26923 Postbuckling Analysis of End Supported Rods under Self-Weight Using Intrinsic Coordinate Finite Elements
Authors: C. Juntarasaid, T. Pulngern, S. Chucheepsakul
Abstract:
A formulation of postbuckling analysis of end supported rods under self-weight has been presented by the variational method. The variational formulation involving the strain energy due to bending and the potential energy of the self-weight, are expressed in terms of the intrinsic coordinates. The variational formulation is accomplished by introducing the Lagrange multiplier technique to impose the boundary conditions. The finite element method is used to derive a system of nonlinear equations resulting from the stationary of the total potential energy and then Newton-Raphson iterative procedure is applied to solve this system of equations. The numerical results demonstrate the postbluckled configurations of end supported rods under self-weight. This finite element method based on variational formulation expressed in term of intrinsic coordinate is highly recommended for postbuckling analysis of end-supported rods under self-weight.Keywords: postbuckling, finite element method, variational method, intrinsic coordinate

Authors: Can Hu, Huixia Shi, Hongcheng Mei, Jun Zhu, Hongling Guo
Abstract:
Textile fibers are important trace evidence and frequently encountered in criminal investigations. A significant aspect of fiber evidence examination is the determination of fiber dyes. Although several instrumental methods have been developed for dyes detection, the analysis speed is not fast enough yet. A rapid dye analysis method is still needed to further improve the efficiency of case handling. Capillary electrophoresis has the advantages of high separation speed and high separation efficiency and is an ideal method for the rapid analysis of fiber dyes. In this paper, acid dyes used for protein fiber dyeing were determined by a developed short-end injection capillary electrophoresis technique. Five acid red dyes with similar structures were successfully baseline separated within 5 min. The separation reproducibility is fairly good for the relative standard deviation of retention time is 0.51%. The established method is rapid and accurate which has great potential to be applied in forensic setting. Keywords: acid dyes, capillary electrophoresis, fiber evidence, rapid determination

Authors: Moumita Sit, Chaitali Ray
Abstract:
The present article deals with the free vibration analysis of hybrid laminated composite structures with initial stresses developed in the laminates. Generally initial stresses may be developed in the laminates by temperature and moisture effect. In this study, an eight noded isoparametric plate bending element has been used for the finite element analysis of composite plates. A numerical model has been developed to assess the geometric nonlinear response of composite plates based on higher order shear deformation theory (HSDT) considering the Green–Lagrange type nonlinearity. A computer code based on finite element method (FEM) has also been developed in MATLAB to perform the numerical calculations. To validate the accuracy of the proposed numerical model, the results obtained from the present study are compared with those available in published literature. Effects of the side to thickness ratio, different boundary conditions and initial stresses on the natural frequency of composite plates have been studied. The free vibration analysis of a hollow stiffened hybrid laminated panel has also been carried out considering initial stresses and presented as case study. Keywords: geometric nonlinearity, higher order shear deformation theory (HSDT), hybrid composite laminate, the initial stress

Authors: Strahinja Kovacevic, Milica Karadzic Banjac, Jasmina Vitas, Stefan Vukmanovic, Radomir Malbasa, Lidija Jevric, Sanja Podunavac-Kuzmanovic
Abstract:
The present study deals with chemometric regression analysis of quality parameters and the radical scavenging ability of kombucha fermented kefir-like products obtained with winter savory (WS), peppermint (P), stinging nettle (SN) and wild thyme tea (WT) kombucha inoculums. Each analyzed sample was described by milk fat content (MF, %), total unsaturated fatty acids content (TUFA, %), monounsaturated fatty acids content (MUFA, %), polyunsaturated fatty acids content (PUFA, %), the ability of free radicals scavenging (RSA Dₚₚₕ, % and RSA.ₒₕ, %) and pH values measured after each hour from the start until the end of fermentation. The aim of the conducted regression analysis was to establish chemometric models which can predict the radical scavenging ability (RSA Dₚₚₕ, % and RSA.ₒₕ, %) of the samples by correlating it with the MF, TUFA, MUFA, PUFA and the pH value at the beginning, in the middle and at the end of fermentation process which lasted between 11 and 17 hours, until pH value of 4.5 was reached. The analysis was carried out applying univariate linear (ULR) and multiple linear regression (MLR) methods on the raw data and the data standardized by the min-max normalization method. The obtained models were characterized by very limited prediction power (poor cross-validation parameters) and weak statistical characteristics. Based on the conducted analysis it can be concluded that the resulting radical scavenging ability cannot be precisely predicted only on the basis of MF, TUFA, MUFA, PUFA content, and pH values, however, other quality parameters should be considered and included in the further modeling. This study is based upon work from project: Kombucha beverages production using alternative substrates from the territory of the Autonomous Province of Vojvodina, 142-451-2400/2019-03, supported by Provincial Secretariat for Higher Education and Scientific Research of AP Vojvodina. Keywords: chemometrics, regression analysis, kombucha, quality control

