205 Designing Next Generation Platforms for Recombinant Protein Production by Genome Engineering of Escherichia coli
Authors: Priyanka Jain, Ashish K. Sharma, Esha Shukla, K. J. Mukherjee
Abstract:
We propose a paradigm shift in our approach to design improved platforms for recombinant protein production, by addressing system level issues rather than the individual steps associated with recombinant protein synthesis like transcription, translation, etc. We demonstrate that by controlling and modulating the cellular stress response (CSR), which is responsible for feedback control of protein synthesis, we can generate hyper-producing strains. We did transcriptomic profiling of post-induction cultures, expressing different types of protein, to analyze the nature of this cellular stress response. We found significant down-regulation of substrate utilization, translation, and energy metabolism genes due to generation CSR inside the host cell. However, transcription profiling has also shown that many genes are up-regulated post induction and their role in modulating the CSR is unclear. We hypothesized that these up-regulated genes trigger signaling pathways, generating the CSR and concomitantly reduce the recombinant protein yield. To test this hypothesis, we knocked out the up-regulated genes, which did not have any downstream regulatees, and analyzed their impact on cellular health and recombinant protein expression. Two model proteins i.e., GFP and L-Asparaginase were chosen for this analysis. We observed a significant improvement in expression levels, with some knock-outs showing more than 7-fold higher expression compared to control. The 10 best single knock-outs were chosen to make 45 combinations of all possible double knock-outs. A further increase in expression was observed in some of these double knock- outs with GFP levels being highest in a double knock-out ΔyhbC + ΔelaA. However, for L-Asparaginase which is a secretory protein, the best results were obtained using a combination of ΔelaA+ΔcysW knock-outs. We then tested all the knock outs for their ability to enhance the expression of a 'difficult-to-express' protein. The Rubella virus E1 protein was chosen and tagged with sfGFP at the C-terminal using a linker peptide for easy online monitoring of expression of this fusion protein. Interestingly, the highest increase in Rubella-sGFP levels was obtained in the same double knock-out ΔelaA + ΔcysW (5.6 fold increase in expression yield compared to the control) which gave the highest expression for L-Asparaginase. However, for sfGFP alone, the ΔyhbC+ΔmarR knock-out gave the highest level of expression. These results indicate that there is a fair degree of commonality in the nature of the CSR generated by the induction of different proteins. Transcriptomic profiling of the double knock out showed that many genes associated with the translational machinery and energy biosynthesis did not get down-regulated post induction, unlike the control where these genes were significantly down-regulated. This confirmed our hypothesis of these genes playing an important role in the generation of the CSR and allowed us to design a strategy for making better expression hosts by simply knocking out key genes. This strategy is radically superior to the previous approach of individually up-regulating critical genes since it blocks the mounting of the CSR thus preventing the down-regulation of a very large number of genes responsible for sustaining the flux through the recombinant protein production pathway. Keywords: cellular stress response, GFP, knock-outs, up-regulated genes

Authors: Muhammad Saleem
Abstract:
The research paper entitled above would be a comprehensive and critical study of translations of the meanings of the Holy Qur’an. The discussion will deal with the targets & purposes of Urdu (National Language of Pakistan) translators of the meanings of the Holy Qur’an. There are more than 400 translations of the meanings of the Holy Qur’an in the Urdu Language. Muslims, non-Muslims and some organizations have made translations of the meanings of the Holy Qur’an to meet various targets. It is observed that all Urdu translators have not translated the Qur’an with a single objective and motivation; rather, some are biased and strive to discredit the Qur’an. Thus, they have made unauthentic and fabricated translations of the Qur’an. Some optimistically believe that they intend to do a service, whereas others pessimistically hold that they treacherously seek to further their rule. Some of them have been observed to be against Islam, starting their activities with spite, but after perceiving the truths of Islam and the miracle and greatness of the Holy Qur’an, they submitted to Islam, embracing it with pure hearts. Some translators made their translations of the meanings of the Holy Qur’an to serve Allah, and some of them have done their translations to earn only. All these translations vary from one to another due to style, trend, type, method and style. Some Urdu translations have been made to fulfill the lingual requirements. Some translations have been made by Muslim scholars to reduce the influence of Urdu translations of the meanings of the Holy Qur’an by Non-Muslims. The article deals with the various purposes of the translators of the meanings of the Holy Qur’an.Keywords: Qur'an, translation, urdu, language

