1867 Defective Autophagy Leads to the Resistance to PP2 in ATG5 Knockout Cells Generated by CRISPR-Cas9 Endonuclease
Authors: Sung-Hee Hwang, Michael Lee
Abstract:
Upregulated Src activity has been implicated in a variety of cancers. Thus, Src family tyrosine kinase (SFK) inhibitors are often effective cancer treatments. Here, we investigate the role of autophagy in ATG5 knockout cell lines generated by the Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR)/Cas mediated genome editing. The CRISPR-associated protein Cas9 is an RNA-guided DNA endonuclease that uses RNA–DNA complementarity to identify target sites for sequence specific double-stranded DNA (dsDNA) cleavage. Interestingly, ATG5 KO cells clearly showed a greater proliferation rate than WT NIH 3T3 cells, implying that autophagy induction is cytotoxic. Also, the clonogenic survival of ATG5 KO cells was greater than WT cells. The MTT assay revealed that the cytotoxic effect of PP2 was weaker on ATG5 knockout cells than that WT cells. The conversion of non-autophagic LC3-I to autophagic LC3-II and RT-PCR confirmed the functional gene knockout. Furthermore, Cyto-ID autophagy assay also revealed that PP2 failed to induce autophagy in ATG5 knockout cells. Together, our findings suggest that the resistance to PP2 in ATG5 knockout cells is associated with defective autophagy. Keywords: ATG5 knockout, Autophagy, CRISPR/Cas9, PP2

Authors: R. Odoh, M. S. Dauda, E. A. Kamba, N. C. Igwemmar
Abstract:
Honey and honey trade is an important economic activity for many tropical rural and urban areas worldwide. In West Africa and other part of the world, honey and honey products holds high socio–cultural, religious, medicinal and traditional values. Therefore, to maximize benefits or to enhance profit, a variety of components are added to the raw, fresh and unprocessed honey, introducing the possibility of heavy metals contaminants. Therefore the honey sold in various places, markets and shops in some states in Northern Nigeria (Benue, Nassarawa and Taraba) including Abuja FCT, in Nigeria was analyzed to determine the level of heavy metals (Cd, Co, Cr, Cu, Fe, Mn, Ni, Pb, and Zn). All the honey samples contain heavy metals. The results ranged from 0.028–0.070, 0.023–0.058, 0.042–0.092, 4.231–8.589, 8.115–14.892, 0.078–0.922, 0.044–0.092, 0.041–0.087 and 18.234–28.654 μg/L for Cd, Co, Cr, Cu, Fe, Mn, Ni, Pb and Zn respectively. The mean concentration (μg/L) of the heavy metals Cd, Co, Cr, Cu, Fe, Mn, Ni, Pb, and Zn of the regularly marketed honey is significantly higher than the mean concentration observed in raw, fresh and unprocessed honey. However, continued consumption of honey with high heavy metal content might lead to exposure to chronic heavy metal poisoning. Keywords: honey, health, mineral profile adulteration, contamination
Procedia PDF Downloads 4261864 Review on Green Synthesis of Gold Nanoparticles
Authors: Shabnam, Jagdeep Kumar
Abstract:
Because of the impact of their greater surface area and smaller quantum sizes in comparison with other metal atoms or bulk metals, metal nanoparticles, such as those formed of gold, exhibit a variety of unusual chemical and physical properties. The size- and shape-dependent properties of gold nanoparticles (GNPs) are particularly notable. Metal nanoparticles have received a lot of attention due to their unique properties and exciting prospective uses in photonics, electronics, biological sensing, and imaging. The latest developments in GNP synthesis are discussed in this review. Green chemistry measures were used to assess the production of gold nanoparticles, with a focus on Process Mass Intensity (PMI). Based on these measurements, opportunities for improving synthetic approaches were found. With PMIs that were often in the thousands, solvent usage was found to be the main obstacle for nanoparticle synthesis, even ones that were otherwise considered to be environmentally friendly. Since ligated metal nanoparticles are the most industrially relevant but least environmentally friendly, their synthesis by arrested precipitation was chosen as the best chance for significant advances. Gold nanoparticles of small sizes and bio-stability are produced biochemically, and they are used in many biological applications. Keywords: gold, nanoparticles, green synthesis, AuNP

