Authors: Refal Hussain, Saifuddin M. Nomanbhay
Abstract:
Glycerol is a valuable raw material for the production of industrially useful metabolites. Among many promising applications for the use of glycerol is its bioconversion to high value-added compounds, such as bioethanol through microbial fermentation. Bioethanol is an important industrial chemical with emerging potential as a biofuel to replace vanishing fossil fuels. The yield of liquid fuel in this process was greatly influenced by various parameters viz, temperature, pH, glycerol concentration, organic concentration, and agitation speed were considered. The present study was undertaken to investigate optimum parameters for bioethanol production from raw glycerol by immobilized mutant Escherichia coli (E.coli) (ATCC11505) strain on chitosan cross linked glutaraldehyde optimized by Taguchi statistical method in shake flasks. The initial parameters were set each at four levels and the orthogonal array layout of L16 (45) conducted. The important controlling parameters for optimized the operational fermentation was temperature 38 °C, medium pH 6.5, initial glycerol concentration (250 g/l), and organic source concentration (5 g/l). Fermentation with optimized parameters was carried out in a custom fabricated shake flask. The predicted value of bioethanol production under optimized conditions was (118.13 g/l). Immobilized cells are mainly used for economic benefits of continuous production or repeated use in continuous as well as in batch mode. Keywords: bioethanol, Escherichia coli, immobilization, optimization

Abstract:
Melia azedarach L. is freshly fruited small to medium sized tree native to China and North western India. It is growing in Pakistan and Turkey in various areas facing great environmental changes to maintain its survival. The species is valued for its high quality wood, medicinal, ornamental and shade purposes. The present work was aimed to estimate the genetic variation among the populations of Melia azedarach L. leaf samples that were collected from five different locations of Turkey and three different areas of Pakistan. These populations were chosen on the random bases by applying RAPD primers in order to construct a dendogram using UPGMA method to show genetic diversity. After that appropriate conservation strategies were suggested. 14 primers producing polymorphic and monomorphic bands were analyzed. Genetic distances were calculated for all the species studied by RAPD-PCR methods. According to the results the lowest genetic identity values and the highest genetic polymorphic values were determined. It is observed that there was a clear split among populations from different areas in Turkey and Pakistan. These differences may be due to eco-geographical association with genetic variation and should be conserved to retain the genetic variation of the species. Keywords: melia azedarach L., genetic diversity, conservation, RAPD-PCR, medicinal plant

Abstract:
Many places worldwide, especially arid and semi-arid remote regions, are suffering from the lack of drinkable water and the situation will be aggravated in the near future. Furthermore, remote areas are characterised by lack of conventional energy sources, skilled personnel and maintenance facilities. Therefore, the development of small to medium size, stand-alone and robust solar desalination systems is needed to provide independent fresh water supply in remote areas. This paper is focused on experimental studies on compact membrane distillation (MD) solar desalination prototype located at the Mechanical Engineering Department site, Kairouan University, Kairouan, Tunisia. The pilot system is designed and manufactured as a part of a research and development project funded by the MESRS/BMBF. The pilot system is totally autonomous. The electrical energy required to operate the unit is generated through a field of 4 m² of photovoltaic panels, and the heating of feed water is provided by a field of 6 m² of solar collectors. The Kairouan plant performance of the first few months of operation is presented. The highest freshwater production of 150 L/d is obtained on a sunny day in July of 633 W/m²d.Keywords: experimental, membrane distillation, solar desalination, Permeat gap

Authors: Fatemeh Soltani, Simindokht Shirvani Arani, Ali Bahrami Samani, Mahdi Sadeghi, Kamal Yavari
Abstract:
Cerium-141 [T1/2 = 32.501 days, Eβ (max) = 0.580 (29.8%) and 0.435(70.2%) MeV, Eγ=145.44 (48.2%) keV] possesses radionuclidic properties suitable for use in palliative therapy of bone metastases. 141Ce also has gamma energy of 145.44 keV, which resembles that of 99mTc. Therefore, the energy window is adjustable on the Tc-99m energy because of imaging studies. 141Ce can be produced through a relatively easy route that involves thermal neutron bombardment on natural CeO2 in medium flux research reactors (4–5×1013 neutrons/cm2•s). The requirement for an enriched target does not arise. Ethylenediamine tetramethylene phosphonic acid (EDTMP) was synthesized and radiolabeled with 141Ce. Complexation parameters were optimized to achieve maximum yields (>99%). The radiochemical purity of 141Ce-EDTMP was evaluated by radio-thin layer chromatography. The stability of the prepared formulation was monitored for one week at room temperature, and results showed that the preparation was stable during this period (>99%). Biodistribution studies of the complexes carried out in wild-type rats exhibited significant bone uptake with rapid clearance from blood. The properties of produced 141Ce-EDTMP suggest applying a new efficient bone pain palliative therapeutic agent to overcome metastatic bone pains. Keywords: bone pain palliative, cerium-141, EDTMP, radiopharmaceutical