Procedia PDF Downloads 438200 A Refinement Strategy Coupling Event-B and Planning Domain Definition Language (PDDL) for Planning Problems
Authors: Sabrine Ammar, Mohamed Tahar Bhiri
Abstract:
Automatic planning has a de facto standard language called Planning Domain Definition Language (PDDL) for describing planning problems. It aims to formalize the planning problems described by the concept of state space. PDDL-related dynamic analysis tools, namely planners and validators, are insufficient for verifying and validating PDDL descriptions. Indeed, these tools made it possible to detect errors a posteriori by means of test activity. In this paper, we recommend a formal approach coupling the two languages Event-B and PDDL, for automatic planning. Event-B is used for formal modeling by stepwise refinement with mathematical proofs of planning problems. Thus, this paper proposes a refinement strategy allowing to obtain reliable PDDL descriptions from an ultimate Event-B model correct by construction. The ultimate Event-B model, correct by construction which is supposed to be translatable into PDDL, is automatically translated into PDDL using our MDE Event-B2PDDL tool. Keywords: code generation, event-b, PDDL, refinement strategy, translation rules
Procedia PDF Downloads 196199 Perspectives of Computational Modeling in Sanskrit Lexicons
Authors: Baldev Ram Khandoliyan, Ram Kishor
Abstract:
India has a classical tradition of Sanskrit Lexicons. Research work has been done on the study of Indian lexicography. India has seen amazing strides in Information and Communication Technology (ICT) applications for Indian languages in general and for Sanskrit in particular. Since Machine Translation from Sanskrit to other Indian languages is often the desired goal, traditional Sanskrit lexicography has attracted a lot of attention from the ICT and Computational Linguistics community. From Nighaŋţu and Nirukta to Amarakośa and Medinīkośa, Sanskrit owns a rich history of lexicography. As these kośas do not follow the same typology or standard in the selection and arrangement of the words and the information related to them, several types of Kośa-styles have emerged in this tradition. The model of a grammar given by Aṣṭādhyāyī is well appreciated by Indian and western linguists and grammarians. But the different models provided by lexicographic tradition also have importance. The general usefulness of Sanskrit traditional Kośas is well discussed by some scholars. That is most of the matter made available in the text. Some also have discussed the good arrangement of lexica. This paper aims to discuss some more use of the different models of Sanskrit lexicography especially focusing on its computational modeling and its use in different computational operations. Keywords: computational lexicography, Sanskrit Lexicons, nighanṭu, kośa, Amarkosa

Authors: Amal Alshahrani, Allan Ramsay
Abstract:
Paraphrasing is one of the important tasks in natural language processing; i.e. alternative ways to express the same concept by using different words or phrases. Paraphrases can be used in many natural language applications, such as Information Retrieval, Machine Translation, Question Answering, Text Summarization, or Information Extraction. To obtain pairs of sentences that are paraphrases we create a system that automatically extracts paraphrases from a corpus, which is built from different sources of news article since these are likely to contain paraphrases when they report the same event on the same day. There are existing simple standard approaches (e.g. TF-IDF vector space, cosine similarity) and alignment technique (e.g. Dynamic Time Warping (DTW)) for extracting paraphrase which have been applied to the English. However, the performance of these approaches could be affected when they are applied to another language, for instance Arabic language, due to the presence of phenomena which are not present in English, such as Free Word Order, Zero copula, and Pro-dropping. These phenomena will affect the performance of these algorithms. Thus, if we can analysis how the existing algorithms for English fail for Arabic then we can find a solution for Arabic. The results are promising. Keywords: natural language processing, TF-IDF, cosine similarity, dynamic time warping (DTW)

Authors: Ezayra Dubria, Leonora Yambao
Abstract:
Mother Tongue–Based Multilingual Education (MTB-MLE) is a salient part of the recent reform in the country’s Education system which is the implementation of the K to 12 Basic Education Program. Its importance is highlighted by the passing of Republic Act 10523, otherwise known as the ‘Enhanced Basic Education Act of 2013’. However, teachers, especially new teachers encounter problems in using mother tongue as medium of instruction. Fortunately, teachers are able to create strategies which address these problems. Specifically, this paper gathered the viewpoints of teachers in using mother tongue and analyzed the different problems and strategies used. The problems encountered by teachers are lack of instructional materials written in mother tongue, especially books, lack of vocabulary, lack of teacher training, and influences of social media to learners. The strategies which address these problems are translation of literary pieces and other instructional materials, vocabulary enrichment through the use of word-of-the-day and picture-word association, remedial class, storytelling, differentiated instruction, explicit teaching, individual and group activities, and utilization of multilingual teaching. Keywords: mother tongue-based instruction, multilingualism, problems, strategies

