1547 In silico Analysis of Isoniazid Resistance in Mycobacterium tuberculosis

Abstract:
Altered drug binding may be an important factor in isoniazid (INH) resistance, rather than major changes in the enzyme’s activity as a catalase or peroxidase (KatG). The identification of structural or functional defects in the mutant KatGs responsible for INH resistance remains as an area to be explored. In this connection, the differences in the binding affinity between wild-type (WT) and mutants of KatG were investigated, through the generation of three mutants of KatG, Ser315Thr [S315T], Ser315Asn [S315N], Ser315Arg [S315R] and a WT [S315]) with the help of software-MODELLER. The mutants were docked with INH using the software-GOLD. The affinity is lower for WT than mutant, suggesting the tight binding of INH with the mutant protein compared to WT type. These models provide the in silico evidence for the binding interaction of KatG with INH and implicate the basis for rationalization of INH resistance in naturally occurring KatG mutant strains of Mycobacterium tuberculosis.

Abstract:
Hanging to the trapezoidal sheet by decking hanger is a very widespread solution used in civil engineering to lead the distribution of energy, sanitary, air distribution system etc. under the roof or floor structure. The trapezoidal decking hanger is usually a part of the whole installation system for specific distribution medium. The leading companies offer installation systems for each specific distribution e.g. pipe rings, sprinkler systems, installation channels etc. Every specific part is connected to the base connector which is decking hanger. The own connection has three main components: decking hanger, threaded bar with nuts and web of trapezoidal sheet. The aim of this contribution is determinate the failure mechanism of each component in connection. Load bearing capacity of most components in connection could be calculated by formulas in European codes. This contribution is focused on problematic of bearing resistance of threaded bar in web of trapezoidal sheet. This issue is studied by experimental research and numerical modelling. This contribution presented the initial results of experiment which is compared with numerical model of specimen.

Abstract:
This paper describes a method for preparing metallic Cu nanoparticles in aqueous solution, and a metal-metal bonding technique using the Cu particles.Preparation of the Cu particle colloid solution was performed in water at room temperature in air using a copper source (0.01 M Cu(NO3)2), a reducing reagent (0.2 - 1.0 M hydrazine), and stabilizers (0.5×10-3 M citric acid and 5.0×10-3 M cetyltrimethylammonium bromide). The metallic Cu nanoparticles with sizes of ca. 60nm were prepared at all the hydrazine concentrations examined. A stage and a plate of metallic Cu were successfully bonded under annealing at 400oC and pressurizing at 1.2 MPa for 5min in H2 gas with help of the metallic Cu particles. A shear strength required for separating the bonded Cu substrates reached the maximum value at a hydrazine concentration of 0.8M, and it decreased beyond the concentration. Consequently, the largest shear strength of 22.9 MPa was achieved at the 0.8 M hydrazine concentration.

Abstract:
The agriculture lignocellulosic by-products are receiving increased attention, namely in the search for filter materials that retain contaminants from water. These by-products, specifically almond and hazelnut shells are abundant in Portugal once almond and hazelnuts production is a local important activity. Hazelnut and almond shells have as main constituents lignin, cellulose and hemicelluloses, water soluble extractives and tannins. Along the adsorption of heavy metals from contaminated waters, water soluble compounds can leach from shells and have a negative impact in the environment. Usually, the chemical characterization of treated water by itself may not show environmental impact caused by the discharges when parameters obey to legal quality standards for water. Only biological systems can detect the toxic effects of the water constituents. Therefore, the evaluation of toxicity by biological tests is very important when deciding the suitability for safe water discharge or for irrigation applications.
The main purpose of the present work was to assess the potential impacts of waters after been treated for heavy metal removal by hazelnut and almond shells adsorption systems, with short term acute toxicity tests.
To conduct the study, water at pH 6 with 25 mg.L-1 of lead, was treated with 10 g of shell per litre of wastewater, for 24 hours. This procedure was followed for each bark. Afterwards the water was collected for toxicological assays; namely bacterial resistance, seed germination, Lemna minor L. test and plant grow. The effect in isolated bacteria strains was determined by disc diffusion method and the germination index of seed was evaluated using lettuce, with temperature and humidity germination control for 7 days. For aquatic higher organism, Lemnas were used with 4 days contact time with shell solutions, in controlled light and temperature. For terrestrial higher plants, biomass production was evaluated after 14 days of tomato germination had occurred in soil, with controlled humidity, light and temperature.
Toxicity tests of water treated with shells revealed in some extent effects in the tested organisms, with the test assays showing a close behaviour as the control, leading to the conclusion that its further utilization may not be considered to create a serious risk to the environment.

