5203 Immunocytochemical Stability of Antigens in Cytological Samples Stored in In-house Liquid-Based Medium

Abstract:
The decision for immunocytochemistry (ICC) is usually made in the basis of the findings in Giemsa- and/or Papanicolaou- smears. More demanding diagnostic cases require preparation of additional cytological preparations. Therefore, it is convenient to suspend cytological samples in a liquid based medium (LBM) that preserve antigen and morphological properties. However, the duration of these properties being preserved in the medium is usually unknown. Eventually, cell morphology becomes impaired and altered, as well as antigen properties may be lost or become diffused. In this study, the influence of cytological sample storage length in in-house liquid based medium on antigen properties and cell morphology is evaluated. The question is how long the cytological samples in this medium can be stored so that the results of immunocytochemical reactions are still reliable and can be safely used in routine cytopathological diagnostics. The stability of 6 ICC markers that are most frequently used in everyday routine work were tested; Cytokeratin AE1/AE3, Calretinin, Epithelial specific antigen Ep-CAM (MOC-31), CD 45, Oestrogen receptor (ER), and Melanoma triple cocktail were tested on methanol fixed cytospins prepared from fresh fine needle aspiration biopsies, effusion samples, and disintegrated lymph nodes suspended in in-house cell medium. Cytospins were prepared on the day of the sampling as well as on the second, fourth, fifth, and eight day after sample collection. Next, they were fixed in methanol and immunocytochemically stained. Finally, the percentage of positive stained cells, reaction intensity, counterstaining, and cell morphology were assessed using two assessment methods: the internal assessment and the UK NEQAS ICC scheme assessment. Results show that the antigen properties for Cytokeratin AE1/AE3, MOC-31, CD 45, ER, and Melanoma triple cocktail were preserved even after 8 days of storage in in-house LBM, while the antigen properties for Calretinin remained unchanged only for 4 days. The key parameters for assessing detection of antigen are the proportion of cells with a positive reaction and intensity of staining. Well preserved cell morphology is highly important for reliable interpretation of ICC reaction. Therefore, it would be valuable to perform a similar analysis for other ICC markers to determine the duration in which the antigen and morphological properties are preserved in LBM.Keywords: cytology samples, cytospins, immunocytochemistry, liquid-based cytology

Abstract:
The liquid-liquid extraction of copper (II) from aqueous solution by capric acid (HL) in chloroform at 25°C has been studied. The ionic strength effect of the aqueous phase shows that the extraction of copper(II) increases with the increase in ionic strength. with different ionic strengths 1, 0.5, 0.25, 0.125 and 0.1M in the aqueous phase. Welding is one of the most important technological processes used in many branches of industry such as industrial engineering, shipbuilding, pipeline fabrication among others. Generally, welding is the preferred joining method and most common steels are weldable. This investigation is a contribution to scientific work of welding of low carbon steel. This work presents the results of the isothermal heat treatment effect at 200, 400 and 600 °C on microstructural evolution in weld region of X70 pipeline steel. The welding process has been realized in three passes by industrial arc welding. Abstract:
The steel alloy Introduced after becoming carbon-steel does not meet the requirements of engineering industry; and it cannot be obtained tensile strength from carbon-steel higher than (700MPa), the low alloy steel enters in a lot of heavy engineering equipment parts, molds, agricultural equipment and other industry. In addition, that may be exposed to in-service failure, which may require returned to work, to do the repairs or maintenance by one of the welding methods available. The ability of steel weld determined through palpation of the cracks, which can reduce by many ways. These ways are often expensive and difficult to implement, perhaps the control to choose the type of electrode welding user is one of the easiest and least expensive applications. It has been welding the steel low alloys high resistance by manual metal arc (MMA), and by using a set of welding electrodes which varying in chemical composition and in their prices as well and test their effect on tensile strength. Results showed that using the poles of welding, which have a high proportion of iron powder and low hydrogen. The Tensile resistance is (484MPa) and the weld joint efficiency was (56.9%), but when (OK 47.04) electrode was used the tensile strength increased to (720MPa) and the weld joint efficiency to (84.7%). Using the cheapest electrode (OK 45.00) the weld joint efficiency did not exceed (24.2%), but when using the most expensive electrode (OK 91.28) the weld joint efficiency is (38.1%).Keywords: steel low alloys high resistance, electrodes welding, tensile test

