13865 Synthesis and Pharmaco-Potential Evaluation of Quinoline Hybrids
Authors: Paul Awolade, Parvesh Singh
Abstract:
The global threat of pathogenic resistance to available therapeutic agents has become a menace to clinical practice, public health and man’s existence inconsequential. This has therefore led to an exigency in the development of new molecular scaffolds with profound activity profiles. In this vein, a versatile synthetic tool for accessing new molecules by incorporating two or more pharmacophores into a single entity with the unique ability to be recognized by multiple receptors hence leading to an improved bioactivity, known as molecular hybridization, has been explored with tremendous success. Accordingly, aware of the similarity in pharmacological activity spectrum of quinoline and 1,2,3-triazole pharmacophores such as; anti-Alzheimer, anticancer, anti-HIV, antimalarial and antimicrobial to mention but a few, the present study sets out to synthesize hybrids of quinoline and 1,2,3-triazole. The hybrids were accessed via click chemistry using copper catalysed azide-alkyne 1,3-dipolar cycloaddition reaction. All synthesized compounds were evaluated for their pharmaco-potential in an antimicrobial assay out of which the 3-OH derivative emerged as the most active with MIC value of 4 μg/mL against Cryptococcus neoformans; a value superior to standard Fluconazole and comparable to Amphotericin B. Structures of synthesized hybrids were elucidated using appropriate spectroscopic techniques (1H, 13C and 2D NMR, FT-IR and HRMS).Keywords: bioisostere, click chemistry, molecular hybridization, quinoline, 1, 2, 3-triazole

Authors: Meidesta Pitria, Akiko Okabe
Abstract:
In high-density informal settlement, extension area at the outside of the houses could primarily happen as a spatial modification response. House extension in high-density informal settlement is not only becoming a physical spatial modification that makes a blur zone between private and public but also supporting the growth and existence of informal economy and other daily activities in both individuals and communities. This research took a case study in an informal settlement named Kampung Cikini, a densely populated area in Central Jakarta. The aim of this study is to identify and clarify house extension as a strategy in dealing with urbanization in an informal settlement. By using the perspective and information from housewives, the analysis is based on the assumption that land ownership transformation and the activities in house extension area influence the different kinds of house extension’s spatial modification and local planning policy in relation with the implementation of house extension strategy. The data collection was done in four sites, two sites are located in outer-wide alley and another two sites are located in inner-narrow alley. In this research, data of 104 housewives in 86 houses were collected through representatives of housewives and local leader of each sites. The research was started from participatory mapping process, deep interview with local leader, and initiated collaboration with housewives community in having a celebration as communal event to cultivate together the issue. This study shows that land ownership, activities, and alley are indispensable in the decision of extension space making. The more permanency status of land ownership the more permanent and various extension could be implemented. However, in some blocks, the existence of origin house or first land owner also has a significant role in coordination and agreement in using and modifying extension space. In outer-wide alley, the existence of more various activities in front area of the houses is significantly related with the chance given by having wider alley, particularly for informal income generating activities. In the inner-narrow alley, limited space in front of the houses affects more negotiations in the community for having more shared spaces, even inside their private space. Keywords: house extension, housewives, informal settlement, kampung, high density

Authors: Wen Shyong Hsieh, Chih Hsueh Lin
Abstract:
In this paper, we combine the concepts of chameleon hash function (CHF) and identification based cryptography (IBC) to build a message authentication environment for VANET under sparse RSUs. Based on the CHF, TA keeps two common secrets that will be embedded to all identities to be as the evidence of mutual trusting. TA will issue one original identity to every RSU and vehicle. An identity contains one public ID and one private key. The public ID, includes three components: pseudonym, random key, and public key, is used to present one entity and can be verified to be a legal one. The private key is used to claim the ownership of the public ID. Based on the concept of IBC, without any negotiating process, a CHF pairing key multiplied by one private key and other’s public key will be used for mutually trusting and to be utilized as the session key of secure communicating between RSUs and vehicles. To help the vehicles to do message authenticating, the RSUs are assigned to response the vehicle’s temple identity request using two short time secretes that are broadcasted by TA. To light the loading of request information, one day is divided into M time slots. At every time slot, TA will broadcast two short time secretes to all valid RSUs for that time slot. Any RSU can response the temple identity request from legal vehicles. With the collected announcement of public IDs from the neighbor vehicles, a vehicle can set up its neighboring set, which includes the information about the neighbor vehicle’s temple public ID and temple CHF pairing key that can be derived by the private key and neighbor’s public key and will be used to do message authenticating or secure communicating without the help of RSU.Keywords: Internet of Vehicles (IOV), Vehicular Ad-hoc Networks (VANETs), Chameleon Hash Function (CHF), message authentication

