116 Role of Oxidative DNA Damage in Pathogenesis of Diabetic Neuropathy
Authors: Ireneusz Majsterek, Anna Merecz, Agnieszka Sliwinska, Marcin Kosmalski, Jacek Kasznicki, Jozef Drzewoski
Abstract:
Oxidative stress is considered to be the cause for onset and the progression of type 2 diabetes mellitus (T2DM) and complications including neuropathy. It is a deleterious process that can be an important mediator of damage to cell structures: protein, lipids and DNA. Data suggest that in patients with diabetes and diabetic neuropathy DNA repair is impaired, which prevents effective removal of lesions. Objective: The aim of our study was to evaluate the association of the hOGG1 (326 Ser/Cys) and XRCC1 (194 Arg/Trp, 399 Arg/Gln) gene polymorphisms whose protein is involved in the BER pathway with DNA repair efficiency in patients with diabetes type 2 and diabetic neuropathy compared to the healthy subjects. Genotypes were determined by PCR-RFLP analysis in 385 subjects, including 117 with type 2 diabetes, 56 with diabetic neuropathy and 212 with normal glucose metabolism. The polymorphisms studied include codon 326 of hOGG1 and 194, 399 of XRCC1 in the base excision repair (BER) genes. Comet assay was carried out using peripheral blood lymphocytes from the patients and controls. This test enabled the evaluation of DNA damage in cells exposed to hydrogen peroxide alone and in the combination with the endonuclease III (Nth). The results of the analysis of polymorphism were statistically examination by calculating the odds ratio (OR) and their 95% confidence intervals (95% CI) using the ¤ç2-tests. Our data indicate that patients with diabetes mellitus type 2 (including those with neuropathy) had higher frequencies of the XRCC1 399Arg/Gln polymorphism in homozygote (GG) (OR: 1.85 [95% CI: 1.07-3.22], P=0.3) and also increased frequency of 399Gln (G) allele (OR: 1.38 [95% CI: 1.03-1.83], P=0.3). No relation to other polymorphisms with increased risk of diabetes or diabetic neuropathy. In T2DM patients complicated by neuropathy, there was less efficient repair of oxidative DNA damage induced by hydrogen peroxide in both the presence and absence of the Nth enzyme. The results of our study suggest that the XRCC1 399 Arg/Gln polymorphism is a significant risk factor of T2DM in Polish population. Obtained data suggest a decreased efficiency of DNA repair in cells from patients with diabetes and neuropathy may be associated with oxidative stress. Additionally, patients with neuropathy are characterized by even greater sensitivity to oxidative damage than patients with diabetes, which suggests participation of free radicals in the pathogenesis of neuropathy. Keywords: Diabetic neuropathy, oxidative stress, gene polymorphisms, oxidative DNA damage.

Abstract:
Supersonic hydrogen-air cylindrical mixing layer is numerically analyzed to investigate the effect of inlet swirl on ignition time delay in scramjets. Combustion is treated using detail chemical kinetics. One-equation turbulence model of Spalart and Allmaras is chosen to study the problem and advection upstream splitting method is used as computational scheme. The results show that swirling both fuel and oxidizer streams may drastically decrease the ignition distance in supersonic combustion, unlike using the swirl just in fuel stream which has no helpful effect. Keywords: Ignition delay, Supersonic combustion, Swirl, Numerical simulation, Turbulence.

Abstract:
The design and plantwide control of an integrated plant where the endothermic 1,4-butanediol dehydrogenation and the exothermic furfural hydrogenation is simultaneously performed in a single reactor is studied. The reactions can be carried out in an adiabatic reactor using small hydrogen excess and with reduced parameter sensitivity. The plant is robust and flexible enough to allow different production rates of γ-butyrolactone and 2-methyl furan, keeping high product purities. Rigorous steady state and dynamic simulations performed in AspenPlus and AspenDynamics to support the conclusions.

Abstract:
The photochemical and photo-Fenton oxidation of 1,3-dichloro-2-propanol was performed in a batch reactor, at room temperature, using UV radiation, H2O2 as oxidant, and Fenton-s reagent. The effect of the oxidative agent-s initial concentration was investigated as well as the effect of the initial concentration of Fe(II) by following the target compound degradation, the total organic carbon removal and the chloride ion production. Also, from the kinetic analysis conducted and proposed reaction scheme it was deduced that the addition of Fe(II) significantly increases the production and the further oxidation of the chlorinated intermediates.

