385 Regulation of PKA-Dependent Calcineurin as a Switch in Cell Secretion

Abstract:
This study will investigate cyclic adenosine monophosphate (cAMP)/protein kinase A (PKA) dependent calcineurin (Cn), known as protein phosphatase 2 B (PP2B) as well, regulation of chloride ion (Cl⁻) secretion and the release of pro-inflammatory molecules in immune cells such as cytokines. THP-1-derived monocytes, primary human monocytes and the bronchial epithelial cell line (16HBE14o-) were used in this study. The 16HBE14o- cells were chosen as positive control. Hence, to further confirm the expression of cystic fibrosis transmembrane conductance regulator (CFTR), calcium binding protein (S100A10), annexin A2 (AnxA2) and calcineurin A subunit (CnA) in all three cell types, cell lysate was probed against corresponding primary antibodies by immunoblotting. Western blot analyses show the expression of CFTR, AnxA2, CnA and S100A10 in THP-1-derived monocytes and primary human monocytes. In conclusion, CFTR, S100A10, CnA and AnxA2 are expressed in THP-1-derived monocytes and primary human monocytes and regulate Cl⁻ secretion. Also, they may play a role in the pro-inflammatory molecules release. Abstract:
In recent decades with the development of technology and lack of food sources, sewage sludge in production of human foods is inevitable. Various sources of municipal and industrial sewage sludge that is produced can provide the requirement of plant nutrients. Soils in arid, semi-arid climate of central Iran that most affected by water drainage, iron and zinc deficiencies, using of sewage sludge is helpful. Therefore, the aim of this study is investigation of sewage sludge and manure application on Ni and Zn uptake by Savory. An experiment in a randomized complete block design with three replications was performed. Sewage sludge treatments consisted of four levels, control, 15, 30, 80 tons per hectares, the manure was used in four levels of control, 20, 40 and 80 tons per hectare. Results showed that the wet and dry weights was not affected by sewage sludge using, while, manure has significant effect on them. The effect of sewage sludge on the cadmium and lead concentrations were significant. Interactions of sewage sludge and manure on dry weight values were not significant. Compare mean analysis showed that increasing the amount of sewage sludge had no significant effect on cadmium concentration and it reduced when sewage sludge usage increased. This is probably due to increased plant growth and reduced concentrations of these elements in the plant.Keywords: savory, lead, cadmium, sewage sludge, manure

Commercially available poly(acrylonitrile-co-vinyl acetate) P(AN-VA) or poly(acrylonitrile-co-methyl acrylate) P(AN-MA) are not satisfactory to meet the demand in flame and fire-resistance. In this work, vinylphosphonic acid is used during polymerization of acrylonitrile, vinyl acetate, methacrylic acid to produce fire-retardant polymers. These phosphorus containing polymers are successfully spun in the form of nanofibers. Abstract:
Underground space is becoming a necessity nowadays, especially in highly congested urban areas. Retaining underground excavations using shoring systems is essential in order to protect adjoining structures from potential damage or collapse. Reinforced Concrete Piles (RCP) supported by multiple rows of tie-back anchors are commonly used type of shoring systems in deep excavations. However, executing anchors can sometimes be challenging because they might illegally trespass neighboring properties or get obstructed by infrastructure and other underground facilities. A technique is proposed in this paper, and it involves the addition of eccentric high-strength steel strands to the RCP section through ducts without providing the pile with lateral supports. The strands are then vertically stressed externally on the pile cap using a hydraulic jack, creating a compressive strengthening force in the concrete section. An experimental study about the behavior of the shoring wall by pre-stressed piles is presented during the execution of an open excavation in an urban area (Beirut city) followed by numerical analysis using finite element software. Based on the experimental results, this technique is proven to be cost-effective and provides flexible and sustainable construction of shoring works.Keywords: deep excavation, prestressing, pre-stressed piles, shoring system