Abstract:
In precision engineering, including precision positioning, micro-manipulation, robotic systems... a majority of these applications actuated by piezo stack used the compliant amplifier mechanism to amplifying motion and guiding it as needed utilize the flexibility of their components, in this paper, we present a novel approach introducing a symmetric structure comprising three stages, featuring rectangular flexure hinges with a compact size of 77mm×42mm×10mm. This design provides the capability for rotation, translation or a combination of both movements in both directions. The system allows for a displacement of 2107.5 μm when the input displacement of PZT is 50 μm while considering the material constraints of the aluminum alloy (7075 T6) which has a maximum admissible stress of 500 MPa However, our proposed design imposes additional constraints to ensure the stress remains below 361 MPa for optimal performance. These findings were obtained through finite element simulations conducted using ANSYS Workbench. Furthermore, our module facilitates precise control of various components within robotic systems, allowing for adjustable speeds based on specific requirements or desired outcomes. Keywords: robotic, piezoelectric, compliant mechanism, flexure hinge

Authors: Sonakshi Saxena, Moosath Harishankar Vasudevan
Abstract:
The current study seeks to investigate the differences in the psychological impact of movies in their original and translated versions. International cinema is exemplar of the success of globalization. The multitude of languages in the global village does not seem to impede the common cinematic goal of filmmakers across linguistic boundaries. To understand, hence, whether the psychological impact of movies, intentional or otherwise, is preserved when the original is translated into a different language, an experimental design was adopted. Multilingual participants in the age group 18-25 years were recruited for the same. A control group and an experimental group were randomly assigned and the psychological impacts of movies were studied under two conditions- a) watching the movie in its original language, and b) watching the movie in its original language as well as translated version. For the second condition, the experimental group was further divided into two groups randomly to balance order effects. The major aspects of psychological impact assessed were emotional impact and attitude towards the movie. The scores were compared for the two groups. It is further discussed whether the experience is salient across language or do languages inherently possess the ability to alter experiences of the audience. Keywords: experimental design, movies, psychological impact, translation
Procedia PDF Downloads 397194 Experimental Investigation of the Failure Behavior of a Retaining Wall Constructed with Soil Bags
Authors: Kewei Fan, Sihong Liu, Yi Pik Cheng
Abstract:
This paper aims to analyse the failure behaviour of the retaining wall constructed with soil bags that are formed by filling river sand into woven bags (geosynthetics). Model tests were conducted to obtain the failure mode of the wall, and shear tests on two-layers and five-layers of soil bags were designed to investigate the mechanical characteristics of the interface of soil bags. The test results show that the slip surface in the soil bags-constructed retaining wall is ladder-like due to the inter-layer insertion of soil bags, and the wall above the ladder-like surface undergoes a rigid body translation. The insertion strengthens the shear strength of two-layer staggered-stacked soil bags. Meanwhile, it affects the shape of the slip surface of the five-layer staggered-stacked soil bags. Finally, the interlayer resisting friction of soil bags is found to be related to the shape of the slip surface. Keywords: geosynthetics, retaining wall, soil bag, failure mode, interface, shear strength

Authors: Junichiro Kawaguchi
Abstract:
Space debris problems have emerged and threatened the use of low earth orbit around the Earth owing to a large number of spacecraft. In debris removal, a number of research and patents have been proposed and published so far. They assume servicing spacecraft, robots to be built for accessing the target debris objects. The robots should be sophisticated enough automatically to access the debris articulating the attitude and the translation motion with respect to the debris. This paper presents the idea of using the torpedo-like third unsophisticated and disposable body, in addition to the first body of the servicing robot and the second body of the target debris. The third body is launched from the first body from a distance farer than the size of the second body. This paper presents the method and the system, so that the third body is launched from the first body. The third body carries both a net and an inflatable or extendible drag deceleration device and is built small and light. This method enables even a micro to nano satellite to perform contactless and multiple debris removal even via a single flight. Keywords: ballute, debris removal, echo satellite, gossamer, gun-net, inflatable space structure, small satellite, un-cooperated target