Authors: Qingqing Feng, S. Thomas Ng, Frank Xu
Abstract:
Energy consumption in domestic sector has been increasing rapidly in China all along these years. Confronted with environmental challenges, the international society has made a concerted effort by setting the Paris Agreement, the Sustainable Development Goals, and the New Urban Agenda. Thus it’s very important for China to put forward reasonable countermeasures to boost building energy conservation which necessitates looking into the actuality of residential energy end-use profile and its influence factors. In this study, questionnaire surveys have been conducted in Zhuhai city in China, a typical city in hot summer warm winter climate zone. The data solicited mainly include the occupancy schedule, building’s information, residents’ information, household energy uses, the type, quantity and use patterns of appliances and occupants’ satisfaction. Over 200 valid samples have been collected through face-to-face interviews. Descriptive analysis, clustering analysis, correlation analysis and sensitivity analysis were then conducted on the dataset to understand the energy end-use profile. The findings identify: 1) several typical clusters of occupancy patterns and appliances utilization patterns; 2) the top three sensitive factors influencing energy consumption; 3) the correlations between satisfaction and energy consumption. For China with many different climates zones, it’s difficult to find a silver bullet on energy conservation. The aim of this paper is to provide a theoretical basis for multi-stakeholders including policy makers, residents, and academic communities to formulate reasonable energy saving blueprints for hot and humid urban residential buildings in China. Keywords: residential building, energy end-use profile, questionnaire survey, sustainability

Authors: Dale Dzemydiene, Aurelija Burinskiene, Arunas Miliauskas, Kristina Ciziuniene
Abstract:
Data flow and the purpose of reporting the data are different and dependent on business needs. Different parameters are reported and transferred regularly during freight delivery. This business practices form the dataset constructed for each time point and contain all required information for freight moving decisions. As a significant amount of these data is used for various purposes, an integrating methodological approach must be developed to respond to the indicated problem. The proposed methodology contains several steps: (1) collecting context data sets and data validation; (2) multi-objective analysis for optimizing freight transfer services. For data validation, the study involves Grubbs outliers analysis, particularly for data cleaning and the identification of statistical significance of data reporting event cases. The Grubbs test is often used as it measures one external value at a time exceeding the boundaries of standard normal distribution. In the study area, the test was not widely applied by authors, except when the Grubbs test for outlier detection was used to identify outsiders in fuel consumption data. In the study, the authors applied the method with a confidence level of 99%. For the multi-objective analysis, the authors would like to select the forms of construction of the genetic algorithms, which have more possibilities to extract the best solution. For freight delivery management, the schemas of genetic algorithms' structure are used as a more effective technique. Due to that, the adaptable genetic algorithm is applied for the description of choosing process of the effective transportation corridor. In this study, the multi-objective genetic algorithm methods are used to optimize the data evaluation and select the appropriate transport corridor. The authors suggest a methodology for the multi-objective analysis, which evaluates collected context data sets and uses this evaluation to determine a delivery corridor for freight transfer service in the multi-modal transportation network. In the multi-objective analysis, authors include safety components, the number of accidents a year, and freight delivery time in the multi-modal transportation network. The proposed methodology has practical value in the management of multi-modal transportation processes. Keywords: multi-objective, analysis, data flow, freight delivery, methodology

Authors: Farid Amidi-Fazli
Abstract:
Problems and disadvantages of using conventional plastics are more apparent in recent years and have attracted researchers' attention. Polymers from natural resources or bio polymers represent a suitable replacement to overcome to the disadvantages of plastics. But due to the some flaws of bio polymers, using suitable filler almost seems necessary. Nanocrystalline cellulose with low cost and availability can be applied as appropriate filler. In this study nanocrystalline cellulose was produced from cotton Linter and was characterized. The cotton Linter was hydrolyzed in sulfuric acid then neutralized by the two different concentrations of NaOH. The resulted suspension was treated by ultrasound waves. Process efficiency was determined as 90%. The final product was studied using scanning electron microscopy and x-ray diffraction technique. The obtained diagram of XRD experiment confirmed that the produced material was nanocrystalline cellulose. Also percentage of crystallinity was calculated as 84% in the obtained material as well as the size of crystals. It can be said that the applied method is a rapid and easy method for the production of nanocrystalline cellulose. Keywords: nanocrystalline cellulose, crystallinity, XRD, cotton linter

