474 Utilization of 3-N-trimethylamino-1-propanol by Rhodococcus sp. strain A4 isolated from Natural Soil

Authors: Isam A. Mohamed Ahmed, Jiro Arima, Tsuyoshi Ichiyanagi, Emi Sakuno, Nobuhiro Mori

Abstract:

The aim of this study was to screen for microorganism that able to utilize 3-N-trimethylamino-1-propanol (homocholine) as a sole source of carbon and nitrogen. The aerobic degradation of homocholine has been found by a gram-positive Rhodococcus sp. bacterium isolated from soil. The isolate was identified as Rhodococcus sp. strain A4 based on the phenotypic features, physiologic and biochemical characteristics, and phylogenetic analysis. The cells of the isolated strain grown on both basal-TMAP and nutrient agar medium displayed elementary branching mycelia fragmented into irregular rod and coccoid elements. Comparative 16S rDNA sequencing studies indicated that the strain A4 falls into the Rhodococcus erythropolis subclade and forms a monophyletic group with the type-strains of R. opacus, and R. wratislaviensis. Metabolites analysis by capillary electrophoresis, fast atom bombardment-mass spectrometry, and gas chromatography- mass spectrometry, showed trimethylamine (TMA) as the major metabolite beside β-alanine betaine and trimethylaminopropionaldehyde. Therefore, the possible degradation pathway of trimethylamino propanol in the isolated strain is through consequence oxidation of alcohol group (-OH) to aldehyde (-CHO) and acid (-COOH), and thereafter the cleavage of β-alanine betaine C-N bonds yielded trimethylamine and alkyl chain.

473 Structural Behavior of Lightweight Concrete Made With Scoria Aggregates and Mineral Admixtures

Authors: M. Shannag, A. Charif, S. Naser, F. Faisal, A. Karim

Abstract:

Structural lightweight concrete is used primarily to reduce the dead-load weight in concrete members such as floors in high-rise buildings and bridge decks. With given materials, it is generally desired to have the highest possible strength/unit weight ratio with the lowest cost of concrete. The work presented herein is part of an ongoing research project that investigates the properties of concrete mixes containing locally available Scoria lightweight aggregates and mineral admixtures. Properties considered included: workability, unit weight, compressive strength, and splitting tensile strength. Test results indicated that developing structural lightweight concretes (SLWC) using locally available Scoria lightweight aggregates and specific blends of silica fume and fly ash seems to be feasible. The stress-strain diagrams plotted for the structural LWC mixes developed in this investigation were comparable to a typical stress-strain diagram for normal weight concrete with relatively larger strain capacity at failure in case of LWC.

Authors: Togzhan D. Mukasheva, Anel A. Omirbekova, Raikhan S. Sydykbekova, Ramza Zh. Berzhanova, Lyudmila V. Ignatova

Abstract:

Following plants-barley (Hordeum sativum), alfalfa (Medicago sativa), grass mixture (red fescue-75%, long-term ryegrass - 20% Kentucky bluegrass - 10%), oilseed rape (Brassica napus biennis), resistant to growth in the contaminated soil with oil content of 15.8 g / kg 25.9 g / kg soil were used. Analysis of the population showed that the oil pollution reduces the number of bacteria in the rhizosphere and rhizoplane of plants and enhances the amount of spore-forming bacteria and saprotrophic micromycetes. It was shown that regardless of the plant, dominance of Pseudomonas and Bacillus genera bacteria was typical for the rhizosphere and rhizoplane of plants. The frequency of bacteria of these genera was more than 60%. Oil pollution changes the ratio of occurrence of various types of bacteria in the rhizosphere and rhizoplane of plants. Besides the Pseudomonas and Bacillus genera, in the presence of hydrocarbons in the root zone of plants dominant and most typical were the representatives of the Mycobacterium and Rhodococcus genera. Together the number was between 62% to 72%.

