52 Emperical Correlation for Measurement of Thermal Diffusivity of Spherical Shaped Food Products under Forced Convection Environment

Abstract:
The present work is the development of an experimental method for determining the thermal diffusivity variations with temperature of selected regular shaped solid fruits and vegetables subjected to forced convection cooling. Experimental investigations were carried on the sample chosen (potato and brinjal), which is approximately of spherical geometry. The variation of temperature within the food product is measured at several locations from centre to skin, under forced convection environment using a deep freezer, maintained at -10°C.This method uses one dimensional Fourier equation applied to regular shapes. For this, the experimental temperature data obtained from cylindrical and spherical shaped products during pre-cooling was utilised. Such temperature and thermal diffusivity profiles can be readily used with other information such as degradation rate, etc. to evaluate thermal treatments based on cold air cooling methods for storage of perishable food products.Keywords: thermal diffusivity, skin temperature, precooling, forced convection, regular shaped

Abstract:
Potato (Solanum tuberosum L.) is ranked among the top leading staple foods in the world. Salinity adversely affects potato crop yield and quality. Therefore, increased level of salt tolerance is a key factor to ensure high yield. The present study focused on the Agrobacterium-mediated transformation of Atriplex canescens betaine aldehyde dehydrogenase (BADH) gene, using single, double and triple CAMV35s promoter to improve salt tolerance in potato. Detection of seven potato lines harboring BADH gene, followed by identification of T-DNA insertions, determination of transgenes copies no through Southern Hybridization and quantification of BADH protein through Enzyme Linked Immunosorbent Assay were considered in this study. The results clearly depict that the salt tolerance of potato was found to be promoter-dependent, as the potato transgenic lines with triple promoter showed 4.4 times more glycine betaine production which consequently leads towards high resistance to salt stress as compared to transgenic potato lines with single and double promoters having least production of glycine betaine. Moreover, triple promoter transgenic potato lines have also shown lower levels of H2O2, malondialdehyde (MDA), relative electrical conductivity, high proline and chlorophyll content as compared other two lines having a single and double promoter. Insilco analysis also confirmed that Atriplex canescens BADH has the tendency to interact with sodium ions and water molecules. Taken together these facts it can be concluded that over-expression of BADH under triple CAMV35s promoter with more glycine betaine, chlorophyll & MDA contents, high relative quantities of other metabolites results in an enhanced level of salt tolerance in potato.Keywords: Atriplex canescens, BADH, CAMV35s promotor, potato, Solanum tubersum

