169 New Kinetic Approach to the Enzymatic Hydrolysis of Proteins – A Case of Thermolysin-Catalyzed Albumin

Abstract:
Using an enzyme of known specificity the hydrolysis of protein was carried out in a controlled manner. The aim was to obtain oligopeptides being the so-called active peptides or their direct precursors. An original way of expression of the protein hydrolysis kinetics was introduced. Peptide bonds contained in the protein were recognized as a diverse-quality substrate for hydrolysis by the applied protease. This assumption was positively verified taking as an example the hydrolysis of albumin by thermolysin. Peptide linkages for this system should be divided into at least four groups. One of them is a group of bonds non-hydrolyzable by this enzyme. These that are broken are hydrolyzed at a rate that differs even by tens of thousands of times. Designated kinetic constants were k'F = 10991.4 L/g.h, k'M = 14.83L/g.h, k'S about 10-1 L/g.h for fast, medium and slow bonds, respectively. Moreover, a procedure for unfolding of the protein, conducive to the improved susceptibility to enzymatic hydrolysis (approximately three-fold increase in the rate) was proposed.

Abstract:
In this work, the influence of temperature on the different parameters of solar cells based on organic semiconductors are studied. The short circuit current Isc increases so monotonous with temperature and then saturates to a maximum value before decreasing at high temperatures. The open circuit voltage Vco decreases linearly with temperature. The fill factor FF and efficiency, which are directly related with Isc and Vco follow the variations of the letters. The phenomena are explained by the behaviour of the mobility which is a temperature activated process.Keywords: cells parameters, organic materials, solar cells, temperature effect

Abstract:
Alpfa-fetoprotein and its fragments may be an important vehicle for targeted delivery of radionuclides to the tumor. We investigated the effect of conditions on the labeling of biologically active synthetic peptide based on the (F-afp) with technetium-99m. The influence of the nature of the buffer solution, pH, concentration of reductant, concentration of the peptide and the reaction temperature on the yield of labeling was examined. As a result, the following optimal conditions for labeling of (F-afp) are found: pH 8.5 (phosphate and bicarbonate buffers) and pH from 1.7 to 7.0 (citrate buffer). The reaction proceeds with sufficient yield at room temperature for 30 min at the concentration of SnCl2 and (Fafp) (F-afp) is to be less than 10 mkg/ml and 25 mkg/ml, respectively. Investigations of the test drug accumulation in the tumor cells of human breast cancer were carried out. Results can be assumed that the in vivo study of the (F-afp) in experimental tumor lesions will show concentrations sufficient for imaging these lesions by SPECT.

Abstract:
In neural networks, when new patterns are learned by a network, the new information radically interferes with previously stored patterns. This drawback is called catastrophic forgetting or catastrophic interference. In this paper, we propose a biologically inspired neural network model which overcomes this problem. The proposed model consists of two distinct networks: one is a Hopfield type of chaotic associative memory and the other is a multilayer neural network. We consider that these networks correspond to the hippocampus and the neocortex of the brain, respectively. Information given is firstly stored in the hippocampal network with fast learning algorithm. Then the stored information is recalled by chaotic behavior of each neuron in the hippocampal network. Finally, it is consolidated in the neocortical network by using pseudopatterns. Computer simulation results show that the proposed model has much better ability to avoid catastrophic forgetting in comparison with conventional models.

Abstract:
In this paper we investigate the influence of external noise on the inference of network structures. The purpose of our simulations is to gain insights in the experimental design of microarray experiments to infer, e.g., transcription regulatory networks from microarray experiments. Here external noise means, that the dynamics of the system under investigation, e.g., temporal changes of mRNA concentration, is affected by measurement errors. Additionally to external noise another problem occurs in the context of microarray experiments. Practically, it is not possible to monitor the mRNA concentration over an arbitrary long time period as demanded by the statistical methods used to learn the underlying network structure. Abstract:
We report here structural, mechanical and I-V characteristics of Zn1-xMxO ceramic samples with various x and M. It is found that the considered dopants does not influence the wellknown peaks related to wurtzite structure of ZnO ceramics, while the shape and size of grains are clearly affected. Average crystalline diameters, deduced from XRD are between 42 nm and 54 nm, which are 70 times lower than those obtained from SEM micrographs. Interestingly, the potential barrier could be formed by adding Cu up to 0.20, and it is completely deformed by 0.025 Ni additions. The breakdown field could be enhanced up to 4138 V/cm by 0.025 Cu additions, followed by a decrease with further increase of Cu . On the other hand a gradual decrease in VHN is reported for both dopants and their values are higher in Ni samples as compared to Cu samples. The electrical conductivity is generally improved by Ni, while addition of Cu improved it only in the over doped region (≥ 0.10). These results are discussed in terms of the difference of valency and ferromagnetic ordering for both dopants as compared to undoped sample.Keywords: Semiconductors, Chemical Synthesis, Impurities and Electronic Transport

