1396 Identification of Nutrient Sensitive Signaling Pathways via Analysis of O-GlcNAcylation

Abstract:
The majority of glucose metabolism proceeds through glycolytic pathways such as glycolysis or pentose phosphate pathway, however, about 5% is shunted through the hexosamine biosynthetic pathway, producing uridine diphosphate N-acetyl glucosamine (UDP-GlcNAc). This precursor can then be incorporated into complex oligosaccharides decorating the cell surface or remain as an intracellular post-translational-modification (PTM) of serine/threonine residues (O-GlcNAcylation, OGN), which has been identified on over 4,000 cytosolic or nuclear proteins. Intracellular OGN has major implications on cellularprocesses, typically by modulating protein localization, protein-protein interactions, protein degradation, and gene expression. Additionally, OGN is known to have an extensive cross-talk with phosphorylation, be in a competitive or cooperative manner. Unlike other PTMs there are only two cycling enzymes that are capable of adding or removing the GlcNAc moiety, O-linked N-aceytl glucosamine Transferase (OGT) and O-linked N-acetyl glucoamidase (OGA), respectively. The activity of OGT has been shown to be sensitive to cellular UDP-GlcNAc levels, even changing substrate affinity. Owing to this and that the concentration of UDP-GlcNAc is related to the metabolisms of glucose, amino acid, fatty acid, and nucleotides, O-GlcNAc is often referred to as a nutrient sensing rheostat. Indeed OGN is known to regulate several signaling pathways as a result of nutrient levels, such as insulin signaling. Dysregulation of OGN is associated with several disease states such as cancer, diabetes, and neurodegeneration. Improvements in glycomics over the past 10-15 years has significantly increased the OGT substrate pool, suggesting O-GlcNAc’s involvement in a wide variety of signaling pathways. However, O-GlcNAc’s role at the receptor level has only been identified in a case-by-case basis of known pathways. Examining the OGN of the plasma membrane (PM) may better focus our understanding of O-GlcNAc-effected signaling pathways. In this current study, PM fractions were isolated from several cell types via ultracentrifugation, followed by purification and MS/MS analysis in several cell lines. This process was repeated with or without OGT/OGA inhibitors or with increased/decreased glucose levels in media to ascertain the importance of OGN. Abstract:
This study tested a structural model which investigates the relationships among tourists’ need for uniqueness, perceived authenticity (object-based authenticity and existential authenticity) and behavioral intentions to consume cultural and heritage destinations. The sample of the study comprised of 281 participants in a cultural heritage site, in Cappadocia, Turkey. The data were provided via face to face interviews in two months (September and October) which considered the high season. Structural equation modeling was employed to test the causal relationships among the hypotheses. Findings revealed tourists’ creative choice had an influence on object-based authenticity and existential authenticity. Tourists’ avoidance had an influence on object-based authenticity. The study concluded that two dimensions, namely, the object based authenticity and existential authenticity had significant impact on behavioral intentions.Keywords: needs for uniqueness, perceived existential authenticity, emotions, behavioral intentions