Abstract:
Membrane separations are mentioned frequently as a possibility for CO2 capture. Selectivity of ionic liquid membranes is strongly determined by different solubility of separated gases in ionic liquids. The solubility of separated gases usually varies over an order of magnitude, differently from diffusivity of gases in ionic liquids, which is usually of the same order of magnitude for different gases. The present work evaluates the selection of an appropriate ionic liquid for the selective membrane preparation based on the gas solubility in an ionic liquid. The current state of the art of CO2 capture patents and technologies based on the membrane separations was considered. An overview is given of the discussed transport mechanisms. Ionic liquids seem to be promising candidates thanks to their tunable properties, wide liquid range, reasonable thermal stability, and negligible vapor pressure. However, the uses of supported liquid membranes are limited by their relatively short lifetime from the industrial point of view. On the other hand, ionic liquids could overcome these problems due to their negligible vapor pressure and their tunable properties by adequate selection of the cation and anion.Keywords: biogas upgrading, carbon dioxide separation, ionic liquid membrane, transport properties

Abstract:
In the present investigation, 75Cr3C2-25NiCr coating was deposited on T91 boiler tube steel substrate by high velocity oxy-fuel (HVOF) process to enhance high-temperature corrosion resistance. High-temperature performance of bare, as well as HVOF-coated steel specimens was evaluated for 1500 h under cyclic conditions in the platen superheater zone coal-fired boiler, where the temperature was around 900 °C. Experiments were carried out for 15 cycles each of 100 h duration followed by 1 h cooling at ambient temperature. The performance of the bare and coated specimens was assessed via metal thickness loss corresponding to the corrosion scale formation and the depth of internal corrosion attack. 75Cr3C2-25NiCr coating deposited on T91 steel imparted better hot corrosion resistance than the uncoated steel. Inferior resistance of bare T91 steel is attributed to the formation of pores and loosely bounded oxide scale rich in Fe2O3.Keywords: 75Cr3C2-25NiCr, HVOF process, boiler steel, coal fired boilers

This paper propose the method for optimal scheduling for battery energy storage system with reliability constraint of energy storage system in reliability aspect. The optimal scheduling problem is solved by dynamic programming with proposed transition matrix. Proposed optimal scheduling method guarantees the minimum fuel cost within specific reliability constraint. For evaluating proposed method, the timely capacity outage probability table (COPT) is used that is calculated by convolution of probability mass function of each generator. Steel bridges deterioration has been one of the problems in North America for the last years. Steel bridges deterioration mainly attributed to the difficult weather conditions. Steel bridges suffer fatigue cracks and corrosion, which necessitate immediate inspection. Visual inspection is the most common technique for steel bridges inspection, but it depends on the inspector experience, conditions, and work environment. So many Non-destructive Evaluation (NDE) models have been developed use Non-destructive technologies to be more accurate, reliable and non-human dependent. Non-destructive techniques such as The Eddy Current Method, The Radiographic Method (RT), Ultra-Sonic Method (UT), Infra-red thermography and Laser technology have been used. Digital Image processing will be used for Corrosion detection as an Alternative for visual inspection. Different models had used grey-level and colored digital image for processing. However, color image proved to be better as it uses the color of the rust to distinguish it from the different backgrounds. The detection of the rust is an important process as it’s the first warning for the corrosion and a sign of coating erosion. To decide which is the steel element to be repainted and how urgent it is the percentage of rust should be calculated. In this paper, an image processing approach will be developed to detect corrosion and its severity. Abstract:
Thin liquid jets on micrometer scale play an important role in processing such as in fiber fabrication, inkjet printing, but also for sample delivery in modern synchrotron X-ray devices. In all these cases the liquid jets contain solvents and dissolved materials such as polymers, nanoparticles, fibers pigments or proteins. As liquid flow in liquid jets differs significantly from flow in capillaries and microchannels, particle localization and orientation will also be different. This is of critical importance for applications, which depend on well-defined homogeneous particle and fiber distribution and orientation in liquid jets. Investigations of particle orientation in liquid microjets of diluted solutions have been rare, despite their importance. With the arise of micro-focused X-ray beams it has become possible to scan across samples with micrometer resolution to locally analyse structure and orientation of the samples. In the present work, we used this method to scan across liquid microjets to determine the local distribution and orientation of anisotropic particles. The compromise wormlike block copolymer micelles as an example of long flexible fibrous structures, hectorite materials as a model of extended nanosheet structures, and gold nanorods as an illustration of short stiff cylinders to comprise all relevant anisotropic geometries. We find that due to the different velocity profile in the liquid jet, which resembles plug flow, the orientation of the particles which was generated in the capillary is lost or changed into non-oriented or bi-axially orientations depending on the geometrical shape of the particle.Keywords: anisotropic particles, liquid microjets, reorientation, SAXS

