1931 Isolation and Screening of Fungal Strains for β-Galactosidase Production

Abstract:
Enzymes are the biocatalysts which catalyze the biochemical processes and thus have a wide variety of applications in the industrial sector. β-Galactosidase (E.C. 3.2.1.23) also known as lactase, is one of the prime enzymes, which has significant potential in the dairy and food processing industries. It has the capability to catalyze both the hydrolytic reaction for the production of lactose hydrolyzed milk and transgalactosylation reaction for the synthesis of prebiotics such as lactulose and galactooligosaccharides. These prebiotics have various nutritional and technological benefits. Although, the enzyme is naturally present in almonds, peaches, apricots and other variety of fruits and animals, the extraction of enzyme from these sources increases the cost of enzyme. Therefore, focus has been shifted towards the production of low cost enzyme from the microorganisms such as bacteria, yeast and fungi. As compared to yeast and bacteria, fungal β-galactosidase is generally preferred as being extracellular and thermostable in nature. Keeping the above in view, the present study was carried out for the isolation of the β-galactosidase producing fungal strain from the food as well as the agricultural wastes. A total of more than 100 fungal cultures were examined for their potential in enzyme production. All the fungal strains were screened using X-gal and IPTG as inducers in the modified Czapek Dox Agar medium. Among the various isolated fungal strains, the strain exhibiting the highest enzyme activity was chosen for further phenotypic and genotypic characterization. The strain was identified as Rhizomucor pusillus on the basis of 5.8s RNA gene sequencing data.

Abstract:
A macroscopic constitutive equation is developed for a high-density cellulose insulation material with emphasis on the outof- plane stress relaxation behavior. A hypothesis is proposed where the total stress is additively composed by an out-of-plane visco-elastic isotropic contribution and an in-plane elastic orthotropic response. The theory is validated against out-of-plane stress relaxation, compressive experiments and in-plane tensile hysteresis, respectively. For large scale finite element simulations, the presented model provides a balance between simplicity and capturing the materials constitutive behaviour.

Abstract:
One of the biggest concerns in the field of agriculture is around the energy efficiency of robots that will perform agriculture’s activity and their charging methods. In this paper, two different charging methods for agricultural standalone docking stations are shown that will take into account various variants as field size and its irregularities, work’s nature to which the robot will perform, deadlines that have to be respected, among others. Its features also are dependent on the orchard, season, battery type and its technical specifications and cost. First charging base method focuses on wireless charging, presenting more benefits for small field. The second charging base method relies on battery replacement being more suitable for large fields, thus avoiding the robot stop for recharge. Existing many methods to charge a battery, the CC CV was considered the most appropriate for either simplicity or effectiveness. The choice of the battery for agricultural purposes is if most importance. While the most common battery used is Li-ion battery, this study also discusses the use of graphene-based new type of batteries with 45% over capacity to the Li-ion one. A Battery Management Systems (BMS) is applied for battery balancing. All these approaches combined showed to be a promising method to improve a lot of technical agricultural work, not just in terms of plantation and harvesting but also about every technique to prevent harmful events like plagues and weeds or even to reduce crop time and cost.

This paper proposes a method to vibration analysis in order to on-line monitoring and predictive maintenance during the milling process. Adapting envelope method to diagnostics and the analysis for milling tool materials is an important contribution to the qualitative and quantitative characterization of milling capacity and a step by modeling the three-dimensional cutting process. An experimental protocol was designed and developed for the acquisition, processing and analyzing three-dimensional signal. The vibration envelope analysis is proposed to detect the cutting capacity of the tool with the optimization application of cutting parameters. Abstract:
A model is presented to find the optimal design of the mixed renewable warranty policy for non-repairable Weibull life products. The optimal design considers the conflict of interests between the customer and the manufacturer: the customer interests are longer full rebate coverage period and longer total warranty coverage period, the manufacturer interests are lower warranty cost and lower risk. The design factors are full rebate and total warranty coverage periods. Results showed that mixed policy is better than full rebate policy in terms of risk and total warranty coverage period in all of the three bathtub regions. In addition, results showed that linear policy is better than mixed policy in infant mortality and constant failure regions while the mixed policy is better than linear policy in ageing region of the model. Furthermore, the results showed that using burn-in period for infant mortality products reduces warranty cost and risk.Keywords: Reliability, Mixed warranty policy, Optimization, Weibull Distribution.

