27249 Production of Novel Antibiotics by Importing eryK and eryG Genes in Streptomyces fradiae
Authors: Neda Gegar Goshe, Hossein Rassi
Abstract:
The antibacterial properties of macrolide antibiotics (such as erythromycin and tylosin) depend ultimately on the glycosylation of otherwise inactive polyketide lactones. Among the sugars commonly found in such macrolides are various 6-deoxyhexoses including the 3-dimethylamino sugars mycaminose and desosamine (4-deoxymycaminose). Some macrolides (such as tylosin) possess multiple sugar moieties, whereas others (such as erythromycin) have two sugar substituents. Streptomyces fradiae is an ideal host for development of generic polyketide-overproducing strains because it contains three of the most common precursors-malonyl-CoA, methylmalonyl-CoA and ethylmalonyl-CoA-used by modular PKS, and is a host that is amenable to genetic manipulation. As patterns of glycosylation markedly influence a macrolide's drug activity, there is considerable interest in the possibility of using combinatorial biosynthesis to generate new pairings of polyketide lactones with sugars, especially 6-deoxyhexoses. Here, we report a successful attempt to alter the aminodeoxyhexose-biosynthetic capacity of Streptomyces fradiae (a producer of tylosin) by importing genes from the erythromycin producer Saccharopolyspora erythraea. The biotransformation of erythromycin-D into the desired major component erythromycin-A involves two final enzymatic reactions, EryK-catalyzed hydroxylation at the C-12 position of the aglycone and EryG-catalyzed O methylation at the C-3 position of macrose .This engineered S. fradiae produced substantial amounts of two potentially useful macrolides that had not previously been obtained by fermentation. Keywords: Streptomyces fradiae, eryK and eryG genes, tylosin, antibiotics

Authors: Mengmeng Liu, J. David Frost
Abstract:
Interstate highway system within the Atlanta Perimeter plays an important role in residents’ daily life. The serious influence of Atlanta I-85 Collapses implies that transportation system in the region lacks a cohesive and comprehensive transportation plan. Therefore, disaster resilience analysis of the transportation system is necessary. Resilience is the system’s capability to persist or to maintain transportation services when exposed to changes or shocks. This paper analyzed the resilience of the whole transportation system within the Perimeter and see how removing interstates within the Perimeter will affect the resilience of the transportation system. The data used in the paper are Atlanta transportation networks and LEHD Origin-Destination Employment Statistics data. First, we calculate the traffic flow on each road section based on LEHD data assuming each trip travel along the shortest travel time paths. Second, we calculate the measure of resilience, which is flow-based connectivity and centrality of the transportation network, and see how they will change if we remove each section of interstates from the current transportation system. Finally, we get the resilience function curve of the interstates and identify the most resilient interstates section. The resilience analysis results show that the framework of calculation resilience is effective and can provide some useful information for the transportation planning and sustainability analysis of the transportation infrastructures. Keywords: connectivity, interstate highway system, network analysis, resilience analysis

Authors: P. C. Tewari, Rajiv Kumar, Dinesh Khanduja
Abstract:
This paper discusses the performance modeling and availability analysis of Yarn Dyeing System of a Textile Industry. The Textile Industry is a complex and repairable engineering system. Yarn Dyeing System of Textile Industry consists of five subsystems arranged in series configuration. For performance modeling and analysis of availability, a performance evaluating model has been developed with the help of mathematical formulation based on Markov-Birth-Death Process. The differential equations have been developed on the basis of Probabilistic Approach using a Transition Diagram. These equations have further been solved using normalizing condition in order to develop the steady state availability, a performance measure of the system concerned. The system performance has been further analyzed with the help of decision matrices. These matrices provide various availability levels for different combinations of failure and repair rates for various subsystems. The findings of this paper are, therefore, considered to be useful for the analysis of availability and determination of the best possible maintenance strategies which can be implemented in future to enhance the system performance. Keywords: performance modeling, markov process, steady state availability, availability analysis