Abstract:
The modified Claus process is the major technology for the recovery of elemental sulfur from hydrogen sulfide. The chemical reactions that can occur in the reaction furnace are numerous and many byproducts such as carbon disulfide and carbon carbonyl sulfide are produced. These compounds can often contribute from 20 to 50% of the pollutants and therefore, should be hydrolyzed in the catalytic converter. The inlet temperature of the first catalytic reactor should be maintained over than 250 °C, to hydrolyze COS and CS2. In this paper, the various configurations for the first converter reheating of sulfur recovery unit are investigated. As a result, the performance of each method is presented for a typical clause unit. Abstract:
The elimination of ranitidine (a pharmaceutical compound) has been carried out in the presence of UV-C radiation. After some preliminary experiments, it has been experienced the no influence of the gas nature (air or oxygen) bubbled in photolytic experiments. From simple photolysis experiments the quantum yield of this compound has been determined. Two photolytic approximation has been used, the linear source emission in parallel planes and the point source emission in spherical planes. The quantum yield obtained was in the proximity of 0.05 mol Einstein-1 regardless of the method used. Addition of free radical promoters (hydrogen peroxide) increases the ranitidine removal rate while the use of photocatalysts (TiO2) negatively affects the process. Keywords: Quantum yield, photolysis, ranitidine, watertreatment.

Abstract:
A study was carried out to evaluate the growth and yield performance of Pleurotus ostreatus spawn on different organic substrates in Lafia, Nasarawa State, Nigeria. 50 g each of four different substrates namely; corncobs, rice straw, sugarcane bagasse and sawdust sourced locally from farmlands and processing sites, were amended with 2% calcium carbonate and calcium sulphide and sterilized using three sterilization methods namely; hot water, steam, and lime. Five grams of P. ostreatus spawn were inoculated unto treated substrates, incubated in the dark for 16 days and in light for 19 days at 25 0C for the commencement of pinhead and fruit body formations respectively. Growth and yield parameters such as days to full colonization, days to pinhead formation and days to fruit body formation were recorded. Cap diameter and fresh weight of mature mushrooms were also measured for a total count of four flushes. P. ostreatus spawn grown on sugarcane bagasse recorded the highest mean cap diameter (4.69 cm), highest mean fresh weight (34.68 g), highest biological efficiency (69.37%) and highest production rate (2.83 g per day). Spawn grown on rice straw recorded the least number of days to full substrate colonization (11.00). Spawn grown on corn cobs recorded the least mean number of days to pin head (18.75) and fruiting body formations (20.25). There were no significant differences (P ≤ 0.05) among the evaluated substrates with respect to growth and yield performance of P. ostreatus. Substrates sterilized with hot water supported the highest mean cap diameter (5.64 cm), highest biological efficiency (87.04%) and highest production rate (3.43 g per day) of P. ostreatus. Significant differences (P ≤ 0.05) were observed in cap diameter, fresh weight, biological efficiency and production rates among the evaluated sterilization methods. Depending on the number of photons absorbed, the electron radius of rotation around the proton is established. Until now, the principle of photons absorption by electrons and the electron transition to a new energy level, namely to a higher radius of rotation around the proton, is not clarified in physics. This paper aims to demonstrate that radiation, photons respectively, have mass and negative electrostatic charge similar to electrons but infinitely smaller. Abstract:
Formaldehyde is the illegal chemical substance used for food preservation in fish and vegetable. It can promote carcinogenesis. Superoxide dismutases are the important antioxidative enzymes that catalyze the dismutation of superoxide anion into oxygen and hydrogen peroxide. The resultant level of oxidative stress in formaldehyde-treated lymphocytes was investigated. The formaldehyde concentrations of 0, 20, 40, 60, 80 and 120μmol/L were treated in human lymphocytes for 12 hours. After 12 treated hours, the superoxide dismutase activity change was measured in formaldehyde-treated lymphocytes. The results showed that the formaldehyde concentrations of 60, 80 and 120μmol/L significantly decreased superoxide dismutase activities in lymphocytes (P < 0.05). The change of superoxide dismutase activity in formaldehyde-treated lymphocytes may be the biomarker for detect cellular injury, such as damage to DNA, due to formaldehyde exposure.

Abstract:
The influence of SiC powder addition on densification of Al-SiC compacts during sintering in different atmospheres was investigated. It was performed in a dilatometer in flowing nitrogen, nitrogen/hydrogen (95/5 by volume) and argon. Fine, F500 grade of SiC powder was used. Mixtures containing 10 and 30 vol.% of SiC reinforcement were prepared in a Turbula mixer. Green compacts of about 82% of theoretical density were made of each mixture. For comparison, compacts made of pure aluminum powder were also investigated. It was shown that nitrogen is the best sintering atmosphere because only in this atmosphere did shrinkage take place. Its amount is lowered by ceramic powder addition, i.e. the more SiC the less densification occurs. Additionally, the formation of clusters, enhanced in compacts containing 30 vol.% SiC, is also responsible for limiting the shrinkage. Microstructural examinations of sintered composites revealed that sintering of compacts occurs in the presence of the liquid phase exclusively in nitrogen.
