8724 Simulation of a Cost Model Response Requests for Replication in Data Grid Environment
Authors: Kaddi Mohammed, A. Benatiallah, D. Benatiallah
Abstract:
Data grid is a technology that has full emergence of new challenges, such as the heterogeneity and availability of various resources and geographically distributed, fast data access, minimizing latency and fault tolerance. Researchers interested in this technology address the problems of the various systems related to the industry such as task scheduling, load balancing and replication. The latter is an effective solution to achieve good performance in terms of data access and grid resources and better availability of data cost. In a system with duplication, a coherence protocol is used to impose some degree of synchronization between the various copies and impose some order on updates. In this project, we present an approach for placing replicas to minimize the cost of response of requests to read or write, and we implement our model in a simulation environment. The placement techniques are based on a cost model which depends on several factors, such as bandwidth, data size and storage nodes. Keywords: response time, query, consistency, bandwidth, storage capacity, CERN

Authors: Hammadi Larbi
Abstract:
The treatment of wastewater in sewage plants usually results in the formation of a large amount of sludge. These appear at the outlet of the treatment plant as a viscous fluid loaded with a high concentration of dry matter. This sludge production presents environmental, ecological, and economic risks. That is why it is necessary to find many solutions for minimizing these risks. In the present article, the effect of hydrogen peroxide, thermal treatment, and quicklime on the characteristics of the activated sludge produced in urban wastewater plant were evaluated in order to avoid any risk in the plants. The study shows increasing of the dose of H2O2 from 0 to 0.4 g causes an increase in the solubilization rate of COD from 12% to 45% and a reduction in the organic matter content of sludge (VM/SM) from 74% to 36% . The results also show that the optimum efficiency of the heat treatment corresponds to a temperature of 80 ° C for a treatment time of 40 min is 47% and 51.82% for a temperature equal to 100 ° C and 76.30 % for a temperature of 120 ° C, and 79.38% for a temperature of 140 ° C. The treatment of sludge by quicklime gives the optimum efficiency of 70.62 %. It was shown the increasing of the temperature from 80°C to 140°C, the pH of sludge was increased from 7.12 to 9.59. The obtained results showed that with increasing the dose of quicklime from 0 g/l to 1g/l in activated sludge led to an increase of their pH from 7.12 to 12.06. The study shows the increasing the dose of quicklime from 0 g/l to 1g/l causes also an increase in the solubilization of COD from 0% to 70.62 %Keywords: activated sludge, hydrogen peroxide, thermal treatment, quicklime

Authors: S. C. Santos, S. R. Dionísio, A. L. D. De Andrade, L. R. Roque, A. C. Da Costa, J. L. Ienczak
Abstract:
In this work, two fermentations at different temperatures (25 and 30 ºC), with cell recycling, were accomplished to produce ethanol, using a mix of commercial substrates, xylose (70%) and glucose (30%), as organic source for Scheffersomyces stipitis. Five consecutive fermentations of 80 g L-1 (1º, 2º and 3º recycles), 96 g L-1 (4º recycle) and 120 g L-1 (5º recycle)reduced sugars led to a final maximum ethanol concentration of 17.2 and 34.5 g L-1, at 25 and 30 ºC, respectively. Glucose was the preferred substrate; moreover xylose startup degradation was initiated after a remaining glucose presence in the medium. Results showed that yeast acid treatment, performed before each cycle, provided improvements on cell viability, accompanied by ethanol productivity of 2.16 g L-1 h-1 at 30 ºC. A maximum 36% of xylose was retained in the fermentation medium and after five-cycle fermentation an ethanol yield of 0.43 g ethanol/g sugars was observed. S. stipitis fermentation capacity and tolerance showed better results at 30 ºC with 83.4% of theoretical yield referenced on initial biomass. Keywords: 5-carbon sugar, cell recycling fermenter, mixed sugars, xylose-fermenting yeast

