160 Enhancing the Sensitivity of Antigen Based Sandwich ELISA for COVID-19 Diagnosis in Saliva Using Gold Conjugated Nanobodies

Abstract:
Development of sensitive non-invasive tests for detection of SARS-CoV-2 antigens is imperative to manage the extent of infection throughout the population, yet, it is still challenging. Here, we designed and optimized a sandwich enzyme-linked immunosorbent assay (ELISA) for SARS-CoV-2 S1 antigen detection in saliva. Both saliva samples and nasopharyngeal swapswere collected from 170 PCR-confirmed positive and negative cases. Gold nanoparticles (AuNPs) were conjugated with S1protein receptor binding domain (RBD) nanobodies. Recombinant S1 monoclonal antibodies (S1mAb) as primery antibody and gold conjugated nanobodies as secondary antibody were employed in sandwich ELISA. Our developed system were optimized to achieve 87.5 % sensitivity and 100% specificity for saliva samples compared to 89 % and 100% for nasopharyngeal swaps, respectively. This means that saliva could be a suitable replacement for nasopharyngeal swaps No cross reaction was detected with other corona virus antigens. These results revealed that our developed ELISAcould be establishedas a new, reliable, sensitive, and non-invasive test for diagnosis of SARS-CoV-2 infection, using the easily collected saliva samples.Keywords: COVID 19, diagnosis, ELISA, nanobodies

Abstract:
The experimental study was conducted on sediments dredging from the dam of Bni Haroun, the most important and the largest dam in Algeria. The first phase of the work was to substitution of crushed sand with sediments to study the workability and compressive strength of ordinary concretes. The second phase of the work is to study the behavior of concrete with sediment under the effect of the freeze-thaw cycles. Abstract:
In the recent COVID 19 pandemic, experts of public health have emphasized testing, tracking infected people, and tracing their contacts as an effective strategy to reduce the spread of the virus. Development of rapid and sensitive diagnostic assays to replace reverse transcription polymerase chain reaction (RT-PCR) is mandatory..Our innovative test strip relying on the application of nanoparticles conjugated to recombinant nanobodies for SARS-COV-2 spike protein (S1) & angiotensin-converting enzyme 2 (that is responsible for the virus entry into host cells) for rapid detection of SARS-COV-2 spike protein (S1) in saliva or sputum specimens. Comparative tests with RT-PCR will be held to estimate the significant effect of using COVID 19 nanobodies for the first time in the development of lateral flow test strip. The SARS-CoV-2 S1 (3 ng of recombinant proteins) was detected by our developed LFIA in saliva specimen of COVID-19 Patients No cross-reaction was detected with Middle East respiratory syndrome coronavirus (MERS-CoV) or SARS- CoV antigens..Our developed system revealed 96 % sensitivity and 100% specificity for saliva samples compared to 89 % and 100% sensitivity and specificity for nasopharyngeal swabs. providing a reliable alternative for the painful and uncomfortable nasopharyngeal swab process and the complexes, time consuming PCR test. Abstract:
The objective of this article is to improve the passive vibration damping of solar array (SA) used in space structures, by the effective application of numerical optimization. A case study of a SA is used for demonstration. A finite element (FE) model was created and verified by experimental testing. Optimization was then conducted by implementing the FE model with the genetic algorithm, to find the optimal placement of aluminum circular patches, to suppress the first two bending mode shapes. The results were verified using experimental testing. Finally, a parametric study was conducted using the FE model where patch locations, material type, and shape were varied one at a time, and the results were compared with the optimal ones. The results clearly show that through the proper application of FE modeling and numerical optimization, passive vibration damping of space structures has been successfully achieved.Keywords: damping optimization, genetic algorithm optimization, passive vibration damping, solar array vibration damping

