1157 The Qualitative and Quantitative Detection of Pistachio in Processed Food Products Using Florescence Dye Based PCR

Abstract:
Pistachio nuts, the fruits of the pistachio tree (Pistacia vera), are edible tree nuts highly valued for their organoleptic properties. Pistachio nuts used in snack foods, chocolates, baklava, meat products, ice-cream industries and other gourmet products as ingredients. Undeclared pistachios may be present in food products as a consequence of fraudulent substitution. Control of food samples is very important for safety and fraud. Mix of pistachio, peanut (Arachis hypogaea), pea (Pisum sativum L.) used instead of pistachio in food products, because pistachio is a considerably expensive nut. To solve this problem, a sensitive polymerase chain reaction PCR has been developed. A real-time PCR assay for the detection of pea, peanut and pistachio in baklava was designed by using EvaGreen fluorescence dye. Primers were selected from powerful regions for identification of pea, peanut and pistachio. DNA from reference samples and industrial products were successfully extracted with the GIDAGEN® Multi-Fast DNA Isolation Kit. Genomes were identified based on their specific melting peaks (Mp) which are 77°C, 85.5°C and 82.5°C for pea, peanut and pistachio, respectively. Homogenized mixtures of raw pistachio, pea and peanut were prepared with the ratio of 0.01%, 0.1%, 1%, 10%, 40% and 70% of pistachio. Quantitative detection limit of assay was 0.1% for pistachio. Also, real-time PCR technique used in this study allowed the qualitative detection of as little as 0.001% level of peanut DNA, 0,000001% level of pistachio DNA and 0.000001% level of pea DNA in the experimental admixtures. This assay represents a potentially valuable diagnostic method for detection of nut species adulterated with pistachio as well as for highly specific and relatively rapid detection of small amounts of pistachio in food samples.Keywords: pea, peanut, pistachio, real-time PCR

Abstract:
Salinity is one of the most environmental factors limiting crop productivity. It has a negative effect on both germination and plant growth processes (photosynthesis, respiration, and transpiration), nutrient balance, membrane properties and cellular homeostasis, enzymatic and metabolic activities. Among the strategic crops in the world and more mainly in Algeria, durum wheat is very affected by this abiotic stress. For that, this study focuses on an evaluation of salt stress effect on the germination process of durum wheat as well as its response after application of lavender hydrosol and aqueous pistachio extract. The results have shown that all the physicochemical parameters of germination have been affected by this stress. However, lavender hydrosol and aqueous pistachio extract, considered as organic compounds, significantly improved the germination of wheat seeds. Finally, this study has highlighted the importance of using organic products as an ideal alternative to reduce the effect of abiotic stress on durum wheat productivity.Keywords: salinity, wheat durum, extract, lavender hydrosol, aqueous pistachio

Abstract:
In this study, the effect of various drying methods (sun drying, shade drying and industrial drying) on final moisture content, shell splitting degree, shrinkage and color change were studied. Sun drying resulted higher degree of pistachio nuts shell splitting on pistachio nuts relative other drying methods. The ANOVA results showed that the different drying methods did not significantly effects on color change of dried pistachio nut. The results illustrated that pistachio nut dried by industrial drying had the lowest moisture content. After the end of drying process, initially, the experimental drying data were fitted with five famous drying models namely Newton, Page, Silva et al., Peleg and Henderson and Pabis. The results indicated that Peleg and Page models gave better results compared with other models to monitor the moisture ratio’s pistachio nut in industrial drying and open sun (or shade drying) methods, respectively.Keywords: industrial drying, pistachio, quality properties, traditional drying