Authors: Khalid Kaddoura, Tarek Zayed
Abstract:
Infrastructure assets are essential in urban cities; their purpose is to facilitate the public needs. As a result, their conditions and states shall always be monitored to avoid any sudden malfunction. Sewer systems, one of the assets, are an essential part of the underground infrastructure as they transfer sewer medium to designated areas. However, their conditions are subject to deterioration due to ageing. Therefore, it is of great significance to assess the conditions of pipelines to avoid sudden collapses. Current practices of sewer pipeline assessment rely on industrial protocols that consider distinct defects and grades to conclude the limited average or peak score of the assessed assets. This research aims to enhance the evaluation by integrating the Quality Function Deployment (QFD) and the Decision-Making Trial and Evaluation Laboratory (DEMATEL) methods in assessing the condition of sewer pipelines. The methodology shall study the cause and effect relationship of the systems’ defects to deduce the relative influence weights of each defect. Subsequently, the overall grade is calculated by aggregating the WHAT’s and HOW’s of the House of Quality (HOQ) using the computed relative weights. Thus, this study shall enhance the evaluation of the assets to conclude informative rehabilitation and maintenance plans for decision makers. Keywords: condition assessment, DEMATEL, QFD, sewer pipelines

Procedia PDF Downloads 441267 Reduced Switch Count Asymmetrical Multilevel Inverter Topology
Authors: Voodi Kalandhar, Veera Reddy, Yuva Tejasree
Abstract:
Researchers have become interested in multilevel inverters (MLI) because of their potential for medium- and high-power applications. MLIs are becoming more popular as a result of their ability to generate higher voltage levels, minimal power losses, small size, and low price. These inverters used in high voltage and high-power applications because the stress on the switch is low. Even though many traditional topologies, such as the cascaded H-bridge MLI, the flying capacitor MLI, and the diode clamped MLI, exist, they all have some drawbacks. A complicated control system is needed for the flying capacitor MLI to balance the voltage across the capacitor and diode clamped MLI requires more no of diodes when no of levels increases. Even though the cascaded H-Bridge MLI is popular in terms of modularity and simple control, it requires more no of isolated DC source. Therefore, a topology with fewer devices has always been necessary for greater efficiency and reliability. A new single-phase MLI topology has been introduced to minimize the required switch count in the circuit and increase output levels. With 3 dc voltage sources, 8 switches, and 13 levels at the output, this new single- phase MLI topology was developed. To demonstrate the proposed converter's superiority over the other MLI topologies currently in use, a thorough analysis of the proposed topology will be conducted. Keywords: DC-AC converter, multi-level inverter (MLI), diodes, H-bridge inverter, switches

Authors: Mohammed Gamil Montasser, Angelo Battaglia
Abstract:
Entrepreneurship is mostly related to the beginning of organization. In growing business organizations, entrepreneurship expands its conceptualization. It reveals itself through new business creation in the active organization, through renewal, change, innovation, creation and development of current organization, through breaking and changing of established rules inside or outside the organization and becomes more flexible, adaptive and competitive, also improving effectiveness of organization activity. Therefore, the topic of entrepreneurship, relates the creation of firms to personal / individual characteristics of the entrepreneurs and their social context. This paper is an empirical study, which aims to address these two gaps in the literature. For this endeavor, we use the latest available data from the Global Entrepreneurship Monitor (GEM) project. This data set is widely regarded as a unique source of information about entrepreneurial activity, as well as the aspirations and attitudes of individuals across a wide number of countries and territories worldwide. This paper tries to contribute to fill this gap, by exploring the key drivers of innovative entrepreneurship in the tourism sector. Our findings are consistent with the existing literature in terms of the individual characteristics of entrepreneurs, but quite surprisingly we find an inverted U-shape relation between human development and innovative entrepreneurship in tourism sector. It has been revealed that tourism entrepreneurs are less likely to have innovative products, compared with entrepreneurs in medium developed countries. Keywords: GEM, human development, innovative entrepreneurship, occupational choice, tourism

