180 Immobilization of Aspergillus awamori 1-8 for Subsequent Pectinase Production

Authors: Zh. B. Suleimenova, Zh. K. Rakhmetova, R. K. Blieva, A. E. Nurlybayeva

Abstract:

The overall objective of this research is a strain improvement technology for efficient pectinase production. A novel cells cultivation technology by immobilization of fungal cells has been studied in long time continuous fermentations. Immobilization was achieved by using of new material for absorption of stores of immobilized cultures which was for the first time used for immobilization of microorganisms. Effects of various conditions of nitrogen and carbon nutrition on the biosynthesis of pectolytic enzymes in Aspergillus awamori 1-8 strain were studied. Proposed cultivation technology along with optimization of media components for pectinase overproduction led to increased pectinase productivity in Aspergillus awamori 1-8 from 7 to 8 times. Proposed technology can be applied successfully for production of major industrial enzymes such as α-amylase, protease, collagenase etc.

Keywords: Aspergillus awamori, immobilization, pectolytic enzymes.

179 The Influence of Thermic Plastic Films on Vegetative and Reproductive Growth of Iceberg Lettuce ‘Dublin’

Authors: Wael M. Semida, P. Hadley, W. Sobeih, N. A. El-Sawah, M. A. S. Barakat

Abstract:

Photoselective plastic films with thermic properties are now available so that greenhouses clad with such plastics exhibit a higher degree of “Greenhouse Effect” with a consequent increase in night time temperature. In this study, we investigate the potential benefits of a range of thermic plastic films used as greenhouse cover materials on the vegetative and reproductive growth and development of Iceberg lettuce (Lactuca sativa L). Transplants were grown under thermic films and destructively harvested 4, 5, and 6 weeks after transplanting. Thermic films can increase night temperatures up to 2 ⁰C reducing the wide fluctuation in greenhouse temperature during winter compared to the standard commercial film and consequently increased the yield (leaf number, fresh weight, and dry weight) of lettuce plants. Lettuce plants grown under Clear film respond to cold stress by the accumulation of secondary products (phenolics, and flavonoids).

178 Matching-Based Cercospora Leaf Spot Detection in Sugar Beet

Authors: Rong Zhou, Shun’ich Kaneko, Fumio Tanaka, Miyuki Kayamori, Motoshige Shimizu

Abstract:

In this paper, we propose a robust disease detection method, called adaptive orientation code matching (Adaptive OCM), which is developed from a robust image registration algorithm: orientation code matching (OCM), to achieve continuous and site-specific detection of changes in plant disease. We use two-stage framework for realizing our research purpose; in the first stage, adaptive OCM was employed which could not only realize the continuous and site-specific observation of disease development, but also shows its excellent robustness for non-rigid plant object searching in scene illumination, translation, small rotation and occlusion changes and then in the second stage, a machine learning method of support vector machine (SVM) based on a feature of two dimensional (2D) xy-color histogram is further utilized for pixel-wise disease classification and quantification. The indoor experiment results demonstrate the feasibility and potential of our proposed algorithm, which could be implemented in real field situation for better observation of plant disease development.

177 Estimation of the Moisture Diffusivity and Activation Energy in Thin Layer Drying of Ginger Slices

Authors: Ebru Kavak Akpinar, Seda Toraman

Abstract:

In the present work, the effective moisture diffusivity and activation energy were calculated using an infinite series solution of Fick-s diffusion equation. The results showed that increasing drying temperature accelerated the drying process. All drying experiments had only falling rate period. The average effective moisture diffusivity values varied from 2.807x10-10 to 6.977x10-10m2 s_1 over the temperature and velocity range. The temperature dependence of the effective moisture diffusivity for the thin layer drying of the ginger slices was satisfactorily described by an Arrhenius-type relationship with activation energy values of 19.313- 22.722 kJ.mol-1 within 40–70 °C and 0.8-3 ms-1 temperature range.

176 Baking Quality of Hulled Wheat Species in Organic Farming

Authors: P. Konvalina, I. Capouchová, Z. Stehno

Abstract:

The organic farmers use wider range of crop varieties than the conventional farming. Bread wheat is the most favorite and the most common food crop. The organic bread wheat is usually of worse technological quality. Therefore, it is supposed to be an attractive alternative to the hulled wheat species (einkorn, emmer wheat and spelt). Twenty-five hulled bread wheat varieties and control bread wheat ones were grown on the certified organic parcel in České Budějovice (the Czech Republic) between 2009 and 2012. Their baking quality was measured and evaluated with standard methods, and in accordance with ICC. The results have shown that the grain of hulled wheat varieties contain a lot of proteins in grains (up to 18 percent); even the organic hulled bread wheat varieties are characterized by such good baking quality. Einkorn and emmer wheat are of worse technological quality of proteins (low values of gluten index and Zeleny test), which is a disadvantage of these two wheat species. On the other hand, spelt wheat is of better technological quality and is similar to the control bread wheat varieties. Mixtures consisting of bread wheat, among others, are considered good alternatives; they may contribute to wider range of use of the hulled wheat species. It is one of the possibilities which may increase the proportion of proteins in bread wheat grains; the nutrition-rich hulled wheat grains may be also used in such way at the same time.

