70 Antioxidant Characteristics of Serbian Conifers

Abstract:

Many plants possess antioxidant ingredients that provides efficacy by additive or synergistic activities. Present article highlights an antioxidant capacity of Serbian conifer plants. Antioxidant activities of the crude extracts were assessed using different assays. In this study, quantities of phenolic compounds (total phenols, flavonoids, tannins and proanthocyanidins), contents of photosynthetic pigments (chlorophyll a and b and carotenoids), soluble proteins and proline were examined. MDA quantities and ability of extracts to remove reactive nitrogen and oxygen species (RNOS) were also investigated. Furthermore, antioxidant activities of extracts against DPPH∙, ferric reducing antioxidant power, permanganate reducing antioxidant capacity were also determined. According to almost all used assays, antioxidant and scavenging capacities of silver fir (Abies alba Mill.), and Douglas fir (Pseudotsuga menziesii) were superior compared to spruce. Presented results implicated that leaves of Douglas fir and silver fir possessed outstanding antioxidant characteristics that could diminish damage caused by oxygen radicals which are responsible for many of the bodily changes and susceptibility to different diseases.

69 In-Vivo Association of Multivalent 11 Zinc Fingers Transcriptional Factors CTCF and Boris to YB-1 in Multiforme Glioma-RGBM Cell Line

Abstract:

CTCF is a unique, highly conserved and ubiquitously expressed 11 zinc finger (ZF) transcriptional factor with multiple target sites. It is able to bind to various target sequences to perform different regulatory roles including promoter activation or repression, creating hormone-responsive gene silencing element, and functional block of enhancer-promoter interactions. The binding of CTCF to the essential binding site is through the combination of different ZF domain. On the other hand, BORIS for brother of the regulator of imprinted sites, which expressed only in the testis and certain cancer cell line is homology to CTCF 11 ZF domains. Since both transcriptional factors share the same ZF domains hence there is a possibility for both to bind to the same target sequences. In this study, the interaction of these two proteins to multi-functional Y-box DNA/RNA-binding factor, YB-1 was determined. The protein-protein interaction between CTCF/YB-1 and BORIS/YB-1 were discovered by Co-immuno-precipitation (CO-IP) technique through reciprocal experiment from RGBM total cell lysate. The results showed that both CTCF and BORIS were able to interact with YB-1 in Glioma RGBM cell line. To the best of our knowledge, this is the first findings demonstrating the ability of BORIS and YB-1 to form a complex in vivo.

68 Bioactivity Evaluation of Cucurbitin Derived Enzymatic Hydrolysates

Abstract:

After cold pressing of pumpkin oil, the defatted oil cake (PUOC) was utilized as raw material for processing of bio-functional hydrolysates. In this study, the in vitro bioactivity of an alcalase (AH) and a pepsin hydrolysate (PH) prepared from the major pumpkin 12S globulin (cucurbitin) are compared. The hydrolysates were produced at optimum reaction conditions (temperature, pH) for the enzymes, during 60min. The bioactivity testing included antioxidant and angiotensin I converting enzyme inhibitory activity assays. The hydrolysates showed high potential as natural antioxidants and possibly antihypertensive agents in functional food or nutraceuticals. Additionally, preliminary studies have shown that both hydrolysates could exhibit modest α-amylase inhibitory activity, which indicates on their hypoglycemic potential.

67 Green Delivery Systems for Fruit Polyphenols

Abstract:

Green solvents are environmentally friendly and greatly improve the sustainability of chemical processes. There is a growing interest in the green extraction of polyphenols from fruits. In this study, we consider three Natural Deep Eutectic Solvents (NADES) systems based on choline chloride as a hydrogen bond acceptor and malic acid, urea, and fructose as hydrogen bond donors. NADES systems were prepared by heating and stirring, ultrasound, and microwave (MW) methods. Sour cherry pomace was used as a natural source of polyphenols. Polyphenol extraction from cherry pomace was performed by ultrasound-assisted extraction and microwave-assisted extraction and compared with conventional heat and stirring method extraction. It was found that MW-assisted preparation of NADES was the fastest, requiring less than 30 s. Also, MW extraction of polyphenols was the most rapid, with less than 5 min necessary for the extract preparation. All three NADES systems were highly efficient for anthocyanin extraction, but the most efficient was the system with malic acid as a hydrogen bond donor (yield of anthocyanin content was enhanced by 62.33% after MW extraction with NADES compared with the conventional solvent).

