36 The Effectiveness of Tebuconazole and Chitosan in Inhibiting the Growth of Fusarium Species on Winter Wheat Grain under Field Conditions
Authors: Urszula Wachowska, Anna Daria Stasiulewicz-Paluch, Katarzyna Kucharska
Abstract:
A three-year field experiment (2010-2012) was conducted to determine the abundance of epiphytic and endophytic filamentous fungi colonizing the grain of winter wheat cv. Bogatka. Wheat spikes were protected with tebuconazole or chitosan at the watery ripe stage. Untreated plants served as control. Tebuconazole exerted an inhibitory effect primarily on F. culmorum and F. graminearum, and its effectiveness was determined by the pressure from pathogens that infected wheat spikes during the growing season. Chitosan did not suppress the growth of Fusarium species and Alternaria alternata.

Authors: F. Nejati Hafdani, N. Sadeghinia
Abstract:
In recent years application of natural antimicrobials instead of conventional ones, due to their hazardous effects on health, has got serious attentions. On the basis of the results of different studies, chitosan, a natural bio-degradable and non-toxic biopolysaccharide derived from chitin, has potential to be used as a natural antimicrobial. Chitosan has exhibited high antimicrobial activity against a wide variety of pathogenic and spoilage microorganisms, including fungi, and Gram-positive and Gramnegative bacteria. The antimicrobial action is influenced by intrinsic factors such as the type of chitosan, the degree of chitosan polymerization and extrinsic factors such as the microbial organism, the environmental conditions and presence of the other components. The use of chitosan in food systems should be based on sufficient knowledge of the complex mechanisms of its antimicrobial mode of action. In this article we review a number of studies on the investigation of chitosan antimicrobial properties and application of them in culture and food mediums. Keywords: Antimicrobial, Chitosan, Preservative

Abstract:
The spores of entomopathogenic fungi, Beauveria bassiana was evaluated for their compatibility with four surfactants; SDS (sodium dodyl sulphate) and CABS-65 (calcium alkyl benzene sulphonate), Tween 20 (polyethylene sorbitan monolaureate) and Tween 80 (polyoxyethylene sorbitan monoleate) at six different concentrations (0.1%, 0.5%, 1%, 2.5%, 5% and 10%). Incubated spores showed decrease in concentrations due to conversion of spores to hyphae. The maximum germination recorded in 72 h incubated spores varied with surfactant concentration at 49-68% (SDS), 39- 53% (CABS), 78-92% (Tween 80) and 80-92% (Tween 20), while the optimal surfactant concentration for spore germination was found to be 2.5-5%. The surfactant effect on spores was more pronounced with SDS and CABS-65, where significant deterioration and loss in viability of the incubated spores was observed. The effect of Tween 20 and Tween 80 were comparatively less inhibiting. The results of the study would help in surfactant selection for B. bassiana emulsion preparation.

Authors: M. Grabowski, K. Macnar, J. Skrzyński
Abstract:
Huge losses in apple production are caused by pathogens that cannot be seen shortly after harvest. After-harvest thermotherapy treatments can considerably improve control of storage diseases on apples and become an alternative to chemical pesticides. In the years 2010-2012 carried out research in this area. Apples of 'Topaz' cultivar were harvested at optimal maturity time for long storage and subject to water bath treatment at 45, 50, 52, 55°C for 60, 120, 180 and 240 seconds. The control was untreated fruits. After 12 and 24 weeks and during so called simulated trade turnover the fruits were checked for their condition and the originators of diseases were determined by using the standard phytopathological methods. The most common originator of 'Topaz' apple infection during storage were the fungi of genus Gloeosporium. In this paper it was proven that for effective protection of 'Topaz' apples against diseases, thermotherapy by using water treatments at temperature range of 50-52°C is quite sufficient.

Abstract:
Wood decay is addressed continuously within the wood industry through use and development of wood preservatives. The increasing awareness on the negative effects of many chemicals towards the environment is causing political restrictions in their use and creating more urgent need for research on green alternatives. This paper discusses some of the possible natural extracts for wood preserving applications and compares the analytical methods available for testing their behavior and efficiency against decay fungi. The results indicate that natural extracts have interesting chemical constituents that delay fungal growth but vary in efficiency depending on the chemical concentration and substrate used. Results also suggest that presence and redistribution of preservatives in wood during exposure trials can be assessed by spectral imaging methods although standardized methods are not available. This study concludes that, in addition to the many standard methods available, there is a need to develop new faster methods for screening potential preservative formulation while maintaining the comparability and relevance of results.