Authors: Massimo Morichi
Abstract:
Over the last five years, starting in 2019, several nuclear measurement systems have been conceived and realized for specific nuclear safeguards applications, as well as for nuclear security, implementing some innovative technologies and methods for attended and unattended nuclear measurements. Some of those technologies imply the integration of combined gamma and neutron detection systems both for counting and spectroscopic applications that allow the SNM (Special Nuclear Material) verification and quantification through specific simultaneous measurements (gamma and neutron) from standard to high count rate due to high flux irradiation. IAEA has implemented some of these technologies in key international safeguards inspections worldwide, like a Fast Neutron Collar Monitor for fresh fuel verification of U235 mass (used during inspections for material declaration verification) or for unattended measuring systems with a distinct shift register installed in an anti-tampering sealed housing in unattended mode (remote inspection and continuous monitoring) together with an Unattended Multichannel Analyzer for spectroscopy analysis of SNM like canisters. Such developments, realized with integrated mid-resolution scintillators (FWHM: <3,5%) together with organic scintillators such as Stilbene detectors or Liquid sealed scintillators like EJ-309 with great pulse shape discrimination managed by a fast DAQ and with a high level of system integration, are offering in the near term the possibility to enhance further their implementation, reducing the form factor in order to facilitate their implementation in many critical parts of the Nuclear Fuel Operations as well as of the Next generation of Nuclear Reactors. This will facilitate embedding these advanced technical solutions in the next generation of nuclear installations, assuring the implementation of the Safeguards by Design requested by IAEA for all future/novel nuclear installations. This work presents the most recent designs/systems and provides some clear examples of ongoing applications on the Fuel Cycle-Fuel Fabrication as well as for the SMR/AMR and microreactors. Detailed technology testing and validation in different configuration if provided together with some case-studies and operational implications. Keywords: nuclear safeguard, gamma and neutron detection systems, spectroscopy analysis, nuclear fuel cycle, nuclear power reactors, decommissioning dismantling, nuclear security
Procedia PDF Downloads 138719 Surface Nanocrystalline and Hardening Effects of Ti–Al–V Alloy by Electropulsing Ultrasonic Shock
Authors: Xiaoxin Ye, Guoyi Tang
Abstract:
The effect of electropulsing ultrasonic shock (EUS) on the surface hardening and microstructure of Ti6Al4V alloy was studied. It was found that electropulsing improved the microhardness dramatically both in the influential depth and maximum value, compared with the only ultrasonic-shocked sample. It’s indicated that refined surface layer with nanocrystalline and improved microhardness were obtained on account of surface severe plastic deformation, dynamic recrystallization (DRX) and phase change, which was implemented at relative low temperature and high strain rate/capacity due to the coupling of the thermal and athermal effects of EUS. It’s different from conventional experiments and theory. It’s discussed that the positive contributions of EPT in the thermodynamics and kinetics of microstructure and properties change were attributed to the reduction of nucleation energy barrier and acceleration of atomic diffusion. Therefore, it’s supposed that EUS is an energy-saving and high-efficiency method of surface treatment technique with the help of high-energy electropulses, which is promising in cost reduction of the surface engineering and energy management. Keywords: titanium alloys, electropulsing, ultrasonic shock, microhardness, nanocrystalline

Authors: Talebi Aliasghar, Ebrahimpour Komeleh Hooman, Maghsoudi Ali Akbar
Abstract:
In reinforced concrete (RC) structures, beam-column connection region has a considerable effect on the behavior of structures. Using fiber reinforcement polymer (FRP) for the strengthening of connections in RC structures can be one of the solutions to retrofitting this zone which result in the enhanced behavior of structure. In this paper, these changes in behavior by using FRP for high strength concrete beam-column connection have been studied by finite element modeling. The concrete damage plasticity (CDP) model has been used to analyze the RC. The results illustrated a considerable development in load-bearing capacity but also a noticeable reduction in ductility. The study also assesses these qualities for several modes of strengthening and suggests the most effective mode of strengthening. Using FRP in flexural zone and FRP with 45-degree oriented fibers in shear zone of joint showed the most significant change in behavior. Keywords: HSC, beam-column connection, Fiber Reinforcement Polymer, FRP, Finite Element Modeling, FEM