Seismic performance of historical structures damaged by the earthquake effects can be enhanced by repair and retrofitting applications. However, repair and retrofitting applications of historical structures are more complicated compared with the traditional structures. For this reason, they need much more attention in repair and retrofitting applications to preserve the spirit of historical structures. Abstract:
This paper investigates the energy storage technologies that can potentially enhance the use of solar energy. Water electrolysis systems are seen as the principal means of producing a large amount of hydrogen in the future. Starting from the analysis of the models of the system components, a complete simulation model was realized in the Matlab-Simulink environment. Results of the numerical simulations are provided. The operation of electrolysis and photovoltaic array combination is verified at various insulation levels. In the estimation of elastic settlement, the Schmertmann’s method that is a very popular method in the elastic settlement estimation of shallow foundations is employed. In order to consider affect of neighboring footings on elastic settlement of main footing in different configurations, a MATLAB script has been generated. The studies concentrated on the historical background of Van show the fact that Van has had a significant position as a settlement since ancient times and that it has hosted many civilizations during history. With the dominance of Ottoman Empire in 16th century, the region had been re-constructed by building new walls at the southern side of Van Castle. These construction activities had mostly been fulfilled by the usage of adobe which had been a fundamental material for thousands of years. As a result of natural disasters, battles and the move at the threshold of 20th century to the new settlement which is 9 kilometers away from the Ancient City Van is an open-air museum with the ruins of churches, mosques and baths. Abstract:
Citizens, businesses, and public authorities all around the world are becoming aware of the impact that they have on the environment. Currently, climate change is an apparent cause to urge everyone to act and move to sustainable energy solutions. After the Paris Climate Agreement, every country has thought of a way to cut down carbon emissions. The Netherlands formulated the National Climate Agreement. “The government’s central goal with the National Climate Agreement is to reduce greenhouse gas emissions in the Netherlands by 49% compared to 1990 levels. At a European level, the government is advocating a 55% reduction of greenhouse gas emissions by 2030.” [5]. From a survey of the CBS, it is apparent that citizens are not putting in as much effort into the transition to sustainable energy as the government would like them to. After analysing the data, it became clear that the citizens miss the motivation to switch to sustainable energy because they do not believe it is urgent at this point and it is too expensive for them [2]. This needs to be changed. The citizens need to be aware of their impact on the climate and the advantages that this process will bring them. For example, the implementation of smart home displays 4 for real time energy measuring will give the citizens an overview of their energy usage so they are aware of the impact they have. Researchers have also found that the citizens must be included in the decision-making aimed at changing their behaviour [4, 3, 1]. In the future, the government will need to include the citizens when they create campaigns, strategies or introduce new policies [7, 6]. By including and informing the citizens about the policies it will be more attractive for them to choose sustainable energy. Abstract:
The present work highlights some of our up-to-date findings on the effect of vanadium addition on the mechanical properties and microstructure of one of the most versatile aluminum-silicon alloys, i.e., A319. In terms of microstructure, it was found that in addition to its ability to refine some of the constituent phases, vanadium also helps in retarding the formation of some of the detrimental intermetallic compounds, such as those involving Al-Fe-Si. Preliminary studies of the effect of vanadium on the mechanical properties of A319 have shown that vanadium additions up to 0.4% cause slight increase in the yield and tensile strength. Abstract:
The electrodeposition of Zn-Ni alloy is mostly studied for its high degree of corrosion and mechanical properties. In this work, the zinc–nickel alloy coatings elaborated from sulfate bath have been carried out under low and high applied magnetic field. The effect of alloy stuctural parameters upon corrosion behavior is studied. It has been found that the magnetically induced convection changes the phase composition, promoting the zinc phase in spite of the γ-Ni₅Zn₂₁. Low magnetic field acts also on the morphology of the deposits as a levelling agent and a refiner by lowering the deposit roughness Ra and the spot size. For alloy obtained with low magnetic field (up to 1T) superimposition, surface morphology modification has no significant influence on corrosion behavior whereas for low nickel content alloy, the modification of phase composition, induced by applied magnetic field, favours higher polarization resistance. When high magnetic field amplitude is involved (up to12T), the phase composition modifications are the same that for low applied B and the morphology is not largely modified. In this case, the hydrogen reduction current dramatically decreases that leads to a large shift of the corrosion potential. It is suggested that the surface reactivity of electrodeposited alloys depends on the magnetically induced convection that is efficient during the codeposition process.Keywords: magnetic field, Zn-Ni alloy, corrosion, corrosive medium

