1328 Influence of AgNO3 Treatment on the Flavonolignan Production in Cell Suspension Culture of Silybum marianum (L.) Gaertn

Abstract:
The abiotic elicitation is one of the methods for increasing the secondary metabolites production in plant tissue cultures and it seems to be more effective than traditional strategies. This study verified the use of silver nitrate as elicitor to enhance flavonolignans and flavonoid taxifolin production in suspension culture of Sylibum marianum (L.) Gaertn. Silver nitrate in various concentrations (5.887.10-3 mol/L, 5.887.10-4 mol/L, 5.887.10-5 mol/L) was used as elicitor. The content of secondary metabolites in cell suspension cultures was determined by high performance liquid chromatography. The samples were taken after 6, 12, 24, 48, 72 and 168 hours of treatment. The highest content of taxifolin production (2.2 mg.g-1) in cell suspension culture of Silybum marianum (L.) Gaertn. was detected after silver nitrate (5.887.10-4 mol/L) treatment and 72 h application. Flavonolignans such as silybinA, silybin B, silydianin, silychristin, isosilybin A, isosilybin B were not produced by cell suspension culture of S. marianum after elicitor treatment. Our results show that the secondarymetabolites could be released from S. marianum cells into the nutrient medium by changed permeability of cell wall.Keywords: Silybum marianum (L.) Organizational culture is crucial for innovation. Literature reveals that research of organizational culture mostly confirm already existing conceptions and models, but those help to make profile of innovation culture.
Research summarize previous research of organizational culture as culture which foster innovation and provide profile of innovation culture, which may be used by managers to improve cultural environment to increase performance of their companies. Research also leads to hypothesis for further research.

Abstract:
Cell processing techniques for gene and cell therapies use several separate procedures for gene transfer and cell separation or elimination, because no current technology can offer simultaneous multi-functional processing of specific cell sub-sets in heterogeneous cell systems. Using our novel on-demand nonstationary intracellular events instead of permanent materials, plasmonic nanobubbles, generated with a short laser pulse only in target cells, we achieved simultaneous multifunctional cell-specific processing with the rate up to 50 million cells per minute.

Abstract:
To help overcome limits to the density of conventional SRAMs and leakage current of SRAM cell in nanoscaled CMOS technology, we have developed a four-transistor SRAM cell. The newly developed CMOS four-transistor SRAM cell uses one word-line and one bit-line during read/write operation. This cell retains its data with leakage current and positive feedback without refresh cycle. The new cell size is 19% smaller than a conventional six-transistor cell using same design rules. Also the leakage current of new cell is 60% smaller than a conventional sixtransistor SRAM cell. Simulation result in 65nm CMOS technology shows new cell has correct operation during read/write operation and idle mode.

Abstract:
Compared to oil production from microorganisms, little work has been performed for mixed culture of microalgae and yeast. In this article it is aimed to show high oil accumulation potential of mixed culture of microalgae Chlorella sp. KKU-S2 and oleaginous yeast Torulaspora maleeae Y30 using sugarcane molasses as substrate. The monoculture of T. maleeae Y30 grew faster than that of microalgae Chlorella sp. KKU-S2. In monoculture of yeast, a biomass of 6.4g/L with specific growth rate (m) of 0.265 (1/d) and lipid yield of 0.466g/L were obtained, while 2.53g/L of biomass with m of 0.133 (1/d) and lipid yield of 0.132g/L were obtained for monoculture of Chlorella sp. KKU-S2. The biomass concentration in the mixed culture of T. maleeae Y30 with Chlorella sp. KKU-S2 increased faster and was higher compared with that in the monoculture and mixed culture of microalgae. In mixed culture of microalgae Chlorella sp. KKU-S2 and C. vulgaris TISTR8580, a biomass of 3.47g/L and lipid yield of 0.123 g/L were obtained. In mixed culture of T. maleeae Y30 with Chlorella sp. KKU-S2, a maximum biomass of 7.33 g/L and lipid yield of 0.808g/L were obtained. Maximum cell yield coefficient (YX/S, 0.229g/L), specific yield of lipid (YP/X, 0.11g lipid/g cells) and volumetric lipid production rate (QP, 0.115 g/L/d) were obtained in mixed culture of yeast and microalgae. Clearly, T. maleeae Y30 and Chlorella sp. KKU-S2 use sugarcane molasses as organic nutrients efficiently in mixed culture under mixotrophic growth. The biomass productivity and lipid yield are notably enhanced in comparison with monoculture.