This paper presents a speed sensorless direct torque control scheme using space vector modulation (DTC-SVM) for permanent magnet synchronous motor (PMSM) drive based a Model Reference Adaptive System (MRAS) algorithm and stator resistance estimator. The MRAS is utilized to estimate speed and stator resistance and compensate the effects of parameter variation on stator resistance, which makes flux and torque estimation more accurate and insensitive to parameter variation. In other hand the use of SVM method reduces the torque ripple while achieving a good dynamic response. Negative pressures of liquids have been expected to contribute many kinds of technology. Nevertheless, experiments for subjecting liquids which have not too small volumes to negative pressures are difficult even now. The reason of the difficulties is because the liquids tend to generate cavities easily. In order to remove cavitation nuclei, an apparatus for enclosing water into a metal Berthelot tube under vacuum conditions was developed. By using the apparatus, negative pressures for water rose to ca. -20 MPa. This is the highest value for water in metal Berthelot tubes. Results were explained by a traditional crevice model. A research project dealing with the phytoremediation of a soil polluted by some heavy metals is currently running. The case study is represented by a mining area in Hamedan province in the central west part of Iran. The potential of phytoextraction and phytostabilization of plants was evaluated considering the concentration of heavy metals in the plant tissues and also the bioconcentration factor (BCF) and the translocation factor (TF). Also the several established criteria were applied to define hyperaccumulator plants in the studied area. Results showed that none of the collected plant species were suitable for phytoextraction of Cu, Zn, Fe and Mn, but among the plants, Euphorbia macroclada was the most efficient in phytostabilization of Cu and Fe, while, Ziziphora clinopodioides, Cousinia sp. and Chenopodium botrys were the most suitable for phytostabilization of Zn and Chondrila juncea and Stipa barbata had the potential for phytostabilization of Mn. Using the most common criterion, Euphorbia macroclada and Verbascum speciosum were Fe hyperaccumulator plants. In the recent years, aluminum metal matrix composites were most widely used, which are finding wide applications in various field such as automobile, aerospace defense etc., due to their outstanding mechanical properties like low density, light weight, exceptional high levels of strength, stiffness, wear resistance, high temperature resistance, low coefficient of thermal expansion and good formability. In the present work, an effort is made to study the effect of heat treatment on mechanical properties of aluminum 7075 alloy reinforced with constant weight percentage of naturally occurring mineral beryl and varying weight percentage of graphene. The hybrid composites are developed with 0.5 wt. %, 1wt.%, 1.5 wt.% and 2 wt.% of graphene and 6 wt.% of beryl by stir casting liquid metallurgy route. The cast specimens of unreinforced aluminum alloy and hybrid composite samples were prepared for heat treatment process and subjected to solutionizing treatment (T6) at a temperature of 490±5 oC for 8 hours in a muffle furnace followed by quenching in boiling water. The microstructure analysis of as cast and heat treated hybrid composite specimens are examined by scanning electron microscope (SEM). The tensile test and hardness test of unreinforced aluminum alloy and hybrid composites are examined. The wear behavior is examined by pin-on disc apparatus. The results of as cast specimens and heat treated specimens were compared. The heat treated Al7075-Beryl-Graphene hybrid composite had better properties and significantly improved the ultimate tensile strength, hardness and reduced wear loss when compared to aluminum alloy and as cast hybrid composites.

Abstract:
Magnetically controlled microelectromechanical system (MCMEMS) switches is one of the directions in the field of micropower switching technology. MCMEMS switches are a promising alternative to Hall sensors and reed switches. The most important parameter for MCMEMS is the contact resistance, which should have a minimum value and is to be stable for the entire duration of service life. The value and stability of the contact resistance is mainly determined by the contact coating material. This paper presents the research results of a contact coating based on nanoscale ruthenium films obtained by electrolytic deposition. As a result of the performed investigations, the deposition modes of ruthenium films are chosen, the regularities of the contact resistance change depending on the number of contact switching, and the coating roughness are established. It is shown that changing the coating roughness makes it possible to minimize the contact resistance.