Authors: Nargiz Ismayilova
Abstract:
In Kamal Abdulla's "The Adventure of Mysteries", the plot develops on two parallel lines. While reading the work, the future looks hazy on the background of the present and the past. It is impossible to predict the end of the work in particular. This can be considered the success of the author. The novel has reflected the features of postmodernism. The novel is characterized by a richness of intertwined plots, themes, meta- submission, device (fiction) typical of postmodern prose technique. The introduction and progress of the work takes the reader to the place, which is an unrecognizable unknown for him but at the same time, its native for him very well. Parts of the novel, divided into chapter techniques, force the reader to distinguish mystical repetitions from the artistic circulation of reality. This makes people think directly. Intertextual communication and the variety of fiction, intelligence, and informativeness determine the perspective of the exemplary reader. As is well known, “postmodern novels, which often use intertextual communication and superstructure techniques, focus on expression rather than on the subject, and benefit from history by combining fiction with historical facts, are able to attract attention with their extraordinary foreign fiction. Keywords: Kamal Abdulla, postmodernism, parallelism, labyrinth, comparison, novel

Authors: Ning Xue, Srinivas Merugu, Ignacio Delgado Martinez, Tao Sun, John Tsang, Shih-Cheng Yen
Abstract:
We have developed a polyimide based neural interface electrode to record nerve signals from the sciatic nerve of a rat. The neural interface electrode has a split-ring shape, with four protruding gold electrodes for recording, and two reference gold electrodes around the split-ring. The split-ring electrode can be opened up to encircle the sciatic nerve. The four electrodes can be bent to sit on top of the nerve and hold the device in position, while the split-ring frame remains flat. In comparison, while traditional cuff electrodes can only fit certain sizes of the nerve, the developed device can fit a variety of rat sciatic nerve dimensions from 0.6 mm to 1.0 mm, and adapt to the chronic changes in the nerve as the electrode tips are bendable. The electrochemical impedance spectroscopy measurement was conducted. The gold electrode impedance is on the order of 10 kΩ, showing excellent charge injection capacity to record neural signals. Keywords: impedance, neural interface, split-ring electrode, neural signal recording

Procedia PDF Downloads 1921852 A Deep Learning Based Approach for Dynamically Selecting Pre-processing Technique for Images
Authors: Revoti Prasad Bora, Nikita Katyal, Saurabh Yadav
Abstract:
Pre-processing plays an important role in various image processing applications. Most of the time due to the similar nature of images, a particular pre-processing or a set of pre-processing steps are sufficient to produce the desired results. However, in the education domain, there is a wide variety of images in various aspects like images with line-based diagrams, chemical formulas, mathematical equations, etc. Hence a single pre-processing or a set of pre-processing steps may not yield good results. Therefore, a Deep Learning based approach for dynamically selecting a relevant pre-processing technique for each image is proposed. The proposed method works as a classifier to detect hidden patterns in the images and predicts the relevant pre-processing technique needed for the image. This approach experimented for an image similarity matching problem but it can be adapted to other use cases too. Experimental results showed significant improvement in average similarity ranking with the proposed method as opposed to static pre-processing techniques. Keywords: deep-learning, classification, pre-processing, computer vision, image processing, educational data mining

Authors: Maamar Fatouma
Abstract:
Mirror optical performance can experience varying levels of stress and tolerances, which can have a notable impact on optical parametric systems. to ensure proper optical figure and position of mirror mounting within design tolerances, it is crucial to have a robust support structure in place for optical systems. The optical figure tolerance determines the allowable deviation from the ideal form of the mirror and the position tolerance determines the location and orientations of the optical axis of the optical systems. A variety of factors influence the optical figure of the mirror. Included are self-weight (Deflection), excitation from temperature change, temperature gradients and dimensional instability. This study employs an analytical approach and finite element method to examine the effects of stress resulting from mirror mounting on the wavefront passing through the mirror. The combined effect of tolerance and deflection on mirror performance is represented by an error budget. Numerical mirror mounting is presented to illustrate the space application of performance techniques. Keywords: opto-mechanical, bonded optic, tolerance, self-weight distortion, Rayleigh criteria

