418 Analysis and Remediation of Fecal Coliform Bacteria Pollution in Selected Surface Water Bodies of Enugu State of Nigeria

Abstract:

The assessment of surface waters in Enugu metropolis for fecal coliform bacteria was undertaken. Enugu urban was divided into three areas (A1, A2 and A3), and fecal coliform bacteria analysed in the surface waters found in these areas for four years (2005-2008). The plate count method was used for the analyses. Data generated were subjected to statistical tests involving; Normality test, Homogeneity of variance test, correlation test, and tolerance limit test. The influence of seasonality and pollution trends were investigated using time series plots. Results from the tolerance limit test at 95% coverage with 95% confidence, and with respect to EU maximum permissible concentration show that the three areas suffer from fecal coliform pollution. To this end, remediation procedure involving the use of saw-dust extracts from three woods namely; Chlorophora-Excelsa (C-Excelsa),Khayan-Senegalensis,(CSenegalensis) and Erythrophylum-Ivorensis (E-Ivorensis) in controlling the coliforms was studied. Results show that mixture of the acetone extracts of the woods show the most effective antibacterial inhibitory activities (26.00mm zone of inhibition) against E-coli. Methanol extract mixture of the three woods gave best inhibitory activity (26.00mm zone of inhibition) against S-areus, and 25.00mm zones of inhibition against E-Aerogenes. The aqueous extracts mixture gave acceptable zones of inhibitions against the three bacteria organisms.

417 Molecular Identification of ESBL Genesbla GES-1, blaVEB-1, blaCTX-M blaOXA-1, blaOXA-4,blaOXA-10 and blaPER-1 in Pseudomonas aeruginosa Strains Isolated from Burn Patientsby PCR, RFLP and Sequencing Techniques

Abstract:

Fourty one strains of ESBL producing P.aeruginosa which were previously isolated from burn patients in Kerman University general hospital, Iran were subjected to PCR, RFLP and sequencing in order to determine the type of extended spectrum β- lactamases (ESBL), the restriction digestion pattern and possibility of mutation among detected genes. DNA extraction was carried out by phenol chloroform method. PCR for detection of bla genes was performed using specific primer for each gene. Restriction Fragment Length Polymorphism (RFLP) for ESBL genes was carried out using EcoRI, NheI, PVUII, EcoRV, DdeI, and PstI restriction enzymes. The PCR products were subjected to direct sequencing of both the strands for identification of the ESBL genes.The blaCTX-M, blaVEB-1, blaPER-1, blaGES-1, blaOXA-1, blaOXA-4 and blaOXA-10 genes were detected in the (n=1) 2.43%, (n=41)100%, (n=28) 68.3%, (n=10) 24.4%, (n=29) 70.7%, (n=7)17.1% and (n=38) 92.7% of the ESBL producing isolates respectively. The RFLP analysis showed that each ESBL gene has identical pattern of digestion among the isolated strains. Sequencing of the ESBL genes confirmed the genuinety of PCR products and revealed no mutation in the restriction sites of the above genes. From results of the present investigation it can be concluded that blaVEB-1 and blaCTX-M were the most and the least frequently isolated ESBL genes among the P.aeruginosa strains isolated from burn patients. The RFLP and sequencing analysis revealed that same clone of the bla genes were indeed existed among the antibiotic resistant strains.

416 Study the Biological Activities of Tribulus Terrestris Extracts

Abstract:

In this study the extracts of the Iraqi herb Tribulus terrestris (Al-Hassage or Al-Kutub) was done by using of polar and non polar solvents, then the biological activity of these extractants was studied in three fields, First, the antibacterial activity (in vitro) on gram positive bacteria (Staphylococcus aureus), and gram negative bacteria (E. coli, Proteus vulgaris, Pseudomonas aerugiuosa, and Klebsiella), all extracts showed considerable activity against all bacteria. Second, the effect of extracts on free serum testosterone level in male mice (in vivo), the alcoholic, and acetonitrilic extracts showed significant (P < 0.05) increase in free serum testosterone level, and we found that the extracts contained compounds with less genotoxic effects in mice germ cells. 3rd, was to study the effect of methanolic extract of T. terrestris in diabetes management.

415 Pentachlorophenol Removal via Adsorption and Biodegradation

Abstract:

Removal of PCP by a system combining biodegradation by biofilm and adsorption was investigated here. Three studies were conducted employing batch tests, sequencing batch reactor (SBR) and continuous biofilm activated carbon column reactor (BACCOR). The combination of biofilm-GAC batch process removed about 30% more PCP than GAC adsorption alone. For the SBR processes, both the suspended and attached biomass could remove more than 90% of the PCP after acclimatisation. BACCOR was able to remove more than 98% of PCP-Na at concentrations ranging from 10 to 100 mg/L, at empty bed contact time (EBCT) ranging from 0.75 to 4 hours. Pure and mixed cultures from BACCOR were tested for use of PCP as sole carbon and energy source under aerobic conditions. The isolates were able to degrade up to 42% of PCP under aerobic conditions in pure cultures. However, mixed cultures were found able to degrade more than 99% PCP indicating interdependence of species.