Authors: Maria Isabel Garcia-Planas, Maria Victoria Garcia-Camba
Abstract:
The use of brain stem auditory evoked potential (BAEP) is a common way to study the auditory function of people, a way to learn the functionality of a part of the brain neuronal groups that intervene in the learning process by studying the behaviour of wave VI. The latest advances in neuroscience have revealed the existence of different brain activity in the learning process that can be highlighted through the use of innocuous, low-cost, and easy-access techniques such as, among others, the BAEP that can help us to detect early possible neurodevelopmental difficulties for their subsequent assessment and cure. To date and to the authors' best knowledge, only the latency data obtained, observing the first to V waves and mainly in the left ear, were taken into account. This work shows that it is essential to take into account both ears; with these latest data, it has been possible had diagnosed more precise some cases than with the previous data had been diagnosed as 'normal' despite showing signs of some alteration that motivated the new consultation to the specialist. Keywords: ear, neurodevelopment, auditory evoked potentials, intervals of normality, learning disabilities

Authors: R. Simões, M. Silveirinha
Abstract:
Driven by the need to understand the results of previous research that clearly shows deep unbalances of the media discourses about women and men in spite of the growing numbers of female journalists, our paper aims to progress from the 'what' to the 'why' of these unbalanced representations. Furthermore, it does so at a time when journalism is undergoing a dramatic change in terms of professional practices and in how media organizations are organized and run, affecting women in particular. While some feminist research points to the fact that female and male journalists evaluate the role of the news and production methods in similar ways feminist theorizing also suggests that thought and knowledge are highly influenced by social identity, which is also inherently affected by the experiences of gender. This is particularly important at a time of deep societal and professional changes. While there are persuasive discussions of gender identities at work in newsrooms in various countries studies on the issue will benefit from cases that focus on the particularities of local contexts. In our paper, we present one such case: the case of Portugal, a country hit hard by austerity measures that have affected all cultural industries including journalism organizations, already feeling the broader impacts of the larger societal changes of the media landscape. Can we gender these changes? How are they felt and understood by female and male journalists? And how are these discourses framed by androcentric, feminist and post-feminist sensibilities? Foregrounding questions of gender, our paper seeks to explore some of the interactions of societal and professional forces, identifying their gendered character and outlining how they shape journalism work in general and the production of unbalanced gender representations in particular. We do so grounded in feminist studies of journalism as well as feminist organizational and work studies, looking at a corpus of 20 in-depth interviews of female and male Portuguese journalists. The research findings illustrate how gender in journalism practices interacts with broader experiences of the cultural and economic contexts and show the ambivalences of these interactions in news organizations. Keywords: gender, journalism, newsroom culture, Portuguese journalists

Authors: K. S. Sreejith, C. Shaji
Abstract:
Though both Arabian Sea and its counterpart Bay of Bengal is forced primarily by the semi-annually reversing monsoons, the spatio-temporal variations of surface waters is very strong in the Arabian Sea as compared to the Bay of Bengal. This study focuses on the inter-annual variability of Sea Surface Temperature (SST) in the Arabian Sea by analysing ERSST dataset which covers 152 years of SST (January 1854 to December 2002) based on the ICOADS in situ observations. To capture the dominant SST oscillations and to understand the inter-annual SST variations at various local regions of the Arabian Sea, wavelet analysis was performed on this long time-series SST dataset. This tool is advantageous over other signal analysing tools like Fourier analysis, based on the fact that it unfolds a time-series data (signal) both in frequency and time domain. This technique makes it easier to determine dominant modes of variability and explain how those modes vary in time. The analysis revealed that pentadal SST oscillations predominate at most of the analysed local regions in the Arabian Sea. From the time information of wavelet analysis, it was interpreted that these cold and warm events of large amplitude occurred during the periods 1870-1890, 1890-1910, 1930-1950, 1980-1990 and 1990-2005. SST oscillations with peaks having period of ~ 2-4 years was found to be significant in the central and eastern regions of Arabian Sea. This indicates that the inter-annual SST variation in the Indian Ocean is affected by the El Niño-Southern Oscillation (ENSO) and Indian Ocean Dipole (IOD) events. Keywords: Arabian Sea, ICOADS, inter-annual variation, pentadal oscillation, SST, wavelet analysis