Authors: Carolina Beres, Laurine Da Silva, Danielle Pereira, Ana Ribeiro, Renata Tonon, Caroline Mellinger-Silva, Karina Dos Santos, Flavia Gomes, Lourdes Cabral
Abstract:
The health benefits of bioactive compounds such as phenolics are well known. The main source of these compounds are fruits and derivates. This study had the objective to study the bioacessibility of phenolic compounds from grape pomace and juçara dried extracts. For this purpose both characterized extracts were submitted to a simulated human digestion and the total phenolic content, total anthocyanins and antioxidant scavenging capacity was determinate in digestive fractions (oral, gastric, intestinal and colonic). Juçara had a higher anthocianins bioacessibility (17.16%) when compared to grape pomace (2.08%). The opposite result was found for total phenolic compound, where the higher bioacessibility was for grape (400%). The phenolic compound increase indicates a more accessible compound in the human gut. The lyophilized process had a beneficial impact in the final accessibility of the phenolic compounds being a more promising technique. Keywords: bioacessibility, phenolic compounds, grape, juçara

Authors: Weiying Wang, Qi Li, Huiwen Deng, Weirong Chen
Abstract:
The abrupt light change and uneven illumination will make the PV system get rid of constant output power, which will affect the efficiency of the grid connected inverter as well as the stability of the system. To solve this problem, this paper presents a strategy to control the stability of photovoltaic power system under the condition of partial shading of PV array, leading to constant power output, improving the capacity of resisting interferences. Firstly, a photovoltaic cell model considering the partial shading is established, and the backtracking search algorithm is used as the maximum power point to track algorithm under complex illumination. Then, the energy storage system based on the constant power control strategy is used to achieve constant power output. Finally, the effectiveness and correctness of the proposed control method are verified by the joint simulation of MATLAB/Simulink and RTLAB simulation platform. Keywords: backtracking search algorithm, constant power control, hybrid system, partial shading, stability

Abstract:
This paper deals with the issue of biomass and sorted municipal waste gasification and cogeneration using hot air turbo set. It brings description of designed pilot plant with electrical output 80 kWe. The generated gas is burned in secondary combustion chamber located beyond the gas generator. Flue gas flows through the heat exchanger where the compressed air is heated and consequently brought to a micro turbine. Except description, this paper brings our basic experiences from operating of pilot plant (operating parameters, contributions, problems during operating, etc.). The principal advantage of the given cycle is the fact that there is no contact between the generated gas and the turbine. So there is no need for costly and complicated gas cleaning which is the main source of operating problems in direct use in combustion engines because the content of impurities in the gas causes operation problems to the units due to clogging and tarring of working surfaces of engines and turbines, which may lead as far as serious damage to the equipment under operation. Another merit is the compact container package making installation of the facility easier or making it relatively more mobile. We imagine, this solution of cogeneration from biomass or waste can be suitable for small industrial or communal applications, for low output cogeneration. Keywords: biomass, combustion, gasification, microcogeneration

