587 Immobilization of Lipase Enzyme by Low Cost Material: A Statistical Approach

Abstract:
Immobilization of lipase enzyme produced from palm oil mill effluent (POME) by the activated carbon (AC) among the low cost support materials was optimized. The results indicated that immobilization of 94% was achieved by AC as the most suitable support material. A sequential optimization strategy based on a statistical experimental design, including one-factor-at-a-time (OFAT) method was used to determine the equilibrium time. Three components influencing lipase immobilization were optimized by the response surface methodology (RSM) based on the face-centered central composite design (FCCCD). On the statistical analysis of the results, the optimum enzyme concentration loading, agitation rate and carbon active dosage were found to be 30 U/ml, 300 rpm and 8 g/L respectively, with a maximum immobilization activity of 3732.9 U/g-AC after 2 hrs of immobilization. Analysis of variance (ANOVA) showed a high regression coefficient (R2) of 0.999, which indicated a satisfactory fit of the model with the experimental data. The parameters were statistically significant at p<0.05.

Abstract:
PCCI engines can reduce NOx and PM emissions simultaneously without sacrificing thermal efficiency, but a low combustion temperature resulting from early fuel injection, and ignition occurring prior to TDC, can cause higher THC and CO emissions and fuel consumption. In conclusion, it was found that the PCCI combustion achieved by the 2-stage injection strategy with optimized calibration factors (e.g. EGR rate, injection pressure, swirl ratio, intake pressure, injection timing) can reduce NOx and PM emissions simultaneously. Abstract:
The main objective of seismic rehabilitation in the foundations is decreasing the range of horizontal and vertical vibrations and omitting high frequencies contents under the seismic loading. In this regard, the advantages of micropiles network is utilized. Reduction in vibration range of foundation can be achieved by using high dynamic rigidness module such as deep foundations. In addition, natural frequency of pile and soil system increases in regard to rising of system rigidness. Accordingly, the main strategy is decreasing of horizontal and vertical seismic vibrations of the structure. In this case, considering the impact of foundation, pile and improved soil foundation is a primary concern. Therefore, in this paper, effective factors are studied on the seismic rehabilitation of foundations applying network micropiles in sandy soils with nonlinear reaction.Keywords: Micropile network, rehabilitation, vibration, seismic load.

Abstract:
Starting from the basic pillars of the supportability analysis this paper queries its characteristics in LCI (Life Cycle Integration) environment. The research methodology contents a review of modern logistics engineering literature with the objective to collect and synthesize the knowledge relating to standards of supportability design in e-logistics environment. The results show that LCI framework has properties which are in fully compatibility with the requirement of simultaneous logistics support and productservice bundle design. The proposed approach is a contribution to the more comprehensive and efficient supportability design process. Also, contributions are reflected through a greater consistency of collected data, automated creation of reports suitable for different analysis, as well as the possibility of their customization according with customer needs. In addition to this, convenience of this approach is its practical use in real time. Abstract:
Continually increasing working temperature and growing need for greater efficiency and reliability of automotive exhaust require systematic investigation into the thermal fatigue properties especially of high temperature stainless steels. In this study, thermal fatigue properties of 300 series austenitic stainless steels have been evaluated in the temperature ranges of 200-800oC and 200-900oC. Systematic methods for control of temperatures within the predetermined range and measurement of load applied to specimens as a function of temperature during thermal cycles have been established. Thermal fatigue tests were conducted under fully constrained condition, where both ends of specimens were completely fixed. Load relaxation behavior at the temperatures of thermal cycle was closely related with the thermal fatigue property.

Abstract:
The winding hot-spot temperature is one of the most critical parameters that affect the useful life of the power transformers. The winding hot-spot temperature can be calculated as function of the top-oil temperature that can estimated by using the ambient temperature and transformer loading measured data. This paper proposes the estimation of the top-oil temperature by using a method based on Least Squares Support Vector Machines approach. The estimated top-oil temperature is compared with measured data of a power transformer in operation. The results are also compared with methods based on the IEEE Standard C57.91-1995/2000 and Artificial Neural Networks. It is shown that the Least Squares Support Vector Machines approach presents better performance than the methods based in the IEEE Standard C57.91-1995/2000 and artificial neural networks.Keywords: Artificial Neural Networks, Hot-spot Temperature, Least Squares Support Vector, Top-oil Temperature.