Authors: Shailaja Grover, Purvi Tiwari, Vigneshwaran S. R., U. Dinesh Kumar
Abstract:
In India, grapes are one of the most important horticulture crops. Grapes are most vulnerable to downy mildew, which is one of the most devasting diseases. In the absence of a precise weather-based advisory system, farmers spray pesticides on their crops extensively. There are two main challenges associated with using these pesticides. Firstly, most of these sprays were panic sprays, which could have been avoided. Second, farmers use more expensive "Preventive and Eradicate" chemicals than "Systemic, Curative and Anti-sporulate" chemicals. When these chemicals are used indiscriminately, they can enter the fruit and cause health problems such as cancer. This paper utilizes decision trees and predictive modeling techniques to provide grape farmers with customized advice on grape disease management. This model is expected to reduce the overall use of chemicals by approximately 50% and the cost by around 70%. Most of the grapes produced will have relatively low residue levels of pesticides, i.e., below the permissible level. Keywords: analytics in agriculture, downy mildew, weather based advisory, decision tree, predictive modelling
Procedia PDF Downloads 7426905 Prevalence of Common Mental Disorders and Its Correlation with Mental Toughness among Professional South African Rugby Players
Authors: H. B. Grobler, K. Du Plooy, P. Kruger, S. Ellis
Abstract:
Objectives: The primary objective of the study was to determine the common mental disorders (CMD) identified by professional South African rugby players and its correlation with their mental toughness, as a first step towards developing such a programme within a larger research project. Design: Survey research, within the theoretical perspective of field theory, was conducted, utilising an adaptation of an already existing mental health questionnaire. The aim was to obtain feedback from as many possible professional South African rugby players in order to make certain generalizations and come to conclusions with regard to the current mental health experiences of these rugby players. Methods: Non-randomized sampling was done, linking it with internet research in the form of the online completion of a questionnaire. A sample of 215 rugby players participated and completed the online questionnaire. Permission was obtained to make use of an existing questionnaire, previously used by the specific authors with retired professional rugby players. A section on mental toughness was added. Data were descriptively analysed by means of the SPSS software platform. Results: Results indicated that the most significant problem that the players are experiencing, is a problem with alcohol (47.9%). Other problems that featured are distress (16.3%), sleep disturbances (7%), as well as anxiety and depression (4.2%). 4.7% of the players indicated that they smoke. 3.3% of the players experience themselves as not being mentally tough. A positive correlation between mental toughness and sound sleep (0.262) was found while a negative correlation was found between mental toughness and the following: anxiety/depression (-0.401), anxiety/depression positive (-0.423), distress (-0.259) and common mental disorder problems in general (-0.220). Conclusions: Although the presence of CMD at first glance do not seem significantly high amongst all the players, it must be considered that if one player in a team experiences the presence of CMD, it will have an impact on his mental toughness and most likely on his performance, as well as on the performance of the whole team. It is therefore important to ensure mental health in the whole team, by addressing individual CMD problems. A mental health support programme is therefore needed to be implemented to the benefit of these players within the South African context. Keywords: common mental disorders, mental toughness, professional athletes, rugby players

Authors: Aasim Siddiqui
Abstract:
In a country where 45.6 per cent of the total population lives below the poverty line, according to the Human Development Report 2014 by UNDP, an increasing number of private companies are now dedicating their resources to remedy this situation of chronic poverty. Most corporations in Pakistan now have a separate and dedicated department for Corporate Social Responsibility (CSR), albeit with varying goals and hence different strategies for achieving those goals. Similarly, Marine Group of Companies (MRGC) also has a robust CSR policy which the group implements through a Non-Government Organization (NGO) called Organization for Social Development Initiatives (OSDI). This organization, which operates under the ambit of MRGC’s CSR division, has a concentrated focus on helping the poorest communities in the rural areas of Pakistan to break out of intergenerational poverty. This paper maps the theoretical strategies as well as practical activities undertaken by OSDI for poverty alleviation via rural development in Pakistan. To obtain in-depth information of demographics, livelihood and socio-economic indicators in OSDI’s focused districts; a combination of quantitative and qualitative research methodologies was used during the course of this research. The paper highlights and explains OSDI’s unique three-pronged approach which aims at reducing poverty through income generation via the livelihood assistance program and through the provision of access to the most basic services (including health and education) via the community development and food security programs. Modeled on the concept of capacity building, OSDI’s modus operandi is centered on disbursing timely microcredit facilities to farmers who can benefit from these funds by investing in productive assets to foster financial capability for the future. With a focus on increasing the income of poor farmers, OSDI’s approach is to integrate all the socio-economic facets: education, health and sanitation and food security, to induce a sustained positive impact on their living standards. Keywords: CSR, poverty, rural, sustainability