Abstract:

Researchers investigate arious strategies to develop composite beams and maximize the structural advantages. This study attempted to conduct experiments and analysis of changes in the neutral axis of positive moments of a Green Beam. Strain compatibility analysis was used, and its efficiency was demonstrated by comparing experimental and analytical values. In the comparison of neutral axis, the difference between experimental and analytical values was found to range from 8.8~26.2%. It was determined that strain compatibility analysis can be useful for predicting the behaviors of composite beams, with the ability to predict the behavior of not only the elastic location of the composite member, but also of the plastic location

Abstract:

The strain intensity and redundant strains, dependent in multistage TRIP wire drawing processes from values used single partial reductions, should influence on the intensity of transformation the retained austenite into martensite and thereby on mechanical properties of drawn wires. The numerical analysis of drawing processes with use of Drawing 2D programme, for steel wires made from TRIP steel with 0,29% has been shown in the work. The change of strain intensity εc and the values of redundant strain εxy, has been determined for particular draws in dependence of used single partial reductions.

469 Re-Examination of Louis Pasteur’s S-Shaped Flask Experiment

Authors: Ming-Hua Fu

Abstract:

No negative control nor control to prevent microbes from escaping was set when the S-shaped flask experiments were performed by Pasteur. Microscope was not used to observe the media in the flasks. Louis Pasteur’s S-shaped flask experiment was re-examined by using U-shaped flasks, modified S-shaped flasks and microscope. A mixture of microbes was isolated from the room air, from which one rod-shaped Bacillus species with proposed name Bacillus gaso-mobilis sp nov and one grape-shaped Staphylococcus species with proposed name of Staphylococcus gaso-mobilis sp nov were identified. Their penicillin and ampicillin resistant strains containing plasmids were isolated. These bacteria could change color, produce odor and automatically move in the air. They did not form colonies on solid media. They had a high suspension capacity in liquid media. Their light absorbance peaked at the wave length of 320 nm.  It was concluded that there were flaws with Louis Pasteur’s S-shaped flask experiments.

468 Modelling, Simulation and Validation of Plastic Zone Size during Deformation of Mild Steel

Authors: S. O. Adeosun, E. I. Akpan, S. A. Balogun, O. O. Taiwo

Abstract:

A model to predict the plastic zone size for material under plane stress condition has been developed and verified experimentally. The developed model is a function of crack size, crack angle and material property (dislocation density). Simulation and validation results show that the model developed show good agreement with experimental results. Samples of low carbon steel (0.035%C) with included surface crack angles of 45o, 50o, 60o, 70o and 90o and crack depths of 2mm and 4mm were subjected to low strain rate between 0.48 x 10-3 s-1 – 2.38 x 10-3 s-1. The mechanical properties studied were ductility, tensile strength, modulus of elasticity, yield strength, yield strain, stress at fracture and fracture toughness. The experimental study shows that strain rate has no appreciable effect on the size of plastic zone while crack depth and crack angle plays an imperative role in determining the size of the plastic zone of mild steel materials.

467 Strain Based Evaluation of Dents in Pressurized Pipes

Authors: Maziar Ramezani, Thomas Neitzert

Abstract:

A dent is a gross distortion of the pipe cross-section. Dent depth is defined as the maximum reduction in the diameter of the pipe compared to the original diameter. Pipeline dent finite element (FE) simulation and theoretical analysis are conducted in this paper to develop an understanding of the geometric characteristics and strain distribution in the pressurized dented pipe. Based on the results, the magnitude of the denting force increases significantly with increasing the internal pressure, and the maximum circumferential and longitudinal strains increase by increasing the internal pressure and the dent depth. The results can be used for characterizing dents and ranking their risks to the integrity of a pipeline.

466 High Strain Rate Characteristics of the Advanced Blast Energy Absorbers

Authors: Martina Drdlová, Michal Frank, Jaroslav Buchar, Josef Krátký

Abstract:

The main aim of the presented experiments is to improve behaviour of sandwich structures under dynamic loading, such as crash or explosion. Several cellular materials are widely used as core of the sandwich structures and their properties influence the response of the entire element under impact load. To optimize their performance requires the characterisation of the core material behaviour at high strain rates and identification of the underlying mechanism. This work presents the study of high strain-rate characteristics of a specific porous lightweight blast energy absorbing foam using a Split Hopkinson Pressure Bar (SHPB) technique adapted to perform tests on low strength materials. Two different velocities, 15 and 30 m.s-1 were used to determine the strain sensitivity of the material. Foams were designed using two types of porous lightweight spherical raw materials with diameters of 30- 100 *m, combined with polymer matrix. Cylindrical specimens with diameter of 15 mm and length of 7 mm were prepared and loaded using a Split Hopkinson Pressure Bar apparatus to assess the relation between the composition of the material and its shock wave attenuation capacity.