Authors: Khalida Boutemak, Nasssima Benali, Nadji Moulai-Mostefa
Abstract:
Effect of enzyme on the yield and chemical composition of Artemisia campestris essential oil is reported in the present study. It was demonstrated that enzyme facilitated the extraction of essential oil with increase in oil yield and did not affect any noticeable change in flavour profile of the volatile oil. Essential oil was tested for antibacterial activity using Escherichia coli; which was extremely sensitive against control with the largest inhibition (29mm), whereas Staphylococcus aureus was the most sensitive against essential oil obtained from enzymatic pre-treatment with the largest inhibition zone (25mm). The antioxidant activity of the essential oil with hemicellulase pre-treatment (EO2) and control sample (EO1) was determined through reducing power. It was significantly lower than the standard drug (vitamin C) in this order: vitamin C˃EO2˃EO1.Keywords: Artemisia campestris, enzyme pre-treatment, hemicellulase, antibacterial activity, antioxidant activity

Authors: George Lin
Abstract:
This study examines a practice in which Chinese municipal governments actively pursue momentary and spectacular urbanism through the hosting of mega-events as an instrument to reproduce urban space for the enhancement of place competitiveness and advancement of political career. Practicing event-driven spectacular urbanism is found to have a short-term impact upon the economy and an effect upon the career advancement of the party secretary more than the mayor. Hosting mega-events has been used as a means to create “a harmonious society” and unified social space whereby grievance and discontents are grossed over, ignored, excluded and marginalized. Geographically, a new urban space has been created for the central city to reassert/consolidate its leading competitive position in the regional and national economy at the expense of the disadvantaged and marginalized. Findings of this research call for a critical re-evaluation of the sophisticated state-space inter-relations in the ongoing processes of planetary urbanization and global urban revolution in which China has taken an important part. Keywords: Chinese cities, mega events, urbanism, urbanization

Authors: Zintle Sikhunyana
Abstract:
Management of public finance in many countries such as South Africa is affected by political decisions and by policies around fiscal decentralization amongst the government spheres. Economic success is said to be determined by efficient management of public finance and by the policies or strategies that are implemented to support efficient public finance management. Policymakers focus on pay attention to how economic policies have been implemented and how they are directed into ensuring stable development. This will allow policymakers to address economic challenges through the usage of fiscal policy parameters that are linked to the achieved rate of economic growth and development. Efficient public finance management reduces the likelihood of corruption and corruption is said to have negative effects on economic growth and development. Corruption in public finance refers to an act of using funds for personal benefits. To achieve macroeconomic objectives, governments make use of government expenditure and government expenditure is financed through tax revenue. The main aim of this paper is to investigate the potential impact of public finance management on economic growth and development in South Africa. The secondary data obtained from the South African Reserve Bank (SARB) and World Bank for 1980- 2020 has been utilized to achieve the research objectives. To test the impact of public finance management on economic growth and development, the study will use Seeming Unrelated Regression Equation (SURE) Modelling that allows researchers to model multiple equations with interdependent variables. The advantages of using SUR are that it efficiently allows estimation of relationships between variables by combining information on different equations and SUR test restrictions that involve parameters in different equations. The findings have shown that there is a positive relationship between efficient public finance management and economic growth/development. The findings also show that efficient public finance management has an indirect positive impact on economic growth and development. Corruption has a negative impact on economic growth and development. It results in an efficient allocation of government resources and thereby improves economic growth and development. The study recommends that governments who aim to stimulate economic growth and development should target and strengthen public finance management policies or strategies. Keywords: corruption, economic growth, economic development, public finance management, fiscal decentralization

Procedia PDF Downloads 44813857 Cost Effectiveness of Transcatheter Aortic Valve Replacement vs Surgical Aortic Valve Replacement in a Low-Middle Income Country
Authors: Vasuki Rayapati, Bhanu Duggal
Abstract:
Trans catheter aortic valve replacement (TAVR) is the recommended treatment over surgical aortic valve replacement (SAVR) for high-risk groups, patients >75 years of age with severe symptomatic Aortic stenosis (AS). In high income countries TAVR is more cost effective because of – i) Reduction in total length of stay including less number of days in ICU ii) Non-procedural costs like cost of general anaesthesia are higher for SAVR. In India, there are two kinds of hospitals – Public and Private. Most patients visit public sector hospitals than private sector hospitals. In a LMIC like India, especially in the Public health sector cost of TAVR is prohibitive. In a small study from three (public) hospitals in India, it was envisaged that cost of TAVR should decrease at least by 2/3 to be a cost effective option in Public health sector for severe AS.Keywords: cost effectiveness, TAVR vs SAVR, LMIC, HTA