Abstract:
Turkey is the third largest producer of pistachio (Pistacia vera L.) after Iran and United States. Harvested pistachio nuts are covered with organic hull which is removed by de-hulling process. Most of the pistachio by-products which are produced during de-hulling process are considered as agricultural waste and often mixed with soil, to a lesser extent are used as feedstuff by local livestock farmers and a small portion is used as herbal medicine. Due to its high organic and phenolic content as well as high solids concentration, pistachio processing wastes create significant waste management problems unless they are properly managed. However, there is not a well-established waste management method compensating the waste generated during the processing of pistachios. This study investigated the anaerobic treatability and biogas generation potential of pistachio hull waste. The effect of pre-treatment on biogas generation potential was investigated. For this purpose, Biochemical Methane Potential (BMP) Assays were conducted for two Chemical Oxygen Demand (COD) concentrations of 22 and 33 g tCOD l-1 at the absence and presence of chemical and thermal pre-treatment methods. The results revealed anaerobic digestion of the pistachio de-hulling wastes and subsequent biogas production as a renewable energy source are possible. The observed percent COD removal and methane yield values of the pre-treated pistachio de-hulling waste samples were significantly higher than the raw pistachio de-hulling waste. Abstract:
The common pistachio psylla, Agonoscena pistaciae Burckhardt and Lauterer (Hemiptera: Aphalaridae), as one of the most detrimental pests in all pistachio producing regions, causes great economic damages to pistachio trees. Nowadays, various pesticides are used to control the common pistachio psylla and robust pesticide exposure has occurred in orchards. In this study, field experiments were conducted during 2018–2021 to assess the effects of sulfur on A. pistaciae. This study compared sulfur with asafoetida extract and pesticide (acetamiprid) on A. pistaciae based on complete randomized blocks with three replications. The analysis results of variance showed that the effect of treatments on egg (F2,24 = 17.61, P = 0.00) and nymphs (F2,24 = 18.29, P = 0.00) had a significant difference at a 1% level. The results demonstrated that sulfur had the highest measure of control on eggs and nymphs significantly compared to the plant extract and pesticide (negative control). These results provide support to the potential use of sulfur as an alternative pest management tool against A. pistaciae. The results clearly indicated that sulfur could control the common pistachio psylla population for six weeks at least.Keywords: Agonoscena pistaciae, pesticide exposure, pistachio, sulfur

Governments Policies play a major role in national and international Markets. Pistachio is one of the most important non-oil export commodity of Iran. Therefore, in this study the relation between the producer support policies and the export of Pistachio was examined. An econometric model (VAR) was applied to test the study hypothesis. According to the estimated coefficient in VAR model, lag of producer support index has a significant and negative effect on variation of Pistachio’s export in short term. The introduction of real pistachio was made in the early sixties by an FAO program in Algeria in several regions in the northern part of Algeria: Tlemcen, Sidi Bel Abbes, Batna, Bouira M'sila . Chemical analyzes of seeds pistachios were made on seeds from an orchard that localizes to Bechloul (Bouira) located in bioclimatic sub-humid temperate winter floor. Analyzes reveal dry matter content of 3.60 ± 0.45%, the water rate is 7.21 ± 0.36%. However, the fat content is 46.00 ± 0.90%, in average blood sugar, it is 4.02 ± 0.47%, the protein reached 29.88 ± 0.76%. Given the very interesting that high-fat food nutritional values, culture pistachio must be considered for its extension in Algeria.Keywords: pistachio, dry matter, fat, sugar, protein

Abstract:
Parkinson’s disease (PD) is a common neurological disorder characterized by motor deficits and loss of dopaminergic neurons. Oxidative stress is said to play a pivotal role in the pathophysiology of the disease. In the present study, PD was induced by injection of rotenone (1.5 mg/kg/day, s.c.) for eight days. Pistachio (800 mg/kg/day, p.o.) was given for two weeks. At the end of treatment brains were dissected out and striatum was isolated for biochemical and neurochemical analysis. Morris water maze (MWM) test and novel object recognition (NOR) task was used to test the memory function while motor behavior was determined by open field test (OFT), Kondziela inverted screen test (KIST), pole test (PT), beam walking test (BWT), inclined plane test (IPT) and footprint (FP) test. Several dietary components have been evaluated as potential therapeutic compounds in many neurodegenerative diseases. Increasing evidence shows that nuts have protective effects against various diseases by improving the oxidative status and reducing lipid peroxidation. Pistachio is the only nut that contains anthocyanin, a potent antioxidant having neuroprotective properties. Results showed that pistachio supplementation significantly restored the rotenone-induced motor deficits and improved the memory performance. Moreover, rats treated with pistachio also exhibited enhanced oxidative status and increased dopamine (DA) and 5-hydroxytryptamine (5-HT) concentration in striatum. In conclusion, to our best knowledge, we have for the first time shown that pistachio nut possesses neuroprotective effects against rotenone-induced motor and cognitive deficits. These beneficial effects of pistachio may be attributed to its high content of natural antioxidant and phenolic compounds. Hence, consumption of pistachio regularly as part of a daily diet can be beneficial in the prevention and treatment of PD.Keywords: rotenone, pistachio, oxidative stress, Parkinson’s disease