Abstract:
The present study has the overall objective of studying the effect of garlic and Argan oil on the growth of Rainbow trout (Oncorhynchus mykiss) fingerlings at the Ras El Ma (Azrou) salmon farming station during the 2023 production period. The fingerlings were distributed in seven tanks at a rate of 1000 per lot. The first control tank (B0) received only the feed without additives. Tanks B1, B2, B3, and B4 received garlic as a feed additive at a rate of 1%, 1.5%, 2% and 2.5% respectively. The fingerlings in tanks B5 and B6, in addition to 2.5% garlic, received 5 and 10ml argon oil, respectively. During this two-month experiment, the weight growth of the fingerlings and the physico-chemical parameters of the water that are favorable for fry rearing (hydrogen potential, temperature, dissolved oxygen, and electrical conductivity) were monitored. The weight growth of fingerlings receiving garlic was positive (mean weight: 4.95g, 5.43g, 5.13g, and 5.06g) compared with control fingerlings (mean weight: 3.88g). The maximum average weight was obtained with 1.5% garlic (average weight: 5.43g). The addition of 5 and 10ml of argon oil to B5 and B6 resulted in a slight increase in weight for the B5 fingerlings (5.37g) compared with the B4 control fingerlings (mean weight: 5.06g) but a minor decrease for the B6 batch (4.73g). In a fire incident involving a Nitrogen storage tank that over-pressured and exploded, resulting in a fire in one of the units in a refinery, lack of data and evidence hampered the investigation to determine the root cause. Instrumentation and fittings were destroyed in the fire. To make it worst, this incident occurred during the COVID-19 pandemic, making collecting and testing evidence delayed. In addition to that, the storage tank belonged to a third-party company which requires legal agreement prior to the refinery getting approval to test the remains. Despite all that, the investigation had to be carried out with stakeholders demanding answers. The investigation team had to devise alternative means to support whatever little evidence came out as the most probable root cause. International standards, practices, and previous incidents on similar tanks were referred. To narrow down to just one root cause from 8 possible causes, transient simulations were conducted to simulate the overpressure scenarios to prove and eliminate the other causes, leaving one root cause. This paper shares the methodology used and details how transient simulations were applied to help solve this. The experience and lessons learned gained from the event investigation and from numerous case studies via transient analysis in finding the root cause of the accident leads to the formulation of future mitigations and design modifications aiming at preventing such incidents or at least minimize the consequences from the fire incident.Keywords: fire, transient, simulation, relief

Abstract:
The chemical composition of desalinated water is modified to make it more acceptable to the end-user. Sometimes, this modification is approached by mixing with brackish water that is known to contain a variety of minerals. Expectedly, besides minerals, brackish water indigenous bacterial communities access the final mixture hence reaching the end consumer. The current project examined the safety of using brackish water as an ingredient in potable water. Pseudomonas aeruginosa strains were detected in potable and brackish water samples collected from storage facilities in residential areas as well as from main water distribution and storage tanks. The application of molecular and biochemical fingerprinting methods, including phylogeny, RFLP (restriction fragment length polymorphism), MLST (multilocus sequence typing) and substrate specificity testing, suggested that the potable water P. aeruginosa strains were most probably originated from brackish water. Additionally, all the sixty-four isolates showed multi-drug resistance (MDR) phenotype and harboured the three genes responsible for biofilm formation. These virulence factors represent serious health hazards compelling the scientific community to revise the WHO (World Health Organization) and USEP (US Environmental Protection Agency) A potable water quality guidelines, particularly those related to the types of bacterial genera that evade the current water quality guidelines.Keywords: potable water, brackish water, pseudomonas aeroginosa, multidrug resistance