414 Isolation and Identification of an Acetobacter Strain from Iranian White-Red Cherry with High Acetic Acid Productivity as a Potential Strain for Cherry Vinegar Production in Foodand Agriculture Biotechnology

Abstract:

According to FDA (Food and Drug Administration of the United States), vinegar is definedas a sour liquid containing at least 4 grams acetic acid in 100 cubic centimeter (4% solution of acetic acid) of solution that is produced from sugary materials by alcoholic fermentation. In the base of microbial starters, vinegars could be contained of more than 50 types of volatile and aromatic substances that responsible for their sweet taste and smelling. Recently the vinegar industry has a great proportion in agriculture, food and microbial biotechnology. The acetic acid bacteria are from the family Acetobacteraceae. Regarding to the latest version of Bergy-s Mannual of Systematic Bacteriology that has categorized bacteria in the base of their 16s RNA differences, the most important acetic acid genera are included Acetobacter (genus I), Gluconacetobacter (genus VIII) and Gluconobacter (genus IX). The genus Acetobacter that is primarily used in vinegar manufacturing plants is a gram negative, obligate aerobe coccus or rod shaped bacterium with the size 0.6 - 0.8 X 1.0 - 4.0 μm, nonmotile or motile with peritrichous flagella and catalase positive – oxidase negative biochemically. Some strains are overoxidizer that could convert acetic acid to carbon dioxide and water.In this research one Acetobacter native strain with high acetic acid productivity was isolated from Iranian white – red cherry. We used two specific culture media include Carr medium [yeast extract, 3%; ethanol, 2% (v/v); bromocresol green, 0.002%; agar, 2% and distilled water, 1000 ml], Frateur medium [yeast extract, 10 g/l; CaCO3, 20 g/l; ethanol, 20 g/l; agar, 20 g/l and distilled water, 1000 ml] and an industrial culture medium. In addition to high acetic acid production and high growth rate, this strain had a good tolerance against ethanol concentration that was examined using modified Carr media with 5%, 7% and 9% ethanol concentrations. While the industrial strains of acetic acid bacteria grow in the thermal range of 28 – 30 °C, this strain was adapted for growth in 34 – 36 °C after 96 hours incubation period. These dramatic characteristics suggest a potential biotechnological strain in production of cherry vinegar with a sweet smell and different nutritional properties in comparison to recent vinegar types. The lack of growth after 24, 48 and 72 hours incubation at 34 – 36 °C and the growth after 96 hours indicates a good and fast thermal flexibility of this strain as a significant characteristic of biotechnological and industrial strains.

413 Safety Assessment of Traditional Ready-to-Eat Meat Products Vended at Retail Outlets in Kebbi and Sokoto States, Nigeria

Abstract:

Food safety is a significant and growing public health problem in the world and Nigeria as a developing country, since food-borne diseases are important contributors to the huge burden of sickness and death of humans. In Nigeria, traditional ready-to-eat meat products (RTE-MPs) like balangu, tsire, guru and dried meat products like kilishi, dambun nama, banda, were reported to be highly appreciated because of their eating qualities. The consumption of these products was considered as safe due to the treatments that are usually involved during their production process. However, during processing and handling, the products could be contaminated by pathogens that could cause food poisoning. Therefore, a hazard identification for pathogenic bacteria on some traditional RTE-MPs was conducted in Kebbi and Sokoto States, Nigeria. A total of 116 RTE-MPs (balangu-38, kilishi-39 and tsire-39) samples were obtained from retail outlets and analyzed using standard cultural microbiological procedures in general and selective enrichment media to isolate the target pathogens. A six-fold serial dilution was prepared and using the pour plating method, colonies were counted. Serial dilutions were selected based on the prepared pre-labeled Petri dishes for each sample. A volume of 10-12 ml of molten Nutrient agar cooled to 42-45°C was poured into each Petri dish and 1 ml each from dilutions of 102, 104 and 106 for every sample was respectively poured on a pre-labeled Petri plate after which colonies were counted. The isolated pathogens were identified and confirmed after series of biochemical tests. Frequencies and percentages were used to describe the presence of pathogens. The General Linear Model was used to analyze data on pathogen presence according to RTE-MPs and means were separated using the Tukey test at 0.05 confidence level. Of the 116 RTE-MPs samples collected, 35 (30.17%) samples were found to be contaminated with some tested pathogens. Prevalence results showed that Escherichia coli, salmonella and Staphylococcus aureus were present in the samples. Mean total bacterial count was 23.82×106 cfu/g. The frequency of individual pathogens isolated was; Staphylococcus aureus 18 (15.51%), Escherichia coli 12 (10.34%) and Salmonella 5 (4.31%). Also, among the RTE-MPs tested, the total bacterial counts were found to differ significantly (P < 0.05), with 1.81, 2.41 and 2.9×104 cfu/g for tsire, kilishi, and balangu, respectively. The study concluded that the presence of pathogenic bacteria in balangu could pose grave health risks to consumers, and hence, recommended good manufacturing practices in the production of balangu to improve the products’ safety.