Abstract:
Text mining techniques are generally applied for classifying the text, finding fuzzy relations and structures in data sets. This research provides plenty text mining capabilities. One common application is text classification and event extraction, which encompass deducing specific knowledge concerning incidents referred to in texts. The main contribution of this paper is the clarification of a concept graph generation mechanism, which is based on a text classification and optimal fuzzy relationship extraction. Furthermore, the work presented in this paper explains the application of fuzzy relationship extraction and branch and bound (BB) method to simplify the texts.

Abstract:
The aim of this work is to study the possible use of stone cutting sludge waste in concrete production, which would reduce both the environmental impact and the production cost .Slurry sludge was used a source of water in concrete production, which was obtained from Samara factory/Jordan, The physico-chemical and mineralogical characterization of the sludge was carried out to identify the major components and to compare it with the typical sand used to produce concrete. Samples analysis showed that 96% of slurry sludge volume is water, so it should be considered as an important source of water. Results indicated that the use of slurry sludge as water source in concrete production has insignificant effect on compression strength, while it has a sharp effect on the slump values. Using slurry sludge with a percentage of 25% of the total water content obtained successful concrete samples regarding slump and compression tests. To clarify slurry sludge, settling process can be used to remove the suspended solid. A settling period of 30 min. obtained 99% removal efficiency. The clarified water is suitable for using in concrete mixes, which reduce water consumption, conserve water recourses, increase the profit, reduce operation cost and save the environment. Additionally, the dry sludge could be used in the mix design instead of the fine materials with sizes < 160 um. This application could conserve the natural materials and solve the environmental and economical problem caused by sludge accumulation.Keywords: Concrete, recycle, sludge, slurry waste, stone cutting waste, waste.

Despite being one of the most significant economic contributors of the country, Canada’s construction industry is lagging behind other sectors when it comes to labor productivity improvements. The construction industry is very collaborative as a general contractor, will hire trade contractors to perform most of a project’s work; meaning low productivity from one contractor can have a domino effect on the shared success of a project. To address this issue and encourage trade contractors to improve their productivity tracking methods, an investigative study was done on the productivity views and tracking methods of various trade contractors. Additionally, an in-depth review was done on four standard tracking methods used in the construction industry: cost codes, benchmarking, the job productivity measurement (JPM) standard, and WorkFace Planning (WFP). The four tracking methods were used as a baseline in comparing the trade contractors’ responses, determining gaps within their current tracking methods, and for making improvement recommendations. 15 interviews were conducted with different trades to analyze how contractors value productivity. The results of these analyses indicated that there seem to be gaps within the construction industry when it comes to an understanding of the purpose and value in productivity tracking. The trade contractors also shared their current productivity tracking systems; which were then compared to the four standard tracking methods used in the construction industry. Gaps were identified in their various tracking methods and using a framework; recommendations were made based on the type of trade on how to improve how they track productivity.

Abstract:
The objective of this work was to examine the changes in non destructive properties caused by carbonation of CEM II mortar. Samples of CEM II mortar were prepared and subjected to accelerated carbonation at 20°C, 65% relative humidity and 20% CO2 concentration. We examined the evolutions of the gas permeability, the thermal conductivity, the thermal diffusivity, the volume of the solid phase by helium pycnometry, the longitudinal and transverse ultrasonic velocities. The principal contribution of this work is that, apart of the gas permeability, changes in other non destructive properties have never been studied during the carbonation of cement materials. These properties are important in predicting/measuring the durability of reinforced concrete in CO2 environment. The carbonation depth and the porosity accessible to water were also reported in order to explain comprehensively the changes in non destructive parameters.Keywords: Carbonation, cement mortar, longitudinal and transverse ultrasonic velocities, non destructive tests.