Legislation & Government laws are stricter day by day & each plant has to think of recycling, reuse &reintegration of pollutants generated during the process of steel making. This paper deals with experiments conducted in Bhilai Steel Plant and Real Ispat and Power Limited for reuse, recycle & reintegrate some of the steel making process fines. Iron ore fines with binders have been agglomerated to be used as a part of the charge for small furnaces. This will improve yield at nominal cost. Rolling mill fines have been recycled to increase the yield of sinter making. This will solve the problems of fine disposal. Huge saving on account of recycling will be achieved. Lime fines after briquetting is used along with prime lime. Lime fines have also been used as a binding material during production of fly ash bricks. These fines serve as low-cost binder. Experiments have been conducted along with coke breeze & gas cleaning plant sludge. As a result, the anti-sloping compound has been developed for converter vessels. Dolo char and Char during Sponge Iron production have been successfully used in power generation and brick making. Pellets have been made with ventilation dust & flue dust. These samples have been tried as a coolant in the converter. Pellets have been made with Sinter Plant electrostatic precipitator micro fines with liquid binder. Trials have been conducted to reuse these pellets in sinter making. Coke breeze from coke-ovens fines and mill scale along with binders were agglomerated. This was used in furnace after attaining required screening and reactivity index. The study was conducted comparatively with specimens exposed to natural conditions free of chloride ion. The effect of chloride ion on the specimens was generated of form accelerated under controlled conditions (3.5% NaCl and 25 ° C temperature). The Impedance data were acquired over a range of 1 mHz to 100 kHz. In this research, we have studied and analyzed the modulation of light and liquid crystal in HPDLCs using Finite Domain Time Difference (FDTD) method. HPDLCs are modeled as a mixture of polymer and liquid crystals (LCs) that categorized as an anisotropic medium. FDTD method is directly solves Maxwell’s equation with less approximation, so this method can analyze more flexible and general approach for the arbitrary anisotropic media. As the results from FDTD simulation, the highest diffraction efficiency occurred at ±19 degrees (Bragg angle) using p polarization incident beam to Bragg grating, Q > 10 when the pitch is 1µm. Abstract:
Precast/prestressed concrete hollow core (PCHC) slabs, especially ones with depth more than 300 mm, are susceptible to web-shear failure. The reasons lie on the fact that the production process of PCHC slabs, i.e., the extrusion method (the most common method to cast PCHC slabs nowadays), does not allow them to contain any shear reinforcement. Moreover, due to the presence of the longitudinal voids, cross sections of PCHC slabs are reduced. Therefore, the shear capacity of the slabs depends solely on the tensile strength of concrete which is relatively low. Given that shear is a major concern in using hollow-core slabs, this paper investigates the possibility of adopting steel fibers in PCHC slabs produced by the extrusion method to enhance the shear capacity of the slabs. Three full-scale PCHC slabs with and without hooked-steel fibers were cast and tested until failure. Three different volumetric fiber contents of 0, 0.51 and 0.89% were investigated. The test results showed that there were substantial increases in shear capacity and ductility with the use of hooked-steel fibers. Ultimate shear strength increased with fiber content. In addition, while the specimen without steel fibers and the one with the steel-fiber volume fraction of 0.51% failed in web-shear mode, the specimen with the higher fiber content (0.89%) collapsed in flexural-shear mode. However, as the hooked-steel fibers with the fiber content of 0.89% were used, difficulties in concrete consolidation were observed while concrete was being cast. This could lead to a lower ultimate shear capacity due to a poorer bond between the concrete and the steel fibers.Keywords: hollow-core slabs, shear strength, steel fibers, web-shear failure

