Authors: Anna Vildová, H. Hendrychová, J. Kubeš, L. Tůmová
Abstract:
The abiotic elicitation is one of the methods for increasing the secondary metabolites production in plant tissue cultures and it seems to be more effective than traditional strategies. This study verified the use of silver nitrate as elicitor to enhance flavonolignans and flavonoid taxifolin production in suspension culture of Sylibum marianum (L.) Gaertn. Silver nitrate in various concentrations (5.887.10-3 mol/L, 5.887.10-4 mol/L, 5.887.10-5 mol/L) was used as elicitor. The content of secondary metabolites in cell suspension cultures was determined by high performance liquid chromatography. The samples were taken after 6, 12, 24, 48, 72 and 168 hours of treatment. The highest content of taxifolin production (2.2 mg.g-1) in cell suspension culture of Silybum marianum (L.) Gaertn. was detected after silver nitrate (5.887.10-4 mol/L) treatment and 72 h application. Flavonolignans such as silybinA, silybin B, silydianin, silychristin, isosilybin A, isosilybin B were not produced by cell suspension culture of S. marianum after elicitor treatment. Our results show that the secondarymetabolites could be released from S. marianum cells into the nutrient medium by changed permeability of cell wall. Keywords: Silybum marianum (L.) Authors: Shan-Jen Cheng, Jr-Ming Miao, Sheng-Ju Wu
Abstract:
The purpose of this paper is applied Taguchi method on the optimization for PEMFC performance, and a representative Computational Fluid Dynamics (CFD) model is selectively performed for statistical analysis. The studied factors in this paper are pressure of fuel cell, operating temperature, the relative humidity of anode and cathode, porosity of gas diffusion electrode (GDE) and conductivity of GDE. The optimal combination for maximum power density is gained by using a three-level statistical method. The results confirmed that the robustness of the optimum design parameters influencing the performance of fuel cell are founded by pressure of fuel cell, 3atm; operating temperature, 353K; the relative humidity of anode, 50%; conductivity of GDE, 1000 S/m, but the relative humidity of cathode and porosity of GDE are pooled as error due to a small sum of squares. The present simulation results give designers the ideas ratify the effectiveness of the proposed robust design methodology for the performance of fuel cell.

Abstract:
Chitosan is a biopolymer composed of glucosamine and N-acetyl glucosamine. Solubility and viscosity pose problems in some applications. These problems can be overcome with unique modifications. In this study, firstly, chitosan was modified by caffeic acid and thioglycolic acid, separately. Then, growing effects of these modified polymers was observed in U937 cell line. Caffeic acid is a phenolic compound and its modifications act carcinogenic inhibitors in drugs. Thiolated chitosans are commonly being used for drugdelivery systems in various routes, because of enhancing mucoadhesiveness property. U937 cell line was used model cell for leukaemia. Modifications were achieved by 1 – 15 % binding range. Increasing binding ratios showed higher radical-scavenging activity and reducing cell growth, in compared to native chitosan. Caffeic acid modifications showed higher radical-scavenging activity than thiolated chitosans at the same concentrations. Caffeic acid and thioglycolic acid modifications inhibited growth of U937, effectively. Keywords: Chitosan, U937 cell, caffeic acid, thioglycolic acid

Abstract:
The counting process of cell colonies is always a long and laborious process that is dependent on the judgment and ability of the operator. The judgment of the operator in counting can vary in relation to fatigue. Moreover, since this activity is time consuming it can limit the usable number of dishes for each experiment. For these purposes, it is necessary that an automatic system of cell colony counting is used. This article introduces a new automatic system of counting based on the elaboration of the digital images of cellular colonies grown on petri dishes. This system is mainly based on the algorithms of region-growing for the recognition of the regions of interest (ROI) in the image and a Sanger neural net for the characterization of such regions. The better final classification is supplied from a Feed-Forward Neural Net (FF-NN) and confronted with the K-Nearest Neighbour (K-NN) and a Linear Discriminative Function (LDF). The preliminary results are shown. Keywords: Automatic cell counting, neural network, region growing, Sanger net.

