Authors: Shakir H. Mohammed Al-Alwany, Saad Hasan M. Ali, Ibrahim Mohammed S. Shnawa
Abstract:
The study was aimed to define the percentage of detection of high-oncogenic risk types of HPV and their genotyping in archival tissue specimens that ranged from apparently healthy tissue to invasive breast cancer by using one of the recent versions of In Situ Hybridization(ISH) 0.2. To find out rational significance of such genotypes as well as over expressed products of mutants P53 and RB genes on the severity of underlying breast cancers. The DNA of HPV was detected in 46.5 % of tissues from breast cancers while HPV DNA in the tissues from benign breast tumours was detected in 12.5%. No HPV positive–ISH reaction was detected in healthy breast tissues of the control group. HPV DNA of genotypes (16, 18, 31 and 33) was detected in malignant group in frequency of 25.6%, 27.1%, 30.2% and 12.4%, respectively. Over expression of p53 was detected by IHC in 51.2% breast cancer cases and in 50% benign breast tumour group, while none of control group showed P53- over expression. Retinoblastoma protein was detected by IHC test in 49.7% of malignant breast tumours, 54.2% of benign breast tumours but no signal was reported in the tissues of control group. The significance prevalence of expression of mutated p53 & Rb genes as well as detection of high-oncogenic HPV genotypes in patients with breast cancer supports the hypothesis of an etiologic role for the virus in breast cancer development. Keywords: human papilloma virus, P53, RB, breast cancer

Authors: Aliyu Adamu, Fahrul Huyop, Roswanira Abdul Wahab, Mohd Shahir Shamsir
Abstract:
Halogenated organic compounds occur in huge amount in biosphere. This is attributable to the diverse use of halogen-based compounds in the synthesis of various industrially important products. Halogenated compound is toxic and may persist in the environment, thereby causing serious health and environmental pollution problems. L-2-haloacid dehalogenases (EC 3.8.1.2) catalyse the specific cleavage of carbon-halogen bond in L-isomers of halogenated compounds, which consequently reverse the effects of environmental halogen-associated pollution. To enhance the efficiency and utility of these enzymes, this study investigates the catalytic amino acid residues and the molecular functional mechanism of DehL, by classical molecular dynamic simulations, MM-PBSA and ab initio fragments molecular orbital (FMO) calculations. The results of the study will serve as the basis for the molecular engineering of the enzyme. Keywords: DehL, Functional mechanism, Catalytic residues, L-2-haloacid dehalogenase

Abstract:
Biodegradable and renewable-based polymeric packaging like cellulose acetate (CA) is an alternative to petroleum-based polymers, in the way of low cost and also creates a positive outcome on both environmentally. The objective of the present research was to develop bioactive packaging films from cellulose acetate incorporated with a low-cost cypress essential oil (EO). We prepared cellulose acetate films via solvent casting method incorporating 0, 10, 30, and 60 % (w/w) of EO, with the purpose of evaluating the possible changes caused by the cypress essential oil on the properties of the packaging. The films were characterized using FTIR, TGA, XRD and other analysis technologies. The mechanical, antibacterial and antioxidant properties of the films were analyzed. FTIR and XRD analysis indicated that cypress EO was homogenously distributed on the film. Meanwhile, TGA analysis demonstrated that the addition of EO had an impact on thermal properties. The impact of EO on mechanical and optical properties was explored. The results displayed that antibacterial activity against Escherichia coli and Staphylococcus aureus increased as cypress essential oil percentage increased in cellulose acetate films. Moreover, free radical scavenger activity by DPPH of cellulose acetate films improved by increasing the cypress essential oil concentration. These results indicate that the films of cellulose acetate containing cypress essential oil have potential for use as active packaging for foods. Keywords: cellulose acetate, essential oil, active packaging, antibacterial, antioxidant