412 Toxicity Study of Two Different Synthesized Silver Nanoparticles on Bacteria Vibrio Fischeri

Abstract:

A comparative evaluation of acute toxicity of synthesized nano silvers using two different procedures (biological and chemical reduction methods) and silver ions on bacteria Vibrio fischeri was investigated. The bacterial light inhibition test as a toxicological endpoint was used by applying of a homemade luminometer. To compare the toxicity effects as a quantitative parameter, a nominal effective concentrations (EC) of chemicals and a susceptibility constant (Z-value) of bacteria, after 5 min and 30 min exposure times, were calculated. After 5 and 30 min contact times, the EC50 values of two silver nanoparticles and the EC20 values were about similar. It demonstrates that toxicity of silvers was independent of their procedure. The EC values of nanoparticles were larger than those of the silver ions. The susceptibilities(Z- Values) of V.fischeri (L/mg) to the silver ions were greater than those of the nano silvers. According to the EC and Z values, the toxicity of silvers decreased in the following order: Silver ions >> silver nanoparticles from chemical reduction method ~ silver nanoparticles from biological method.

411 Identification of the Antimicrobial Effect of Liquorice Extracts on Gram-Positive Bacteria: Determination of Minimum Inhibitory Concentration and Mechanism of Action Using a luxABCDE Reporter Strain

Abstract:

Natural preservatives have been used as alternatives to traditional chemical preservatives; however, a limited number have been commercially developed and many remain to be investigated as sources of safer and effective antimicrobials. In this study, we have been investigating the antimicrobial activity of an extract of Glycyrrhiza glabra (liquorice) that was provided as a waste material from the production of liquorice flavourings for the food industry, and to investigate if this retained the expected antimicrobial activity so it could be used as a natural preservative. Antibacterial activity of liquorice extract was screened for evidence of growth inhibition against eight species of Gram-negative and Gram-positive bacteria, including Listeria monocytogenes, Listeria innocua, Staphylococcus aureus, Enterococcus faecalis and Bacillus subtilis. The Gram-negative bacteria tested include Pseudomonas aeruginosa, Escherichia coli and Salmonella typhimurium but none of these were affected by the extract. In contrast, for all of the Gram-positive bacteria tested, growth was inhibited as monitored using optical density. However parallel studies using viable count indicated that the cells were not killed meaning that the extract was bacteriostatic rather than bacteriocidal. The Minimum Inhibitory Concentration [MIC] and Minimum Bactericidal Concentration [MBC] of the extract was also determined and a concentration of 50 µg ml-1 was found to have a strong bacteriostatic effect on Gram-positive bacteria. Microscopic analysis indicated that there were changes in cell shape suggesting the cell wall was affected. In addition, the use of a reporter strain of Listeria transformed with the bioluminescence genes luxABCDE indicated that cell energy levels were reduced when treated with either 12.5 or 50 µg ml-1 of the extract, with the reduction in light output being proportional to the concentration of the extract used. Together these results suggest that the extract is inhibiting the growth of Gram-positive bacteria only by damaging the cell wall and/or membrane.

410 Utilization of 3-N-trimethylamino-1-propanol by Rhodococcus sp. strain A4 isolated from Natural Soil

Abstract:

The aim of this study was to screen for microorganism that able to utilize 3-N-trimethylamino-1-propanol (homocholine) as a sole source of carbon and nitrogen. The aerobic degradation of homocholine has been found by a gram-positive Rhodococcus sp. bacterium isolated from soil. The isolate was identified as Rhodococcus sp. strain A4 based on the phenotypic features, physiologic and biochemical characteristics, and phylogenetic analysis. The cells of the isolated strain grown on both basal-TMAP and nutrient agar medium displayed elementary branching mycelia fragmented into irregular rod and coccoid elements. Comparative 16S rDNA sequencing studies indicated that the strain A4 falls into the Rhodococcus erythropolis subclade and forms a monophyletic group with the type-strains of R. opacus, and R. wratislaviensis. Metabolites analysis by capillary electrophoresis, fast atom bombardment-mass spectrometry, and gas chromatography- mass spectrometry, showed trimethylamine (TMA) as the major metabolite beside β-alanine betaine and trimethylaminopropionaldehyde. Therefore, the possible degradation pathway of trimethylamino propanol in the isolated strain is through consequence oxidation of alcohol group (-OH) to aldehyde (-CHO) and acid (-COOH), and thereafter the cleavage of β-alanine betaine C-N bonds yielded trimethylamine and alkyl chain.