This paper deals with a nonlinear finite element analysis to examine the behavior up to failure of cantilever composite steel-concrete beams which are prestressed externally. 'Pre-' means stressing the high strength external tendons in the steel beam section before the concrete slab is added. The composite beam contains a concrete slab which is connected together with steel I-beam by means of perfect shear connectors between the concrete slab and the steel beam which is subjected to static loading. A finite element analysis will be done to study the effects of external prestressed tendons on the composite steel-concrete beams by locating the tendons in different locations (profiles). ANSYS version 12.1 computer program is being used to analyze the represented three-dimensional model of the cantilever composite beam. Corrosion inhibitors are widely used in concrete industry to reduce the corrosion rate of steel rebar which is present in contact with aggressive environments. The present work aims to using Zamzam water from well located within the Masjid al-Haram in Mecca, Saudi Arabia 20 m (66 ft) east of the Kaaba, the holiest place in Islam as corrosion inhibitor for steel in rain water and simulated acid rain. The effect of Zamzam water was investigated by electrochemical impedance spectroscopy (EIS) and Potentiodynamic polarization techniques in Department of Civil Engineering - IUT Saint-Nazaire, Nantes University, France. Zamzam water is considered to be one of the most important steel corrosion inhibitor which is frequently used in different industrial applications. An aluminum coating deposited on mild steel substrate by electric arc spray and diffused to the base steel material by diffusion treatment at 800 and 900°C for 1 and 3 hours in a static air. Alloy layers formed by diffusion at both temperatures were investigated, and their features were compared with those of pack cementation aluminized steel. High-temperature oxidation tests were carried out in air at 600 °C for 145 hours. Results indicated that the aluminide coatings obtained from this process have significantly improved the high-temperature oxidation resistance in both methods due to the Al2O3 scale formation. Furthermore, it showed that the isothermal oxidation resistance of arc spray technique is better than pack cementation method. Abstract:
Tofu industry is one of the local food industry which is can being competitive industry in the ASEAN Economic Community (AEC). However, a lot of tofu entrepreneurs just thinking how to produce good quality product without considering the impact of environmental conditions from the production process. Production of tofu per day requires a number of 15 kg with liquid waste generated is 652.5 liters. That liquid waste is discharged directly into waterways, whereas tofu liquid waste contains organic compounds that quickly unraveled, so it can pollute waterways. In addition, tofu liquid waste is high in Biological Oxygen Demand (BOD), Chemical Oxygen Demand (COD), Total Suspended Solid (TSS), nitrogen and phosphorus. This research is aim to create a method of handling liquid waste effectively and efficiently by using water lettuce. The method is done by observation and experiment by using phytoremediation method in the tofu liquid waste using water lettuce and adding aeration to reduce the concentration of contaminants. The results of the research analyzed the waste quality standard parameters based on SNI (National Standardization Agency of Indonesia). The efficiency concentration and parameters average of tofu liquid waste are obtained pH 3,42% (from 4,0 to be 3,3), COD 76,13% (from 3579 ppm to be 854 ppm), BOD 55 % (from 11600 ppm to be 5242 ppm), TSS 93,6% (from 3174 ppm to be 203 ppm), turbidity is 64,8% (from 977 NTU to be 1013 NTU), and temperature 36oC (from 45oC to be 40oC). The efficiency of these parameters indicates a safe value for the effluent to be channeled in waterways. Water lettuce and tofu liquid waste phytoremediation result will be used as biogas as renewable energy.Keywords: aeration, phytoremediation, water letuce, tofu liquid waste

Abstract:
In this paper, simplified equations to predict maximum inter-story drift demands of steel framed buildings are proposed in terms of two ground motion intensity measures based on the acceleration spectral shape. For this aim, the maximum inter-story drifts of steel frames with 4, 6, 8 and 10 stories subjected to narrow-band ground motion records are estimated and compared with the spectral acceleration at first mode of vibration Sa(T1) which is commonly used in earthquake engineering and seismology, and with a new parameter related with the structural response known as INp. It is observed that INp is the parameter best related with the structural response of steel frames under narrow-band motions. Finally, equations to compute maximum inter-story drift demands of steel frames as a function of spectral acceleration and INp are proposed.Keywords: intensity measures, spectral shape, steel frames, peak demands