Abstract:
The prevalence and burden of diabetes are rapidly increasing globally, stemming from changes in lifestyle and dietary habits. Although several drugs are available to treat type 2 diabetes mellitus (T2DM), many are accompanied by several side effects and are often costly. Solanum anguivi Lam. fruits (SALF) are bitter berries that commonly grow in the wild and are traditionally cultivated by many globally as a remedy for T2DM. This effect is likely attributable to the presence of bioactive compounds such as phenolics, flavonoids, saponins, alkaloids, and vitamin C in SALF. In this study, we investigated the morphological characteristics of different SALF accessions and the effect of ripeness stages and thermal treatments on the bioactive compounds contents (BCC) and antioxidant activity (AA) of SALF accessions. Using the fruit fly Drosophila melanogaster (D. melanogaster) model, we explored the potential impact of dietary SALF in preventing and treating T2DM phenotypes. Morphological characterization was conducted based on descriptors of Solanum species. The BCC and AA of SALF at different ripeness stages (unripe, yellow, orange, and red) and after thermal treatments were determined using spectrophotometry, HPLC, and gravimetry. Male and female fruit flies were fed a high-sugar diet (HSD) to induce a T2DM-like phenotype, while control flies were fed on SY10 medium for up to 24 days. Experimental flies were exposed to HSD supplemented with 5 or 10 mg/ml SALF. The therapeutic and prevention effect of SALF in T2DM-like phenotype was investigated on weight, climbing activity, glucose and triglyceride contents, survival, and gene expression of PPARγ co-activator 1α fly homolog Srl and Drosophila insulin-like peptides. Methods in fly studies included Gustatory assay, Climbing assay, Glucose GOD-PAP assay, Triglyceride GPO-PAP assay, Roti-Quant®, and Real Time-PCR analysis. The ripeness stage significantly influenced SALF BCC and AA, and this was dependent on the accession. The unripe stage had the highest AA and total phenolics and flavonoids; the orange stage was rich in saponins, while the red stage had the highest alkaloid contents. Boiling and steaming increased the total phenolics and AA up to 4-fold and 3-fold, respectively. Drying at low temperatures resulted in higher phenolics and AA than the control. In the therapeutic model, the HSD-fed female flies exhibited elevated glucose levels, which exhibited a dose-dependent reduction upon exposure to a SALF-supplemented diet. Female flies fed on a SALF+ HSD exhibited a significant increase in survival compared to HSD-fed and control diet-fed flies. SALF supplementation did not alter the weights, fitness, and triglyceride levels of female flies in comparison with HSD-only-fed flies. The mRNA levels of Srl decreased in HSD-fed flies compared to the control-fed, with no effect observed in females exposed to HSD+SALF. Similarly, in the preventative model, the SALF diet resulted in higher survival of supplemented flies compared to controls. Consumption of boiled unripe SALF may result in the highest health benefits due to the high phenolic contents and antioxidant activity observed. Dietary intake of SALF significantly lowered glucose levels and increased survival of the D. melanogaster model. Abstract:
Fusarium Crown and Root Rot (FCRR) caused by Fusarium oxysporum f. sp. radicis-lycopersici (FORL) is a serious tomato (Solanum lycopersicum L.) disease in Tunisia. Its management is very difficult due to the long survival of its resting structures and to the luck of genetic resistance. In this work, we explored the wild Solanaceae species Withania somnifera, growing in the Tunisian Centre-East, as a potential source of biocontrol agents effective in FCRR suppression and tomato growth promotion. Seven fungal isolates were shown able to colonize tomato roots, crowns, and stems. Used as conidial suspensions or cell-free culture filtrates, all tested fungal treatments significantly enhanced tomato growth parameters by 21.5-90.3% over FORL-free control and by 27.6-93.5% over pathogen-inoculated control. All treatments significantly decreased the leaf and root damage index by 28.5-92.8 and the vascular browning extent 9.7-86.4% over FORL-inoculated and untreated control. The highest disease suppression ability (decrease by 86.4-92.8% in FCRR severity) over pathogen-inoculated control and by 81.3-88.8 over hymexazol-treated control) was expressed by I6 based treatments. This endophytic fungus was morphologically characterized and identified using rDNA sequencing gene as Fusarium sp. I6 (MG835371). This fungus was shown able to reduce FORL radial growth by 58.5–83.2% using its conidial suspension or cell-free culture filtrate. Fusarium sp. I6 showed chitinolytic, proteolytic and amylase activities. The current study clearly demonstrated that Fusarium sp. (I6) is a promising biocontrol candidate for suppressing FCRR severity and promoting tomato growth. Abstract:
Chili (Solanum annum L.) attacks by many fungal pathogens, including members of Oomycetes which are responsible for root rot in different chili growing areas of the world. Oomycetes pathogens cause economic losses in different regions of the Pakistan. Most of the plant tissues, including roots, crowns, fruit, and leaves, are vulnerable to Phytophthora capsici. It is very difficult to manage the Phytophthora root rot of chili as many commercial varieties are tremendously vulnerable to P. capsici. The causal agent of the disease was isolated on corn meal agar (CMA) and identified on a morphological basis by using available taxonomic keys. The pathogen was also confirmed on the molecular basis through internal transcribed spacer region and with other molecular markers.The Blastn results showed 100% homology with already reported sequences of P. capsici in NCBI database. Most of the farmers have conventionally relied on foliar fungicide applications to control Phytophthora root rot in spite of their incomplete effectiveness. In this study, in vitro plate assay, seed soaking and foliar applications of 6 fungicides were evaluated against root rot of chili. In vitro assay revealed that significant inhibition of linear growth was obtained with Triflumizole at 7.0%, followed by Thiophanate methyl (8.9%), Etridiazole (6.0%), Propamocarb (5.9%) and 7.5% with Mefenoxam and Iprodione for P. capsici. The promising treatments of in vitro plate bioassay were evaluated in pot experiments under controlled conditions in the greenhouse. All fungicides were applied after at 6-day intervals. Results of pot experiment showed that all treatments considerably inhibited the percentage of P. capsici root rot incidence. In addition, application of seed soaking with all six fungicides combined with the foliar spray of the same components showed the significant reduction in root rot incidence. The combine treatments of all fungicides as in vitro bioassay, seed soaking followed by foliar spray is considered non-harmful control methods which have advantages and limitation. Hence, these applications proved effective and harmless for the management of soil-borne plant pathogens.Keywords: blastn, bioassay, corn meal agar(CMA), oomycetes

Abstract:
The concept of organic agriculture has been accepted as novelty in Indian society, but there is no data available on the human pathogens colonizing plant parts due to such practices. Also, the pattern and mechanism of their colonization need to be understood in order to devise possible strategies for their prevention. In the present study, human pathogenic bacteria were isolated from organically grown tomato plants and five of them were identified as Klebsiella pneumoniae, Enterobacter ludwigii, Serratia fonticola, Stenotrophomonas maltophilia and Chryseobacterium jejuense. Tomato plants were grown in controlled aseptic conditions with 25±1˚C, 70% humidity and 12 hour L/D photoperiod. Six weeks old plants were divided into 6 groups of 25 plants each and treated as follows: Group 1: K. pneumonia, Group 2: E. ludwigii, Group 3: S. fonticola, Group 4: S. maltophilia, Group 5: C. jejuense, Group 6: Sterile distilled water (control). The inoculums for all treatments were prepared by overnight growth with uniform concentration of 108 cells/ml. Leaf samples from above groups were collected at 0.5, 2, 4, 6 and 24 hours post inoculation for the colony forming unit counts (CFU/cm2 of leaf area) of individual pathogens using leaf impression method. These CFU counts were used for the in vivo colonization assay and adherence assay of individual pathogens. Also, resistance of these pathogens to at least 12 antibiotics was studied. Based on these findings S. fonticola was found to be most prominently colonizing the phylloplane of tomato and was further studied. Tomato plants grown in controlled aseptic conditions same as mentioned above were divided into 2 groups of 25 plants each and treated as follows: Group 1: S. fonticola, Group 2: Sterile distilled water (control). Leaf samples from above groups were collected at 0, 24, 48, 72 and 96 hours post inoculation and homogenized in suitable buffers for surface and cell wall protein isolation. Protein samples thus obtained were subjected to isocratic SDS-gel electrophoresis and analyzed. It was observed that presence of S. fonticola could induce the expression of at least 3 additional cell wall proteins at different time intervals. Surface proteins also showed variation in the expression pattern at different sampling intervals. Further identification of these proteins by MALDI-MS and bioinformatics tools revealed the gene(s) involved in the interaction of S. fonticola with tomato phylloplane.Keywords: cell wall proteins, human pathogenic bacteria, phylloplane, solanum lycopersicum