Abstract:
The underground shopping mall has the constructional problem of the fire evacuation. Also, the people sometimes lose their direction and information of current time in the mall. If the emergencies such as terrorist explosions or gas explosions are happened, they have to go out soon. Under such circumstances, inside of the mall has high risk for life. In this research, the authors propose a way that he/she can go out from the underground shopping mall quickly. If the narrow exits are discovered by using active RFID (Radio Frequency Identification) tags and using cellular phones, they can evacuate as soon as possible. To verify this hypothesis, the authors design the model and carry out the agent-based simulation. They treat, as a case study, the Tenjin mall in Fukuoka Prefecture in Japan. The result of the simulation is that the case of the pedestrian with using active RFID tags and cellular phones reduced the amount of time to spend on the evacuation. Even if the diffusion of RFID tags and cellular phones was not perfect, they could show the effectiveness of reducing the time of evacuation. Keywords: Evacuation, active RFID tag and cellular phone, underground shopping mall, agent-based simulation.

Authors: S. Limpattayanate, M. Hunsom
Abstract:
A comparison of activity and stability of the as-formed Pt/C, Pt-Co and Pt-Pd/C electrocatalysts, prepared by a combined approach of impregnation and seeding, was performed. According to the activity test in a single Proton Exchange Membrane (PEM) fuel cell, the Oxygen Reduction Reaction (ORR) activity of the Pt-M/C electrocatalyst was slightly lower than that of Pt/C. The j0.9 V and E10 mA/cm2 of the as-prepared electrocatalysts increased in the order of Pt/C > Pt-Co/C > Pt-Pd/C. However, in the medium-to-high current density region, Pt-Pd/C exhibited the best performance. With regard to their stability in a 0.5 M H2SO4 electrolyte solution, the electrochemical surface area decreased as the number of rounds of repetitive potential cycling increased due to the dissolution of the metals within the catalyst structure. For long-term measurement, Pt- Pd/C was the most stable than the other three electrocatalysts. Keywords: ORR activity, Stability, Pt-based electrocatalysts, PEM fuel cell.

Authors: Tamer A. Tabet, Fauziah Abdul Aziz, Shahidan Radiman
Abstract:
The spiral angle of the elementary cellulose fibril in the wood cell wall, often called microfibril angle, (MFA). Microfibril angle in hardwood is one of the key determinants of solid timber performance due to its strong influence on the stiffness, strength, shrinkage, swelling, thermal-dynamics mechanical properties and dimensional stability of wood. Variation of MFA (degree) in the S2 layer of the cell walls among Acacia mangium trees was determined using small-angle X-ray scattering (SAXS). The length and orientation of the microfibrils of the cell walls in the irradiated volume of the thin samples are measured using SAXS and optical microscope for 3D surface measurement. The undetermined parameters in the analysis are the MFA, (M) and the standard deviation (σФ) of the intensity distribution arising from the wandering of the fibril orientation about the mean value. Nine separate pairs of values are determined for nine different values of the angle of the incidence of the X-ray beam relative to the normal to the radial direction in the sample. The results show good agreement. The curve distribution of scattered intensity for the real cell wall structure is compared with that calculated with that assembly of rectangular cells with the same ratio of transverse to radial cell wall length. It is demonstrated that for β = 45°, the peaks in the curve intensity distribution for the real and the rectangular cells coincide. If this peak position is Ф45, then the MFA can be determined from the relation M = tan-1 (tan Ф45 / cos 45°), which is precise for rectangular cells. It was found that 92.93% of the variation of MFA can be attributed to the distance from pith to bark. Here we shall present our results of the MFA in the cell wall with respect to its shape, structure and the distance from pith to park as an important fast check and yet accurate towards the quality of wood, its uses and application. Keywords: Small-Angle X-Ray Scattering, Microfibril Angle, MFA, rectangular cell wall and real cell wall, Acacia mangium.