It is not easy to imagine how the existing city can be converted to the principles of sustainability, however, the need for innovation, requires a pioneering phase which must address the main problems of rehabilitation of the operating models of the city. Today, however, there is a growing awareness that the identification and implementation of policies and measures to promote the adaptation, resilience and reversibility of the city, require the contribution of our discipline. This breakthrough is present in some recent international experiences of Climate Plans, in which the envisaged measures are closely interwoven with those of urban planning. Abstract:
In the 1980s, companies began to feel the effect of three major influences on their product development: newer and innovative technologies, increasing product complexity and larger organizations. And therefore companies were forced to look for new product development methods. This paper tries to focus on the two of new product development methods (DFM and CE). The aim of this paper is to see and analyze different product development methods specifically on Design for Manufacturability and Concurrent Engineering. Companies can achieve and be benefited by minimizing product life cycle, cost and meeting delivery schedule. This paper also presents simplified models that can be modified and used by different companies based on the companies- objective and requirements. Methodologies that are followed to do this research are case studies. Two companies were taken and analysed on the product development process. Historical data, interview were conducted on these companies in addition to that, Survey of literatures and previous research works on similar topics has been done during this research. This paper also tries to show the implementation cost benefit analysis and tries to calculate the implementation time. From this research, it has been found that the two companies did not achieve the delivery time to the customer. Some of most frequently coming products are analyzed and 50% to 80 % of their products are not delivered on time to the customers. The companies are following the traditional way of product development that is sequentially design and production method, which highly affect time to market. In the case study it is found that by implementing these new methods and by forming multi disciplinary team in designing and quality inspection; the company can reduce the workflow steps from 40 to 30.

Hence, service quality, in general, is a complicated process to be measured and evaluated. Hotels, as a service sector and part of the hospitality industry, are growing rapidly. This research paper was carried out to identify the quality determinants that may affect hotel guests’ service quality perception. In this research paper, each quality determinant will be discussed, illustrated, and justified thoroughly via a systematic literature review. This paper sets the stage to measure the significant influence of the service quality determinants on guest satisfaction. The knowledge contribution from this study proposes to practitioners and/or hotel service providers, fundamental elements to adopt the implications into their policies.

Abstract:
Hydraulic structures such as gravity dams are classified as essential structures, and have the vital role in providing strong and safe water resource management. Three major aspects must be considered to achieve an effective design of such a structure: 1) The building cost, 2) safety, and 3) accurate analysis of seepage characteristics. Due to the complexity and non-linearity relationships of the seepage process, many approximation theories have been developed; however, the application of these theories results in noticeable errors. The analytical solution, which includes the difficult conformal mapping procedure, could be applied for a simple and symmetrical problem only. Therefore, the objectives of this paper are to: 1) develop a surrogate model based on numerical simulated data using SEEPW software to approximately simulate seepage process related to a hydraulic structure, 2) develop and solve a linked simulation-optimization model based on the developed surrogate model to describe the seepage occurring under a concrete gravity dam, in order to obtain optimum and safe design at minimum cost. The virtual container yard is an effective solution to the container inventory imbalance problem which is a global issue. It causes substantial cost to carriers, which inadvertently adds to the prices of consumer goods. The virtual container yard is rooted in the fundamentals of container interchange between carriers. If carriers opt to interchange their excess containers with those who are deficit, a substantial part of the empty reposition cost could be eliminated. Unlike in other types of ships, cargo cannot be directly loaded to a container ship. Slots and containers are supplementary components; thus, without containers, a carrier cannot ship cargo if the containers are not available and vice versa. Few decades ago, carriers recognized slot (the unit of space in a container ship) interchange as a viable solution for the imbalance of shipping space. Carriers interchange slots among them and it also increases the advantage of scale of economies in container shipping. Some of these service agreements between mega carriers have provisions to interchange containers too. However, the interchange mechanism is still not popular among carriers for containers. This is the paradox that prevails in the liner shipping industry. At present, carriers reposition their excess empty containers to areas where they are in demand. This research applied factor analysis statistical method. The paper reveals that five major components may influence the virtual container yard namely organisation, practice and culture, legal and environment, international nature, and marketing. There are 12 variables that may impact the virtual container yard, and these are explained in the paper.

