2642 The Diffusion of Membrane Nanodomains with Specific Ganglioside Composition
Authors: Barbora Chmelova, Radek Sachl
Abstract:
Gangliosides are amphipathic membrane lipids. Due to the composition of bulky oligosaccharide chains containing one or more sialic acids linked to the hydrophobic ceramide base, gangliosides are classified among glycosphingolipids. This unique structure induces a high self-aggregating tendency of gangliosides and, therefore, the formation of nanoscopic clusters called nanodomains. Gangliosides are preferentially present in an extracellular membrane leaflet of all human tissues and thus have an impact on a huge number of biological processes, such as intercellular communication, cell signalling, membrane trafficking, and regulation of receptor activity. Defects in their metabolism, impairment of proper ganglioside function, or changes in their organization lead to serious health conditions such as Alzheimer´s and Parkinson´s diseases, autoimmune diseases, tumour growth, etc. This work mainly focuses on ganglioside organization into nanodomains and their dynamics within the plasma membrane. Current research investigates static ganglioside nanodomains characterization; nevertheless, the information about their diffusion is missing. In our study, fluorescence correlation spectroscopy is implemented together with stimulated emission depletion (STED-FCS), which combines the diffraction-unlimited spatial resolution with high temporal resolution. By comparison of the experiments performed on model vesicles containing 4 % of either GM1, GM2, or GM3 and Monte Carlo simulations of diffusion on the plasma membrane, the description of ganglioside clustering, diffusion of nanodomains, and even diffusion of ganglioside molecules inside investigated nanodomains are described. Keywords: gangliosides, nanodomains, STED-FCS, flourescence microscopy, membrane diffusion

Authors: Gizem Buldum, Alexander Bismarck, Athanasios Mantalaris
Abstract:
Cellulose is the most abundant biopolymer on earth and it is currently being utilised in a multitude of industrial applications. Over the last 30 years, attention has been paid to the bacterial cellulose (BC), since BC exhibits unique physical, chemical and mechanical properties when compared to plant-based cellulose, including high purity and biocompatibility. Although Acetobacter xylinum is the most efficient producer of BC, it’s long doubling time results in insufficient yields of the cellulose production. This limits widespread and continued use of BC. In this study, E. coli BL21 (DE3) or E. coli HMS cells are selected as host organisms for the expression of bacterial cellulose synthase operon (bcs) of A.xylinum. The expression system is created based on pET-Duet1 and pCDF plasmid vectors, which carry bcs operon. The results showed that all bcs genes were successfully transferred and expressed in E.coli strains. The expressions of bcs proteins were shown by SDS and Native page analyses. The functionality of the bcs operon was proved by congo red binding assay. The effect of culturing temperature and the inducer concentration (IPTG) on cell growth and plasmid stability were monitored. The percentage of plasmid harboring cells induced with 0.025 mM IPTG was obtained as 85% at 22˚C in the end of 10-hr culturing period. It was confirmed that the high output cellulose production machinery of A.xylinum can be transferred into other organisms. Keywords: bacterial cellulose, biopolymer, recombinant expression system, production

Authors: Ji Hye Im, Nguyen T. T. Phuong, Keon Wook Kang
Abstract:
Estrogens are important for the development and growth of estrogen receptor (ER)-positive breast cancer, for which anti-estrogen therapy is one of the most effective treatments. However, its efficacy can be limited by either de novo or acquired resistance. Aromatase is a key enzyme for the biosynthesis of estrogens, and inhibition of this enzyme leads to profound hypoestrogenism. Here, we found that the basal expression and activity of aromatase were significantly increased in tamoxifen (TAM)-resistant human breast cancer (TAMR-MCF-7) cells compared to control MCF-7 cells. We further revealed that aromatase immunoreactivity in tumor tissues was increased in recurrence group after TAM therapy compared to non-recurrence group after TAM therapy. Phosphorylation of Akt, extracellular signal-regulated kinase (ERK), and p38 kinase were all increased in TAMR-MCF-7 cells. Inhibition of phosphoinositide 3-kinase (PI3K) suppressed the transactivation of the aromatase gene and its enzyme activity. Furthermore, we have also shown that PI3K/Akt-dependent cAMP-response element binding protein (CREB) activation was required for the enhanced expression of aromatase in TAMR-MCF-7 cells. Our findings suggest that aromatase expression is up-regulated in TAM-resistant breast cancer via PI3K/Akt-dependent CREB activation. Keywords: TAMR-MCF-7, CREB, estrogen receptor, aromatase
Procedia PDF Downloads 4142639 The Relationship between the Parameters of Laser 3D Printing of Titanium Alloy and Its Strength Properties
Authors: Lubov Magerramova, Vladimir Isakov, Michail Petrov
Abstract:
A methodology for calculating and modeling technological modes of laser 3D printing of Ti6Al4V powder alloy samples has been developed. ProXDPM320 3D printer was used. The technological model that takes into account the multifactorial influence of modes and conditions of additive cultivation on characteristics and strength properties of titanium samples has been created. Process control parameters and an order parameter, to which the others are subordinate, were established. Using the iterative method, the optimal technological parameters for the additive growth of cylindrical samples were calculated. The calculations were combined with data obtained during virtual 3D printing in the Altair Inspire software environment. The samples were subjected to short-term tensile strength tests at normal temperature on a servo-hydraulic machine “LFV-100”. As a result, deformation diagrams were constructed, and mechanical characteristics such as proportionality limit, conditional yield strength, tensile strength, elastic modulus, relative elongation, and stress at break were obtained. Comparison of these characteristics with those for the industrial alloy Ti6Al4V showed acceptable agreement. Some of the synthesized samples were subjected to laser shock treatment to increase fatigue strength. The results obtained were used to validate the mathematical model of 3D printing of titanium alloys. Keywords: additive technology, titanium alloy, numerical simulation, strength tests