Abstract:
Microarrays are made it possible to simultaneously monitor the expression profiles of thousands of genes under various experimental conditions. It is used to identify the co-expressed genes in specific cells or tissues that are actively used to make proteins. This method is used to analysis the gene expression, an important task in bioinformatics research. Cluster analysis of gene expression data has proved to be a useful tool for identifying co-expressed genes, biologically relevant groupings of genes and samples. In this work K-Means algorithms has been applied for clustering of Gene Expression Data. Further, rough set based Quick reduct algorithm has been applied for each cluster in order to select the most similar genes having high correlation. Then the ACV measure is used to evaluate the refined clusters and classification is used to evaluate the proposed method. They could identify compact clusters with feature selection method used to genes are selected.

Abstract:
Wireless sensor networks are consisted of hundreds or thousands of small sensors that have limited resources. Energy-efficient techniques are the main issue of wireless sensor networks. This paper proposes an energy efficient agent-based framework in wireless sensor networks. We adopt biologically inspired approaches for wireless sensor networks. Agent operates automatically with their behavior policies as a gene. Agent aggregates other agents to reduce communication and gives high priority to nodes that have enough energy to communicate. Agent behavior policies are optimized by genetic operation at the base station. Simulation results show that our proposed framework increases the lifetime of each node. Each agent selects a next-hop node with neighbor information and behavior policies. Biologically active peptides are of particular interest in food science and human nutrition because they have been shown to play several physiological roles. In vitro gastrointestinal digestion of lentil and whey proteins in this study produced high angiotensin-I converting enzyme inhibitory activity with 75.5±1.9 and 91.4±2.3% inhibition, respectively. High ACE inhibitory activity was observed in lentil after 5 days of germination (84.3±1.2%). Fractionation by reverse phase chromatography gave inhibitory activities as high as 86.3±2.0 for lentil, 94.8±1.8% for whey and 93.7±1.7% at 5th day of germination. Further purification by HPLC resulted in several inhibitory peptides with IC50 values ranging from 0.064 to 0.164 mg/ml. These results demonstrate that lentil proteins are a good source of peptides with ACE inhibitory activity that can be released by germination or gastrointestinal digestion. Despite the lower bioactivity in comparison with whey proteins, incorporation of lentil proteins in functional food formulations and natural drugs look promising.Keywords: ACE inhibitory peptides, digestion, germination, lentil proteins, whey proteins

Abstract:
Inferring the network structure from time series data is a hard problem, especially if the time series is short and noisy. DNA microarray is a technology allowing to monitor the mRNA concentration of thousands of genes simultaneously that produces data of these characteristics. In this study we try to investigate the influence of the experimental design on the quality of the result. More precisely, we investigate the influence of two different types of random single gene perturbations on the inference of genetic networks from time series data. To obtain an objective quality measure for this influence we simulate gene expression values with a biologically plausible model of a known network structure. Abstract:
The microstructure and texture evolution of cryo rolled and annealed ductile TaHfNbZrTi refractory high entropy alloy was investigated. To obtain that, the alloy is severely cryo rolled and subsequently annealed for the recrystallization process. The cryo rolled – 90% shows the presence of very fine grains and microstructural heterogeneity. The cryo rolled samples are annealed at a temperature ranging from 800°C to 1400°C, the partial recrystallization is observed at 800°C annealed condition, and at higher annealing temperatures the complete recrystallization process is noticed. The development of ND fiber texture is observed after the annealing.

