1287 Impact of Herbicides on Soil Biology in Rapeseed
Authors: M. Eickermann, M. K. Class, J. Junk
Abstract:
Winter oilseed rape, Brassica napus L., is characterized by a high number of herbicide applications. Therefore, its cultivation can lead to massive contamination of ground water and soil by herbicide and their metabolites. A multi-side long-term field experiment (EFFO, Efficient crop rotation) was set-up in Luxembourg to quantify these effects. Based on soil sampling and laboratory analysis, preliminary results showed reduced dehydrogenase activities of several soil organisms due to herbicide treatments. This effect is highly depending on the soil type. Relation between the dehydrogenase activity and the amount of microbial carbon showed higher variability on the test side with loamy Brown Earth, based on Bunter than on those with sandy-loamy Brown Earth, based on calciferous Sandstone. Keywords: Cropping system, dehydrogenase activity, herbicides, mechanical weed control, oilseed rape.

Authors: Shuvasish Roy Choudhury, Rita Mahanta, Aparajita Borkotoki
Abstract:
Fishes, in general, follow ammonotelic mode of excretion. However, certain stress factors may provoke them to excrete urea. In the present study, the possible role of ureogenesis to avoid accumulation of toxic ammonia under water-restricted condition was tested in Heteropneustes fossilis. A total of hundred fishes were collected and sacrificed. Excretory urea and ammonia were estimated in the water of the aquarium and glutamate dehydrogenase acitivity was measured in the hepatic tissue. During the experimental period, excretory ammonia in Heteropneustes fossilis was found between 931% to 16% above the baseline ammonia and excretory urea was found between 112% to 898% above the baseline urea. A high degree of correlation with r (coefficient of correlation) above 0.9 is observed between excretory ammonia and urea in Heteropneustes fossilis. However, only a moderate degree of correlation is observed between the activity of glutamate dehydrogenase and excretory ammonia and urea.

Authors: Y. Hong, H. Y. Hu, A. Sakoda, M. Sagehashi
Abstract:
In this study, inhibition of Microcystis aeruginosa by antialgal alleochemical gramine, was studied by analyzing algal metabolic activity (represented by esterase and total dehydrogenase activities) and cell ultrastructure (showing morphological and ultrastructure alterations using transmission electron microscopy and DNA ladder analysis). After gramine exposure, esterase and total dehydrogenase activities were increased firstly but decreased later. In contrast with the controls, the cells exposed to gramine showed apparent ultrastructure alterations with thylakoids in breakage, phycobilins in decrease, lipid and cyanophycin granules abundant firstly but dissolved afterwards, DNA in fragementation. The occurrence of increase of metabolic activity and specific granules reflected that the resistance of cellular response to gramine was initiated. DNA fragementation associated with the increase of metabolic activity and specific granules hinted that gramine caused M. aeruginosa cells to initiate some morphotype of programmed cell death. Keywords: Allelochemical, gramine, metabolic activity, Microcystis aeruginosa, ultrastructure.

Abstract:
Malate dehydrogenase-glutamate oxaloacetate aminotransferase (MDh-GOAT) enzyme complex (the EC) was isolated and purified from wheat and rise, their some main physicchemical properties were studied. Michael-s constants of the EC MDh-GOAT to malate, glutamate and NAD were investigated. This kinetic results show a high relationship to glutamate. Taking into account important role of the the EC in catabolism of glutamate – the central amino acid of a nitric exchange, there is a sharp necessity of deeper studying of this enzyme complex. Therefore the basic purpose of the work is studying the basic physical and chemical properties of this enzyme complex discovered by us, which would be very important for understanding the mechanisms of reaction catalyzed by the EC.Keywords: Malate dehydrogenase-glutamate oxaloacetate aminotransferase, enzyme complex, glutamate.

