5307 Antifungal Protein ~35kDa Produced by Bacillus cereus Inhibits the Growth of Some Molds and Yeasts

Abstract:

An antifungal protein synthesized by Bacillus cereus has been partially purified by the use of ammonium sulfate precipitation and Sephadex-G-200 column chromatography. The protein was produced from Bacillus cereus grown in potato Dextrose Broth Medium (PDB) at 30 ºC for 3 days at 100 rpm. The protein showed antagonistic effect against some fungi and yeasts. Crude extract from medium and semi-purified protein were tested in vitro against both fungi and yeasts using the disc diffusion method in order to detect the inhibitory effect of the protein. Zones of inhibition of the following diameter were found (mm) were Alternaria alternate (28), Rhodotorula glutinis (20), Fusarium sp. (16), Rhizopus sp. (15), Penicillium digitatum (13), Mucor sp. (13) and Aspergillus niger (10). The isolated protein was found to have a molecular weight of ~35kDa by sodium deodecyl sulfate-poly acrylamide gel electrophoresis. The data showed that the protein of Bacillus cereus has antifungal activity, a fact which points to the possibility of using it as a bio-control agent against some fungi, findings which emphasize the potential role of B. cereus as an important bio-control agent.

5306 Experimental Study of Heat Transfer and Pressure Drop in Serpentine Channel Water Cooler Heat Sink

Abstract:

With the high power density and high integration of electronic devices, their heat flux has been increasing rapidly. Therefore, an effective cooling technology is essential for the reliability and efficient operation of electronic devices. Liquid cooling is studied increasingly widely for its higher heat transfer efficiency. Serpentine channels are superior in the augmentation of single-phase convective heat transfer because of their better channel velocity distribution. In this paper, eight different frame sizes water-cooled serpentine channel heat sinks are designed to study the heat transfer and pressure drop characteristics. With water as the working fluid, experiment setup is established and the results showed the effect of different channel width, fin thickness and number of channels on thermal resistance and pressure drop.

5305 Jet Impingement Heat Transfer on a Rib-Roughened Flat Plate

Abstract:

Cooling by impingement jet is known to have a significant high local and average heat transfer coefficient which make it widely used in industrial cooling systems. The heat transfer characteristics of an impinging jet on rib-roughened flat plate has been investigated numerically. This paper was set out to investigate the effect of rib height on the heat transfer rate. Since the flow needs to have enough spacing after passing the rib to allow reattachment especially for high Reynolds numbers, this study focuses on finding the optimum rib height which would be the best to maximize the heat transfer rate downstream the plate. This investigation employs a round nozzle with hydraulic diameter (Dh) of 13.5 mm, Jet-to-target distance of (H/D) of 4, rib location=1.5D and and finally jet angels of 45˚ and 90˚ under the influence of Re =10,000.

5304 Analyzing the Effect of Design of Pipe in Shell and Tube Type Heat Exchanger by Measuring Its Heat Transfer Rate by Computation Fluid Dynamics and Thermal Approach

Abstract:

Shell and tube type heat exchangers are predominantly used in heat exchange between two fluids and other applications. This paper projects the optimal design of the pipe used in the heat exchanger in such a way to minimize the vibration occurring in the pipe. Paper also consists of the comparison of the different design of the pipe to get the maximize the heat transfer rate by converting laminar flow into the turbulent flow. By the updated design the vibration in the pipe due to the flow is also decreased. Computational Fluid Dynamics and Thermal Heat Transfer analysis are done to justifying the result. Currently, the straight pipe is used in the shell and tube type of heat exchanger where as per the paper the pipe consists of the curvature along with the pipe. Hence, the heat transfer area is also increased and result in the increasing in heat transfer rate. Curvature type design is useful to create turbulence and minimizing the vibration, also. The result will give the output comparison of the effect of laminar flow and the turbulent flow in the heat exchange mechanism, as well as, inverse effect of the boundary layer in heat exchanger is also justified.