Abstract:
The natural convection cooling system of a fully immersed server in a dielectric liquid is studied numerically. In the present case study, the dielectric liquid represents working fluid and it is in contact with server inside capsule. The capsule includes electronic component and fluid which can be modeled as saturated porous media. This medium follow Darcy flow regime and assumed to be in balance between its components. The study focus is on role of spatial parameters on thermal behavior of convective heat transfer. Based on server known unit, which is 1U, two parameters Ly and S are changed to test their effect. Meanwhile, wide-range of modified Rayleigh number, which is 0.5 to 300, are covered to better understand thermal performance. Navier-Stokes equations are used to model physical domain. Furthermore, successive over-relaxation and time marching techniques are used to solve momentum and energy equation. From obtained correlation, the in-between distance S is more effective on Nusselt number than distance to edge Ly by approximately 14%. In addition, as S increases, the average Nusselt number of the upper unit increases sharply, whereas the lower one keeps on the same level. Keywords: convective cooling of server, Darcy flow, liquid-immersed server, porous media

Authors: Min-Shiuan Liou, Yi Shan Yang, Yang-Zhan Huang, Chia-Wen Hsieh
Abstract:
A high speed growing and butanol-tolerant Clostridium acetobutylicum HOL1 mutant was screened throughout continuous adaption culture with C. acetobutylicum ATCC 824. The HOL1 strain can grow well in 10 g/L butanol contained CGM medium and can produce about 12.8 g /L butanol during 24 hrs. The C. acetobutylicum HOL1 strain was able to produce 166 mM butanol with 21 mM acetone at pH 4.8, resulting in a butanol selectivity (a molar ratio of butanol to total solvents) of 0.79, which is much higher than that (0.6) of the wild-type strain C. acetobutylicum ATCC 824. The acetate and butyrate accumulation were not observed during fermentation of the HOL1 strain. A hyper-butanol producing C. acetobutylicum HOL1 (pBPHS-3), which was created to overexpress the Bacillus psychrosaccharolyticus originated specific heat-shock protein gene, hspX, from a clostridial phosphotransbutyrylase promoter, was studied for its potential to produce a high titer of butanol. Overexpression of hspX resulted in increased final butanol yield 47% and 30% higher than those of the the ATCC824 and the HOL1 strains, respectively. The remarkable high-speed growth and butanol tolerance of strain HOL1 (pBPHS-3) demonstrates that overexpression of heterogeneous stress protein-encoding gene, hspX, could help C. acetobutylicum to effectively produce a high concentration of butanol. Keywords: Clostridium acetobutylicum, butanol, heat-shock protein, resistance

Authors: Fung Chiat Loo, Fung Ying Loo
Abstract:
This paper discusses, suggests, and constructs a notation system to facilitate the learning and understanding of the two aspects of music and movement in a sports routine. This model serves to provide a simple and logical notation that does not require training in both music and choreography. Notation is an important medium in many art forms, particularly in music and dance, transmitting information that cannot easily be expressed using words or language. Another field that is closely associated with dance and music is sports routine, which equally requires choreography and music. However, from the perspective of music, it is common to observe many incongruencies appearing between the music used and the choreography that impede an optimal perception of the performance. The concept of the notation proceeds with a discussion and review of existing dance notations that could contribute to sports routines, along with rules and a code of points in selected sports routines. The author's involvement as an insider of numerous musical theatre productions also contributed to this study. The notation constructed includes time (tempo), significances of musical accents, direction, and phrasing, along with significances of movements (jump, punch, shape). It is believed that the level of congruence between music and movement will provide optimal visualization, and in that, the notation serves to provide adequate information on both entities for the understanding of athletes and coaches. Keywords: notation, choreography, music learning, sports routines, congruence

Authors: Aymen H. Khalil, Ashraf M. Heniegal, Bassam A. Abdelsalam
Abstract:
There are many problems related to cantilever slabs such as the time-dependent deformation, corrosion problems of steel reinforcement, and lack of experimental studies on the strength of strengthened cantilever slabs. This paper presents an investigation to evaluate the behavior of reinforced concrete cantilever slabs after strengthening with different techniques. Six medium scale specimens, divided into three groups, were tested along with a control slab. The first group consists of two specimens which were repaired and strengthened using reinforced concrete jacket above with and without shear connector bars, whereas the second group contained two slabs which were strengthened using two strips of two layers of glass fiber reinforced polymer (GFRP) covering 60% and 90% from the cantilever length. The last group involves two specimens strengthened with two steel plates. In one specimen, the steel plates were glued to the surface using epoxy resin. The second specimen, the steel plates were affixed to the concrete surface using expansion bolts. The loading was conducted in two phases. Firstly, the samples were subjected to 40% of the ultimate load of the control slab. Secondly, the specimens reloaded after being strengthened up to failure. The load-deflection, steel strain, concrete strain, failure mode, toughness, and ductility index are discussed in this paper. Keywords: repair, strengthened, GFRP layers, reloaded, jacketing, cantilever slabs