Abstract:
Forty-five dairy cows were used to compare the enzyme activity of alkaline phosphatase (ALP), lactate dehydrogenase (LDH), α -amylase in the cervical mucus of cows during spontaneous and induced estrus using progestagen or PGF2 α and to determine whether these enzymes affect the fertility in cows with induced estrus, at the time of Al. The animals were assigned to 3 groups (no treatment, a Crestar® for 12 days, a double im injection of PGF2 α). The cows were artificially inseminated (AI). Cervical mucus samples were collected from all cows 3 to 5 min before the AI. The results are summarized as follows: ALP and α -amylase activity for spontaneous estrus were similar to those for induced estrus (P>0.05) . LDH activity levels during spontaneous and PGF2 α induced estrus was significantly lower (P < 0.001) than that in progestagene induced estrus groups. While no difference was found between the first and the third groups. Our result showed a significant difference in LDH activity levels between cows conceived with 2 or more AI and those conceived with 1 AI. The result of this study showed that the enzyme activity in cervical mucus is helpful for detection of ovulation and time of AI.Keywords: cervical mucus, dairy cow, enzyme, induced, estrus, ovulation, AI

Abstract:
The kinetic properties of enzymes are often reported using the apparent KM and Vmax appropriate to the standard Michaelis-Menten enzyme. However, this model is inappropriate to enzymes that have more than one substrate or where the rate expression does not apply for other reasons. Consequently, it is desirable to have a means of estimating the appropriate kinetic parameters from the apparent values of KM and Vmax reported for each substrate. We provide a means of estimating the range within which the parameters should lie and apply the method to data for glutamate dehydrogenase from the nematode parasite of sheep Teladorsagia circumcincta. Keywords: enzyme kinetics, glutamate dehydrogenase, intervalanalysis, parameter estimation.

Authors: N. Aliasgharzad, A. Molaei, S. Oustan
Abstract:
Soil microbial activity is adversely affected by pollutants such as heavy metals, antibiotics and pesticides. Organic amendments including sewage sludge, municipal compost and vermicompost are recently used to improve soil structure and fertility. But, these materials contain heavy metals including Pb, Cd, Zn, Ni and Cu that are toxic to soil microorganisms and may lead to occurrence of more tolerant microbes. Among these, Pb is the most abundant and has more negative effect on soil microbial ecology. In this study, Pb levels of 0, 100, 200, 300, 400 and 500 mg Pb [as Pb(NO3)2] per kg soil were added to the pots containing 2 kg of a loamy soil and incubated for 6 months at 25°C with soil moisture of - 0.3 MPa. Dehydrogenase activity of soil as a measure of microbial activity was determined on 15, 30, 90 and 180 days after incubation. Triphenyl tetrazolium chloride (TTC) was used as an electron acceptor in this assay. PICTs (IC50 values) were calculated for each Pb level and incubation time. Soil microbial activity was decreased by increasing Pb level during 30 days of incubation but the induced tolerance appeared on day 90 and thereafter. During 90 to 180 days of incubation, the PICT was gradually developed by increasing Pb level up to 200 mg kg-1, but the rate of enhancement was steeper at higher concentrations.

Abstract:
This study was conducted to examine the possible role of serum Gamma-glutamyltransferase (GGT) and Lactate dehydrogenase (LDH) in the prediction of severity of preeclampsia. The study group comprised of 40 preeclamptic cases (22 with mild and 18 with severe) and 40 healthy normotensive pregnant controls. Serum samples of all the cases were assayed for GGT and LDH. Demographic, hemodynamic and laboratory data as well as serum GGT and LDH levels were compared among the three groups.
The results indicated that severe preeclamptic cases had significantly increased levels of serum GGT and LDH. The symptoms in severe preeclamptic women were significantly increased in patients with GGT > 70 IU/L and LDH >800 IU/L. Elevated levels of serum GGT and LDH can be used as biochemical markers which reflects the severity of preeclampsia and useful for the management of preeclampsia to decrease maternal and fetal morbidity and mortality.

