1730 Cloning and Over Expression of an Aspergillus niger XP Phytase Gene (phyA) in Pichia pastoris

Abstract:
A. niger XP isolated from Vietnam produces very low amount of acidic phytase with optimal pH at 2.5 and 5.5. The phytase production of this strain was successfully improved through gene cloning and expression. A 1.4 - kb DNA fragment containing the coding region of the phyA gene was amplified by PCR and inserted into the expression vector pPICZαA with a signal peptide α- factor, under the control of AOX1 promoter. The recombined plasmid was transformed into the host strain P. pastoris KM71H and X33 by electroporation. Both host strains could efficiently express and secret phytase. The multicopy strains were screened for over expression of phytase. All the selected multicopy strains of P. pastoris X33 were examined for phytase activity, the maximum phytase yield of 1329 IU/ml was obtained after 4 days of incubation in medium BMM. The recombinant protein with MW of 97.4 KW showed to be the only one protein secreted in the culture broth. Multicopy transformant P. pastoris X33 supposed to be potential candidate for producing the commercial preparation of phytase.

Abstract:
In this paper, an automated system is presented for identification and separation of plastic resins based on near infrared (NIR) reflectance spectroscopy. For identification and separation among resins, a "Two-Filter" identification method is proposed that is capable to distinguish among polyethylene terephthalate (PET), high density polyethylene (HDPE), polyvinyl chloride (PVC), polypropylene (PP) and polystyrene (PS). Through surveying effects of parameters such as surface contamination, sample thickness, label and cap existence, it was obvious that the "Two-Filter" method has a high efficiency in identification of resins. It is shown that accurate identification and separation of five major resins can be obtained through calculating the relative reflectance at two wavelengths in the NIR region.Keywords: Identification, Near Infrared, Plastic, Separation, Spectroscopy

Abstract:
Pulse width modulation (PWM) techniques have been the subject of intensive research for different industrial and power sector applications. A large variety of methods, different in concept and performance, have been newly developed and described. This paper analyzes the comparative merits of Sinusoidal Pulse Width Modulation (SPWM) and Space Vector Pulse Width Modulation (SVPWM) techniques and the suitability of these techniques in a Shunt Active Filter (SAF). The objective is to select the scheme that offers effective utilization of DC bus voltage and also harmonic reduction at the input side. The effectiveness of the PWM techniques is tested in the SAF configuration with a non linear load. The performance of the SAF with the SPWM and (SVPWM) techniques are compared with respect to the THD in source current. The study reveals that in the context of closed loop SAF control with the SVPWM technique there is only a minor improvement in THD. The utilization of the DC bus with SVPWM is also not significant compared to that with SPWM because of the non sinusoidal modulating signal from the controller in SAF configuration.Keywords: Voltage source inverter, Shunt active filter, SPWM, SVPWM, Matlab/SIMULINK.

Abstract:
Palynology is a field of interest in many disciplines due to its multiple applications: chronological dating, climatology, allergy treatment, and honey characterization. Unfortunately, the analysis of a pollen slide is a complicated and time consuming task that requires the intervention of experts in the field, which are becoming increasingly rare due to economic and social conditions. In this context, the automation of this task is urgent. In this work, we compare classical feature extraction methods (Shape, GLCM, LBP, and others) and Deep Learning (CNN and Transfer Learning) to perform a recognition task over 80 regional pollen species. It has been found that the use of Transfer Learning seems to be more precise than the other approaches.

Abstract:
We present results from experimental price-setting oligopolies in which green firms undertake different levels of energy-saving investments motivated by public subsidies and demand-side advantages. We find that consumers reveal higher willingness to pay for greener sellers’ products. This observation in conjunction to the fact that greener sellers set higher prices is compatible with the use and interpretation of energy-saving behaviour as a differentiation strategy. However, sellers do not exploit the resulting advantage through sufficiently high price-cost margins, because they seem trapped into “run to stay still” competition. Regarding the use of public subsidies to energy-saving sellers we uncover an undesirable crowding-out effect of consumers’ intrinsic tendency to support green manufacturers. Namely, consumers may be less willing to support a green seller whose energy-saving strategy entails a direct financial benefit. Finally, we disentangle two alternative motivations for consumer’s attractions to pro-social firms; first, the self-interested recognition of the firm’s contribution to the public and private welfare and, second, the need to compensate a firm for the cost entailed in each pro-social action. Our results show the prevalence of the former over the latter.