Abstract:
This paper presents the effect of corrugation profile geometry on the crushing behavior, energy absorption, failure mechanism, and failure mode of woven roving glass fibre/epoxy laminated composite tube. Experimental investigations were carried out on composite tubes with three different profile shapes: sinusoidal, triangular and trapezoidal. The tubes were subjected to lateral compressive loading. On the addition to a radial corrugated composite tube, cylindrical composite tube, were fabricated and tested under the same condition in order to know the effect of corrugation geometry. Typical histories of their deformation are presented. Behavior of tubes as regards the peak crushing load, energy absorbed and mode of crushing has been discussed. The results show that the behavior of the tube under lateral compression load is influenced by the geometry of the tube itself.Keywords: Corrugated composite specimens, Energy absorption, Lateral crushing.

In this paper, an inventory model with finite and constant replenishment rate, price dependant demand rate, time value of money and inflation, finite time horizon, lead time and exponential deterioration rate and with the objective of maximizing the present worth of the total system profit is developed. Using a dynamic programming based solution algorithm, the optimal sequence of the cycles can be found and also different optimal selling prices, optimal order quantities and optimal maximum inventories can be obtained for the cycles with unequal lengths, which have never been done before for this model. Abstract:
Atrazine, a herbicide widely used in sugarcane and corn production, is a frequently detected groundwater contaminant. An atrazine-degrading bacterium, strain KB02, was obtained from long-term atrazine-treated sugarcane field soils in Kanchanaburi province of Thailand. Strain KB02 had a rod-to-coccus morphological cycle during growth. Sequence analysis of the PCR product indicated that the 16S rRNA gene in strain KB02 was ranging from 97-98% identical to the same region in Klebsiella sp. Based on biochemical, physiological analysis and 16S rDNA sequence analysis of one representative isolate, strain KB02, the isolates belong to the genus Klebsiella in the family Enterobacteriaceae. Interestingly that the various primers for atzA, B and C failed to amplify genomic DNA of strain KB02. Whereas the expected PCR product of atzA, B and C were obtained from the reference strain, Arthrobacter sp. strain KU001.

Abstract:
This paper gives an intuition regarding glowworm swarm optimization (GSO) technique to solve dynamic economic dispatch (DED) problems of thermal generating units. The objective of the problem is to schedule optimal power generation of dedicated thermal units over a specific time band. In this study, Glowworm swarm optimization technique enables a swarm of agents to split into subgroup, exhibit simultaneous taxis towards each other and rendezvous at multiple optima (not necessarily equal) of a given multimodal function. The feasibility of the GSO method has been tested on ten-unit-test systems where the power balance constraints, operating limits, valve point effects, and ramp rate limits are taken into account. The results obtained by the proposed technique are compared with other heuristic techniques. The results show that GSO technique is capable of producing better results.

Abstract:
In this paper, a study on the modes of collapse of compress- expand members are presented. Compress- expand member is a compact, multiple-combined cylinders, to be proposed as energy absorbers. Previous studies on the compress- expand member have clarified its energy absorption efficiency, proposed an approximate equation to describe its deformation characteristics and also highlighted the improvement that it has brought. However, for the member to be practical, the actual range of geometrical dimension that it can maintain its applicability must be investigated. In this study, using a virtualized materials that comply the bilinear hardening law, Finite element Method (FEM) analysis on the collapse modes of compress- expand member have been conducted. Water Sensitive Urban Design (WSUD) features are increasingly used to treat and manage polluted stormwater runoff in urbanised areas. It is important to monitor and evaluate the effectiveness of the infrastructure in achieving their intended performance targets after constructing and operating these features overtime. The paper presents the various methods of analysis used to assess the effectiveness of the in-situ WSUD features, such as: onsite visual inspections during operational and non operational periods, maintenance audits and periodic water quality testing. Abstract:
Pervious concrete combines considerable permeability with adequate strength, which makes it very beneficial in pavement construction and also in ground improvement projects. In this paper, a single pervious concrete pile subjected to vertical and lateral loading is analysed using a verified three dimensional finite element code. A parametric study was carried out in order to investigate load bearing capacity of a single unreinforced pervious concrete pile in saturated soft soil and also gain insight into the failure mechanism of this rather new soil improvement technique. The results show that concrete damaged plasticity constitutive model can perfectly simulate the highly brittle nature of the pervious concrete material and considering the computed vertical and horizontal load bearing capacities, some suggestions have been made for ground improvement projects.