Abstract:
Crop yield prediction is a paramount issue in agriculture. The main idea of this paper is to find out efficient way to predict the yield of corn based meteorological records. The prediction models used in this paper can be classified into model-driven approaches and data-driven approaches, according to the different modeling methodologies. The model-driven approaches are based on crop mechanistic modeling. They describe crop growth in interaction with their environment as dynamical systems. But the calibration process of the dynamic system comes up with much difficulty, because it turns out to be a multidimensional non-convex optimization problem. An original contribution of this paper is to propose a statistical methodology, Multi-Scenarios Parameters Estimation (MSPE), for the parametrization of potentially complex mechanistic models from a new type of datasets (climatic data, final yield in many situations). It is tested with CORNFLO, a crop model for maize growth. On the other hand, the data-driven approach for yield prediction is free of the complex biophysical process. But it has some strict requirements about the dataset. A second contribution of the paper is the comparison of these model-driven methods with classical data-driven methods. For this purpose, we consider two classes of regression methods, methods derived from linear regression (Ridge and Lasso Regression, Principal Components Regression or Partial Least Squares Regression) and machine learning methods (Random Forest, k-Nearest Neighbor, Artificial Neural Network and SVM regression). The dataset consists of 720 records of corn yield at county scale provided by the United States Department of Agriculture (USDA) and the associated climatic data. A 5-folds cross-validation process and two accuracy metrics: root mean square error of prediction(RMSEP), mean absolute error of prediction(MAEP) were used to evaluate the crop prediction capacity. The results show that among the data-driven approaches, Random Forest is the most robust and generally achieves the best prediction error (MAEP 4.27%). It also outperforms our model-driven approach (MAEP 6.11%). However, the method to calibrate the mechanistic model from dataset easy to access offers several side-perspectives. The mechanistic model can potentially help to underline the stresses suffered by the crop or to identify the biological parameters of interest for breeding purposes. The adaptation of social networking sites within higher education has garnered significant interest in the recent years with numerous researches considering it as a possible shift from the traditional classroom based learning paradigm. Notwithstanding this increase in research and conducted studies however, the adaption of SNS based modules have failed to proliferate within Universities. This paper commences its contribution by analyzing the various models and theories proposed in literature and amalgamate together various effective aspects for the inclusion of social technology within e-Learning. A three phased framework is further proposed which details the necessary considerations for the successful adaptation of SNS in enhancing the students learning experience. This proposal outlines the theoretical foundations which will be analyzed in practical implementation across international university campuses.

The concept of converting the kinetic energy of quadcopter propellers into electrical energy is considered in this contribution following the feasibility study of the propeller vibrations, theoretical energy conversion, and simulation techniques. Analysis of the propeller vibration performance is presented via graphical representation of calculated and simulated parameters, in order to demonstrate the possibility of recovering the harvested energy from the propeller vibrations of the quadcopter while the quadcopter is in operation. Consideration of using piezoelectric materials in such concept, converting the mechanical energy of the propeller into the electrical energy, is given. Abstract:
This paper presents an approach based on the adoption of a distributed cognition framework and a non parametric multicriteria evaluation methodology (DEA) designed specifically to compare e-commerce websites from the consumer/user viewpoint. In particular, the framework considers a website relative efficiency as a measure of its quality and usability. A website is modelled as a black box capable to provide the consumer/user with a set of functionalities. When the consumer/user interacts with the website to perform a task, he/she is involved in a cognitive activity, sustaining a cognitive cost to search, interpret and process information, and experiencing a sense of satisfaction. The degree of ambiguity and uncertainty he/she perceives and the needed search time determine the effort size – and, henceforth, the cognitive cost amount – he/she has to sustain to perform his/her task. On the contrary, task performing and result achievement induce a sense of gratification, satisfaction and usefulness. In total, 9 variables are measured, classified in a set of 3 website macro-dimensions (user experience, site navigability and structure). The framework is implemented to compare 40 websites of businesses performing electronic commerce in the information technology market. A questionnaire to collect subjective judgements for the websites in the sample was purposely designed and administered to 85 university students enrolled in computer science and information systems engineering undergraduate courses.Keywords: Website, e-commerce, DEA, distributed cognition, evaluation, comparison.