Abstract:
Influence of octane and benzene on plant cell ultrastructure and enzymes of basic metabolism, such as nitrogen assimilation and energy generation have been studied. Different plants: perennial ryegrass (Lolium perenne) and alfalfa (Medicago sativa); crops- maize (Zea mays L.) and bean (Phaseolus vulgaris); shrubs – privet (Ligustrum sempervirens) and trifoliate orange (Poncirus trifoliate); trees - poplar (Populus deltoides) and white mulberry (Morus alba L.) were exposed to hydrocarbons of different concentrations (1, 10 and 100 mM). Destructive changes in bean and maize leaves cells ultrastructure under the influence of benzene vapour were revealed at the level of photosynthetic and energy generation subcellular organells. Different deviations at the level of subcellular organelles structure and distribution were observed in alfalfa and ryegrass root cells under the influence of benzene and octane, absorbed through roots. The level of destructive changes is concentration dependent. Benzene at low 1 and 10 mM concentration caused the increase in glutamate dehydrogenase (GDH) activity in maize roots and leaves and in poplar and mulberry shoots, though to higher extent in case of lower, 1mM concentration. The induction was more intensive in plant roots. The highest tested 100mM concentration of benzene was inhibitory to the enzyme in all plants. Octane caused induction of GDH in all grassy plants at all tested concentrations; however the rate of induction decreased parallel to increase of the hydrocarbon concentration. Octane at concentration 1 mM caused induction of GDH in privet, trifoliate and white mulberry shoots. The highest, 100mM octane was characterized by inhibitory effect to GDH activity in all plants. Octane had inductive effect on malate dehydrogenase in almost all plants and tested concentrations, indicating the intensification of Trycarboxylic Acid Cycle. The data could be suggested for elaboration of criteria for plant selection for phytoremediation of oil hydrocarbons contaminated soils. Keywords: Higher plants, hydrocarbons, cell ultrastructure, glutamate and malate dehydrogenases.

Authors: Fatima Zohra Ibn Majdoub Hassani, Ivan Lavandera, Joseph Kreit
Abstract:
This study explored the effects of different organic solvents, temperature, and the amount of glycerol on the alcohol dehydrogenase (ADH)-catalysed stereoselective reduction of different ketones. These conversions were then analyzed by gas chromatography. It was found that when the amount of deep eutectic solvents (DES) increases, it can improve the stereoselectivity of the enzyme although reducing its ability to convert the substrate into the corresponding alcohol. Moreover, glycerol was found to have a strong stabilizing effect on the ADH from Ralstonia sp. (E. coli/ RasADH). In the case of organic solvents, it was observed that the best conversions into the alcohols were achieved with DMSO and hexane. It was also observed that temperature decreased the ability of the enzyme to convert the substrates into the products and also affected the selectivity. In addition to that, the recycling of DES up to three times gave good conversions and enantiomeric excess results and glycerol showed a positive effect in the stability of various ADHs. Using RasADH, a good conversion and enantiomeric excess into the S-alcohol were obtained. It was found that an enhancement of the temperature disabled the stabilizing effect of glycerol and decreased the stereoselectivity of the enzyme. However, for other ADHs a temperature increase had an opposite positive effect, especially with ADH-T from Thermoanaerobium sp. One of the objectives of this study was to see the effect of cofactors such as NAD(P) on the biocatlysis activities of ADHs.

Abstract:
The aim of this study was to examine current levels of physical activity determined via heart rate monitoring. A total of 176 children (85 boys, 91 girls) aged 5-13 years wore sealed Polar heart rate monitors for at least 10 hours per day on at least 3 days. Mean daily minutes of moderate to vigorous-intensity physical activity was 65 ± 43 (mean ± SD) for boys and 54 ± 37 for girls. Daily minutes of vigorous-intensity activity was 31 ± 24 and 24 ± 21 for boys and girls respectively. Significant differences in physical activity levels were observed between school day and weekends, boys and girls, and among age and geographical groups. Only 36% of boys and 22% of girls met the New Zealand physical activity guideline. Authors: Young-Seok Choi
Abstract:
This paper provides a quantitative measure of the time-varying multiunit neuronal spiking activity using an entropy based approach. To verify the status embedded in the neuronal activity of a population of neurons, the discrete wavelet transform (DWT) is used to isolate the inherent spiking activity of MUA. Due to the de-correlating property of DWT, the spiking activity would be preserved while reducing the non-spiking component. By evaluating the entropy of the wavelet coefficients of the de-noised MUA, a multiresolution Shannon entropy (MRSE) of the MUA signal is developed. The proposed entropy was tested in the analysis of both simulated noisy MUA and actual MUA recorded from cortex in rodent model. Simulation and experimental results demonstrate that the dynamics of a population can be quantified by using the proposed entropy.