5303 Folding Pathway and Thermodynamic Stability of Monomeric GroEL

Abstract:

Chaperonin GroEL is a tetradecameric Escherichia coli protein having identical subunits of 57 kDa. The elucidation of thermodynamic parameters related to stability for the native GroEL is not feasible as it undergoes irreversible unfolding because of its large size (800kDa) and multimeric nature. Nevertheless, it is important to determine the thermodynamic stability parameters for the highly stable GroEL protein as it helps in folding and holding of many substrate proteins during many cellular stresses. Properly folded monomers work as building-block for the formation of native tetradecameric GroEL. Spontaneous refolding behavior of monomeric GroEL makes it suitable for protein-denaturant interactions and thermodynamic stability based studies. The urea mediated unfolding is a three state process which means there is the formation of one intermediate state along with native and unfolded states. The heat mediated denaturation is a two-state process. The unfolding process is reversible as observed by the spontaneous refolding of denatured protein in both urea and head mediated refolding processes. Analysis of folding/unfolding data provides a measure of various thermodynamic stability parameters for the monomeric GroEL. The proposed mechanism of unfolding of monomeric GroEL is a three state process which involves formation of one stable intermediate having folded apical domain and unfolded equatorial, intermediate domains. Research in progress is to demonstrate the importance of specific residues in stability and oligomerization of GroEL protein. Several mutant versions of GroEL are under investigation to resolve the above mentioned issue.

5302 Experimental Study of Heat Transfer Enhancement Using Protruded Rectangular Fin

Abstract:

The investigation deals with the study of heat transfer enhancement using protruded square fin. This study is enough to determine whether protrusion in forced convection is enough to enhance the rate of heat transfer. It includes the results after performing experiments by using a plane rectangular fin of aluminum material and the same dimension rectangular fin of the same material but having protruded circular shape extended normally. The fins made by a sand casting method. The results clearly mentioned that the protruded surface is effective enough to enhance the rate of heat transfer. This research investigates a modern fin topologies heat transfer characteristics that will clearly outdated the conventional fin to increase the rate of heat transfer. Protruded fins improve the rate of heat transfer compared to solid fin by varying shape of the protrusion in diameter and height.

5301 Experimental Investigation of Heat Pipe with Annular Fins under Natural Convection at Different Inclinations

Abstract:

Heat pipe is characterised as superconductor of heat because of its excellent heat removal ability. The operation of several engineering system results in generation of heat. This may cause several overheating problems and lead to failure of the systems. To overcome this problem and to achieve desired rate of heat dissipation, there is need to study the performance of heat pipe with annular fins under free convection at different inclinations. This study demonstrates the effect of different mass flow rate of hot fluid into evaporator section on the condenser side heat transfer coefficient with annular fins under natural convection at different inclinations. In this study annular fins are used for the experimental work having dimensions of length of fin, thickness of fin and spacing of fin as 10 mm, 1 mm and 6 mm, respectively. The main aim of present study is to discover at what inclination angles the maximum heat transfer coefficient shall be achieved. The heat transfer coefficient on the external surface of heat pipe condenser section is determined by experimental method and then predicted by empirical correlations. The results obtained from experimental and Churchill and Chu relation for laminar are in fair agreement with not more than 22% deviation. It is elucidated the maximum heat transfer coefficient of 31.2 W/(m2-K) at 25˚ tilt angle and minimal condenser heat transfer coefficient of 26.4 W/(m2-K) is seen at 45˚ tilt angle and 200 ml/min mass flow rate. Inclination angle also affects the thermal performance of heat pipe. Beyond 25o inclination, heat transport rate starts to decrease.

Abstract:

The development of composite materials and the related design and manufacturing technologies is one of the most important advances in the history of materials. Composites are multifunctional materials having unprecedented mechanical and physical properties that can be tailored to meet the requirements of a particular application. Some composites also exhibit great resistance to high-temperature corrosion, oxidation, and wear. Polymers are widely used indoors and outdoors, therefore they are exposed to a chemical environment which may include atmospheric oxygen, acidic fumes, acidic rain, moisture heat and thermal shock, ultra-violet light, high energy radiation, etc. Different polymers are affected differently by these factors even though the amorphous polymers are more sensitive. Ageing is also important and it is defined as the process of deterioration of engineering materials resulting from the combined effects of atmospheric radiation, heat, oxygen, water, micro-organisms and other atmospheric factors.

5299 Study of Natural Convection Heat Transfer of Plate-Fin Heat Sink

Abstract:

This study applies the inverse method and three-dimensional CFD commercial software in conjunction with the experimental temperature data to investigate the heat transfer and fluid flow characteristics of the plate-fin heat sink in a rectangular closed enclosure. The inverse method with the finite difference method and the experimental temperature data is applied to determine the approximate heat transfer coefficient. Later, based on the obtained results, the zero-equation turbulence model is used to obtain the heat transfer and fluid flow characteristics between two fins. To validate the accuracy of the results obtained, the comparison of the heat transfer coefficient is made. The obtained temperature at selected measurement locations of the fin is also compared with experimental data. The effect of the height of the rectangular enclosure on the obtained results is discussed.

