2294 Cloning, Expression and Protein Purification of AV1 Gene of Okra Leaf Curl Virus Egyptian Isolate and Genetic Diversity between Whitefly and Different Plant Hosts

Abstract:
Begomoviruses are economically important plant viruses that infect dicotyledonous plants and exclusively transmitted by the whitefly Bemisia tabaci. Here, replicative form was isolated from Okra, Cotton, Tomato plants and whitefly infected with Begomoviruses. Using coat protein specific primers (AV1), the viral infection was verified with amplicon at 450 bp. The sequence of OLCuV-AV1 gene was recorded and received an accession number (FJ441605) from Genebank. The phylogenetic tree of OLCuV was closely related to Okra leaf curl virus previously isolated from Cameroon and USA with nucleotide sequence identity of 92%. The protein purification was carried out using His-Tag methodology by using Affinity Chromatography. The purified protein was separated on SDS-PAGE analysis and an enriched expected size of band at 30 kDa was observed. Furthermore, RAPD and SDS-PAGE were used to detect genetic variability between different hosts of okra leaf curl virus (OLCuV), cotton leaf curl virus (CLCuV), tomato yellow leaf curl virus (TYLCuV) and the whitefly vector. Finally, the present study would help to understand the relationship between the whitefly and different economical crops in Egypt.

Abstract:
One of the major problems in liberalized power markets is loss allocation. In this paper, a different method for allocating transmission losses to pool market participants is proposed. The proposed method is fundamentally based on decomposition of loss function and current projection concept. The method has been implemented and tested on several networks and one sample summarized in the paper. Abstract:
The nickel-manganese (Ni-Mn) alloy coating prepared from DC electrodeposition process in sulphamate bath was studied. The effects of process parameters, such as current density and electrolyte composition, on the cathodic current efficiency, microstructure, internal stress and mechanical properties were investigated. Because of its crucial effect on the application to the electroforming of microelectronic components, the development of low internal stress coating with high leveling power was emphasized. It was found that both the coating’s manganese content and the cathodic current efficiency increased with the raise in current density. In addition, the internal stress of the deposited coating showed compressive nature at low current densities while changed to tensile one at higher current densities. Moreover, the metallographic observation, X-ray diffraction measurement, and polarization curve measurement were conducted. It was found that the Ni-Mn coating consisted of nano-sized columnar grains and the maximum hardness of the coating was associated with (111) preferred orientation in the microstructure. The grain size was refined along with the increase in the manganese content of the coating, which accordingly, raised its hardness and resistance to annealing softening. In summary, the Ni-Mn coating prepared at lower current density of 1-2 A/dm2 had low internal stress, high leveling power, and better corrosion resistance.Keywords: DC plating, internal stress, leveling power, Ni-Mn coating.

Abstract:
A new low-voltage floating gate MOSFET (FGMOS) based squarer using square law characteristic of the FGMOS is proposed in this paper. The major advantages of the squarer are simplicity, rail-to-rail input dynamic range, low total harmonic distortion, and low power consumption. The proposed circuit is biased without body effect. The circuit is designed and simulated using SPICE in 0.25μm CMOS technology. The squarer is operated at the supply voltages of ±0.75V . The total harmonic distortion (THD) for the input signal 0.75Vpp at 25 KHz, and maximum power consumption were found to be less than 1% and 319μW respectively.Keywords: Analog signal processing, floating gate MOSFETs, low-voltage, Spice, squarer.

Abstract:
A method based on the power series solution is proposed to solve the natural frequency of flapping vibration for the rotating inclined Euler beam with constant angular velocity. The vibration of the rotating beam is measured from the position of the corresponding steady state axial deformation. In this paper the governing equations for linear vibration of a rotating Euler beam are derived by the d'Alembert principle, the virtual work principle and the consistent linearization of the fully geometrically nonlinear beam theory in a rotating coordinate system. The governing equation for flapping vibration of the rotating inclined Euler beam is linear ordinary differential equation with variable coefficients and is solved by a power series with four independent coefficients. Substituting the power series solution into the corresponding boundary conditions at two end nodes of the rotating beam, a set of homogeneous equations can be obtained. The natural frequencies may be determined by solving the homogeneous equations using the bisection method. Numerical examples are studied to investigate the effect of inclination angle on the natural frequency of flapping vibration for rotating inclined Euler beams with different angular velocity and slenderness ratio.Keywords: Flapping vibration, Inclination angle, Natural frequency, Rotating beam.