In this study rack systems that are structural storage units of warehouses have been analyzed as structural with Finite Element Method (FEA). Each cell of discussed rack system storages pallets which have from 800 kg to 1000 kg weights and 0.80x1.15x1.50 m dimensions. Under this load, total deformations and equivalent stresses of structural elements and principal stresses, tensile stresses and shear stresses of connection elements have been analyzed. The results of analyses have been evaluated according to resistance limits of structural and connection elements. Abstract:
The present paper concerns with the influence of fiber packing on the transverse plastic properties of metal matrix composites. A micromechanical modeling procedure is used to predict the effective mechanical properties of composite materials at large tensile and compressive deformations. Microstructure is represented by a repeating unit cell (RUC). Two fiber arrays are considered including ideal square fiber packing and random fiber packing defined by random sequential algorithm. The micromechanical modeling procedure is implemented for graphite/aluminum metal matrix composite in which the reinforcement behaves as elastic, isotropic solids and the matrix is modeled as an isotropic elastic-plastic solid following the von Mises criterion with isotropic hardening and the Ramberg-Osgood relationship between equivalent true stress and logarithmic strain. The deformation is increased to a considerable value to evaluate both elastic and plastic behaviors of metal matrix composites. In this paper are illustrated the principal aspects connected with the numerical evaluation of thermal stress induced by high gradient temperature in the concrete beam. The reinforced concrete beam has many advantages over steel beam, such as high resistance to high temperature, high resistance to thermal shock, Better resistance to fatigue and buckling, strong resistance against, fire, explosion, etc. The main drawback of the reinforced concrete beam is its poor resistance to tensile stresses. Abstract:
In this study, Friction Stir Processing (FSP) a recent grain refinement technique was employed to disperse micron-sized (2 *m) SiCp particles into aluminum alloy AA6063. The feasibility to fabricate bulk composites through FSP was analyzed and experiments were conducted at different traverse speeds and wider volumes of the specimens. Micro structural observation were carried out by employing optical microscopy test of the cross sections in both parallel and perpendicular to the tool traverse direction. Mechanical property including micro hardness was evaluated in detail at various regions on the specimen. The composites had an excellent bonding with aluminum alloy substrate and a significant increase of 30% in the micro hardness value of metal matrix composite (MMC) as to that of the base metal has observed. The observations clearly indicate that SiC particles were uniformly distributed within the aluminum matrix.

Abstract:
In recent years, rehabilitation has been the subject of extensive research due to increased spending on building work and repair of built works. In all cases, it is absolutely essential to carry out methods of strengthening or repair of structural elements, and that following an inspection analysis and methodology of a correct diagnosis. The reinforced concrete columns are important elements in building structures. They support the vertical loads and provide bracing against the horizontal loads. This research about the behavior of reinforced concrete rectangular columns, rehabilitated by concrete liner, confinement FRP fabric, steel liner or cage formed by metal corners. It allows comparing the contributions of different processes used perspective section resistance elements rehabilitated compared to that is not reinforced or repaired. The different results obtained revealed a considerable gain in bearing capacity failure of reinforced sections cladding concrete, metal bracket, steel plates and a slight improvement to the section reinforced with fabric FRP. The use of FRP does not affect the weight of the structures, but the use of different techniques cladding increases the weight of elements rehabilitated and therefore the weight of the building which requires resizing foundations.

Abstract:
Automotive experimental measurements in wind tunnel are often conducted on reduced scale. Depending on the study, different similitude parameters are used by researchers to best reproduce the flow at full scale. In this paper, two parameters are investigated, which are Reynolds number and upstream velocity when dealing with airflow of typical urban speed range, below 15 m.s-1. Their impact on flow structures and aerodynamic drag in the wake of a heavy truck model are explored. To achieve this, Computational Fluid Dynamics (CFD) simulations have been conducted with the aim of modeling the wake airflow of full- and reduced-scaled heavy trucks (1/4 and 1/28). The Reynolds Average Navier-Stokes (RANS) approach combined to the Reynolds Stress Model (RSM) as the turbulence model closure was used. Both drag coefficients and upstream velocity profiles (flow topology) were found to be close one another for the three investigated scales, when the dynamical similitude Reynolds is achieved. Moreover, the difference is weak for the simulations based on the same inlet air velocity. Hence, for the relative low velocity range investigated here, the impact of the scale factor is limited.