66 Neural Network Based Approach of Software Maintenance Prediction for Laboratory Information System

Abstract:

Software maintenance phase is started once a software project has been developed and delivered. After that, any modification to it corresponds to maintenance. Software maintenance involves modifications to keep a software project usable in a changed or a changing environment, to correct discovered faults, and modifications, and to improve performance or maintainability. Software maintenance and management of software maintenance are recognized as two most important and most expensive processes in a life of a software product. This research is basing the prediction of maintenance, on risks and time evaluation, and using them as data sets for working with neural networks. The aim of this paper is to provide support to project maintenance managers. They will be able to pass the issues planned for the next software-service-patch to the experts, for risk and working time evaluation, and afterward to put all data to neural networks in order to get software maintenance prediction. This process will lead to the more accurate prediction of the working hours needed for the software-service-patch, which will eventually lead to better planning of budget for the software maintenance projects.

65 Mentha piperita Formulations in Natural Deep Eutectic Solvents: Phenolic Profile and Biological Activity

Abstract:

Natural deep eutectic solvents (NADES) represent a class of modern systems that have been developed as a green alternative to toxic organic solvents, which are commonly used as extraction media. It has been considered that hydrogen bonding is the main interaction leading to the formation of NADES. The aim of this study was phytochemical characterization and determination of the antioxidant and antibacterial activity of Mentha piperita leaf extracts obtained by six choline chloride-based NADES. NADES were prepared by mixing choline chloride with different hydrogen bond donors in 1:1 molar ratio following the addition of 30% (w/w) water. The mixtures were then heated (60 °C) and stirred (650 rpm) until the clear homogenous liquids were obtained. The Mentha piperita extracts were prepared by mixing 75 mg of peppermint leaves with 1 mL of NADES following by the heating and stirring (60 °C, 650 rpm) within 30 min. The content of six phenolics in extracts was determined using HPLC-PDA. The dominant compounds presented in peppermint leaves - rosmarinic acid and luteolin 7-O-glucoside, were extracted by NADES at a similar level as 70% ethanol. The microdilution method was applied to test the antibacterial activity of extracts. Compared with 70% ethanol, all NADES systems showed higher antibacterial activity towards Pseudomonas aeruginosa (Gram -), Staphylococcus aureus (Gram +), Escherichia coli (Gram -), and Salmonella enterica (Gram -), especially NADES containing organic acids. The majority of NADES extracts showed a better ability to neutralize DPPH radical than conventional solvent and similar ability to reduce Fe3+ to Fe2+ ions in FRAP assay. The obtained results introduce NADES systems as the novel, sustainable, and low-cost solvents with a variety of applications.

64 Study of Electron Cyclotron Resonance Acceleration by Cylindrical TE₀₁₁ Mode

Abstract:

In this work, we present results from analytical and numerical studies of the electron acceleration by a TE₀₁₁ cylindrical microwave mode in a static homogeneous magnetic field under electron cyclotron resonance (ECR) condition. The stability of the orbits is analyzed using the particle orbit theory. In order to get a better understanding of the interaction wave-particle, we decompose the azimuthally electric field component as the superposition of right and left-hand circular polarization standing waves. The trajectory, energy and phase-shift of the electron are found through a numerical solution of the relativistic Newton-Lorentz equation in a finite difference method by the Boris method. It is shown that an electron longitudinally injected with an energy of 7 keV in a radial position r=Rc/2, being Rc the cavity radius, is accelerated up to energy of 90 keV by an electric field strength of 14 kV/cm and frequency of 2.45 GHz. This energy can be used to produce X-ray for medical imaging. These results can be used as a starting point for study the acceleration of electrons in a magnetic field changing slowly in time (GYRAC), which has some important applications as the electron cyclotron resonance ion proton accelerator (ECR-IPAC) for cancer therapy and to control plasma bunches with relativistic electrons.