Abstract:
A field study was conducted to evaluate the efficacy of safflower plant for phytoremediation of contaminated soils. The experiment was performed on an agricultural fields contaminated by the Non-Ferrous-Metal Works near Plovdiv, Bulgaria. Field experiments with randomized complete block design with five treatments (control, compost amendments added at 20 and 40 t/daa, and vermicompost amendments added at 20 and 40 t/daa) were carried out. The quality of safflower seeds and oil (heavy metals and fatty acid composition) were determined. Tested organic amendments significantly influenced the chemical composition of safflower seeds and oil. The compost and vermicompost treatments significantly reduced heavy metals concentration in safflower seeds and oils, but the effect differed among them. Addition of vermicompost and compost leads to an increase in the content of palmitic acid and linoleic acid, and a decrease in the stearic and oleic acids compared with the control. A significant increase in the quantity of saturated acids was observed in the variants with 20 t/daa of compost and 20 t/daa of vermicompost (9.1 and 8.9% relative to the control). Safflower is a plant which is tolerant to heavy metals and can be successfully used in the phytoremediation of heavy metal contaminated soils. The processing of seeds to oil and using the obtained oil for nutritional purposes will greatly reduce the cost of phytoremediation.Keywords: Heavy metals, organic amendments, phytoremediation, safflower.

This paper describes the development of a WLAN propagation model, using Spectral Analyzer measurements. The signal is generated by two Access Points (APs) on the base floor at the administrative Communication School of ESPOL building. In general, users do not have a Q&S reference about a wireless network; however, this depends on the level signal as a function of frequency, distance and other path conditions between receiver and transmitter. Then, power density of the signal decrease as it propagates through space and data transfer rate is affected. Diabetes mellitus (DM) is frequently characterized by autonomic nervous dysfunction. Analysis of heart rate variability (HRV) has become a popular noninvasive tool for assessing the activities of autonomic nervous system (ANS). In this paper, changes in ANS activity are quantified by means of frequency and time domain analysis of R-R interval variability. Electrocardiograms (ECG) of 16 patients suffering from DM and of 16 healthy volunteers were recorded. Frequency domain analysis of extracted normal to normal interval (NN interval) data indicates significant difference in very low frequency (VLF) power, low frequency (LF) power and high frequency (HF) power, between the DM patients and control group. Time domain measures, standard deviation of NN interval (SDNN), root mean square of successive NN interval differences (RMSSD), successive NN intervals differing more than 50 ms (NN50 Count), percentage value of NN50 count (pNN50), HRV triangular index and triangular interpolation of NN intervals (TINN) also show significant difference between the DM patients and control group.Keywords: Autonomic nervous system, diabetes mellitus, frequency domain and time domain analysis, heart rate variability.

Abstract:
Thermal performance is considered to be a key measure in building sustainability. One of the technologies used in the current building sustainable design is the rooftop solar PV power generators. The application of this type of technology has expanded vastly during the last five years in many countries. This paper studies the effect of roof shading developed by the solar PV panels on dwellings’ thermal performance. The analysis in this work is performed by using two types of packages: “AccuRate Sustainability” for rating the energy efficiency of residential building design, and “PVSYST” for the solar PV power system design. The former package is used to calculate the annual heating and cooling load, and the later package is used to evaluate the power production from the roof top PV system. The analysis correlates the electrical energy generated from the PV panels to the change in the heating and cooling load due to roof shading. Different roof orientation, roof inclination, roof insulation, as well as PV panel area are considered in this study. The analysis shows that the drop in energy efficiency due to the shaded area of the roof by PV panels is negligible compared to the energy generated by these panels.