Abstract:
Jet grouting strengthened pile (JPP) is one of composite piles used in soft ground improvement. It may improve the vertical and lateral bearing capacity effectively and it has been practically used in a considerable scale. In order to make a further research on load transfer mechanism of single JPP with and without cap under lateral loads, JPP is analyzed by means of FEM analysis. It is resulted that the JPP pile could improve lateral bearing capacity by compared with bored concrete pile which is higher for shorter pile and the biggest bending moment of JPP pile is located in the depth of around 48% of embedded length of the pile. Meanwhile, increase of JPP pile length causes to increase of peak mobilized bending moment. Also, by cap addition, JPP piles will have a much higher lateral bearing capacity and increasing in cohesion of soil layer resulted to increase of lateral bearing capacity of JPP pile. Abstract:
Without uncertainty by applying external loads on beams, bending is created. The created bending in I-beams, puts one of the flanges in tension and the other one in compression. With increasing of bending, compression flange buckled and beam in out of its plane direction twisted, this twisting well-known as Lateral Torsional Buckling. Providing bending moment varieties along the beam, the critical moment is greater than the case its under pure bending. In other words, the value of bending gradient coefficient is always greater than unite. In this article by the use of " ANSYS 10.0" software near 80 3-D finite element models developed for the propose of analyzing beams` lateral torsional buckling and surveying influence of slenderness on beams' bending gradient coefficient. Results show that, presented Cb coefficient via AISC is not correct for some of beams and value of this coefficient is smaller than what proposed by AISC. Abstract:
In this work, bending fatigue life of notched specimens with various notch geometries and dimensions is investigated by experiment and Manson-Caffin theoretical method. In this theoretical method, fatigue life of notched specimens is calculated using the fatigue life obtained from the experiments for plain specimens (without notch). Three notch geometries including ∪-shape, ∨-shape and C -shape notches are considered in this investigation. The experiments are conducted on a rotary bending Moore machine. The specimens are made of a low carbon steel alloy, which has wide application in industry. The stress- life curves are captured for all notched specimen by experiment. The results indicate that Manson-Caffin analytical method cannot adequately predict the fatigue life of notched specimen. However, it seems that the difference between the experiments and Manson-Caffin predictions can be compensated by a proportional factor.Keywords: fatigue life, Mason-Caffin method, notchedspecimen, stress-life curve.

Abstract:
The goal of this paper is to develop a model to integrate “pricing" and “advertisement" for short life cycle products, such as branded fashion clothing products. To achieve this goal, we apply the concept of “Dynamic Pricing". There are two classes of advertisements, for the brand (regardless of product) and for a particular product. Advertising the brand affects the demand and price of all the products. Thus, the model considers all these products in relation with each other. We develop two different methods to integrate both types of advertisement and pricing. The first model is developed within the framework of dynamic programming. However, due to the complexity of the model, this method cannot be applicable for large size problems. Therefore, we develop another method, called hieratical approach, which is capable of handling the real world problems. Diesel vehicle should be equipped with emission after-treatment devices as NOx reduction catalyst and particulate filtersin order to meet more stringer diesel emission standard. Urea-SCR is being developed as the most efficient method of reducing NOx emissions in the after-treatment devices of diesel engines, and recent studies have begun to mount the Urea-SCR device for diesel passenger cars and light duty vehicles. In the present study, the effects of the mixer on the efficiency of urea-SCR System (i.e., NH3uni- formityindex (NH3 UI) is investigated by predicting the transport phenomena in the urea-SCR system. The three dimensional Eulerian-Lagrangian CFD simulationfor internal flow and spray characteristics in front of SCR is carried out by using STAR-CCM+ 7.06 code. In addition, the paper proposes a method to minimize the wall-wetting around the urea injector in order to prevent injector blocks caused by solid urea loading.

This paper investigates the effects of lubrication on the quantity of heat emission of two spur gear. System with and without lubrication effected on the quantity of heat induced on the gear box (oil - bearings – gears). Both of lubrication and speed of motor are affected on the performance of gears. Research investigated the lubrication on the system with and without loading as well as the wear of gears and bearing's conditions. Gear box investigated includes the motor, pump, two spur gears, two shafts; speed change used pulleys and belts. Load used equal one weight ones of gear. Lubrication mechanism used jet system (upper and lower jet). Gear box we used system of jet lubrication is perpendicular direction of the contact line between two teeth. Results appeared in this work that the lubrication is the vital parameter which is affected on the performance and durability of gears and bearings. In macroscopic observation, we noted that damage of bearings happened during the absence of lubrication as well as abrasive of wear of teeth. Precast residential houses are normally constructed in Malaysia using precast shear-key wall panel and this panel is designed using BS8110 where there is no provision for earthquake. However, the safety of this house under moderate and strong earthquake is still questionable. Consequently, the full-scale of residential house are designed, constructed, tested and analyzed under in-plane lateral quasi-static cyclic loading. Hysteresis loops are plotted based on the experimental work and compared with modeling of hysteresis loops using HYSTERES in RUAUMOKO 2D program. Modified Takeda hysteresis model is chosen to behave a similar pattern with experimental work. This program will display the earthquake excitations, spectral displacements, pseudo spectral acceleration, mode shape and deformation of the structure. It can be concluded that this building is suffering severe cracks and damage under moderate and severe earthquake.