CADmapper is a significant tool of software for transforming geographic data into realistic CAD drawings. It speeds up and simplifies the conversion process by automating it. This allows architects, urban planners, engineers, and geographic information system (GIS) experts to solely concentrate on the imaginative and scientific parts of their projects. While the future incorporation of AI has the potential for further improvements, CADmapper's current capabilities make it an indispensable asset in the business. It covers a combination of 2D and 3D city and urban area models. The user can select a specific square section of the map to view, and the fee is based on the dimensions of the area being viewed. Abstract:
In action recognition, previous studies have demonstrated the effectiveness of using multiple features to improve the recognition performance. We focus on two practical issues: i) most studies use a direct way of concatenating/accumulating multi features to evaluate the similarity between two actions. This way could be too strong since each kind of feature can include different dimensions, quantities, etc; ii) in many studies, the employed classification methods lack of a flexible and effective mechanism to add new feature(s) into classification. In this paper, we explore an unified scheme based on recently-proposed multi-dimensional dynamic time warping (MD-DTW). Experiments demonstrated the scheme's effectiveness of combining multi-feature and the flexibility of adding new feature(s) to increase the recognition performance. Abstract:
In this paper, we began in the first step by two-dimensional simulation of a CIGS solar cell, in order to increase the current record efficiency of 20.48% for a single CIGS cell. Was created by utilizing a set of physical and technological parameters a solar cell of reference (such as layer thicknesses, gallium ratio, doping levels and materials properties) documented in bibliography and very known in the experimental field. This was accomplished through modeling and simulation using Atlas SILVACO-TCAD, an tool two and three dimensions very powerful and very adapted. This study has led us to determine the influence of different environmental parameters such as illumination (G) and temperature (T). In the second step, we continued our study by determining the influence of physical parameters (the acceptor of concentration NA) and geometric (thickness t) of the CIGS absorber layer, were varied to produce an optimum efficiency of 24.36%. Abstract:
The purpose of this study was to determine the construct validity of the morale sprit scale (MSS). Ninety faculty members from colleges of physical education at Jordanian universities were chosen to participate in this study. The design of this study was an ex-post facto. The MSS consists of (48) items that measure different dimensions of morale spirit among faculty members. Principle axis factoring with oblique rotation was utilized to uncover the underlying structure of the instrument. The findings revealed eight factor solution explaining (72.825%). Seven factors were accepted according to the conditions of accepting factors. The seven factors were named morale as reflection of faculty and department's administration, regulations and instructions, working environment and conditions, promotions and incentives and salaries, relations between the faculty member's, the trend toward the college and university, the trend toward self factors.Keywords: Factorial validity, morale sprit, faculty members, Jordanian Universities

Abstract:
Wireless Sensor Networks (WSNs) are an emerging technology that opens up a new field of research. The significant advance in WSN leads to an increasing prevalence of various monitoring applications and real-time assistance in labs and factories. Selective surface activation induced by laser (SSAIL) is a promising technology that adapts to the WSN design freedom of shape, dimensions, and material. This article proposes and implements a WSN-based temperature and humidity monitoring system, and its deployed architectures made for the monitoring task are discussed. Experimental results of newly developed sensor nodes implemented in university campus laboratories are shown. Then, the simulation and the implementation results obtained through monitoring scenarios are displayed. At last, a convenient solution to keep the WSN alive and functional as long as possible is proposed. Unlike other existing models, on success, the node is self-powered and can utilise minimal power consumption for sensing and data transmission to the base station.Keywords: IoT, network formation, sensor nodes, SSAIL technology

In many Islamic societies, women face restrictions and limitations when it comes to their participation in the political sphere. This paper investigates the reasons behind these restrictions and analyzes how Islamic law can be used to curb any excesses that may arise from women's involvement in politics. By examining various case studies and analyzing the principles of Islamic law, this paper sheds light on the issue of women's participation in politics and provides potential solutions for balancing their rights and duties according to Islamic law. The methodology includes primary sources through in-depth oral interviews and secondary sources like textbooks and journals, aiming for a holistic understanding of the ethical dimensions of support initiatives within the context of Islamic Law to establish the fairness of Islam Law in its approach to women in politics. Abstract:
This paper studies free vibration behavior of single-walled carbon nanotubes (SWCNTs) embedded on elastic medium based on three-dimensional theory of elasticity. To accounting the size effect of carbon nanotubes, nonlocal theory is adopted to shell model. The nonlocal parameter is incorporated into all constitutive equations in three dimensions. The surrounding medium is modeled as two-parameter elastic foundation. By using Fourier series expansion in axial and circumferential direction, the set of coupled governing equations are reduced to the ordinary differential equations in thickness direction. Then, the state-space method as an efficient and accurate method is used to solve the resulting equations analytically. The different process engineering applications of vibratory finishing technology have led to its versatile use in the development of aviation components. The most noteworthy applications of vibratory finishing include deburring and imparting the required surface finish. In this paper, vibratory finishing has been used to study its effectiveness in removal of laser shock peened (LSP) layers from Titanium workpieces. A vibratory trough operating at a frequency of 25 Hz, amplitude 3.5 mm and titanium specimens (Ti-6Al-4V, Grade 5) of dimensions 50 x 50 x 10 mm³ were utilized for the experiments. A vibrating fixture operating at 200 Hz was used to provide vibration to the test piece and was immersed in the vibratory trough. It was evident that there is an increase in efficiency of removal of the complex featured layer and smoother surface finish with the introduction of the vibrating fixture in the vibratory finishing setup as compared to the conventional vibratory finishing setup wherein the fixture is not vibrating.Keywords: laser shock peening, material removal, surface roughness, vibrating fixture, vibratory finishing