Abstract:
A hydrogel from cellulose acetate cross linked with ethylenediaminetetraacetic dianhydride (HAC-EDTA) was synthesized by our research group, and submitted to characterization and biological tests. Cytocompatibility analysis was performed by confocal microscopy using human adipocyte derived stem cells (ASCs). The FTIR analysis showed characteristic bands of cellulose acetate and hydroxyl groups and the tensile tests evidence that HAC-EDTA present a Young’s modulus of 643.7 MPa. The confocal analysis revealed that there was cell growth at the surface of HAC-EDTA. After one day of culture the cells presented spherical morphology, which may be caused by stress of the sequestration of Ca2+ and Mg2+ ions at the cell medium by HAC-EDTA, as demonstrated by ICP-MS. However, after seven days and 14 days of culture, the cells present fibroblastoid morphology, phenotype expected by this cellular type. The results give efforts to indicate this new material as a potential biomaterial for tissue engineering, in the future in vivo approach.

Abstract:
Saccharomyces cerevisiae (baker-s yeast) can exhibit sustained oscillations during the operation in a continuous bioreactor that adversely affects its stability and productivity. Because of heterogeneous nature of cell populations, the cell population balance models can be used to capture the dynamic behavior of such cultures. In this paper an unstructured, segregated model is used which is based on population balance equation(PBE) and then in order to simulation, the 4th order Rung-Kutta is used for time dimension and three methods, finite difference, orthogonal collocation on finite elements and Galerkin finite element are used for discretization of the cell mass domain. The results indicate that the orthogonal collocation on finite element not only is able to predict the oscillating behavior of the cell culture but also needs much little time for calculations. Therefore this method is preferred in comparison with other methods. In the next step two controllers, a globally linearizing control (GLC) and a conventional proportional-integral (PI) controller are designed for controlling the total cell mass per unit volume, and performances of these controllers are compared through simulation. A new sythetic gene coding for a Human Elastin-Like Polypeptide was constructed and expressed. The recombinant product was tested as coating agent to realize a surface suitable for cell growth. Coatings showed peculiar features and different human cell lines were seeded and cultured. Abstract:
The iron environment in Fe-doped Vycor Anode was investigated with EXAFS using Brookhaven Synchrotron Light Source. The iron-reducing Shewanella oneidensis culture was grown in a microbial fuel cell under anaerobic respiration. The Fe bond length was found to decrease and correlate with the amount of biofilm growth on the Fe-doped Vycor Anode. Abstract:
Lawsone is a pigment that occurs naturally in plants. It has been used as a skin and hair dye for a long time. Moreover, its different biological activities have been reported. The present study focused on the effect of lawsone on a plant cell model represented by tobacco BY-2 cell suspension culture, which is used as a model comparable with the HeLa cells. It has been shown that lawsone inhibits the cell growth in the concentration-dependent manner. In addition, changes in DNA methylation level have been determined. We observed decreasing level of DNA methylation in the presence of increasing concentrations of lawsone. These results were accompanied with overproduction of reactive oxygen species (ROS). Since epigenetic modifications can be caused by different stress factors, there could be a connection between the changes in the level of DNA methylation and ROS production caused by lawsone.

Abstract:
The primary tool currently used to pre-process 10X chromium single-cell ATAC-seq data is Cell Ranger, which can take very long to run on standard datasets. To facilitate rapid pre-processing that enables reproducible workflows, we present a suite of tools called scATAK for pre-processing single-cell ATAC-seq data that is 15 to 18 times faster than Cell Ranger on mouse and human samples. Our tool can also calculate chromatin interaction potential matrices and generate open chromatin signal and interaction traces for cell groups. We use scATAK tool to explore the chromatin regulatory landscape of a healthy adult human brain and unveil cell-type specific features, and show that it provides a convenient and computational efficient approach for pre-processing single-cell ATAC-seq data.

