred in piping system. The design is said to be safe if all these are in allowable range as per code. In this study, a sample problem is considered for analysis as per power piping ASME B31.1 code and the results thus obtained are compared.

The performance of schedules released to a shop floor may greatly be affected by unexpected disruptions. Thus, this paper considers the flexible job shop scheduling problem when processing times of some operations are represented by a uniform distribution with given lower and upper bounds. The objective is to find a predictive schedule that can deal with this uncertainty. The paper compares two genetic approaches to obtain predictive schedule. To determine the performance of the predictive schedules obtained by both approaches, an experimental study is conducted on a number of benchmark problems.

Experimental investigation of the effect of hydrophobic injection on siloxane basis on the properties of oldfashioned type of ceramic brick is presented in the paper. At the experimental testing, the matrix density, total open porosity, pore size distribution, sorptivity, water absorption coefficient, sorption and desorption isotherms are measured for the original, as well as the hydrophobic-injection treated brick. The purpose of this study is to review representative cases of green space development in order to compare the Garden City concept and Green Belt concept as applied and to examine its direction in major Asian and Oceanic cities. The results of previous studies and this study show that there are two major directions in such green-oriented city planning. One direction is toward Multi-Regional Development, and the other focuses on an Environmentally Symbiotic City based on the Garden City concept. In this paper multivariable predictive PID controller has been implemented on a multi-inputs multi-outputs control problem i.e., quadruple tank system, in comparison with a simple multiloop PI controller. One of the salient feature of this system is an adjustable transmission zero which can be adjust to operate in both minimum and non-minimum phase configuration, through the flow distribution to upper and lower tanks in quadruple tank system. Stability and performance analysis has also been carried out for this highly interactive two input two output system, both in minimum and non-minimum phases. The objective of the manufactures in their supply network is to reduce inventory costs and increase customer satisfaction levels. One way of doing that is to synchronize delivery performance. A supply network can be described by nodes representing the companies and the links (relationships) between these nodes. Uncertainty in delivery time depends on type of network relationship between suppliers. Abstract:
The Želazny Most tailing pond is one of the largest facilities worldwide for waste disposal from the copper mines located in South-West Poland. A potential failure of the dam would allow more than 10 million cubic meters of contaminated slurry to flow to the valley, causing immense environmental problems to the surrounding area. Thus, the determination of the strength properties of the dam's soils and their variability is of utmost importance. An extensive site investigation consisting of more than 480 cone penetration tests (CPTs) with or without pore water pressure measurements were conducted within a period of 13 years to study the mechanical properties of the tailings body. Abstract:
Liquid spray heat exchanger is the critical equipment of temperature regulating system by gaseous nitrogen which realizes the environment temperature in the range of -180 ℃~+180 ℃. Liquid nitrogen is atomized into smaller liquid drops through liquid nitrogen sprayer and then contacts with gaseous nitrogen to be cooled. By adjusting the pressure of liquid nitrogen and gaseous nitrogen, the flowrate of liquid nitrogen is changed to realize the required outlet temperature of heat exchanger. The temperature accuracy of shrouds is ±1 ℃. Liquid nitrogen spray heat exchanger is simulated by CATIA, and the numerical simulation is performed by FLUENT. The comparison between the tests and numerical simulation is conducted. Abstract:
A vacuum fractionation technique was introduced to remove ethanol from fermentation broth. The effect of initial glucose and ethanol concentrations were investigated for specific productivity. The inhibitory ethanol concentration was observed at 100 g/L. In order to increase the fermentation performance, the ethanol product was removed as soon as it is produced. The broth was boiled at 35oC by reducing the pressure to 65 mBar. The ethanol/water vapor was fractionated for up to 90 wt% before leaving the column. Ethanol concentration in the broth was kept lower than 25 g/L, thus minimized the product inhibition effect to the yeast cells. For batch extractive fermentation, a high substrate utilization rate was obtained at 26.6 g/L.h and most of glucose was consumed within 21 h. For repeated-batch extractive fermentation, addition of glucose was carried out up to 9 times and ethanol was produced more than 8-fold higher than batch fermentation.

Abstract:
In this paper, we study the performance of the strong stability method of the univariate classical risk model. We interest to the stability bounds established using two approaches. The first based on the strong stability method developed for a general Markov chains. The second approach based on the regenerative processes theory . By adopting an algorithmic procedure, we study the performance of the stability method in the case of exponential distribution claim amounts. Abstract:
This paper presents the experimental results of comparison between leakage currents and discharge currents. The leakage currents were obtained on polluted porcelain insulator. Whereas, the discharge currents were obtained on lightly artificial polluted porcelain specimen. The conducted measurements were leakage current or discharge current and applied voltage. The insulator or specimen was in a hermetically sealed chamber, and the current waveforms were analyzed using FFT. The result indicated that the leakage current (LC) on low RH condition the fifth harmonic would be visible, and followed by the seventh harmonic. The insulator had capacitive property. Otherwise, on 99% relative humidity, the fifth harmonic would also be visible, and the phase angle reached up to 12.2 degree. Whereas, on discharge current, the third harmonic would be visible, and followed by fifth harmonic. The third harmonic would increase as pressure reduced. Abstract:
This study examines the stack ventilation performance of an office building located in Taipei, Taiwan. Atriums in this building act as stacks that facilitate buoyancy-driven ventilation. Computational Fluid Dynamic (CFD) simulations are used to identify interior airflow patterns, and then used these patterns to assess the building’s heat expulsion efficiency. Ambient temperatures of 20°C were adopted as the typical seasonal spring temperature range in Taipei. Further, “zero-wind” conditions are established to ensure simulation results reflected only the buoyancy effect. After checking results against neutral pressure level (NPL) level, airflow, air velocity, and indoor temperature stratification, the lower stack is modified to reduce the NPL in order to remove heat accumulated on the top floor.