Authors: Ondrej Slach, Jan Machacek, Jan Zenka, Lucie Hyllova, Petr Rumpel
Abstract:
The aim of the paper is to identify and asses different trajectories of two fashionable urban policies –creative and smart cities– in specific post-socialistic context. Drawing on the case of Czechia, we employ the concept of policy mobility research. More specifically, we employ a discourse analysis in order to identify the so-called 'infrastructure' of both policies (such as principal actors, journals, conferences, events), with the special focus on 'agents of transfer' in a multiscale perspective. The preliminary results indicate faster and more aggressive spatial penetration of smart cities policy compared to creative cities policy in Czechia. Further, it seems that existed translation and implementation of smart cities policy into the national and urban context resulted in deliberated fragmented policy of smart cities in Czechia (pure technocratic view), which might be a threat for the future development of social sustainability, especially in cities that are facing increasing social polarisation. Last but not least, due to the fast spatial penetration of the concept and policies of smart cities, it seems that creative cities policy has almost been crowded out of the Czech urban agenda. Keywords: policy mobility, smart cities, creative cities, Czechia

Authors: Bhuyain Mobarok Hossain
Abstract:
Image classification and localization research is currently an important strategy in the field of computer vision. The evolution and advancement of deep learning and convolutional neural networks (CNN) have greatly improved the capabilities of object detection and image-based classification. Target detection is important to research in the field of computer vision, especially in video surveillance systems. To solve this problem, we will be applying a convolutional neural network of multiple scales at multiple locations in the image in one sliding window. Most translation networks move away from the bounding box around the area of interest. In contrast to this architecture, we consider the problem to be a classification problem where each pixel of the image is a separate section. Image classification is the method of predicting an individual category or specifying by a shoal of data points. Image classification is a part of the classification problem, including any labels throughout the image. The image can be classified as a day or night shot. Or, likewise, images of cars and motorbikes will be automatically placed in their collection. The deep learning of image classification generally includes convolutional layers; the invention of it is referred to as a convolutional neural network (CNN).Keywords: image classification, object detection, localization, particle filter

Type of study: Observational cross section study. Background and purpose: Forward head posture (FHP) is a common sagittal faulty posture with anterior head translation relative to vertical posture line. FHP related to temporomandibular joint dysfunctions, neck pain and headache. Sports persons usually overuse one side of the body in training and playing leading to postural imbalance, yet the effect of playing football or bodybuilding on measurements of FHP has never been studied. Participants: Thirty six subjects divided into 3 groups of 12 football players, 12 body builders and 12 students. Method: FHP severity was assessed by measuring the craniovertebral (CVA) and gaze angles, using the photogrammetric method. Photos were taken from right side of subjects while assuming standing position. Analysis of variance was used to assess angles difference between the three groups. Results: No significant differences were found in CVA and gaze angles between the three groups (P > 0.05). Conclusion: Playing football or body building doesn't impose significant FHP.Keywords: craniovertebral angle, gaze angle, football, body building
Procedia PDF Downloads 416189 Advances in Axonal Biomechanics and Mechanobiology: A Nanotechnology-Based Approach to the Study of Mechanotransduction of Axonal Growth
Authors: Alessandro Falconieri, Sara De Vincentiis, Vittoria Raffa
Abstract:
Mechanical force regulates axonal growth, elongation and maturation processes. This force is opening new frontiers in the field, contributing to a general understanding of the mechanisms of axon growth that, in the past, was thought to be governed exclusively by the growth cone and its ability to influence axonal growth in response to chemical signals. A method recently developed in our laboratory allows, through the labeling of neurons with magnetic nanoparticles (MNPs) and the use of permanent magnets, to apply extremely low mechanical forces, similar to those generated endogenously by the growth cone or by the increase of body mass during the organism growth. We found that these extremely low forces strongly enhance the spontaneous axonal elongation rate as well as neuronal sprouting. Data obtained don’t exclude that local phenomena, such as local transport and local translation, may be involved. These new advances could shed new light on what happens when the cell is subjected to external mechanical forces, opening new interesting scenarios in the field of mechanobiology. Keywords: axon, external mechanical forces, magnetic nanoparticles, mechanotransduction
Procedia PDF Downloads 122188 Perceptions of Senior Academics in Teacher Education Colleges Regarding the Integration of Digital Games during the Pandemic
Authors: Merav Hayakac, Orit Avidov-Ungarab
Abstract:
The current study adopted an interpretive-constructivist approach to examine how senior academics from a large sample of Israeli teacher education colleges serving general or religious populations perceived the integration of digital games into their teacher instruction and what their policy and vision were in this regard in the context of the COVID-19 pandemic. Half the participants expressed a desire to integrate digital games into their teaching and learning but acknowledged that this practice was uncommon. Only a small minority believed they had achieved successful integration, with doubt and skepticism expressed by some religious colleges. Most colleges had policies encouraging technology integration supported by ongoing funding. Although a considerable gap between policy and implementation remained, the COVID-19 pandemic was viewed as having accelerated the integration of digital games into pre-service teacher instruction. The findings suggest that discussions around technology-related vision and policy and their translation into practice should relate to the specific cultural needs and academic preparedness of the population(s) served by the college. Keywords: COVID-19, digital games, pedagogy, teacher education colleges