5298 To Examine Perceptions and Associations of Shock Food Labelling and to Assess the Impact on Consumer Behaviour: A Quasi-Experimental Approach

Abstract:

Shock and fear tactics have been used to encourage consumer behaviour change within the UK regarding lifestyle choices such as smoking and alcohol abuse, yet such measures have not been applied to food labels to encourage healthier purchasing decisions. Obesity levels are continuing to rise within the UK, despite efforts made by government and charitable bodies to encourage consumer behavioural changes, which will have a positive influence on their fat, salt, and sugar intake. We know that taking extreme measures to shock consumers into behavioural changes has worked previously; for example, the anti-smoking television adverts and new standardised cigarette and tobacco packaging have reduced the numbers of the UK adult population who smoke or encouraged those who are currently trying to quit. The USA has also introduced new front-of-pack labelling, which is clear, easy to read, and includes concise health warnings on products high in fat, salt, or sugar. This model has been successful, with consumers reducing purchases of products with these warning labels present. Therefore, investigating if shock labels would have an impact on UK consumer behaviour and purchasing decisions would help to fill the gap within this research field. This study aims to develop an understanding of consumer’s initial responses to shock advertising with an interest in the perceived impact of long-term effect shock advertising on consumer food purchasing decisions, behaviour, and attitudes and will achieve this through a mixed methodological approach taken with a sample size of 25 participants ages ranging from 22 and 60. Within this research, shock mock labels were developed, including a graphic image, health warning, and get-help information. These labels were made for products (available within the UK) with large market shares which were high in either fat, salt, or sugar. The use of online focus groups and mouse-tracking experiments results helped to develop an understanding of consumer’s initial responses to shock advertising with interest in the perceived impact of long-term effect shock advertising on consumer food purchasing decisions, behaviour, and attitudes. Preliminary results have shown that consumers believe that the use of graphic images, combined with a health warning, would encourage consumer behaviour change and influence their purchasing decisions regarding those products which are high in fat, salt and sugar. Preliminary main findings show that graphic mock shock labels may have an impact on consumer behaviour and purchasing decisions, which will, in turn, encourage healthier lifestyles. Focus group results show that 72% of participants indicated that these shock labels would have an impact on their purchasing decisions. During the mouse tracking trials, this increased to 80% of participants, showing that more exposure to shock labels may have a bigger impact on potential consumer behaviour and purchasing decision change. In conclusion, preliminary results indicate that graphic shock labels will impact consumer purchasing decisions. Findings allow for a deeper understanding of initial emotional responses to these graphic labels. However, more research is needed to test the longevity of these labels on consumer purchasing decisions, but this research exercise is demonstrably the foundation for future detailed work.

5297 Determination of Forced Convection Heat Transfer Performance in Lattice Geometric Heat Sinks

Abstract:

In this experimental study, the effects of heat transfer and flow characteristics on lattice geometric heat sinks, where high rates of heat removal are required, were investigated. The design parameters were Reynolds number, the height of heat sink (H), horizontal (Sy) and vertical (Sx) distances between heat sinks. In the experiments, the Reynolds number ranged from 4000 to 20000; heat sink heights were (H) 20 mm and 40 mm; the distances (Sy) between the heat sinks in the flow direction were45 mm, 32 mm, 23.3 mm; the distances (Sx) between the heat sinks perpendicular to the flow direction were selected to be 23.3 mm, 12.5 mm and 6 mm. A total of 90 experiments were conducted and the maximum Nusselt number and minimum friction coefficient were targeted. Experimental results have shown that heat sinks in lattice geometry have a significant effect on heat transfer enhancement. Under the different experimental conditions, the highest increase in Nusselt number was 283% while the lowest increase was calculated as 66% as compared with the straight channel results. The lowest increase in the friction factor was also obtained as 173% according to the straight channel results. It is seen that the increase in heat sink height and flow velocity increased the level of turbulence in the channel, leading to higher Nusselt number and friction factor values.

5296 Effect of Solution Heat Treatment on Intergranular Corrosion Resistance of Welded Stainless Steel AISI 321

Abstract:

In this investigation, AISI321 steel after welding by Shilded Metal Arc Welding (SMAW) was solution heat treated in various temperatures and times, and then was sensitizied. Results indicated, increasing of temperature in solution heat treatment raises the sensitization and creates the cavity structure in grain boundaries. Besides, in order to examine the effect of time on solution heat treatment, all samples were solution heat treated at different times and fixed temperature (1050°C). By increasing the time, more chrome carbides were created due to dissolution of delta ferrite phase and reproduce titanium carbides. Additionally, the best process for solution heat treatment for this steel was suggested.