The designed model has been applied on Vegetable Oil Purification System (neutralization system) to help the specialist user based on the concept of FMEA (Failure Mode and Effect Analysis) to estimate the reliability of the repairable system at the vegetable oil industry. The fuzzy model has been used to predict the system reliability for a future time period, depending on a historical database for the two past years. Reducing the quantity of cement in cementitious composites can help to reduce the environmental effect of construction materials. By-products such as ferronickel slags (FNS), fly ash (FA), and Crepidula fornicata (CR) are promising options for cement replacement. In this work, we investigated the relevance of substituting cement with FNS-CR and FA-CR on the mechanical properties of mortar and on the thermal properties of concrete. Foraging intervals ranging from 2 to 28 days, the mechanical properties are obtained by 3-point bending and compression tests. The chosen mix is used to construct a prototype in order to study the material’s hygrothermal performance. Decryption failure of the Rabin cryptosystem caused by the 4-to-1 decryption output is overcome efficiently in this work. In this research, we have linked the concept of rough set and topological structure to the creation of a new topological structure that assists in the analysis of the information systems of some electrical engineering issues. We used non-specific information whose boundaries do not have an empty set in the top topological structure is rough set. It is characterized by the fact that it does not contain a large number of elements and facilitates the establishment of rules. We used this structure in reducing the specifications of electrical information systems. We have provided a detailed example of this method illustrating the steps used. Arab Grammarians stood at variance with regard to the definition of kana (which might equal was, were, the past form of “be” in English). Kana was considered as a verb, a particle, or a quasi-verb by different scholars; others saw it as an auxiliary verb; while some other scholars categorized kana as one of the incomplete verbs or (Afa’al naqisa) based on two different claims: first, a considerable group of grammarians saw kana as fie’l naqis or an incomplete verb since it indicates time, but not the event or action itself. Second, kana requires a predicate (xabar) to complete the meaning, i.e., it does not suffice itself with a noun in the nominal sentence. This study argues that categorizing the verb kana as fie’l naqis or an incomplete verb is inaccurate and confusing since the term “incomplete” does not agree with its characteristics, meanings, and temporal indications. Moreover, interpreting kana as a past verb is also inaccurate. kana كان (derived from the absolute action of being كون) is considered unique and the most comprehensive verb, encompassing all tenses of the past, present, and future within the dimensions of continuity and eternity of all possible actions under “being”.Keywords: pragmatics, kana, context, Arab grammarians, meaning, fie’l naqis

Due to this diverse of mixed goals and requirements, e-Government systems need to be designed based on special design considerations in order to eliminate the risks of failure to compliance to government regulations, citizen dissatisfaction, or market repulsion. In this article we suggest a framework with guidelines for designing government information systems that takes into consideration the special requirements of the public sector. In this paper, numerical solutions for the nonlinear coupled Korteweg-de Vries, (abbreviated as KdV) equations are calculated by Sinc-collocation method. This approach is based on a global collocation method using Sinc basis functions. First, discretizing time derivative of the KdV equations by a classic finite difference formula, while the space derivatives are approximated by a $\theta-$weighted scheme. Sinc functions are used to solve these two equations. Soliton solutions are constructed to show the nature of the solution. Abstract:
Hydrogen production from cake wastewater by anaerobic dark fermentation via upflow anaerobic staged reactor (UASR) was investigated in this study. The reactor was continuously operated for four months at constant hydraulic retention time (HRT) of 21.57 hr, PH value of 6 ± 0.6, temperature of 21.1°C, and organic loading rate of 2.43 gCOD/l.d. The hydrogen production was 5.7 l H2/d and the hydrogen yield was 134.8 ml H2 /g CODremoved. The system showed an overall removal efficiency of TCOD, TBOD, TSS, TKN, and Carbohydrates of 40 ± 13%, 59 ± 18%, 84 ± 17%, 28 ± 27%, and 85 ± 15% respectively during the long term operation period. Based on the available results, the system is not sufficient for the effective treatment of cake wastewater, and the effluent quality of UASR is not complying for discharge into sewerage network, therefore a post treatment is needed (not covered in this study).Keywords: cake wastewater industry, chemical oxygen demand (COD), hydrogen production, up-flow anaerobic staged reactor (UASR)

Cytotoxic activity of the total methanol extract against breast cancer cell line MCF-7 was amazing IC50 at 54 ug/ml. 130 polyphenolic compounds were tentatively identified in pomegranate peel (Punica granatum L.) methanol extract using HPLC-MS/MS technique. The antiestrogenic activity of the polyphenolic constituents found in pomegranate extract was confirmed experimentally in-vitro and by the in-silico molecular docking using gallagic acid, ellagic acid, and Punicalagin as these are considered model compounds confirmed in pomegranate peel extract. Many recent studies show the positive effect of phenolic compounds in olive oil on health. They are known for their biological properties, where they have shown potential activity as an antioxidant, anti-inflammatory, and antimicrobial agents. However, this characteristic is rarely studied in olive oil from different regions of Algeria. Different samples collected from the western region of Algeria were evaluated for their polyphenol content, antioxidant activity, and antimicrobial effect. The obtained results demonstrated that this oil is rich in polyphenols and revealed high antimicrobial activity against Staphylococcus aureus and Escherichia coli. With the increase of network virtualization and the disparity of vendors, the continuous monitoring and detection of anomalies cannot rely on static rules. An advanced analytical methodology is needed to discriminate between ordinary events and unusual anomalies. In this paper, we focus on log data (textual data), which is a crucial source of information for network performance. Then, we introduce an algorithm used as a pipeline to help with the pretreatment of such data, group it into patterns, and dynamically label each pattern as an anomaly or not. Such tools will provide users and experts with continuous real-time logs monitoring capability to detect anomalies and failures in the underlying system that can affect performance. The treatment of the geotechnical database of the region of Casablanca was difficult to achieve due to the heterogeneity of the nomenclature of the lithological formations composing its soil. It appears necessary to harmonize the nomenclature of the facies and to produce cartographic documents useful for construction projects and studies before any investment program. To achieve this, more than 600 surveys made by the Public Laboratory for Testing and Studies (LPEE) in the agglomeration of Casablanca, were studied. Moreover, some local observations were made in different places of the metropolis. Each survey was the subject of a sheet containing lithological succession, macro and microscopic description of petrographic facies with photographic illustration, as well as measurements of geomechanical tests. Sprinkler irrigation system became more popular to reduce water consumption and increase irrigation efficiency. The water distribution uniformity plays an important role in the performance of the sprinkler irrigation system. The use of low operating pressure instead of high operating pressure can be achieved many benefits including energy and water saving. An experimental study was performed to investigate the water distribution uniformity of the solid-set sprinkler irrigation system under low operating pressure. Different low operating pressures (62, 82, 102 and 122 kPa) were selected. The range of operating pressure was lower than the recommended in the previous studies to investigate the effect of low pressure on the water distribution uniformity. Different nozzle diameters (4, 5, 6 and 7 mm) were used. The outdoor single sprinkler test was performed. The water distribution of single sprinkler, the coefficients of uniformity such as coefficient of uniformity (CU), distribution uniformity of low quarter (DUlq), distribution uniformity of low half (DUlh), coefficient of variation (CV) and the distribution characteristics like rotation speed, throw radius and overlapping distance are presented in this paper.Keywords: low operating pressure, sprinkler irrigation system, water distribution uniformity