Abstract:
The edge waviness in hot rolled steel is a common defect. Variables that affect such defect include raw material and machine. These variables are necessary to consider to understand such defect. This research studied the defect of edge waviness for SS 400 of metal sheet manufacture. Defect of metal sheets were divided into two groups. The specimens were investigated on chemical composition and mechanical properties to find the difference. The results of investigation showed that the difference was not significant. Therefore the roll mill machine should be used to adjust to support another location on a roller to avoide edge waviness.

Abstract:
Contact width and contact stress are important parameters related to the leakage behavior of corrugated metal gasket. In this study, contact width and contact stress of three-layer corrugated metal gasket are investigated due to the modulus of elasticity and thickness of surface layer for 2 type gasket (0-MPa and 400-MPa mode). A finite element method was employed to develop simulation solution to analysis the effect of each parameter. The result indicated that lowering the modulus of elasticity ratio of surface layer will result in better contact width but the average contact stresses are smaller. When the modulus of elasticity ratio is held constant with thickness ratio increase, its contact width has an increscent trend otherwise the average contact stress has decreased trend.

Abstract:
Most HWRs currently use natural uranium fuel. Using enriched uranium fuel results in a significant improvement in fuel cycle costs and uranium utilization. On the other hand, reactivity changes of HWRs over the full range of operating conditions from cold shutdown to full power are small. This reduces the required reactivity worth of control devices and minimizes local flux distribution perturbations, minimizing potential problems due to transient local overheating of fuel. Analyzing heavy water effectiveness on neutronic parameters such as enrichment requirements, peaking factor and reactivity is important and should pay attention as primary concepts of a HWR core designing. Two nuclear nuclear reactors of CANDU-type and hexagonal-type reactor cores of 33 fuel assemblies and 19 assemblies in 1.04 P/D have been respectively simulated using MCNP-4C code. Using heavy water and light water as moderator have been compared for achieving less reactivity insertion and enrichment requirements. Two fuel matrixes of (232Th/235U)O2 and (238/235U)O2 have been compared to achieve more economical and safe design. Heavy water not only decreased enrichment needs, but it concluded in negative reactivity insertions during moderator density variations. Thorium oxide fuel assemblies of 2.3% enrichment loaded into the core of heavy water moderator resulted in 0.751 fission to absorption ratio and peaking factor of 1.7 using. Heavy water not only provides negative reactivity insertion during temperature raises which changes moderator density but concluded in 2 to 10 kg reduction of enrichment requirements, depend on geometry type.

At the previous study of new metal gasket, contact width and contact stress were important design parameter for optimizing metal gasket performance. However, the range of contact stress had not been investigated thoroughly. In this study, we conducted a gasket design optimization based on an elastic and plastic contact stress analysis considering forming effect using FEM. The gasket model was simulated by using two simulation stages which is forming and tightening simulation. The optimum design based on an elastic and plastic contact stress was founded. Final evaluation was determined by helium leak quantity to check leakage performance of both type of gaskets. The helium leak test shows that a gasket based on the plastic contact stress design better than based on elastic stress design.Keywords: Contact stress, metal gasket, plastic, elastic

Silicon photonics is merging as a unified platform for driving photonic based telecommunications and for local photonic based interconnect but it suffers from large footprint as compared with the nanoelectronics. Plasmonics is an attractive alternative for nanophotonics. In this work, two CMOS compatible plasmonic waveguide platforms are compared. One is the horizontal metal-insulator-Si-insulator-metal nanoplasmonic waveguide and the other is metal-insulator-Si hybrid plasmonic waveguide. Various passive and active photonic devices have been experimentally demonstrated based on these two plasmonic waveguide platforms.