5295 Experimental Investigation of Nanofluid Heat Transfer in a Plate Type Heat Exchanger

Abstract:

In this study, it was aimed to determine the convective heat transfer characteristics of water-based silicon dioxide nanofluids (SiO₂) with particle volume fractions of 0.2 and 0.4% vol. Nanofluids were tested in a plate type heat exchanger with six plates. Plate type heat exchanger was manufactured from stainless steel. Water was driven in the hot flow side, and nanofluids were driven in the cold flow side. The thermal energy of the hot water was taken by nanofluids. Effect of the inlet temperature of the hot water was investigated on heat transfer performance of the nanofluids while the inlet temperature of the nanofluids was fixed. In addition, the effects of the particle volume fraction and the cold flow rate on the performance of the system were tested. Results showed that increasing inlet temperature of the hot flow caused heat transfer to enhance. The suspended solid particles into the carrier fluid also remarkably enhanced heat transfer, and, an increase in the particle volume fraction resulted in an increase in heat transfer.

5294 Myeloid Zinc Finger 1/Ets-Like Protein-1/Protein Kinase C Alpha Associated with Poor Prognosis in Patients with Hepatocellular Carcinoma

Abstract:

Protein kinase C alpha (PKCα) is a key signaling molecule in human cancer development. As a therapeutic strategy, targeting PKCα is difficult because the molecule is ubiquitously expressed in non-malignant cells. PKCα is regulated by the cooperative interaction of the transcription factors myeloid zinc finger 1 (MZF-1) and Ets-like protein-1 (Elk-1) in human cancer cells. By conducting tissue array analysis, herein, we determined the protein expression of MZF-1/Elk-1/PKCα in various cancers. The data show that the expression of MZF-1/Elk-1 is correlated with that of PKCα in hepatocellular carcinoma (HCC), but not in bladder and lung cancers. In addition, the PKCα down-regulation by shRNA Elk-1 was only observed in the HCC SK-Hep-1 cells. Blocking the interaction between MZF-1 and Elk-1 through the transfection of their binding domain MZF-160–72 decreased PKCα expression. This step ultimately depressed the epithelial-mesenchymal transition potential of the HCC cells. These findings could be used to develop an alternative therapeutic strategy for patients with the PKCα-derived HCC.

5293 Different Stages for the Creation of Electric Arc Plasma through Slow Rate Current Injection to Single Exploding Wire, by Simulation and Experiment

Abstract:

This work simulates the voltage drop and resistance of the explosion of copper wires of diameters 25, 40, and 100 µm surrounded by 1 bar nitrogen exposed to a 150 A current and before plasma formation. The absorption of electrical energy in an exploding wire is greatly diminished when the plasma is formed. This study shows the importance of considering radiation and heat conductivity in the accuracy of the circuit simulations. The radiation of the dense plasma formed on the wire surface is modeled with the Net Emission Coefficient (NEC) and is mixed with heat conductivity through PLASIMO® software. A time-transient code for analyzing wire explosions driven by a slow current rise rate is developed. It solves a circuit equation coupled with one-dimensional (1D) equations for the copper electrical conductivity as a function of its physical state and Net Emission Coefficient (NEC) radiation. At first, an initial voltage drop over the copper wire, current, and temperature distribution at the time of expansion is derived. The experiments have demonstrated that wires remain rather uniform lengthwise during the explosion and can be simulated utilizing 1D simulations. Data from the first stage are then used as the initial conditions of the second stage, in which a simplified 1D model for high-Mach-number flows is adopted to describe the expansion of the core. The current was carried by the vaporized wire material before it was dispersed in nitrogen by the shock wave. In the third stage, using a three-dimensional model of the test bench, the streamer threshold is estimated. Electrical breakdown voltage is calculated without solving a full-blown plasma model by integrating Townsend growth coefficients (TdGC) along electric field lines. BOLSIG⁺ and LAPLACE databases are used to calculate the TdGC at different mixture ratios of nitrogen/copper vapor. The simulations show both radiation and heat conductivity should be considered for an adequate description of wire resistance, and gaseous discharges start at lower voltages than expected due to ultraviolet radiation and the exploding shocks, which may have ionized the nitrogen.

5292 Numerical Investigation of Natural Convection of Pine, Olive and Orange Leaves

Abstract:

Heat transfer of leaves is a crucial factor in optimal operation of metabolic functions in plants. In order to quantify this phenomenon in different leaves and investigate the influence of leaf shape on heat transfer, natural convection for pine, orange and olive leaves was simulated as representatives of different groups of leaf shapes. CFD techniques were used in this simulation with the purpose to calculate heat transfer of leaves in similar environmental conditions. The problem was simulated for steady state and three-dimensional conditions. From obtained results, it was concluded that heat fluxes of all three different leaves are almost identical, however, total rate of heat transfer have highest and lowest values for orange leaves and pine leaves, respectively.