Abstract:
This paper presents an effective technique for harmonic current mitigation using an adaptive notch filter (ANF) to estimate current harmonics. The proposed filter consists of multiple units of ANF connected in parallel structure; each unit is governed by two ordinary differential equations. The frequency estimation is carried out based on the output of these units. The simulation and experimental results show the ability of the proposed tracking scheme to accurately estimate harmonics. The proposed filter was implemented digitally in TMS320F2808 and used in the control of hybrid active power filter (HAPF). The theoretical expectations are verified and demonstrated experimentally.

Abstract:
A healthcare monitoring system is presented in this paper. This system is based on ultra-low power sensor nodes and a personal server, which is based on hardware and software extensions to a Personal Digital Assistant (PDA)/Smartphone. The sensor node collects data from the body of a patient and sends it to the personal server where the data is processed, displayed and made ready to be sent to a healthcare network, if necessary. The personal server consists of a compact low power receiver module and equipped with a Smartphone software. The receiver module takes less than 30 × 30 mm board size and consumes approximately 25 mA in active mode.Keywords: healthcare monitoring, sensor node, personal server, wireless.

Abstract:
Plant tissue culture is an important in vitro technology applied for agricultural and industrial production. A sterile condition of culture medium is one of the main aspects. The alternative technique for medium sterilization to replace autoclaving was carried out. For sterilization of plant tissue culture medium without autoclaving, ten commercial pure essential oils and 5 disinfectants were tested. Each essential oil or disinfectant was added to a 20-mL Murashige and Skoog (MS) medium before medium was solidified in a 120-mL container, kept for 2 weeks before evaluating sterile conditions. Treated media, supplemented with essential oils or disinfectants, were compared to control medium, autoclaved at 121 degree Celsius for 15 min. Sterile conditions of MS medium were found 100% from betel oil or clove oil (18 mL/20 mL medium), cinnamon oil (36 mL/20 mL medium), lavender oil or holy basil oil (108 mL/20 mL medium), and lemon oil or tea tree oil or turmeric oil (252 mL/20 mL medium), compared to 100% sterile condition from autoclaved medium. For disinfectants, 2% iodine + 2.4% potassium iodide, 2% merbromine solution, 10% povidone-iodine, 6% sodium hypochlorite or 0.1% thimerosal at 36 mL/20 mL medium provided 100% sterile conditions. Furthermore, growth of new shoots from chrysanthemum node explants on treated media (fresh weight, shoot length, root length and number of node) were also reported and discussed in the comparison of those on autoclaved medium.

This paper presents analysis and characterization of a piezoelectric micro-generator for energy harvesting application. A low-cost experimental prototype was designed to operate as piezoelectric micro-generator in the laboratory. An input acceleration of 9.8m/s2 using a sine signal (peak-to-peak voltage: 1V, offset voltage: 0V) at frequencies ranging from 10Hz to 160Hz generated a maximum average power of 432.4μW (linear mass position = 25mm) and an average power of 543.3μW (angular mass position = 35°). These promising results show that the prototype can be considered for low consumption load application as an energy harvesting micro-generator.Keywords: Piezoelectric, microgenerator, energy harvesting, cantilever beam.