There is a requirement for registered edible seed pumpkin suitable for eating in Turkey. A total of 81 genotypes collected from the researchers in 2005 originated from Eskisehir, Konya, Nevsehir, Tekirdag, Sakarya, Kayseri and Kirsehir provinces were utilized. The used genetic materials were brought to S5 generation by the research groups among 2006 and 2010 years. In this research, S5 stage reached in the genotype given some of the morphological features, and selection of promising genotypes generated scale were made. Results showed that the A-1 (420), A-7 (410), A-8 (420), A-32 (420), B-17 (410), B-24 (410), B-25 (420), B-33 (400), C-24 (420), C-25 (410), C-26 (410) and C-30 (420) genotypes are expected to be promising varieties.Keywords: candidate cultivar, edible seed pumpkin, morphologic parameters, selection

Historical buildings may face various impacts throughout their life cycle. There have been environmental, structural, public works actions on old monuments influencing sustainability and maintenance issues. As a result, ancient monuments can have been undergone various changes in the context of restoration and repair. Currently, these buildings face integrated conditions including city planning macro solutions, old intervention methods, modifications in building envelope and artefacts in terms of conservation. Moreover, documentation of phases is an essential for assessing the historical building, yet it can result in highly complicated and interwoven issues. Herein, two monuments constructed in the 16th century are selected as case studies in Kocaeli, Turkey which are located in different micro climatic conditions and/or exposed to different interventions and which are important for the city as cultural property. Pertev Paşa Mosque (also known as Yenicuma Mosque) -constructed by Architect Sinan-; Gebze Çoban Mustafa Paşa Mosque -constructed in 1523 and known as the work of Architect Sinan but various names asserted as the architect of building according to resources. Active water infiltration and damages, recent material interventions, hidden niches, and foundation techniques of the mosque are investigated via Infrared Thermography under the project of 114K284, “Non-Destructive Test Applications, in the Context of Planned Conservation, through Historical Mosques of Kocaeli: Coban Mustafa Pasa Mosque, Fevziye Mosque and Pertev Pasa Mosque” funded by TUBITAK. It is aimed to reveal active deteriorations on building elements generated by unwanted effects of structural and climatic conditions, historical interventions, and modifications by monitoring the variation of surface temperature and humidity by IRT visualization method which is an important non- destructive process for investigation of monuments in the conservation field in the context of planned conservation. It is also concluded that in-situ monitoring process via IRT through different climatic conditions give substantial information on the behaviour of the envelope to the physical environmental conditions by observation of thermal performance, degradations. Masonry buildings constitute major part of building stock in Turkey. Masonry buildings were built up especially in rural areas and underdeveloped regions due to economic reasons. Almost all of these masonry buildings are not designed and detailed according to any design guidelines by designers. As a result of this, masonry buildings were totally collapsed or heavily damaged when subjected to destructive earthquake effects. Thus, these masonry buildings that were built up in our country must be retrofitted to improve their seismic performance. The main objective of this research is to optimize the power produced by a solar chimney wind turbine. The cut out speed and the maximum possible production are considered while performing the optimization. Solar chimney is one of the solar technologies that can be used in rural areas at cheap cost. With over 50% of rural areas still yet to have access to electricity. The OptimTool in MATLAB is used to maximize power produced by the turbine subject to certain constraints. The results show that an optimized turbine produces about ten times the power of the normal turbine which is 111 W/h. Van province which has a very rich historical heritage is located in eastern part of Turkey, between Lake Van and the Iranian border. Many civilizations prevailing in Van until today have built up many historical structures such as castles, mosques, churches, bridges, baths, etc. In 2011, a devastating earthquake with magnitude 7.2 Mw, epicenter in Tabanlı Village, occurred in Van, where a large part of the city locates in the first-degree earthquake zone. As a result of this earthquake, 644 people were killed; a lot of reinforced, unreinforced and historical structures were badly damaged. Many historical structures damaged due to this earthquake have been restored. In the standards of IEC 60076-2 and IEC 60076-7, three different hot-spot temperature estimation methods are suggested. In this study, the algorithms which used in hot-spot temperature calculations are analyzed by comparing the algorithms with the results of an experimental set-up made by a Transformer Monitoring System (TMS) in use. In tested system, TMS uses only top oil temperature and load ratio for hot-spot temperature calculation. And also, it uses some constants from standards which are on agreed statements tables. Abstract:
The dimensional stability and static bending properties of epoxy composite reinforced with amorphous and crystalline silica were investigated. The amorphous and crystalline silica was obtained by the precipitation method from carbonisation process of the rice husk at a temperature of 600 °C and 1000 °C for 7 hours respectively. The epoxy resin was mixed with 5%, 10% and 15% concentrations of amorphous and crystalline silica. The mixture was stirred for 10 minutes and cured at 28 °C for 72 hours and oven dried at 80 °C for 72 hours. The scanning electron microscope image showed the silica sized of 10-30nm was obtained. The water absorption and thickness swelling of epoxy/amorphous silica composite was not significantly different with silica concentration ranged from 0.08% to 0.09% and 0.17% to 0.20% respectively. The maximum modulus of rupture (85 MPa) and modulus of elasticity (3284 MPa) were achieved for 10% silica concentration. For epoxy/crystalline silica composite; the water absorption and thickness swelling were also not significantly different with silica concentration, ranged from 0.08% to 0.11% and 0.16% to 0.18% respectively. The maximum modulus of rupture (47.9 MPa) and modulus of elasticity (2760 MPa) were achieved for 10% silica concentration. Overall, the water absorption and thickness swelling were almost identical for epoxy composite made from either amorphous or crystalline silica. In this study, the effect of pre-stressed strand diameters, providing the beam-to-column connections, was investigated from both experimental, and analytical aspects. In the experimental studies, the strength and stiffness, the capacities of the precast specimens were compared. The precast specimen with strands of 15.24 mm reached an equal strength of the reference specimen. Parallel results were obtained during the analytical studies from the aspects of strength, and behavior, but in terms of stiffness, it was seen that the initial stiffness of the analytical models was lower than that of the tested specimen.Keywords: post-tensioned connections, beam to column connections, finite element method, strand diameter