Authors: Andleeb Zahra, Itrat Rubab, Sumaira Malik, Amina Khan, Muhammad Jawad Khan, M. Qaiser Fatmi
Abstract:
Oral squamous cell carcinoma (OSCC) is one of the most common cancers worldwide. Recent studies have highlighted the role of miRNA in disease pathology, indicating its potential use in an early diagnostic tool. miRNAs are small, double stranded, non-coding RNAs that regulate gene expression by deregulating mRNAs. miRNAs play important roles in modifying various cellular processes such as cell growth, differentiation, apoptosis, and immune response. Dis-regulated expression of miRNAs is known to affect the cell growth, and this may function as tumor suppressors or oncogenes in various cancers. Objectives: The main objectives of this study were to characterize the extracellular miRNAs involved in oral cancer (OC) to assist early detection of cancer as well as to propose a list of genes that can potentially be used as biomarkers of OC. We used gene expression data by microarrays already available in literature. Materials and Methods: In the first step, a total of 318 miRNAs involved in oral carcinoma were shortlisted followed by the prediction of their target genes. Simultaneously, the differentially expressed genes (DEGs) of oral carcinoma from all experiments were identified. The common genes between lists of DEGs of OC based on experimentally proven data and target genes of each miRNA were identified. These common genes are the targets of specific miRNA, which is involved in OC. Finally, a list of genes was generated which may be used as biomarker of OC. Results and Conclusion: In results, we included some of pathways in cancer to show the change in gene expression under the control of specific miRNA. Ingenuity pathway analysis (IPA) provided a list of major biomarkers like CDH2, CDK7 and functional enrichment analysis identified the role of miRNA in major pathways like cell adhesion molecules pathway affected by cancer. We observed that at least 25 genes are regulated by maximum number of miRNAs, and thereby, they can be used as biomarkers of OC. To better understand the role of miRNA with respect to their target genes further experiments are required, and our study provides a platform to better understand the miRNA-OC relationship at genomics level. Keywords: biomarkers, gene expression, miRNA, oral carcinoma

Authors: Zahra Shokrolahi, Mohammad Reza Atashzar
Abstract:
The goals of treating patients with breast cancer are to cure the disease, prolong survival, and improve quality of life. Immune cells in the tumor microenvironment have an important role in regulating tumor progression. The term of cellular immunotherapy refers to the administration of living cells to a patient; this type of immunotherapy can be active, such as a dendritic cell (DC) vaccine, in that the cells can stimulate an anti-tumour response in the patient, or the therapy can be passive, whereby the cells have intrinsic anti-tumour activity; this is known as adoptive cell transfer (ACT) and includes the use of autologous or allogeneic lymphocytes that may, or may not, be modified. The most important breast cancer cellular immunotherapies involving the use of T cells and natural killer (NK) cells in adoptive cell transfer, as well as dendritic cells vaccines. T cell-based therapies including tumour-infiltrating lymphocytes (TILs), engineered TCR-T cells, chimeric antigen receptor (CAR T cell), Gamma-delta (γδ) T cells, natural killer T (NKT) cells. NK cell-based therapies including lymphokine-activated killers (LAK), cytokine-induced killer (CIK) cells, CAR-NK cells. Adoptive cell therapy has some advantages and disadvantages some. TILs cell strictly directed against tumor-specific antigens but are inactive against tumor changes due to immunoediting. CIK cell have MHC-independent cytotoxic effect and also need concurrent high dose IL-2 administration. CAR T cell are MHC-independent; overcome tumor MHC molecule downregulation; potent in recognizing any cell surface antigen (protein, carbohydrate or glycolipid); applicable to a broad range of patients and T cell populations; production of large numbers of tumor-specific cells in a moderately short period of time. Meanwhile CAR T cells capable of targeting only cell surface antigens; lethal toxicity due to cytokine storm reported. Here we present the most popular cancer cellular immunotherapy approaches and discuss their clinical relevance referring to data acquired from clinical trials .To date, clinical experience and efficacy suggest that combining more than one immunotherapy interventions, in conjunction with other treatment options like chemotherapy, radiotherapy and targeted or epigenetic therapy, should guide the way to cancer cure. Keywords: breast cancer , cell therapy , CAR T cell , CIK cells