Abstract:
The structure is made using different members and combining them with each other. These members are basically based on technical and engineering principles and are combined in different ways and have their own unique effects on the building. Trusses are one of the most common and important members of the structure, accounting for a large percentage of the power transmission structure in the building. Different types of trusses are based on structural needs and evaluating and making complete comparisons between them is one of the most important engineering analyses. In the present study, four types of trusses have been studied; 1) Hawe truss, 2) Pratt truss, 3) k truss, and 4) warren truss, under cyclic loading for 80 seconds. The trusses are modeled in 3d using st37 steel. The results showed that Hawe trusses had higher values than all other trusses (k, Pratt and Warren) in all the studied indicators. Indicators examined in the study include; 1) von Mises stresses, 2) displacement, 3) support force, 4) velocity, 5) acceleration, 6) capacity (hysteresis curve) and 7) energy diagram. Pratt truss in indicators; Mises stress, displacement, energy have the least amount compared to other trusses. K truss in indicators; support force, speed and acceleration are the lowest compared to other trusses.

Abstract:
The purpose of this paper is to investigate the relationship among the key macroeconomic variables and Islamic stock market in India. This study is based on the time series data of financial years 2009-2015 to explore the consistency of relationship between macroeconomic variables and Shariah Indices. The ADF (Augmented Dickey–Fuller Test Statistic) and PP (Phillips–Perron Test Statistic) tests are employed to check stationarity of the data. The study depicts the long run relationship between Shariah indices and macroeconomic variables by using the Johansen Co-integration test. BSE Shariah and Nifty Shariah have uni-direct Granger causality. The outcome of VECM is significantly confirming the applicability of best fitted model. Thus, Islamic stock indices are proficiently working for the development of Indian economy. This research study aims to present a retrospective study about speech recognition systems and artificial intelligence. Speech recognition has become one of the widely used technologies, as it offers great opportunity to interact and communicate with automated machines. Precisely, it can be affirmed that speech recognition facilitates its users and helps them to perform their daily routine tasks, in a more convenient and effective manner. This research intends to present the illustration of recent technological advancements, which are associated with artificial intelligence. Recent researches have revealed the fact that speech recognition is found to be the utmost issue, which affects the decoding of speech. In order to overcome these issues, different statistical models were developed by the researchers. Some of the most prominent statistical models include acoustic model (AM), language model (LM), lexicon model, and hidden Markov models (HMM). The research will help in understanding all of these statistical models of speech recognition. Researchers have also formulated different decoding methods, which are being utilized for realistic decoding tasks and constrained artificial languages. These decoding methods include pattern recognition, acoustic phonetic, and artificial intelligence. The paper presents the results of clusterization by Kohonen self-organizing maps (SOM) applied for analysis of array of Raman spectra of multi-component solutions of inorganic salts, for determination of types of salts present in the solution. It is demonstrated that use of SOM is a promising method for solution of clusterization and classification problems in spectroscopy of multicomponent objects, as attributing a pattern to some cluster may be used for recognition of component composition of the object.