63 Cognitive Semantics Study of Conceptual and Metonymical Expressions in Johnson's Speeches about COVID-19

Abstract:

The study is an attempt to investigate the conceptual metonymies is used in political discourse about COVID-19. Thus, this study tries to analyze and investigate how the conceptual metonymies in Johnson's speech about coronavirus are constructed. This study aims at: Identifying how are metonymies relevant to understand the messages in Boris Johnson speeches and to find out how can conceptual blending theory help people to understand the messages in the political speech about COVID-19. Lastly, it tries to Point out the kinds of integration networks are common in political speech. The study is based on the hypotheses that conceptual blending theory is a powerful tool for investigating the intended messages in Johnson's speech and there are different processes of blending networks and conceptual mapping that enable the listeners to identify the messages in political speech. This study presents a qualitative and quantitative analysis of four speeches about COVID-19; they are said by Boris Johnson. The selected data have been tackled from the cognitive-semantic perspective by adopting Conceptual Blending Theory as a model for the analysis. It concludes that CBT is applicable to the analysis of metonymies in political discourse. Its mechanisms enable listeners to analyze and understand these speeches. Also the listener can identify and understand the hidden messages in Biden and Johnson's discourse about COVID-19 by using different conceptual networks. Finally, it is concluded that the double scope networks are the most common types of blending of metonymies in the political speech.

62 On the Application of Heuristics of the Traveling Salesman Problem for the Task of Restoring the DNA Matrix

Abstract:

The traveling salesman problem (TSP) is a well-known optimization problem that seeks to find the shortest possible route that visits a set of points and returns to the starting point. In this paper, we apply some heuristics of the TSP for the task of restoring the DNA matrix. This restoration problem is often considered in biocybernetics. For it, we must recover the matrix of distances between DNA sequences if not all the elements of the matrix under consideration are known at the input. We consider the possibility of using this method in the testing of distance calculation algorithms between a pair of DNAs to restore the partially filled matrix.

61 Application of Signature Verification Models for Document Recognition

Abstract:

In modern economic conditions, the question of the possibility of correct recognition of a signature on digital documents in order to verify the expression of will or confirm a certain operation is relevant. The additional complexity of processing lies in the dynamic variability of the signature for each individual, as well as in the way information is processed because the signature refers to biometric data. The article discusses the issues of using artificial intelligence models in order to improve the quality of signature confirmation in document recognition. The analysis of several possible options for using the model is carried out. The results of the study are given, in which it is possible to correctly determine the authenticity of the signature on small samples.

60 Solving the Pseudo-Geometric Traveling Salesman Problem with the “Union Husk” Algorithm

Abstract:

This study explores the pseudo-geometric version of the extensively researched Traveling Salesman Problem (TSP), proposing a novel generalization of existing algorithms which are traditionally confined to the geometric version. By adapting the "onion husk" method and introducing auxiliary algorithms, this research fills a notable gap in the existing literature. Through computational experiments using randomly generated data, several metrics were analyzed to validate the proposed approach's efficacy. Preliminary results align with expected outcomes, indicating a promising advancement in TSP solutions.

59 Beginner Steps of the First Dendrochronology Lab in Montenegro - Dendrochronology Research in The Bosnian Pine (Pinus heldreichii) Forests

Abstract:

Officially, 60% of Montenegrin territory is covered in forests, but they are continually being destroyed by illegal cutting, concession politics and wildfires. Montenegrin Ecologists Society started the first dendrochronology lab in Montenegro, and data collection began in the Summer of 2021. The cores were taken from 3 localities around the peak Lisac on the mt. Prekornica, where biggest P.heldreichii forests existed until recent huge wildfires. This research is the first step towards comprehensive dendrochronology research in Montenegro. It will show how old certain forest stands of Pinus heldreichii on mountain Prekornica are, that were not destroyed in huge wildfires from the recent years. It will also show how do they correlate between each other. Per locality 15 trees were sampled. Electric sanders (150 - 2000) were used for preparation. Cores were scanned, then measured in CooRecorder. Analysis is done in Cofecha. Process will be repeated with Lintab 6 and TSAP (Time Series Analysis and Presentation for Dendrochronology and Related Applications) - Win Scientific software by Rinntech. Since this is the first dendrochronology research entirely done in Montenegro it is a foundation for the dendroclimatology research. Besides, it’ll contribute to the understanding of the life of these forests in this part of its areal, and in designing good management practices.