In this paper, we propose an architecture for easily constructing a robot controller. The architecture is a multi-agent system which has eight agents: the Man-machine interface, Task planner, Task teaching editor, Motion planner, Arm controller, Vehicle controller, Vision system and CG display. The controller has three databases: the Task knowledge database, the Robot database and the Environment database. Powerline Communications –PLC– as an alternative method for broadband networking, has the advantage of transmitting over channels already used for electrical distribution or even transmission. But these channels have been not designed with usual wired channels requirements for broadband applications such as stable impedance or known attenuation, and the network have to reject noises caused by electrical appliances that share the same channel. Noise control standards are difficult to complain or simply do not exist on Latin-American environments. This paper deals with rheological behavior of tomato paste from the view point of time independent properties inclusive of processing variables such as sample temperature which influence on rheological properties as well as breaking temperature and concentration which beside the rheological properties, influence on the quality of final product. With this aim 10 tomato paste samples at various concentrations (17-25%) and breaking temperatures (65- 85 C o ) have been produced. Distributed Power generation has gained a lot of attention in recent times due to constraints associated with conventional power generation and new advancements in DG technologies .The need to operate the power system economically and with optimum levels of reliability has further led to an increase in interest in Distributed Generation. However it is important to place Distributed Generator on an optimum location so that the purpose of loss minimization and voltage regulation is dully served on the feeder. This paper investigates the impact of DG units installation on electric losses, reliability and voltage profile of distribution networks. In this paper, our aim would be to find optimal distributed generation allocation for loss reduction subjected to constraint of voltage regulation in distribution network. The system is further analyzed for increased levels of Reliability. Distributed Generator offers the additional advantage of increase in reliability levels as suggested by the improvements in various reliability indices such as SAIDI, CAIDI and AENS. Comparative studies are performed and related results are addressed. An analytical technique is used in order to find the optimal location of Distributed Generator. The suggested technique is programmed under MATLAB software. In this research, firstly, a commercial gas sweetening unit with methyl-di-ethanol-amine (MDEA) solution is simulated and comprised in an integrated model in accordance with Aspen HYSYS software. For evaluation purposes, in the second step, the results of the simulation are compared with operating data gathered from South Pars Gas Complex (SPGC). According to the simulation results, the considerable energy potential contributed to the pressure difference between absorber and regenerator columns causes this energy driving force to be applied in power recovery turbine (PRT). In the last step, the amount of waste hydraulic energy is calculated, and its recovery methods are investigated.

This paper describes a 2.4 GHz passive switch mixer and a 5/2.5 GHz voltage-controlled negative Gm oscillator (VCO) with an inversion-mode MOS varactor. Both circuits are implemented using a 1P8M 0.13 μm process. The switch mixer has an input referred 1 dB compression point of -3.89 dBm and a conversion gain of -0.96 dB when the local oscillator power is +2.5 dBm. The VCO consumes only 1.75 mW, while drawing 1.45 mA from a 1.2 V supply voltage. In order to reduce the passives size, the VCO natural oscillation frequency is 5 GHz. A clocked CMOS divideby- two circuit is used for frequency division and quadrature phase generation. The VCO has a -109 dBc/Hz phase noise at 1 MHz frequency offset and a 2.35-2.5 GHz tuning range (after the frequency division), thus complying with ZigBee requirements.Keywords: Switch Mixers, Varactors, IEEE 802.15.4 (ZigBee), Direct Conversion Receiver, Wireless Sensor Networks.

This paper presents an inverting high step-up DC/DC converter. Basically, this high step-up DC/DC converter is an appealing interface for solar applications. The proposed topology takes advantage of using coupled inductors. Due to the leakage inductances of these coupled inductors, the power MOSFET has the zero current switching (ZCS) condition, which results in decreased switching losses. This will substantially improve the overall efficiency of the power converter. Furthermore, employing coupled inductors has led to a higher voltage gain. The paper presents actual benefits and drawbacks of a multidirectional autonomous hovercraft conceived with limited resources and designed for indoor exploration. Recent developments in the field have led to the apparition of very powerful automotive systems capable of very high calculation and exploration in complex unknown environments. They usually propose very complex algorithms, high precision/cost sensors and sometimes have heavy calculation consumption with complex data fusion. These systems are usually powerful but have a certain price, and the benefits may not be worth the cost, especially considering their hardware limitations and their power consumption. The present approach is to build a compromise between cost, power consumption and results preciseness.

Although lighting systems powered by Photovoltaic (PV) cells have existed for many years, they are not widely used, especially in lighting for buildings, due to their high initial cost and low conversion efficiency. One of the technical challenges facing PV powered lighting systems has been how to use dc power generated by the PV module to energize common light sources that are designed to operate efficiently under ac power. Usually, the efficiency of the dc light sources is very poor compared to ac light sources. Rapid developments in LED lighting systems have made this technology a potential candidate for PV powered lighting systems. Abstract:
Doubly-fed induction generator (DFIG) is currently the choice for many wind turbines. These generators, when connected to the grid through a converter, is subjected to varied power system conditions like voltage variation, frequency variation, short circuit fault conditions, etc. Further, many countries like Canada, Germany, UK, Scotland, etc. have distinct grid codes relating to wind turbines. Accordingly, following the network faults, wind turbines have to supply a definite reactive current. To satisfy the requirements including reactive current capability, an optimum electrical design becomes a mandate for DFIG to function. This paper intends to optimize the equivalent circuit parameters of an electrical design for satisfactory DFIG performance. Direct search method has been used for optimization of the parameters. The variables selected include electromagnetic core dimensions (diameters and stack length), slot dimensions, radial air gap between stator and rotor and winding copper cross section area. Optimization for 2 MW DFIG has been executed separately for three objective functions - maximum reactive power capability (Case I), maximum efficiency (Case II) and minimum weight (Case III). In the optimization analysis program, voltage variations (10%), power factor- leading and lagging (0.95), speeds for corresponding to slips (-0.3 to +0.3) have been considered. The optimum designs obtained for objective functions were compared. It can be concluded that direct search method of optimization helps in determining an optimum electrical design for each objective function like efficiency or reactive power capability or weight minimization.