Authors: Hassan. A. Abas, Saad A. Aiban
Abstract:
Soil improvement by means of stone column method is used to improve sabkha soils in order to limit total and differential settlement and to achieve the required bearing capacity. Full-scale plate test was performed on site to confirm the achievement of required bearing capacity at the specified settlement. Despite the fact that this technique is widely used to improve sabkha soils, there are no studies focusing on the behavior of stone columns in such problematic soils. Sabkha soils are known for its high compressibility, low strength and water sensitivity due to loss of salt cementation upon flooding during installation of stone columns. Numerical modeling of plate load test assist to understand complicated behavior of sabkha – stone column interaction. This paper presents a three-dimensional Finite element model, using PLAXIS 3D software, to simulate vertical plate load tests on a stone column installed in sabkha. The predicted settlement values are in reasonable agreement with the field measure values and the field load - settlement curve can be predicted with good accuracy. Keywords: soil improvement, stone column, sabkha, PLAXIS 3D

Authors: Wang Yanping Karen, Mohd Rafeah Siti, Park MI Kyoung
Abstract:
Formalin-fixed paraffin embedded (FFPE) tissue is important in the area of oncological diagnostics. This method of preserving tissues enabling them to be stored easily at ambient temperature for a long time. This decreases the risk of losing the DNA quantity and quality after extraction, reducing sample wastage, and making FFPE more cost effective. However, extracting DNA from FFPE tissue is a challenge as DNA purified is often highly cross-linked, fragmented, and degraded. In addition, this causes problems for many downstream processes. In this study, there will be a comparison of DNA extraction efficiency between One BioMed’s Xceler8 automated platform with commercial available extraction kits (Qiagen and Roche). The FFPE tissue slices were subjected to deparaffinization process, pretreatment and then DNA extraction using the three mentioned platforms. The DNA quantity were determined with real-time PCR (BioRad CFX ) and gel electrophoresis. The amount of DNA extracted with the One BioMed’s X8 platform was found to be comparable with the other two manual extraction kits. Keywords: DNA extraction, FFPE tissue, qiagen, roche, one biomed X8

Abstract:
Transcriptome retrieved from SRA database of different tissues and treatments of C. roseus was assembled in order to detect tissue-specific transcription factors (TFs) and TFs possibly related to terpenoid indole alkaloids (TIA) pathway. A number of 290 TF-like transcripts along with 12 transcripts related to TIA biosynthetic pathway were divided in terms of co-expression in the different tissues, treatments and genotypes. Three transcripts encoding peroxidases 1 and 12 were downregulated in hairy root, while upregulated in mature leaf. Eight different transcripts of the TIA pathway co-expressed with TFs either functioning downstream tryptophan biosynthesis, e.g., tdc, str1 and sgd, or upstream vindoline biosynthesis, e.g., t16h, omt, nmt, d4h and dat. The results showed no differential expression of TF transcripts in hairy roots knocked down for tdc gene (TDCi) as compared to their wild type controls. There were several evidences of tissue-specific expression of TF transcripts in flower, mature leaf, root/hairy root, stem, seedling, hairy root and immature/mature leaves. Regulation included transcription factor families, e.g., bHLH, MYB and WRKY mostly induced by ABA and/or JA (or MeJA) and regulated during abiotic or biotic stress. The information of tissue-specific regulation and co-expression of TFs and genes in the TIA pathway can be utilized in manipulating alkaloid biosynthesis in C. roseus. Keywords: SRA database, bHLH, MYB, WRKY, co-expression