Abstract:
Using Fourier transform and based on the Mindlin's 2nd gradient model that involves two length scale parameters, the Green's function, the Eshelby tensor, and the Eshelby-like tensor for a spherical inclusion are derived. It is proved that the Eshelby tensor consists of two parts; the classical Eshelby tensor and a gradient part including the length scale parameters which enable the interpretation of the size effect. When the strain gradient is not taken into account, the obtained Green's function and Eshelby tensor reduce to its analogue based on the classical elasticity. The Eshelby tensor in and outside the inclusion, the volume average of the gradient part and the Eshelby-like tensor are explicitly obtained. Unlike the classical Eshelby tensor, the results show that the components of the new Eshelby tensor vary with the position and the inclusion dimensions. Goal theory perspective as an emerging trend in students’ motivation explores reasons why students engage in achievement related behaviour. While previous research typifies students’ goal orientations into two dimensions of mastery and performance orientations in various other parts of the world, not much has been done in this regard in Nigeria and specifically in Bayelsa state to the best of the researcher’s knowledge. To this end, the study explores the achievement goal orientations of schooling adolescents in Bayelsa State. The sample of the study consists of 220 schooling adolescents drawn from four urban schools in the state. A modified form of the Patterns of Adaptive learning survey (PALS) questionnaire was used to elicit data. Results indicated that schooling adolescents in Bayelsa state are mastery as well as performance oriented. The students also did not differ in goal orientations by gender. The main objective of structural design is to ensure safety and functional performance requirements of a structural system for its target reliability levels. In this study, the reliability index for the reinforcement concrete slender columns with rectangular cross section is studied. The variable parameters studied include the loads, the concrete compressive strength, the steel yield strength, the dimensions of concrete cross-section, the reinforcement ratio, and the location of steel placement. Risk analysis program was used to perform the analytical study. The effect of load eccentricity on the reliability index of reinforced concrete slender column was studied and presented. Abstract:
The expression ‘Porto School’, usually associated with an educational institution, the School of Fine Arts of Porto, is applied for the first time with the sense of an architectural trend by Nuno Portas in a text published in 1983. The expression is used to characterize a set of works by Porto architects, in which common elements are found, namely the desire to reuse languages and forms of the German and Dutch rationalism of the twenties, using the work of Alvar Aalto as a mediation for the reinterpretation of these models. In the same year, Álvaro Siza classifies the Finnish architect as a miscegenation agent who transforms experienced models and introduces them to different realities in a text published in Jornal de Letras, Artes e Ideias. The influence of foreign models and their adaptation to the context has been a recurrent theme in Portuguese architecture, which finds important contributions in the writings of Alexandre Alves Costa, at this time. However, the identification of these characteristics in Siza’s work is not limited to the Portuguese theoretical production: it is the recognition of this attitude towards the context that leads Kenneth Frampton to include Siza in the restricted group of architects who embody Critical Regionalism (in his book Modern architecture: a critical history). For Frampton, his work focuses on the territory and on the consequences of the intervention in the context, viewing architecture as a tectonic fact rather than a series of scenographic episodes and emphasizing site-specific aspects (topography, light, climate). Therefore, the motto of this paper is the dichotomous opposition between foreign influences and adaptation to the context in the early work of Álvaro Siza (designed in the sixties) in which the influence (theoretical, methodological, and formal) of Alvar Aalto manifests itself in the form and the language: the pool at Quinta da Conceição, the Seaside Pools and the Tea House (three works in Leça da Palmeira) and the Lordelo Cooperative (in Porto). In Serbia as a continental country, the tourism industry relies on city-break, spa and mountain tourism, where ski resorts have primacy during the winter season. Even though the number of tourists has recently increased, the share of domestic tourists remained predominant. It is also noticed that tourists from Serbia eagerly travel abroad, which was so far researched in the context of summer holidays but not in the framework of ski resorts. Therefore, this paper examines the competitiveness of ski resorts in Serbia from the perspective of domestic tourists. A survey was used as a data collection method, covering various competitiveness dimensions. The aim is to recognize the main motives of consumers when choosing a ski resort in Serbia or abroad. The results showed that the choices of Serbian tourists are predominantly shaped by the cost of an offer – of accommodation above all others. They are attentive by estimating the value for money, which is the most common reason to choose a ski resort abroad over a domestic one. The crowd at ski resorts and ski runs appears to be a result of unbalanced accommodation capacities on the one hand and ski infrastructure on the other, which is currently the most notable competitiveness drawback of ski resorts in Serbia.Keywords: mountain tourism, Serbia, ski resorts, tourism competitiveness