58 Frobenius Manifolds Pairing and Invariant Theory

Abstract:

The orbit space of an irreducible representation of a finite group is a variety with the ring of invariant polynomials as a coordinate ring. The invariant ring is a polynomial ring if and only if the representation is a reflection representation. Boris Dubrovin shows that the orbits spaces of irreducible real reflection representations acquire the structure of polynomial Frobenius manifolds. Dubrovin's method was also used to construct different examples of Frobenius manifolds on certain reflection representations. By successfully applying Dubrovin’s method on non-polynomial invariant rings of linear representations of dicyclic groups, it gives some results that magnify the relation between invariant theory and Frobenius manifolds.

57 Using High Performance Computing for Online Flood Monitoring and Prediction

Abstract:

The main goal of this article is to describe the online flood monitoring and prediction system Floreon+ primarily developed for the Moravian-Silesian region in the Czech Republic and the basic process it uses for running automatic rainfall-runoff and hydrodynamic simulations along with their calibration and uncertainty modeling. It takes a long time to execute such process sequentially, which is not acceptable in the online scenario, so the use of high-performance computing environment is proposed for all parts of the process to shorten their duration. Finally, a case study on the Ostravice river catchment is presented that shows actual durations and their gain from the parallel implementation.

56 The Locus of Action - Tinted Windows

Abstract:

This research is about the ways artists and scientists deal with (and endure) new meaning and comprehend and construct the world. The project reflects on the intense connection between comprehension and construction and their place of creation – the ‘locus of action’. It seeks to define a liquid form of understanding and analysis capable of approaching our complex liquid world as discussed by Zygmunt Bauman. The aim is to establish a multi-viewpoint theoretical approach based on the dynamic concept of the Flâneur as introduced by Baudelaire, replacing single viewpoint categorization. This is coupled with the concept of thickening as proposed by Clifford Geertz with its implication of interaction between multi-layers of meaning. Here walking and looking is introduced as a method or strategy, a model or map, providing a framework of understanding in conditions of hybridity and change.

55 On Some Properties of Maximal Prefix Codes

Abstract:

We study the properties of maximal prefix codes. The codes have many applications in computer science, theory of formal languages, data processing and data classification. Practical application is based on the representation of the maximal prefix codes as a sequence of words in a specific order. Our approach to study uses finite state automata (so-called flower automata) for the representation of prefix codes. An important task is the decomposition of prefix codes into prime prefix codes (factors). We discuss the properties of such prefix code decompositions. A linear time algorithm is designed which find the prime decomposition. To verify the correctness of the proposed algorithms, we implemented a system computer algebra GAP.

54 A 5-V to 30-V Current-Mode Boost Converter with Integrated Current Sensor and Power-on Protection

Abstract:

This paper presents a 5-V to 30-V current-mode boost converter for powering the drive circuit of a micro-electro-mechanical sensor. The design of a transconductance amplifier and an integrated current sensing circuit are presented. In addition, essential building blocks for power-on protection such as a soft-start and clamp block and supply and clock ready block are discussed in details. The chip is fabricated in a 0.18-μm CMOS process. Measurement results show that the soft-start and clamp block can effectively limit the inrush current during startup and protect the boost converter from startup failure.

53 National Project 'Environment' of Russian Federation as a Management Tool in Achieving SDGs

Abstract:

Priority national projects have become an essential phenomenon in the Russian Federation. Both regional and local government institutions and a significant part of the society have been involved in their implementation. The scale and multispectricity of the national projects give a reason to believe that their concept is beyond the scope of the individual state programs. The national project “environment” contains federal projects on waste management, water, and air quality, ecotourism development, and biodiversity conservation highlights the importance of the preservation and restoration of Volga River and Lake Baikal ecosystems. This study assesses the national projects according to their relativeness with the current SDGs (i.e., SGD 14 and 15), evaluates the methodology of the projects. The paper considers the peculiarities of the national projects as strategic management tools as well as the possibility of amending the project objective indicators. Conclusion on the effectiveness of NP in terms of achieving SDGs is provided.