409 Screening and Identification of Microorganisms – Potential Producers of Arachidonic Acid

Abstract:

Microorganisms isolated from water and soil of Kazakhstan to identify potential high-effective producers of the arachidonic acid, exhibiting a wide range of physiological activity and having practical applications were screened. Based on the results of two independent tests (the test on the sensitivity of the growth processes of microorganisms to acetylsalicylic acid - an irreversible inhibitor of PGH-synthase involved in the metabolism of arachidonic acid and its derivatives, the test for inhibition of peroxidase activity of membrane-bounding fraction of PGH - synthase by acetylsalicylic acid) were selected microbial cultures which are potential highproducer of arachidonic acid. They are characterized by a stable strong growth in the laboratory conditions. Identification of microorganism cultures based on morphological, physiological, biochemical and molecular genetic characteristics was performed.

408 Analysis of Sulphur-Oxidizing Bacteria Attack on Concrete Based On Waste Materials

Abstract:

Concrete durability as an important engineering property of concrete, determining the service life of concrete structures very significantly, can be threatened and even lost due to the interactions of concrete with external environment. Bio-corrosion process caused by presence and activities of microorganisms producing sulphuric acid is a special type of sulphate deterioration of concrete materials. The effects of sulphur-oxidizing bacteria Acidithiobacillus thiooxidans on various concrete samples, based on silica fume and zeolite, were investigated in laboratory during 180 days. A laboratory study was conducted to compare the performance of concrete samples in terms of the concrete deterioration influenced by the leaching of calcium and silicon compounds from the cement matrix. The changes in the elemental concentrations of calcium and silicon in both solid samples and liquid leachates were measured by using X – ray fluorescence method. Experimental studies confirmed the silica fume based concrete samples were found out to have the best performance in terms of both silicon and calcium ions leaching.

407 Biological Methods to Control Parasitic Weed Phelipanche ramosa L. Pomel in the Field Tomato Crop

Abstract:

Phelipanche ramosa L. Pomel is a root holoparasitic weed plant of many cultivations, particularly of tomato (Lycopersicum esculentum L.) crop. In Italy, Phelipanche problem is increasing, both in density and in acreage. The biological control of this parasitic weed involves the use of living organisms as numerous fungi and bacteria that can infect the parasitic weed, while it may improve the crop growth. This paper deals with the biocontrol with microorganism, including Arbuscular mycorrhizal (AM) fungi and fungal pathogens as Fusarium oxisporum spp. Colonization of crop roots by AM fungi can provide protection of crops against parasitic weeds because of a reduction in their seed germination and attachment, while F. oxisporum, isolated from diseased broomrape tubercles, proved to be highly virulent on P. ramosa. The experimental trial was carried out in open field at Foggia province (Apulia Region, Southern Italy), during the spring-summer season 2016, in order to evaluate the effect of four biological treatments: AM fungi and Fusarium oxisporum applied in the soil alone or combined together, and Rizosum Max® product, compared with the untreated control, to reduce the P. ramosa infestation in processing tomato crop. The principal results to be drawn from this study under field condition, in contrast of those reported previously under laboratory and greenhouse conditions, show that both AM fungi and F. oxisporum do not provide the reduction of the number of emerged shoots of P. ramosa. This can arise probably from the low efficacy seedling of the agent pathogens for the control of this parasite in the field. On the contrary, the Rizosum Max® product, containing AM fungi and some rizophere bacteria combined with several minerals and organic substances, appears to be most effective for the reduction of P. ramosa infestation.

406 Probiotics’ Antibacterial Activity on Beef and Camel Minced Meat at Altered Ranges of Temperature

Abstract:

Because of their inhibitory effects, selected probiotic Lactobacilli may be used as antimicrobial against some hazardous microorganisms responsible for spoilage of fresh minced beef (cattle) minced meat and camel minced meat. Lactic acid bacteria were isolated from camel meat. These included 10 isolates; 1 Lactobacillus fermenti, 4 Lactobacillus plantarum, 4 Lactobacillus pulgaricus, 3 Lactobacillus acidophilus and 1 Lactobacillus brevis. The most efficient inhibitory organism was Lactobacillus plantarum which can be used as a propiotic with antibacterial activity. All microbiological analyses were made at the time 0, first day and the second day at altered ranges of temperature [4±2 ⁰C (chilling temperature), 25±2 ⁰C, and 38±2 ⁰C]. Results showed a significant decrease of pH 6.2 to 5.1 within variant types of meat, in addition to reduction of Total Bacterial Count, Enterococci, Bacillus cereus and Escherichia coli together with the stability of Coliforms and absence of Staphylococcus aureus.

