3215 Indecisiveness in 'The Road Not Taken' by Robert Frost: An Expressive Critical Analysis
Authors: Kurt S. Candilas
Abstract:
This expressive critical study is an effort to bring in light new interpretation of Robert Frost poem 'The Road Not Taken' as a reflection of his indecisiveness in life. Specifically, it aims at examining Frost’s inner being, emphasizing his own self and experiences in the poem or text. The study employs the qualitative research design which made use of discourse analysis using the critical theory of expressivism as the main guide. In acquiring the data of the study, the art of historiography is used such as autobiographical and/or biographical notes, sources documents, and web information. In executing the methods involved in this study, it is observed that the poem shows a naturalist implicatures, expressing Frost’s strong feelings and emotions being devoid of free will and a narrow bit of confusions and ambiguities with his indecisions in life. Keywords: The Road Not Taken, expressivism, indecisiveness, naturalist implicatures
Procedia PDF Downloads 3443214 An Enhanced Hybrid Backoff Technique for Minimizing the Occurrence of Collision in Mobile Ad Hoc Networks
Authors: N. Sabiyath Fatima, R. K. Shanmugasundaram
Abstract:
In Mobile Ad-hoc Networks (MANETS), every node performs both as transmitter and receiver. The existing backoff models do not exactly forecast the performance of the wireless network. Also, the existing models experience elevated packet collisions. Every time a collision happens, the station’s contention window (CW) is doubled till it arrives at the utmost value. The main objective of this paper is to diminish collision by means of contention window Multiplicative Increase Decrease Backoff (CWMIDB) scheme. The intention of rising CW is to shrink the collision possibility by distributing the traffic into an outsized point in time. Within wireless Ad hoc networks, the CWMIDB algorithm dynamically controls the contention window of the nodes experiencing collisions. During packet communication, the backoff counter is evenly selected from the given choice of [0, CW-1]. At this point, CW is recognized as contention window and its significance lies on the amount of unsuccessful transmission that had happened for the packet. On the initial transmission endeavour, CW is put to least amount value (C min), if transmission effort fails, subsequently the value gets doubled, and once more the value is set to least amount on victorious broadcast. CWMIDB is simulated inside NS2 environment and its performance is compared with Binary Exponential Backoff Algorithm. The simulation results show improvement in transmission probability compared to that of the existing backoff algorithm. Keywords: backoff, contention window, CWMIDB, MANET

Authors: Seyed Habib A. Rahmati, Parsa Fallah Sheikhlari, Morteza Musakhani
Abstract:
This paper studies the structure of financial investment and efficiency on the subway would be created between Hashtgerd and Qazvin in Iran. Regarding ascending rate of transportation between Tehran and Qazvin which directly air pollution, it clearly implies to public transportation requirement between these two cities near Tehran. The railway transportation like subway can help each country to terminate traffic jam which has some advantages such as speed, security, non-pollution, low cost of public transport, etc. This type of transportation needs national infrastructures which require enormous investment. It couldn’t implement without leading and managing funds and investments properly. In order to response 'needs', clear norms or normative targets have to be agreed and obviously it is important to distinguish costs from investment requirements critically. Implementation phase affects investment requirements and financing needs. So recognizing barrier related to investment and the quality of investment (what technologies and services are invested in) is as important as the amounts of investment. Different investment methods have mentioned as follows loan, leasing, equity participation, Line of financing, finance, usance, bay back. Alternatives survey before initiation and analyzing of risk management is one of the most important parts in this project. Observation of similar project cities each country has the own specification to choose investment method. Keywords: subway project, project investment, project contract, project management