Authors: Sanjay Singh, Karunanithi Priyanka, Ramoji Kosuru, Raju Prasad Sharma
Abstract:
Herbal drugs are well known for their mixed pharmacological activities with the benefit of no harmful side effects. The use of herbal drugs is limited because of their higher dose requirement, frequent drug administration, poor bioavailability of phytochemicals and delayed onset of action. Ficus religiosa, a potent anti-oxidant plant useful in the treatment of diabetes and cancer was selected for the study. Solid lipid nanoparticles (SLN) of Ficus religiosa extract was developed for the enhancement in oral bioavailability of stigmasterol and β-sitosterol-d-glucoside, principal components present in the extract. Hot homogenization followed by ultrasonication method was used to develop extract loaded SLN. Developed extract loaded SLN were characterized for particle size, PDI, zeta potential, entrapment efficiency, in vitro drug release and kinetics, fourier transform infra-red spectroscopy, differential scanning calorimetry, powder X-ray diffractrometry and stability studies. Entrapment efficiency of optimized extract loaded SLN was found to be 68.46 % (56.13 % of stigmasterol and 12.33 % of β-sitosteryl-d-glucoside, respectively). RP HPLC method development was done for simultaneous estimation of stigmasterol and β-sitosterol-d-glucoside in Ficus religiosa extract in rat plasma. Bioavailability studies were carried out for extract in suspension form and optimized extract loaded SLN. AUC of stigmasterol and β-sitosterol-d-glucoside were increased by 6.7-folds by 9.2-folds, respectively in rats treated with extract loaded SLN compared to extract suspension. Also, Cmax of stigmasterol and β-sitosterol-d-glucoside were increased by 4.3-folds by 3.9-folds, respectively in rats treated with extract loaded SLN compared to extract suspension. Mean residence times (MRT) for stigmasterol were found to be 12.3 ± 0.67 hours from extract and 7.4 ± 2.1 hours from SLN and for β-sitosterol-d-glucoside, 10.49 ± 2.9 hours from extract and 6.4 ± 0.3 hours from SLN. Hence, it was concluded that SLN enhanced the bioavailability and reduced the MRT of stigmasterol and β-sitosterol-d-glucoside in Ficus religiosa extract which in turn may lead to reduction in dose of Ficus religiosa extract, prolonged duration of action and also enhanced therapeutic efficacy. Keywords: Ficus religiosa, phytosterolins, bioavailability, solid lipid nanoparticles, stigmasterol and β-sitosteryl-d-glucoside

Abstract:
This paper focuses on the analysis of tensile stresses perpendicular to the grain in simply supported beams with different geometry made of glued laminated timber. Two types of beams are considered: standard double-tapered beams described in Eurocode 5 and non-standard glulam beams with a flattened apex. The beams are analyzed using two methodology approaches: a code design verification method and a finite element method (FEM) in terms of the linear theory of elasticity with plane stress assumption. The performed analyses proved that both methodologies lead to consistent results in case of standard glulam beams and therefore, the FEM can be used in case of non-standard structures, which are not included in Eurocode 5. Moreover, the FE analysis of the glulam beam with a flattened apex showed that it can be treated as a structure with two apex zones. Keywords: double-tapered beams, finite element analysis, glued laminated timber, perpendicular to grain stress

Based on the findings of this study, the following conclusion were drawn: The majority of the respondents are aged 18 and 21 old, female, are third year students, and have monthly allowance of P 2,000 above. On the respondents’ usage of Facebook, the majority of use the Facebook on a daily basis for one to two (1-2) hours everyday. And most users used Facebook by renting a computer in an internet cafe. On the use of Facebook, most users have created their profiles mainly to connect with people and gain new friends. The most commonly used features of Facebook, are: photos application, like button, wall, notification, friend, chat, network, groups and “like” pages status updates, messages and inbox and events. While the other Facebook features that are seldom used by the respondents are games, news feed, user name, video sharing and notes. And the least used Facebook features are questions, poke feature, credits and the market place. The respondents stated that the major benefit that the Facebook has given to its users is its ability to keep in touch with family members or friends while the main risk identified is that the users can become addicted to the Internet. On the tests of relationships between the respondents’ use of Facebook and the four (4) socio-demographic profile variables: age, sex, year level, and month allowance, were found to be not significantly related to the respondents’ use of the Facebook. While the variable found to be significantly related was gender. Keywords: Facebook, sociology, social networking, exploratory study