465 Effect of Heat Treatment on the Portevin-Le Chatelier Effect of Al-2.5%Mg Alloy

Authors: A. Chatterjee, A. Sarkar, N. Gayathri, P. Mukherjee, P. Barat

Abstract:

An experimental study is presented on the effect of microstructural change on the Portevin-Le Chatelier effect behaviour of Al-2.5%Mg alloy. Tensile tests are performed on the as received and heat treated (at 400 ºC for 16 hours) samples for a wide range of strain rates. The serrations observed in the stress-time curve are investigated from statistical analysis point of view. Microstructures of the samples are characterized by optical metallography and X-ray diffraction. It is found that the excess vacancy generated due to heat treatment leads to decrease in the strain rate sensitivity and the increase in the number of stress drop occurrences per unit time during the PLC effect. The microstructural parameters like domain size, dislocation density have no appreciable effect on the PLC effect as far as the statistical behavior of the serrations is considered.

464 Influence of Inter-tube Connections on the Stress-Strain Behavior of Nanotube-Polymer Composites: Molecular Dynamics

Authors: Jianwei Zhang, Dazhi Jiang, Huaxin Peng, Chunqi Wang

Abstract:

Stress-strain curve of inter-tube connected carbon nanotube (CNT) reinforced polymer composite under axial loading generated from molecular dynamics simulation is presented. Comparison of the response to axial mechanical loading between this composite system with composite systems reinforced by long, continuous CNTs (replicated via periodic boundary conditions) and short, discontinuous CNTs has been made. Simulation results showed that the inter-tube connection improved the mechanical properties of short discontinuous CNTs dramatically. Though still weaker than long CNT/polymer composite, more remarkable increase in the stiffness relative to the polymer was observed in the inter-tube connected CNT/polymer composite than in the discontinuous CNT/polymer composite. The manually introduced bridge break process resulted in a stress-strain curve of ductile fracture mode, which is consistent with the experimental result.

463 Numerical Simulation of Fiber Bragg Grating Spectrum for Mode-І Delamination Detection

Authors: O. Hassoon, M. Tarfoui, A. El Malk

Abstract:

Fiber Bragg optic sensor is embedded in composite material to detect and monitor the damage that occurs in composite structures. In this paper, we deal with the mode-Ι delamination to determine the material strength to crack propagation, using the coupling mode theory and T-matrix method to simulate the FBGs spectrum for both uniform and non-uniform strain distribution. The double cantilever beam test is modeled in FEM to determine the longitudinal strain. Two models are implemented, the first is the global half model, and the second is the sub-model to represent the FBGs with higher refined mesh. This method can simulate damage in composite structures and converting strain to a wavelength shifting in the FBG spectrum.

462 Numerical Study of Fiber Bragg Grating Sensor: Longitudinal and Transverse Detection of Temperature and Strain

Authors: K. Khelil, H. Ammar, K. Saouchi

Abstract:

Fiber Bragg Grating (FBG) structure is an periodically modulated optical fiber. It acts as a selective filter of wavelength whose reflected peak is called Bragg wavelength and it depends on the period of the fiber and the refractive index. The simulation of FBG is based on solving the Coupled Mode Theory equation by using the Transfer Matrix Method which is carried out using MATLAB. It is found that spectral reflectivity is shifted when the change of temperature and strain is uniform. Under non-uniform temperature or strain perturbation, the spectrum is both shifted and destroyed. In case of transverse loading, reflectivity spectrum is split into two peaks, the first is specific to X axis, and the second belongs to Y axis. FBGs are used in civil engineering to detect perturbations applied to buildings.

Abstract:

In this study, a general approach to the reliability based limit analysis of laterally loaded piles is presented. In engineering practice the uncertainties play a very important role. The aim of this study is to evaluate the lateral load capacity of free-head and fixed-head long pile when plastic limit analysis is considered. In addition to the plastic limit analysis to control the plastic behaviour of the structure, uncertain bound on the complementary strain energy of the residual forces is also applied. This bound has significant effect for the load parameter. The solution to reliability-based problems is obtained by a computer program which is governed by the reliability index calculation.