Abstract:
Mushrooms or macro-fungi, as an important superfood, contain many bioactive compounds, particularly bio-peptides. In this research, mushroom proteins were extracted by buffer or buffer plus salt (0.15 M), along with ultrasound bath to extract the intercellular protein. As a result, the highest amount of proteins in mushrooms were categorized into albumin. Proteins were also hydrolyzed and changed into peptides through endogenous and exogenous proteases, including gastrointestinal enzymes. The potency of endogenous proteases was also higher in Agaricus bisporus than Terfezia claveryi, as their activity ended at 75 for 15 min. The blanching process, endogenous enzymes, the mixture of gastrointestinal enzymes (pepsin-trypsin-α-chymotrypsin or trypsin- α-chymotrypsin) produced the different antioxidant and antibacterial hydrolysates. The peptide fractions produced with different cut-off ultrafilters also had various levels of radical scavenging, lipid peroxidation inhibition, and antibacterial activities. The bio-peptides with the superior bio-activities (less than 3 kD of T. claveryi) were resistance to various environmental conditions (pH and temperatures). Therefore, they are good options to be added in nutraceutical and pharmaceutical preparations or functional foods, even during processing.

Abstract:
This study was conducted to evaluate the effect of Aspergillus species on acridid populations which are major agricultural pests of rice, sugarcane, wheat, maize and fodder crops in Pakistan. Three and replicates i.e. Aspergillus flavus, A. fumigatus and A. niger, excluding the control, were held under laboratory conditions. It was observed that consumption faecal production of acridids was significantly reduced after the pathogenic application of Aspergillus. In the control replicate, the mortality ratio for stage (N4-N6) was maximum on day 2nd i.e. [F10.7 = 18.33, P < 0.05] followed by [F4.20 = 07.85, P < 0.05] and [F3.77 = 06.11, P < 0.05] on 4th and 3rd day, respectively. Similarly, it was a minimum i.e. [F0.48 = 84.65, P < 0.05] on the 1st day. It was also noted that faecal production of Acridid nymphs was not significantly affected when treated with conidial concentration in H2O formulation; however, it was significantly reduced after the contamination with conidial concentration in oil. The high morality of acridids after contamination of Aspergillus supports their use as bio-control agent for reducing pest population. The present study recommends that exploration and screening must be conducted to provide additional pathogens for evaluation as potential biological control against grasshoppers and locusts.

Abstract:
Anaerobic digestion process is one of the alternative methods to convert organic waste into methane gas which is a fuel and energy source. Activities of various kinds of microorganisms are the main factor for anaerobic digestion which produces methane gas. Therefore, in this study a modified Anaerobic Baffled Reactor (ABR) with working volume of 50 liters was designed to identify the microorganisms through biogas production. The mixture of 75% kitchen waste and 25% sewage sludge was used as substrate. Observations on microorganisms in the ABR showed that there exists a small amount of protozoa (5%) and fungi (2%) in the system, but almost 93% of the microorganism population consists of bacteria. It is definitely clear that bacteria are responsible for anaerobic biodegradation of kitchen waste. Results show that in the acidification zone of the ABR (front compartments of reactor) fast growing bacteria capable of growth at high substrate levels and reduced pH was dominant. A shift to slower growing scavenging bacteria that grow better at higher pH was occurring towards the end of the reactor. Due to the ability of activity in acetate environment the percentages of Methanococcus, Methanosarcina and Methanotrix were higher than other kinds of methane former in the system. Keywords: Anaerobic microorganism identification, Kitchenwaste, Biogas.