Abstract:
Salinity is a major constraint limiting crop productivity. It has been predicted that by the year 2050, more than 50% of the arable land will be affected by salinity. Two similar salinity experiments were conducted in two seasons under greenhouse condition. Six levels of salinity plus control (viz; control, 2, 4, 8, 16, 32 and 64 % NaCl and CaCl2 at 3:1 ratio) were applied in a form of irrigation water in a single factor experiment arranged in a complete block design with 20 replications. Plant growth and yield were negatively affected by salinity treatments especially at the high levels of salinity. For example, our results suggest that the 32 and 64% of NaCl and CaCl2 treatment were too much for the plant to withstand as determined by reduced dry shoot mass, stem diameter and plant height in both seasons. On the other hand, stomatal conductance and chlorophyll content increased with an increased level of salinity.Keywords: growth, salinity, season, yield

Abstract:
Organic farming is a fundamental principle in sustainable agriculture. Preventing excessive contamination of water and soil with pesticides and chemical fertilizers is important in order to produce healthy food. For this purpose, two potato cultivars (Sante and Marfona) and seven levels of fertilizer (non-fertilizer, chemical fertilizer, granulated chicken manure, common manure, compost, vermicompost and tea compost) were evaluated by factorial experiment based on randomized complete block design (RCBD) with three replications. According to the results, the effect of different manure was significant on number of tubers per plant, tuber weight per plant and tuber yield. The highest value of these traits was obtained by using of chicken manure which was significantly superior to other treatments. However, there was no significant difference between the two varieties. According to the results, the use of chicken manure has produced the highest potato yield even in comparison with the use of chemical fertilizer.Keywords: organic farming, organic manure, potato, tuber yield

Potato (Solanum tuberosum L.) is one of the major vegetable crops that are grown world wide because of its economic importance. This experiment investigated the effect of local sands (River Base, Al-Ekader and Karbala) on number and total weight of mini tubers. Statistical analysis revealed that there were no significant differences among sand cultures in number of stem/plant, chlorophyll index and tubers dry weight. River Base sand had the highest plant height (74.9 cm), leaf number/plant number (39.3), leaf area (84.4 dcm2⁄plant), dry weight/plant (26.31), tubers number/plant (8.5), tubers weight/plant (635.53 gm) and potato tuber yields/trove (28.60 kg), whereas the Karbala sand had lower performance. All the characters had positive and significant correlation with yields except the traits number of stem and tuber dry weight.Keywords: correlation, potato, sand culture, yield

Background: Oxidative stress has been an important health problem as itinduceschronic diseases such as cancer, cardiovascular, diabetics, and neurodegenerative disease. Plant source natural antioxidant has gained attention as synthetic antioxidant negatively impact human health. Wild edible plants arecheap source of dietary-medicine in mainly rural communityin south-west Ethiopia and elsewhere the country. Thus, the study aimed to determine total pheneol,flavoinoids, antioxidant, vitamin C, and beta-carotene content from wild edible plants Solanum nigrum L., Vigna membranacea A. Rich, Dioscorea praehensilis Benth., Trilepisium madagascariense D.C.andCleome gynandra L. Methods: Methanol was used to extract samples of oven-dried edible plants. Total phenolic compound (TPC) was determined using a Folin Ciocalteu method, whereas total flavonoid content (TFC) was determined using the Aluminium chloride colorimetric method. By using 2, 2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP) tests, antioxidant activities were evaluated in vitro. Additionally, beta-carotene was assessed using a spectrophotometric technique, whilst vitamin C was determined using a titration approach. Results: Total flavonoid contentranged from 0.85±0.03 to 11.25±0.01 mg CE/g in D. praehensilis Benth. tuber and C. gynandra L, respectively. Total phenolic compounds varied from 0.25±0.06 GAE/g in D. praehensilis Benth tuber to 35.73±2.52 GAE/g in S.nigrum L. leaves. In the DPPH test, the highest antioxidant value (87.65%) was obtained in the S.nigrum L. leaves, whereas the smallest amount of antioxidant (50.12%)was contained in D. praehensilis Benth tuber. Similarly in FRAP assay,D. praehensilis Benth tuber showed the least reducing potential(49.16± 2.13mM Fe2+/100 g)whilst the highest reducing potential was presented in the S.nigrum L. leaves(188.12±1.13 mM Fe2+/100 g). The beta-carotene content was found between 11.81±0.00 mg/100g in D. praehensilis Benth tubers to 34.49±0.95 mg/100g in V. membranacea A. Rich leaves. The concentration of vitamin C ranged from 10.00±0.61 in D. praehensilis Benth tubers to 45±1.80 mg/100g in V. membranacea A. Rich leaves. The results showed that high positive linear correlations between TPC and TFC of WEPs (r=0.828), as well as between FRAP and total phenolic contents (r = 0.943) and FRAP and vitamin C (r= 0.928). Conclusion: These findings showed the total phenolic and flavonoid contents of Solanum nigrum L. and Cleome gynandra L, respectively, are abundant. The outcome may be used as a natural supply of dietary antioxidants, which may be useful in preventing oxidative stress. The study's findings also showed that Vigna membranacea A. Rich leaves were cheap source of vitamin C and beta-carotene for people who consumed these wild green. Additional research on the in vivo antioxidant activity, toxicological analysis, and promotion of these wild food plants for agricultural production should be taken into consideration.Keywords: antioxidant activity, beta-carotene, flavonoids, phenolic content, and vitamin c