Authors: Anwar Elsadat Elmahal, Ahmed Abdalla
Abstract:
The population in Sudan has recently grown to a significant level, Khartoum city the capital has the major portion of this growth. Khartoum is separated by three Niles and linked by eight bridges to Khartoum North and Omdurman. The construction of these bridges disrupted the natural flow of water and sediments which will consequently be reflected on the geomorphological settings of fluvial islands including erosion and sedimentation patterns. The objective of this study is to monitor and assess the development of fluvial islands in Khartoum State up to Sabaloka Gorge using Remote Sensing (RS) and Geographical Information System (GIS) techniques. Landsat Images captured during the period from 1975-2015 with standard false color and standardized 30 m resolution were found useful in understanding the impacts of bridges on disrupting the fluvial cycle. Consequently, the rates, trends of erosions and deposition, and the development of fluvial islands are explained. GIS provides the-state-of-the-art tools in mapping, delineating the fluvial islands during different periods and in quantifying the changes that occurred to fluvial islands as well as creating the geographic databases for the Islands in Khartoum State. It was found that, the developments, shapes and sizes of the islands are directly affected by the construction of bridges, specifically in the Nile River from Tutti Island to Sabaloka gorge. Keywords: fluvial islands, fluvial cycle, GIS and remote Sensing, Khartoum State, landsat, Sabaloka Gorge

Authors: Athena Pedro, Laura Bradfield, Mike Dare, Zandile Bantwana, Ashley Nayman
Abstract:
The purpose of the research was to explore mother’s unique experience and knowledge of mothering in the first 1000 day of their child’s life, from birth to age 2. The study used a qualitative research methodology with an exploratory research design. A sample of 12 mothers was used, comprising different racial backgrounds from low income areas in the Western Cape. The data was collected by means of semi-structured, in-depth interviews, which were transcribed verbatim, analysed using Braun’s and Clark’s (2006) six phases of thematic analysis. Some of the findings revealed that the mothers who participated in the study were consistently unable to feed their children and themselves due to profound and extreme situations of poverty, stress, and lack of infrastructural support. These mothers residing in low-income communities are not adequately supported both financially and socially and are often unable to meet the needs of their infants within the first 1000 days. Given the consequential nature of this period, it is imperative that mothers are able to access such support. Single mothers especially are in need of social and financial support. Appropriate interventions are required to assist mothers generally but more specifically, mothers who have children within the first 1000 days of life. By implementing appropriate interventions to address these needs, it will assist mothers to ensure optimal developmental growth of their children. This will positively impact the developmental trajectory of children in South Africa. Keywords: caring, economic hardship, first one thousand days, mothers

Abstract:
According to the terminology of Angle, the functional mandibular prognathism belongs to the class III malocclusion. Considered as a functional anomaly with considerable aesthetic prejudice, it needs to be detected and treated early to prevent it from becoming skeletal anomaly. With this in mind, early treatment enables a correct anterior guide to be restored quickly, and above all to rehabilitate craniofacial growth. Material and methodology Patient G.H, aged 11 years, who was treated in the Department of Dentofacial Orthopaedics at Oran University Hospital. He presented with functional mandibular prognathism and a sever crowding, which put the upper canines at risk of impaction. Results The orthodontic treatment of this case was divided into two distinct phases, the first of which aimed to correct the functionnal mandibular prognathism and thus re-establish a correct anterior articulation, using a Biourge appliance. The second phase aimed to create the needed space for the eruption of the upper canines, thus eliminating the risk of impaction thanks to multi-attachment fixed therapy. Conclusion Through this clinical case, we have once again been able to demonstrate the value of early caring for young patients. Although these are simple methods, the key is to know how to introduce them in the right way and at the right time. It would therefore seem useful to democratise this type of practice as part of the day-to-day management carried out in the dental practice. We'll finish by quoting Jean Rotrou, the French poet, who said ‘When you can prevent, it's weakness to wait’.Keywords: functional mandibular prognathism, early treatment, impaction, orthodontics