Authors: Hami Ashraf, Farid Kosari
Abstract:
Graft-versus-host disease (GvHD) is a common complication of allogeneic hematopoietic stem cell transplantation that can lead to significant morbidity and mortality. Histopathological examination of affected tissues is an essential tool for diagnosing and grading GvHD in animal models, which are used to study disease mechanisms and evaluate new therapies. In this systematic review, we identified and analyzed original research articles in PubMed, Scopus, Web of Science, and Google Scholar that described grading systems for GvHD in animal models based on histopathological features. We found that several grading systems have been developed, which vary in the tissues and criteria they assess, the severity scoring scales they use, and the level of detail they provide. Skin, liver, and gut are the most commonly evaluated tissues, but lung and thymus are also included in some systems. Our analysis highlights the need for standardized criteria and consistent use of grading systems to enable comparisons between studies and facilitate the translation of preclinical findings to clinical practice. Keywords: graft-versus-host disease, GvHD, animal model, histopathology, grading system

Authors: Khaing Su Su Than, Hibino Yo
Abstract:
Seismic evaluation is needed upon the buildings in Myanmar. Microtremor measurement was conducted in the main cities, Mandalay and Yangon. In order to evaluate the seismic properties of buildings currently under construction, seismic information was gathered for six buildings in Yangon city and four buildings in Mandalay city. The investigated buildings vary from 12m-80 m in height, and mostly public residence structures. The predominant period obtained from frequency results of the investigated buildings were given by horizontal to vertical spectral ratio (HVSR) for each building. The fundamental period results have been calculated in the form of Fourier amplitude spectra of translation along with the main structure. Based on that, the height (H)-period(T) relationship was observed as T=0.012H-0.017H in the buildings of Yangon and, observed the relationship as T=0.014H-0.019H in the buildings of Mandalay. The results showed that the relationship between height and natural period was slightly under the relationship T=0.02H that is used for Japanese reinforced concrete buildings, which indicated that the results depend on the properties and characteristics of materials used. Keywords: HVSR, height-period relationship, microtremor, Myanmar earthquake, reinforced concrete structures
Procedia PDF Downloads 156182 Corporate Social Responsibility vs Corporate Social Reactivity: An Exploration of Corporate Social Responsibility Planning in a Multinational Oil and Gas in Indonesia
Authors: Endang Ghani Ashfiya
Abstract:
This study explores corporate social responsibility (CSR) planning in a downstream business of multinational oil and gas company in Indonesia from managerial perspectives. The institutional logic is employed in this research to gain a comprehensive understanding of the way the MNC manages the socio-cultural aspects in the host countries, especially in the process of translation and adaptation of the company’s CSR global guidelines. The interviews are conducted with fifteen managers in that company, both at the top managerial level and operational level. In the beginning, this research explains the Indonesian society’s conception of CSR from the managerial standpoints. The society’s understanding of the CSR concept becomes the fundamental foundations of the company in developing CSR programs. This study found the company’s approach to its CSR in two ways. First, proactive CSR which reflects the global CSR guidelines. Second, reactive CSR which do not show any explicit relations to the global guidelines, but conform with society’s demands. The findings stimulate discussions regarding the power of an MNC vis-à-vis the socio-cultural implication in society’s demand for CSR.Keywords: corporate social responsibility planning, Indonesia, institutional logic, multinational company, oil and gas company, socio-cultural aspects