Authors: Ludivine Gueho, Sylvain Fleury, Eric Jamet
Abstract:
Driver distraction is known to be a major contributing factor of car accidents. Since many years, constructors have been designing embedded technologies to face this problem and reduce distraction. Being able to predict user acceptance would further be helpful in the development process to build appropriate systems. The present research aims at modelling the acceptability of a specific system, an innovative personalized and contextualized embedded radio, through an online survey of 202 people in France that assessed the psychological variables determining intentions to use the system. The questionnaire instantiated the dimensions of the extended version of the UTAUT acceptability model. Because of the specific features of the system assessed, we added 4 dimensions: perceived security, anxiety, trust and privacy concerns. Results showed that hedonic motivation, i.e., the fun or pleasure derived from using a technology, and performance expectancy, i.e., the degree to which individuals believe that the characteristics of the system meet their needs, are the most important dimensions in determining behavioral intentions about the innovative radio. To a lesser extent, social influence, i.e., the degree to which individuals think they can use the system while respecting their social group’s norms and while giving a positive image of themselves, had an effect on behavioral intentions. Moreover, trust, that is, the positive belief about the perceived reliability of, dependability of, and confidence in a person, object or process, had a significant effect, mediated by performance expectancy. In an applicative way, the present research reveals that, to be accepted, in-car embedded new technology has to address individual needs, for instance by facilitating the driving activity or by providing useful information. If it shows hedonic qualities by being entertaining, pretty or comfortable, this may improve the intentions to use it. Therefore, it is clearly important to include reflection about user experience in the design process. Finally, the users have to be reassured on the system’s reliability. For example, improving the transparency of the system by providing information about the system functioning, could improve trust. These results bring some highlights on determinant of acceptance of an in-vehicle technology and are useful for constructors to design acceptable systems. Keywords: acceptability, innovative embedded radio, structural equation, user-centric evaluation, UTAUT

Procedia PDF Downloads 3438704 Assessing Flexural Damage Mechanisms Induced by Mesoscopic Buckle Defects in Textile-Reinforced Polymer Matrix Composites Using Acoustic Emission Analysis
Authors: Christopher Okechukwu Ndukwe
Abstract:
This paper investigates and categorizes the flexural damage mechanisms in composite materials caused by mesoscopic out-of-plane buckle defects that occur during the initial stage of the resin transfer molding (RTM) process. The findings of this study have significant practical implications for the manufacturing and use of composite materials, as they provide a deeper understanding of these damage mechanisms and their analysis. During the initial stage of shaping a preform, alterations, and distortions in the reinforcement sample can significantly lead to defects, such as buckling, especially when forming double-curvature geometries. These recurring mesoscopic defects have been investigated using a specialized laboratory bench designed to reproduce buckle defects like those found in complex geometric shapes, such as tetrahedrons. The study examined two sample configurations with buckle defects in the longitudinal and transverse directions alongside a reference sample for comparison. An acoustic emission (AE) system, a well-regarded non-contact method for monitoring structural health, was used to analyze the mechanical behavior of material samples in detail. An unsupervised K-means algorithm was employed to classify the damage mechanisms—such as matrix cracking, interface damage, and fiber breakage linked to the samples' failure. A standard was established based on three AE parameters: absolute energy, amplitude, and the number of AE events. This standard helped identify the origin and sequence of damage propagation. Initially, the results of the AE parameters were superimposed with the flexural loading curves to pinpoint the loading phases during which damage began and the specific points at which the samples ultimately failed. The normalized density of AE events related to different damage mechanisms was evaluated by analyzing the number of AE events within the amplitude domain of the AE signals. The ranges of the identified damage mechanisms in the amplitude plane illustrate the progression and order of load transfer among the elements of the composite material. In the reference sample, the AE event signals corresponding to the three classes of damage mechanisms partially overlap with adjacent signals. In contrast, the two defective sample configurations showed that the overlapping AE event signals for the respective damage mechanisms converged within the intermediate damage mode area at specific points, depending on the sample configuration. The convergence points in the samples with transverse defects were identified relatively earlier than in the other samples. Low and high amplitude ranges characterize the matrix cracking and fiber breakage damage mechanisms. The low amplitude damage occurred over a more extended length, while the high amplitude damage began much earlier. This results in the signals from both damage mechanisms converging at the center of the interface damage zone. This convergence suggests that all individual composite components fail concurrently at specific points in the defective samples, resulting in rapid fragmentation and ultimately contributing to failure. Overall, the results show that mesoscopic out-of-plane buckling in all directions affects the composite's flexural response, with more severe effects observed when the load is applied transversely. Keywords: acoustic emission, composite reinforcement, damage mechanisms, mesoscopic buckle defects