405 Antibacterial and Antifungal Activity of Essential Oil of Eucalyptus camendulensis on a Few Bacteria and Fungi

Abstract:

Red River Gum (Eucalyptus camaldulensis) is a tree of the genus Eucalyptus widely distributed in Algeria and in the world. The value of its aromatic secondary metabolites offers new perspectives in the pharmaceutical industry. This strategy can contribute to the sustainable development of our country. Preliminary tests performed on the essential oil of Eucalyptus camendulensis showed that this oil has antibacterial activity vis-à-vis the bacterial strains (Enterococcus feacalis, Enterobacter cloaceai, Proteus microsilis, Escherichia coli, Klebsiella pneumonia, and Pseudomonas aeruginosa) and antifungic (Fusarium sporotrichioide and Fusarium graminearum). The culture medium used was nutrient broth Muller Hinton. The interaction between the bacteria and the essential oil is expressed by a zone of inhibition with diameters of MIC indirectly expression of. And we used the PDA medium to determine the fungal activity. The extraction of the aromatic fraction (essentially oilhydrolat) of the fresh aerian part of the Eucalyptus camendulensis was performed by hydrodistillation. The average essential oil yield is 0.99%. The antimicrobial and fungal study of the essential oil and hydrosol showed a high inhibitory effect on the growth of pathogens.

The primary aim of this paper is to present our research focused on theoretical and experimental analysis of potable water and in-building water distribution systems from the point of view of microbiological risk on the basis of confrontation between the theoretical analysis and synthesis of gathered information in conditions of the Slovak Republic. The presence of the bacteria Legionella in water systems, especially in hot water distribution system, represents in terms of health protection of inhabitants the crucial problem which cannot be overlooked. Legionella pneumophila discovery, its classification and its influence on installations inside buildings are relatively new. There are a lot of guidelines and regulations developed in many individual countries for the design, operation and maintenance for tap water systems to avoid the growth of bacteria Legionella pneumophila, but in Slovakia we don-t have any. The goal of this paper is to show the necessity of prevention and regulations for installations inside buildings verified by simulation methods.

403 Segmentation and Recognition of Handwritten Numeric Chains

Abstract:

In this paper we present an off line system for the recognition of the handwritten numeric chains. Our work is divided in two big parts. The first part is the realization of a recognition system of the isolated handwritten digits. In this case the study is based mainly on the evaluation of neural network performances, trained with the gradient back propagation algorithm. The used parameters to form the input vector of the neural network are extracted on the binary images of the digits by several methods: the distribution sequence, the Barr features and the centred moments of the different projections and profiles. The second part is the extension of our system for the reading of the handwritten numeric chains constituted of a variable number of digits. The vertical projection is used to segment the numeric chain at isolated digits and every digit (or segment) will be presented separately to the entry of the system achieved in the first part (recognition system of the isolated handwritten digits). The result of the recognition of the numeric chain will be displayed at the exit of the global system.

402 Analysis of Microalgae Lipids Isolated from Basin of Kazakhstan, to Assess the Prospects of Practical Use

Abstract:

It was analyzed of fatty acid composition of 16 strains of microalgae lipid fractions isolated from different basins of Kazakhstan and characterized by stable active growth in the laboratory. Three species of green microalgae (Oocystis rhomboideus, Chlorococcum infusionum, Dictyochlorella globosa) and three species of diatoms (Synedra sp., Nitzshia sp., Pleurosigma attenuatum) are characterized by a high content of lipids and are promising for further study as a source of polyunsaturated fatty acids.

401 Identification Common Microbes Observed on Polyester Tufting

Abstract:

Tufting carpet is a very suitable substrate for growing microorganism such as pathogenic microbes, due to the direct touch with human body, long washing periods and laying on the floor; in fact there are 3 major problems: To risk human health, Prepare bad odors and Destruction of the products.. In the presented research, for investigation of presence most common microbes on polyester tufting, first goods laid in a public place (in the corridor fair) for 30 days and the existence of some microbes were investigate on it with two methods of enrichment in nutrient environments such as thioglycolate and noutrunt brath, and shake the dust off the polyester tufting onto cultivation mediums such as blood agar and noutrunt agar. After the microorganism colonics are grown, the colonies were separated and six microbial tests such as cataloes and sitrat were carried out in five phases on the colonics for identifying the varieties of bacteria. As a result of tests, 5 type of bacteria, such as Escherichia coli, staphylococcus saprophytic as were identified. Each of the mentioned bacteria can be seriously harmful for the heath of human.