Authors: Sanjay Kumar, Lillie Dewan
Abstract:
The study of complex dynamic systems has advanced through various scientific approaches with the help of computer modeling. The common design trends in aerospace system design can be applied to quadcopter design. A quadcopter is a nonlinear, under-actuated system with complex aerodynamics parameters and creates challenges that demand new, robust, and effective control approaches. The flight control stability can be improved by planning and tracking the trajectory and reducing the effect of sensors and the operational environment. This paper presents a modern design Simmechanics visual modeling approach for a mechanical model of a quadcopter with three degrees of freedom. The Simmechanics model, considering inertia, mass, and geometric properties of a dynamic system, produces multiple translation and rotation maneuvers. The proportional, integral, and derivative (PID) controller is integrated with the Simmechanics model to follow a predefined quadcopter rotational trajectory for a fixed time interval. The results presented are satisfying. The simulation of the quadcopter control performed operations successfully. Keywords: nonlinear system, quadcopter model, simscape modelling, proportional-integral-derivative controller

Authors: Mingshu Liu, Laura Santamaria, Xavier Carmaniu Mainadé
Abstract:
As one of the sectors most affected by the COVID-19 pandemic, tourism is facing challenges in revitalizing the industry. But at the same time, it would be a good opportunity to take advantage of digital communication as an effective tool for tourism promotion. Our proposal aims to verify the linguistic operations on online platforms in China. The research is carried out based on the theory of Eco-traductology put forward by Gengshen Hu, whose contribution focuses on the translator's adaptation to the ecosystem environment and the three elaborated parameters (linguistic, cultural and communicative). We also relate it to Even-Zohar's and Toury's theoretical postulates on the Polysystem to elaborate on interdisciplinary methodology. Such a methodology allows us to analyze personal treatments and phraseology in the target text. As for the corpus, we adopt the official Spanish-language website of Turismo de España as the source text and the postings on the two major social networks in China, Weibo and Wechat, in 2019. Through qualitative analysis, we conclude that, in the tourism advertising campaign on Chinese social networks, chengyu (Chinese phraseology) and honorific titles are used very frequently. Keywords: digital communication, eco-traductology, polysystem theory, tourism advertising

Authors: Alexandra Cheira
Abstract:
Cultural appropriation has been theorised as a form of colonialism in which members of a dominant culture reduce cultural elements that are deeply meaningful to a minority culture to the category of the “exotic other” since they do not experience the oppression and discriminations faced by members of the minority culture. Yet, in the particular case of literature, writers such as Lionel Shriver and Bernardine Evaristo have argued that authors from a cultural majority have a right to write in the voice of someone from a cultural minority, hence attacking the idea that this is a form of cultural appropriation. By definition, Shriver and Evaristo claim, writers are supposed to write beyond their own culture, gender, class, and/ or race. In this light, this paper discusses the limits of cultural appropriation vis-à-vis the ethics of narrative empathy by addressing the mixed critical reception of Kathryn Stockett’s The Help (2009) and Jeanine Cummins’s American Dirt (2020). In fact, both novels were acclaimed as global eye-openers regarding the struggles of respectively South American migrants and African American maids. At the same time, both novelists have been accused of cultural appropriation by telling a story that is not theirs to tell, given the fact that they are white women telling these stories in what critics have argued is really an American voice telling a story to American readers. These claims will be investigated within the framework of Edward Said’s foundational examination of Orientalism in the field of postcolonial studies as a Western style for authoritatively restructuring the Orient. This means that Orientalist stereotypes regarding Eastern cultures have implicitly validated colonial and imperial pursuits, in the specific context of literary representations of African American and Mexican cultures by white writers. At the same time, the conflicted reception of American Dirt and The Help will be examined within the critical framework of narrative empathy as theorised by Suzanne Keen. Hence, there will be a particular focus on the way a reader’s heated perception that the author’s perspective is purely dishonest can result from a friction between an author’s intention and a reader’s experience of narrative empathy, while a shared sense of empathy between authors and readers can be a rousing momentum to move beyond literary response to social action. Finally, in order to assess that “the key question should not be who can write about whom, but how the writer tells the story”, the recent controversy surrounding Dutch author Marieke Lucas Rijneveld’s decision to resign the translation of American poet Amanda Gorman’s work into Dutch will be duly investigated. In fact, Rijneveld stepped out after journalist and activist Janice Deul criticised Dutch publisher Meulenhoff for choosing a translator who was not also Black, despite the fact that 22-year-old Gorman had selected the 29-year-old Rijneveld herself, as a fellow young writer who had likewise come to fame early on in life. In this light, the critical argument that the controversial reception of The Help reveals as much about US race relations in the early twenty-first century as about the complex literary transactions between individual readers and the novel itself will also be discussed in the extended context of American Dirt and white author Marieke Rijneveld’s withdrawal from the projected translation of Black poet Amanda Gorman. Keywords: cultural appropriation, cultural stereotypes, narrative empathy, race relations