This paper applies a modified Laplace Adomian decomposition method to solve the time-fractional JaulentMiodek system. The method produce convergent series solutions with easily compatible components. This paper considers the Caputo fractional derivative. The effectiveness and applicability of the method are demonstrated by comparing its results with those of prior studies. Results are presented in tables and figures. These solutions might be imperative and significant for the explanation of some practical physical phenomena. All computations and figures in the work are done using MATHEMATICA. Using HPLC-MS/MS technique, 133 polyphenolic compounds were identified in the methanol extract of pomegranate rind (Punica granatum L.). In-vitro cytotoxic activity against breast cancer cell line MCF-7 was investigated, with an IC50 of 54 ug/ml. In-silico molecular docking using ellagic acid, gallagic acid, and Punicalagin as model compounds identified in pomegranate rind extract confirmed the intriguing anti-estrogenic action of the key polyphenolic components in pomegranate rind extract. Surprisingly, taxol showed low activity compared to pomegranate compounds as ERα antagonist and ERβ agonist. To assure the viability of a numerical simulation, an experimental study of a binary mixture of monodiperse particles taken as free flowing material in a V blender was developed on the basis of relative standard deviation curves, and the arrangement of the particles in the vessel. So understanding teachers’ beliefs and their use of computers is an important way to create effective motivational systems for teachers to use computers in the classroom in an effective way.A qualitative study (6 focus group) was carried out among Saudi High school teachers, both male and female, to examine their attitudes towards computers and to find out their computer skills and usage. The study showed a gender difference in that females were less likely to attend computer workshops, females also had less computer skills, and they have more negative attitudes towards computers than males. Also, the study found that low computer skills in the classroom made students unlikely to have the lessons presented using computers. Furthermore, the study found some factors that affected teachers’ attitudes towards computers. These factors were computer experience and confidence as much having skills and good experience in computer use, the role and importance of computers had become in their life and in teaching as well.Keywords: attitude, education, student, teacher, technology

Abstract:
In Algeria, Opuntia ficus Indica production is important. This seasonal fruit is a characteristic of arid and semi-arid regions. Taking into account its high content in antioxidants, it has an excellent nutritional value. The aim of this research is the prickly pear morphological and physicochemical characterization study which is widely present in the Arris (Batna, Algeria) area. The results of this experimental study are comparative to those of the same species from other world regions. The whole fruit weight is estimated to reach 63.38 g with a juice ratio of 71.42%, a pH of 5.54, moisture of 89.3% and a brix of 10.4°. The quantitative amount of the phenolic compounds of the fruit revealed contents of 20.65-45.70 mg / 100 g of MF for total polyphenols and 0.519 -0.591 mg / 100 g of MF for the flavonoids.Keywords: functional composition, nutritionals properties, opuntia ficus indica, phenolic compounds

Abstract:
We present solitary wave solutions for the perturbed nonlinear Schrodinger (PNLS) equation describing propagation of femtosecond light pulses through the fiber Bragg grating structure where the pulse dynamics is governed by the nonlinear-coupled mode (NLCM) equations. Using the multiple scale analysis, we reduce the NLCM equations into the perturbed nonlinear Schrodinger (PNLS) type equation. Unlike the reported solitary wave solutions of the PNLS equation, the novel ones can describe W shaped of solitons and their properties.Keywords: fiber bragg grating, nonlinear-coupled mode equations, w shaped of solitons, PNLS