Authors: Anwar Ul Haque, Waqar Asrar, Erwin Sulaeman, Jaffar S. M. Ali
Abstract:
Pressure distribution data of the standard airfoils is usually used for the calibration purposes in subsonic wind tunnels. Results of such experiments are quite old and obtained by using the model in the spanwise direction. In this manuscript, pressure distribution over NACA 4412 airfoil model was presented by placing the 3D model in the lateral direction. The model is made of metal with pressure ports distributed longitudinally as well as in the lateral direction. The pressure model was attached to the floor of the tunnel with the help of the base plate to give the specified angle of attack to the model. Before the start of the experiments, the pressure tubes of the respective ports of the 128 ports pressure scanner are checked for leakage, and the losses due to the length of the pipes were also incorporated in the results for the specified pressure range. Growth rate maps of the boundary layer thickness were also plotted. It was found that with the increase in the velocity, the dynamic pressure distribution was also increased for the alpha seep. Plots of pressure distribution so obtained were overlapped with those obtained by using XFLR software, a low fidelity tool. It was found that at moderate and high angles of attack, the distribution of the pressure coefficients obtained from the experiments is high when compared with the XFLR ® results obtained along with the span of the wing. This under-prediction by XFLR ® is more obvious on the windward than on the leeward side. Keywords: subsonic flow, boundary layer, wind tunnel, pressure testing
Procedia PDF Downloads 3222634 Design and Application of NFC-Based Identity and Access Management in Cloud Services
Authors: Shin-Jer Yang, Kai-Tai Yang
Abstract:
In response to a changing world and the fast growth of the Internet, more and more enterprises are replacing web-based services with cloud-based ones. Multi-tenancy technology is becoming more important especially with Software as a Service (SaaS). This in turn leads to a greater focus on the application of Identity and Access Management (IAM). Conventional Near-Field Communication (NFC) based verification relies on a computer browser and a card reader to access an NFC tag. This type of verification does not support mobile device login and user-based access management functions. This study designs an NFC-based third-party cloud identity and access management scheme (NFC-IAM) addressing this shortcoming. Data from simulation tests analyzed with Key Performance Indicators (KPIs) suggest that the NFC-IAM not only takes less time in identity identification but also cuts time by 80% in terms of two-factor authentication and improves verification accuracy to 99.9% or better. In functional performance analyses, NFC-IAM performed better in salability and portability. The NFC-IAM App (Application Software) and back-end system to be developed and deployed in mobile device are to support IAM features and also offers users a more user-friendly experience and stronger security protection. In the future, our NFC-IAM can be employed to different environments including identification for mobile payment systems, permission management for remote equipment monitoring, among other applications. Keywords: cloud service, multi-tenancy, NFC, IAM, mobile device
Procedia PDF Downloads 4392633 The Quality of Public Space in Mexico City: Current State and Trends
Authors: Mildred Moreno Villanueva
Abstract:
Public space is essential to strengthen the social and urban fabric and the social cohesion; there lies the importance of its study. Hence, the aim of this paper is to analyze the quality of public space in the XXI century in both quantitative and qualitative terms. In this article, the concept of public space includes open spaces such as parks, public squares and walking areas. To make this analysis we take Mexico City as the case study. It has a population of nearly 9 million inhabitants and it is composed of sixteen boroughs. For this analysis, we consider both, existing public spaces and the government intervention for building and improvement of new and existent public spaces. Results show that on the one hand, quantitatively there is not an equitable distribution of public spaces because of both, the growth of the city itself, as well as for the absence of political will to create public spaces. Another factor is the evolution of this city, which has been growing merely in a 'patched pattern', where public space has played no role at all with a total absence of urban design. On the other hand, qualitatively, even the boroughs with the most public spaces have not shown interest in making these spaces qualitatively inclusive and open to the general population aiming for integration. Therefore, urban projects that privatize public space seem to be the rule, rather than a rehabilitation effort of the existent public spaces. Hence, state intervention should reinforce its role as an agent of social change acting in the benefit of the majority of the inhabitants with the promotion of more inclusive public spaces. Keywords: exclusion, inclusion, Mexico City, public space