Abstract:
A comparison of activity and stability of the as-formed Pt/C, Pt-Co and Pt-Pd/C electrocatalysts, prepared by a combined approach of impregnation and seeding, was performed. According to the activity test in a single Proton Exchange Membrane (PEM) fuel cell, the Oxygen Reduction Reaction (ORR) activity of the Pt-M/C electrocatalyst was slightly lower than that of Pt/C. The j0.9 V and E10 mA/cm2 of the as-prepared electrocatalysts increased in the order of Pt/C > Pt-Co/C > Pt-Pd/C. However, in the medium-to-high current density region, Pt-Pd/C exhibited the best performance. With regard to their stability in a 0.5 M H2SO4 electrolyte solution, the electrochemical surface area decreased as the number of rounds of repetitive potential cycling increased due to the dissolution of the metals within the catalyst structure. For long-term measurement, Pt- Pd/C was the most stable than the other three electrocatalysts. Keywords: ORR activity, Stability, Pt-based electrocatalysts, PEM fuel cell.

Abstract:
Candida albicans ATCC 10231 had low endogenous activity of the alternative oxidase compared with that of C. albicans ATCC 10261. In C. albicans ATCC 10231 the endogenous activity declined as the cultures aged. Alternative oxidase activity could be induced in C. albicans ATCC 10231 by treatment with cyanide, but the induction of this activity required the presence of oxygen which could be replaced, at least in part, with high concentrations of potassium ferricyanide. We infer from this that the expression of the gene encoding the alternative oxidase is under the control of a redoxsensitive transcription factor.

Abstract:
The purpose of this study was to determine the influence of physical activity and dietary fat intake on Body Mass Index (BMI) of lecturers within a higher learning institutionalized setting. The study adopted a Cross-sectional Correlational Design and included 120 lecturers selected proportionately by simple random sampling techniques from a population of 600 lecturers. Data was collected using questionnaires, which had sections including physical activity checklist adopted from the international physical activity questionnaire (IPAQ), 24-hour food recall, anthropometric measurements mainly weight and height. Analysis involved the use of bivariate correlations and linear regression. A significant inverse association was registered between BMI and duration (in minutes) spent doing moderate intense physical activity per day (r=-0.322, p<0.01). Physical activity also predicted BMI (r2=0.096, F=13.616, β=-3.22, t=-3.69, n=120, P<0.01). However, the association between Body Mass Index and dietary fat was not significant (r=0.038, p>0.05). Physical activity emerged as a more powerful determinant of BMI compared to dietary fat intake. Keywords: Physical activity, dietary fat intake, Body MassIndex, Kenya.

Abstract:
Malting is usually carried out on intact barley seed, while hull is still attached to it. In this study, oat grain with and without hull was subjected to controlled germination to optimize its enzymes activity, in such a way that lipase has the lowest and α- amylase and proteinase the highest activities. Since pH has a great impact on the activity of the enzymes, the pH of germination media was set up to 3 to 8. In dehulled oats, lipase and α-amylase had the lowest and highest activities in pHs 3 and 6, respectively whereas the highest proteinase activity was evidenced at pH 7 and 4 in the oats with and without hull respectively. While measurements indicated that the effect of hull on the enzyme activities particularly in lipase and amylase at each level of the pH are significantly different, the best results were obtained in those samples in which their hull had been removed. However, since the similar lipase activity in germinated dehulled oat were recorded at the pHs 4 and 5, therefore it was concluded that pH 5 in dehulled oat seed may provide the optimum enzyme activity for all the enzymes. Keywords: Enzyme activity, malting, oat, optimization.

Abstract:
In recent years, there has been a decline in physical activity among adults. Motivation has been shown to be a crucial factor in maintaining physical activity. The purpose of this study was to whether PA motives measured by the Physical Activity and Leisure Motivation Scale PALMS predicted the actual amount of PA at a later time to provide evidence for the construct validity of the PALMS. A quantitative, cross-sectional descriptive research design was employed. The Demographic Form, PALMS, and International Physical Activity Questionnaire Short form (IPAQ-S) questionnaires were used to assess motives and amount for physical activity in adults on two occasions. A sample of 489 male undergraduate students aged 18 to 25 years (mean ±SD; 22.30±8.13 years) took part in the study. Participants were divided into three types of activities, namely exercise, racquet sport, and team sports and female participants only took part in one type of activity, namely team sports. After 14 weeks, all 489 undergraduate students who had filled in the initial questionnaire (Occasion 1) received the questionnaire via email (Occasion 2). Of the 489 students, 378 males emailed back the completed questionnaire. The results showed that not only were pertinent sub-scales of PALMS positively related to amount of physical activity, but separate regression analyses showed the positive predictive effect of PALMS motives for amount of physical activity for each type of physical activity among participants. This study supported the construct validity of the PALMS by showing that the motives measured by PALMS did predict amount of PA. This information can be obtained to match people with specific sport or activity which in turn could potentially promote longer adherence to the specific activity. Keywords: Physical activity, motivation, the level of physical activity, types of physical activities.