Abstract:
Interactive effects of cucurbitacin-containing phytonematicides and biocontrol agents on growth and nematode suppression on tomato (Solanum lycopersicum) had not been documented. The objective of this study was to determine the interactive effects of Nemafric-BL phytonematicide, Trichoderma harzianum and Steinernema feltiae on growth of tomato plants and suppression of root-knot (Meloidogyne species) nematodes. A 2x2x2 trial was conducted using tomato cv. ‘HTX’ on a field infested with Meloidogyne species. The treatments were applied at commercial rates. At 56 days after treatments, interactions were significant (P ≤ 0.05) for selected plant variables, without significant interactions on nematode variables. In conclusion, results of the current study did not support the combination of the test products for nematode suppression, except that some combinations improved plant growth.Keywords: cucumis africanus, cucurbitacin b, ethnobotanicals, entomopathogenic nematodes, natural enemies, plant extracts

Small Heat Shock Proteins (sHSPs) are low molecular weight chaperones that play an important role during stress response and development in all living organisms. Fruit maturation and oxidative stress can induce sHSP synthesis both in Arabidopsis and tomato plants. RNA-Seq technology is becoming widely used in various transcriptomics studies; however, analyzing and interpreting the RNA-Seq data face serious challenges. In the present work, we de novo assembled the Solanum lycopersicum transcriptome for three different maturation stages (mature green, breaker and red ripe). Differential gene expression analysis was carried out during tomato fruit development. We identified 12 sHSPs differentially expressed that might be involved in breaker and red ripe fruit maturation. Interestingly, these sHSPs have different subcellular localization and suggest a complex regulation of the fruit maturation network process.Keywords: sHSPs, maturation, tomato, RNA-Seq, assembly

Abstract:
Fusarium solani and its variants cause root and stem rot of plants. Dry rot is the most common disease of potato tubers during storage. The causative agents of fusariosis in contact with plants behave as antagonists, growth stimulants or parasites. The diversity of host-parasite relationships is explained by the parasite’s ability to produce a wide spectrum of biologically active compounds including toxins, enzymes, oligosaccharides, antibiotic substances, enniatins and gibberellins. Many of these metabolites contribute to the creation of compatible relations; others behave as elicitors, inducing various protective responses in plants. An important part of the strategy for developing plant resistance against pathogens is the activation of protein synthesis to produce protective ‘pathogenesis-related’ proteins. The family of PR-proteins known to confer the most protective response is chitinases (EC 3.2.1.14, Cht) and β-1,3-glucanases (EC 3.2.1.39, Glu). PR-proteins also include a large multigene family of peroxidases (EC 1.11.1.7, Pod), and increased activity of Pod and expression of the Pod genes leads to the development of resistance to a broad class of pathogens. Despite intensive research on the role of PR-proteins, the question of their participation in the mechanisms of formation of the F.solani–S.tuberosum pathosуstem is not sufficiently studied. Our aim was to investigate the effect of different classes of F. solani metabolites on the activity of chitinase, β-1,3-glucanases and peroxidases in tubers of Solanum tuberosum. Metabolite culture filtrate (CF) and cytoplasmic components were fractionated by extraction of the mycelium with organic solvents, salting out techniques, dialysis, column chromatography and ultrafiltration. Protein, lipid, carbohydrate and polyphenolic fractions of fungal metabolites were derived. Using enzymatic hydrolysis we obtained oligo glycans from fungal cell walls with different molecular weights. The activity of the metabolites was tested using potato tuber discs (d = 16mm, h = 5mm). The activity of PR-proteins of tubers was analyzed in a time course of 2–24 hours. The involvement of the analysed metabolites in the modulation of both early non-specific and late related to pathogenesis reactions was demonstrated. The most effective inducer was isolated from the CF (fraction of total phenolic compounds including naphtazarins). Induction of PR-activity by this fraction was: chitinase - 340-360%, glucanase - 435-450%, soluble forms of peroxidase - 400-560%, related forms of peroxidase - 215-237%. High-inducing activity was observed by the chloroform and acetonitrile extracts of the mycelium (induction of chitinase and glucanase activity was 176-240%, of soluble and bound forms of peroxidase - 190-400%). The fraction of oligo glycans mycelium cell walls of 1.2 kDa induced chitinase and β-1,3-glucanase to 239-320%; soluble forms and related peroxidase to 198-426%. Oligo glycans cell walls of 5-10 kDa had a weak suppressor effect - chitinase (21-25%) and glucanase (25-28%) activity; had no effect on soluble forms of peroxidase, but induced to 250-270% activity related forms. Abstract:
Cucurbitacin-containing phytonematicides had been consistent in suppressing root-knot (Meloidogyne species) when used in dried crude form, with limited evidence whether the efficacy could be affected when fresh fruits were used during fermentation. The objective of this study was to determine the influence of Nemafric-BL phytonematicide prepared using fermented crude extracts of fresh fruit from wild watermelon (Cucumis africanus) on the growth of tomato (Solanum lycopersicum) plants and suppression of Meloidogyne species. Seedlings of tomato cultivar ‘Floradade’ were inoculated with 3 000 eggs and second-stage juveniles (J2) of M. incognita race 2 in pot trials, with treatments comprising 0, 2, 4, 8, 16, 32 and 64 % Nemafric-BL phytonematicide. At 56 days after inoculation, the phytonematicide reduced eggs and J2 in roots by 84-97%, J2 in soil by 49-96% and total nematodes by 70-97%. Plant variables and concentrations of Nemafric-BL phytonematicide exhibited positive quadratic relations, with 74-98% associations. In conclusion, fresh fruit of C. africanus could be used for the preparation of Nemafric-BL phytonematicide, particularly in cases where the dry infrastructure is not available.Keywords: Cucurbitacin B, density-dependent growth, effective microorganisms, quadratic relations