Authors: Sara Kamalisiahroudi, Zhang Jianbo, Bin Wu, Jun Huang, Laisuo Su
Abstract:
The temperature rising within a battery cell depends on the level of heat generation, the thermal properties and the heat transfer around the cell. The rising of temperature is a serious problem of Lithium-Ion batteries and the internal resistance of battery is the main reason for this heating up, so the heat generation rate of the batteries is an important investigating factor in battery pack design. The delivered power of a battery is directly related to its capacity, decreases in the battery capacity means the growth of the Solid Electrolyte Interface (SEI) layer which is because of the deposits of lithium from the electrolyte to form SEI layer that increases the internal resistance of the battery. In this study two identical cylindrical Lithium-Ion (NCR18650)batteries from the same company with noticeable different in capacity (a fresh and a used battery) were compared for more focusing on their heat generation parameters (entropy coefficient and internal resistance) according to Brandi model, by utilizing potentiometric method for entropy coefficient and EIS method for internal resistance measurement. The results clarify the effect of capacity difference on cell electrical (R) and thermal (dU/dT) parameters. It can be very noticeable in battery pack design for its Safety. Keywords: heat generation, Solid Electrolyte Interface (SEI), potentiometric method, entropy coefficient

Authors: Panpan Feng
Abstract:
The concept of green infrastructure originated in Europe and the United States. It aims to ensure smart growth of urban and rural ecosystems and achieve sustainable urban and rural ecological, social, and economic development by combining it with gray infrastructure in traditional planning. Based on the literature review of the theoretical origin, value connotation, and measurement methods of green infrastructure, this study summarizes the research content of green infrastructure at different scales from the three spatial levels of region, city, and block and divides it into functional dimensions, spatial dimension, and strategic dimension. The results show that in the functional dimension, from region-city-block, the research on green infrastructure gradually shifts from ecological function to social function. In the spatial dimension, from region-city-block, the research on the spatial form of green infrastructure has shifted from two-dimensional to three-dimensional, and the spatial structure of green infrastructure has shifted from single ecological elements to multiple composite elements. From a strategic perspective, green infrastructure research is more of a spatial planning tool based on land management, environmental livability and ecological psychology, providing certain decision-making support. Keywords: green infrastructure, multi-scale, social and ecological functions, spatial strategic decision-making tools

Abstract:
Solar energy is thought as main source of all energy sources on the world and it can be used in many applications like agricultural areas, heating cooling or direct electricity production directly or indirectly. Greenhousing is the first one of the agricultural activities that solar energy can be used directly in. Greenhouses offer us suitable conditions which can be controlled easily for the growth of the plant and they are made by using a covering material that allows the sun light entering into the system. Covering material can be glass, fiber glass, plastic or another transparent element. This study investigates the solar energy usability rates and solar energy benefiting rates of a semi-spherical (modified arch) type greenhouse system according to different orientations and positions which exists under climatic conditions of Bayburt. In the concept of this study it is tried to determine the best direction and best sizes of a semi-spherical greenhouse to get best solar benefit from the sun. To achieve this aim a modeling study is made by using MATLAB. However this modeling study is running for some determined shapes and greenhouses it can be used for different shaped greenhouses or buildings. The basic parameters are determined as greenhouse azimuth angle, the rate of size of long edge to short and seasonal solar energy gaining of greenhouse. Keywords: greenhousing, solar energy, direct radiation, renewable energy

Authors: Yen-Ni Teng, Hsing-Yi Chen, Hsien-An Pan, Yung-Ming Lin, Hany A. Omar, Jui-Hsiang Hung
Abstract:
Leucine-rich repeats and WD repeat domain containing 1 (LRWD1) is highly expressed in the testes of healthy males. On the other hand, LRWD1 is significantly down-regulated in the testicular tissues of patients with severe spermatogenic defects. In our study, the downregulation of LRWD1 expression by shRNA caused a significant reduction of cell growth and mitosis and a noteworthy increase in the cell microtubule atrophy rate. Here, we used EMBOSS CpG plot analysis to explore the promoter region of LRWD1 gene. We found that CpG islands are located between positions -253 to +5 nucleotides upstream from the LRWD1 transcription start site. Luciferase reporter assay revealed that the hypermethylation of the LRWD1 promoter reduced the transcription activity in cells. In addition, quantitative methylation-specific PCR and immunostaining showed that the methylation inhibitor, 5-Aza-2'-deoxycytidine, increased LRWD1 promoter activity, LRWD1 mRNA, protein expression and cell viability. Whereas, the methylation activator, S-adenosylmethionine, caused opposite effects. The overexpression of p53 and Nrf2 in NT2/D1 cells increased LRWD1 promoter activity while 5-fluorodeoxyuridine decreased it. In conclusion, this study highlights evidence that the methylation status of LRWD1 promoter is associated with LRWD1 expression. Since the expression level of LRWD1 plays an important role in spermatogenesis, the methylation status of LRWD1 may serve as a novel molecular diagnostic or therapeutic approach in male's infertility. Keywords: LRWD1, DNA methylation, p53, Nrf2