Authors: Aliyu Uthman Abdulkadir
Abstract:
Insurgent reports in Nigeria have become a recurring focus in the media due to the significance of language choices. This paper investigates these reports with the aim of identifying various pragmatic practices and exploring the role of the media in shaping public perception of insurgency. Three Nigerian newspapers The Punch, This Day, and The Guardian were selected for analysis between December 2022 and January 2023. Five media reports were examined to uncover the pragmatic functions embedded in the discourse. The study reveals that the media employ implicit acts such as exposing, sensitizing, informing, castigating, reprimanding, and shaming to depict insurgent activities in the country. The analysis also highlights how the use of presupposed ideologies enhances the delivery and acceptance of information related to insurgent actions. The study concludes that the media's portrayal of insurgency is often biased, as reflected in the data analysis. Keywords: insurgency, pragmatic acts, bias, framing, ideoligies
Procedia PDF Downloads 1527239 Potential Role of Arbuscular Mycorrhizal (AM) Fungi in CO₂-Sequestration During Bipartite Interaction with Host Plant Oryza Sativa
Authors: Sadhana Shukla, Pushplata Singh, Nidhi Didwania
Abstract:
Arbuscular mycorrhizal (AM) fungi are a highly advantageous and versatile group of fungi that significantly contribute to the formation of soil organic matter by creating a demand for plant carbon (C) and distributing it through below-ground hyphal biomass, regardless of their substantial contribution in enhancing net primary productivity and accumulating additional photosynthetic fixed C in the soil. The genetic role of AM fungi in carbon cycling is largely unexplored. In our study, we propose that AM fungi significantly interact with the soil, particularly: the provision of photosynthates by plants. We have studied the expression of AM fungi genes involved in CO₂ sequestration during host-plant interaction was investigated by qPCR studies. We selected Rhizophagus proliferus (AM fungi) and Oryza sativa (Rice) (inoculated with or without 200ppg AMF inoculums per plant) and investigated the effect of AM fungi on soil organic carbon (SOC) and rice growth under field conditions. Results thus provided faster SOC turnover, 35% increased nutrient uptake in plants and pronounced hyphal biomass of AM fungi which enhanced soil carbon storage by 15% in comparison to uninoculated plants. This study will offer a foundation for delving into various carbon-soil studies while also advancing our comprehension of the relationship between AM fungi and the sustainability of agricultural ecosystems. Keywords: arbuscular mycorrhizal (AM) fungi, carbon sequestration, gene expression, soil health, plant development.

Authors: Irakli Gamkrelidze, David Shengelia, Giorgi Chichinadze, Tamara Tsutsunava, Giorgi Beridze, Tamara Tsamalashvili, Ketevan Tedliashvili, Irakli Javakhishvili
Abstract:
The Caucasus is situated between the Eurasian and Africa-Arabian plates and represents a component of the Mediterranean (Alpine-Himalayan) collision belt. The Loki crystalline massif crops out within one of the terranes of the Caucasus – Baiburt-Sevanian terrane. By the end of 2018, a digital geological map (1:50 000) of the Loki massif was compiled. The presented map is of great importance for the region since there is no large-scale geological map which reflects the present standards of the geological study of the massif up to the last time. The existing State Geological Map of the Loki massif is very outdated. A new map drown by using GIS (Geographic Information System) technology is loaded with multi-informative details that include: specified contours of geological units and separate tectonic scales, key mineral assemblages and facies of metamorphism, temperature conditions of metamorphism, ages of metamorphism events and the massif rocks, genetic-geodynamic types of magmatic rocks. Explanatory note, attached to the map includes the large specter of scientific information. It contains characterization of the geological setting, composition and petrogenetic and geodynamic models of the massif formation. To create a geological map of the Loki crystalline massif, appropriate methodologies were applied: a sampling of rocks, GIS technology-based mapping of geological units, microscopic description of the material, composition analysis of rocks, microprobe analysis of minerals and a new interpretation of obtained data. To prepare a digital version of the map the appropriated activities were held including the creation of a common database. Finally, the design was created that includes the elaboration of legend and the final visualization of the map. The results of the study presented in the explanatory note are given below. The autochthonous gneissose quartz diorites of normal alkalinity and sub-alkaline gabbro-diorites included in them belong to different phases of magmatism. They represent “igneous” granites corresponding to mixed mantle-crustal type granites. Four tectonic plates of the allochthonous metamorphic complex–Lower Gorastskali, Sapharlo–Lok-Jandari, Moshevani, and Lower Gorastskali differ from each other by structure and degree of metamorphism. The initial rocks of these plates are formed in different geodynamic conditions and during the Early Bretonian orogeny while overthrusting due to tectonic compression they form a thick tectonic sheet. The Lower Gorastskali overthrust sheet is a fragment of ophiolitic association corresponding to the Paleotethys oceanic crust. The protolith of the ophiolitic complex basites corresponds to the tholeiitic series of basalts. The Sapharlo–Lok-Jandari overthrust sheet is metapelites, metamorphosed in conditions of greenschist facies of regional metamorphism. The regional metamorphism of Moshevani overthrust sheet crystalline schists quartzites corresponds to a range from greenschist to hornfels facies. The “mélange” is built of rock fragments and blocks of above-mentioned overthrust sheets. Sub-alkaline and normal alkaline post-metamorphic granites of the Loki crystalline massif belong to “igneous” and rarely to “sialic” and “anorogenic” types of granites. Keywords: digital geological map, 1:50 000 scale, crystalline massif, the caucasus