Abstract:
Vision based user interfaces to control TVs and PCs have the advantage of being able to perform natural control without being limited to a specific device. Accordingly, various studies on hand gesture recognition using RGB cameras or depth cameras have been conducted. However, such cameras have the disadvantage of lacking in accuracy or the construction cost being large. The proposed method uses a low cost IR camera to accurately differentiate between the hand and the background. Also, complicated learning and template matching methodologies are not used, and the correlation between the fingertips extracted through curvatures is utilized to recognize Click and Move gestures. Keywords: recognition, hand gestures, infrared camera, RGB cameras

Authors: Meysam Saidi
Abstract:
While the specific externalities and required regulatory measures in relation to Islamic banking are fairly uncertain, the business is growing across the world. Unofficial data indicate that the Islamic Finance market is growing with annual rate of 15% and it has reached 1.3 $ trillion size. This trend is associated with inherent systematic connection of Islamic financial institutions to other entities and different sectors of economies. Islamic banking has been subject of market development policies in major economies, most notably the UK. This trend highlights the need for identification of distinct risk features of Islamic banking and crafting customized regulatory measures. So far there has not been a significant systemic crisis in this market which can be attributed to its distinct nature. However, the significant growth and spread of its products worldwide necessitate an in depth study of its nature for customized congruent regulatory measures. In the post financial crisis era some market analysis and reports suggested that the Islamic banks fairly weathered the crisis. As far as heavily blamed conventional financial products such as subprime mortgage backed securities and speculative credit default swaps were concerned the immunity claim can be considered true, as Islamic financial institutions were not directly exposed to such products. Nevertheless, similar to the experience of the conventional banking industry, it can be only a matter of time for Islamic banks to face failures that can be specific to the nature of their business. Using the experience of conventional banking regulations and identifying those peculiarities of Islamic banking that need customized regulatory approach can aid to prevent major failures. Frank Knight has stated that “We perceive the world before we react to it, and we react not to what we perceive, but always to what we infer”. The debate over congruent Islamic banking regulations might not be an exception to Frank Knight’s statement but I will try to base my discussion on concrete evidences. This paper first analyzes both theoretical and actual features of Islamic banking in order to ascertain to its peculiarities in terms of market stability and other externalities. Next, the paper discusses distinct features of Islamic financial transactions and banking which might require customized regulatory measures. Finally, the paper explores how a more transparent path for the Islamic banking regulations can be drawn. Keywords: Islamic banking, regulation, risks, capital requirements, customer protection, financial stability

This proposed framework-An Evaluation Framework for Mobile Health Education Apps-consists of a hybrid of 5 metrics designated from a larger set in usability evaluation and heuristic evaluation, illuminated grounded on 15 unstructured interviews from software developers (SD), health professionals (HP) and patients (P). These five metrics corresponding to explicit facets of usability recognised through a requirements analysis of typical stakeholders of mobile health apps. These five hybrid selected metrics were scattered across 24 specific questionnaire questions, which are available on request from first author. This questionnaire has been sent to 81 participants distributed in three sets of stakeholders from software developers (SD), health professionals (HP) and patients/general users (P/GU) on the purpose of ranking three sets of mobile health education applications. Finally, the outcomes from the questionnaire data helped us to approach our aims which are finding the profile for different stakeholders, finding the profile for different mobile health educations application packages, ranking different mobile health education application and guide us to build the select guidance too which is apart from the Evaluation Framework for Mobile Health Education Apps. Keywords: evaluation framework, heuristic evaluation, usability evaluation, metrics