Authors: N. N. A. K. Shah, R. A. Rahman, R. Shamsuddin, N. M. Adzahan

Abstract:

Pummelo is known to be the largest of all citrus fruits, with expected ratio of 2:1 (w/v) of producing juice, is an attractive opportunity for Malaysia to expand pummelo-s influence and marketability over the international market of juices. The purpose of this study is to identify and quantify the phenolic compounds in two Malaysian varieties of pummelo fruit juice: Ledang (PO55) and Tambun (PO52). Identifications of polyphenols composition were done using High Performance Liquid Chromatography Diode Array Detection (HPLC-DAD). The phenolic compounds that were found in both varieties were hydroxycinnamic acids and flavonones. This study proved that Tambun variety has the highest antioxidant and phenolic compounds in comparison to Ledang variety. However, considerations have to be made to suit consumer-s taste bud and the amount of enzyme needed to clarify the juice for its marketability.

173 Experimental Investigation of Drying Behavior of Rosehip in a Cyclone-Type Dryer

Authors: Ayse Bicer, Filiz Kar

Abstract:

This paper describes an experimental investigation of the drying behavior and conditions of rosehip in a convective cyclone-type dryer. Drying experiments were conducted at air inlet temperatures of 50, 60 and 70 o C and air velocities of 0.5, 1 and 1.5 ms–1. The parametric values obtained from the experiments were fitted to the Newton mathematical models. Consequently, the drying model developed by Newton model showed good agreement with the data obtained from the experiments. Concluding, it was obtained that; (i) the temperature is the major effect on the drying process, (ii) air velocity has low effect on the drying of rosehip, (iii) the C-vitamin is observed to change according to the temperature, moisture, drying time and flow types. The changing ratio is found to be in the range of 0.70-0.74.

171 The Impacts of Food Safety Standards on China Export of Vegetables and Fruits

Authors: Lei Dou, Mitsuhiro Nakagawa, Fei Yan, Ping Li

Abstract:

Participation in global trade means that Chinas vegetables and fruits industry faces international food safety standards and increased scrutiny worldwide. The objectives of this paper were to investigate how existing food safety standards and regulations in the importing countries impact the export of vegetables and fruits from China. This paper discussed the current and historical situations of Chinas vegetables and fruits export from 1996 to 2010, analyzed the Maximum Residual Limit (MRL) standards of pesticides imposed by importing countries, quantitatively estimated the impacts of food safety standards on Chinas vegetables and fruits export based on a gravity model. The results showed that although transportation distance between trade partners and tariff rates on vegetables and fruits were still the importantly resistant factors for China export, vegetables and fruits export was sensitive to the number of regulated pesticides, the strictness, and the level of food safety standards imposed by importing countries, which showed a significant trade flow effect, stricter food safety standards, increased number of regulated pesticides significantly inhibit China export of vegetables and fruits. Moreover, Chinas food safety standards also showed a significantly effect on vegetables and fruits export, which inhibited export to some extent. 170 Ripper and Sugar Effects on Hydroxymethylfurfural Formation in Gingerbread Biscuits

Authors: A. Komarovska, V. Kreicbergs, F. Diminš

Abstract:

Hydroxymethylfurfural (HMF) is formed by thermally treating products rich in carbohydrates. HMF and other furan derivatives are toxic. The aim of the research was to establish the content of HMF in gingerbread biscuits with honey and sugar syrup additives by using three leavening agents— ammonium carbonate (NH4HCO3 and (NH4)2CO3), baking powder, and baking soda (NaHCO3). The content of HMF is significantly affected by the leavening agent used. The content of HMF with honey where ammonium carbonate was used as additive range from 5.7 to 27.3mg 100g-1, but when sugar syrup is used the content varies from 2.3 to 7.4mg 100g-1. When baking powder and baking soda are used as leavening agents, the content of HMF exceeds 4mg 100g-1 in the event honey had been added and the baking time had been longer (10 minutes at 180°C or 9 minutes at 200°C).