Abstract:
With the increasing popularity of document analysis and recognition systems, text detection (TD) and optical character recognition (OCR) in document images become challenging tasks. However, according to our best knowledge, no publicly available datasets for these particular problems exist. In this paper, we introduce a Distorted Document Images dataset (DDI-100) and provide a detailed analysis of the DDI-100 in its current state. To create the dataset we collected 7000 unique document pages, and extend it by applying different types of distortions and geometric transformations. In total, DDI-100 contains more than 100,000 document images together with binary text masks, text and character locations in terms of bounding boxes. We also present an analysis of several state-of-the-art TD and OCR approaches on the presented dataset. Lastly, we demonstrate the usefulness of DDI-100 to improve accuracy and stability of the considered TD and OCR models. Keywords: document analysis, open dataset, optical character recognition, text detection

Authors: Himanshu Pavagadhi, Maunik Jani, S. M. Vyas, Jaymin Ray, Vimal Patel, Piyush Patel, Jignesh P. Raval
Abstract:
The preparation of pure and Sn-doped cadmium sulfide (CdS) pellets was carried out using a compression technique with a pelletizer. The energy dispersive X-ray (EDX) analysis is used to confirm the purity and stoichiometric ratio of Cd, S, and Sn in the prepared pellets. The surface morphology of the pellets was examined using a scanning electron microscope. Both XRD and Raman scattering spectrum analysis confirmed the doping effect in the CdS pellets. The X-ray diffraction (XRD) analysis confirmed the hexagonal structure and revealed that the grain size decreases with increasing Sn dopant concentration in the parent CdS pellet. The optical properties of the pellets were evaluated by measuring diffuse reflectance using a UV-vis spectrophotometer. The analysis indicated that as the Sn concentration increases in the parent CdS pellet, the optical band gap decreases. This implies that the optical properties of the CdS material are also affected by the Sn dopant. Keywords: CdS, Sn dopant, UV-Spetrophotometer, XRD

Authors: Yefrem Yefremov
Abstract:
In Kazakhstan, there are approximately 100,000 ethnic Koreans with the ethnonym "Koryo Saram". They are part of the global Korean diaspora around the world, deported to Kazakhstan by Stalin’s decree in 1937. Koryo Saram's diasporic identity is a composite of numerous identities based on a shared cultural heritage of the USSR and independent Kazakhstan and has mosaic character. The author has conducted a sociological survey to find out the main features of the identity of the Koryo Saram diaspora. The purpose of this paper is to depict the degree of ethnic, cultural, and diasporic identity of Koryo Saram and which effect on the preserving Korean diaspora in Kazakhstna do they have. The following elements impacting the above-mentioned identities were investigated in the survey: criteria by which Koryo Saram perceive themselves to be Korean, attitude of Koryo Saram to their ethnicity, degree of feeling of ethnocultural similarity between Koreans of Kazakhstan and Koreans of the Republic of Korea, degree of association of Koreans of Kazakhstan with other Koreans living in other CIS countries, degree of practicing Korean traditions Koryo Saram's attitudes towards interethnic marriages. The primary factor in defining the identity among the respondents is the factor of ethnic origin. Nationality is the second most significant component in establishing Koryo Saram’s identity. The maintenance of "Koreanness" of Koryo Sarams in the context of a multiethnic community, particularly in Kazakhstan, is based on genetic elements as well as the preservation of the culture. In conclusion, the high level of preserving Korean identity is being observed in the Korean Diaspora of Kazakhstan. Keywords: diasporic identity, diaspora, ethnic identity, identity markers, korean diaspora, koreans of kazakhstan, koryo saram, multiethnicity