Abstract:
A total of 115 yeast strains isolated from local cassava processing wastes were measured for crude protein content. Among these strains, the strain MSY-2 possessed the highest protein concentration (>3.5 mg protein/mL). By using molecular identification tools, it was identified to be a strain of Pichia kudriavzevii based on similarity of D1/D2 domain of 26S rDNA region. In this study, to optimize the protein production by MSY-2 strain, Response Surface Methodology (RSM) was applied. The tested parameters were the carbon content, nitrogen content, and incubation time. Here, the value of regression coefficient (R2) = 0.7194 could be explained by the model which is high to support the significance of the model. Under the optimal condition, the protein content was produced up to 3.77 g per L of the culture and MSY-2 strain contains 66.8 g protein per 100 g of cell dry weight. These results revealed the plausibility of applying the novel strain of yeast in single-cell protein production.Keywords: Single cell protein, response surface methodology, yeast, cassava processing waste.

Epstein-Barr virus (EBV) is implicated in the pathogenesis of the endemic Burkitt-s lymphoma (BL). The EBVpositive BL-derived cell lines initially maintain the original tumor phenotype of EBV infection (latency I, LatI), but most of them drift toward a lymphoblast phenotype of EBV latency III (LatIII) during in vitro culturing. The aim of the present work was to characterize the B-cell subsets in EBV-positive BL cell lines and to verify whether a particular cell subset correlates with the type of EBV infection. The phenotype analysis of two EBV-negative and eleven EBV-positive (three of LatI and eight of LatIII) BL cell lines was performed by polychromatic flow cytomery, based on expression pattern of CD19, CD10, CD38, CD27, and CD5 markers. Two cell subsets, CD19+CD10+ and CD19+CD10-, were defined in LatIII BL cell lines. In both subsets, the CD27 and CD5 cell surface expression was detected in a proportion of the cells.

Abstract:
Since the heart of the hybrid system is the fuel cell and it has vital impact on efficiency and performance of cycle, in this study, the major modeling of electrochemical reaction within the fuel cell is analyzed. Also, solid oxide fuel cell is integrated with the gas turbine and thermodynamic analysis on different elements of hybrid system is applied. Next, in predefined operational points of hybrid cycle, the simulation results are obtained. Then, different source of irreversibility in fuel cell is modeled and influence of different major parameters on different irreversibility is computed and applied. Then, the effect of important parameters such as thickness and surface of electrolyte fuel cell are simulated in fuel cell and its dependency to these parameters is explained. Abstract:
Aloe Vera is a short-stemmed succulent plant which is commonly used in Myanmar traditional medicine. A. vera gel was also used as food addictive. This study aims to improve the Myanmar folk medicine to a functional beverage. In this research, Aloe vera was fermented with Saccharomyces cerevisiae for 6 months. Three different processes were carried out. Process I contains A. vera 10%, sugar 30%, water 50%, and starter culture 10%, process II contains A. vera 10%, sugar 15%, honey 15%, and water 50%, starter culture 10%; process III contains A. vera 10%, honey 30%, water 50%, starter culture 10%. During wine fermentation, the wine parameters such as alcohol content, total soluble solid (ºBrix), pH, color and cell population were analyzed. After 30 days of fermentation, total cell population remained 2.8x106 in P-I, P-II and 3.2x106 in P-III. Total soluble solid content dropped to 15.8 in P-I, P-II and 15.7 in P-III. After 30 days, clear wine was transferred to other vassals for racking. After 6 months of racking, microbial population reached under detectable level and alcohol content was round about 11% but not significantly different among these processes. P-II was found to have the highest color intensity at 450 nm and it got the most taster satisfaction when sensory evaluation was carried out using five hedonic scales after 6 month of racking.Keywords: Aloe vera, fermentation, S. cerevisiae, functional beverage, folk medicine.