Authors: Soumitri Bag Majumder, Anindita Chaudhuri
Abstract:
This exploratory study delves into the perception of job dignity among delivery agents in Kolkata, focusing on both food and grocery delivery sectors. The rapid expansion of online delivery platforms in India has led to a significant rise in the delivery service industry. Despite its growth, there is a dearth of research addressing the multifaceted challenges faced by delivery agents. This study aims to bridge this gap by shedding light on their experiences. The study’s objectives include exploring the lived experiences of delivery agents, their work-life balance, and their perception of job dignity. Using a qualitative research approach, the study will conduct semi-structured in-depth interviews with a purposive sample of 10 participants from each sector, consisting of individuals with lower socio-economic backgrounds aged between 18 and 35 years. The Three-Layer Coding framework proposed by Charmaz will guide the data analysis process, encompassing open coding, axial coding, and selective coding. Through this method, the study seeks to uncover emergent themes and patterns that illuminate the participants’ perspectives on job dignity, recognition, and the challenges they encounter. By uncovering their perceptions of job dignity and the challenges they face, the research aims to contribute to the well-being of these workers and inform relevant stakeholders for a more equitable work environment. Keywords: delivery agents, equitable work environment, perception of job dignity, work-life balance
Procedia PDF Downloads 672626 A Quantitative Study on the Structure of Corporate Social Responsibility in India
Authors: Raj C. Aparna
Abstract:
In India, the mandatory clause on Corporate Social Responsibility (CSR) in Companies Act, 2013 has led to varying responses from the companies. From excessive spending to resistance, the private and the public stakeholders have been considering the law from different perspectives. This paper tends to study the characteristics of CSR spending in India with emphasis on the locations to which the funds are routed. This study examines the effects of CSR fund flow on regional development by considering the growth in Gross State Domestic Product (GSDP), agriculture, education and healthcare using panel data for the 29 States in the country. The results confirm that the CSR funds have been instrumental in improving the quality of teaching and healthcare in the areas around the industrial hubs. However, the study shows that the corporates mostly invest in regions which are easily accessible to them, by their physical presence, irrespective of whether the area is developed or not. Such a skewness is visible in the extensive spending in and around the metropolitan cities, the established centers, in the country to which large chunks of CSR funds are channeled. The results show that there is a variation from what the government had proposed while initiating the CSR law to promote social inclusion and equality in the rural and isolated areas in the country. The implication is that even though societal improvement is the aim of CSR, ease of access to the needy is an essential factor in corporate choices. As poverty and lack of facilities are found in the innermost parts, it is vital to have government policies for their aid as corporate help. Keywords: corporate social responsibility, geographic spread, panel data analysis, strategic implementation

Abstract:
The fall army worm (FAW), Spodoptera frugiperda (J. E. Smith) (Lepidoptera, Noctuidae) has recently become one of the major threats to maize production in the world. It is native to tropical and subtropical America and began to spread to many African and a few Asian Countries. A survey for the observation of infestation and collection of fall armyworm was conducted in field planted with maize in the northern part of Katsina state. Eggs and immature stages were collected, place in a plastic container and brought to the laboratory for observation and study of developmental stages. FAW was identified based on the morphological characteristics, i.e. the “Y” inverted shape on the head capsule and the patterns of black spots on the abdominal segments (square and trapezoidal forms). Different growing stage of maize are affected by fall armyworm, but the damage is greatest during the early growing phase of corn. Heavy infestation on the leaves also cause defoliation. Four developmental stages (eggs larvae, pupae and adults) of the FAW were studied when fed with young corn under laboratory conditions. Furthermore, effective scouting or monitoring of FAW could be practice at early stage of growth of maize. Keywords: infestation, katsina, maize, fall armyworm

Abstract:
Background: Despite breakthrough therapeutics in breast cancer, it is one of the main causes of mortality among women worldwide. Thus, drug therapies for treating breast cancer have recently been developed by scientists. Metformin and Sorafenib are well-known therapeutic in breast cancer. In the present study, we combined Sorafenib and PCL-sorafenib with metformin to improve drug absorption and promote therapeutic efficiency. Methods: The MCF-7 cells were treated with Metformin, Sorafenib, or PCL-sorafenib. The growth inhibitory effect of these drugs and cell viability were assessed using MTT and flow cytometry assays, respectively. The expression of targeted genes involved in cell proliferation, signaling, and the cell cycle was measured by Real-time PCR. Results: The results showed that MCF-7 cells treated with Metformin/Sorafenib and PCL-sorafenib/Metformin co-treatment contributed to 50% viability compared to untreated group. Moreover, PI and Annexin V staining tests showed that the cells viability for Metformin/Sorafenib and PCL-sorafenib/Metformin was 38% and 17%, respectively. Furthermore, Sorafenib/Metformin and PCL-sorafenib/Metformin leads to p53 gene expression increase by which they can increase ROS, thereby decreasing GPX4 gene expression. In addition, they affected the expression of BCL2, and BAX genes and altered the cell cycle. Conclusion: Together, the combination of PCL-sorafenib/Metformin and Sorafenib/Metformin increased Sorafenib absorption at lower doses and also leads to apoptosis and oxidative stress increases in MCF-7 cells. Keywords: breast cancer, metformin, nanotechnology, sorafenib