Abstract:

This work is devoted to a problem of aesthetic quality determinism in the context of contemporary art. The object of study is photography regarding as a kind of art which demands specific system of quality marking. Objective: To define aesthetic criteria in photography art. For current searching different kind of texts by such powerful authors like Clement Greenberg and Rosalind Krauss, Theodor Adorno and Herbert Marcuse, Charlott Cotton and Boris Groys, Viktor Miziano and Ekaterina Degot' were analyzed. Before all, there are two different kinds of photography: the classic art photography (by Ansel Adams) and the photography as kind of art (by Andreas Gursky). In this text we are talking about the photography as kind of art. The main principle of current searching is synthesis of two different approaches: modernism and postmodernism. This method helps us to define uniform criteria of aesthetic quality in photography as kind of art. The criteria mentioned in conclusion paragraph are: aesthetic rationality, aesthetic economy, awareness (using photographic technics or references), and intention to go beyond form, practice and method.

The idea of a networked economy has gained momentum in the data domain as collaborative approaches for data management emerge. Traditional organizational knowledge consequently needs to be extended by comprehensive knowledge about data. The knowledge about data is vital for organizations to ensure that data quality requirements are met and data can be effectively utilized and sovereignly governed. As this specific knowledge has been paid little attention to so far by academics, the aim of the research presented in this paper is to conceptualize it by proposing a “data knowledge model”. Relevant model entities have been identified based on a design science research (DSR) approach that iteratively integrates insights of various industry case studies and literature research.

50 The Impact of Ship Traffic and Harbor Activities on the Atmospheric Pollution in Two Northern Adriatic Ports: Venice and Rijeka

Abstract:

The aim of the POSEIDON project is to quantify the relative contribution of maritime traffic and harbor activities to atmospheric pollutants concentration in four port-cities of the Adriatic Sea. This study focuses on the harbors of Venice and Rijeka. In order to investigate the main pollution sources, emission inventories were used as input for receptor models: PMF (positive matrix factorization) and PCA (principal components analysis); moreover source identification was also conducted using PAHs diagnostic ratios. The ship traffic impact was quantified: i) on gaseous and particulate PAHs, collected using a new method which consisted in a double simultaneous sampling, in different wind sectors; ii) applying PMF to data of metals, PAHs and ions in PM10; iii) using the vanadium concentration according to the Agrawal methodology.

49 Clash of Institutions: Role of Constitutional Courts in Mediating between Institutions

Abstract:

Brexit nudged the British executive towards overriding parliamentary sovereignty in the UK. In 2019, Prime Minister Boris Johnson sought to prorogue parliament to prevent it from debating withdrawal from the UK. In 2022, Pakistan's Prime Minister also tried to nullify the ability of parliament to vote on the constitutional mechanism of a no-confidence vote. In both cases, the apex courts intervened and restored the supremacy of Parliament, averting constitutional crises. This paper examines the legitimacy and power of said courts to intervene in sensitive political and constitutional questions. The research focuses on the administrative law area of judicial review. It examines how in UK and Pakistan practice of judicial review helps mediate constitutional deadlocks between institutions comparatively. This is secondary research employing qualitative, comparative, doctrinal, and analytical methodologies to research a specific area of law from two jurisdictions, using primary and secondary sources.

48 On the Study of All Waterloo Automaton Semilattices

Abstract:

The aim is to study the set of subsets of grids of the Waterloo automaton and the set of covering automata defined by the grid subsets. The study was carried out using the library for working with nondeterministic finite automata NFALib implemented by one of the authors (M. Abramyan) in C#. The results are regularities obtained when considering semilattices of covering automata for the Waterloo automaton. A complete description of the obtained semilattices from the point of view of equivalence of the covering automata to the original Waterloo automaton is given, the criterion of equivalence of the covering automaton to the Waterloo automaton in terms of properties of the subset of grids defining the covering automaton is formulated. The relevance of the subject area under consideration is due to the need to research a set of regular languages and, in particular, a description of their various subclasses. Also relevant are the problems that may arise in some subclasses. This will give, among other things, the possibility of describing new algorithms for the equivalent transformation of nondeterministic finite automata.