Authors: Zakia Hammouni
Abstract:
Healthcare professionals’ well-being is becoming a priority during this Covid-19 pandemic due to stress, fatigue, and workload. Well before this pandemic, contemporary hospitals are endowed with environmental attributes that contribute to achieving well-being within their environment with the emphasis on the patient. The patient-centered care approach has been followed by the patient-centered design approach. Studies that have focused on the physical environment in hospitals have dealt with the patient's recovery process and his well-being. Prior scientific literature has placed less emphasis on the healthcare professionals’ interactions within the physical environment and to guide hospital designers to make evidence-based design choices to meet the needs and expectations of hospital users by considering, in addition to patients, healthcare professionals. This paper examines these issues related to the daily stress of professionals who provide care in a hospital environment. In this exploratory study, the interest was to grasp the issues related to this environment and explores the current realities of newly built hospitals based on design approaches and what attributes of the physical setting support healthcare professional’s well-being. Within a constructivist approach, this study was conducted in two care units in a new hospital in a big city in Canada before the Covid-19 pandemic (august 2nd to November 2nd 2018). A spatial evaluation of these care units allowed us to understand the interaction of health professionals in their work environment, to understand the spatial behavior of these professionals, and the narratives from 44 interviews of various healthcare professionals. The mental images validated the salient components of the hospital environment as perceived by these healthcare professionals. Thematic analysis and triangulation of the data set were conducted. Among the key attributes promoting the healthcare professionals’ well-being as revealed by the healthcare professionals are the overall light-color atmosphere in the hospital and care unit, particularly in the corridors and public areas of the hospital, the maintenance and cleanliness. The presence of the art elements also brings well-being to the health professionals as well as panoramic views from the staff lounge and corridors of the care units or elevator lobbies. Despite the overall positive assessment of this environment, some attributes need to be improved to ensure the well-being of healthcare professionals and to provide them with a restructuring environment. These are the supply of natural light, softer colors, sufficient furniture, comfortable seating in the restroom, and views, which are important in allowing these healthcare professionals to recover from their work stress. Noise is another attribute that needs to be further improved in the hospital work environment, especially in the nursing workstations and consultant's room. In conclusion, this study highlights the importance of providing healthcare professionals with work and rest areas that allow them to resist the stress they face, particularly during periods of extreme stress and fatigue such as a Covid-19 pandemic. Keywords: healthcare facilities, healthcare professionals, physical environment, well-being

Authors: Taha Bensiradj, Samira Moussaoui
Abstract:
Hybrid Sensors and Vehicular Networks (HSVN), represent a hybrid network, which uses several generations of Ad-Hoc networks. It is used especially in Intelligent Transport Systems (ITS). The HSVN allows making collaboration between the Wireless Sensors Network (WSN) deployed on the border of the road and the Vehicular Network (VANET). This collaboration is defined by messages exchanged between the two networks for the purpose to inform the drivers about the state of the road, provide road safety information and more information about traffic on the road. Moreover, this collaboration created by HSVN, also allows the use of a network and the advantage of improving another network. For example, the dissemination of information between the sensors quickly decreases its energy, and therefore, we can use vehicles that do not have energy constraint to disseminate the information between sensors. On the other hand, to solve the disconnection problem in VANET, the sensors can be used as gateways that allow sending the messages received by one vehicle to another. However, because of the short communication range of the sensor and its low capacity of storage and processing of data, it is difficult to ensure the exchange of road messages between it and the vehicle, which can be moving at high speed at the time of exchange. This represents the time where the vehicle is in communication range with the sensor. This work is the proposition of a communication protocol between the sensors and the vehicle used in HSVN. The latter has as the purpose to ensure the exchange of road messages in the available time of exchange. Keywords: HSVN, ITS, VANET, WSN
Procedia PDF Downloads 3633208 Multi-Level Clustering Based Congestion Control Protocol for Cyber Physical Systems
Authors: Manpreet Kaur, Amita Rani, Sanjay Kumar
Abstract:
The Internet of Things (IoT), a cyber-physical paradigm, allows a large number of devices to connect and send the sensory data in the network simultaneously. This tremendous amount of data generated leads to very high network load consequently resulting in network congestion. It further amounts to frequent loss of useful information and depletion of significant amount of nodes’ energy. Therefore, there is a need to control congestion in IoT so as to prolong network lifetime and improve the quality of service (QoS). Hence, we propose a two-level clustering based routing algorithm considering congestion score and packet priority metrics that focus on minimizing the network congestion. In the proposed Priority based Congestion Control (PBCC) protocol the sensor nodes in IoT network form clusters that reduces the amount of traffic and the nodes are prioritized to emphasize important data. Simultaneously, a congestion score determines the occurrence of congestion at a particular node. The proposed protocol outperforms the existing Packet Discard Network Clustering (PDNC) protocol in terms of buffer size, packet transmission range, network region and number of nodes, under various simulation scenarios. Keywords: internet of things, cyber-physical systems, congestion control, priority, transmission rate