An experiment was conducted at Agronomic Research Farm, University of Agriculture Faisalabad. The experiment was laid out in randomized complete block designee (RCBD) with three replications. Treatments studied were clodinafop (Topic-15 WG) at 37 g a.i. ha-1, clodinafop (Topaz-15 WG) at 45 g a.i. ha-1, fenoxaprop-p-ethyl (Puma Super-75 EW) at 45 g a.i. ha-1, fenoxaprop-p-ethyl (Gramicide-6.9 EW) at 85 g a.i. ha-1, fenoxaprop-p-ethyl (Chinlima-6.9 EW) at 85 g a.i. ha-1 and weedy check. Abstract:
EU and UK Government targets for minimising and recycling household waste has led the responsible authorities to research the alternatives to landfill. In the work reported here the local waste collection authority (Charnwood Borough Council) has adopted the aspirational strategy of becoming a “Zero Waste Borough” to lead the drive for public participation. The work concludes that the separate collection of food waste would be needed to meet the two regulatory standards on recycling and biologically active wastes.
An analysis of a neighbouring Authority (Newcastle-Under-Lyne Borough Council (NBC), a similar sized local authority that has a successful weekly food waste collection service was undertaken. Results indicate that the main challenges for Charnwood Borough Council would be gaining householder co-operation, the extra costs of collection and organising alternative treatment. The analysis also demonstrated that there was potential offset value via anaerobic digestion for CBC to overcome these difficulties and improve its recycling performance.

Abstract:
The grain quality is a decisive factor in its use. In Poland, spring wheat is characterized by more favorable quality parameters in relation to the winter form of this species. In the present study, the effects of three different sowing dates (autumn, delayed autumn, and spring) and cultivar (Tybalt, Cytra, Bombona, Monsun, and Parabola) on the selected technological value parameters of spring wheat over three years were studied. The field trials were carried out in two locations (Bezek, Czesławice) in the Lubelskie Vivodeship, Poland. It was found that the falling number of spring wheat grains from autumn sowing dates was at a similar level to wheat sown in spring. The amount of wet gluten in the grain was variable in years, and its quality was better in wheat sown in spring. Sedimentation index was dependent upon on the cultivar.

Abstract:
Attempts were made to identify anuran glial cells. They were found as nervous tissue resident. Having stage dependent morphotype changes, whereby, appeared as an ovoid to oval in resting state and amoeboid mrophotypes in activated state, stained fairly with methylene blue and take up Pelikane blue 10% aqueous solution, as well as having the ability to phagocytize heat killed Staphylococcus aureus. They were delineated from the migrating peripheral monocytes by morphotypic and morphometeric differences. Such criteria were consistence with glial cells. Thus, the anuran glial cells are being identified in the frog Rana ridibunda Pallas 1771 and this animal can be of use as a simple model for the immunobiology of glial cells.

In order to increase in chickpea quality and agroecosystem sustainability, field experiments were carried out in 2007 and 2008 growing seasons. In this research the effects of different organic, chemical and biological fertilizers were investigated on grain yield and quality of chickpea. Experimental units were arranged in split-split plots based on randomized complete blocks with three replications. The highest amounts of yield and yield components were obtained in G1×N5 interaction. Significant increasing of N, P, K, Fe and Mg content in leaves and grains emphasized on superiority of mentioned treatment because each one of these nutrients has an approved role in chlorophyll synthesis and photosynthesis ability of the crop. The combined application of compost, farmyard manure and chemical phosphorus (N5) had the best grain quality due to high protein, starch and total sugar contents, low crude fiber and reduced cooking time.Keywords: Agroecosystem, sustainability, chickpea, naturalresources.

The aim of this investigation is to study the performance of the new generation of the PVD coated grade and to map the influence of cutting conditions on the tool life in milling of ADI (Austempered Ductile Iron). The results show that chipping is the main wear mechanism which determines the tool life in dry condition and notch wear in wet condition for this application. This due to the different stress mechanisms and preexisting cracks in the coating. The wear development shows clearly that the new PVD coating (C20) has the best ability to delay the chipping growth. It was also found that a high content of Al in the new coating (C20) was especially favorable compared to a TiAlN multilayer with lower Al content (C30) or CVD coating. This is due to fine grains and low compressive stress level in the coating which increase the coating ability to withstand the mechanical and thermal impact. It was also found that the use of coolant decreases the tool life with 70-80% compare to dry milling.Keywords: Austempered Ductile Iron (ADI), coating, chipping, milling, tool performance.

Celiac disease is an immune-mediated disease, triggered in genetically susceptible individuals by ingested gluten from wheat, rye, barley and other closely related cereal grains. The only effective treatment is a strict gluten free diet for life. Latvian producers do not offer gluten-free products. In this research, use of extruded maize flour was tested for substituting rice, maize or buckwheat flour in gluten-free bread formulations at different ratios. Also the influence of extruded maize flour on the quality parameters of gluten-free bread was investigated. The aim of research was to study the influence of extruded maize flour on gluten-free bread quality. Addition of extruded maize flour affect gluten-free bread crumb color, structure of crumb, weight loss and dry off of bread.