The pistachio fruit is a strategic product in Iran. One of the problems caused the reduction of pistachio proceeds is related to biennial- bearing or alternative bearing. Biennial- bearing is very important and is happened because of the fallen female bloom buds in vintage year. This test was done according to random blocks of 6 orchards in the type of Ahmad Aghaie with 4 iterations. Vegetative properties of branch are investigated. The results are shown that if the bunch numbers are increased, the possibility of falling is increased in bloom buds. The effect of two different transplanting date (March and November) in five pistachio cultivars ('Ahmad Aghaei', 'Akbri', Momtaz', 'Ghazvini', and 'Ohadi') were evaluated in Rafsanjan Azad University, Iran. Seeds were planted in plastic bags in March 2012. The seedlings were transplanted to a field in November 2012 and March 2013. Vegetative growing factors such as plant length, stem diameter, number of leaves and fresh and dry weight were measured in date. The results obtained from collected data indicated that no significant differences were found for the traits of interest. Proper management of these symbiosis causes to reduce the usage of chemical fertilizers and absorb nutrition especially phosphor. Pistacia vera is endemic in Iran and is one of the most important products for this country. In recent years, food allergies which cause serious health problems affect to public health around the world. Foodstuffs which contain allergens are either intentionally used as ingredients or are encased as contaminant in food products. The prevalence of clinical allergy to peanuts and nuts is estimated at about 0.4%-1.1% of the adult population, representing the allergy to pistachio the 7% of the cases of tree nut causing allergic reactions. In order to protect public health and enforce the legislation, methods for sensitive analysis of pistachio and peanut contents in food are required. Pea, pistachio and peanut are used together, to reduce the cost in food production such as baklava, snack foods.DNA technology-based methods in food analysis are well-established and well-roundedtools for species differentiation, allergen detection. Especially, the probe-based TaqMan real-time PCR assay can amplify target DNA with efficiency, specificity, and sensitivity.In this study, pistachio, peanut and pea were finely ground and three separate series of triplet mixtures containing 0.1, 1, 10, 100, 1000, 10,000 and 100,000 mg kg-1 of each sample were prepared for each series, to a final weight of 100 g. DNA from reference samples and industrial products was successfully extracted with the GIDAGEN® Multi-Fast DNA Isolation Kit. TaqMan probes were designed for triplex determination of ITS, Ara h 3 and pea lectin genes which are specific regions for identification pistachio, peanut and pea, respectively.The real-time PCR as quantitative detected pistachio, peanut and pea in these mixtures down to the lowest investigated level of 0.1, 0.1 and 1 mg kg-1, respectively. Also, the methods reported here are capable of detecting of as little as 0.001% level of peanut DNA, 0,000001% level of pistachio DNA and 0.000001% level of pea DNA. Abstract:
Storage pests with quantitative, qualitative, and hygienic losses to storage products lead to heavy damage to these products. One of the best methods of controlling storage pests is microwave heating, which is an environmentally friendly method and can be used to replace chemical methods to control storage pests. Pistachios and almonds are the most important dried fruit items in Iran, which account for a significant part of Iran's exports every year. In this study, which along with Pistachio and almond samples were exposed to microwave radiation at 320, 720, 900 watts with times of 10, 20, 30 seconds. Qualitative evaluation of product changes due to the above treatments was performed in the form of changes in colorimetric factors and organoleptic properties of the product. The results showed that in microwave treatment, power, and time factors had a significant effect on the taste and overall acceptance of pistachio product, polymer and power interaction, polymer and time, time and power had no significant effect on pistachio product quality. In almond products, the factors of polymer, time, power, interaction of polymer and power, polymer and time, and power had no significant effect on almond quality.Keywords: microwave, qualitative, pistachio, almond