This work concerns the coacervate extraction of industrial dye, namely BezanylGreen - F2B, from an aqueous solution by nonionic surfactant “Lutensol AO7 and TX-114” (readily biodegradable). Binary water/surfactant and pseudo-binary (in the presence of solute) phase diagrams were plotted. The extraction results as a function of wt.% of the surfactant and temperature are expressed by the following four quantities: percentage of solute extracted, E%, residual concentrations of solute and surfactant in the dilute phase (Xs,w, and Xt,w, respectively) and volume fraction of coacervate at equilibrium (Фc). For each parameter, whose values are determined by a design of experiments, these results are subjected to empirical smoothing in three dimensions. The aim of this study is to find out the best compromise between E% and Фc. E% increases with surfactant concentration and temperature in optimal conditions, and the extraction extent of TA reaches 98 and 96 % using TX-114 and Lutensol AO7, respectively. The effect of sodium sulfate or cetyltrimethylammonium bromide (CTAB) addition is also studied. The current paper shows the application of the boundary element method for the analysis of plates under shear stress causing plasticity. In this case, the shear deformation of a plate is considered by means of the Reissner’s theory. The probability of failure of a Reissner’s plate due to a proposed index plastic behavior is calculated taken into account the uncertainty in mechanical and geometrical properties. The problem is developed in two dimensions. The classic plasticity’s theory is applied and a formulation for initial stresses that lead to the boundary integral equations due to plasticity is also used. For the plasticity calculation, the Von Misses criteria is used. To solve the non-linear equations an incremental method is employed. The results show a relatively small failure probability for the ranges of loads between 0.6 and 1.0. However, for values between 1.0 and 2.5, the probability of failure increases significantly. Consequently, for load bigger than 2.5 the plate failure is a safe event. The results are compared to those that were found in the literature and the agreement is good.Keywords: boundary element method, failure, plasticity, probability

In the present study, a design of the suspended polymeric microfluidic platform is introduced that is fabricated with three polymeric layers. Changing the microchannel plane to be perpendicular to microcantilever plane, drastically decreases moment of inertia in that direction. In addition, the platform is made of polymer (around five orders of magnitude less compared to silicon). It causes significant increase in the sensitivity of the cantilever deflection. Next, although the dimensions of this platform are constant, by misaligning the embedded microchannels laterally in the suspended microfluidic platform, the sensitivity can be highly increased. The investigation is studied on four fluids including water, seawater, milk, and blood for flow ranges from low rate of 5 to 70 µl/min to obtain the best design with the highest sensitivity. The best design in this study shows the sensitivity increases around 50% for water, seawater, milk, and blood at the flow rate of 70 µl/min by just misaligning the embedded microchannels in the suspended polymeric microfluidic platform.Keywords: microfluidic, MEMS, biosensor, microresonator