Authors: Abootaleb Shirvani, Svetlozar Rachev
Abstract:
ESG stands for Environmental, Social, and Governance and is a non-financial factor that investors use to specify material risks and growth opportunities in their analysis process. ESG ratings provide a quantitative measure of socially responsible investment, and it is essential to incorporate ESG ratings when modeling the dynamics of asset returns. In this article, we propose a triple subordinated Lévy process for incorporating numeric ESG ratings into dynamic asset pricing theory to model the time series properties of the stock returns. The motivation for introducing three layers of subordinator is twofold. The first two layers of subordinator capture the skew and fat-tailed properties of the stock return distribution that cannot be explained well by the existing Lévy subordinated model. The third layer of the subordinator introduces ESG valuation and incorporates numeric ESG ratings into dynamic asset pricing theory and option pricing. We employ the triple subordinator Lévy model for developing the ESG-valued stock return model, derive the implied ESG score surfaces for Microsoft, Apple, and Amazon stock returns, and compare the shape of the ESG implied surface scores for these stocks. Keywords: ESG scores, dynamic asset pricing theory, multiple subordinated modeling, Lévy processes, option pricing

Authors: Kai-Cheng Hsu, Tzu-Ying Sung, Jinn-Moon Yang
Abstract:
Colorectal cancer (CRC) is one of the main causes of cancer death in the world. Although several drugs have been developed to treat colorectal cancer, such as Regorafenib and 5-FU, their efficacy is often limited by the development of drug resistance. Therefore, development of new drugs with new scaffolds is necessary to treat CRC. Here, we used site-moiety maps to identify inhibitors against PIM1, LIMK1, SRC, and mTOR, which are often overexpressed in CRC. A site-moiety map represents physicochemical properties and moiety preferences of a binding site through anchors. An anchor contains three elements: (1) conserved interacting residues of a binding pocket; (2) moiety preference of the binding pocket; and (3) the type (e.g., hydrogen-bonding or van der Waals interactions) of interaction between the moieties and the binding pocket. Then, we performed a structure-based virtual screening of ~260,000 compounds and selected compound candidates with high site-moiety map scores for bioassays. Among these candidates, compound 1 and compound 2 inhibited the growth of CRC cells with IC50 values of <10 μM. The experimental result of enzyme-based assays indicated that compound 1 is a dual inhibitor against PIM1 (IC50 6 μM) and LIMK1(IC50 11 μM). Compound 2 was predicted as a SRC inhibitor and will be further validated. The compounds inhibited different protein targets compared to the current drugs. We believe that the compounds provide a starting point to design new drugs for CRC treatment. Keywords: colorectal cancer, drug discovery, site-moiety map, virtual screening, PIM1, LIMK1

Authors: Xin Zhao
Abstract:
3D printing is used in creating bone grafts of various architectures by printing materials in a layer-by-layer manner. Traditionally, to make materials printable, heating up or dissolving materials in organic solvents have been used, compromising their capability in loading biomolecules. Photocrosslinkable materials which are initially liquid and printable, and solidified upon light exposure are therefore developed. However, the existing photocrosslinkable materials are either too soft to bear load or non-degradable with potential long-term biocompatibility problems. Here, photocrosslinkable nanocomposite ink is developed composed of poly (lactide-co-propylene glycol-co-lactide) dimethacrylate (PmLnDMA) and hydroxyethyl methacrylate-functionalized hydroxyapatite nanoparticles (nHAMA) mimicking the hairy setae of gecko that can strongly interact with its surroundings to bear high load. Incorporation of nHAMA into PmLnDMA endows the nanocomposite ink with several advantages in (1) improved organic/inorganic interfacial compatibility to increase mechanical strength, (2) readily modulated rheological behaviors, wettability, and biodegradation, (3) enhanced osteoconductivity and osteoinductivity. Abstract:
Tbilisi, a large city of the South Caucasus, is a junction point connecting Asia and Europe, Russia and republics of the Asia Minor. Over the last years, its atmosphere has been experienced an increasing anthropogenic load. Numerical modeling method is used for study of Tbilisi atmospheric air pollution. By means of 3D non-linear non-steady numerical model a peculiarity of city atmosphere pollution is investigated during background western light air. Dust concentration spatial and time changes are determined. There are identified the zones of high, average and less pollution, dust accumulation areas, transfer directions etc. By numerical modeling, there is shown that the process of air pollution by the dust proceeds in four stages, and they depend on the intensity of motor traffic, the micro-relief of the city, and the location of city mains. In the interval of time 06:00-09:00 the intensive growth, 09:00-15:00 a constancy or weak decrease, 18:00-21:00 an increase, and from 21:00 to 06:00 a reduction of the dust concentrations take place. The highly polluted areas are located in the vicinity of the city center and at some peripherical territories of the city, where the maximum dust concentration at 9PM is equal to 2 maximum allowable concentrations. The similar investigations conducted in case of various meteorological situations will enable us to compile the map of background urban pollution and to elaborate practical measures for ambient air protection. Keywords: air pollution, dust, numerical modeling, urban