460 High Temperature Deformation Behavior of Cr-containing Superplastic Iron Aluminide

Authors: Seok Hong Min, Woo Young Jung, Tae Kwon Ha

Abstract:

Superplastic deformation and high temperature load relaxation behavior of coarse-grained iron aluminides with the composition of Fe-28 at.% Al have been investigated. A series of load relaxation and tensile tests were conducted at temperatures ranging from 600 to 850oC. The flow curves obtained from load relaxation tests were found to have a sigmoidal shape and to exhibit stress vs. strain rate data in a very wide strain rate range from 10-7/s to 10-2/s. Tensile tests have been conducted at various initial strain rates ranging from 3×10-5/s to 1×10-2/s. Maximum elongation of ~500 % was obtained at the initial strain rate of 3×10-5/s and the maximum strain rate sensitivity was found to be 0.68 at 850oC in binary Fe-28Al alloy. Microstructure observation through the optical microscopy (OM) and the electron back-scattered diffraction (EBSD) technique has been carried out on the deformed specimens and it has revealed the evidences for grain boundary migration and grain refinement to occur during superplastic deformation, suggesting the dynamic recrystallization mechanism. The addition of Cr by the amount of 5 at.% appeared to deteriorate the superplasticity of the binary iron aluminide. By applying the internal variable theory of structural superplasticity, the addition of Cr has been revealed to lower the contribution of the frictional resistance to dislocation glide during high temperature deformation of the Fe3Al alloy.

459 Elastic Strain-Concentration Factor of Cylindrical Bars with Circumferential Flat-Bottom Groove under Static Tension

Authors: Hitham M. Tlilan

Abstract:

Using finite element method (FEM), the elastic new strain-concentration factor (SNCF) of cylindrical bars with circumferential flat-bottom groove is studied. This new SNCF has been defined under triaxial stress state. The employed specimens have constant groove depth with net section and gross diameters of 10.0 and 16.7 mm, respectively. The length of flatness ao has been varied form 0.0 ~12.5 mm to study the elastic SNCF of this type of geometrical irregularities. The results that the elastic new SNCF rapidly drops from its elastic value of the groove with ao = 0.0, i.e. circumferential U-notch, and reaches minimum value at ao = 2 mm. After that the elastic new SNCF becomes nearly constant with increasing flatness length (ao). The value of tensile load at yielding at the groove root increases with increasing ao. The current results show that severity of the notch decreases with increasing flatness length ao.

Authors: Rosarin Rujananon, Poonsuk Prasertsan, Amornrat Phongdara, Tanate Panrat, Jibin Sun, Sugima Rappert, An-Ping Zeng

Abstract:

In this study, a synthetic pathway was created by assembling genes from Clostridium butyricum and Escherichia coli in different combinations. Among the genes were dhaB1 and dhaB2 from C. butyricum VPI1718 coding for glycerol dehydratase (GDHt) and its activator (GDHtAc), respectively, involved in the conversion of glycerol to 3-hydroxypropionaldehyde (3-HPA). The yqhD gene from E.coli BL21 was also included which codes for an NADPHdependent 1,3-propanediol oxidoreductase isoenzyme (PDORI) reducing 3-HPA to 1,3-propanediol (1,3-PD). Molecular modeling analysis indicated that the conformation of fusion protein of YQHD and DHAB1 was favorable for direct molecular channeling of the intermediate 3-HPA. According to the simulation results, the yqhD and dhaB1 gene were assembled in the upstream of dhaB2 to express a fusion protein, yielding the recombinant strain E. coliBL21 (DE3)//pET22b+::yqhD-dhaB1_dhaB2 (strain BP41Y3). Strain BP41Y3 gave 10-fold higher 1,3-PD concentration than E. coliBL21 (DE3)//pET22b+::yqhD-dhaB1_dhaB2 (strain BP31Y2) expressing the recombinant enzymes simultaneously but in a non-fusion mode. This is the first report using a gene fusion approach to enhance the biological conversion of glycerol to the value added compound 1,3- PD.