400 Fermentation of Germinated Native Black Rice Milk Mixture by Probiotic Lactic Acid Bacteria

Abstract:

This research aimed to demonstrate probiotic germinated native black rice juice fermentation by lactic acid bacteria (Lactobacillus casei TISTR 390). Germinated native black rice juice was inoculated with a 24-h old lactic culture and incubated at 30 °C for 72 hours. Changes in pH, acidity, total soluble solid, and viable cell counts during fermentation under controlled conditions at 0-h, 24-h, 48-h, and 72-h fermentations were evaluated. The study found out that the change in pH and total soluble solid of probiotic germinated black rice juice significantly (p ≤ 0.05) decreased at 72-h fermentation (5.67±0.12 to 2.86±0.04 and 7.00±0.00 to 6.40±0.00 ºbrix at 0-h and 72-h fermentations, respectively). On the other hand, the amount of titratable acidity expressed as lactic acid and the viable cell count significantly (p≤0.05) increased at 72-h fermentation (0.11±0.06 to 0.43±0.06 (% lactic acid) and 3.60 x 106 to 2.75 x 108 CFU/ml at 0-h and 72-h fermentations, respectively). Interestingly, the amount of γ-Amino Butyric Acid (GABA) had a significant difference (p≤0.05) twice as high as that of the control group (0.25±0.01 and 0.13±0.01 mg/100g, respectively). In addition, the free radical scavenging capacity assayed by DPPH method also showed that the IC50 values were significantly (p≤0.05) higher than the control (147.71±0.96 and 202.55±1.24 mg/ml, respectively). After 4 weeks of cold storage at 4 °C, the viable cell counts of lactic acid bacteria reduced to 1.37 x 106 CFU/ml. In conclusion, fermented germinated native black rice juice could be served as a healthy beverage for vegans and people who are allergic to cow milk products.

399 Communities of Ammonia-oxidizing Archaea and Bacteria in Enriched Nitrifying Activated Sludge

Abstract:

In this study, communities of ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB) in nitrifying activated sludge (NAS) prepared by enriching sludge from a municipal wastewater treatment plant in three continuous-flow reactors receiving an inorganic medium containing different ammonium concentrations of 2, 10, and 30 mM NH4 +-N (NAS2, NAS10, and NAS30, respectively) were investigated using molecular analysis. Results suggested that almost all AOA clones from NAS2, NAS10, and NAS30 fell into the same AOA cluster and AOA communities in NAS2 and NAS10 were more diverse than those of NAS30. In contrast to AOA, AOB communities obviously shifted from the seed sludge to enriched NASs and in each enriched NAS, communities of AOB varied particularly. The seed sludge contained members of N. communis cluster and N. oligotropha cluster. After it was enriched under various ammonium loads, members of N. communis cluster disappeared from all enriched NASs. AOB with high affinity to ammonia presented in NAS 2, AOB with low affinity to ammonia presented in NAS 30, and both types of AOB survived in NAS 10. These demonstrated that ammonium load significantly influenced AOB communities, but not AOA communities in enriched NASs.

398 Non-Isolated Direct AC-DC Converter Design with BCM-PFC Circuit

Abstract:

This paper proposes two types of non-isolated direct AC-DC converters. First, it shows a buck-boost converter with an H-bridge, which requires few components (three switches, two diodes, one inductor and one capacitor) to convert AC input to DC output directly. This circuit can handle a wide range of output voltage. Second, a direct AC-DC buck converter is proposed for lower output voltage applications. This circuit is analyzed with output voltage of 12V. We describe circuit topologies, operation principles and simulation results for both circuits.

397 Study on Nitrite Accumulation Characteristics and Nitrifying Population Dynamics at Different Growth Environments

Abstract:

Novel nitrogen removal technologies via nitrite pathway attract increasing interest in recent years. In this study, batch experiments were performed to investigate nitrite accumulation characteristics and shifts in nitrifying community structure at different growth environments including ammonia concentration, pH and alkalinity. It was found that nitrite accumulation ratios were maintained at around 95% at studied conditions, and the optimum pH and Alk/N (ratio between alkalinity and nitrogen) for ammonium oxidization were 8.5 and 8.33, respectively. Fluorescence in situ hybridization analysis of nitrifying bacteria showed that high free ammonia (from influent ammonium or caused by high pH) significantly altered the structure of nitrifying community, leading to abundance of ammonia-oxidizing bacteria (AOB), especially Nitrososmonas, and inhibition of nitrite-oxidizing bacteria (NOB). The results suggest that free ammonia plays more important role than other studied conditions on nitrite accumulation.