Authors: Djouhou Fowe Michelle Carole
Abstract:
In the last few decades, there have been considerable efforts by the international community to develop strategies that reduce global poverty and hunger. Despite the modest success in reducing food insecurity, there are still around 795 million people worldwide who remain undernourished, the majority of whom are in sub-Saharan Africa. In many of these impoverished communities, agriculture still remains one of the most important sectors in driving economic growth and reducing poverty. For the growing population, with higher food demand and fixed agricultural land, sustainable intensification is proposed as an important strategy to respond to the challenges of low yields, environmental degradation, and adaptation to climate change. Adoption of agroforestry technologies is increasingly being promoted as a promising solution. This study was conducted to determine the perceptions of the Cameroonian population and farmers on agroforestry. The methodology used was based on a survey to determine their knowledge level of agroforestry, their representation of its advantages and disadvantages, and the reasons that might motivate them whether or not to adopt agroforestry. Participants were randomly selected and received a questionnaire. Data were subjected to a descriptive analysis using SPSS software. The obtained results showed that less than 50% of the general population had already heard about agroforestry at least once; they have basic knowledge about this concept and its advantages. Farmers had been particularly sensitive to tree's food production function and seemed to value their environmental assets. However, various constraints could affect the possible adoption of agroforestry techniques. Keywords: agroforestry, quality and sustainable agriculture, perceptions, advantages, limits
Procedia PDF Downloads 1772618 An Innovative Equipment for ICU Infection Control
Authors: Ankit Agarwal
Abstract:
Background: To develop a fully indigenous equipment which is an innovation in critical care, which can effectively scavenge contaminated ICU ventilator air. Objectives: Infection control in ICUs is a concern the world over. Various modalities from simple hand hygiene to costly antibiotics exist. However, one simple and scientific fact has been unnoticed till date, that the air exhaled by patients harboring MDR and other microorganisms, is released by ventilators into ICU atmosphere itself. This increases infection in ICU atmosphere and poses risk to other patients. Material and Methods: Some parts of the ventilator are neither disposable nor sterilizable. Over time, microorganisms accumulate in ventilator and act as a source of infection and also contaminate ICU air. This was demonstrated by exposing microbiological culture plates to air from expiratory port of ventilator, whereby dense growth of pathogenic microorganisms was observed. The present prototype of the equipment is totally self-made. It has a mechanism of controlled negative pressure, active and passive systems and various alarms and is versatile to be used with any ventilator. Results: This equipment captures the whole of contaminated exhaled air from the expiratory port of the ventilator and directs it out of the ICU space. Thus, it does not allow contaminated ventilator air to release into the ICU atmosphere. Therefore, there is no chance of exposure of other patients to contaminated air. Conclusion: The equipment is first of its kind the world over and is already under patent process. It has rightly been called ICU Ventilator Air Removal System (ICU VARS). It holds a chance that this technique will gain widespread acceptance shall find use in all the ventilators in most of the ICUs throughout the world. Keywords: innovative, ICU Infection Control, microorganism, negative pressure
Procedia PDF Downloads 3532617 Development and Power Characterization of an IoT Network for Agricultural Imaging Applications
Authors: Jacob Wahl, Jane Zhang
Abstract:
This paper describes the development and characterization of a prototype IoT network for use with agricultural imaging and monitoring applications. The sensor and gateway nodes are designed using the ESP32 SoC with integrated Bluetooth Low Energy 4.2 and Wi-Fi. A development board, the Arducam IoTai ESP32, is used for prototyping, testing, and power measurements. Google’s Firebase is used as the cloud storage site for image data collected by the sensor. The sensor node captures images using the OV2640 2MP camera module and transmits the image data to the gateway via Bluetooth Low Energy. The gateway then uploads the collected images to Firebase via a known nearby Wi-Fi network connection. This image data can then be processed and analyzed by computer vision and machine learning pipelines to assess crop growth or other needs. The sensor node achieves a wireless transmission data throughput of 220kbps while consuming 150mA of current; the sensor sleeps at 162µA. The sensor node device lifetime is estimated to be 682 days on a 6600mAh LiPo battery while acquiring five images per day based on the development board power measurements. This network can be utilized by any application that requires high data rates, low power consumption, short-range communication, and large amounts of data to be transmitted at low-frequency intervals. Keywords: Bluetooth low energy, ESP32, firebase cloud, IoT, smart farming