Authors: Frederik Schulte, Stefan Voß
Abstract:
The adverse effects of air pollution on public health are an increasingly vital problem in planning for urban regions in many parts of the world. The issue is addressed from various angles and by distinct disciplines in research. Epidemiological studies model the relative increase of numerous diseases in response to an increment of different forms of air pollution. A significant share of air pollution in urban regions is related to transport emissions that are often measured and stored in emission inventories. Though, most approaches in transport planning, engineering, and operational design of transport activities are restricted to general emission limits for specific air pollutants and do not consider more nuanced exposure models. We conduct an extensive literature review on exposure models and emission inventories used to study the health impact of transport emissions. Furthermore, we review methods applied in both domains and use emission inventory data of transportation hubs such as ports, airports, and urban traffic for an in-depth analysis of public health impacts deploying medical exposure models. The results reveal specific urban health risks related to transport emissions that may improve urban planning for environmental health by providing insights in actual health effects instead of only referring to general emission limits. Keywords: emission inventories, exposure models, transport emissions, urban health

Authors: Muhammad Suradi, Sugiarto, Abdullah Latip
Abstract:
Many roadside slope failures occur during the rainy season, particularly in the period of extreme rainfall along Connecting National Road of Salubatu-Mambi, West Sulawesi, Indonesia. These occurrences cause traffic obstacles and endanger people along and around the road. Research collaboration between P2JN (National Road Construction Board) West Sulawesi Province, who authorize to supervise the road condition, and Ujung Pandang State Polytechnic (Applied University) was established to cope with the landslide problem. This research aims to determine factors triggering roadside slope failures and their optimum stabilizing solution. To achieve this objective, site observation and soil investigation were carried out to obtain parameters for analyses of rainfall-induced slope instability and reinforcement design using the SV Flux and SV Slope software. The result of this analysis will be taken into account for the next analysis to get an optimum design of the slope reinforcement. The result indicates some factors such as steep slopes, sandy soils, and unvegetated slope surface mainly contribute to the slope failures during intense rainfall. With respect to the contributing factors as well as construction material and technology, cantilever/butressing retaining wall becomes the optimum solution for the roadside slope reinforcement. Keywords: roadside slope, failure, rainfall, slope reinforcement, optimum solution

Authors: Ruchi Makani, B. V. R. Reddy
Abstract:
Mobile Adhoc Networks (MANETs) are gaining popularity due to their potential of providing low-cost mobile connectivity solutions to real-world communication problems. Integrating Intrusion Detection Systems (IDS) in MANETs is a tedious task by reason of its distinctive features such as dynamic topology, de-centralized authority and highly controlled/limited resource environment. IDS primarily use automated soft-computing techniques to monitor the inflow/outflow of traffic packets in a given network to detect intrusion. Use of machine learning techniques in IDS enables system to make decisions on intrusion while continuous keep learning about their dynamic environment. An appropriate IDS model is essential to be selected to expedite this application challenges. Thus, this paper focused on fuzzy-logic based machine learning IDS technique for MANETs and presented their applicability for achieving effectiveness in identifying the intrusions. Further, the selection of appropriate protocol attributes and fuzzy rules generation plays significant role for accuracy of the fuzzy-logic based IDS, have been discussed. This paper also presents the critical attributes of MANET’s routing protocol and its applicability in fuzzy logic based IDS.Keywords: AODV, mobile adhoc networks, intrusion detection, anomaly detection, fuzzy logic, fuzzy membership function, fuzzy inference system