Aerial and satellite images are information rich. They are also complex to analyze. For GIS systems, many features require fast and reliable extraction of roads and intersections. In this paper, we study efficient and reliable automatic extraction algorithms to address some difficult issues that are commonly seen in high resolution aerial and satellite images, nonetheless not well addressed in existing solutions, such as blurring, broken or missing road boundaries, lack of road profiles, heavy shadows, and interfering surrounding objects. The new scheme is based on a new method, namely reference circle, to properly identify the pixels that belong to the same road and use this information to recover the whole road network. This feature is invariable to the shape and direction of roads and tolerates heavy noise and disturbances. Road extraction based on reference circles is much more noise tolerant and flexible than the previous edge-detection based algorithms. The scheme is able to extract roads reliably from images with complex contents and heavy obstructions, such as the high resolution aerial/satellite images available from Google maps.

Speed sensorless systems are intensively studied during recent years; this is mainly due to their economical benefit and fragility of mechanical sensors and also the difficulty of installing this type of sensor in many applications. These systems suffer from instability problems and sensitivity to parameter mismatch at low speed operation. In this paper an analysis of adaptive observer stability with stator resistance estimation is given.

In this paper we propose new method for simultaneous generating multiple quantiles corresponding to given probability levels from data streams and massive data sets. This method provides a basis for development of single-pass low-storage quantile estimation algorithms, which differ in complexity, storage requirement and accuracy. Abstract:
Physical activity and diet are factors that influence the body's structure. The purpose of this study was to compare the effects of four weeks of resistance training, and glutamine supplement consumption on growth hormone (GH), and Insulin-like growth factor 1 (IGF-1) Axis. 40 amateur male bodybuilders, participated in this study. They were randomly divided into four equal groups, Resistance (R), Glutamine (G), Resistance with Glutamine (RG), and Control (C). The R group was assigned to a four week resistance training program, three times/week, three sets of 10 exercises with 6-10 repetitions, at the 80-95% 1RM (One Repetition Maximum), with 120 seconds rest between sets), G group is consuming l-glutamine (0.1 g/kg-1/day-1), RG group resistance training with consuming L-glutamine, and C group continued their normal lifestyle without exercise training. GH, IGF1, IGFBP-III plasma levels were measured before and after the protocol. One-way ANOVA indicated significant change in GH, IGF, and IGFBP-III between the four groups, and the Tukey test demonstrated significant increase in GH, IGF1, IGFBP-III plasma levels in R, and RG group. Based upon these findings, we concluded that resistance training at 80-95% 1RM intensity, and resistance training along with oral glutamine shows significantly increase secretion of GH, IGF-1, and IGFBP-III in amateur males, but the addition of oral glutamine to the exercise program did not show significant difference in GH, IGF-1, and IGFBP-III.

Abstract:
Urbanization remains one of the unique predominant factors which is linked to the destruction of urban environment and its associated cases of soil contamination by heavy metals through the natural and anthropogenic activities. These activities are important sources of toxic heavy metals such as arsenic (As), cadmium (Cd), chromium (Cr), copper (Cu), iron (Fe), manganese (Mn), and lead (Pb), nickel (Ni) and zinc (Zn). Often, these heavy metals lead to increased levels in some areas due to the impact of atmospheric deposition caused by their proximity to industrial plants or the indiscriminately burning of substances. Information gathered on potentially hazardous levels of these heavy metals in soils leads to establish serious health and urban agriculture implications. However, characterization of spatial variations of soil contamination by heavy metals in Ghana is limited. Kumasi is a Metropolitan city in Ghana, West Africa and is challenged with the recent spate of deteriorating soil quality due to rapid economic development and other human activities such as “Galamsey”, illegal mining operations within the metropolis. The paper seeks to use both univariate and multivariate geostatistical techniques to assess the spatial distribution of heavy metals in soils and the potential risk associated with ingestion of sources of soil contamination in the Metropolis. Geostatistical tools have the ability to detect changes in correlation structure and how a good knowledge of the study area can help to explain the different scales of variation detected. To achieve this task, point referenced data on heavy metals measured from topsoil samples in a previous study, were collected at various locations. Linear models of regionalisation and coregionalisation were fitted to all experimental semivariograms to describe the spatial dependence between the topsoil heavy metals at different spatial scales, which led to ordinary kriging and cokriging at unsampled locations and production of risk maps of soil contamination by these heavy metals. Results obtained from both the univariate and multivariate semivariogram models showed strong spatial dependence with range of autocorrelations ranging from 100 to 300 meters. The risk maps produced show strong spatial heterogeneity for almost all the soil heavy metals with extremely risk of contamination found close to areas with commercial and industrial activities. Hence, ongoing pollution interventions should be geared towards these highly risk areas for efficient management of soil contamination to avert further pollution in the metropolis.