This contribution examines the relationship between the family environment and the level of young pupils’ scholastic success. It comments on the partial results of a research probe carried out in the year 2012 on a sample of 412 Czech Republic primary school pupils of the fourth, fifth and sixths forms within the Project IGA 43 201 15 0004 01. The key links of this project were monitored in relation to the highest education level achieved by the learners´ parents, as well as to the type of family it is (in particular its ability to function), to component factors specific to the family climate (their willingness to share information, communication, parental control) and, finally, to the number of children in the family as an important socialization constituent.

This research has two objectives: The first is to make a contribution by focusing on the development of a competence approach, based on continuous improvement. It allows the enterprise to spot key competencies, mobilize them in order to serve its strategic objectives and to develop future competencies. The second is to propose a method to evaluate the Level of Collective Competence. The issue of unintentional islanding in PV grid interconnection still remains as a challenge in grid-connected photovoltaic (PV) systems. This paper discusses the overview of popularly used anti-islanding detection methods, practically applied in PV grid-connected systems. Anti-islanding methods generally can be classified into four major groups, which include passive methods, active methods, hybrid methods and communication base methods. Active methods have been the preferred detection technique over the years due to very small non-detected zone (NDZ) in small scale distribution generation. Passive method is comparatively simpler than active method in terms of circuitry and operations. However, it suffers from large NDZ that significantly reduces its performance. Communication base methods inherit the advantages of active and passive methods with reduced drawbacks. Hybrid method which evolved from the combination of both active and passive methods has been proven to achieve accurate anti-islanding detection by many researchers. For each of the studied anti-islanding methods, the operation analysis is described while the advantages and disadvantages are compared and discussed. It is difficult to pinpoint a generic method for a specific application, because most of the methods discussed are governed by the nature of application and system dependent elements. This study concludes that the setup and operation cost is the vital factor for anti-islanding method selection in order to achieve minimal compromising between cost and system quality.Keywords: Active method, hybrid method, islanding detection, passive method, photovoltaic (PV), utility method

Abstract:
The use of hydroelectric pump-storage system at large scale, MW-size systems, is already widespread around the world. Designed for large scale applications, pump-storage station can be scaled-down for small, remote residential applications. Given the cost and complexity associated with installing a substation further than 100 miles from the main transmission lines, a remote, independent and self-sufficient system is by far the most feasible solution. This article is aiming at the design of wind and solar power generating system, by means of pumped-storage to replace the wind and /or solar power systems with a battery bank energy storage. Wind and solar pumped-storage power generating system can reduce the cost of power generation system, according to the user's electricity load and resource condition and also can ensure system reliability of power supply. Wind and solar pumped-storage power generation system is well suited for remote residential applications with intermittent wind and/or solar energy. This type of power systems, installed in these locations, could be a very good alternative, with economic benefits and positive social effects. The advantage of pumped storage power system, where wind power regulation is calculated, shows that a significant smoothing of the produced power is obtained, resulting in a power-on-demand system’s capability, concomitant to extra economic benefits.Keywords: Battery bank, photo-voltaic, pump-storage, wind energy.