Abstract:

Each year nearly nine hundred persons die in head injuries and over fifty thousand persons are severely injured due to non wearing of helmets. In motor cycle accidents, the human head is exposed to heavy impact loading against natural protection. In this work, an attempt has been made for analyzing the helmet with all the standard data. The simulation software ‘ANSYS’ is used to analyze the helmet with different conditions such as bottom fixed-load on top surface, bottom fixed -load on top line, side fixed –load on opposite surface, side fixed-load on opposite line and dynamic analysis. The maximum force of 19.5 kN is applied on the helmet to study the model in static and dynamic conditions. The simulation has been carried out for the static condition for the parameters like total deformation, strain energy, von-Mises stress for different cases. The dynamic analysis has been performed for the parameter like total deformation and equivalent elastic strain. The result shows that these values are concentrated in the retention portion of the helmet. These results have been compared with the standard experimental data proposed by the BIS and well within the acceptable limit.

455 The Effect of Maximum Strain on Fatigue Life Prediction for Natural Rubber Material

Authors: Chang S. Woo, Hyun S. Park, Wan D. Kim

Abstract:

Fatigue life prediction and evaluation are the key technologies to assure the safety and reliability of automotive rubber components. The objective of this study is to develop the fatigue analysis process for vulcanized rubber components, which is applicable to predict fatigue life at initial product design step. Fatigue life prediction methodology of vulcanized natural rubber was proposed by incorporating the finite element analysis and fatigue damage parameter of maximum strain appearing at the critical location determined from fatigue test. In order to develop an appropriate fatigue damage parameter of the rubber material, a series of displacement controlled fatigue test was conducted using threedimensional dumbbell specimen with different levels of mean displacement. It was shown that the maximum strain was a proper damage parameter, taking the mean displacement effects into account. Nonlinear finite element analyses of three-dimensional dumbbell specimens were performed based on a hyper-elastic material model determined from the uni-axial tension, equi-biaxial tension and planar test. Fatigue analysis procedure employed in this study could be used approximately for the fatigue design.

Abstract:

Champs Bourcin black grape originated from Aquitaine, France and planted in Sapa, Lao cai provice, exhibited high total acidity (11.72 g/L). After 9 days of alcoholic fermentation at 25oC using Saccharomyces cerevisiae UP3OY5 strain, the ethanol concentration of wine was 11.5% v/v, however the sharp sour taste of wine has been found. The malolactic fermentation (MLF) was carried out by Oenococcus oeni ATCCBAA-1163 strain which had been preadapted to acid (pH 3-4) and ethanol (8-12%v/v) conditions. We obtained the highest vivability (83.2%) upon malolactic fermentation after 5 days at 22oC with early stationary phase O. oeni cells preadapted to pH 3.5 and 8% v/v ethanol in MRS medium. The malic acid content in wine was decreased from 5.82 g/L to 0.02 g/L after MLF (21 days at 22oC). The sensory quality of wine was significantly improved.

453 Image Processing on Geosynthetic Reinforced Layers to Evaluate Shear Strength and Variations of the Strain Profiles

Authors: S. K. Khosrowshahi, E. Güler

Abstract:

This study investigates the reinforcement function of geosynthetics on the shear strength and strain profile of sand. Conducting a series of simple shear tests, the shearing behavior of the samples under static and cyclic loads was evaluated. Three different types of geosynthetics including geotextile and geonets were used as the reinforcement materials. An image processing analysis based on the optical flow method was performed to measure the lateral displacements and estimate the shear strains. It is shown that besides improving the shear strength, the geosynthetic reinforcement leads a remarkable reduction on the shear strains. The improved layer reduces the required thickness of the soil layer to resist against shear stresses. Consequently, the geosynthetic reinforcement can be considered as a proper approach for the sustainable designs, especially in the projects with huge amount of geotechnical applications like subgrade of the pavements, roadways, and railways.

452 Shape Sensing and Damage Detection of Thin-Walled Cylinders Using an Inverse Finite Element Method

Authors: Ionel D. Craiu, Mihai Nedelcu

Abstract:

Thin-walled cylinders are often used by the offshore industry as columns of floating installations. Based on observed strains, the inverse Finite Element Method (iFEM) may rebuild the deformation of structures. Structural Health Monitoring uses this approach extensively. However, the number of in-situ strain gauges is what determines how accurate it is, and for shell structures with complicated deformation, this number can easily become too high for practical use. Any thin-walled beam member's complicated deformation can be modeled by the Generalized Beam Theory (GBT) as a linear combination of pre-specified cross-section deformation modes. GBT uses bar finite elements as opposed to shell finite elements. This paper proposes an iFEM/GBT formulation for the shape sensing of thin-walled cylinders based on these benefits. This method significantly reduces the number of strain gauges compared to using the traditional inverse-shell finite elements. Using numerical simulations, dent damage detection is achieved by comparing the strain distributions of the undamaged and damaged members. The effect of noise on strain measurements is also investigated.