The present study has been conducted to characterize the prophenoloxidase (PPO) system of the desert locust, Schistocerca gregaria following injection of Bacillus thuringiensis kurstaki (Bt). The bulk of PPO system was associated with haemocytes and a little amount was found in plasma. This system was activated by different activators such as laminarin, lipopolysaccharide (LPS) and trypsin suggesting that the stimulatory mechanism may involve an enzyme cascade of one or more associated molecules. These activators did not activate all the molecules of the cascade. A strain of Monascus purpureus CMU001 was used to prepare red yeast rice from Thai glutinous rice Korkor 6 (RD 6). Adding of different amounts of histidine (156, 312, 625 and 1250 mg in 100 g of rice grains)) under aerobic and air limitation (air-lock) condition were used in solid fermentation. Determination of the yield as well as monacolin K content was done. Citrinin content was also determined in order to confirm the safety use of prepared red yeast rice. It was found that under air-lock condition with 1250 mg of histidine addition gave the highest yield of 37.40 g of dried red yeast rice prepared from 100 g of rice. Highest 5.72 mg content of monacolin K was obtained under air-lock condition with 312 mg histidine addition. In the other hand, citrinin content was found to be less than 24462 ng/g of all dried red yeast rice samples under the experimental methods used in this work.

The localized corrosion behavior of laser surface melted 304L austenitic stainless steel was studied by potentiodynamic polarization test. The extent of improvement in corrosion resistance was governed by the preferred orientation and the percentage of delta ferrite present on the surface of the laser melted sample. It was established by orientation imaging microscopy that the highest pitting potential value was obtained when grains were oriented in the most close- packed [101] direction compared to the random distribution of the base metal and other laser surface melted samples oriented in [001] direction. Waste of certain process can be the input source of other sectors in order to reduce environmental pollution. Today there are more and more solid wastes are generated, but only very small amount of those are recycled. So, the threatening of environmental pressure to public health is very serious. The methods considered for the treatment of solid waste are biogas tanks or processing to make animal feed and fertilizer, however, they did not perform well. An alternative approach is growing mushrooms on waste residues. This is regarded as an environmental friendly solution with potential economical benefit. The substrate producers do their best to produce quality substrate at low cost. Apart from other methods, this can be achieved by employing biologically degradable wastes used as the resource material component of the substrate. Mushroom growing is a significant tool for the restoration, replenishment and remediation of Earth’s overburdened ecosphere. One of the rational methods of waste utilization involves locally available wastes. The present study aims to find out the yield of mushroom grown on locally available waste for free and to conserve our environment by recycling wastes.

The available register of farmers association was the sampling frame work where ten percent (61) farmers per state were randomly sampled. Data were collected and analysed using z-test. The research findings revealed tree crops and grains production enterprises ranked higher in Osun (rain fed zones) and Taraba states (savannah zones) respectively. Osun state entrepreneur felt the effect of the conflict on their enterprises more than Tarba state. The reasons adduced for severity of the conflict on enterprises are majority (77.0%) migrated and (75.5%) of them were not allowed to enter their farms during and when conflict deescalated unlike situation in Taraba state. The different in enterprises production level between the two agroecological zone was statistically significant at p<0.05. Abstract:
Wheat germ has a balanced amino acid composition of the protein, which is well digested by enzymes in the gastrointestinal tract of humans, a high content of vitamins, minerals and unsaturated acids. Introduction components grain food products will enrich their biologically important substances, giving these products a number of valuable properties and reducing their caloric. A complex natural system of substances in foods will help replenish the body's need of essential nutrients, increasing its resistance to the harmful effects of the environment, prolong life. In this regard, there was a need for the development of production technology of protein complexes from wheat germ and then applying them in food, particularly in the dairy industry. Experimental studies were conducted to determine the number of herbal supplements on the sensory characteristics of the product. Studies have been conducted to determine the optimal process parameters of water activity and moisture content of the investigational product.

Abstract:
Paddy being cultivated since about 10,000 years B.C in Ganga Valley in India, its production reached up to 99 million tons in the year 2012. BGA are of much ecological importance for maintaining the soil fertility and reclaiming the alkalinity. In present investigation attempts were made to identify the local cyanobacterial genera from the paddy fields, BGA application for green farming enabling the paddy to utilize more amount of nitrogen released and to examine its impact along with Urea upon growth and yield responses of the Paddy crop. It was observed that combined treatment of BGA with Urea proved better response in almost all growth parameters and yield attributes except number of tillers/ Plant and grains/ panicle as compared to application of either Urea or BGA alone. The Paddy growers should be encouraged to adopt BGA along with Urea as source of Nitrogen for Paddy cultivation.