Pistachio is like almond but the second cycle of growth (third phase) has rather fast growth. This is caused to add final mass of product. When the germ grows, it and its cover are reached to the final size during six week period. As starting the second phase, the lignifications of pericarp is begun and continued for 4 or 6 weeks. Physiological maturity or easy separation of green from scutum is specified. This test was done according to random blocks of 6 orchards in the type of Ahmad Aghaie with 4 iterations. Vegetative properties of branch are investigated. The results of the bunch numbers on the growth of branch in current year are shown that the most growth of branch is happened by trimming of one and two bunches of the branch and the most diameter of the branch is happened by trimming of one to four bunches of branch. Abstract:
Water quality is the critical factor that influence on human health and quantity and quality of grain production in semi-humid and semi-arid area. Pistachio is a main crop that accounts for a considerable portion of Iranian agricultural exports. Give that pistachio tree is a tolerant type of tree to saline and alkaline soil and water conditions, but groundwater and irrigation water quality play important roles in main production this crop. For this purpose, 94 well water samples were taken from 25 wells and samples were analyzed. The results showed give that region’s geological, climatic characteristics, statistical analysis, and based on dominant cations and anions in well water samples (piper diagram); four main types of water were found: Na-Cl, K-Cl, Na-SO4, and K-SO4. It seems that most wells in terms of water quality (salinity and alkalinity) and based on Wilcox diagram have critical status. The analysis suggested that more than eighty-seven percentage of the well water samples have high values of EC that these values are higher than into critical limit EC value for irrigation water, which may be due to the sandy soils in this area. Most groundwater were relatively unsuitable for irrigation but it could be used by application of correct management such as removing and reducing the ion concentrations of Cl‾, SO42‾, Na+ and total hardness in groundwater and also the concentrated deep groundwater was required treatment to reduce the salinity and sodium hazard. Given that irrigation water quality in this area was relatively unsuitable for most agriculture production but pistachio tree was adapted to this area conditions. The integrated management of groundwater for irrigation is the way to solve water quality issues not only in Rafsanjan area, but also in other arid and semi-arid areas.Keywords: groundwater quality, irrigation water quality, salinity, alkalinity, Rafsanjan plain, pistachio