Abstract:
Voltage stability has become an important issue to many power systems around the world due to the weak systems and long line on power system networks. In this paper, MATLAB load flow program is applied to obtain the weak points in the system combined with finding the voltage stability limit. The maximum permissible loading of a system, within the voltage stability limit, is usually determined. The methods for varying tap ratio (using tap changing transformer) and applying different values of shunt capacitor injection to improve the voltage stability within the limit are also provided.

Abstract:
An innovative approach to develop modified scaling free CORDIC based two parallel pipelined Multipath Delay Commutator (MDC) FFT and IFFT architectures for radix 22 FFT algorithm is presented. Multipliers and adders are the most important data paths in FFT and IFFT architectures. Multipliers occupy high area and consume more power. In order to optimize the area and power overhead, modified scaling-free CORDIC based complex multiplier is utilized in the proposed design. In general twiddle factor values are stored in RAM block. In the proposed work, modified scaling-free CORDIC based twiddle factor generator unit is used to generate the twiddle factor and efficient switching units are used. In addition to this, four point FFT operations are performed without complex multiplication which helps to reduce area and power in the last two stages of the pipelined architectures. The design proposed in this paper is based on multipath delay commutator method. The proposed design can be extended to any radix 2n based FFT/IFFT algorithm to improve the throughput. The work is synthesized using Synopsys design Compiler using TSMC 90-nm library. The proposed method proves to be better compared to the reference design in terms of area, throughput and power consumption. Abstract:
Cereals are considered as a strategic product in human life and their demand is increasing with the growth of world population. Increasing wheat production is important for the country. One of the ways to solve the problem is to develop and implement new, environmentally and economically acceptable technologies. Such technologies include pre-sowing treatment of seed with a laser and associative nitrogen-fixing bacteria - Azospirillum brasilense. In the region there are the wheat varieties - Dika and Lomtagora, which are among the most common in Georgia. Dika is a frost-resistant wheat, with a high ability to adapt to the environment, resistant to falling and it is sown in highlands. Lomtagora 126 differs with its winter and drought resistance, and it has a great ability to germinate. Lomtagora is characterized by a strong root system and a high budding capacity. It is an early variety, fall-resistant, easy to thresh and suitable for mechanized harvesting with large and red grains. This paper presents some preliminary experimental results where a continuous CO2 laser with a power of 25-40 W was used to radiate grains at a flow rate of 10 and 15 cm/sec. The treatment was carried out on grains of the Triticum aestivum L. var. Lutescens (local variety name - Lomtagora 126), and Triticum carthlicum Nevski (local variety name - Dika). Here the grains were treated with A. brasilense isolate (108-109 CFU/ml), which was isolated from the rhizosphere of wheat. It was observed that the germination of the wheat was not significantly influenced by either laser or bacteria treatment. The results of our research show that combined treatment with laser and A. brasilense significantly influenced the germination of wheat. In the case of the Lomtagora 126 variety, grains were exposed to the beam on a speed of 10 cm/sec, only slightly improved the growth for 38-day seedlings, in case of exposition of grains with a speed of 15 cm/sec - by 23%. Treatment of seeds with A. brasilense in both exposed and non-exposed variants led to an improvement in the growth of seedlings, with A. brasilense alone - by 22%, and with combined treatment of grains - by 29%. In the case of the Dika variety, only exposure led to growth by 8-9%, and the combined treatment - by 10-15%, in comparison with the control variant. Superior effect on growth of seedlings of different varieties was achieved with the combinations of laser treatment on grains in a beam of 15 cm/sec (radiation power 30-40 W) and in addition of A. brasilense - nitrogen fixing bacteria. Therefore, this is a promising application of A. brasilense as active agents of bacterial fertilizers due to their ability of molecular nitrogen fixation in cereals in combination with laser irradiation: choosing a proper strain gives a good ability to colonize roots of agricultural crops, providing a high nitrogen-fixing ability and the ability to mobilize soil phosphorus, and laser treatment stimulates natural processes occurring in plant cells, will increase the yield.