In this work, radio frequency (RF) coupling between two HF antennas on a shipboard platform is minimized by determining an optimal antenna placement. Unlike the other works, the coupling is minimized not only at single frequency but over the whole frequency band of operation. Similarly, GAO and PSO, are used in order to determine optimal antenna placement. Throughout this work, outputs of two optimization techniques are compared with each other in terms of antenna placements and coupling results. The purpose of the research was to study the relationship between self-esteem, self-Efficacy with psychological distress in the high school teachers. A total of 245 teachers (92 male and 153 female) in the high school of Kabul and Mazar-e-Sharif from Afghanistan completed inventories General Self-Efficacy, Rosenberg Self-Esteem Scale, and General Health Questionnaire-12 and that assessed their Self-Efficacy, self-esteem with psychological distress. Correlational analysis showed that self-efficacy and self-esteem were significantly and positively correlated with each other. The results of the study indicated that psychological distress is negatively related to self-esteem, and self-efficacy. Abstract:
The studies concentrated on the historical background of Van city show the fact that Van city has had a significant position as a settlement since ancient times and that it has hosted many civilizations during history. With the dominance of Ottoman Empire in 16th century, the region had been re-constructed by building new walls at the southern side of Van Castle. These construction activities had mostly been fulfilled by the usage of mudbrick which had been a fundamental material for thousands of years. As a result of natural disasters, battles and the move at the threshold of 20th century to the new settlement which is 9 kilometers away from the Ancient City Van is an open-air museum with the ruins of churches, mosques and baths. In this study, the usage of mudbrick from past till today in historical structures of Van city is evaluated in detail.Keywords: historical structures, history, mudbrick, Van city