Abstract:
This article presents an interactive system for data acquisition in vehicle testing according to the test process defined in automotive directive 2004/104/EC. The project has been designed and developed by authors for the Spanish company Applus-LGAI. The developed project will result in a new process, which will involve the creation of braking cycle test defined in the aforementioned automotive directive. It will also allow the analysis of new vehicle features that was not feasible, allowing an increasing interaction with the vehicle. Potential users of this system in the short term will be vehicle manufacturers and in a medium term the system can be extended to testing other automotive components and EMC tests.

The influence of three different types of halloysite nanotubes (HNTs) with different dimensions, namely as camel lake (CLA), Jarrahdale (JA) and Matauri Bay (MB), on their reinforcing ability of ethylene propylene dine monomer (EPDM) were investigated by varying the HNTs loading (from 0-15 phr). Mechanical properties of the nanocomposites improved with addition of all three HNTs, but CLA based nanocomposites exhibited a significant enhancement compared to the other HNTs. For instance, tensile properties of EPDM nanocomposites increased by 120%, 256% and 340% for MB, JA and CLA, respectively, with addition of 15 phr of HNTs. This could be due to the higher aspect ratio and higher surface area of CLA compared to others. Scanning electron microscopy (SEM) of nanocomposites at 15 phr of HNT loadings showed low amounts of pulled-out nanotubes which confirmed the presence of more embedded nanotubes inside the EPDM matrix, as well as aggregates within the fracture surface of EPDM/HNT nanocomposites

Abstract:
In this study, a new constitutive model is developed to describe the hyperelastic behavior of collagenous tissues with a parallel arrangement of collagen fibers such as ligaments and tendons. The model is formulated using a continuum approach incorporating the structural changes of the main tissue components: collagen fibers, proteoglycan-rich matrix and fiber-matrix interaction. The mechanical contribution of the interaction between the fibers and the matrix is simply expressed by a coupling term. The structural change of the collagen fibers is incorporated in the constitutive model to describe the activation of the fibers under tissue straining. Finally, the constitutive model can easily describe the stress-stretch nonlinearity which occurs when a ligament/tendon is axially stretched. This study shows that the interaction between the fibers and the matrix contributes to the mechanical tissue response. Abstract:
This paper seeks to give a general idea of the universe of project portfolio management, from its multidisciplinary nature, to the many challenges it raises, passing through the different techniques, models and tools used to solve the multiple problems known. It is intended to contribute to the clarification, with great depth, of the impacts and relationships involved in managing the projects- portfolio. It aims at proposing a technique for the project alignment with the organisational strategy, in order to select projects that later on will be considered in the analysis and selection of the portfolio. We consider the development of a methodology for assessing the project alignment index very relevant in the global market scenario. It can help organisations to gain a greater awareness of market dynamics, speed up the decision process and increase its consistency, thus enabling the strategic alignment and the improvement of the organisational performance.

Abstract:
In this paper the vibration behaviors of a structure equipped with a tuned liquid column damper (TLCD) under a harmonic type of earthquake loading are studied. However, due to inherent nonlinear liquid damping, it is no doubt that a great deal of computational effort is required to search the optimum parameters of the TLCD, numerically. Therefore by linearization the equation of motion of the single degree of freedom structure equipped with the TLCD, the closed form solutions of the TLCD-structure system are derived. To find the reliability of the analytical method, the results have been compared with other researcher and have good agreement. Further, the effects of optimal design parameters such as length ratio and mass ratio on the performance of the TLCD for controlling the responses of a structure are investigated by using the harmonic type of earthquake excitation. Finally, the Citicorp Center which has a very flexible structure is used as an example to illustrate the design procedure for the TLCD under the earthquake excitation.