Abstract:
This paper analyses the torsional efforts in gas turbine-generator shafts caused by high speed automatic reclosing of transmission lines. This issue is especially important for cases of three phase short circuit and unsuccessful reclosure of lines in the vicinity of the thermal plant. The analysis was carried out for the thermal plant TERMOPERNAMBUCO located on Northeast region of Brazil. It is shown that stress level caused by lines unsuccessful reclosing can be several times higher than terminal three-phase short circuit. Simulations were carried out with detailed shaft torsional model provided by machine manufacturer and with the “Alternative Transient Program – ATP" program [1]. Unsuccessful three phase reclosing for selected lines in the area closed to the plant indicated most critical cases. Also, reclosing first the terminal next to the gas turbine gererator will lead also to the most critical condition. Considering that the values of transient torques are very sensible to the instant of reclosing, simulation of unsuccessful reclosing with statistics ATP switch were carried out for determination of most critical transient torques for each section of the generator turbine shaft.

Classifying data hierarchically is an efficient approach to analyze data. Data is usually classified into multiple categories, or annotated with a set of labels. To analyze multi-labeled data, such data must be specified by giving a set of labels as a semantic range. There are some certain purposes to analyze data. This paper shows which multi-labeled data should be the target to be analyzed for those purposes, and discusses the role of a label against a set of labels by investigating the change when a label is added to the set of labels. Abstract:
This paper presents reliability indices evaluation of the rotor core magnetization of the induction motor operated as a self excited induction generator by using probability distribution approach and Monte Carlo simulation. Parallel capacitors with calculated minimum capacitive value across the terminals of the induction motor operated as a SEIG with unregulated shaft speed have been connected during the experimental study. A three phase, 4 poles, 50Hz, 5.5 hp, 12.3A, 230V induction motor coupled with DC Shunt Motor was tested in the electrical machine laboratory with variable reactive loads. Based on this experimental study, it is possible to choose a reliable induction machines operated as a SEIG for unregulated renewable energy application in remote area or where grid is not available. Failure density function, cumulative failure distribution function, survivor function, hazard model, probability of success and probability of failure for reliability evaluation of the three phase induction motor operating as a SEIG have been presented graphically in this paper.

Abstract:
The dielectric properties and ionic conductivity of novel "ceramic state" polymer electrolytes for high capacity lithium battery are characterized by Radio frequency and Microwave methods in two broad frequency ranges from 50 Hz to 20 KHz and 4 GHz to 40 GHz. This innovative solid polymer electrolyte which is highly ionic conductive (10-3 S/cm at room temperature) from -40oC to +150oC can be used in any battery application. Such polymer exhibits properties more like a ceramic rather than polymer. The various applied measurement methods produced accurate dielectric results for comprehensive analysis of electrochemical properties and ion transportation mechanism of this newly invented polymer electrolyte. Two techniques and instruments employing air gap measurement by Capacitance Bridge and in-waveguide measurement by vector network analyzer are applied to measure the complex dielectric spectra. The complex dielectric spectra are used to determine the complex alternating current electrical conductivity and thus the ionic conductivity.

In this paper we propose an intelligent agent approach to control the electric power grid at a smaller granularity in order to give it self-healing capabilities. We develop a method using the influence model to transform transmission substations into information processing, analyzing and decision making (intelligent behavior) units. We also develop a wireless communication method to deliver real-time uncorrupted information to an intelligent controller in a power system environment. A combined networking and information theoretic approach is adopted in meeting both the delay and error probability requirements. We use a mobile agent approach in optimizing the achievable information rate vector and in the distribution of rates to users (sensors). We developed the concept and the quantitative tools require in the creation of cooperating semiautonomous subsystems which puts the electric grid on the path towards intelligent and self-healing system.Keywords: Mobile agent, power system operation and control, real time, wireless communication.

Nowadays, manufacturing industries augment their production lines with modern machining centers backed by CAM software. Several attempts are being made to cut down the programming time for machining complex geometries. Special programs/software have been developed to generate the digital numerical data and to prepare NC programs by using suitable post-processors for different machines. By selecting the tools and manufacturing process then applying tool paths and NC program are generated. More and more complex mechanical parts that earlier were being cast and assembled/manufactured by other processes are now being machined. Majority of these parts require lots of pocketing operations and find their applications in die and mold, turbo machinery, aircraft, nuclear, defense etc. Pocketing operations involve removal of large quantity of material from the metal surface. The modeling of warm cast and clamping a piece of food processing parts which the used of Pro-E and MasterCAM® software. Pocketing operation has been specifically chosen for toolpath optimization. Then after apply Pocketing toolpath, Multi Tool Selection and Reduce Air Time give the results of software simulation time and experimental machining time.Keywords: Toolpath, part program, optimization, pocket.