Abstract:
Malware-detecting approaches suffer many limitations, due to which all anti-malware solutions have failed to be reliable enough for detecting zero-day malware. Signature-based solutions depend upon the signatures that can be generated only when malware surfaces at least once in the cyber world. Another approach that works by detecting the anomalies caused in the environment can easily be defeated by diligently and intelligently written malware. Solutions that have been trained to observe the behavior for detecting malicious files have failed to cater to the malware capable of detecting the sandboxed or protected environment. Machine learning and deep learning-based approaches greatly suffer in training their models with either an imbalanced dataset or an inadequate number of samples. AI-based anti-malware solutions that have been trained with enough samples targeted a selected feature vector, thus ignoring the input of leftover features in the maliciousness of malware just to cope with the lack of underlying hardware processing power. Our research focuses on producing an anti-malware solution for detecting malicious PE files by circumventing the earlier-mentioned shortcomings. Our proposed framework, which is based on automated feature engineering through autoencoders, trains the model over a fairly large dataset. It focuses on the visual patterns of malware samples to automatically extract the meaningful part of the visual pattern. Our experiment has successfully produced a state-of-the-art accuracy of 99.54 % over test data.Keywords: malware, auto encoders, automated feature engineering, classification

Abstract:
Islam has been an important topic in Turkey’s institutional identity. Since the dawn of the Turkish Republic, at the end of the First World War, the new Turkish leadership was urged to deal with the religious heritage of the Sultanate. Mustafa Kemal Ataturk, Turkey’s first President, led the country in a process of internal change, substantially modifying not merely the democratic stance of it, but also the way politics was addressing the Muslim faith. Islam was banned from the public sector of the society and was drastically marginalized to the mere private sphere of citizens’ lives. Headscarves were banned from institutional buildings together with any other religious practice, while the country was proceeding down a path of secularism and Westernization. This issue is demonstrated by the fact that even a new elected Prime Minister, Recep Tayyip Erdoğan, was initially barred from taking the institutional position, because of allegations that he had read a religious text while campaigning. Over the years, thanks to this initial internal shift, Turkey has often been seen by Western partners as one of the few countries that had managed to find a perfect balance between a democratic stance and an Islamic inherent nature. In the early 2000s, this led many academics to believe that Ankara could eventually have become the next European capital. Since then, the internal and external landscape of Turkey has drastically changed. Today, religion has returned to be an important point of reference for Turkish politics, considering also the failure of the European negotiations and the always more unstable external environment of the country. This paper wants to address this issue, looking at the important role religion has covered in the Turkish society and the way it has been politicized since the early years of the Republic. It will evolve from a more theoretical debate on secularism and the path of political westernization of Turkey under Ataturk’s rule to a more practical analysis of today’s situation, passing through the failure of Ankara’s accession into the EU and the current tense political relation with its traditional NATO allies. The final objective of this research, therefore, is not to offer a meticulous opinion on Turkey’s current international stance. This issue will be left entirely to the personal consideration of the reader. Rather, it will supplement the existing literature with a comprehensive and more structured analysis on the role Islam has played on Turkish politics since the early 1920s up until the political domestic revolution of the early 2000s, after the first electoral win of the Justice and Development Party (AKP).Keywords: democracy, Islam, Mustafa Kemal Atatürk, Recep Tayyip Erdoğan, Turkey

Abstract:
In this paper we present a quick technique to measure the similarity between binary images. The technique is based on a probabilistic mapping approach and is fast because only a minute percentage of the image pixels need to be compared to measure the similarity, and not the whole image. We exploit the power of the Probabilistic Matching Model for Binary Images (PMMBI) to arrive at an estimate of the similarity. We show that the estimate is a good approximation of the actual value, and the quality of the estimate can be improved further with increased image mappings. Furthermore, the technique is image size invariant; the similarity between big images can be measured as fast as that for small images. For the systems like sliding door, designers should predict not only strength but also dynamic behavior of the system and this prediction usually becomes more critical if design has radical changes refer to previous designs. Also, sometimes physical tests could cost more than expected, especially for rail geometry changes, since this geometry affects design of the body. The aim of the study is to observe and understand the dynamics of the sliding door in virtual environment. For this, multibody dynamic model of the sliding door was built and then affects of various parameters like rail geometry, roller diameters, or center of mass detected.