451 Application of Acinetobacter sp. KKU44 for Cellulase Production from Agricultural Waste

Authors: Surasak Siripornadulsil, Nutt Poomai, Wilailak Siripornadulsil

Abstract:

Due to a high ethanol demand, the approach for  effective ethanol production is important and has been developed  rapidly worldwide. Several agricultural wastes are highly  abundant in celluloses and the effective cellulase enzymes do exist  widely among microorganisms. Accordingly, the cellulose  degradation using microbial cellulase to produce a low-cost substrate  for ethanol production has attracted more attention. In this  study, the cellulase producing bacterial strain has been isolated  from rich straw and identified by 16S rDNA sequence analysis as Acinetobacter sp. KKU44. This strain is able to grow and exhibit the cellulase activity. The optimal temperature for its growth and  cellulase production is 37°C. The optimal temperature of bacterial  cellulase activity is 60°C. The cellulase enzyme from  Acinetobacter sp. KKU44 is heat-tolerant enzyme. The bacterial culture of 36h. showed highest cellulase activity at 120U/mL when  grown in LB medium containing 2% (w/v). The capability of  Acinetobacter sp. KKU44 to grow in cellulosic agricultural wastes as a sole carbon source and exhibiting the high cellulase activity at high temperature suggested that this strain could be potentially developed further as a cellulose degrading strain for a production of low-cost substrate used in ethanol production.

450 Effects of Ophiocordyceps dipterigena BCC 2073 β-Glucan as a Prebiotic on the in vitro Growth of Probiotic and Pathogenic Bacteria

Authors: Wai Prathumpai, Pranee Rachtawee, Sutamat Khajeeram, Pariya Na Nakorn

Abstract:

The  β-glucan produced by Ophiocordyceps dipterigena BCC 2073 is a (1, 3)-β-D-glucan with highly branching O-6-linkedside chains that is resistant to acid hydrolysis (by hydrochloric acid and porcine pancreatic alpha-amylase). This β-glucan can be utilized as a prebiotic due to its advantageous structural and biological properties. The effects of using this β-glucan as the sole carbon source for the in vitro growth of two probiotic bacteria (L. acidophilus BCC 13938 and B. animalis ATCC 25527) were investigated. Compared with the effect of using 1% glucose or fructo-oligosaccharide (FOS) as the sole carbon source, using 1% β-glucan for this purpose showed that this prebiotic supported and stimulated the growth of both types of probiotic bacteria and induced them to produce the highest levels of metabolites during their growth. The highest levels of lactic and acetic acid, 10.04 g·L-1 and 2.82 g·L-1, respectively, were observed at 2 h of cultivation using glucose as the sole carbon source. Furthermore, the fermentation broth obtained using 1% β-glucan as the sole carbon source had greater antibacterial activity against selected pathogenic bacteria (B. subtilis TISTR 008, E. coli TISTR 780, and S. typhimurium TISTR 292) than did the broths prepared using glucose or FOS as the sole carbon source. The fermentation broth obtained by growing L. acidophilus BCC 13938 in the presence of β-glucan inhibited the growth of B. subtilis TISTR 008 by more than 70% and inhibited the growth of both S. typhimurium TISTR 292 and E. coli TISTR 780 by more than 90%. In conclusion, O. dipterigena BCC 2073 is a potential source of a β-glucan prebiotic that could be used for commercial production in the near future.