Abstract:
It is known that the heart interacts with and adapts to its venous and arterial loading conditions. Various experimental studies and modeling approaches have been developed to investigate the underlying mechanisms. This paper presents a model of the left ventricle derived based on nonlinear stress-length myocardial characteristics integrated over truncated ellipsoidal geometry, and second-order dynamic mechanism for the excitation-contraction coupling system. The results of the model presented here describe the effects of the viscoelastic damping element of the electromechanical coupling system on the hemodynamic response. Different heart rates are considered to study the pacing effects on the performance of the left-ventricle against constant preload and afterload conditions under various damping conditions. The results indicate that the pacing process of the left ventricle has to take into account, among other things, the viscoelastic damping conditions of the myofilament excitation-contraction process.Keywords: Myocardial sarcomere, cardiac pump, excitationcontraction coupling, viscoelasicity

Abstract:
The development of shape and size of a crack in a pressure vessel under uniaxial and biaxial loadings is important in fitness-for-service evaluations such as leak-before-break. In this work finite element modelling was used to evaluate the mean stress and the J-integral around a front of a surface-breaking crack. A procedure on the basis of ductile tearing resistance curves of high and low constrained fracture mechanics geometries was developed to estimate the amount of ductile crack extension for surface-breaking cracks and to show the evolution of the initial crack shape. The results showed non-uniform constraint levels and crack driving forces around the crack front at large deformation levels. It was also shown that initially semi-elliptical surface cracks under biaxial load developed higher constraint levels around the crack front than in uniaxial tension. However similar crack shapes were observed with more extensions associated with cracks under biaxial loading.Keywords: biaxial load, crack shape, fracture toughness, surface crack, uniaxial load.

Abstract:
Lightweight construction became more and more important over the last decades in several applications, e.g. in the automotive or aircraft sector. This is the result of economic and ecological constraints on the one hand and increasing safety and comfort requirements on the other hand. In the field of lightweight design, different approaches are used due to specific requirements towards the technical systems. The use of endless carbon fiber reinforced plastics (CFRP) offers the largest weight saving potential of sometimes more than 50% compared to conventional metal-constructions. However, there are very limited industrial applications because of the cost-intensive manufacturing of the fibers and production technologies. Other disadvantages of pure CFRP-structures affect the quality control or the damage resistance. One approach to meet these challenges is hybrid materials. This means CFRP and sheet metal are combined on a material level. Therefore, new opportunities for innovative process routes are realizable. Hybrid lightweight design results in lower costs due to an optimized material utilization and the possibility to integrate the structures in already existing production processes of automobile manufacturers. In recent and current research, the advantages of two-layered hybrid materials have been pointed out, i.e. the possibility to realize structures with tailored mechanical properties or to divide the curing cycle of the epoxy resin into two steps. Current research work at the Chair for Automotive Lightweight Design (LiA) at the Paderborn University focusses on production processes for fiber-metal-laminates. The aim of this work is the development and qualification of a large-scale production process for high-performance fiber-metal-laminates (FML) for industrial applications in the automotive or aircraft sector. Therefore, the prepreg-press-technology is used, in which pre-impregnated carbon fibers and sheet metals are formed and cured in a closed, heated mold. The investigations focus e.g. on the realization of short process chains and cycle times, on the reduction of time-consuming manual process steps, and the reduction of material costs. This paper gives an overview over the considerable steps of the production process in the beginning. Afterwards experimental results are discussed. This part concentrates on the influence of different process parameters on the mechanical properties, the laminate quality and the identification of process limits. Concluding the advantages of this technology compared to conventional FML-production-processes and other lightweight design approaches are carried out.

Abstract:
Dynamic analysis of composite doubly curved panels with variable thickness subjected to different pulse types using Generalized Differential Quadrature method (GDQ) is presented in this study. Panels with variable thickness are used in the construction of aerospace and marine industry. Giving variable thickness to panels can allow the designer to get optimum structural efficiency. For this reason, estimating the response of variable thickness panels is very important to design more reliable structures under dynamic loads. Dynamic equations for composite panels with variable thickness are obtained using virtual work principle. Partial derivatives in the equation of motion are expressed with GDQ and Newmark average acceleration scheme is used for temporal discretization. Several examples are used to highlight the effectiveness of the proposed method. Results are compared with finite element method. Effects of taper ratios, boundary conditions and loading type on the response of composite panel are investigated.

Automated material handling is given prime importance in the semi automated and automated facilities since it provides solution to the gigantic problems related to inventory and also support the latest philosophies like just in time production JIT and lean production. Automated storage and retrieval system is an antidote (if designed properly) to the facility sufferings like getting the right material , materials getting perished, long cycle times or many other similar kind of problems. A working model of automated storage and retrieval system (AS/RS) is designed and developed under the design parameters specified by Material Handling Industry of America (MHIA). Later on analysis was carried out to calculate the throughput and size of the machine.