Authors: Arif Ali, Divya Raj Sapkota
Abstract:
In this paper, we explain gravitational attraction as the consequence of the dynamics of four-dimensional bodies and the consequent distortion of space. This approach provides an alternative direction to understand various physical phenomena based on the existence of the fourth spatial dimension. For this interpretation, we formulate the acceleration due to gravity and orbital velocity based on the accelerating expansion of three-dimensional symmetric bodies. It is also shown how distortion in space caused by the dynamics of four-dimensional bodies counterbalances the effect of expansion. We find that the motion of four-dimensional bodies through four-dimensional space leads to gravitational attraction, and the expansion of bodies leads to surface gravity. Thus, dynamics in the fourth spatial dimension provide an alternative explanation to gravity. Keywords: dimensions, four, gravity, voluceleration

Authors: Yessenia Parrado, Ana Barbosa, Daniela Mahe, Sebastian Toro, Jhon Garcia
Abstract:
The purpose of this work is to present the identification and characterization of the barriers and enablers to public innovation in the Central Region of Colombia from a mixed methodology in a research carried out in 2020 by the Laboratory of Innovation, Creativity and New Technologies of the National University of Colombia in alliance with the National Planning Department. Based on the research, the index of barriers to regional and departmental public innovation was built, which reflects the level of difficulty of the territorial entities to overcome the barriers present around three dimensions: organizational structure of the entity, generation of public value, and governance processes. The index was built from the item response methodology and the multiple correspondence analysis from the application of an institutional information form for public entities and a perception form for public servants. This investigation had the participation of 36 entities and 1038 servers and servants from the departments of Huila, Meta, Boyacá, Cundinamarca, Tolima, and the Capital District. In this exercise, it was identified that the departmental indices range between 13 and 44 and that the regional index was 30 out of 100. From the analysis of the information, it was possible to establish that the main barriers are the lack of specialized agencies for public innovation exercises, lack of qualified personnel and work methodologies for public innovation, inadequate information management, lack of feedback between the learning from governmental and non-governmental entities, the inability of the initiatives to generate binding participation mechanisms and the lack of qualification of citizens to participate in these processes. Keywords: item response, public innovation, quantitative analysis, compared analysis

Authors: Omaimah Algohary
Abstract:
Kaolin is one of the principle intestinal adsorbents, has traditionally been used internally in the treatment of various enteric disorders, colitis, enteritis, dysentery, and diarrhea associated with food and alkaloidal poisoning and in traveler’s diarrhea. It binds to and traps bacteria and its toxins and gases in the gut. It also binds to water in the gut, which helps to make the stools firmer, hence giving symptomatic relief. Metronidazole is a synthetic antibacterial agent that is used primarily in the treatment of various anaerobic infections such as intra-abdominal infections, antiprotozoal, and as amebicidal. The need for safe, therapeutically effective antidiarrheal combination continuously lead to effective treatment. Metronidazol used for treatment of anaerobic bacteria and kaolin , when administered simultaneously, Metronidazole–Kaolin interactions have been reported by FDA but not studied. This project is the first to study the effect of Metronidazole–Kaolin interactions on the antimicrobial activity of metronidazole. Agar diffusion method performed to test the antimicrobial activity of metronidazole–kaolin antidiarrheal combination from aqueous solutions at an in-vivo simulated pHs conditions that obtained at 37+0.5 °C on Helicobacter pylori as anaerobic bacteria and E.coli as aerobic bacteria and used as a control for the technique. The antimicrobial activity of metronidazole combination as 1:1 and 1:2 with kaolin was abolished in acidic media as no zones of inhibition shown compared to only metronidazole that used as a control. In alkaline media metronidazole combination as 1:1 and 1:2 with kaolin showed diminutive activity compared to the control. These results proved that the kaolin adsorb metronidazole and abolish its antimicrobial activity and such combination should be avoided. Keywords: kaolin, metronidazole, interaction, Helicobacter pylori. Procedia PDF Downloads 44813851 The Effects of Acid Rain, Smog Cars on Antioxidant Systems, Associated Enzyme and H⁺-ATPase Activity in Rice Cultivars (Oriza sativa L.)
Authors: Heidarali Malmir
Abstract:
The effects of acid rain (AR), smog’s cars (SC), and combined AR+SC on the antioxidants enzymes, lipid-soluble antioxidants, and water-soluble antioxidants were studied in the two cultivars of rice. The results showed that simulated AR significantly increased the total glutathione (TGSH), thiobarbituric acid (TBA), and α-tocopherol, accompanied by decreases in dry weight and leaves area in the two cultivars, and this change was more obvious in Shirudi cultivar than in Aus cultivar (p≤0.05). Under SC stress cultivar shirudi had higher H+-ATPase, glutathione peroxidase (GSH-px), and catalase (CAT) activities than cultivar Aus. The results of superoxide dismutase (SOD) activity, TGSH, and α-tocopherol levels affected by AR treatments were very different to those of SOD activity, TGSH, and α-tocopherol levels, as shown in SC treatment. It seems that SOD activity coupled with the water-soluble antioxidants and α-tocopherol levels correlated with the lipid-soluble antioxidants. It is suggested that α-tocopherol increases H+-ATPase activity. Keywords: H+-ATPase, membrane permeability, lipid soluble antioxidants, water soluble antioxidants, associated enzyme