Authors: Adetunji Olusegun Aderigibigbe
Abstract:
Shales, especially black shales, are of great geological significance, in the study of heavy/trace metal contamination. This is due to their abundance in occurrence and high concentration of heavy metals embedded which are released during their weathering. Heavy metals constitute one of the most dangerous pollution known to human because they are toxic (i.e., carcinogenic), non-biodegradable and can enter the global eco-biological circle. In the past, heavy metal contamination in aquatic environment and agricultural top soil has been attributed to industrial wastes, mining extractions and pollution from traffic vehicles; only a few studies have focused on weathering of shale as possible source of heavy metal contamination. Based on the above background, this study attempts to establish weathering of shale as possible source of trace/heavy metal contaminations. This was done by carefully selecting fresh and their corresponding weathered shale samples from selected localities in Bida and Anambra Basins. The samples were analysed in Activation Laboratories Ltd; Ontario, Canada for trace/heavy metal. It was observed that some major and trace metals were released during weathering, i.e., some were depleted and some enriched. By this contamination of water zones and agricultural top soils are not only traceable to biogenic processes but geogenic inputs (weathering of shale) as well. Keywords: contamination, fresh samples, heavy metals, pollution, shales, trace metals, weathered samples
Procedia PDF Downloads 1343201 Building a Dynamic News Category Network for News Sources Recommendations
Authors: Swati Gupta, Shagun Sodhani, Dhaval Patel, Biplab Banerjee
Abstract:
It is generic that news sources publish news in different broad categories. These categories can either be generic such as Business, Sports, etc. or time-specific such as World Cup 2015 and Nepal Earthquake or both. It is up to the news agencies to build the categories. Extracting news categories automatically from numerous online news sources is expected to be helpful in many applications including news source recommendations and time specific news category extraction. To address this issue, existing systems like DMOZ directory and Yahoo directory are mostly considered though they are mostly human annotated and do not consider the time dynamism of categories of news websites. As a remedy, we propose an approach to automatically extract news category URLs from news websites in this paper. News category URL is a link which points to a category in news websites. We use the news category URL as a prior knowledge to develop a news source recommendation system which contains news sources listed in various categories in order of ranking. In addition, we also propose an approach to rank numerous news sources in different categories using various parameters like Traffic Based Website Importance, Social media Analysis and Category Wise Article Freshness. Experimental results on category URLs captured from GDELT project during April 2016 to December 2016 show the adequacy of the proposed method. Keywords: news category, category network, news sources, ranking

Procedia PDF Downloads 873199 Hourly Solar Radiations Predictions for Anticipatory Control of Electrically Heated Floor: Use of Online Weather Conditions Forecast
Authors: Helene Thieblemont, Fariborz Haghighat
Abstract:
Energy storage systems play a crucial role in decreasing building energy consumption during peak periods and expand the use of renewable energies in buildings. To provide a high building thermal performance, the energy storage system has to be properly controlled to insure a good energy performance while maintaining a satisfactory thermal comfort for building’s occupant. In the case of passive discharge storages, defining in advance the required amount of energy is required to avoid overheating in the building. Consequently, anticipatory supervisory control strategies have been developed forecasting future energy demand and production to coordinate systems. Anticipatory supervisory control strategies are based on some predictions, mainly of the weather forecast. However, if the forecasted hourly outdoor temperature may be found online with a high accuracy, solar radiations predictions are most of the time not available online. To estimate them, this paper proposes an advanced approach based on the forecast of weather conditions. Several methods to correlate hourly weather conditions forecast to real hourly solar radiations are compared. Results show that using weather conditions forecast allows estimating with an acceptable accuracy solar radiations of the next day. Moreover, this technique allows obtaining hourly data that may be used for building models. As a result, this solar radiation prediction model may help to implement model-based controller as Model Predictive Control. Keywords: anticipatory control, model predictive control, solar radiation forecast, thermal storage