They are highly nutritious with good amount of quality proteins, vitamins and minerals. An edible mushroom, Calocybe indica was selected to validate its nutritional and medicinal properties. Since tissue damage in hyperglycemia has been related to oxidative stress, we evaluated the enzymatic and non-enzymatic antioxidant status in the serum, liver and kidney since they are the target organs in diabetic complications. From the results, increased oxidative stress and decreased antioxidants might be related to the causation of diabetes mellitus. The treatment in the diabetic rats with the Calocybe indica showed an increase in the antioxidant system and decrease in the production of free radicals. Abstract:
Mycotoxins are secondary metabolic products of fungi. These are poisonous, carcinogens and mutagens in nature and pose a serious health threat to both humans and animals, causing severe illnesses and even deaths. The rapid, simple and cheap detection methods of mycotoxins are of immense importance and in great demand in the food and beverage industry as well as in agriculture and environmental monitoring. Lateral flow immunochromatographic strips (ICSTs) have been widely used in food safety, environment monitoring. 46 papers were identified and reviewed on Google Scholar and Scopus for their limit of detection and nanomaterial on Lateral flow ICSTs on different types of mycotoxins. The papers were dated 2001-2021. 25 papers were compared to identify the lowest limit of detection of among different mycotoxins (Aflatoxin B1: 10, Zearalenone: 5, Fumonisin B1: 5, Trichothecene-A: 5). Most of these highly sensitive strips are competitive. Sandwich structures are usually used in large scale detection. In conclusion, the limit of detection of Aflatoxin B1 is the lowest among these mycotoxins. Gold-nanoparticle based immunochromatographic test strips have the lowest limit of detection. Five papers involve smartphone detection and they all detect aflatoxin B1 with gold nanoparticles.

Abstract:
Phytases are enzymes used as an important component in monogastric animals feeds in order to improve phosphorous availability, since it is not readily assimilated by these animals in the form of the phytate presented in plants and grains. As these enzymes are used in industrial activities, they must retain its catalytic activities during a certain storage period. This study presents information about the stability of 4 different phytases, produced by four macromycetes fungi through solid-state fermentation (SSF). There is a lack of data in literature concerning phytase from macromycetes shelf-life in storage conditions at room, cooling and freezing temperatures. The 4 phytases from macromycetes still had enzymatic activities around 100 days of storage at room temperature. At cooling temperature in 146 days of studies, the phytase from G. stipitatum was the most stable with 44% of the initial activity, in U.gds (units per gram of dried fermented substrate). The freezing temperature was the best condition storage for phytases from G. stipitatum and T. versicolor. Each condition provided a study for each mushroom phytase, totalizing 12 studies. Authors: R. Weinzierl, R. Estes, N. Tinsley, M. Keshlaf
Abstract:
The corn earworm, Helicoverpa zea Boddie, is a serious pest of corn. Larval feeding in ear tips destroys kernels and allows growth of fungi and production of mycotoxins. Infested sweet corn is not marketable. Development of improved transgenic hybrids expressing insecticidal toxins from Bacillus thuringiensis (Bt) may limit or prevent crop losses. The effectiveness of Attribute® II Bt resistance and applications of Voliam Xpress insecticide were evaluated for effectiveness in controlling corn earworm in plots near Urbana, IL, USA, in 2013. Where no insecticides were applied, ear infestations and kernel damage in Attribute® II ‘Protector’ plots were consistently lower (near zero) than in plots of the non-Bt isoline ‘Garrison.’ Multiple applications of Voliam Xpress significantly reduced the number of corn earworm larvae and kernel damage in the Garrison plots, but infestations and damage in these plots were greater than in Protectorplots that did not receive insecticide applications. Our results indicate that Attribute® II Bt resistance is more effective than multiple applications of an insecticide for preventing losses caused by corn earworm in sweet corn.

Abstract:
Nowadays there is a growing interest in biofuel production in most countries because of the increasing concerns about hydrocarbon fuel shortage and global climate changes, also for enhancing agricultural economy and producing local needs for transportation fuel. Ethanol can be produced from biomass by the hydrolysis and sugar fermentation processes. In this study ethanol was produced without using expensive commercial enzymes from sugarcane bagasse. Alkali pretreatment was used to prepare biomass before enzymatic hydrolysis. The comparison between NaOH, KOH and Ca(OH)2 shows NaOH is more effective on bagasse. The required enzymes for biomass hydrolysis were produced from sugarcane solid state fermentation via two fungi: Trichoderma longibrachiatum and Aspergillus niger. The results show that the produced enzyme solution via A. niger has functioned better than T. longibrachiatum. Ethanol was produced by simultaneous saccharification and fermentation (SSF) with crude enzyme solution from T. longibrachiatum and Saccharomyces cerevisiae yeast. To evaluate this procedure, SSF of pretreated bagasse was also done using Celluclast 1.5L by Novozymes. The yield of ethanol production by commercial enzyme and produced enzyme solution via T. longibrachiatum was 81% and 50% respectively.