Abstract:
This paper is mainly concerned with the application of a novel technique of data interpretation to the characterization and classification of measurements of plasma columns in Tokamak reactors for nuclear fusion applications. The proposed method exploits several concepts derived from soft computing theory. In particular, Artifical Neural Networks have been exploited to classify magnetic variables useful to determine shape and position of the plasma with a reduced computational complexity. The proposed technique is used to analyze simulated databases of plasma equilibria based on ITER geometry configuration. As well as demonstrating the successful recovery of scalar equilibrium parameters, we show that the technique can yield practical advantages compares with earlier methods.

Abstract:
In this paper we present an off line system for the recognition of the handwritten numeric chains. Our work is divided in two big parts. The first part is the realization of a recognition system of the isolated handwritten digits. In this case the study is based mainly on the evaluation of neural network performances, trained with the gradient back propagation algorithm. The used parameters to form the input vector of the neural network are extracted on the binary images of the digits by several methods: the distribution sequence, the Barr features and the centred moments of the different projections and profiles. The second part is the extension of our system for the reading of the handwritten numeric chains constituted of a variable number of digits. The vertical projection is used to segment the numeric chain at isolated digits and every digit (or segment) will be presented separately to the entry of the system achieved in the first part (recognition system of the isolated handwritten digits). Public sector tendering has traditionally been conducted using manual paper-based processes which are known to be inefficient, less transparent and more prone to manipulations and errors. The advent of the Internet and the World Wide Web has led to the development of numerous e-Tendering systems that addressed some of the problems associated with the manual paper-based tendering system. However, most of these systems rarely support the evaluation of tenders and where they do it is mostly based on the single decision maker which is not suitable in public sector tendering, where for the sake of objectivity, transparency, and fairness, it is required that the evaluation is conducted through a tender evaluation committee. Currently, in Nigeria, the public tendering process in general and the evaluation of tenders, in particular, are largely conducted using manual paper-based processes. Automating these manual-based processes to digital-based processes can help in enhancing the proficiency of public sector tendering in Nigeria. This paper is part of a larger study to develop an electronic tendering system that supports the whole tendering lifecycle based on Nigerian procurement law. Specifically, this paper presents the design and implementation of part of the system that supports group evaluation of tenders based on a technique called fuzzy multi-attributes group decision making. The system was developed using Object-Oriented methodologies and Unified Modelling Language and hypothetically applied in the evaluation of technical and financial proposals submitted by bidders. The system was validated by professionals with extensive experiences in public sector procurement. The results of the validation showed that the system called NPS-eTender has an average rating of 74% with respect to correct and accurate modelling of the existing manual tendering domain and an average rating of 67.6% with respect to its potential to enhance the proficiency of public sector tendering in Nigeria. Thus, based on the results of the validation, the automation of the evaluation process to support tender evaluation committee is achievable and can lead to a more proficient public sector tendering system.

Partial discharge (PD) detection is an important method to evaluate the insulation condition of metal-clad apparatus. Non-intrusive sensors which are easy to install and have no interruptions on operation are preferred in onsite PD detection. However, it often lacks of accuracy due to the interferences in PD signals. In this paper a novel PD extraction method that uses frequency analysis and entropy based time-frequency (TF) analysis is introduced. The repetitive pulses from convertor are first removed via frequency analysis. Then, the relative entropy and relative peak-frequency of each pulse (i.e. time-indexed vector TF spectrum) are calculated and all pulses with similar parameters are grouped. According to the characteristics of non-intrusive sensor and the frequency distribution of PDs, the pulses of PD and interferences are separated. Finally the PD signal and interferences are recovered via inverse TF transform. The de-noised result of noisy PD data demonstrates that the combination of frequency and time-frequency techniques can discriminate PDs from interferences with various frequency distributions.Keywords: Entropy, Fourier analysis, non-intrusive measurement, time-frequency analysis, partial discharge