Abstract:
The influence of two PGPR strains, Pseudomonas aeruginosa and Stenotrophomonas rhizophilia, on fruit yields, pomological traits and chemical contents of tomato (Solanum lycopersicum) fruits were studied. The study was conducted separately on two different cultivar varieties of tomato, namely Supermarmande and UC82B. The results indicated that the presence of the PGPR almost doubled the average yield per plant. There was a significant improvement in the pomological qualities of the PGPR treated tomato fruits compared to the corresponding healthy treatments especially in traits such as the average fruit weight, height, and fruit volume. The chemical analysis of tomato fruits revealed that the presence of the PGPRs increased the total protein, lycopene, alkalinity and phenol content of the tomato fruits compared to the healthy controls. They had no influence on the reduced sugar, total soluble solids or the titerable acid content of fruits. However their presence reduced the amount of ascorbic acid in tomato fruits compared to the healthy controls.Keywords: PGPR, tomato, fruit quality

Abstract:
Starch is a highly biocompatible, non-toxic, and biodegradable polymer. It is widely used in biomedical applications, including drug delivery systems and tissue engineering scaffolds. Curcumin, a phenolic compound found in the dried root of Curcuma longa, has been used as a nutritional supplement due to its antimicrobial, anti-inflammatory, and antioxidant effects. However, the major problem with ingesting curcumin by itself is its poor bioavailability due to its poor absorption and rapid metabolism. In this study, we report a novel methodology to prepare starch nanoparticles loaded with curcumin. The nanoparticles were synthesized via nanoprecipitation of starch granules extracted from native Andean potatoes (Solanum tuberosum ssp. and Andigena var Huamantanga varieties). The nanoparticles were crosslinked and stabilized by using sodium tripolyphosphate and Tween®80, respectively. The characterization of the nanoparticles loaded with curcumin was assessed by Fourier Transform Infrared Spectroscopy, Dynamic Light Scattering, Zeta potential, and Differential scanning calorimetry. UV-vis spectrophotometry was used to evaluate the loading efficiency and capacity of the samples. One of the main plants cultivated in protected environment is tomato crop, which presents significant growth in its demand, because it is a tasty fruit, rich in nutrients and of high added value, however, poor management of fertilizers induces the process of soil salinization, causing several consequences, from reduced productivity to even soil infertility. These facts are derived from the increased concentration of salts, which hampers the process of water absorption by the plant, resulting in a biochemical and nutritional imbalance in the plant. Thus, this study aimed to investigate the effects of untreated and electromagnetically treated water in salinized soils on physical, physicochemical, and biochemical parameters in tomato fruits. The experiment was conducted at the Faculty of Science and Engineering, Tupã Campus (FCE/UNESP). A randomized complete block design with two types of treated water was adopted, with five different levels of initial salinity (0; 1.5; 2.5; 4; 5.5; 7 dS m⁻¹) by fertigation. Although the effects of salinity on fruit quality parameters are evident, no beneficial effects on increasing or maintaining postharvest quality of fruits whose plants were treated with electromagnetized water were evidenced.Keywords: Solanum lycopersicum, soil salinization, protected environment, fertigation