Authors: Sofia Figueroa, Efrain Bernal, Silvia Del Pilar Forero, Humberto Ramirez
Abstract:
This work shows a detailed review of the scope of global research on the road infrastructure using materials from Construction and Demolition Waste (C&DW), also called RCD. In the first phase of this research, a segment of road was designed using recycled materials such as Reclaimed Asphalt Pavement (RAP) on the top, the natural coarse base including 30% of RAP and recycled concrete blocks. The second part of this segment was designed using regular materials for each layer of the pavement. Both structures were built next to each other in order to analyze and measure the material properties as well as performance and environmental factors in the pavement under real traffic and weather conditions. Different monitoring devices were installed among the structure, based on the literature revision, such as soil cells, linear potentiometer, moisture sensors, and strain gauges that help us to know the C&DW as a part of the pavement structure. This research includes not only the physical characterization but also the measured parameters in a field such as an asphalt mixture (RAP) strain (ετ), vertical strain (εᵥ) and moisture control in coarse layers (%w), and the applied loads and strain in the subgrade (εᵥ). The results will show us what is happening with these materials in order to obtain not only a sustainable solution but also to know its behavior and lifecycle. Keywords: sustainable pavements, construction & demolition waste-C&DW, recycled rigid concrete, reclaimed asphalt pavement-rap

Procedia PDF Downloads 4333196 Understanding Surface Failures in Thick Asphalt Pavement: A 3-D Finite Element Model Analysis
Authors: Hana Gebremariam Liliso
Abstract:
This study investigates the factors contributing to the deterioration of thick asphalt pavements, such as rutting and cracking. We focus on the combined influence of traffic loads and pavement structure. This study uses a three-dimensional finite element model with a Mohr-Coulomb failure criterion to analyze the stress levels near the pavement's surface under realistic conditions. Our model considers various factors, including tire-pavement contact stresses, asphalt properties, moving loads, and dynamic analysis. This research suggests that cracking tends to occur between dual tires. Some key discoveries include the risk of cracking increases as temperatures rise; surface cracking at high temperatures is associated with distortional deformation; using a uniform contact stress distribution underestimates the risk of failure compared to realistic three-dimensional tire contact stress, particularly at high temperatures; the risk of failure is higher near the surface when there is a negative temperature gradient in the asphalt layer; and debonding beneath the surface layer leads to increased shear stress and premature failure around the interface. Keywords: asphalt pavement, surface failure, 3d finite element model, multiaxial stress states, Mohr-Coulomb failure criterion
Procedia PDF Downloads 593195 Providing a Road Pricing and Toll Allocation Method for Toll Roads
Authors: Ali Babaei
Abstract:
There is a worldwide growing tendency toward construction of infrastructures with the possibility of private sector participation instead of free exploitation of public infrastructures. The construction and development of roads through private sector participation is performed by different countries because of appropriate results and benefits such as compensation of public budget deficit in road construction and maintenance and responding to traffic growth (demand). Toll is the most definite form of budget provision in road development. There are two issues in the toll rate assignment: A. costing of transport, B. Cost allocation and distribution of cost between different types of vehicles as each vehicle pay its own share. There can be different goals in toll collection and its extent is variable according to the strategy of toll collection. Costing principles in different countries are based on inclusion of the whole transport and not peculiar to the toll roads. For example, fuel tax policy functions where the road network users pay transportation cost (not just users of toll road). Whereas transportation infrastructures in Iran are free, these methods are not applicable. In Iran, different toll freeways have built by public investment and government provides participation in the road construction through encouragement of financial institutions. In this paper, the existing policies about the toll roads are studied and then the appropriate method of costing and cost allocation to different vehicles is introduced. Keywords: toll allocation, road pricing, transportation, financial and industrial systems