Abstract:
The interaction of the blade tip with the casing boundary layer and the leakage flow may lead to a kind of cavitation namely tip vortex cavitation. In this study, the onset of tip vortex cavitation was experimentally investigated in an axial flow pump. For a constant speed and a fixed angle of attack and by changing the flow rate, the pump head, input power, output power and efficiency were calculated and the pump characteristic curves were obtained. The cavitation phenomenon was observed with a camera and a stroboscope. Finally, the critical flow region, which tip vortex cavitation might have occurred, was identified. The results show that just by adjusting the flow rate, out of the specified region, the possibility of occurring tip vortex cavitation, decreases to a great extent.Keywords: Axial flow pump, Gap cavitation, Leakage vortex, Tip vortex cavitation.

Various genes which control or affect heading time have been found in rice. Out of them, Se1 and E1 loci play important roles in determining heading time by controlling photosensitivity. An isogenic-line pair of late and early lines were developed from progenies of the F1 from Suweon 258 × 36U. A lateness gene tentatively designated as “Ex” was found to control the difference in heading time between the early and late lines mentioned above. The present study was conducted to examine the effect of Ex on yield and related traits. Indica-type variety Suweon 258 was crossed with 36U, which is an Ur1 (Undulate rachis-1) isogenic line of IR36. In the F2 population, comparatively early-heading, late-heading and intermediate-heading plants were segregated. Segregation similar to that by the three types of heading was observed in the F3 and later generations. A late-heading plant and an early-heading plant were selected in the F8 population from an intermediate-heading F7 plant, for developing L and E of the isogenic-line pair, respectively. Experiments for L and E were conducted by randomized block design with three replications. Transplanting was conducted on May 3 at a planting distance of 30 cm × 15 cm with two seedlings per hill to an experimental field of the Faculty of Agriculture, Kochi University. Chemical fertilizers containing N, P2O5 and K2O were applied at the nitrogen levels of 4 g/m2, 9 g/m2 and 18 g/m2 in total being denoted by "N4", "N9" and "N18", respectively. Yield, yield components and other traits were measured. Ex delayed 80%-heading by 17 or 18 days in L as compared with E. In total brown rice yield (g/m2), L was 635, 606 and 590, and E was 577, 548 and 501, respectively, at N18, N9 and N4, indicating that Ex increased this trait by 10% to 18%. Ex increased yield-1.5 mm sieve (g/m2) b 9% to 15% at the three fertilizer levels. Ex increased the spikelet number per panicle by 16% to 22%. As a result, the spikelet number per m2 was increased by 11% to 18% at the three fertilizer levels. Ex decreased 1000-grain weight (g) by 2 to 4%. L was not significantly different from E in ripened-grain percentage, fertilized-spikelet percentage and percentage of ripened grains to fertilized spikelets. Hence, it is inferred that Ex increased yield by increasing spikelet number per panicle. Hence, Ex could be utilized to develop high yielding varieties for warmer districts.

Abstract:
This paper explains a project based learning method where autonomous mini-robots are developed for research, education and entertainment purposes. In case of remote systems wireless sensors are developed in critical areas, which would collect data at specific time intervals, send the data to the central wireless node based on certain preferred information would make decisions to turn on or off a switch or control unit. Such information transfers hardly sums up to a few bytes and hence low data rates would suffice for such implementations. As a robot is a multidisciplinary platform, the interfacing issues involved are discussed in this paper. The paper is mainly focused on power supply, grounding and decoupling issues.