Abstract:
This study investigated how to reduce bio-compounds and amino acids in V. amygdalina leaf during processing as a functional food ingredient. Fresh V. amygdalina leaf was processed using thermal oil-aqueous mixtures (soybean oil: aqueous and palm oil: aqueous) at 1:40 and 130 (v/v), respectively. Results indicated that the hot soybean oil-aqueous mixture was the most effective in preserving the bio-compounds and amino acids with retention potentials of 80.95% of the bio-compounds at the rate of 90-100%. Hot palm oil-aqueous mixture retained 61.90% of the bio-compounds at the rate of 90-100% and hot aqueous retained 9.52% of the bio-compounds at the same rate. During the debittering process, seven new bio-compounds were formed in the leaves treated with hot soybean oil-aqueous mixture, six in palm oil-aqueous mixture, and only four in hot aqueous leaves. The bio-compounds in the treated leaves have potential functions as antitumor, antioxidants, antihistaminic, anti-ovarian cancer, anti-inflammatory, antiarthritic, hepatoprotective, antihistaminic, haemolytic 5-α reductase inhibitor, nt, immune-stimulant, diuretic, antiandrogenic, and anaemiagenic. Alkaloids and polyphenols were retained at the rate of 81.34-98.50% using oil: aqueous mixture while aqueous recorded the rate of 33.47-41.46%. Most of the essential amino acids were retained at a rate above 90% through the aid of oil. Public concerns on Extremely Low Frequency (ELF) Electromagnetic Field (EMF) exposure have been elongated since the last few decades. Electrical substations and high tension rooms (HT room) in commercial buildings were among the contributing factors emanating ELF magnetic fields. This paper discussed various shielding methods conventionally used in mitigating the ELF exposure. Nevertheless, the standard methods were found to be impractical and incapable of meeting currents shielding demands. In response to that, remarkable researches were conducted in effort to invent novel methods which is more convenient and efficient such as magnetic aqueous shielding or paint, textiles and papers shielding. A mitigation method using magnetic aqueous substrate in shielding application was proposed in this paper for further investigation. using Manganese Zinc Ferrite (Mn0.4Zn0.6Fe2O4). Abstract:
Efficient extraction for separation of V and Cr in the leaching/aqueous solution is essential to the reuse of V and Cr in the V-Cr slag. Trioctylamine, a common tertiary amine extractant, with some good characters (e.g., weak base, insoluble in water and good stability) different from N1923, was investigated in this paper. The separation factor of Cr and V can be reached to 230.71 when initial pH of the aqueous solution is 0.5, so trioctylamine can be used for extracting Cr from the leaching/aqueous solution contained V and Cr. The highest extraction percentages of Cr and V were 98.73% and 90.22% when the initial pH values were 0.5 and 1.5, respectively. Abstract:
Aqueous peroxotitanium acid solution was prepared by the reaction between H₂O₂ solution and TiO₂ fluorinated using F₂ gas. The coating of TiO₂/SiO₂ multilayer on the surface of polycarbonate (PC) resin was carried out step by step using the TEOS solution and aqueous peroxotitanium acid solution. We confirmed each formation of SiO₂ and TiO₂ layer by scanning electron microscopy and energy-dispersive X-ray spectroscopy, and x-ray photoelectron spectroscopy results. The formation of a TiO₂ thin layer on SiO₂ coated on polycarbonate (PC) was carried out at 120 ℃ and for 15 min ~ 3 h with aqueous peroxotitanium acid solution using a hydrothermal synthesis autoclave reactor. The morphology TiO₂ coating layer largely depended on the reaction time, as shown in the results of SEM-EDS analysis. Increasing the reaction times, the TiO₂ layer expanded uniformly. Moreover, the surface fluorination of the SiO₂ layer can promote the formation of the TiO₂ layer on the surface.Keywords: aqueous peroxotitanium acid solution, photocatalytic activity, polycarbonate, surface fluorination

Abstract:
Glycerol is a byproduct of biodiesel production that can be used for aqueous-phase reforming to obtain hydrogen. Iridium is a material that has high activity and hydrogen selectivity for steam phase reforming. Nevertheless, a drawback for the use of iridium in aqueous-phase reforming is the low activity in water-gas shift reaction. Therefore, in this work, it is proposed the use of nickel and copper as a second metal in the catalyst to reach a synergetic effect. Iridium, iridium-nickel and iridium-copper catalysts were prepared by incipient wetness impregnation and evaluated in the aqueous-phase reforming of glycerol using CeO₂ or La₂O₃ as support. The catalysts were characterized by XRD, XPS, and EDX. The reactions were carried out in a fixed bed reactor feeding a solution of glycerol 10 wt% in water at 270°C, and reaction products were analyzed by gas chromatography. It was found that IrNi/CeO₂ reached highest glycerol conversion and hydrogen production, slightly above 70% and 43 vol% respectively. In terms of conversion, iridium is a promising metal, and its activity for hydrogen production can be enhanced when adding a second metal.Keywords: aqueous-phase reforming, glycerol, hydrogen production, iridium

Abstract:
Acetic acid is one of the simplest and most widely used carboxylic acids having many important chemical and industrial applications. Total worldwide production of acetic acid is about 6.5 million tonnes per year. A great deal of efforts has been made in developing feasible and economic method for recovery of carboxylic acids. Among them, Liquid-liquid extraction using aqueous two-phase systems (ATPS) has been demonstrated to be a highly efficient separation technique. The study of efficiently separating and recovering Acetic acid from aqueous solutions is an important significance on industry and environmentally sustainable development. Many research groups in different countries are working in this field and some methods are proposed in the literature. In this work, effect of sodium chloride with different content (5%, 10% and 20%) on the liquid-liquid equilibrium data of (water+ acetic acid+ DCM) system is investigated. The addition of the salt in an aqueous solution introduces ionic forces which affect liquid-liquid equilibrium and which influence directly the distribution coefficient of the solute. From the experimental results, it can be concluded that when the percentage of salt increases in the aqueous solution, the equilibrium between phases is modified in favor of the extracted phase.Keywords: acetic acid recovery, aqueous solution, salting-effect, sodium chloride