Abstract:
Pollution of the environment is a phenomenon which has taken a big part of importance of the world governments since the second half of the last century, this takes dangerous environmental, economic and social ranges dimensions especially after industrial advancement in industrialized country and good industrial expansion supported with modern technology and as chromium is known to be used in tannery factories. Chromium is considered a harm element to the environment due to its danger and transference through food, air, and water to the plants, animals and people. In this study the capacity of Arundo donax against chromium pollution was conducted. A. donax plants were grown-up under greenhouse conditions in pots contain nursery soil and feeding by Cr synthetic wastewater (0, 0.1, 1.0 and 2.0 mg L-1 ) for four weeks. Leaves, roots and stems dry matter production, color degree values, chlorophyll, growth parameters, and morphological characters were measured. The high Cr concentration was in roots was 1.15 mg kg-1 . Similarly, Cr concentration in stem was 0.469 mg kg-1 at 2.0 mg L-1 supplied Cr. In case of leaves, the maximum Cr concentration was 0.345 mg kg-1 at 2.0 g L-1 supplied Cr. The bioaccumulation and translocation factors was calculated. The macrophyte A. donax L. may be considered to be the most promising plant species in remediation of Cr-contaminated soil and wastewater due to its deeper root system as well as has higher efficiency to absorb chromium and other heavy metals as well. Keywords: Arundo donax, Chromium pollution, heavy metals, phytoremediation, wastewater

Authors: Patcharakamon Nooeaid, Piyachat Chuysrinuan
Abstract:
Electrospun natural polymers-based nanofibers are one of the most interesting materials used in tissue engineering and drug delivery applications. Bead-on-string nanofibers have gained considerable interest for sustained drug release. Vancomycin was used as the model drug and sodium alginate (SA)/soy protein isolated (SPI) as the polymer blend to fabricate the bead-on-string nanofibers by aqueous-based electrospinning. The bead-on-string SA/SPI nanofibers were successfully fabricated by the addition of poly(ethylene oxide) (PEO) as a co-blending polymer. SA-PEO with mass ratio of 70/30 showed the best spinnability with continuous nanofibers without the occurrence of beads. Bead structure formed with the addition of SPI and bead number increased with increasing SPI content. The electrospinning of 80/20 SA-PEO/SPI was obtained as a great promising bead-on-string nanofibers for drug loading, while the solution of 50/50 was not able to obtain continuous fibers. In vitro release tests showed that a more sustainable release profile up to 14 days with less initial burst release on day 1 could be obtained from the bead-on-string fibers than from smooth fibers with uniform diameter. In addition, vancomycin-loaded beaded fibers inhibited the growth of Staphylococcus aureus (S. aureus) bacteria. Therefore, the SA-PEO/SPI nanofibers showed the potential to be used as biomaterials for tissue engineering and drug delivery. Keywords: bead-on-string fibers, electrospinning, drug delivery, tissue engineering

Abstract:
Sulfur-oxidizing bacteria were isolated and then grown on snakefruits seeds forming biofilm. Their performance in sulfide removal were experimentally observed. Snakefruit seeds were then used as packing material in a cylindrical tube. Biological treatment of hydrogen sulfide from biogas was investigated using biofilm on packed bed of snakefruits seeds. Biogas containing 27,9512 ppm of hydrogen sulfide was flown through the bed. Then the hydrogen sulfide concentrations in the outlet at various times were analyzed. A set of simple kinetics model for the rate of the sulfide removal and the bacterial growth was proposed. The axial sulfide concentration gradient in the flowing liquid are assumed to be steady-state. Mean while the biofilm grows on the surface of the seeds and the oxidation takes place in the biofilm. Since the biofilm is very thin, the sulfide concentration in the biofilm is assumed to be uniform. The simultaneous ordinary differential equations obtained were then solved numerically using Runge-Kutta method. The acuracy of the model proposed was tested by comparing the calcultion results using the model with the experimental data obtained. It turned out that the model proposed can be applied to describe the removal of sulfide liquid using bio-filter in packed bed. The values of the parameters were also obtained by curve-fitting. The biofilter could remove 89,83 % of the inlet of hydrogen sulfide from biogas for 2.5 h, and optimum loading of 8.33 ml/h.Keywords: Sulfur-oxidizing bacteria, snakefruits seeds, biofilm, packing material, biogas