396 Biodegradation of PCP by the Rhizobacteria Isolated from Pentachlorophenol-tolerant Crop Species

Abstract:

Pentachlorophenol (PCP) is a polychlorinated aromatic compound that is widespread in industrial effluents and is considered to be a serious pollutant. Among the variety of industrial effluents encountered, effluents from tanning industry are very important and have a serious pollution potential. PCP is also formed unintentionally in effluents of paper and pulp industries. It is highly persistent in soils and is lethal to a wide variety of beneficial microorganisms and insects, human beings and animals. The natural processes that breakdown toxic chemicals in the environment have become the focus of much attention to develop safe and environmentfriendly deactivation technologies. Microbes and plants are among the most important biological agents that remove and degrade waste materials to enable their recycling in the environment. The present investigation was carried out with the aim of developing a microbial system for bioremediation of PCP polluted soils. A number of plant species were evaluated for their ability to tolerate different concentrations of pentachlorophenol (PCP) in the soil. The experiment was conducted for 30 days under pot culture conditions. The toxic effect of PCP on plants was studied by monitoring seed germination, plant growth and biomass. As the concentration of PCP was increased to 50 ppm, the inhibition of seed germination, plant growth and biomass was also increased. Although PCP had a negative effect on all plant species tested, maize and groundnut showed the maximum tolerance to PCP. Other tolerating crops included wheat, safflower, sunflower, and soybean. From the rhizosphere soil of the tolerant seedlings, as many as twenty seven PCP tolerant bacteria were isolated. From soybean, 8; sunflower, 3; safflower 8; maize 2; groundnut and wheat, 3 each isolates were made. They were screened for their PCP degradation potentials. HPLC analyses of PCP degradation revealed that the isolate MAZ-2 degraded PCP completely. The isolate MAZ-1 was the next best isolate with 90 per cent PCP degradation. These strains hold promise to be used in the bioremediation of PCP polluted soils.

395 Antibacterial Activity of Some Medicinal Plant Extracts

Abstract:

Medicinal plants are now gaining attractiveness in treatment of bacterial infections and food preservation. The objective of this study was to assess antibacterial activity of some medicinal plants on pathogenic bacteria. Screening of antibacterial activity of aqueous and methanol extracts of some plants: Jojoba, Ginger, Sage, Thyme and Clove against Bacillus cereus, Salmonella typhimurium, Staphylococcus aureus, Clostridium perfringens and Escherichia coli were investigated. Antibacterial activity was performed by agar diffusion and disc diffusion method. Jatropha, Jojoba, Clove and Ginger extracts showed notable bacterial activity in the first screening step then selected to be tested against Bacillus cereus (Gram+), Staphylococcus aureus (Gram+) and Salmonella typhimurium (Gram−) and their effect was compared using antibiotics as control. Screening results showed potential antibacterial activity of the tested plant extracts against the screened bacterial strains. It was found that methanol extracts exhibited higher antibacterial activity than aqueous extracts. Methanol extract of Jatropha showed the highest inhibition zone against Staphylococcus aureus (Gram+) with 24.00 mm diameter, compared to the other plant extracts followed by clove. Meanwhile, the inhibition zones of methanol extracts of Jojoba and Ginger were the same (12mm).The Gram-positive bacteria were found to be more sensitive to aqueous and methanol extracts than Gram-negative bacteria.

394 Aureobasidium pullulans Used as a Biological Control Agent under Field Conditions Affects the Microbial Quality of Winter Wheat Grain

Abstract:

The biological activity of A. pullulans isolates against species of the genus Fusarium, bacteria of the genus Azotobacter and pseudomonads colonizing wheat kernels was evaluated. A field experiment was carried out in 2009-2011, in north-eastern Poland. Winter wheat (cv. Bogatka) plants were sprayed with a cell suspension of A. pullulans at a density of 106 - 108 per cm3 water at the stem elongation stage and the heading stage. Untreated plants served as control. The abundance of epiphytic yeasts, bacteria of the genus Azotobacter, pseudomonads and Fusarium pathogens on wheat grain was estimated at harvest and after six months’ storage. The average size of yeast communities was significantly greater on wheat kernels treated with a cell suspension of A. pullulans, compared with control samples. In 2010-2011, biological control reduced the abundance of some species of the genus Fusarium.

393 Influence of Yeast Strains on Microbiological Stability of Wheat Bread

Abstract:

Problem of food preservation is extremely important for mankind. Viscous damage ("illness") of bread results from development of Bacillus spp. bacteria. High temperature resistant spores of this microorganism are steady against 120°C) and remain in bread during pastries, potentially causing spoilage of the final product. Scientists are interested in further characterization of bread spoiling Bacillus spp. species. Our aim was to find weather yeast Saccharomyces cerevisiae strains that are able to produce natural antimicrobial killer factor can preserve bread illness. By diffusion method, we showed yeast antagonistic activity against spore-forming bacteria. Experimental technological parameters were the same as for bakers' yeasts production on the industrial scale. Risograph test during dough fermentation demonstrated gas production. The major finding of the study was a clear indication of the presence of killer yeast strain antagonistic activity against rope in bread causing bacteria. After demonstrating antagonistic effect of S. cerevisiae on bacteria using solid nutrient medium, we tested baked bread under provocative conditions. We also measured formation of carbon dioxide in the dough, dough-making duration and quality of the final products, when using different strains of S. cerevisiae. It is determined that the use of yeast S. cerevisiae RCAM 01730 killer strain inhibits appearance of rope in bread. Thus, natural yeast antimicrobial killer toxin, produced by some S. cerevisiae strains is an anti-rope in bread protector.