Abstract:
One part of the total employee-s reward is apart from basic wages or salary, employee-s benefits and intangible elements also so called contingent (variable) pay. Contingent pay is connected to performance, contribution, capcompetency or skills of individual employees, and to team-s or company-wide performance or to combination of few of the mentioned possibilities. Main aim of this article is to define, based on available information, contingent pay, describe reasons for its implementation and arguments for and against this type of remuneration, but also bring information not only about its extent and level of utilization by organizations of the Czech Republic operating in the field of environmental protection, but also mention their practical experience with this type of remuneration.

The growth of population, rising fossil fuel prices (limited and decreasing day by day), pollution problem due to use of fossil fuels and increasing electrical demand are important factors that encourage the use of green and renewable energy technologies. Among the different renewable energy technologies, hydro power generation (large and small scale) is the prime choice in terms of contribution to the world's electricity generation by using water current turbines. Currently, researchers mainly focused on design and development of different kind of turbines to capture hydropower to generate electricity as clean and reliable energy. This paper is a review of the status of research on water current turbines carried out to generate electricity from hydrokinetic energy especially in places where there is no electricity, but there is access to flowing water.

This contribution presents a friction estimator for industrial purposes which identifies Coulomb friction in a steering system. The estimator only needs a few, usually known, steering system parameters. Friction occurs on almost every mechanical system and has a negative influence on high-precision position control. This is demonstrated on a steering angle controller for highly automated driving. In this steering system the friction induces limit cycles which cause oscillating vehicle movement when the vehicle follows a given reference trajectory. When compensating the friction with the introduced estimator, limit cycles can be suppressed. Abstract:
This contribution aims to compare legislation adjusting the course of insolvency proceedings in France, Germany and Slovakia. On the basis of an investigation of the legislative adjustment of this problem, an attempt is made to ascertain in the given countries the extent to which the outcome of the entire proceedings is influenced by legislation and to determine the fundamental moments that influence costs, recovery rate and the duration of proceedings. A comparative analysis was utilized in order to achieve the set goal. The results of the survey could be used to improve legislation so as to lead in the best and most expedient way to a departure from the market of those subjects that are for economic reasons unable to continue with their activities whilst burdening the entire process with the lowest possible costs, which would lead to a high level of satisfaction for creditors.

Abstract:
Silicon nanowire (SiNW) based thermoelectric device (TED) has potential applications in areas such as chip level cooling/ energy harvesting. It is a great challenge however, to assemble an efficient device with these SiNW. The presence of parasitic in the form of interfacial electrical resistance will have a significant impact on the performance of the TED. In this work, we explore the effect of the electrical contact resistance on the performance of a TED. Numerical simulations are performed on SiNW to investigate such effects on its cooling performance. Intrinsically, SiNW individually without the unwanted parasitic effect has excellent cooling power density. However, the cooling effect is undermined with the contribution of the electrical contact resistance.

Abstract:
In this paper, we observe that developed countries are generally equipped with innovation capabilities and produce major chunk of the world-s knowledge and technology. The contribution of developing countries, on the other hand, is insignificant, and most of them far behind the global technological front. More specifically, we empirically observe that the developing world neither contributes substantially to the world-s scientific publications nor to the R&D activities. They also have lesser “absorptive capacity" and “technological capability", and their “innovation systems" are plagued with many problems. Finally, we argue that these countries can break the shackles and improve their innovation capabilities by pursuing genuine innovation policies on long-term basis with honesty and commitment.

Abstract:
In the recent years, high dynamic range imaging has gain popularity with the advancement in digital photography. In this contribution we present a subjective evaluation of various tone production and tone mapping techniques by a number of participants. Firstly, standard HDR images were used and the participants were asked to rate them based on a given rating scheme. After that, the participant was asked to rate HDR image generated using linear and nonlinear combination approach of multiple exposure images. The experimental results showed that linearly generated HDR images have better visualization than the nonlinear combined ones. In addition, Reinhard et al. and the exponential tone mapping operators have shown better results compared to logarithmic and the Garrett et al. tone mapping operators.Keywords: tone mapping, high dynamic range, low dynamic range, bits per pixel.