Abstract:
The demand for electric power in Saudi Arabia is steadily increasing with economic growth. More power plants should be installed to increase generation capacity and meet demand. Electricity in Saudi Arabia is mainly dependent on fossil fuels, which are a major problem as they deplete natural resources and increase CO₂ emissions. In this research work, performance and techno-economic analyzes are conducted to evaluate a microgrid system based on hybrid PV/wind diesel power sources as a stand-alone system for rural electrification in Saudi Arabia. The total power flow, maximum power point tracking (MPPT) efficiency, effectiveness of the proposed control strategy, and total harmonic distortion (THD) are analyzed in MATLAB/Simulink environment. Various simulation studies have been carried out under different irradiation conditions. The sizing, optimization, and economic feasibility analysis were performed using Homer energy software. Keywords: WIND, solar, microgrid, energy

Abstract:
Biological methylation is a methyl group transfer from S-adenosyl-L-methionine (AdoMet) onto N-, C-, O- or S-nucleophiles in DNA, RNA, proteins or small biomolecules. The reaction is catalyzed by enzymes called AdoMet-dependent methyltransferases (MTases), which represent more than 3 % of the proteins in the cell. As a general mechanism, the methyl group from AdoMet replaces a hydrogen atom of nucleophilic center producing methylated DNA and S-adenosyl-L-homocysteine (AdoHcy). Recently, DNA methyltransferases have been used for the sequence-specific, covalent labeling of biopolymers. Two types of MTase catalyzed labeling of biopolymers are known, referred as two-step and one-step. During two-step labeling, an alkylating fragment is transferred onto DNA in a sequence-specific manner and then the reporter group, such as biotin, is attached for selective visualization using suitable chemistries of coupling. This approach of labeling is quite difficult and the chemical hitching does not always proceed at 100 %, but in the second step the variety of reporter groups can be selected and that gives the flexibility for this labeling method. In the one-step labeling, AdoMet analog is designed with the reporter group already attached to the functional group. Thus, the one-step labeling method would be more comfortable tool for labeling of biopolymers in order to prevent additional chemical reactions and selection of reaction conditions. Also, time costs would be reduced. However, effective AdoMet analog appropriate for one-step labeling of biopolymers and containing cleavable bond, required for reduction of PCR interferation, is still not known. To expand the practical utility of this important enzymatic reaction, cofactors with activated sulfonium-bound side-chains have been produced and can serve as surrogate cofactors for a variety of wild-type and mutant DNA and RNA MTases enabling covalent attachment of these chains to their target sites in DNA, RNA or proteins (the approach named methyltransferase-directed Transfer of Activated Groups, mTAG). Compounds containing hex-2-yn-1-yl moiety has proved to be efficient alkylating agents for labeling of DNA. Herein we describe synthetic procedures for the preparation of N-biotinoyl-N’-(pent-4-ynoyl)cystamine starting from the coupling of cystamine with pentynoic acid and finally attaching the biotin as a reporter group. The synthesis of the first AdoMet based cofactor containing a cleavable reporter group and appropriate for one-step labeling was developed. Keywords: adoMet analogs, DNA alkylation, cofactor, methyltransferases

Authors: Samira Alvandi
Abstract:
In the context of the circular economy, production planning aims to eliminate waste and emissions and maximize resource efficiency. Historically production planning is challenged through arrays of uncertainty and complexity arising from the interdependence and variability of products, processes, and systems. Manufacturers worldwide are facing new challenges in tackling various environmental issues such as climate change, resource depletion, and land degradation. In managing the inherited complexity and uncertainty and yet maintaining profitability, the manufacturing sector is in need of a holistic framework that supports energy efficiency and carbon emission reduction schemes. The proposed framework addresses the current challenges and integrates simulation modeling with optimization for finding optimal machine-job allocation to maximize throughput and total energy consumption while minimizing lead time. The aluminium refinery facility in western Sydney, Australia, is used as an exemplar to validate the proposed framework. Keywords: smart production planning, simulation-optimisation, energy aware capacity planning, energy intensive industries