47 Modelling of Polymeric Fluid Flows between Two Coaxial Cylinders Taking into Account the Heat Dissipation

Abstract:

Mathematical model based on the mesoscopic theory of polymer dynamics is developed for numerical simulation of the flows of polymeric liquid between two coaxial cylinders. This model is a system of nonlinear partial differential equations written in the cylindrical coordinate system and coupled with the heat conduction equation including a specific dissipation term. The stationary flows similar to classical Poiseuille ones are considered, and the resolving equations for the velocity of flow and for the temperature are obtained. For solving them, a fast pseudospectral method is designed based on Chebyshev approximations, that enables one to simulate the flows through the channels with extremely small relative values of the radius of inner cylinder. The numerical analysis of the dependance of flow on this radius and on the values of dissipation constant is done.

46 Mixed Natural Adsorbents and Oxides for Oil Remediation

Abstract:

The importance of the crude oil refining process is due to the demand for petroleum products such as gasoline, kerosene, asphalt, etc., which are used in daily activities and have a high impact on the global economy. In the processes of oil obtaining and refining, it is common to find problems such as spills on seabed and high energy consumption in processing. In order to quickly and efficiently attack these problems, the use of adsorbents has taken on great importance due to its ease of implementation, as well as the possibility of their regeneration to be reused. In this work, the use of two types of adsorbents is proposed: the first is a natural adsorbent such as aloe vera or nopal, which were lyophilized and hydrophobized to achieve a selectivity in oil adsorption in oil / water mixtures. The second is a mixed iron/nickel oxide, which is specially designed to adsorb the asphaltenes in the heavy fractions of the oil; in addition, this type of adsorbents presents catalytic properties that manage to decompose the heavier fractions of the petroleum in light hydrocarbons, descending thus the energy required for the oil refining process.

45 Efficient Reconstruction of DNA Distance Matrices Using an Inverse Problem Approach

Abstract:

We continue to consider one of the cybernetic methods in computational biology related to the study of DNA chains. Namely, we are considering the problem of reconstructing the not fully filled distance matrix of DNA chains. When applied in a programming context, it is revealed that with a modern computer of average capabilities, creating even a small-sized distance matrix for mitochondrial DNA sequences is quite time-consuming with standard algorithms. As the size of the matrix grows larger, the computational effort required increases significantly, potentially spanning several weeks to months of non-stop computer processing. Hence, calculating the distance matrix on conventional computers is hardly feasible, and supercomputers are usually not available. Therefore, we started publishing our variants of the algorithms for calculating the distance between two DNA chains; then, we published algorithms for restoring partially filled matrices, i.e., the inverse problem of matrix processing. In this paper, we propose an algorithm for restoring the distance matrix for DNA chains, and the primary focus is on enhancing the algorithms that shape the greedy function within the branches and boundaries method framework.

44 A Systematic Review of Prevalence, Gender and Age Differences in Cyberbullying Studies in Croatia

Abstract:

Background: Cyberbullying has become a prevalent issue worldwide, including in Croatia. However, a comprehensive understanding of the extent and nature of cyberbullying in the Croatian context is lacking. Objective: The objective of this systematic review is to evaluate the quality of current research conducted in Croatia on the subject of cyberbullying, identify any gaps in the research, and provide suggestions for future investigations. It examines the prevalence gender and age differences of cyberbullying in Croatia. Participants and Setting: Research is done on secondary data resources (published studies) of cyberbullying in Croatia. The participants in these studies that were included in systematic review are children and youth of all ages residing in Croatia who have been involved in cyberbullying incidents. The setting includes various environments where cyberbullying may occur, such as social media platforms and educational institutions. Methods: To identify pertinent studies on cyberbullying in Croatia, a comprehensive exploration of both international and domestic electronic databases was systematically undertaken. Relevant studies were chosen according to predefined criteria that determined inclusion and exclusion. Key findings from the selected studies were extracted and synthesized, enabling the identification of patterns in the data. Results: A total of 43 studies that fulfilled the inclusion criteria were identified in the review. The prevalence of cyberbullying victimization in Croatia ranged from 7% - 55.3%, with adolescents being the most affected group. The prevalence of cyberbullying perpetration was ranging from 3.2% - 30.3%. The most prevalent form of cyberbullying included gossiping and mocking others. Gender and age differences are highlighted. Conclusions: The outcomes of this systematic review highlight the pressing need for targeted interventions and preventative measures to address cyberbullying in Croatia. Additionally, it is crucial to conduct further research to investigate the long-term impacts and potential factors that can help mitigate cyberbullying in the context of Croatia.