Abstract:
SRSF1 governs splicing of over 1,500 mRNA transcripts. SRSF1 contains two RNA-recognition motifs (RRMs) and a C-terminal Arg/Ser-rich region (RS). It has been thought that SRSF1 RRMs exclusively recognize single-stranded exonic splicing enhancers, while RS lacks RNA-binding specificity. With our success in solving the insolubility problem of SRSF1, we can explore the unknown RNA-binding landscape of SRSF1. We find that SRSF1 RS prefers purine over pyrimidine. Moreover, SRSF1 binds to the G-quadruplex (GQ) from the ARPC2 mRNA, with both RRMs and RS being crucial. Our binding assays show that the traditional RNA-binding sites on the RRM tandem and the Arg in RS are responsible for GQ binding. Interestingly, our FRET and circular dichroism data reveal that SRSF1 unfolds the ARPC2 GQ, with RS leading unfolding and RRMs aiding. Our saturation transfer difference NMR results discover that Arg residues in SRSF1 RS interact with the guanine base but with other nucleobases, underscoring the uniqueness of the Arg/guanine interaction. Our luciferase assays confirm that SRSF1 can alleviate the inhibitory effect of GQ on gene expression in the cell. Given the prevalence of RNA GQ and SR proteins, our findings unveil unexplored SR protein functions with broad implications in RNA splicing and translation. Keywords: SR, SRSF!, RNA G-quadruplex, unfolding, RNA binding

Authors: Debora Gottardello, Mireia Valverde Aparicio, Juan Llopis Taverner
Abstract:
Globalization has given rise to a great diversity in the composition of people in organizations. Diversity management is therefore key to create growth in today’s competitive global marketplace. This work develops a literature review related to the existing models for managing diversity covering the period from 1980 until 2014. Furthermore, it identifies limitations in previous models. More specifically, the literature review reveals that there is a lack of information about how these models can be adapted from the headquarters to the subsidiaries. Therefore, the contribution of this paper is to suggest how the models should be adapted when they are directed to host countries. Our aim is to highlight the limitations of the developed models with regards to the translation of the diversity management practices to the subsidiaries. Accordingly, a model that will enable MNCs to ensure a global strategy is suggested. Taking advantage of the potential incorporated in a culturally diverse work team should be at the top of every international company’s aims. Executives from headquarters need to use different attitudes when transferring diversity practices towards their subsidiaries. Further studies should reassess local practices of diversity management to find out how this universal management model is translated. Keywords: culture diversity, diversity management, human resources management, MNCs, subsidiaries, workforce diversity

Authors: Hadjoui Abdelhamid, Saimi Ahmed
Abstract:
The hybrid h-p finite element method for the dynamic behavior of nonlinear rotors is described in this paper. The standard h-version method of discretizing the problem is retained, but modified to allow the use of polynomially-enriched beam elements. A hierarchically enriching element will thus not affect the nodal displacement and rotation, but will influence the values of the nodal bending moment and shear force is used. The deterministic movements of rotation and translation of the support which are coupled to the excitations due to unbalance are also taken into account. We study also the geometric dissymmetry of the shaft and the disc, thus the equations of motion of the rotor contain variable parametric coefficients over time that can lead to a lateral dynamic instability. The effects of movements combined support for bearings are analyzed and discussed through Campbell diagrams and spectral analyses. A program is made in Matlab. After validation of the program, several examples are studied. The influence of physical and geometric parameters on the natural frequencies of the shaft is determined through the study of these examples. Among these parameters, we include the variation in the diameter and the thickness of the rotor, the position of the disc. Keywords: Campbell diagram, critical speeds, nonlinear rotor, version h-p of FEM