Abstract:
The conservation programme of various vascular plant species has been started in the Botanical garden o fEszterházy College in Eger cooperating with two national parks in the Northern mountain region and Botanical garden of Eötvös Lóránd University in Budapest. The seeds of the species were collected in the chosen habitats with the permission determined by the National Parks and the conservation specialists. Now we have different numbers of individuals from mainly endemic and relict species. We took some experiments to know how can we germinate and grow up this species succesfully up to blooming and fruiting. In the temperate zone the majority of species after ripening the seeds or corps get dormancy to avoid the inadequate period to germinate. The seeds of species need variously pre-treatment (for example pre-chill) and suitable environment (for example basic medium) to unlock the seed dormancy and germinate in large scale. This impacts are often similar to in their originally habitat. To bloom the plants need suitable types of soil, but we couldn’t grow them in the most fruitful soil of habitat. Suitable microclimate is usually more important for some relict species than the soil, that’s why should we make experiments to find the suitable essential conditions for different species and know all of fenological states of them. These experiments can start a method for growing common wild native plants as food materials. Keywords: ex situ conservation, germination success, soil preference Hungary, regionality, native wild plants

Abstract:
Based on the geodynamic environment, basic geological characteristics of minerals and so on, gold deposits in China are divided into 11 categories, of which tectonic fracture altered rock, mid-intrudes and contact zone, micro-fine disseminated and continental volcanic types are the main prospecting kinds. The metallogenic age of gold deposits in China is dominated by the Mesozoic and Cenozoic. According to the geotectonic units, geological evolution, geological conditions, spatial distribution, gold deposits types, metallogenic factors etc., 42 gold concentration areas are initially determined and have a concentrated distribution feature. On the basis of the gold exploration density, gold concentration areas are divided into high, medium and low level areas. High ones are mainly distributed in the central and eastern regions. 93.04% of the gold exploration drillings are within 500 meters, but there are some problems, such as less and shallower of drilling verification etc.. The paper discusses the resource potentials of gold deposits and proposes the future prospecting directions and suggestions. The deep and periphery of old mines in the central and eastern regions and western area, especially in Xinjiang and Qinghai, will be the future key prospecting one and have huge potential gold reserves. If the exploration depth is extended to 2,000 meters shallow, the gold resources will double. Keywords: gold deposits, gold deposits types, gold concentration areas, prospecting, resource potentiality

Authors: Maryam Aloshan, Imam Mohammad Ibn Saud, Ahmad Khudair
Abstract:
This study assesses the data governance maturity of nonprofit organizations in Riyadh, Saudi Arabia, using the National Data Maturity Index (NDI) framework developed by the Saudi Data and Artificial Intelligence Authority (SDAIA). Employing a survey designed around the NDI model, data maturity levels were evaluated across 14 dimensions using a 5-point Likert scale. The results reveal a spectrum of maturity levels among the organizations surveyed: while some medium-sized associations reached the ‘Defined’ stage, others, including large associations, fell within the ‘Absence of Capabilities’ or ‘Building’ phases, with no organizations achieving the advanced ‘Established’ or ‘Pioneering’ levels. This variation suggests an emerging recognition of data governance but underscores the need for targeted interventions to bridge the maturity gap. The findings point to a significant opportunity to elevate data governance capabilities in Saudi nonprofits through customized capacity-building initiatives, including training, mentorship, and best practice sharing. This study contributes valuable insights into the digital transformation journey of the Saudi nonprofit sector, aligning with national goals for data-driven governance and organizational efficiency. Keywords: nonprofit organizations-national data maturity index (NDI), Saudi Arabia- SDAIA, data governance, data maturity