Abstract:
The rate of genetic progress in wheat production must be improved to meet global food security targets. However, past selection for domestication traits has reduced the genetic variation in modern wheat cultivars, a fact that could severely limit the future rate of genetic gain. The genetic variation in agronomically important traits for the wild relatives and progenitors of wheat is far greater than that of the current domesticated cultivars, but transferring these traits into modern cultivars is not straightforward. Between the elite cultivars of wheat, photosynthetic capacity is a key trait for which there is limited variation. Early screening of wheat wild relative and progenitors has shown differences in photosynthetic capacity and efficiency not only between wild relative species but marked differences between the accessions of each species. By identifying wild relative accessions with improved photosynthetic traits and characterising the genetic variation responsible, it is possible to incorporate these traits into advanced breeding programmes by wide crossing and introgression programmes. To identify the potential variety of photosynthetic capacity and efficiency available in the secondary and tertiary genepool, a wide scale survey was carried out for over 600 accessions from 80 species including those from the genus Aegilops, Triticum, Thinopyrum, Elymus, and Secale. Genotype data were generated for each accession using a ‘Wheat Wild Relative’ Single Nucleotide Polymorphism (SNP) genotyping array composed of 35,000 SNP markers polymorphic between wild relatives and elite hexaploid wheat. This genotype data was combined with phenotypic measurements such as gas exchange (CO₂, H₂O), chlorophyll fluorescence, growth, morphology, and RuBisCO activity to identify potential breeding material with enhanced photosynthetic capacity and efficiency. The data and associated analysis tools presented here will prove useful to anyone interested in increasing the genetic diversity in hexaploid wheat or the application of complex genotyping data to plant breeding.Keywords: wheat, wild relatives, pre-breeding, genomics, photosynthesis

Stress causes deleterious effects to the physical, psychological and organizational levels, which highlight the need to use effective coping strategies to deal with it. Several coping models exist, but they don’t integrate the different strategies in a coherent way nor do they take into account the new research on the emotional coping and acceptance of the stressful situation. To fill these gaps, an integrative model incorporating the main coping strategies was developed. This model arises from the review of the scientific literature on coping and from a qualitative study carried out among workers with low or high levels of stress, as well as from an analysis of clinical cases. The model allows one to understand under what circumstances the strategies are effective or ineffective and to learn how one might use them more wisely. It includes Specific Strategies in controllable situations (the Modification of the Situation and the Resignation-Disempowerment), Specific Strategies in non-controllable situations (Acceptance and Stubborn Relentlessness) as well as so-called General Strategies (Wellbeing and Avoidance). This study is intended to undertake and present the process of development and validation of an instrument to measure coping strategies based on this model. An initial pool of items has been generated from the conceptual definitions and three expert judges have validated the content. Of these, 18 items have been selected for a short form questionnaire. A sample of 300 students and employees from a Quebec university was used for the validation of the questionnaire. Concerning the reliability of the instrument, the indices observed following the inter-rater agreement (Krippendorff’s alpha) and the calculation of the coefficients for internal consistency (Cronbach's alpha) are satisfactory. To evaluate the construct validity, a confirmatory factor analysis using MPlus supports the existence of a model with six factors. The results of this analysis suggest also that this configuration is superior to other alternative models. The correlations show that the factors are only loosely related to each other. Overall, the analyses carried out suggest that the instrument has good psychometric qualities and demonstrates the relevance of further work to establish predictive validity and reconfirm its structure. This instrument will help researchers and clinicians better understand and assess coping strategies to cope with stress and thus prevent mental health issues.Keywords: acceptance, coping strategies, stress, validation process

Abstract:
This paper analyses how different educational backgrounds affect the mathematics performance of middle and high school students in terms of three dimensions: parental involvement, school teaching ability, and demographic variables and personal attributes of the student. Based on the analysis of Beijing High School Mathematics Competition in 2022, it was found that students from high level schools won significantly more awards than those from low level schools. In addition, a significant positive correlation (p<0.05) was identified between school level and students' mathematics performance. This study also confirms that parents' education level and family environment show a significant impact on the next generation’s mathematics learning performance. The findings suggest that interest and student’s habits, the family environment and the quality of teaching and learning at school are the main factors affecting the mathematics performance of middle and high school students.Keywords: educational background, academic performance, middle and high school education, teenager