Abstract:
Enzymes are the biocatalysts which catalyze the biochemical processes and thus have a wide variety of applications in the industrial sector. β-Galactosidase (E.C. 3.2.1.23) also known as lactase, is one of the prime enzymes, which has significant potential in the dairy and food processing industries. It has the capability to catalyze both the hydrolytic reaction for the production of lactose hydrolyzed milk and transgalactosylation reaction for the synthesis of prebiotics such as lactulose and galactooligosaccharides. These prebiotics have various nutritional and technological benefits. Although, the enzyme is naturally present in almonds, peaches, apricots and other variety of fruits and animals, the extraction of enzyme from these sources increases the cost of enzyme. Therefore, focus has been shifted towards the production of low cost enzyme from the microorganisms such as bacteria, yeast and fungi. As compared to yeast and bacteria, fungal β-galactosidase is generally preferred as being extracellular and thermostable in nature. Keeping the above in view, the present study was carried out for the isolation of the β-galactosidase producing fungal strain from the food as well as the agricultural wastes. A total of more than 100 fungal cultures were examined for their potential in enzyme production. All the fungal strains were screened using X-gal and IPTG as inducers in the modified Czapek Dox Agar medium. Among the various isolated fungal strains, the strain exhibiting the highest enzyme activity was chosen for further phenotypic and genotypic characterization. The strain was identified as Rhizomucor pusillus on the basis of 5.8s RNA gene sequencing data.

Abstract:
The quality and condition of perishable products delivered to the market and their subsequent selling prices are directly affected by the care taken during harvesting and handling. Mechanical injury, in fact, occurs at all stages, from pre-harvest operations through post-harvest handling, packing and transport to the market. The main implications of this damage are the reduction of the product’s quality and economical losses related to the shelf life diminution. For most perishable products, the shelf life is relatively short and it is typically dictated by microbial growth related to the application of dynamic and static loads during transportation. This paper presents the correlation between vibration levels and microbiological growth on strawberries and woodland strawberries and detects the presence of volatile organic compounds (VOC) in order to develop an intelligent logistic unit capable of monitoring VOCs using a specific sensor system. Fresh fruits were exposed to vibrations by means of a vibrating table in a temperature-controlled environment. Microbiological analyses were conducted on samples, taken at different positions along the column of the crates. The values obtained were compared with control samples not exposed to vibrations and the results show that different positions along the column influence the development of bacteria, yeasts and filamentous fungi.

Abstract:
The present study has been taken to explore the screening of in vitro antimicrobial activities of D-galactose-binding sponge lectin (HOL-30). HOL-30 was purified from the marine demosponge Halichondria okadai by affinity chromatography. The molecular mass of the lectin was determined to be 30 kDa with a single polypeptide by SDS-PAGE under non-reducing and reducing conditions. HOL-30 agglutinated trypsinized and glutaraldehydefixed rabbit and human erythrocytes with preference for type O erythrocytes. The lectin was subjected to evaluation for inhibition of microbial growth by the disc diffusion method against eleven human pathogenic gram-positive and gram-negative bacteria. The lectin exhibited strong antibacterial activity against gram-positive bacteria, such as Bacillus megaterium and Bacillus subtilis. However, it did not affect against gram-negative bacteria such as Salmonella typhi and Escherichia coli. The largest zone of inhibition was recorded of Bacillus megaterium (12 in diameter) and Bacillus subtilis (10 mm in diameter) at a concentration of the lectin (250 μg/disc). On the other hand, the antifungal activity of the lectin was investigated against six phytopathogenic fungi based on food poisoning technique. The lectin has shown maximum inhibition (22.83%) of mycelial growth of Botrydiplodia theobromae at a concentration of 100 μg/mL media. These findings indicate that the lectin may be of importance to clinical microbiology and have therapeutic applications. Keywords: Antibacterial, Halichondria okadai, Inhibition zone, Lectin.