Authors: Debora B. Moretti, Wiolene M. Nordi, Thaline Maira P. Cruz, José Eurico P. Cyrino, Raul Machado-Neto
Abstract:
Enzyme activity was evaluated in the intestine of juvenile dourado (Salminus brasiliensis) fed with diets containing 0, 10 or 20% of lyophilized bovine colostrum (LBC) inclusion for either 30 or 60 days. The intestinal enzymes acid and alkaline phosphatase (ACP and ALP, respectively), non-specific esterase (NSE), lipase (LIP), dipeptidyl aminopeptidase IV (DAP IV) and leucine aminopeptidase (LAP) were studied using histochemistry in four intestinal segments (S1, S2, S3 and posterior intestine). Weak proteolitic activity was observed in all intestinal segments for DAP IV and LAP. The activity of NSE and LIP was also weak in all intestines, except for the moderate activity of NSE in the S2 of 20% LBC group after 30 days and in the S1 of 0% LBC group after 60 days. The ACP was detected only in the S2 and S3 of the 10% LBC group after 30 days. Moderate and strong staining was observed in the first three intestinal segments for ALP and weak activity in the posterior intestine. The activity of DAP IV, LAP and ALP were also present in the cytoplasm of the enterocytes. In the present results, bovine colostrum feeding did not cause alterations in activity of intestinal enzymes.

Authors: Sh. Khoshemehry, M. J. Pourvaghar, M. E. Bahram
Abstract:
In modern life, daily physical activity is relatively reduced, which is why the incidence of some diseases associated with overweight and obesity, such as hypertension, diabetes and other chronic illnesses, even in young people are observed. Obesity and overweight is one of the most common metabolic disorders in industrialized countries and in developing countries. One consequence of pathological obesity is cardiovascular disease and metabolic syndrome. In the past, it was believed that adipose tissue was ineffective and served only for storing triglycerides. In this review article, it was tried to refer to the esteemed scientific sources about physical activity and responses of leptin.

Authors: Jacques J. Scheepers, Edison Muzenda
Abstract:
This work focused on the interactions which occur between ester solvents and alcohol solutes. The alcohols selected ranged from the simplest alcohol (methanol) to C10-alcohols, and solubility predictions in the form of infinite dilution activity coefficients were made using the Modified UNIFAC Dortmund group contribution model. The model computation was set up on a Microsoft Excel spreadsheet specifically designed for this purpose. It was found that alcohol/ ester interactions yielded an increase in activity coefficients (i.e. became less soluble) with an increase in the size of the ester solvent molecule. Furthermore, activity coefficients decreased with an increase in the size of the alcohol solute. The activity coefficients also decreased with an increase in the degree of unsaturation of the ester hydrocarbon tail. Tertiary alcohols yielded lower activity coefficients than primary alcohols. Finally, cyclic alcohols yielded higher activity coefficients than straight-chain alcohols until a point is reached where the trend is reversed, referred to as the ‘crossover’ point.

Authors: M. Yaldagard, S.A. Mortazavi, F. Tabatabaie
Abstract:
In the present study, the effects of ultrasound as emerging technology were investigated on germination stimulation, amount of alpha-amylase activity on dry barley seeds before steeping stage of malting process. All experiments were carried out at 20 KHz on the ultrasonic generator in 3 different ultrasonic intensities (20, 60 and 100% setting from total power of device) and time (5, 10 and 15 min) at constant temperature (30C). For determining the effects of these parameters on enzyme the Fuwa method assay based on the decreased staining value of blue starch–iodine complexes employed for measurement an activity. The results of these assays were analyzed by Qualitek4 software using the Taguchi statistical method to evaluate the factor-s effects on enzyme activity. It has been found that when malting barley is irradiated with an ultrasonic power, a stimulating effect occurs as to the enzyme activity. Keywords: ultrasound, alpha-amylase activity, stimulationand Taguchi statistical method.