Abstract:
Abnormal adipocyte growth, in terms of increased cell numbers and increased cell differentiation, is considered to be a major pathological feature of obesity. Thus, the inhibition of preadipocyte mitogenesis and differentiation could help prevent and suppress obesity. The aim of this study was to assess whether extracts from Weissella koreensis 521 cells isolated from kimchi could exert anti-adipogenic effects in 3T3-L1 cells (fat cells). Differentiating 3T3-L1 cells were treated with W. koreensis 521 cell extracts (W. koreensis 521_CE), and cell viability was assessed by MTT assays. At concentrations below 0.2 mg/ml, W. koreensis 521_CE did not exert any cytotoxic effect in 3T3-L1 cells. However, treatment with W. koreensis 521_CE significantly inhibited adipocyte differentiation, as assessed by morphological analysis and Oil Red O staining of fat. W. koreensis 521_CE treatment (0.2 mg/ml) also reduced lipid accumulation by 24% in fully differentiated 3T3-L1 adipocytes. These findings collectively indicate that Weissella koreensis 521 may help prevent obesity.

Abstract:
Nowadays, e-commerce shopping websites have experienced noticeable growth. These websites have gained consumers’ trust. After purchasing a product, many consumers share comments where opinions are usually embedded about the given product. Research on the automatic management of opinions that gives suggestions to potential consumers and portrays an image of the product to manufactures has been growing recently. After launching the product in the market, the reviews generated around it do not usually contain helpful information or generic opinions about this product (e.g. telephone: great phone...); in the sense that the product is still in the launching phase in the market. Within time, the product becomes old. Therefore, consumers perceive the advantages/ disadvantages about each specific product feature. Therefore, they will generate comments that contain their sentiments about these features. In this paper, we present an unsupervised method to extract different product features hidden in the opinions which influence its purchase, and that combines Time Weighting (TW) which depends on the time opinions were expressed with Term Frequency-Inverse Document Frequency (TF-IDF). We conduct several experiments using two different datasets about cell phones and hotels. The results show the effectiveness of our automatic feature extraction, as well as its domain independent characteristic.

Complex assemblies of interacting proteins carry out most of the interesting jobs in a cell, such as metabolism, DNA synthesis, mitosis and cell division. These physiological properties play out as a subtle molecular dance, choreographed by underlying regulatory networks that control the activities of cyclin-dependent kinases (CDK). The network can be modeled by a set of nonlinear differential equations and its behavior predicted by numerical simulation. In this paper, an innovative approach has been proposed that uses genetic algorithms to mine a set of behavior data output by a biological system in order to determine the kinetic parameters of the system. In our approach, the machine learning method is integrated with the framework of existent biological information in a wiring diagram so that its findings are expressed in a form of system dynamic behavior. By numerical simulations it has been illustrated that the model is consistent with experiments and successfully shown that such application of genetic algorithms will highly improve the performance of mathematical model of the cell division cycle to simulate such a complicated bio-system.Keywords: Cell cycle, Cyclin-dependent kinase, Fission yeast, Genetic algorithms, Mathematical modeling, Wiring diagram

Abstract:
Migration in breast cancer cell wound healing assay had been studied using image fractal dimension analysis. The migration of MDA-MB-231 cells (highly motile) in a wound healing assay was captured using time-lapse phase contrast video microscopy and compared to MDA-MB-468 cell migration (moderately motile). The Higuchi fractal method was used to compute the fractal dimension of the image intensity fluctuation along a single pixel width region parallel to the wound. The near-wound region fractal dimension was found to decrease three times faster in the MDA-MB- 231 cells initially as compared to the less cancerous MDA-MB-468 cells. The inner region fractal dimension was found to be fairly constant for both cell types in time and suggests a wound influence range of about 15 cell layer. The box-counting fractal dimension method was also used to study region of interest (ROI). The MDAMB- 468 ROI area fractal dimension was found to decrease continuously up to 7 hours. The MDA-MB-231 ROI area fractal dimension was found to increase and is consistent with the behavior of a HGF-treated MDA-MB-231 wound healing assay posted in the public domain. A fractal dimension based capacity index has been formulated to quantify the invasiveness of the MDA-MB-231 cells in the perpendicular-to-wound direction. Our results suggest that image intensity fluctuation fractal dimension analysis can be used as a tool to quantify cell migration in terms of cancer severity and treatment responses. Keywords: Higuchi fractal dimension, box-counting fractal dimension, cancer cell migration, wound healing.