Abstract:
Fuel and oxidant gas delivery plate, or fuel cell plate, is a key component of a Proton Exchange Membrane (PEM) fuel cell. To manufacture low-cost and high performance fuel cell plates, advanced computer modeling and finite element structure analysis are used as virtual prototyping tools for the optimization of the plates at the early design stage. The present study examines thermal stress analysis of the fuel cell plates that are produced using a patented, low-cost fuel cell plate production technique based on screen-printing. Design optimization is applied to minimize the maximum stress within the plate, subject to strain constraint with both geometry and material parameters as design variables. The study reveals the characteristics of the printed plates, and provides guidelines for the structure and material design of the fuel cell plate.Keywords: Design optimization, FEA, PEM fuel cell, Thermal stress

Fuel cells have become one of the major areas of research in the academia and the industry. The goal of most fish farmers is to maximize production and profits while holding labor and management efforts to the minimum. Risk of fish kills, disease outbreaks, poor water quality in most pond culture operations, aeration offers the most immediate and practical solution to water quality problems encountered at higher stocking and feeding rates. Many units of aeration system are electrical units so using a continuous, high reliability, affordable, and environmentally friendly power sources is necessary. Aeration of water by using PEM fuel cell power is not only a new application of the renewable energy, but also, it provides an affordable method to promote biodiversity in stagnant ponds and lakes. This paper presents a new design and control of PEM fuel cell powered a diffused air aeration system for a shrimp farm in Mersa Matruh in Egypt. Also Artificial intelligence (AI) techniques control is used to control the fuel cell output power by control input gases flow rate. Moreover the mathematical modeling and simulation of PEM fuel cell is introduced. A comparison study is applied between the performance of fuzzy logic control (FLC) and neural network control (NNC). This paper analyzes different techniques of the fine grained security of relational databases for the two variables-data accessibility and inference. Data accessibility measures the amount of data available to the users after applying a security technique on a table. Inference is the proportion of information leakage after suppressing a cell containing secret data. A row containing a secret cell which is suppressed can become a security threat if an intruder generates useful information from the related visible information of the same row. This paper measures data accessibility and inference associated with row, cell, and column level security techniques. Cell level security offers greatest data accessibility as it suppresses secret data only. But on the other hand, there is a high probability of inference in cell level security. Row and column level security techniques have least data accessibility and inference. This paper introduces cell plus innocent security technique that utilizes the cell level security method but suppresses some innocent data to dodge an intruder that a suppressed cell may not necessarily contain secret data. Four variations of the technique namely cell plus innocent 1/4, cell plus innocent 2/4, cell plus innocent 3/4, and cell plus innocent 4/4 respectively have been introduced to suppress innocent data equal to 1/4, 2/4, 3/4, and 4/4 percent of the true secret data inside the database. Results show that the new technique offers better control over data accessibility and inference as compared to the state-of-theart security techniques. This paper further discusses the combination of techniques together to be used. The paper shows that cell plus innocent 1/4, 2/4, and 3/4 techniques can be used as a replacement for the cell level security.

In-vitro mouse co-culture of E14 embryonic stem cells (ESCs) and OP9 stromal cells can recapitulate the earliest stages of haematopoietic development, not accessible in human embryos, supporting both haemogenic precursors and their primitive haematopoietic progeny. 1α, 25-Dihydroxy-vitamin D3 (VD3) has been demonstrated to be a powerful differentiation inducer for a wide variety of neoplastic cells, and could enhance early differentiation of ESCs into blood cells in E14/OP9 co-culture. This study aims to ascertain whether VD3 is key in promoting differentiation and suppressing proliferation, by separately investigating the effects of VD3 on the proliferation phase of the E14 cell line and on stromal OP9 cells.The results showed that VD3 inhibited the proliferation of the cells in a dose-dependent manner, quantitatively by decreased cell number, and qualitatively by alkaline-phosphatase staining that revealed significant differences between VD3-treated and untreated cells, characterised by decreased enzyme expression (colourless cells). Propidium-iodide cell-cycle analyses showed no significant percentage change in VD3-treated E14 and OP9 cells within their G and S-phases, compared to the untreated controls, despite the increased percentage of G-phase compared to the S-phase in a dosedependent manner. These results with E14 and OP9 cells indicate that adequate VD3 concentration enhances cellular differentiation and inhibits proliferation. The results also suggest that if E14 and OP9 cells were co-cultured andVD3-treated, there would be furtherenhanced differentiation of ESCs into blood cells.