Wild edible food plants contain a number of organic phytochemical that have been linked to the promotion of good health. These plants used by the local people of Arunachal Pradesh (Northeast India) are found to have high nutritional potential to maintain general balance diet. A study was conducted to evaluate the nutritional potential of five commonly found, unexplored wild food plants namely, Piper pedicellatum C. DC (leaves), Gonostegia hirta (Blume ex Hassk.) Miq. (leaves), Mussaenda roxburghii Hook. f. (leaves), Solanum spirale Roxb. (leaves and fruits) and Cyathea spinulosa Wall. ex Hook. (pith portion and tender rachis) from East Siang District of Arunachal Pradesh Northeast (India) for ascertaining their suitability for utilization as supplementary food. Solanumnigrum L. (Family: Solanaceae), is an important Indian medicinal plant and have been used in various traditional formulations for hepato-protection. It has been reported to contain significant amount of steroidal glycosides such as solamargine and solasonine as well as their aglycone part solasodine. Being important pharmacologically active metabolites of several members of Solanaceae these markers have been attempted various times for their extraction and quantification but separately for glycoside and aglycone part because of their opposite polarity. Here, we propose for the first time simultaneous extraction and quantification of aglycone (solasodine)and glycosides (solamargine and solasonine) inleaves and berries of S.nigrumusing solvent extraction followed by HPTLC analysis. Simultaneous extraction was carried out by sonication in mixture of chloroform and methanol as solvent. The quantification was done using silica gel 60F254HPTLC plates as stationary phase and chloroform: methanol: acetone: 0.5 % ammonia (7: 2.5: 1: 0.4 v/v/v/v) as mobile phaseat 400 nm, after derivatization with an isaldehydesul furic acid reagent. The method was validated as per ICH guideline for calibration, linearity, precision, recovery, robustness, specificity, LOD, and LOQ. The statistical data obtained for validation showed that method can be used routinely for quality control of various solanaceous drugs reported for these markers as well as traditional formulations containing those plants as an ingredient.Keywords: solanumnigrum, solasodine, solamargine, solasonine, quantification

Abstract:
Asparagine is vital amino acid which is required for the development of brain and it regulates the equilibrium of central nervous system. Asparagine is the chief amino acid that forms acrylamide in baked food by reacting with reducing sugars at high temperature ( Millard Reaction i.e. amino acids and sugars give new flavors at high temperature). It can also be a parameter of freshness in fruit juices because on storage of juices at 37°C caused an 87% loss in the total free amino acids and major decrease was recorded in asparagine contents. With this significance of monitoring asparagine, in the present work a biosensor for determining asparagine in fruit juices is developed. For the construction of biosensor L-asparaginase enzyme (0.5 IU) was co-immobilized with phenol red on TEOS chitosan sol-gel plastic disc and fixed on the fiber optic tip. Tip was immersed in a cell having 5ml of substrate and absorption was noted at response time of 5 min with 10-1 - 10-10 M concentrations of asparagine at 538 nm. L-asparaginase was extracted and from Solanum nigrum Asparagine biosensor was applied fruit juices on the monitoring asparagine contents. L-asparagine concentration found to be present in fruit juices like Guava Juice, Apple Juice, Mango Juice, Litchi juice, Strawberry juice, Pineapple juice Lemon juice, and Orange juice. Colloidal metal nanoparticles are widely applied in various areas and have great potential in different biotechnological applications. Their particular properties associated with both the antiseptic, antioxidant and anti aging properties as well as ability to penetrate most of the biological barriers, synergy in the absorption of nutrients and nontoxic to plants. The properties make them potentially useful in the fast and safe production of healthy, certified starting material in the production of plants exposed to many pathogenic microorganisms causing serious diseases, significantly affecting yield and causing the economic losses. In this case it is crucial to provide appropriate conditions for the production, storage and distribution of the plant material. Therefore, the aim of the proposed research was to develop and identify the influence of four colloidal metal nanoparticles on growth and proliferation of in vitro cultures of potato (Solanum tuberosum) - one of the most economically important strategic crops in the world. The research on different varieties of potato was performed by placing the explants of the in vitro cultures on sterile Murashige and Skoog (MS) type medium. The influence of the nanocolloids was evaluated using the MS medium impregnated with the examinated nanoparticles. The vigour of growth and the rate of proliferation was examinated for 6-8 weeks with both night/day-length and temperature over the ranges 8/16 h and 20–22 °C respectively. Potato is a high-potential food security crop in Ethiopia. However, the yield and productivity of the crop have been far below the world average. This is due to several factors, including appropriate agronomic practices, such as time of earthing-up and inflorescence management. A field experiment was conducted at Jimma, Southwest Ethiopia, during 2016/17 under irrigation to determine the effect of time of earthing-up and inflorescence removal on the growth, yield, and quality of potatoes. The treatments consisted of a time of earthing-up (no earthing-up, earthing-up at 15, 30, and 45 days after complete plant emergence) and inflorescence removal (inflorescence removed and not removed). Potato variety (Belete) was used for this experiment. A 2x4 factorial experiment was laid out with three replications. Data collected on the growth, yield, and quality components of potatoes were analyzed using SAS Version 9.3 statistical software. Inflorescence removal affected the majority of the growth and yield parameters, while the time of earthing-up affected all growth, yield, and quality (green tuber number) parameters. Earthing-up at 15 days in combination with inflorescence removal (at 60 days after complete plant emergence) gave better plant growth and maximum tuber yield of the Belete potato variety under irrigated conditions. Since the current research was conducted at one location, in one season, and with one potato cultivar (Belete), it would be advisable to repeat the experiment so as to arrive at a final conclusion and subsequent recommendation.Keywords: Belete, earthing-up, inflorescence, yield