But being a part of global trends while constantly pushing the boundaries of storytelling in virtual reality and standing on the edge of technology also brings huge responsibility for game designers around the globe. A very recent emerging topic over the last years was an individual's mental state. In recent years there has been a shift in mental problems representations in commercial game releases such as Hell blade: Senua's Sacrifice and Sea of Solitude. The aim of this study is to research the approach of mental illness representation in media and digital games over the years and to suggest alternatives for putting characters who suffer from mental illness at the forefront of the storyline. This study traces dominant representations of characters with mental illness in digital games, reflecting the major change of the game industry toward inclusiveness. At the same time, the research embraces a hybrid approach to the academic study of digital games and includes the development of a game that follows a post-traumatic young girl, forcing the users to live her life through her eyes. The game prototype was developed as part of the Mdes Game Design and Development program and consisted of academic research and game development practices. Keywords: framing analysis, mental condition, up keying, game mechanics

Abstract:
Microplastics (MPs; < 5 mm) have been cited as exceptionally detrimental to marine organisms and ocean health. They can carry other pollutants and abundant microbes that can serve as food for other organisms. Their small particle size and high abundance means that non-discriminatory feeders may ingest MPs involuntarily and microbial colonization of the particles (a niche coined ‘Plastisphere’) could facilitate particle ingestion. To assess how marine nematodes, the most abundant member of the meiofauna (32-500 um), are affected by microbe-MP associations, an experiment was conducted with three MP concentrations (low, medium, and expected high values of MPs in a local bay system), and two levels of microbe-MP associations (absence or presence). MPs were introduced into sediment microcosms and treatments were removed at three distinct time points (0, 3, and 7 days) to measure mean MP consumption/individual nematode. The quantitative results from this work should inform on microbial facilitation of MP ingestion and MP effects on seafloor ecology. As most MP feeding experiments use straight-from-package or sterile MPs, this work represents an important step in realizing the effects of MPs and their plastispheres in coastal sediments where they likely accumulate microbial biofilms prior to their ingestion by marine metazoans. Furthermore, the results here convey realistic effects of MPs on faunal behaviors, as the MP concentrations used are based on field measurements rather than artificially high levels. Keywords: ecosystem function, microbeads, plastisphere, pollution, polyethylene

Abstract:
Thirty fungi were tested for their degradation ability on low-density polyethylene (LDPE) plastic film. Biodegradation of all fungi was screened in mineral salt medium broth containing LDPE film as the sole carbon source for 30 days. Diaporthe italiana, Thyrostroma jaczewskii, Colletotrichum fructicola, and Stagonosporopsis citrulli were able to colonize and cover the surface of LDPE film in media. The degradation test result was compared to those obtained from Aspergillus niger. LDPE films cocultured with D. italiana, T. jaczewskii, C. fructicola, S. citrulli, A. niger, and control showed weight loss of 43.90%, 46.34%, 48.78%, 45.12%, 28.78%, and 10.85%, respectively. The tensile strength of degraded LDPE films cocultured with D. italiana, T. jaczewskii, C. fructicola, S. citrulli, A. niger, and control also reduced significantly by 1.56 MPa, 1.78 MPa, 0.43 MPa, 1.86 MPa, 3.34 MPa, and 9.98 MPa, respectively. Analysis of LDPE films by Fourier transform infrared spectroscopy and scanning electron microscopy confirmed the biodegradation by the presence of morphological changes such as cracks, scions, and holes on the surface of the film. These fungi have the ability to break down and consume the LDPE film, especially C. fructicola. These findings showed the potential of fungi in Thailand that play an important role in LDPE film degradation. Keywords: plastic biodegradation, LDPE film, fungi, Fourier transform infrared, scanning electron microscopy

Abstract:
Additive Manufacturing (AM) provides promising opportunities to optimize and to produce tooling by integrating near-contour tempering channels for more efficient cooling. To enhance the properties of the produced tooling using additive manufacturing, prototypes should be produced in short periods. Thereby, this requires a small amount of tailored powders, which either has a high production cost or is commercially unavailable. Hence, in this study, an arc spray atomization approach to produce a tailored metal powder at a lower cost and even in small quantities, in comparison to the conventional powder production methods, was proposed. This approach involves converting commercially available metal wire into powder by modifying the wire arc spraying process. The influences of spray medium and gas pressure on the powder properties were investigated. As a result, particles with smooth surface and lower porosity were obtained, when nonoxidizing gases are used for thermal spraying. The particle size decreased with increasing of the gas pressure, and the particles sizes are in the range from 10 to 70 µm, which is desirable for selective laser melting (SLM). A comparison of microstructure and mechanical behavior of SLM generated parts using arc sprayed powders (alloy: X5CrNiCuNb 16-4) and commercial powder (alloy: X5CrNiCuNb 16-4) was also conducted. Keywords: additive manufacturing, arc spraying, powder production, selective laser melting