Abstract:
Brachistinae Föerster, 1862 is a subfamily of the family Braconidae (order Hymenoptera) with about 410 species distributed all around the world. Brachistinae includes the genera, Eubazus Nees von Esenbeck 1814, Foersteria Szépligeti 1896, Chelostes van Achterberg 1990, Triaspis Haliday 1835 and Schizoprymnus Förster 1862. Members of the subfamily live as parasitoids on the families Curculionidae and Apionidae (Coleoptera), which also include very important agricultural pests. In generally, members of the genus Triaspis are poorly known biologically. The genus is represented by 37 species in the West Palearctic region and 118 species worldwide. Adult specimens of Triaspis were collected from as wide a range of habitats as possible at different altitudes in different parts of Turkey between 1982 and 2010. Samples collected from short plants using standard insect sweeping nets were transferred into tubes containing 70% ethanol and labelled following their preparations according to museum techniques. Seven Triaspis species have been reported from Turkey in this study. Five of these species are new to the fauna of Turkey.

Abstract:
This paper presents a mixed design trial of structural concretes with laterites from Benin. These materials are often the only granular resources readily available in many tropical regions. In the first step concretes were designed with raw laterites, but the performances obtained were rather disappointing in spite of high cement dosages. A detailed physical characterization of these materials then showed that they contained a significant proportion of fine clays, and that the coarsest fraction (gravel) contained a variety of facies, some of which were not very dense or indurated. Washing these laterites, and even the elimination of the most friable grains of the gravel fraction, made it possible to obtain concretes with satisfactory properties in terms of workability, density and mechanical strength. However, they were found to be slightly less stiff than concretes made with more traditional aggregates. It is therefore possible to obtain structural concretes with only laterites and cement but at the cost of eliminating some of their granular constituents.

Abstract:
In order to evaluation the effects of natural, biological and chemical fertilizers on grain yield and chickpea quality, field experiments were carried out in 2007 and 2008 growing seasons. In this research the effects of different organic, chemical and biological fertilizers were investigated on grain yield and quality of chickpea. Experimental units were arranged in split-split plots based on randomized complete blocks with three replications. The highest amounts of yield and yield components were obtained in G1×N5 interaction. Significant increasing of N, P, K, Fe and Mg content in leaves and grains emphasized on superiority of mentioned treatment because each one of these nutrients has an approved role in chlorophyll synthesis and photosynthesis ability of the crop. The combined application of compost, farmyard manure and chemical phosphorus (N5) had the best grain quality due to high protein, starch and total sugar contents, low crude fiber and reduced cooking time.Keywords: soil fertility, grain yield, chickpea, natural resources.

Abstract:
The paper presents the results of research on trends in shaping of multifamily buildings in Poland on the example of Wrocław, after Polish accession to the European Union. The study is conducted within the research project: “Trends in creating of multifamily housing development since 2004, on the Wrocław study case" supported by Polish Ministry of Science and Higher Education and will be completed in November 2011. The research involves multifamily buildings completed in the last decade, in term of fundamental urbanization factors such as: building-s coefficient area, useable area, green area (biologically active surface), intensity of building development, amount of dwellings, dwelling area, amount of parking places, numbers of floors, etc. The analysis of these indicators was conducted based on the date obtained in the study of approximately one hundred new housing units, completed in Wroclaw. The analysis attempts to formulate the main trends in creating of housing policy in Poland during the last 10 years in reference to local urban policy.

Abstract:
Biodiesel production with used frying by transesterification reaction with methanol, using a commercial kaolinite thermally-activated solid acid catalyst was investigated. The surface area, the average pore diameter and pore volume of the kaolinite catalyst were 10 m2/g, 13.0 nm and 30 mm3/g, respectively. The optimal conditions for the transesterification reaction were determined to be oil/methanol, in a molar ratio 1:31, temperature 160 ºC and catalyst concentration of 3% (w/w). The yield of fatty acids methyl esters (FAME) was 92.4% after 2 h of reaction. This method of preparation of biodiesel can be a positive alternative for utilizing used frying corn oil for feedstock of biodiesel combined with the inexpensive catalyst.Keywords: Biodiesel, frying corn oil, kaolinite, transesterification