169 Managing Meat Safety at South African Abattoirs

Authors: R. Govender, D. Naidoo, E. M. Buys

Abstract:

The importance of ensuring safe meat handling and processing practices has been demonstrated in global reports on food safety scares and related illness and deaths. This necessitated stricter meat safety control strategies. Today, many countries have regulated towards preventative and systematic control over safe meat processing at abattoirs utilizing the Hazard Analysis Critical Control Point (HACCP) principles. HACCP systems have been reported as effective in managing food safety risks, if correctly implemented. South Africa has regulated the Hygiene Management System (HMS) based on HACCP principles applicable to abattoirs. Regulators utilise the Hygiene Assessment System (HAS) to audit compliance at abattoirs. These systems were benchmarked from the United Kingdom (UK). Little research has been done them since inception as of 2004. This paper presents a review of the two systems, its implementation and comparison with HACCP. Recommendations are made for future research to demonstrate the utility of the HMS and HAS in assuring safe meat to consumers.

168 Production of WGHs and AFPHs using Protease Combinations at High and Ambient Pressure

Authors: Namsoo Kim, So-Hee Son, Jin-Soo Maeng, Yong-Jin Cho, Chul-Jin Kim, Chong-Tai Kim

Abstract:

Wheat gluten hydrolyzates (WGHs) and anchovy fine powder hydrolyzates (AFPHs) were produced at 300 MPa using combinations of Flavourzyme 500MG (F), Alcalase 2.4L (A), Marugoto E (M) and Protamex (P), and then were compared to those produced at ambient pressure concerning the contents of soluble solid (SS), soluble nitrogen and electrophoretic profiles. The contents of SS in the WGHs and AFPHs increased up to 87.2% according to the increase in enzyme number both at high and ambient pressure. Based on SS content, the optimum enzyme combinations for one-, two-, three- and four-enzyme hydrolysis were determined as F, FA, FAM and FAMP, respectively. Similar trends were found for the contents of total soluble nitrogen (TSN) and TCA-soluble nitrogen (TCASN). The contents of SS, TSN and TCASN in the hydrolyzates together with electrophoretic mobility maps indicates that the high-pressure treatment of this study accelerated protein hydrolysis compared to ambient-pressure treatment.

167 Effect of Process Parameters on the Proximate Composition, Functional and Sensory Properties

Authors: C. I. Omohimi, O. P. Sobukola, K. O. Sarafadeen, L.O. Sanni

Abstract:

Flour from Mucuna beans (Mucuna pruriens) were used in producing texturized meat analogue using a single screw extruder to monitor modifications on the proximate composition and the functional properties at high moisture level. Response surface methodology based on Box Behnken design at three levels of barrel temperature (110, 120, 130°C), screw speed (100,120,140rpm) and feed moisture (44, 47, 50%) were used in 17 runs. Regression models describing the effect of variables on the product responses were obtained. Descriptive profile analyses and consumer acceptability test were carried out on optimized flavoured extruded meat analogue. Responses were mostly affected by barrel temperature and moisture level and to a lesser extent by screw speed. Optimization results based on desirability concept indicated that a barrel temperature of 120.15°C, feed moisture of 47% and screw speed of 119.19 rpm would produce meat analogue of preferable proximate composition, functional and sensory properties which reveals consumers` likeness for the product.

156 Food Safety and Perceived Risk: A Case Study of Khao San Road, Bangkok, Thailand

Authors: Siripen Yiamjanya, Kevin Wongleedee

Abstract:

Food safety is an important concern for holiday makers in foreign and unfamiliar tourist destinations. In fact, risk from food in these tourist destinations has an influence on tourist perception. This risk can potentially affect physical health and lead to an inability to pursue planned activities. The objective of this paper was to compare foreign tourists- demographics including gender, age and education level, with the level of perceived risk towards food safety. A total of 222 foreign tourists during their stay at Khao San Road in Bangkok were used as the sample. Independent- samples ttest, analysis of variance, and Least Significant Difference or LSD post hoc test were utilized. The findings revealed that there were few demographic differences in level of perceived risk among the foreign tourists. The post hoc test indicated a significant difference among the old and the young tourists, and between the higher and lower level of education. Ranks of tourists- perceived risk towards food safety unveiled some interesting results. Tourists- perceived risk of food safety in established restaurants can be ranked as i) cleanliness of dining utensils, ii) sanitation of food preparation area, and iii) cleanliness of food seasoning and ingredients. 155 Analysis of Food Security Situation among Nigerian Rural Farmers

Authors: Victoria A. Okwoche, Benjamin C. Asogwa

Abstract:

This paper analysed the food security situation among Nigerian rural farmers. Data collected on 202 rural farmers from Benue State were analysed using descriptive and inferential statistics. The study revealed that majority of the respondents (60.83%) had medium dietary diversity. Furthermore, household daily calorie requirement for the food secure households was 10,723 and the household daily calorie consumption was 12,598, with a surplus index of 0.04. The food security index was 1.16. The Household daily per capita calorie consumption was 3,221.2. For the food insecure households, the household daily calorie requirement was 20,213 and the household daily calorie consumption was 17,393. The shortfall index was 0.14. The food security index was 0.88. The Household daily per capita calorie consumption was 2,432.8. The most commonly used coping strategies during food stress included intercropping (99.2%), reliance on less preferred food (98.1%), limiting portion size at meal times (85.8%) and crop diversification (70.8%).