Authors: Nidhi Sharotri, Dhiraj Sud
Abstract:
Water pollution by pesticides constitutes a serious ecological problem due to their potential toxicity and bioaccumulation. The widespread use of pesticides in industry and agriculture along with their resistance to natural decomposition, biodegradation, chemical and photochemical degradation under typical environmental conditions has resulted in the emergence of these chemicals and their transformed products in natural water. Among AOP’s, heterogeneous photocatalysis using TiO2 as photocatalyst appears as the most emerging destructive technology for mineralization of the pollutant in aquatic streams. Among the various semiconductors (TiO2, ZnO, CdS, FeTiO3, MnTiO3, SrTiO2 and SnO2), TiO2 has proven to be the most efficient photocatalyst for environmental applications due to its biological and chemical inertness, high photo reactivity, non-toxicity, and photo stability. Semiconductor photocatalysts are characterized by an electronic band structure in which valence band and conduction band are separated by a band gap, i.e. a region of forbidden energy. Semiconductor based photocatalysts produces e-/h+ pairs which have been employed for degradation of organic pollutants. The present paper focuses on modification of TiO2 photocatalyst in order to shift its absorption edge towards longer wavelength to make it active under natural light. Semiconductor TiO2 photocatalysts was prepared by doping with anion (N), cation (Mn) and double doped (Mn, N) using greener approach. Titanium isopropoxide is used as titania precursor and ethanedithiol, hydroxyl amine hydrochloride, manganous chloride as sulphur, nitrogen and manganese precursors respectively. Synthesized doped TiO2 nanomaterials are characterized for surface morphology (SEM, TEM), crystallinity (XRD) and optical properties (absorption spectra and band gap). EPR data confirms the substitutional incorporation of Mn2+ in TiO2 lattice. The doping influences the phase transformation of rutile and anatase phase crystal and thereby the absorption spectrum changes were observed. The effect of variation of reaction parameters such as solvent, reaction time and calcination temperature on the yield, surface morphology and optical properties was also investigated. The TEM studies show the particle size of nanomaterials varies from 10-50 nm. The calculated band gap of nanomaterials varies from 2.30-2.60 eV. The photocatalytic degradation of organic pollutant organophosphate pesticide (Quinalphos) has been investigated by studying the changes in UV absorption spectrum and the promising results were obtained under visible light. The complete mineralization of quinalphos has occurred as no intermediates were recorded after 8 hrs of degradation confirmed from the HPLC studies. Keywords: quinalphos, doped-TiO2, mineralization, EPR

Authors: Mohsen Abotalebi Esfahani, Amin Rahimi
Abstract:
Maintenance and management of route and roads infrastructure is one of the most important and the most fundamental principles of the countries. Several methods have been under investigation as preventive proceedings for the maintenance of asphalt pavements for many years. Using a mixture of salt, sand and gravel is the most common method of deicing, which could have numerous harmful consequences. Icy or snow-covered road is one of the major reasons of accidents in rainy seasons, which causes substantial damages such as loss of time and energy, environmental pollution, destruction of buildings, traffic congestion and rising possibility of accidents. Regarding this, every year the government incurred enormous costs to secure traverses. In this study, asphalt pavements have been cured, in terms of compressive strength, tensile strength and resilient modulus of asphalt samples, under the influence of Magnesium Chloride, Calcium Chloride, Sodium Chloride, Urea and pure water; and showed that de-icing with the calcium chloride solution and urea have the minimum negative effect and de-icing with pure water has most negative effect on laboratory specimens. Hence some simple techniques and new equipment and less use of sand and salt, can reduce significantly the risks and harmful effects of excessive use of salt, sand and gravel and at the same time use the safer roads. Keywords: maintenance, sodium chloride, icyroad, calcium chloride

Abstract:
Energy consumption efficiency is a major concern for the material processing industry such as thermoforming process and molding. Indeed, these systems should deliver the right amount of energy at the right time to the processed material. Recent technical development, as well as the particularities of the heating system dynamics, made the Model Predictive Control (MPC) one of the best candidates for thermal control of several production processes like molding and composite thermoforming to name a few. The main principle of this technique is to use a dynamic model of the process inside the controller in real time in order to anticipate the future behavior of the process which allows the current timeslot to be optimized while taking future timeslots into account. This study presents a procedure based on a predictive control that brings balance between optimality, simplicity, and flexibility of its implementation. The development of this approach is progressive starting from the case of a single zone before its extension to the multizone and/or multisource case, taking thus into account the thermal couplings between the adjacent zones. After a quadratic formulation of the MPC criterion to ensure the thermal control, the linear expression is retained in order to reduce calculation time thanks to the use of the ARMAX linear decomposition methods. The effectiveness of this approach is illustrated by experiment and simulation. Keywords: energy efficiency, linear decomposition methods, model predictive control, mold heating systems