Abstract:
Monoculture and mixed cultures of microalgae and the oleaginous yeast for microbial oil productions were investigated using sugarcane juice as carbon substrate. The monoculture of yeast Torulaspora maleeae Y30, Torulaspora globosa YU5/2 grew faster than that of microalgae Chlorella sp. KKU-S2. In monoculture of T. maleeae Y30, a biomass of 8.267g/L with lipid yield of 0.920g/L were obtained, while 8.333g/L of biomass with lipid yield of 1.141g/L were obtained for monoculture of T. globosa YU5/2. A biomass of 1.933g/L with lipid yield of 0.052g/L was found for monoculture of Chlorella sp. KKU-S2. The biomass concentration in the mixed culture of the oleaginous yeast with microalgae increased faster and was higher compared with that in the monocultures. A biomass of 8.733g/L with lipid yield of 1.564g/L was obtained for a mixed culture of T. maleeae Y30 with Chlorella sp. KKU-S2, while 8.010g/L of biomass with lipid yield of 2.424g/L was found for mixed culture of T. globosa YU5/2 with Chlorella sp. KKU-S2. Maximum cell yield coefficient (YX/S, g/L) was found of 0.323 in monoculture of Chlorella sp. KKU-S2 but low level of both specific yield of lipid (YP/X, g lipid/g cells) of 0.027 and volumetric lipid production rate (QP, g/L/d) of 0.003 were observed. While, maximum YP/X (0.303), QP (0.105) and maximum process product yield (YP/S, 0.061) were obtained in mixed culture of T. globosa YU5/2 with Chlorella sp. KKU-S2. The results obtained from the study shows that mixed culture of yeast with microalgae is a desirable cultivation process for microbial oil production.Keywords: Microbial oil, Chlorella sp. Abstract:
One of the tasks in contemporary biotechnology, pharmacology and other fields of human activities is to obtain biologically active substances from plants. They are very essential in the treatment of many diseases due to their actually high therapeutic value without visible side effects. However, sometimes the possibility of obtaining the metabolites is limited due to the reduction of wild-growing plants. That is why the plant cell cultures are of great interest as alternative sources of biologically active substances. Besides, during the monitored cultivation, it is possible to obtain substances that are not synthesized by plants in nature. Isolated culture of Ajuga genevensis with high growth activity and ability of regeneration was obtained using MS nutrient medium. The agar-diffusion method showed that aqueous extracts of callus culture revealed high antimicrobial activity towards various gram-positive (Bacillus subtilis A1WT; B. mesentericus WDCM 1873; Staphylococcus aureus WDCM 5233; Staph. citreus WT) and gram-negative (Escherichia coli WKPM M-17; Salmonella typhimurium TA 100) microorganisms. The broth dilution method revealed that the minimal and half maximal inhibitory concentration values against E. coli corresponded to the 70 μg/mL and 140 μg/mL concentration of the extract respectively. According to the photochemiluminescent analysis, callus tissue extracts of leaf and root origin showed higher antioxidant activity than the same quantity of A. genevensis intact plant extract. A. genevensis intact plant and callus culture extracts showed no cytotoxic effect on K-562 suspension cell line of human chronic myeloid leukemia. The GC-MS analysis showed deep differences between the qualitative and quantitative composition of callus culture and intact plant extracts. Hexacosane (11.17%); n-hexadecanoic acid (9.33%); and 2-methoxy-4-vinylphenol (4.28%) were the main components of intact plant extracts. 10-Methylnonadecane (57.0%); methoxyacetic acid, 2-tetradecyl ester (17.75%) and 1-Bromopentadecane (14.55%) were the main components of A. genevensis callus culture extracts. Obtained data indicate that callus culture of A. genevensis can be used as an alternative source of biologically active substances.