Over the past few years, the idea of adaptive buildings and specifically, adaptive building energy management systems (ABEMS) has become popular. Well-performed management in terms of energy is to create a balance between energy consumption and user comfort; therefore, in new energy management models, efficient energy consumption is not the sole factor and the user's comfortability is also considered in the calculations. One of the main ways of measuring this factor is by analyzing user feedback on the conditions to understand whether they are satisfied with conditions or not. This paper provides a comprehensive review of recent approaches towards energy management systems based on users' feedbacks and subsequently performs a comparison between them premised upon their efficiency and accuracy to understand which approaches were more accurate and which ones resulted in a more efficient way of minimizing energy consumption while maintaining users' comfortability. It was concluded that the highest accuracy rate among the presented works was 95% accuracy in determining satisfaction and up to 51.08% energy savings can be achieved without disturbing user’s comfort. Considering the growing interest in designing and developing adaptive buildings, these studies can support diverse inquiries about this subject and can be used as a resource to support studies and researches towards efficient energy consumption while maintaining the comfortability of users.

Abstract:
The development of machine learning methods and techniques has opened doors for investigation in many areas such as medicines, economics, finance, etc. One active research area involving machine learning is stock market prediction. This research paper tries to consider multiple techniques and methods for stock movement prediction using historical price or price factors. The paper explores the effectiveness of some deep learning frameworks for forecasting stock. Moreover, an architecture (TimeStock) is proposed which takes the representation of time into account apart from the price information itself. Our model achieves a promising result that shows a potential approach for the stock movement prediction problem.

Abstract:
In conventional reliability assessment, the reliability data of system components are treated as crisp values. The collected data have some uncertainties due to errors by human beings/machines or any other sources. These uncertainty factors will limit the understanding of system component failure due to the reason of incomplete data. In these situations, we need to generalize classical methods to fuzzy environment for studying and analyzing the systems of interest. Fuzzy set theory has been proposed to handle such vagueness by generalizing the notion of membership in a set. Essentially, in a Fuzzy Set (FS) each element is associated with a point-value selected from the unit interval [0, 1], which is termed as the grade of membership in the set. A Vague Set (VS), as well as an Intuitionistic Fuzzy Set (IFS), is a further generalization of an FS. Instead of using point-based membership as in FS, interval-based membership is used in VS. The interval-based membership in VS is more expressive in capturing vagueness of data. In the present paper, vague set theory coupled with conventional Lambda-Tau method is presented for reliability analysis of repairable systems. The methodology uses Petri nets (PN) to model the system instead of fault tree because it allows efficient simultaneous generation of minimal cuts and path sets. The presented method is illustrated with the press unit of the paper mill.

Abstract:
The paper presents a new drugs delivery system, based on the thin film technology. As a model antitumor drug, highly toxic doxorubicin is chosen. The system is based on the technology of obtaining zinc oxide composite of doxorubicin by deposition of nanosize ZnO films on the surface of doxorubicin coating on glass substrate using DC magnetron sputtering of zinc targets in Ar:O2 medium at room temperature. For doxorubicin zinc oxide compositions in the form of coatings and gels with 180-200nm thick ZnO films, higher (by a factor 2) in vivo (ascitic Ehrlich's carcinoma) antitumor activity is observed at low doses of doxorubicin in comparison with that of the initial preparation at therapeutic doses. The vector character of the doxorubicin zinc oxide composite transport to tumor tissues ensures the increase in antitumor activity as well as decrease of toxicity in comparison with the initial drug.