5291 Thrombophilic Risk Factors and Pregnancy Complications

Abstract:

Background: Inherited thrombophilias are a heterogenous group of conditions which have been implicated in a variety of pregnancy complications. More recently, deficiency of protein Z (PZ) has been liked to pregnancy complications, including preterm delivery. Aim: We designed this study to evaluate the association of inherited thrombophilias including [Protein C (PC), Protein S (PS), Anti thrombin III (ATIII) deficiency and activated protein C (APC) resistance] and protein Z deficiency with a variety of pregnancy complications. Patients and Methods: 60 women with different pregnancy complications, including 20 patients with preeclampsia, 20 patients with intrauterine growth resistance (IUGR), and 20 patients with intrauterine fetal death (IUFD), in addition to 30 healthy pregnant women were recruited for the present study. PC and free PS antigen, ATIII activity, modified functional APC-resistance, and PZ levels were determined. Results: There was no significant association between inherited thrombophilias and complicated pregnancies as regards PC deficiency (p=1.0), AT III and PS deficiency (p=0.312), and APC-resistance (P=0.083). PZ was significantly associated with complicated pregnancies (p=0.012). Patients with protein Z levels below 1.5 µg/ml were considered deficient. Accordingly, we demonstrated protein Z deficiency in 30% of complicated pregnancies (RR 6.0, 95% CI 1.29-27.90;p=0.022), 20% of preeclampsia (RR 3.5, 95% CI 0.57 – 21.28; P = 0.174), 40% of IUGR (RR 9.3 95% CI 1.72-50.61; P = 0.010) and 30% of IUFD (RR 6, 95% CI 1.07 – 33.64; P = 0.042). Conclusions: These findings indicate the absence of association of inherited thrombophilias, including PC, PS, AT III deficiency, and APC resistance with pregnancy complications. However, PZ deficiency is associated with increased risk of pregnancy complications, especially intrauterine growth restriction and intrauterine fetal death.

5290 Optimization of Supercritical CO2 Power Cycle for Waste Heat Recovery from Gas Turbine with Respect to Cooling Condition

Abstract:

This study describes the optimization of supercritical carbon dioxide (S-CO2) power cycle for recovering waste heat from a gas turbine. An S-CO2 cycle that recovers heat from small industrial and aeroderivative gas turbines can outperform a steam-bottoming cycle despite its simplicity and compactness. In using S-CO2 power cycles for waste heat recovery, a split cycle was studied to maximize the net output power by incorporating the utilization efficiency of the waste heat (lowering the temperature of the exhaust gas through the heater) along with the thermal efficiency of the cycle (minimizing the temperature difference for the heat transfer, exergy loss). The cooling condition of the S-CO2 WHR system has a great impact on the performance and the optimum low pressure of the system. Furthermore, the optimum high pressure of the S-CO2 WHR systems for the maximum power from the given heat sources is dependent on the temperature of the waste heat source.

Abstract:

Nanofluids have received much more attention since its discovery. They are believed to be promising coolants in heat transfer applications due to their enhanced thermal conductivity and heat transfer characteristics. In this study, the enhancement in heat transfer of CNT-nanofluids under turbulent flow conditions is investigated experimentally. Carbon nanotube (CNTs) concentration was varied between 0.051-0.085 wt%. The nanofluid suspension was stabilized by gum arabic (GA) through a process of homogenisation and sonication. The flow rates of cold fluid (water) is varied from 1.7-3 L/min and flow rates of the hot fluid is varied between 2-3.5 L/min. Thermal conductivity, density and viscosity of the nanofluids were also measured as a function of temperature and CNT concentration. The experimental results are validated with theoretical correlations for turbulent flow available in the literature. Results showed an enhancement in heat transfer range between 9-67% as a function of temperature and CNT concentration.

5288 A Review of the Relation between Thermofludic Properties of the Fluid in Micro Channel Based Cooling Solutions and the Shape of Microchannel

Abstract:

The shape of microchannels in microchannel heat sinks can have a significant impact on both heat transfer and fluid flow properties. Heat Transfer, pressure drop, and Some effects of microchannel shape on these properties. The shape of microchannels can affect the heat transfer performance of microchannel heat sinks. Channels with rectangular or square cross-sections typically have higher heat transfer coefficients compared to circular channels. This is because rectangular or square channels have a larger wetted perimeter per unit cross-sectional area, which enhances the heat transfer from the fluid to the channel walls. The shape of microchannels can also affect the pressure drop across the heat sink. Channels with a rectangular cross-section usually have higher pressure drop than circular channels. This is because the corners of rectangular channels create additional flow resistance, which leads to a higher pressure drop. Overall, the shape of microchannels in microchannel heat sinks can have a significant impact on the heat transfer and fluid flow properties of the heat sink. The optimal shape of microchannels depends on the specific application and the desired balance between heat transfer performance and pressure drop.