Oil leakage from the pipelines and the tanks carrying them, or during oil extraction, could lead to the changes in the characteristics and properties of the soil. In this paper, conducting a series of experimental cyclic triaxial tests, the effects of oil contamination on the liquefaction potential of sandy soils is investigated. The studied specimens are prepared by mixing the Firoozkuh sand with crude oil in 4, 8 and 12 percent by soil dry weight. The results show that the oil contamination up to 8% causes an increase in the soil liquefaction resistance and then with increase in the contamination, the liquefaction resistance decreases.

Abstract:
Fluoroquinolones form the mainstay of therapy for the treatment of infections due to Salmonella enterica subsp. enterica. There is a complex interplay between several resistance mechanisms for quinolones and various fluoroquinolones discs, giving varying results, making detection and interpretation of fluoroquinolone resistance difficult. For detection of fluoroquinolone resistance in Salmonella ssp., we compared the use of pefloxacin and nalidixic acid discs as surrogate marker. Using MIC for ciprofloxacin as the gold standard, 43.5% of strains showed MIC as ≥1 μg/ml and were thus resistant to fluoroquinoloes. Based on the performance of nalidixic acid and pefloxacin discs as surrogate marker for ciprofloxacin resistance, both the discs could correctly detect all the resistant phenotypes; however, use of nalidixic acid disc showed false resistance in the majority of the sensitive phenotypes. We have also tested newer antimicrobial agents like cefixime, imipenem, tigecycline and azithromycin against Salmonella spp. Moreover, there was a comeback of susceptibility to older antimicrobials like ampicillin, chloramphenicol, and cotrimoxazole. We can also use cefixime, imipenem, tigecycline and azithromycin in the treatment of multidrug resistant S. typhi due to their high susceptibility.

Abstract:
Memristor is also known as the fourth fundamental passive circuit element. When current flows in one direction through the device, the electrical resistance increases and when current flows in the opposite direction, the resistance decreases. When the current is stopped, the component retains the last resistance that it had, and when the flow of charge starts again, the resistance of the circuit will be what it was when it was last active. It behaves as a nonlinear resistor with memory. Recently memristors have generated wide research interest and have found many applications. In this paper we survey the various applications of memristors which include non volatile memory, nanoelectronic memories, computer logic, neuromorphic computer architectures low power remote sensing applications, crossbar latches as transistor replacements, analog computations and switches.Keywords: Memristor, non-volatile memory, arithmatic operation, programmable resistor.

Abstract:
Interest in using bacteria in cement materials due to its positive influences has been increased. Cement materials such as mortar and concrete basically suffer from higher porosity and water absorption compared to other building materials such as steel materials. Because of the negative side-effects of certain chemical techniques, biological methods have been proposed as a desired and environmentally friendly strategy for reducing concrete porosity and diminishing water absorption. This paper presents the results of an experimental investigation carried out to evaluate the influence of Sporosarcina pasteurii bacteria on the behaviour of two types of concretes (light weight aggregate concrete and normal weight concrete). The resistance of specimens to water penetration by testing water absorption and evaluating the electrical resistance of those concretes was examined and compared. As a conclusion, 20% increase in electrical resistance and 10% reduction in water absorption of lightweight aggregate concrete (LWAC) and for normal concrete the results show 7% decrease in water absorption and almost 10% increase in electrical resistance.

Abstract:
Microplasma welding is a less expensive alternative to laser welding in dental technology. The aim of the study was to highlight discontinuities present in the microplasma welded joints of dental base metal alloys by visual analysis. Five base metal alloys designated for fixed prostheses manufacture were selected for the experiments. Using these plates, preliminary tests were conducted by microplasma welding in butt joint configuration, without filler material, bilaterally and with filler material, proper for each base metal. Macroscopic visual inspection was performed to assess carefully the irregularities in the welds. Electron microscopy allowed detection of discontinuities that are not visible to the eye and revealing details regarding location, trajectory, morphology and size of discontinuities. Supplementing visual control with microscopic analysis allows to detect small discontinuities, which escapes the macroscopic control and to make a detailed study of the weld.Keywords: base metal alloys, fixed prosthodontics, microplasmawelding, visual inspection