Authors: Mohammad Ali Farrokhpour
Abstract:
In the current decade and sustainable transportation systems, scooter sharing has attracted widespread attention as an environmentally-friendly means of public transportation which can help develop public transportation. The combination of scooters and subway in the area of sustainable transportation systems can provide a great many opportunities for developing access to public transportation. Of the challenges which have arisen and initiated discussions of interest about the implementation of a scooter-subway system to replace personal vehicles is the issue of routing in the aforementioned system. This has been chosen as the main subject of the present paper. Thus, the present paper provides an account for routing in this system. Because the issue of routing includes multiple factors such as time, costs, traffic, green spaces, etc., the above-mentioned problem is considered to be a multi-objective NP-hard optimization problem. For this purpose, the hybrid optimization approach of PSO-GA has been put forward in the present paper for the provided answers to be of higher accuracy and validity than those of normal optimization methods. The results obtained from modeling and problem solving for the case study in the MATLAB software are indicative of the efficiency and desirability of the model and the proposed approach for solving the modelKeywords: route planning, scooter sharing, public transportation, sharing system

Abstract:
The alternative oxidase (AOX) mediates cyanide-resistant respiration, which bypasses proton-pumping complexes III and IV of the cytochrome pathway to directly transfer electrons from reduced ubiquinone to molecular oxygen. In Chlamydomonas reinhardtii, AOX is a monomeric protein that is encoded by two genes of discrete subfamilies, AOX1 and AOX2. Although AOX has been proposed to play essential roles in stress tolerance of organisms, the role of subfamily AOX2 is largely unknown. In C. reinhardtii, AOX2 was initially identified as one of constitutively low expressed genes. Like other photosynthetic organisms C. reinhardtii cells frequently experience periods of hypoxia. To examine AOX2 transcriptional regulation and role of AOX2 in hypoxia adaptation, real-time PCR analysis and artificial microRNA method were employed. Two experimental approaches have been used to induce the anoxic conditions: dark-anaerobic and light-anaerobic conditions. C. reinhardtii cells exposed to the oxygen deprivation have shown increased AOX2 mRNA levels. By contrast, AOX1 was not an anoxia-responsive gene. In C. reinhardtii, a subset of genes is regulated by transcription factor CRR1 in anaerobic conditions. Notable, the AOX2 promoter region contains the potential motif for CRR1 binding. Therefore, the role of CRR1 in the control of AOX2 transcription was tested. The CRR1-underexpressing strains, that were generated and characterized in this work, exhibited low levels of AOX2 transcripts under anoxic conditions. However, the transformants still slightly induced AOX2 gene expression in the darkness. These confirmed our suggestions that darkness is a regulatory stimulus for AOX genes in C. reinhardtii. Thus, other factors must contribute to AOX2 promoter activity under dark-anoxic conditions. Moreover, knock-down of CRR1 caused a complete reduction of AOX2 expression under light-anoxic conditions. These results indicate that (1) CRR1 is required for AOX2 expression during hypoxia, and (2) AOX2 gene is regulated by CRR1 together with yet-unknown regulatory factor(s). In addition, the AOX2-underexpressing strains were generated. The analysis of amiRNA-AOX2 strains suggested a role of this alternative oxidase in hypoxia adaptation of the alga. In conclusion, the results reported here show that C. reinhardtii AOX2 gene is stress inducible. CRR1 transcriptional factor is involved in the regulation of the AOX2 gene expression in the absence of oxygen. Moreover, AOX2 but not AOX1 functions under oxygen deprivation.