Authors: Eirini Oikonomopoulou
Abstract:
Paper is dealing with gentrification of a poor central historic district of Fener and Balat in Istanbul, Turkey and propose ålans and principles of a neighborhood. Istanbul is located in a special geographic place, just in the meeting of Europe and Asia and it has a long and great history, facts that had affected the urban form of the city. Trough the time different civilizations inhabited in the city and they changed it by giving different character to its parts. The modernization of Istanbul brought western ideas into the historic organic urban fabric and put in the first priority the need for a clear and strong car-road/highway network in order to improve the car accessibility along the city. Following that model, transformation of public spaces was based on the driving experience. New public spaces was formulated to be the new symbol of Turkish Republic, to give a beautiful and clean image of the modern Turkish city, as well as work as landmarks across the highway network. Even if city is upgraded, bad quality neighborhoods still exist, far and near to the historic city center. One of them is Fener/Balat, which is located in Fatih district on the European side of Istanbul. This project aims to analyze the urban qualities of that neighborhood (mahalle) and propose a better, qualitative urban space towards a denser, greener and more inclusive neighborhood which could be an example for the whole city. Keywords: urban design, upgrade neighborhood, Istanbul, sustanability

Authors: Sulamain Alharbi, Mohammad Khiyami, Reda Amasha, Bassam Al-Johny, Hesham Khalil, Milton Wainwrigh
Abstract:
Nematodes, Rotifers and Tardigrades (NRT) are extreme-tolerant invertebrates which can survive long periods of stasis brought about by extreme drying and cold. They can also resist the effects of UV radiation, and as a result could act as vehicles for the panspermic transfer of microorganisms. Here we show that NRT contain a variety of bacteria and fungi within their bodies in which environment they could be protected from the extremes of the space and released into new cosmic environments. Diatoms were also shown to contain viable alga and Escherichia coli and so could also act as panspermic vehicles for the transfer of these and perhaps other microbes through space. Although not studied here, NRT, and possibly diatoms, also carry protozoa and viruses within their bodies and could act as vehicles for the panspermic transfer of an even wider range of microbes than shown here. Keywords: extromophiles, diatoms, panspermia, survival in space

Authors: Han Xiao, Ruan Qi, Zhang Lei, Qi Yan
Abstract:
Space environment simulator is a facility used to conduct thermal test for spacecraft, and vacuum vessel is the main body of it. According to the requirements for thermal tests of the spacecraft and its solar array panels, the primary vessel and the side vessels are designed to be a combinative structure connected with aperture, which ratio reaches 0.7. Since the vacuum vessel suffers 0.1MPa external pressure during the process of thermal test, in order to ensure the simulator’s reliability and safety, it’s necessary to calculate the vacuum vessel’s intensity and stability. Based on the impact of large openings to vacuum vessel structure, this paper explored the reinforce design and analytical way of vacuum vessel with large openings, using a large space environment simulator’s vacuum vessel design as an example. Tests showed that the reinforce structure is effective to fulfill the requirements of external pressure and the gravity. This ensured the reliability of the space environment simulator, providing a guarantee for developing the spacecraft. Keywords: vacuum vessel, large opening, space environment simulator, structure design