5287 Transfer Learning for Protein Structure Classification at Low Resolution

Abstract:

Structure determination is key to understanding protein function at a molecular level. Whilst significant advances have been made in predicting structure and function from amino acid sequence, researchers must still rely on expensive, time-consuming analytical methods to visualise detailed protein conformation. In this study, we demonstrate that it is possible to make accurate (≥80%) predictions of protein class and architecture from structures determined at low (>3A) resolution, using a deep convolutional neural network trained on high-resolution (≤3A) structures represented as 2D matrices. Thus, we provide proof of concept for high-speed, low-cost protein structure classification at low resolution, and a basis for extension to prediction of function. We investigate the impact of the input representation on classification performance, showing that side-chain information may not be necessary for fine-grained structure predictions. Finally, we confirm that high resolution, low-resolution and NMR-determined structures inhabit a common feature space, and thus provide a theoretical foundation for boosting with single-image super-resolution.

5286 Potential Use of Cnidoscolus Chayamansa Leaf from Mexico as High-Quality Protein Source

Abstract:

Poverty and food insecurity are still incident problems in the developing countries, where population´s diet is based on cereals which are lack in protein content. Nevertheless, during last years the use of native plants has been studied as an alternative source of protein in order to improve the nutritional intake. Chaya crop also called Spinach tree, is a prehispanic plant native from Central America and South of Mexico (Mayan culture), which has been especially valued due to its high nutritional content particularly protein and some medicinal properties. The aim of this work was to study the effect of protein isolation processing from Chaya leaf harvest in Yucatan, Mexico on its structure quality in order: i) to valorize the Chaya crop and ii) to produce low-cost and high-quality protein. Chaya leaf was extruded, clarified and recovered using: a) acid precipitation by decreasing the pH value until reach the isoelectric point (3.5) and b) thermal coagulation, by heating the protein solution at 80 °C during 30 min. Solubilized protein was re-dissolved in water and spray dried. The presence of Fraction I protein, known as RuBisCO (Rubilose-1,5-biphosfate carboxylase/oxygenase) was confirmed by gel electrophoresis (SDS-PAGE) where molecular weight bands of 55 KDa and 12 KDa were observed. The infrared spectrum showed changes in protein structure due to the isolation method. The use of high temperatures (thermal coagulation) highly decreased protein solubility in comparison to isoelectric precipitated protein, the nutritional properties according to amino acid profile was also disturbed, showing minor amounts of overall essential amino acids from 435.9 to 367.8 mg/g. Chaya protein isolate obtained by acid precipitation showed higher protein quality according to essential amino acid score compared to FAO recommendations, which could represent an important sustainable source of protein for human consumption.

5285 Making Heat Pumps More Compatible with Environmental and Climatic Conditions

Abstract:

In this study, the effects of air temperature and relative humidity on the operation of the heat pump were examined experimentally. The results were analyzed in an energy and exergetic way. Two heat pumps were used in the experimental system established for experimental analysis. With the first heat pump, the relative humidity and temperature of atmospheric air are reduced. The air at low humidity and temperature is given heat and water vapor to the desired extent on the channel that reaches the other heat pump. Effects of the air reaching the desired humidity and temperature in the 2nd heat pump; temperature, humidity, pressure, flow, and current are detected by meters. The measured values and the exergy yield and thermodynamic favor ratios of the system and its components were determined. In this way, the effects of temperature and relative humidity change in the heat pump and components were tried to be revealed. Relative humidity in the air caused a significant increase in the loss of exergy in the evaporator. This has shown that cooling machines experience greater exergy in areas with high relative humidity. The highest COPSM values were determined to be at 30% and 40%, which is the least relative humidity values. The results showed that heat pump exergy efficiency was affected by increased temperature and relative humidity.

5284 Protein Tertiary Structure Prediction by a Multiobjective Optimization and Neural Network Approach

Abstract:

Protein structure prediction is a challenging task in the bioinformatics field. The biological function of all proteins majorly relies on the shape of their three-dimensional conformational structure, but less than 1% of all known proteins in the world have their structure solved. This work proposes a deep learning model to address this problem, attempting to predict some aspects of the protein conformations. Throughout a process of multiobjective dominance, a recurrent neural network was trained to abstract the particular bias of each individual multiobjective algorithm, generating a heuristic that could be useful to predict some of the relevant aspects of the three-dimensional conformation process formation, known as protein folding.