Abstract:
The natural resistance to biodegradation of polymeric materials prepared from petroleum-based source and the management of their wastes in the environment are the driving forces to replace them by other biodegradable materials from renewable resources. For that, in this work new biocomposites materials have been synthesis from sunflower oil (Helianthus annuus) and alfa plants (Stipatenacissima) as natural based resources. The sunflower oil (SFO) was chemically modified via epoxidation then acrylation reactions to obtain acrylated epoxidized sunflower oil resin (AESFO). The AESFO resin was then copolymerized with styrene as co-monomer in the presence of boron trifluoride (BF3) as cationic initiator and cobalt octoate (Co) as catalyst. The alfa fibers were treated with alkali treatment (5% NaOH) before been used as bio-reinforcement. Biocomposites were prepared by mixing the resin with untreated and treated alfa fibers at different percentages. A biodegradation study was carried out for the synthesized biocomposites in a solid medium (burial in the soil) by evaluated, first, the loss of mass, the results obtained were reached between 7.8% and 11% during one year. Then an observation under an optical microscope was carried out, after one year of burial in the soil, microcracks, brown and black spots were appeared on the samples surface. This results shows that the synthesized biocomposites have a great aptitude for biodegradation. Keywords: alfa fiber, biocomposite, biodegradation, soil, sunflower oil

Abstract:
Because of their effectiveness and environmental safety, many essential oils have been investigated as biopesticides. Nevertheless, the encapsulation process is necessary to improve its physical, chemical, and biological properties. Therefore, the purpose of this paper was to study the physicochemical characteristics, and antifungal activity of the Artemisia Herba-Alba essential oil (HAEO) encapsulated in succinic acid modified β-CD (SACD). A yellowish oil was obtained from plant A. Herba-Alba using hydrodistillation and GC-MS was used to identify the chemical composition, in which α-Thujone (65.0%) was the main component in HAEO. The succinic acid has been esterified via the hydroxyl groups in β-CD to produce SACD. In addition, the inclusion complex formation of HAEO and SACD was generated according to the co-precipitation method and was analyzed by several techniques. The antifungal activity in vitro was examined against Botrytis cinerea by direct contact with a potato dextrose agar culture medium. At a 0.1 % concentration, the HAEO in encapsulated form showed higher potential for the control of B. cinerea when compared to the EO in free form (38.34 to 12%). Thus, these results produced evidence that the encapsulation of EOs in SACD can be useful for the development of B.cinerea inhibitors and a promising alternative biopesticide. Keywords: Artemisia Herba-Alba essential oil, succinic acid modified β-cyclodextrin, inclusion complex, co-precipitation, Botrytis cinerea, direct contact

Abstract:
In this research, enrichment culture using an inorganic liquid medium collected soil contaminated with 1,2-dichlorobenzene (1,2-DCB) in Sendai, Japan, was added 1,2-DCB as the sole carbon source to create a stable consortium. The purpose of this research is to analysis dominant microorganisms in the stable consortium and enzyme system which play a role in the degradation of DCBs. The consortium is now at 30 generation and is still being cultured. By the result of PCR-DGGE and clone library, two bacteria are dominant. The bacteria named sk1 was isolated. 40mg/l of 1,2-DCB and 40mg/l of 1,4-DCB were completely degraded after 32 hours and 50 hours, respectively, but no degradation occurred in the case of 1,3-DCB. By PCR, tecA1 (α-subunit of DCB dioxygenase) gene which plays a role degrading DCB to DCB dihydrodiol, and tecB (dehydrogenase) gene which plays a role degrading DCB dihydrodiol to dichlorocatechol were amplified from strain sk1. Bacteria named sk100 was also isolated. 40mg/l of 1,2-DCB was completely degraded after 32 hours, but no degradation occurred in case of 1,3-DCB and 1,4-DCB. By the result of the catalytic core region of dioxygenase amplified by PCR, gene played a role degrading DCB was analyzed. The results of this study concluded that the isolated strains which have not been reported are able to degrade 1,2-DCB stably, and the characterization of degradation and the genomic analysis which is now in progress is helpful to have an overall view of this microbial degradation. Keywords: DCB, 1, 2-DCB degrading strains, DCB dioxygenase, enrichment culture