449 Equilibrium, Kinetic and Thermodynamic Studies on Biosorption of Cd (II) and Pb (II) from Aqueous Solution Using a Spore Forming Bacillus Isolated from Wastewater of a Leather Factory

Authors: Sh. Kianfar, A. Moheb, H. Ghaforian

Abstract:

The equilibrium, thermodynamics and kinetics of the biosorption of Cd (II) and Pb(II) by a Spore Forming Bacillus (MGL 75) were investigated at different experimental conditions. The Langmuir and Freundlich, and Dubinin-Radushkevich (D-R) equilibrium adsorption models were applied to describe the biosorption of the metal ions by MGL 75 biomass. The Langmuir model fitted the equilibrium data better than the other models. Maximum adsorption capacities q max for lead (II) and cadmium (II) were found equal to 158.73mg/g and 91.74 mg/g by Langmuir model. The values of the mean free energy determined with the D-R equation showed that adsorption process is a physiosorption process. The thermodynamic parameters Gibbs free energy (ΔG°), enthalpy (ΔH°), and entropy (ΔS°) changes were also calculated, and the values indicated that the biosorption process was exothermic and spontaneous. Experiment data were also used to study biosorption kinetics using pseudo-first-order and pseudo-second-order kinetic models. Kinetic parameters, rate constants, equilibrium sorption capacities and related correlation coefficients were calculated and discussed. The results showed that the biosorption processes of both metal ions followed well pseudo-second-order kinetics.

448 Numerical Simulation on Deformation Behaviour of Additively Manufactured AlSi10Mg Alloy

Authors: Racholsan Raj Nirmal, B. S. V. Patnaik, R. Jayaganthan

Abstract:

The deformation behaviour of additively manufactured AlSi10Mg alloy under low strains, high strain rates and elevated temperature conditions is essential to analyse and predict its response against dynamic loading such as impact and thermomechanical fatigue. The constitutive relation of Johnson-Cook is used to capture the strain rate sensitivity and thermal softening effect in AlSi10Mg alloy. Johnson-Cook failure model is widely used for exploring damage mechanics and predicting the fracture in many materials. In this present work, Johnson-Cook material and damage model parameters for additively manufactured AlSi10Mg alloy have been determined numerically from four types of uniaxial tensile test. Three different uniaxial tensile tests with dynamic strain rates (0.1, 1, 10, 50, and 100 s-1) and elevated temperature tensile test with three different temperature conditions (450 K, 500 K and 550 K) were performed on 3D printed AlSi10Mg alloy in ABAQUS/Explicit. Hexahedral elements are used to discretize tensile specimens and fracture energy value of 43.6 kN/m was used for damage initiation. Levenberg Marquardt optimization method was used for the evaluation of Johnson-Cook model parameters. It was observed that additively manufactured AlSi10Mg alloy has shown relatively higher strain rate sensitivity and lower thermal stability as compared to the other Al alloys.

447 Analysis of Cyclic Elastic-Plastic Loading of Shaft Based On Kinematic Hardening Model

Authors: Isa Ahmadi, Ramin Khamedi

Abstract:

In this paper, the elasto-plastic and cyclic torsion of a shaft is studied using a finite element method. The Prager kinematic hardening theory of plasticity with the Ramberg and Osgood stress-strain equation is used to evaluate the cyclic loading behavior of the shaft under the torsional loading. The material of shaft is assumed to follow the non-linear strain hardening property based on the Prager model. The finite element method with C1 continuity is developed and used for solution of the governing equations of the problem. The successive substitution iterative method is used to calculate the distribution of stresses and plastic strains in the shaft due to cyclic loads. The shear stress, effective stress, residual stress and elastic and plastic shear strain distribution are presented in the numerical results.

446 Enhance Halorespiration in Rhodopseudomonas palustris with Cytochrome P450cam System from Pseudomonas putida

Authors: Shou-Chen Lo, Chia-Ching Lin, Chieh-Chen Huang

Abstract:

To decompose organochlorides by bioremediation, co-culture biohydrogen producer and dehalogenation microorganisms is a useful method. In this study, we combined these two characteristics from a biohydrogen producer, Rhodopseudomonas palustris, and a dehalogenation microorganism, Pseudomonas putida, to enchance halorespiration in R. palustris. The genes encoding cytochrome P450cam system (camC, camA, and camB) from P. putida were expressed in R. palustris with designated expression plasmid. All tested strains were cultured to log phase then presented pentachloroethane (PCA) in media. The vector control strain could degrade PCA about 78% after 16 hours, however, the cytochrome P450cam system expressed strain, CGA-camCAB, could completely degrade PCA in 12 hours. While taking chlorinated aromatic, 3-chlorobenzoate, as sole carbon source or present benzoate as co-substrate, CGA-camCAB presented faster growth rate than vector control strain.