Authors: Susana Sousa Machado
Abstract:
The debate on the use of religious symbols has been highlighted in modern societies, especially in the field of labour relationships. As litigiousness appears to be growing, the matter requires a careful study from a legal perspective. In this context, a description and critical analysis of the most recent case law is conducted regarding the use of symbols by the employee in the workplace, delivered both by the European Court of Human Rights and by the Court of Justice of the European Union. From this comparative analysis we highlight the most relevant aspects in order to seek a common core regarding the juridical-argumentative approach of case law. Keywords: religion, religious symbols, workplace, discrimination

Authors: M. A. Adejumobi, J. O. Ojediran
Abstract:
The aim of irrigation is to recharge the available water in the soil. Quality of irrigation water is essential for the yield and quality of crops produced, maintenance of soil productivity and protection of the environment. The analysis of irrigation water arises as a need to know the impact of irrigation water on the yield of crops, the effect, and the necessary control measures to rectify the effect of this for optimum production and yield of crops. This study was conducted to assess the quality of irrigation water with its performance on crop planted, in Josepdam irrigation scheme Bacita, Nigeria. Field visits were undertaken to identify and locate water supply sources and collect water samples from these sources; X1 Drain, Oshin, River Niger loop and Ndafa. Laboratory experiments were then undertaken to determine the quality of raw water from these sources. The analysis was carried for various parameters namely; physical and chemical analyses after water samples have been taken from four sources. The samples were tested in laboratory. Results showed that the raw water sources shows no salinity tendencies with SAR values less than 1me/l and Ecvaules at Zero while the pH were within the recommended range by FAO, there are increase in potassium and sulphate content contamination in three of the location. From this, it is recommended that there should be proper monitoring of the scheme by conducting analysis of water and soil in the environment, preferable test should be carried out at least one year to cover the impact of seasonal variations and to determine the physical and chemical analysis of the water used for irrigation at the scheme. Keywords: irrigation, salinity, raw water quality, scheme

Authors: Ehsan Gholami, Vincent Demers
Abstract:
Low-pressure powder injection molding is an emerging technology for cost-effectively producing complex shape metallic parts with the proper dimensional tolerances, either in high or in low production volumes. In this study, the molding properties of cobalt-chrome-based feedstocks were evaluated for use in a low-pressure powder injection molding process. The rheological properties of feedstock formulations were obtained by mixing metallic powder with a proprietary wax-based binder system. Rheological parameters such as reference viscosity, shear rate sensitivity index, and activation energy for viscous flow, were extracted from the viscosity profiles and introduced into the Weir model to calculate the moldability index. Feedstocks were experimentally injected into a spiral mold cavity to validate the injection performance calculated with the model. Keywords: binder, feedstock, moldability, powder injection molding, viscosity

Authors: Wang Yanping Karen, Mohd Rafeah Siti, Park MI Kyoung
Abstract:
Formalin-fixed paraffin embedded (FFPE) tissue is important in the area of oncological diagnostics. This method of preserving tissues enabling them to be stored easily at ambient temperature for a long time. This decreases the risk of losing the DNA quantity and quality after extraction, reducing sample wastage, and making FFPE more cost effective. However, extracting DNA from FFPE tissue is a challenge as DNA purified is often highly cross-linked, fragmented, and degraded. In addition, this causes problems for many downstream processes. In this study, there will be a comparison of DNA extraction efficiency between One BioMed’s Xceler8 automated platform with commercial available extraction kits (Qiagen and Roche). The FFPE tissue slices were subjected to deparaffinization process, pretreatment and then DNA extraction using the three mentioned platforms. The DNA quantity were determined with real-time PCR (BioRad CFX ) and gel electrophoresis. The amount of DNA extracted with the One BioMed’s X8 platform was found to be comparable with the other two manual extraction kits. Keywords: DNA extraction, FFPE tissue, qiagen, roche, one biomed X8