Abstract:
The refractometric method has been used to determine optical properties of concentration features of aqueous solutions of ethanol, tetrahydrofuran and dimethylformamide at the room temperature. Changes in dielectric permittivity of aqueous solutions of ethanol, tetrahydrofuran and dimethylformamide in a wide range of concentrations (0÷1.0 molar fraction) have been studied using molecular dynamics method. The curves depending on the concentration of experimental data on excess refractive indices and excess dielectric permittivity were compared. It has been shown that stable heteromolecular complexes in binary solutions are formed in the concentration range of 0.3÷0.4 mole fractions. The real and complex part of dielectric permittivity was obtained from dipole-dipole autocorrelation functions of molecules. At the concentrations of C = 0.3 / 0.4 m.f. the heteromolecular structures with hydrogen bonds are formed. This is confirmed by the extremum values of excessive dielectric permittivity and excessive refractive index of aqueous solutions.Keywords: refractometric method, aqueous solution, molecular dynamics, dielectric constant

Acute toxicity test (LD50, oral, rat) of the extract showed no mortality at up to 5000 mg/kg and the animals were found active and healthy. Abstract:
Hexadecyltrimethylammonium bromide (HTAB) modified nano silica were used as pore stabilizer for the preparation of interconnected macroporous copolymer foam of glycidyl methacrylate (GMA), divinylbenzene (DVB) and tert-butyl acrylate (BA). The polymerization of air infused aqueous foam is carried out through free radical thermal initiator. The porosity of the polymerized foam depends on the concentration of HTAB used to control the hydrophobic and hydrophilic behavior of silica nanoparticle. Modified silica particle results to form closed cell foam with 74% of porosity for 60% of air infusion during aqueous foaming. The preliminary structure of microfoam was observed through optical microscopy, whereas for a better understanding of morphology SEM was used. The proposed route is an eco-friendly route for synthesizing polymeric microporous polymer as compared to other chemical and additive-based routes available.Keywords: air-infused, interconnected microporous, porosity, aqueous foam

Abstract:
Fishbone is a type of waste generated from food and food processing industries. Fishbone wastes are usually treated as the source of organic matter for the by-production. It is a rich source of hydroxyapatite (HAP). In this study, the adsorption behavior of fishbone was examined in a batch system as an economically viable adsorbent for the removal of Ni⁺² ions from aqueous solution. The powdered fishbone was characterized using Fourier Transform Infrared (FT-IR) spectrophotometer and Scanning Electron microscope (SEM). The study investigated the influence of adsorbent dosage, solution pH, contact time, and initial metal concentration on the removal of Nickel (II) ions at room temperature. The batch kinetics study showed that the optimum adsorption of Ni(II) was 98% at pH 7, metal ion concentration of 30 mg/L. The results obtained from the experimental work showed that fishbone can be used as an adsorbent for the removal of Ni(II) ions from aqueous solution.Keywords: adsorption, aqueous media, fishbone, kinetic study

Abstract:
A nanocrystalline thin film of ZnSe was successfully electrodeposited on copper substrate using a non-aqueous solution and subsequently annealed in air at 400°C. XRD analysis indicates the polycrystalline deposit of (111) plane in both the cases. The sharpness of the peak increases due to annealing of the film and average grain size increases to 20 nm to 27nm. SEM photograph indicate that grains are uniform and densely distributed over the surface. Due to annealing the average grain size increased by 20%. The EDS spectroscopy shows the ratio of Zn & Se is 1.1 in case of annealed film. AFM analysis indicates the average roughness of the film reduces from 181nm to 165nm due to annealing of the film. The liquid-liquid extraction of copper (II) from aqueous solution by capric acid (HL) in chloroform at 25°C has been studied. The ionic strength effect of the aqueous phase shows that the extraction of copper(II) increases with the increase in ionic strength. with different ionic strengths 1, 0.5, 0.25, 0.125 and 0.1M in the aqueous phase.