Authors: Jan Muhammad, Saad Malik, Fadia W. Al-Azawi, Ali Imran
Abstract:
In the era of climate variability, rising temperatures are the most significant aspect. In this study PRECIS model data and observed data are used for assessing the temperature change scenarios of Sindh province during the first half of present century. Observed data from various meteorological stations of Sindh are the primary source for temperature change detection. The current scenario (1961–1990) and the future one (2010-2050) are acted by the PRECIS Regional Climate Model at a spatial resolution of 25 * 25 km. Regional Climate Model (RCM) can yield reasonably suitable projections to be used for climate-scenario. The main objective of the study is to map the simulated temperature as obtained from climate model-PRECIS and their comparison with observed temperatures. The analysis is done on all the districts of Sindh in order to have a more precise picture of temperature change scenarios. According to results the temperature is likely to increases by 1.5 - 2.1°C by 2050, compared to the baseline temperature of 1961-1990. The model assesses more accurate values in northern districts of Sindh as compared to the coastal belt of Sindh. Procedia PDF Downloads 252418 Failure of PC Wire for PCC Pipes Due to Corrosion in Man-Made River Project Investigation of Possible Causes and Relation of Wire Breaks with Potential Measurement
Authors: Saad A. Bakheet, Salah M. Elkoum, Asharaf A. Almaghribi
Abstract:
Pre-stressed wire is considered the most important in manufacturing pre-stressed concrete pipes, this is because the life of the mentioned pipes depends on the integrity of the wire. When the wire is wrapped around the concrete core, it provides a compressive strength that enables the concrete cores to withstand internal and external pressures. Since LIBYA has constructed different stages of man-made river projects using pre-stressed concrete pipes in different diameters (1.6, 2.2, 3.6 and 4.0 m) to transport water from the south part of the country to the north-populated area, pipe failures due to corrosion have occurred after several years of operation at different locations of the pipeline route. The PC wire is corroded and, in the end, breaks and becomes unable to withstand internal water pressure. The wire breaks recorded using the hydrophone technique added extra pressure on the project management staff and engineers to resolve and study the possible main cause of wire break and pipe failure. Information regarding the PC wire used will be provided in this paper which includes specifications, manufacturing etc. In this paper, the causes of wire corrosion and wire breaks after several years of PCCP operation will be discussed and explained, in addition to that, the correlation between wire breaks and pipe potential will be addressed and highlighted. Keywords: wire technical specification, wire break, corrosion causes, potential measurement, failure concrete pipe

Authors: Tahseen Saad, Halim Ceylan, Jonathan Weaver, Osman Nuri Çelik, Onur Gungor Sahin
Abstract:
A new concept of uniform delay offset dependent mathematical optimization problem is derived as the main objective for this study using a differential evolution algorithm. To control the coordination problem, which depends on offset selection and to estimate uniform delay based on the offset choice in a traffic signal network. The assumption is the periodic sinusoidal function for arrival and departure patterns. The cycle time is optimized at the entry links and the optimized value is used in the non-entry links as a common cycle time. The offset optimization algorithm is used to calculate the uniform delay at each link. The results are illustrated by using a case study and are compared with the canonical uniform delay model derived by Webster and the highway capacity manual’s model. The findings show new model minimizes the total uniform delay to almost half compared to conventional models. The mathematical objective function is robust. The algorithm convergence time is fast. Keywords: area traffic control, traffic flow, differential evolution, sinusoidal periodic function, uniform delay, offset variable

Authors: Saad Mohamed Elsaid Abdelrahman
Abstract:
The adsorption method is one of the best modern techniques used in removing pollutants, especially organic hydrocarbon compounds, from polluted water. Through this research, bentonite clay can be used to remove organic hydrocarbon compounds, such as heptane and octane, resulting from oil spills in seawater. Bentonite clay can be obtained from the Kholayaz area, located north of Jeddah, at a distance of 80 km. Chemical analysis shows that bentonite clay consists of a mixture of silica, alumina and oxides of some elements. Bentonite clay can be activated in order to raise its adsorption efficiency and to make it suitable for removing pollutants using an ionic organic solvent. It is necessary to study some of the factors that could be in the efficiency of bentonite clay in removing oily organic compounds, such as the time of contact of the clay with heptane and octane solutions, pH and temperature, in order to reach the highest adsorption capacity of bentonite clay. The temperature can be a few degrees Celsius higher. The adsorption capacity of the clay decreases when the temperature is raised more than 4°C to reach its lowest value at the temperature of 50°C. The results show that the friction time of 30 minutes and the pH of 6.8 is the best conditions to obtain the highest adsorption capacity of the clay, 467 mg in the case of heptane and 385 mg in the case of octane compound. Experiments conducted on bentonite clay were encouraging to select it to remove heavy molecular weight pollutants such as petroleum compounds under study. Keywords: adsorbent, bentonite clay, oil spills, removal