Abstract:
Modeling of the dynamic behavior and motion are renewed interest in the improved tractive performance of an intelligent air-cushion tracked vehicle (IACTV). This paper presents a new dynamical model for the forces on the developed small scale intelligent air-cushion tracked vehicle moving over swamp peat. The air cushion system partially supports the 25 % of vehicle total weight in order to make the vehicle ground contact pressure 7 kN/m2. As the air-cushion support system can adjust automatically on the terrain, so the vehicle can move over the terrain without any risks. The springdamper system is used with the vehicle body to control the aircushion support system on any undulating terrain by making the system sinusoidal form. Experiments have been carried out to investigate the relationships among tractive efficiency, slippage, traction coefficient, load distribution ratio, tractive effort, motion resistance and power consumption in given terrain conditions. Experiment and simulation results show that air-cushion system improves the vehicle performance by keeping traction coefficient of 71% and tractive efficiency of 62% and the developed model can meet the demand of transport efficiency with the optimal power consumption.Keywords: Air-cushion system, ground contact pressure, slippage, power consumption.

Abstract:
An adaptive Chinese hand-talking system is presented in this paper. By analyzing the 3 data collecting strategies for new users, the adaptation framework including supervised and unsupervised adaptation methods is proposed. For supervised adaptation, affinity propagation (AP) is used to extract exemplar subsets, and enhanced maximum a posteriori / vector field smoothing (eMAP/VFS) is proposed to pool the adaptation data among different models. For unsupervised adaptation, polynomial segment models (PSMs) are used to help hidden Markov models (HMMs) to accurately label the unlabeled data, then the "labeled" data together with signerindependent models are inputted to MAP algorithm to generate signer-adapted models. Experimental results show that the proposed framework can execute both supervised adaptation with small amount of labeled data and unsupervised adaptation with large amount of unlabeled data to tailor the original models, and both achieve improvements on the performance of recognition rate.Keywords: sign language recognition, signer adaptation, eMAP/VFS, polynomial segment model.

Abstract:
A mobile ad hoc network (MANET) represents a system of wireless mobile nodes that can self-organize freely and dynamically into arbitrary and temporary network topology. Unlike a wired network, wireless network interface has limited transmission range. Routing is the task of forwarding data packets from source to a given destination. Ad-hoc On Demand Distance Vector (AODV) routing protocol creates a path for a destination only when it required. This paper describes the implementation of AODV routing protocol using MATLAB-based Truetime simulator. In MANET's node movements are not fixed while they are random in nature. Hence intelligent techniques i.e. fuzzy and ANFIS are used to optimize the transmission range. In this paper, we compared the transmission range of AODV, fuzzy AODV and ANFIS AODV. For soft computing AODV, we have taken transmitted power and received threshold as input and transmission range as output. Abstract:
The treatment of the geotechnical database of the region of Casablanca was difficult to achieve due to the heterogeneity of the nomenclature of the lithological formations composing its soil. It appears necessary to harmonize the nomenclature of the facies and to produce cartographic documents useful for construction projects and studies before any investment program. To achieve this, more than 600 surveys made by the Public Laboratory for Testing and Studies (LPEE) in the agglomeration of Casablanca, were studied. Moreover, some local observations were made in different places of the metropolis. Each survey was the subject of a sheet containing lithological succession, macro and microscopic description of petrographic facies with photographic illustration, as well as measurements of geomechanical tests. In addition, an X-ray diffraction analysis was made in order to characterize the surficial formations of the region.

Abstract:
SoftBoost is a recently presented boosting algorithm, which trades off the size of achieved classification margin and generalization performance. This paper presents a performance evaluation of SoftBoost algorithm on the generic object recognition problem. An appearance-based generic object recognition model is used. The evaluation experiments are performed using a difficult object recognition benchmark. An assessment with respect to different degrees of label noise as well as a comparison to the well known AdaBoost algorithm is performed. The obtained results reveal that SoftBoost is encouraged to be used in cases when the training data is known to have a high degree of noise.