Repeated additions of the unfertilized bacteria led to increase the activity of Nitrogen-fixing bacteria in the root zone with drip irrigation system compared to traditional manual vaccination to increase the proportion of Nitrogen from 29% to 64%, and the efficiency of adding Nitrogen fertilizer did not exceed 9.5% while dropped to 4%, due to the amount of fertilizer added was not exceed 20kg N/h, and the second was the existence of a large amount of available Nitrogen in the soil by fixation, while the efficiency of irrigation system between 2.08 to 2.26 kg/m3.

391 Screening and Evaluation of in vivo and in vitro Generated Insulin Plant (Vernonia divergens) for Antimicrobial and Anticancer Activities

Abstract:

Vernonia divergens Benth., commonly known as “Insulin Plant” (Fam: Asteraceae) is a potent sugar killer. Locally the leaves of the plant, boiled in water are successfully administered to a large number of diabetic patients. The present study evaluates the putative anti-diabetic ingredients, isolated from the in vivo and in vitro grown plantlets of V. divergens for their antimicrobial and anticancer activities. Sterilized explants of nodal segments were cultured on MS (Musashige and Skoog, 1962) medium in presence of different combinations of hormones. Multiple shoots along with bunch of roots were regenerated at 1mg l-1 BAP and 0.5 mg l-1 NAA. Micro-plantlets were separated and sub-cultured on the double strength (2X) of the above combination of hormones leading to increased length of roots and shoots. These plantlets were successfully transferred to soil and survived well in nature. The ethanol extract of plantlets from both in vivo & in vitro sources were prepared in soxhlet extractor and then concentrated to dryness under reduced pressure in rotary evaporator. Thus obtainedconcentrated extracts showed significant inhibitory activity against gram negative bacteria like Escherichia coli and Pseudomonas aeruginosa but no inhibition was found against gram positive bacteria. Further, these ethanol extracts were screened for in vitro percentage cytotoxicity at different time periods (24 h, 48 h and 72 h) of different dilutions. The in vivo plant extract inhibited the growth of EAC mouse cell lines in the range of 65, 66, 78, and 88% at 100, 50, 25 & 12.5μg mL-1 but at 72 h of treatment. In case of the extract of in vitro origin, the inhibition was found against EAC cell lines even at 48h. During spectrophotometric scanning, the extracts exhibited different maxima (ʎ) - four peaks in in vitro extracts as against single in in vivo preparation suggesting the possible change in the nature of ingredients during micropropagation through tissue culture techniques.

390 Comparison between Antibacterial Effects of Ethanolic and Isopropyl: Hexan (7:3) Extracts of Zingiber officinale Rose

In this investigation, the antibacterial effects of ethanolic and 7:3 isopropyl –hexane mixture extracts of Zingiber officinale were evaluated against three Gram positive bacteria, B. cereus, S.epidermidis, S. aureus and three Gram negative bacteria, E. coli, K.pneumonia and P.areuginosa. Utilizing paper disk diffusion and well methods in-vitro, MIC and MBC were determined by macrodilution. The results showed that ethanolic rhizome extract of ginger had significantly active than Isopropyl –hexan extract. Further work needs to be done in these extracts including fractionation to isolate active constituents and subsequent pharmacological evaluation.

389 Influential Effect of Self-Healing Treatment on Water Absorption and Electrical Resistance of Normal and Light Weight Aggregate Concretes

Abstract:

Interest in using bacteria in cement materials due to its positive influences has been increased. Cement materials such as mortar and concrete basically suffer from higher porosity and water absorption compared to other building materials such as steel materials. Because of the negative side-effects of certain chemical techniques, biological methods have been proposed as a desired and environmentally friendly strategy for reducing concrete porosity and diminishing water absorption. This paper presents the results of an experimental investigation carried out to evaluate the influence of Sporosarcina pasteurii bacteria on the behaviour of two types of concretes (light weight aggregate concrete and normal weight concrete). The resistance of specimens to water penetration by testing water absorption and evaluating the electrical resistance of those concretes was examined and compared. As a conclusion, 20% increase in electrical resistance and 10% reduction in water absorption of lightweight aggregate concrete (LWAC) and for normal concrete the results show 7% decrease in water absorption and almost 10% increase in electrical resistance.