Authors: Delele Worku Ayele, Bing-Joe Hwang
Abstract:
A method that does not employ hot injection techniques has been developed for the size-tunable synthesis of high-quality CdSe quantum dots (QDs) with a zinc blende structure. In this environmentally benign synthetic route, which uses relatively less toxic precursors, solvents, and capping ligands, CdSe QDs that absorb visible light are obtained. The size of the as-prepared CdSe QDs and, thus, their optical properties can be manipulated by changing the microwave reaction conditions. The QDs are characterized by XRD, TEM, UV-vis, FTIR, time-resolved fluorescence spectroscopy, and fluorescence spectrophotometry. In this approach, the reaction is conducted in open air and at a much lower temperature than in hot injection techniques. The use of microwaves in this process allows for a highly reproducible and effective synthesis protocol that is fully adaptable for mass production and can be easily employed to synthesize a variety of semiconductor QDs with the desired properties. The possible application of the as-prepared CdSe QDs has been also assessed using deposition on TiO2 films. Keywords: average life time, CdSe QDs, microwave (MW), mass production oleic acid, Na2SeSO3

Abstract:
This paper presents a classification study on yield prediction in semiconductor production using machine learning approaches. A complicated semiconductor production process is generally monitored continuously by signals acquired from sensors and measurement sites. A monitoring system contains a variety of signals, all of which contain useful information, irrelevant information, and noise. In the case of each signal being considered a feature, "Feature Selection" is used to find the most relevant signals. The open-source UCI SECOM Dataset provides 1567 such samples, out of which 104 fail in quality assurance. Feature extraction and selection are performed on the dataset, and useful signals were considered for further study. Afterward, common machine learning algorithms were employed to predict whether the signal yields pass or fail. The most relevant algorithm is selected for prediction based on the accuracy and loss of the ML model. Keywords: deep learning, feature extraction, feature selection, machine learning classification algorithms, semiconductor production monitoring, signal processing, time-series analysis

Abstract:
Variety of different factors could influence on the measure urban perception. Both physical and non-physical factors, at least, make the quality of perception through the urban spaces. The value of lighting is one of the important factors which could make the better quality of environmental perception for the user. The perception of urban space in most of the Iranian cities is offer by different factors during the night time which caused to the death of nightlife and social activities. Therefore, this research is an attempt to study on the different of user perception during day and night in the Koohsangi Street. As the case study area in Iran in order to bring out the main influential factors during perception process. To deal with this good we used chi-square test on a sample size made up of on hundred participants. The result shows that for improving the night quality of urban spaces the legibility, navigation, and role stimulation were in important perception factors. Therefore, by focusing on these factors it would be possible to find out more functional solution for improving the activity of night perception. Keywords: perception, urban space, legibility, imageability, nightscape

Authors: Miroslav Popelka, Jakub Nozicka
Abstract:
The whole work is based on possibility to use Lego Mindstorms robotics systems to reduce costs. Lego Mindstorms consists of a wide variety of hardware components necessary to simulate, programme and test of robotics systems in practice. To programme algorithm, which simulates space using the ultrasonic sensor, was used development environment supplied with kit. Software Matlab was used to render values afterwards they were measured by ultrasonic sensor. The algorithm created for this paper uses theoretical knowledge from area of signal processing. Data being processed by algorithm are collected by ultrasonic sensor that scans 2D space in front of it. Ultrasonic sensor is placed on moving arm of robot which provides horizontal moving of sensor. Vertical movement of sensor is provided by wheel drive. The robot follows map in order to get correct positioning of measured data. Based on discovered facts it is possible to consider Lego Mindstorm for low-cost and capable kit for real-time modelling. Keywords: LEGO Mindstorms, ultrasonic sensor, real-time modeling, 2D object, low-cost robotics systems, sensors, Matlab, EV3 Home Edition Software