This study aimed to investigate and examine the structural behaviour of steel fibre reinforced concrete slabs when subjected to impact loading using drop weight method. A number of compressive tests, tensile splitting tests, as well as impact tests were conducted. The experimental work consists of testing both conventional reinforced slabs and SFRC slabs. Parameters to be considered for carrying out the test will consist of the volume fraction of steel fibre, type of steel fibres, drop weight height and number of blows. Plasma arc welding technology is used for welding SS-304L with A-36. Two different optimize butt welded joints were produced by using austenitic filler alloy E-309L and with direct fusion at 45 A, 2mm/sec by keeping plasma gas flow rate at 0.5LPM. Microstructure analysis of the weld bead was carried out. Discussion and / or findings: In general, central gaze analysis results of water we can conclude that the contents of the various physico-chemical parameters greatly exceed the standards. The research work focuses on exploring better technologies for solar energy storage. The research has the objective of substituting fossil fuels with renewable solar energy technology. This was the reason that motivated the research team to search for alternatives to develop an eco-friendly desalination process, which fully depends on the solar energy source. The Authors also investigated the potential of using different salt mixtures for better solar energy storage and better pure water productivity. Experiments were conducted to understand the impacts of solar energy collection and storage techniques on heat accumulation, heat storage capacity of various compositions of salt mixtures. Based on the experiments conducted, the economic and technical advantages of the integrated water desalination was assessed. In the development of the times, there are many materials used to plan a building structure. Steel became one of the most widely used materials in building construction that works as the main structure. Steel Castellated Beam is a type of innovation in the use of steel in building construction. Steel Castellated Beam is a beam that used for long span construction (more than 10 meters). The Castellated Beam is two steel profiles that unified into one to get the appropriate profile height (more than 10 meters). The profile is perforated to minimize the profile's weight, increase the rate, save costs, and have architectural value. The perforations shape in the Castellated Beam can be circular, elliptical, hexagonal, and rectangular. The Castellated beam has a height (h) almost 50% higher than the initial profile thus increasing the axial bending value and the moment of inertia (Iₓ). In this analysis, there are 3 specimens were used with 12.1 meters span of Castellated Beam as the sample with varied perforation, such us round, hexagon, and octagon. Castellated Beam testing system is done with computer-based applications that named Staad Pro V8i. It is to provide a central load in the middle of the steel beam span. It aims to determine the effect of perforation on bending behavior on the steel Castellated Beam by applying some form of perforations on the steel Castellated Beam with test specimen WF 200.100.5.5.8. From the analysis, results found the behavior of steel Castellated Beam when receiving such central load. From the results of the analysis will be obtained the amount of load, shear, strain, and Δ (deflection). The result of analysis by using Staad Pro V8i shows that with the different form of perforations on the profile of Castellated steel, then we get the different tendency of inertia moment. From the analysis, results obtained the moment of the greatest inertia can increase the stiffness of Castellated steel. By increasing the stiffness of the steel Castellated Beam the deflection will be smaller, so it can withstand the moment and a large strength. The results of the analysis show that the most effective and efficient perforations are the steel beam with a hexagon perforation shape.Keywords: Castellated Beam, the moment of inertia, stress, deflection, bending test

Abstract:
Hydrogen as a sustainable fuel has the highest energy density per mass as compared to conventional non-renewable sources. As the world looks to move towards sustainability, especially in the sectors of aviation and automotive, it becomes important to address the issue of storage of hydrogen as compressed gas in high-pressure tanks. To improve the design for the efficient storage and transportation of Hydrogen, this paper presents the design and stress analysis of Dome Composite Overwrapped Pressure Vessels (COPVs) using the geodesic trajectory approach. The geodesic trajectory approach is used to optimize the dome design, resulting in a lightweight and efficient structure. Python scripting is employed to implement the mathematical modeling of the COPV, and after validating the model by comparison to the published paper, stress analysis is conducted using Abaqus commercial code. The results demonstrate the effectiveness of the geodesic trajectory approach in achieving a lightweight and structurally sound dome design, as well as the accuracy and reliability of the stress analysis using Abaqus commercial code.