The increased number of weeds in tomato crops highly lower yields. Weed identification with the aim of machine learning is important to carry out site-specific control. The last advances in computer vision are a powerful tool to face the problem. The analysis of RGB (Red, Green, Blue) images through Artificial Neural Networks had been rapidly developed in the past few years, providing new methods for weed classification. The development of the algorithms for crop and weed species classification looks for a real-time classification system using Object Detection algorithms based on Convolutional Neural Networks. The site study was located in commercial corn fields. The classification system has been tested. The procedure can detect and classify weed seedlings in tomato fields. The input to the Neural Network was a set of 10,000 RGB images with a natural infestation of Cyperus rotundus l., Echinochloa crus galli L., Setaria italica L., Portulaca oeracea L., and Solanum nigrum L. The validation process was done with a random selection of RGB images containing the aforementioned species. The mean average precision (mAP) was established as the metric for object detection. The results showed agreements higher than 95 %. The system will provide the input for an online spraying system. Thus, this work plays an important role in Site Specific Weed Management by reducing herbicide use in a single step.Keywords: deep learning, object detection, cnn, tomato, weeds

Early blight caused by Alternaria solani Sorauer is one of the most serious foliage diseases of the potato (Solanum tuberosum L.). This disease causes huge crop losses and has major economic importance worldwide. The antifungal activity for three medicinal plants (Foeniculum vulgare, Syzygium aromaticum, and Eucalyptus citriodora) against Alternaria solani has been evaluated. The inhibitory potential of selected essential oils on the radial mycelial growth and germination of spore was measured in vitro at various concentrations (5%, 2.5%. 1.25%, 0.625%, and 0.312%) using agar well diffusion assay. Essential oil of E. citriodora was most effective causing 85% inhibition of mycelial growth and 88% inhibition of spore germination at 0.625% and 1.25% concentrations. Essential oil of Foeniculum vulgare also caused 80% and 82% inhibition of the above mentioned parameters but at double the concentrations 1.25% and 2.5%. While essential oil of Syzygium aromaticum was least effective in controlling the mycelial growth and spore germination with 76% and 77% inhibition at 1.25% and 2.5%. All the selected essential oils, especially E. citriodora, showed marked antimicrobial activity significant at higher concentration. These results suggest that the use of essential oils for the control of A. solani can reduce environmental risks related with commercial fungicides, lower cost for control, and the chances for resistance development. Additional studies are essential to evaluate the potential of essential oils as natural treatments for this disease.Keywords: clove, essential oils, fennel, potato

Salinity stress is one of the major abiotic stresses, restricting plant growth and crop productivity in different world regions, especially in arid and semi-arid regions, including Saudi Arabia. The tomato plant is proven to be moderately sensitive to salt stress. Therefore, two field experiments were conducted using tomato plants (Hybrid 6130) to evaluate the effect of four concentrations of salicylic acid (SA; 0, 20, 40, and 60 µM) applied as foliar spraying in improving plant tolerance to saline soil conditions. Tomato plant growth, yield, osmoprotectants, chloeophyll fluorescence, and ionic contents were determined. The results of this study displayed that growth and yield components and physiological attributes of water-sprayed plants (the control) grown under saline soil conditions were negatively impacted. However, under the adverse conditions of salinity, SA-treated plants had enhanced growth and yield components of tomato plants compared to the control. Free proline, soluble sugars, chlorophyll fluorescence, relative water content, membrane stability index, and nutrients contents (e.g., N, P, K⁺, and Ca²⁺) were also improved significantly, while Na⁺ content was significantly reduced in SA-applied tomato plants. SA at 40 µM was the best treatment, which could be recommended to use for salt-stressed tomato plants to enable them to tolerate the adverse conditions of saline soils.Keywords: tomatoes, salt stress, chlorophyll fluorescence, dehydration tolerance, osmoprotectants

This study, in which crispy salad plants was cultivated between the two rows of tomato, was conducted in Spring 2007 in a research glasshouse at Akdeniz University. Crispy salad (Lactuca sativa var. crispa cv. Bohemia) plants were intercropped with tomato (Solanum lycopersicon cv. Selin F1) plants as the main crop. Tomato seedlings were planted according to double line plantation system with 100 cm large spacing, 50 cm narrow spacing and 50 cm within row plant spacing. In both control and intercropping applications, each plot was 9.75 m2 according to plantation distances and there were 26 plants per each plot for tomato. Crispy salad seedlings were planted with 30 cm spacing as one row in the middle of tomato plants and with 30x30 spacing as two rows between plants rows. Moreover, salad seedlings were transplanted between tomato plants above the tomato rows that were planted in two rows with intervals of 50 cm and also with 25x25 cm spacing as the third row in the middle of tomato rows. While tomato plants were growing during the research, fruit width and height were measured periodically with 15 days in the tomato fruits of the third cluster from the formation of fruit to fruit ripening. According to results, while there were no differences between cropping systems in terms of fruit width, the highest fruit height was found in Control trial in the first measurement. In the second measurement while the highest fruit width was determined with 64.39 mm in Control, there were no differences between cropping systems. In the third measurement, the highest fruit width and height were obtained from Control with 68.47 mm and 55.52 mm, respectively. As a conclusion the trial, which crispy salad seedlings were planted with 30x30 cm spacing as two rows between tomato plants rows, was determined as a best intercropping application.Keywords: crispy salad, glasshouse, intercropping, tomato