Authors: Amira Kalifa, Faïek Errouissi
Abstract:
The seasonal changes and the daily activity of Stomoxys calcitrans (Diptera: Muscidae) were examined, using Vavoua traps, in a dairy cattle farm in Zaouiet Sousse, the Sahel area of Tunisia during May 2014 to October 2014. Over this period, a total of 4366 hematophagous diptera were captured and Stomoxys calcitrans was the most commonly trapped species (96.52%). Analysis of the seasonal activity, showed that S.calcitrans is bivoltine, with two peaks: a significant peak is recorded in May-June, during the dry season, and a second peak at the end of October, which is quite weak. This seasonal pattern would depend on climatic factors, particularly the temperature of the manure and that of the air. The activity pattern of Stomoxys calcitrans was diurnal with seasonal variations. The daily rhythm shows a peak between 11:00 am to 15:00 pm in May and between 11:00 am to 17:00 pm in June. These vector flies are important pests of livestock in Tunisia, where they are known as a mechanical vector of several pathogens and have a considerable economic and health impact on livestock. A better knowledge of their ecology is a prerequisite for more efficient control measures. Keywords: cattle farm, daily rhythm, Stomoxys calcitrans, seasonal activity
Procedia PDF Downloads 27213844 The Associations between Self-Determined Motivation and Physical Activity in Patients with Coronary Heart Disease
Authors: I. Hua Chu, Hsiang-Chi Yu, Hsuan Su
Abstract:
Purpose: To examine the associations between self-determined motivation and physical activity in patients with coronary heart disease (CHD) in a longitudinal study. Methods: Patients with CHD were recruited for this study. Their motivations for exercise were measured by the Behavioral Regulation in Exercise Questionnaire-2 (BREQ-2). Physical activity was assessed using the 7-day physical activity recall questionnaire. Duration and energy expenditure of moderate to vigorous physical activity (MVPA) were used in data analysis. All outcome measures were assessed at baseline and 12 months follow up. Data were analyzed using Pearson correlation analysis and regression analysis. Results: The results of the 45 participants (mean age 60.24 yr; 90.2% male) revealed that there were significant negative correlations between amotivation at baseline and duration (r=-.295, p=.049) and energy expenditure (r=-.300, p=.045) of MVPA at 12 months. In contrast, there were significant positive correlations between calculated relative autonomy index (RAI) at baseline and duration (r=.377, p=.011) and energy expenditure (r=.382, p=.010) of MVPA at 12 months. There was no significant correlation between other subscales of the BREQ-2 and duration or energy expenditure of MVPA. Regression analyses revealed that RAI was a significant predictor of duration (p=.011) and energy expenditure (p=.010) of MVPA at 12 months follow-up. Conclusions: These results suggest that the relative degree of self-determined motivation could predict long-term MVPA behaviors in CHD patients. Physical activity interventions are recommended to target enhancing one’s identified and intrinsic motivation to increase the likelihood of physical activity participation in this population. Keywords: self-determined motivation, physical activity, coronary heart disease, relative autonomy index (RAI)

Authors: Xin Jie Li
Abstract:
Due to population agglomeration, huge scale, and complex activities, megacities have become risk centers. To resist the risks brought by development uncertainty, the construction of resilient cities has become a common strategic choice for megacities. As a key link in promoting the modernization of the national governance system and governance capacity, optimizing the layout of national land space that focuses on ecology, production, and life and improving the rationality of spatial resource allocation are conducive to fundamentally promoting the resilience construction of megacities. Therefore, based on the perspective of territorial space governance, this article explores the potential risks faced by the territorial space of megacities and proposes possible paths for the resilience construction of megacities from four aspects: promoting the construction of a resilience system throughout the entire life cycle, constructing a disaster prevention and control system with ecological resilience, creating an industrial spatial pattern with production resilience, and enhancing community resilience to anchor the front line of risk response in megacities. Keywords: mega cities, potential risks, resilient city construction, territorial and spatial governance

The escalating issues of overcrowding, lack of space, and land shortage for full-body burial in the urbanized area of Kuala Lumpur have raised a concern to this alarming situation. There is a necessity to address these problems through the incorporation of sustainable development in the making of urban cemeteries to ensure a holistic approach. Recorded site observation of cemeteries’ area has been employed as a means of data collection and interpreted by conducting spatial analysis. The spatial analysis entails the assessment of form and function in accordance with sustainable design principles. The finding exhibits the dimensional layout of Muslim cemeteries were problematics due to the tension that exists between ritual practices and space organization set-up by the local authorities. This article concludes by providing conceptual guidelines for the purpose of Muslim cemetery development in the future. Keywords: cemetery, metropolitan, spatial analysis, sustainable design practice