Authors: Sai Sankalp Vemavarapu
Abstract:
This paper discusses the application of machine learning in the field of transportation engineering for predicting engineering properties of pavement more accurately and efficiently. Predicting the elastic properties aid us in assessing the current road conditions and taking appropriate measures to avoid any inconvenience to commuters. This improves the longevity and sustainability of the pavement layer while reducing its overall life-cycle cost. As an example, we have implemented differential evolution (DE) in the back-calculation of the elastic modulus of multi-layered pavement. The proposed DE global optimization back-calculation approach is integrated with a forward response model. This approach treats back-calculation as a global optimization problem where the cost function to be minimized is defined as the root mean square error in measured and computed deflections. The optimal solution which is elastic modulus, in this case, is searched for in the solution space by the DE algorithm. The best DE parameter combinations and the most optimum value is predicted so that the results are reproducible whenever the need arises. The algorithm’s performance in varied scenarios was analyzed by changing the input parameters. The prediction was well within the permissible error, establishing the supremacy of DE.Keywords: cost function, differential evolution, falling weight deflectometer, genetic algorithm, global optimization, metaheuristic algorithm, multilayered pavement, pavement condition assessment, pavement layer moduli back calculation

Authors: Mariam Waheed Mekheimar
Abstract:
Nascent research is conducted to the advancement of discourse analysis to include different modes as images, sound, and text. The focus of this study will be to elucidate how images are embedded with texts in an audio-visual medium as cinema to send political messages; it also seeks to broaden our understanding of politics beyond a relatively narrow conceptualization of the 'political' through studying non-traditional discourses as the cinematic discourse. The aim herein is to develop a systematic approach to film analysis to capture political meanings in films. The method adopted in this research is Multimodal Discourse Analysis (MDA) focusing on embedding visuals with texts. As today's era is the era of images and that necessitates analyzing images. Drawing on the writings of O'Halloran, Kress and Van Leuween, John Bateman and Janina Wildfeuer, different modalities will be studied to understand how those modes interact in the cinematic discourse. 'People at the top movie' is selected as an example to unravel the political meanings throughout film tackling the cinematic representation of the notion of social justice. Keywords: Egyptian cinema, multimodal discourse analysis, people at the top, social justice

Authors: Strahinja Kovačević, Sanja Podunavac-Kuzmanović, Lidija Jevrić, Milica Karadžić
Abstract:
The present study is based on the quantitative structure-activity relationship (QSAR) analysis of eighteen compounds with anti-prion activity. The structures and anti-prion activities (expressed in response units, RU%) of the analyzed compounds are taken from CHEMBL database. In the first step of analysis 85 molecular descriptors were calculated and based on them the hierarchical cluster analysis (HCA) and principal component analysis (PCA) were carried out in order to detect potential significant similarities or dissimilarities among the studied compounds. The calculated molecular descriptors were physicochemical, lipophilicity and ADMET (absorption, distribution, metabolism, excretion and toxicity) descriptors. The first stage of the QSAR analysis was simple linear regression modeling. It resulted in one acceptable model that correlates Henry's law constant with RU% units. The obtained 2D-QSAR model was validated by cross-validation as an internal validation method. The validation procedure confirmed the model’s quality and therefore it can be used for prediction of anti-prion activity. The next stage of the analysis of anti-prion activity will include 3D-QSAR and molecular docking approaches in order to select the most promising compounds in treatment of prion diseases. These results are the part of the project No. 114-451-268/2016-02 financially supported by the Provincial Secretariat for Science and Technological Development of AP Vojvodina. Keywords: anti-prion activity, chemometrics, molecular modeling, QSAR