In this study, the spatial variation of plant and soil nutrition contents of tomato plants grown in greenhouses was investigated in Elmalı region of Antalya. For this purpose, total of 19 sampling points were determined. Coordinates of each sampling points were recorded by using a hand-held GPS device and were transferred to satellite data in GIS. Soil samples were collected from two different depths, 0-20 and 20-40 cm, and leaf were taken from different tomato greenhouses. The soil and plant samples were analyzed for N, P and K. Then, attribute tables were created with the analyses results by using GIS. Data were analyzed and semivariogram models and parameters (nugget, sill and range) of variables were determined by using GIS software. Kriged maps of variables were created by using nugget, sill and range values with geostatistical extension of ArcGIS software. Kriged maps of the N, P and K contents of plant and soil samples showed patchy or a relatively smooth distribution in the study areas. As a result, the N content of plants were sufficient approximately 66% portion of the tomato productions. It was determined that the P and K contents were sufficient of 70% and 80% portion of the areas, respectively. On the other hand, soil total K contents were generally adequate and available N and P contents were found to be highly good enough in two depths (0-20 and 20-40 cm) 90% portion of the areas.Keywords: Elmali, nutrients, springs greenhouses, spatial variation, tomato

Crop fertilizer recommendations are outdated as these are based on the response trails conducted over half a century ago. Further, these recommendations were based on the response trials conducted over large geographical area ignoring the large spatial variability in indigenous nutrient supplying capacity of soils typical of most smallholder systems. Application of fertilizer following such blanket recommendation in fields with varying native nutrient supply capacity leads to under application in some places and over application in others leading to reduced nutrient-use-efficiency (NUE), loss of profitability, and increased environmental risks associated with loss of unutilized nutrient through emissions or leaching. Opportunities exist to further increase yield and profitability through a significant gain in fertilizer use efficiency with commercialization of affordable and precise application technologies. We conducted participatory trails in Maize (Zea Mays), Cauliflower (Brassica oleracea var. botrytis) and Tomato (Solanum lycopersicum) in Mid Hills of Nepal to evaluate the efficacy of Urea Deep Placement (UDP and Polymer Coated Urea (PCU);. UDP contains 46% of N having individual briquette size 2.7 gm each and PCU contains 44% of N . Both PCU and urea briquette applied at reduced amount (100 kg N/ha) during planting produced similar yields (p>0.05) compared with regular urea (200 Kg N/ha). . These fertilizers also reduced N fertilizer by 35 - 50% over government blanket recommendations. Pectobacterium and Dickeya spp which negatively affect a wide range of crops are the main causes of the aggressive diseases of agricultural crops. These aggressive diseases are responsible for a huge economic loss in agriculture including a severe decrease in the quality of the stored vegetables and fruits. Therefore, it is important to detect these pathogenic bacteria at their early stages of infection to control their spread and consequently reduce the economic losses. In addition, early detection is vital for producing non-infected propagative material for future generations. The currently used molecular techniques for the identification of these bacteria at the strain level are expensive and laborious. Other techniques require a long time of ~48 h for detection. Thus, there is a clear need for rapid, non-expensive, accurate and reliable techniques for early detection of these bacteria. In this study, infrared spectroscopy, which is a well-known technique with all its features, was used for rapid detection of Pectobacterium and Dickeya spp. at the strain level. The bacteria were isolated from potato plants and tubers with soft rot symptoms and measured by infrared spectroscopy. The obtained spectra were analyzed using different machine learning algorithms. The performances of our approach for taxonomic classification among the bacterial samples were evaluated in terms of success rates. A greenhouse and laboratory experiment will be conducted at the University of Sussex biotechnology greenhouse and Agriculture laboratory using a growth chamber to study the effect of GA3 on the growth and development attributes of African nightshade. The experiment consists of three replications and 5 treatments and is laid out in a randomized complete block design consisting of various concentrations of GA3. 0ppm, 50ppm, 100ppm, 150ppm and 200ppm. local farmer seed was grown in plastic pots, 6 seeds then hardening off to remain with four plants per pot at the greenhouse to attain purity of germplasm, proper management until maturity of berries then harvesting and squeezing to get seeds, paper dry on the sun for 7 days. In a laboratory, place 5 Whatman filter paper on glass petri-dish subject to different concentrations of stock solution, count 50 certified and clean, healthy seeds, then arrange on the moist filter paper and mark respectively. Spray with the stock solution twice a day and protrusion of radicle termed as germination count and discard to increase the accuracy of precision. Data will be collected on the application of GA3 to compare synergistic effects on the growth, yield, and nutrient contents on African nightshade.Keywords: African nightshade, growth, yield, shoot, gibberellins