Abstract:
Metallic foams have good potential for lightweight structures for impact and blast mitigation. Therefore it is important to find out the optimized foam structure (i.e. cell size, shape, relative density, and distribution) to maximise energy absorption. In this paper, quasistatic compression and microstructural characterization of closed-cell aluminium foams of different pore size and cell distributions have been carried out. We present results for two different aluminium metal foams of density 0.49-0.51 g/cc and 0.31- 0.34 g/cc respectively that have been tested in quasi-static compression. The influence of cell geometry and cell topology on quasistatic compression behaviour has been investigated using optical microscope and computed tomography (micro-CT) analysis. It is shown that the deformation is not uniform in the structure and collapse begins at the weakest point.

Abstract:
The importance of having a good organizational culture that supports employee job satisfaction has fascinated both the business and academic world because of a tantalizing promise: culture can be fundamental to the enhancement of financial performance. This promise has led to growing interest for both researchers and practitioners in attempting to understand the influence of organizational culture on employees’ satisfaction and organizational performance. Even though the relationship between organizational culture and employee job satisfaction have gained attention in the literature, the majority of studies have been conducted within manufacturing organizations and tend to oversee the impact of culture on employee job satisfaction in a service-based environment. Thus, the main driving force of this study was to explore the role of organizational culture types in facilitating employee job satisfaction at Emerald Publishing Group. Interviews qualitative data analysis indicated that Emerald’s culture dominated by adhocracy and clan culture values. In addition, the findings provided evidence, which demonstrated that group and adhocracy organizational culture types play key roles in facilitating employee job satisfaction in a service-based environment.

Fuel cell is an emerging technology in the field of renewable energy sources which has the capacity to replace conventional energy generation sources. Fuel cell utilizes hydrogen energy to produce electricity. The electricity generated by the fuel cell can’t be directly used for a specific application as it needs proper power conditioning. Moreover, the output power fluctuates with different operating conditions. To get a stable output power at an economic rate, power conditioning circuit is essential for fuel cell. This paper implements a two-staged power conditioning unit for fuel cell based distributed generation using hysteresis current control technique.

Relevant agricultural information disseminator (extension agent) ratio of 1:3500 farm families which become a menace to agricultural production capacity in developing countries necessitate this study. Out of 4 zones in the state, 24 extension agents in each zone, 4 extension agents using cell phones and 120 farmers using cell phone and 120 other farmers not using cell phone were purposively selected to give 240 farmers that participated in the research. Data were collected using interview guide and analysized using frequency, percentage and t-test.. Frequency of contact with agricultural information centers revealed that cell phone user farmers had greater means score of X 41.43 contact as against the low mean X19.32 contact recorded by farmers receiving agricultural information from extension agents not using cell phone and their production was statistically significant at P < 0.05. Abstract:
This paper proposes an adaptive discharge time control method to balance cell voltages in alternating battery cell discharging method. In the alternating battery cell discharging method, battery cells are periodically discharged in turn. Recovery effect increases battery output voltage while the given battery cell rests without discharging, thus battery operation time of target system increases. However, voltage mismatch between cells leads two problems. First, voltage difference between cells induces inter-cell current with wasted power. Second, it degrades battery operation time, since system stops when any cell reaches to the minimum system operation voltage. To solve this problem, the proposed method adaptively controls cell discharge time to equalize both cell voltages. In the proposed method, battery operation time increases about 19%, while alternating battery cell discharging method shows about 7% improvement.

This paper presents the development of a compact, portable and easy to handle solar cell characterization device. The presented device reduces the effort and cost of single solar cell characterization to a minimum. It enables realistic characterization of cells under sunlight within minutes. In the field of photovoltaic research the common way to characterize a single solar cell or a module is, to measure the current voltage curve. With this characteristic the performance and the degradation rate can be defined which are important for the consumer or developer.