There is a complicated relation between effective input parameters of cold rolling and output rolling force and exit thickness of strips.in many mathematical models, the effect of some rolling parameters have been ignored and the outputs have not a desirable accuracy. In the other hand, there is a special relation among input thickness of strips,the width of the strips,rolling speeds,mandrill tensions and the required exit thickness of strips with rolling force and the real exit thickness of the rolled strip. Abstract:
The influence of three different types of halloysite nanotubes (HNTs) with different dimensions, namely as camel lake (CLA), Jarrahdale (JA) and Matauri Bay (MB), on their reinforcing ability of ethylene propylene dine monomer (EPDM) were investigated by varying the HNTs loading (from 0-15 phr). Mechanical properties of the nanocomposites improved with addition of all three HNTs, but CLA based nanocomposites exhibited a significant enhancement compared to the other HNTs. For instance, tensile properties of EPDM nanocomposites increased by 120%, 256% and 340% for MB, JA, and CLA, respectively with addition of 15 phr of HNTs. This could be due to the higher aspect ratio and higher surface area of CLA compared to others. Scanning electron microscopy (SEM) of nanocomposites at 15 phr of HNT loadings showed low amounts of pulled-out nanotubes which confirmed the presence of more embedded nanotubes inside the EPDM matrix, as well as aggregates within the fracture surface of EPDM/HNT nanocomposites.Keywords: aspect ratio, halloysite nanotubes (HNTs), mechanical properties, rubber/clay nanocomposites

Abstract:
For studying the overall instability of members of steel structures, there are several methods in which overall buckling and geometrical imperfection effects are considered in analysis. In first section, these methods are compared and ability of software to apply these methods is studied. Buckling loads determined from theoretical methods and software is compared for 2D one bay, one and two stories steel frames. To consider actual condition, buckling loads of three steel frames that have various dimensions are calculated and compared. Also, uncertainties that exist in loading and modeling of structures such as geometrical imperfection, yield stress, and modulus of elasticity in buckling load of 2D framed steel structures have been studied. By performing these uncertainties to each reliability analysis procedures (first-order, second-order, and simulation methods of reliability), one index of reliability from each procedure is determined. Abstract:
Crown enclosure high temperature flares damaged and reduced dimensions crown. Generally crown on EHTF could have a life time up to twenty years. Therefore, this study aims to increase the value of thermal resistance with the effect post treatment on NiCr coated arc spray method. The variation of post treatment temperature, was at 650°C, 750°C, and 850°C. Morphology on the surface and the adhesion strength was analyzed by SEM-EDX, Surface Roughness and Pull - off test. XRD testing was conducted to determine the contained in NiCr coated. Thermal stability of NiCr coated was tested by DSC-TGA. The most optimal results was owned by NiCr coating with post treated at 850°C. It has good thermal stability until 1000°C because of Cr2O3 formation in coated specimen. Aluminum and its alloys are versatile materials which are widely used in industrial and engineering applications due to their good and useful properties e.g. high strength to weight ratio, high thermal and electrical conductivities and good resistance to corrosion. However, against these favorable properties they have the disadvantage they solidifying large grain columnar structure which negatively affects their mechanical properties and surface quality. Aluminum alloys are normally grain refined by some alloying elements, such as Ti, Ti-B or Zr. In this paper, the effect of zirconium addition to Al grain refined by Ti after extrusion on its wear resistance is investigated under different loads and sliding speeds namely at 5,10 and 20 N loads and sliding speeds ranging from m/min. and m/min. the results are presented in three-dimensional wear mode. To the best the authors' knowledge, the wear of aluminum in 3-dimensions has never been tackled before. Encapsulated O-rings are specifically designed to address the problem of sealing the most hostile chemicals and extreme temperature applications. Ultrasonic vibration hot embossing and ultrasonic welding techniques provide a fast and reliable method to fabricate encapsulated O-ring. This paper performs the design and analysis method of the acoustic horns with double extrusion to process tube double side flange simultaneously. The paper deals with study through Finite Element Method (FEM) of ultrasonic stepped horn used to process a capsulated O-ring, the theoretical dimensions of horns, and their natural frequencies and amplitudes are obtained through the simulations of COMOSOL software.