43 Assessing Two Protocols for Positive Reinforcement Training in Captive Olive Baboons (Papio anubis)

Abstract:

Positive Reinforcement Training is a well-known methodology which has been reported frequently to be used in captive non-human primates. As a matter of fact, it is an invaluable tool for different purposes related with animal welfare, such as primate husbandry and environmental enrichment. It is also essential to perform some cognitive experiments. The main propose of this pilot study was to establish an efficient protocol to train captive olive baboons (Papio anubis). This protocol seems to be vital in the context of a larger research program in which it will be necessary to train a complete population of around 40 baboons. Baboons were studied at the Veterinary Research Farm of the University of Murcia. Temporally isolated animals were trained to perform three basic tasks. Firstly, they were required to take food prices directly from the researchers’ hands. Then a clicker sound or bridge stimulus was added each time the animal acceded to the reinforcement. Finally, they were trained to touch a target, consisted of a whip with a red ball in its end, with their hands or their nose. When the subject completed correctly this task, it was also exposed to the bridge stimulus and awarded with a food price, such as a portion of banana, orange, apple, peach or a raisin. Two protocols were tested during this experiment. In both of them, there were 6 series of 2min training periods each day. However, in the first protocol, the series consisted in 3 trials, whereas in the second one, in each series there were 5 trials. A reliable performance was obtained with only 6 days of training in the case of the 5-trials protocol. However, with the 3-trials one, 26 days of training were needed. As a result, the 5-trials protocol seems to be more effective than the 3-trials one, in order to teach these three basic tasks to olive baboons. In consequence, it will be used to train the rest of the colony.

42 Epigenetic Mechanisms Involved in the Occurrence and Development of Infectious Diseases

Abstract:

Infectious diseases are one of the most important causes of morbidity and mortality worldwide. These diseases are caused by micro-pathogenic organisms, such as bacteria, viruses, parasites, and fungi. Heritable changes in gene expression that do not involve changes to the underlying DNA sequence are referred to as epigenetics. Emerging evidence suggests that epigenetic mechanisms are important in the emergence and progression of infectious diseases. Pathogens can manipulate host epigenetic machinery to promote their own replication and evade immune responses. The Human Genome Project has provided new opportunities for developing better tools for the diagnosis and identification of target genes. Several epigenetic modifications, such as DNA methylation, histone modifications, and non-coding RNA expression, have been shown to influence infectious disease outcomes. Understanding the epigenetic mechanisms underlying infectious diseases may result in the progression of new therapeutic approaches focusing on host-pathogen interactions. The goal of this study is to show how different infectious agents interact with host cells after infection.

41 Early Cell Cultures Derived from Human Prostate Cancer Tissue Express Tissue-Specific Epithelial and Cancer Markers

Abstract:

The human prostate gland (PG) samples were obtained from patients who had undergone radical prostatectomy for prostate cancer (PC) and used to extract total RNA and prepare the prostate stromal cell cultures (PSCC) and patients-derived organoids (PDO). Growth of the cell cultures was accessed under microscopic evaluation in transmitted light and the marker expression by reverse polymerase chain reaction (RT-PCR), immunofluorescence, and immunoblotting. Some PCR products from prostate tissue, PSCC, and PDO were cloned and sequenced. We found that the cells of early and late passages of PSCC and corresponding PDO expressed luminal (androgen receptor, AR; cytokeratin 18, CK18) and basal (CK5, p63) epithelial markers, the production of which decreased or disappeared in late PSCC and PDO. The PSCC and PDO of early passages from cancer tissue additionally produced cancer markers AMACR, TMPRSS2-ERG, and Ezh2. The expression of TMPRSS2-ERG fusion transcripts was verified by cloning and sequencing the PCR products. The results obtained suggest that early passages of PSCC might be used as a pre-clinical model for the evaluation of early markers of prostate cancer.