5283 Effect of 17α-Methyltestosterone Hormone on Haematological Profiles of the Sex Reversed, Sarotherodon Melanotheron

Abstract:

The effects of 17α-Methyltestosterone Hormone on blood composition of the Sex Reversed Sarotherodon melanotheron were investigated. S. melanotheron fry were reared in six (6) plastic tanks for three (3) months, of which three (3) tanks served as treatment tanks while the other three (3) served as the control. The fry were fed with 17α-methyl testosterone enzyme, which functions as a sex reversal hormone. The fry were administered this hormone for 30 days, to ensure complete sex reversal. All the S. melanotheron fry were reared to table size for duration of three (3) months, after which, blood samples were taken from both the control and treatment fishes. The blood parameters showed no significant differences with the same values of White Blood Cell count (WBC) and Total plasma protein for the control and experimental fishes. A total protein value for sex reversed specimens was 3.99g/dL, while urea and creatinine values were 0.2g/dL. Alkaline Phosphatase, Aspartate transaminase and Alanine transaminase for the treatment specimen were 183nm/mg protein/min, 98nm/mg protein/min and 105nm/mg protein/min respectively. A total protein value for control specimens was 2.81g/dL, while urea and creatinine values were 0.2g/dL. Alkaline Phosphatase, Aspartate transaminase and Alanine transaminase for the control species were 174nm/mg protein/min, 93nm/mg protein/min and 106nm/mg protein/min respectively. The safety of MT on S. melanotheron is therefore proved since there is no adverse effect on the fish.

5282 The Impact of Different Rhizobium leguminosarum Strains on the Protein Content of Peas and Broad Beans

Abstract:

Legume symbiotic relationship with nitrogen fixating bacteria Rhizobim leguminosarum is an important factor used to improve the productivity of legumes, due to the fact that rhizobia can supply plant with the necessary amount of nitrogen. R. leguminosarum strains have shown different activity in fixing nitrogen. Depending on the chosen R. leguminosarum strain, host plant biochemical content can be altered. In this study we focused particularly on the changes in protein content in beans (using two different varieties) and peas (five different varieties) due to the use of several different R. leguminosarum strains (four strains for both beans and peas). Overall, the protein content increase was observed after seed inoculation with R. leguminosarum. Strain and plant cultivar interaction specification was observed. The effect of R. leguminosarum inoculation on the content of protein was dependent on the R. leguminosarum strain used. Plant cultivar also appeared to have a decisive role in protein content formation with the help of R. leguminosaru.

5281 Coil-Over Shock Absorbers Compared to Inherent Material Damping

Abstract:

Damping accompanies us daily in everyday life and is used to protect (e.g., in shoes) and make our life more comfortable (damping of unwanted motion) and calm (noise reduction). In general, damping is the absorption of energy which is either stored in the material (vibration isolation systems) or changed into heat (vibration absorbers). In case of the last, the damping mechanism can be split in active, passive, as well as semi-active (a combination of active and passive). Active damping is required to enable an almost perfect damping over the whole application range and is used, for instance, in sport cars. In contrast, passive damping is a response of the material due to external loading. Consequently, the material composition has a huge influence on the damping behavior. For elastomers, the material behavior is inherent viscoelastic, temperature, and frequency dependent. However, passive damping is not adjustable during application. Therefore, it is of importance to understand the fundamental viscoelastic behavior and the dissipation capability due to external loading. The objective of this work is to assess the limitation and applicability of viscoelastic material damping for applications in which currently coil-over shock absorbers are utilized. Coil-over shock absorbers are usually made of various mechanical parts and incorporate fluids within the damper. These shock absorbers are well-known and studied in the industry, and when needed, they can be easily adjusted during their product lifetime. In contrary, dampers made of – ideally – a single material are more resource efficient, have an easier serviceability, and are easier manufactured. However, they lack of adaptability and adjustability in service. Therefore, a case study with a remote-controlled sport car was conducted. The original shock absorbers were redesigned, and the spring-dashpot system was replaced by both an elastomer and a thermoplastic-elastomer, respectively. Here, five different formulations of elastomers were used, including a pure and an iron-particle filled thermoplastic poly(urethan) (TPU) and blends of two different poly(dimethyl siloxane) (PDMS). In addition, the TPUs were investigated as full and hollow dampers to investigate the difference between solid and structured material. To get comparative results each material formulation was comprehensively characterized, by monotonic uniaxial compression tests, dynamic thermomechanical analysis (DTMA), and rebound resilience. Moreover, the new material-based shock absorbers were compared with spring-dashpot shock absorbers. The shock absorbers were analyzed under monotonic and cyclic loading. In addition, an impact loading was applied on the remote-controlled car to measure the damping properties in operation. A servo-hydraulic high-speed linear actuator was utilized to apply the loads. The acceleration of the car and the displacement of specific measurement points were recorded while testing by a sensor and high-speed camera, respectively. The results prove that elastomers are suitable in damping applications, but they are temperature and frequency dependent. This is a limitation in applicability of viscous material damper. Feasible fields of application may be in the case of micromobility, like bicycles, e-scooters, and e-skateboards. Furthermore, the viscous material damping could be used to increase the inherent damping of a whole structure, e.g., in bicycle-frames.