154 Optimization of Ethanol Fermentation from Pineapple Peel Extract Using Response Surface Methodology (RSM)

Authors: Nadya Hajar, Zainal, S., Atikah, O., Tengku Elida, T. Z. M.

Abstract:

Ethanol has been known for a long time, being perhaps the oldest product obtained through traditional biotechnology fermentation. Agriculture waste as substrate in fermentation is vastly discussed as alternative to replace edible food and utilization of organic material. Pineapple peel, highly potential source as substrate is a by-product of the pineapple processing industry. Bio-ethanol from pineapple (Ananas comosus) peel extract was carried out by controlling fermentation without any treatment. Saccharomyces ellipsoides was used as inoculum in this fermentation process as it is naturally found at the pineapple skin. In this study, the capability of Response Surface Methodology (RSM) for optimization of ethanol production from pineapple peel extract using Saccharomyces ellipsoideus in batch fermentation process was investigated. Effect of five test variables in a defined range of inoculum concentration 6- 14% (v/v), pH (4.0-6.0), sugar concentration (14-22°Brix), temperature (24-32°C) and time of incubation (30-54 hrs) on the ethanol production were evaluated. Data obtained from experiment were analyzed with RSM of MINITAB Software (Version 15) whereby optimum ethanol concentration of 8.637% (v/v) was determined. The optimum condition of 14% (v/v) inoculum concentration, pH 6, 22°Brix, 26°C and 30hours of incubation. The significant regression equation or model at the 5% level with correlation value of 99.96% was also obtained.

153 Research of the Main Indexes of Freshness Anchovy (Engraulis engrasicolus Linnaeus, 1758) and Sardines (Sardina pilchardus Walbaum 1792) of Mediterranean

Authors: G.R.A. Alberio, D. Scalone, G. Spagna

Abstract:

Anchovy (Engraulis Encrasicholus) and sardine (Sardina Pilchardus) are blue fishes linked to our alimentary tradition of Mediterranean. In our work, particularly, we tested for the first time physical and enzymatic methods to verify the freshness of species of blue fish, anchovy and sardine of Mediterranean. In connection with to the lowering of the pH after post-mortem stage we assisted to a increase in proteolytic activity of calpaine and catpsine. Already after 2 h in post-mortem there was a significant increase.

Authors: W. Samruan, A. Oonsivilai, R. Oonsivilai

Abstract:

Today, people are more interested in the foods beneficial on their health. However, there are still lacks of accurate knowledge in the field of biological properties, functional properties, including the application of legume in foods. This study focused on antioxidant activity of soybean (SB) and fermented soybean (FSB) crude extracts evaluating to have more information in fortification SB and FSB crude extracts in food products and/or dietary supplement. SB and FSB crude extracts were prepared by infusion with water and ethanol. The antioxidant activity of crude extracts was studied with DPPH and ABTS assay including commercial standard. From both DPPH and ABTS assay, the antioxidant activity of SB and FSB water crude extract showed higher antioxidant activity than ethanol crude extract, and FSB crude extract showed higher antioxidant activity than SB crude extract. In DPPH assay, BHT and vitamin C showed IC50 values at 0.241, 0.039 mg/ml, in ABTS assay. In addition, Trolox showed IC50 at 0.058 mg/ml respectively. FSB water crude extract showed high antioxidant activity. Finally, the functional properties study of both water and ethanol crude extracts should be done for beneficial in application of these extracts in food products and dietary supplement in the near future.

Authors: Saadoudi Mouni, Hamebaba Leila, Abdeddaim Mohamed

Abstract:

In Algeria, some fruit trees produce fruits in free nature. Such trees are Celtis australis, Crataegus azarolus, Crataegus monogyna and Zizyphus lotus. In spite of their appreciable consumption, their nutritional value remains unknown. The objective of this study is the determination of sugars in the pulpe and almond of the above fruits. The biochemical analysis shows that these fruits present interesting contents of soluble sugars which confers significant caloric intakes to them. As well as significant fibres which give them therapeutic and industrial benefits? The analysis of the almonds shows that it contains considerable contents of sugars which enable them to be an energetic food.