Abstract:
Eukaryotic protein-coding genes are interrupted by spliceosomal introns, which are removed from the RNA transcripts before translation into a protein. The exon-intron structures of different eukaryotic species are quite different from each other, and the evolution of such structures raises many questions. We try to address some of these questions using statistical analysis of whole genomes. We go through all the protein-coding genes in a genome and study correlations between the net length of all the exons in a gene, the number of the exons, and the average length of an exon. We also take average values of these features for each chromosome and study correlations between those averages on the chromosomal level. Our data show universal features of exon-intron structures common to animals, plants, and protists (specifically, Arabidopsis thaliana, Caenorhabditis elegans, Drosophila melanogaster, Cryptococcus neoformans, Homo sapiens, Mus musculus, Oryza sativa, and Plasmodium falciparum). We have verified linear correlation between the number of exons in a gene and the length of a protein coded by the gene, while the protein length increases in proportion to the number of exons. On the other hand, the average length of an exon always decreases with the number of exons. Finally, chromosome clustering based on average chromosome properties and parameters of linear regression between the number of exons in a gene and the net length of those exons demonstrates that these average chromosome properties are genome-specific features.

Authors: Ashraf S. Hakim, Randa M. Alarousy
Abstract:
The consumption of food contaminated with molds (microscopic filamentous fungi) and their toxic metabolites results in the development of food-borne mycotoxicosis. The spores of molds are ubiquitously spread in the environment and can be detected everywhere. Ochratoxin A is a toxic and potentially carcinogenic fungal toxin found in a variety of food commodities. In this study, the mycological quality of various ready-to-eat local and imported pork meat and meat byproducts sold in Egyptian markets were assessed and the presence of various molds was determined in pork used as a raw material, edible organs as liver and kidney as well as in fermented raw meat by-products. The study assessed the mycological quality of pork raw meat and their by-products sold in commercial shops in Cairo, Egypt. Mycological analysis was conducted on (n=110) samples which included pig’s livers and kidneys from Egyptian Bassatin slaughter house; local and imported processed pork meat by-products from Egyptian pork markets. The isolates were identified using traditional mycological and biochemical tests. All kidney and liver samples were positive to molds growth while all byproducts were negative. Ochratoxin A levels were quantitatively analyzed using the high performance liquid chromatography (HPLC) and the highest results were present in kidney 7.51 part per billion (ppb) followed by minced meat 6.19 ppb generally the local samples showed higher levels than the imported ones. To the best of our knowledge, this is the first report on mycotoxins detection and quantification from pork by-products in Egypt. Keywords: Egypt, imported pork by-products, local, mycotoxins.

Authors: N. S. El-Mougy, M. M. Abdel-Kader, H. M. Abouelnasr
Abstract:
The efficacy of two essential oils applied as a bean seed dressing followed by seedlings foliar spray with four commercial disinfectants against root rot and wilt incidence was evaluated under field conditions. The essential oils, grape seed and peppermint oils and the disinfectants, Gold, Revarest, Klenva, Malva were applied. Chitosan and the fungicide Topsin-M were used as comparison treatment. Essential oils individually or combined with disinfectants were applied as a bean seed dressing. Furthermore, emerged bean plants were sprayed with the same treatments. Under laboratory conditions, growth inhibition effect was observed for the isolated, tested fungi R. solani and F. oxysporum when exposed to essential oils individually or combined with disinfectants. A high inhibitor effect was recorded for peppermint followed by grape seed oils. Concentrations of 1% and 2% of chitosan as well as Topsin M at 400 ppm showed complete reduction (100%) in the two fungal growths. Under field conditions, the obtained results showed that the applied treatments of chitosan had a superior effect on root rot and wilt disease incidence compared with other tested treatments. It was found that seed coating treatment provides good protection of emerged green bean seeds against the root pathogens attack compared with the fungicide and control treatments. Also, the application of seed dressing with essential oils accompanied by seedling spray demonstrated similar results. It was observed that essential oils had an enhancing effect against disease incidence when combined with disinfectants compared with their application. The obvious yield increase was significantly higher in all applied treatments than in fungicide and control.