5280 Text Mining Techniques for Prioritizing Pathogenic Mutations in Protein Families Known to Misfold or Aggregate

Abstract:

Amyloid fibril forming regions, which are known as protein aggregates, in sequences of some protein families are associated with a number of diseases known as amyloidosis. Mutations play a role in forming fibrils by accelerating the fibril formation process. In this paper we want to extract diseases that caused by those mutations as a result of the impact of the mutations on structural and functional properties of the aggregated protein. We propose a text mining system, to automatically extract mutations, diseases and relations between mutations and diseases. We presented an algorithm based on finite state to cluster mutations found in the same sentence as a sentence could contain different mutation cause different diseases. Also, we presented a co reference algorithm that enables cross-link sentences.

5279 Experimental Investigation and Optimization of Nanoparticle Mass Concentration and Heat Input of Loop Heat Pipe

Abstract:

This study presents experimental and optimization of nanoparticle mass concentration and heat input based on the total thermal resistance (Rth) of loop heat pipe (LHP), employed for PC-CPU cooling. In this study, silica nanoparticles (SiO2) in water with particle mass concentration ranged from 0% (pure water) to 1% is considered as the working fluid within the LHP. The experimental design and optimization is accomplished by the design of the experimental tool, Response Surface Methodology (RSM). The results show that the nanoparticle mass concentration and the heat input have a significant effect on the Rth of LHP. For a given heat input, the Rth is found to decrease with the increase of the nanoparticle mass concentration up to 0.5% and increased thereafter. It is also found that the Rth is decreased when the heat input is increased from 20W to 60W. The results are optimized with the objective of minimizing the Rt, using Design-Expert software, and the optimized nanoparticle mass concentration and heat input are 0.48% and 59.97W, respectively, the minimum thermal resistance being 2.66(ºC/W).

5278 Optimising Light Conditions for Recombinant Protein Production in the Microalgal Chlamydomonas reinhardtii Chloroplast

Abstract:

The green alga C. reinhardtii provides a platform for the cheap, scalable, and safe production of complex proteins. Despite gene expression in photosynthetic organisms being tightly regulated by light, most expression studies have analysed chloroplast recombinant protein production under constant light. Here the influence of illumination time and intensity on GFP and a GFP-PlyGBS (bacterial-lysin) fusion protein expression was investigated. The expression of both proteins was strongly influenced by the light regime (6-24 hr illumination per day), the light intensity (0-450 E m⁻²s⁻¹) and growth condition (photoautotrophic, mixotrophic and heterotrophic). Heterotrophic conditions resulted in relatively low recombinant protein yields per unit volume, despite high protein yields per cell, due to low growth rates. Mixotrophic conditions exhibited the highest yields at 6 hrs illumination at 200µE m⁻²s⁻¹ and under continuous low light illumination (13-16 mg L⁻¹ GFP and 1.2-1.6 mg L⁻¹ GFP-PlyGBS), as these conditions supported good cell growth and cellular protein yields. A ~23-fold increase in protein accumulation per cell and ~9-fold increase L⁻¹ culture was observed compared to standard constant 24 hr illumination for GFP-PlyGBS. The highest yields under photoautotrophic conditions were obtained under 9 hrs illumination (6 mg L⁻¹ GFP and 2.1 mg L⁻¹ GFP-PlyGBS). This represents a ~4-fold increase in cellular protein accumulation for GFP-PlyGBS. On a volumetric basis the highest yield was at 15 hrs illumination (~2-fold increase L⁻¹ over the constant light for GFP-PlyGBS). Optimising illumination conditions to balance growth and protein expression can thus significantly enhance overall recombinant protein production in C. reinhardtii cultures.