Authors: Nur Ashida Salim, Muhammad Murtadha Othman, Ismail Musirin, Mohd Salleh Serwan
Abstract:
Over the past few decades, the power system industry in many developing and developed countries has gone through a restructuring process of the industry where they are moving towards a deregulated power industry. This situation will lead to competition among the generation and distribution companies to achieve a certain objective which is to provide quality and efficient production of electric energy, which will reduce the price of electricity. Therefore it is important to obtain an accurate value of the Available Transfer Capability (ATC) and Transmission Reliability Margin (TRM) in order to ensure the effective power transfer between areas during the occurrence of uncertainties in the system. In this paper, the TRM and ATC is determined by taking into consideration the uncertainties of the system operating condition and system cascading collapse by applying the bootstrap technique. A case study of the IEEE RTS-79 is employed to verify the robustness of the technique proposed in the determination of TRM and ATC.Keywords: available transfer capability, bootstrap technique, cascading collapse, transmission reliability margin

Authors: Atef M. Ghaleb, Mohamed Z. Ramadan, Khalid Saad Aljaloud
Abstract:
The aim of this study is to assesses the lifting capabilities of persons experiencing hypoxia. It also examines the behavior of the physiological response induced through the lifting process related to changing in the hypoxia and lifting frequency variables. For this purpose, the study performed two consecutive tests by using; (1) training and acclimatization; and (2) an actual collection of data. A total of 10 male students from King Saud University, Kingdom of Saudi Arabia, were recruited in the study. A two-way repeated measures design, with two independent variables (ambient oxygen (15%, 18% and 21%)) and lifting frequency (1 lift/min and 4 lifts/min) and four dependent variables i.e., maximum acceptable weight of lift (MAWL), Electromyography (EMG) of four muscle groups (anterior deltoid, trapezius, biceps brachii, and erector spinae), rating of perceived exertion (RPE), and rating of oxygen feeling (ROF) were used in this study. The results show that lifting frequency has significantly impacted the MAWL and muscles’ activities. The oxygen content had a significant effect on the RPE and ROE. The study has revealed that acclimatization and training sessions significantly reduce the effect of the hypoxia on the human physiological parameters during the manual materials handling tasks. Keywords: lifting capabilities, muscle activities, oxygen content, perceived exertion

Abstract:
In this study, the numerical investigation of viscous dissipation on convective boundary layer flow towards a horizontal circular cylinder with constant wall temperature is considered. The transformed partial differential equations are solved numerically by using an implicit finite-difference scheme known as the Keller-box method. Numerical solutions are obtained for the reduced Nusselt number and the skin friction coefficient as well as the velocity and temperature profiles. The features of the flow and heat transfer characteristics for various values of the Prandtl number and Eckert number are analyzed and discussed. The results in this paper is original and important for the researchers working in the area of boundary layer flow and this can be used as reference and also as complement comparison purpose in future. Keywords: free convection, horizontal circular cylinder, viscous dissipation, convective boundary layer flow

Abstract:
Application of adaptive online quiz system and a design are performed in this paper. The purpose of adaptive quiz system is to establish different questions automatically for each student and measure their competence on a definite area of discipline. This model determines students competencies in cases like distant-learning which experience challenges frequently. Questions are specialized to allow clear deductions about student gains; they are able to identify student competencies more effectively. Also, negative effects of questions requiring higher knowledge than competency over student’s morale and self-confidence are dismissed. The advantage of the system in the quiz management requires less total time for measuring and is more flexible. Self sufficiency of the system in terms of repeating, planning and assessment of the measurement process allows itself to be used in the individual education sets. Adaptive quiz technique prevents students from distraction and motivation loss, which is led by the questions with quite lower hardness level than student’s competency. Keywords: e-learning, adaptive system, security, quiz database