Authors: Selamawit Mulubrhan Gidey
Abstract:
Adama city, having an area of 29.86 km2, is one of the main cities in Ethiopia experiencing rapid growth in business and construction activities which in turn with an increasing number of vehicles at an alarming rate. For this reason, currently, there is an attempt to develop public transport assignment modeling in the city. Still, there is a huge gap in developing public transport assignments along the road segments of the city with operational and safety performance due to high traffic volume. Thus, the introduction of public transport assignment modeling in Adama City can have a massive impact on the road safety and capacity problem in the city. City transport modeling is important in city transportation planning, particularly in overcoming existing transportation problems such as traffic congestion. In this study, the Adama City transportation model was developed using the PTV VISUM software, whose transportation modeling is based on the four-step model of transportation. Based on the traffic volume data fed and simulated, the result of the study shows that the developed model has better reliability in representing the traffic congestion conditions in Adama city, and the simulation clearly indicates the level of congestion of each route selected and thus, the city road administrative office can take managerial decisions on public transport assignment so as to overcome traffic congestion executed along the selected routes. Keywords: trip modelling, PTV VISUM, public transport assignment, congestion

Authors: Mathew Wakefield, Matthew Mitchell, Lisa Wise, Christopher McCarthy
Abstract:
The properties of memory representations in artificial neural networks have cognitive implications. Distributed representations that encode instances as a pattern of activity across layers of nodes afford memory compression and enforce the selection of a single point in instance space. These encoding schemes also appear to distort the representational space, as well as trading off the ability to validate that input information is within the bounds of past experience. In contrast, a localist representation which encodes some meaningful information into individual nodes in a network layer affords less memory compression while retaining the integrity of the representational space. This allows the validity of an input to be determined. The validity (or familiarity) of input along with the capacity of localist representation for multiple instance selections affords a memory sampling approach that dynamically balances the bias-variance trade-off. When the input is familiar, bias may be high by referring only to the most similar instances in memory. When the input is less familiar, variance can be increased by referring to more instances that capture a broader range of features. Using this approach in a localist instance memory network, an experiment demonstrates a relationship between representational conflict, generalization performance, and memorization demand. Relatively small sampling ranges produce the best performance on a classic machine learning dataset of visual objects. Combining memory validity with conflict detection produces a reliable confidence judgement that can separate responses with high and low error rates. Confidence can also be used to signal the need for supervisory input. Using this judgement, the need for supervised learning as well as memory encoding can be substantially reduced with only a trivial detriment to classification performance. Keywords: artificial neural networks, representation, memory, conflict monitoring, confidence

Authors: Inuwa Abdu Ibrahim
Abstract:
Public officials are required to posses certain values to adequately protect public interest, by being leaders that are servants of the people. The reality in Nigeria is that leaders rule as masters of the people rather than servants. The paper looked at control and control systems of administration in Nigeria, its resultant consequences and ways of achieving true control of administrators and administration. Secondary source of data was adopted for the research. It concludes that the keys to administrative efficiency and effectiveness through control are implementation of the already existing procedures and laws, as well as commitment on the part of public officials. Keywords: Accountability, Fraud, Administration, Nigeria

Authors: Ogbonnaya Nwokoro, Florence O. Anya
Abstract:
Nutritional factors relating to the production of linamarase from Lactobacillus delbrueckii NRRL B–763 were investigated. The microorganism was cultivated in a medium containing 1% linamarin. Enzyme was produced using a variety of carbon substrates but the highest enzyme activity was detected in the presence of salicin (522 U/ml) after 48 h while the lowest yield was observed with CM cellulose (38 U/ml) after 72 h. Enzyme was not produced in the presence of cellobiose. Among a variety of nitrogen substrates tested, peptone supported maximum enzyme production (412 U/ml) after 48 h. Lowest enzyme production was observed with urea (40 U/ml). Organic nitrogen substrates generally supported higher enzyme productivity than inorganic nitrogen substrates. Enzyme activity was observed in the presence of Mn2+ (% relative activity = 216) while Hg2+ was inhibitory (% relative activity = 28). Locally-formulated media were comparable to MRS broth in supporting linamarase production by the bacterium. Higher enzyme activity was produced in media with surfactant than in media without surfactant. The enzyme may be useful in enhanced degradation of cassava cyanide. Keywords: linamarase, locally formulated media, carbon substrates, nitrogen substrates, metal ions
