7663 Review on Application of DVR in Compensation of Voltage Harmonics in Power Systems
Authors: S. Sudhharani
Abstract:
Energy distribution networks are the main link between the energy industry and consumers and are subject to the most scrutiny and testing of any category. As a result, it is important to monitor energy levels during the distribution phase. Power distribution networks, on the other hand, remain subject to common problems, including voltage breakdown, power outages, harmonics, and capacitor switching, all of which disrupt sinusoidal waveforms and reduce the quality and power of the network. Using power appliances in the form of custom power appliances is one way to deal with energy quality issues. Dynamic Voltage Restorer (DVR), integrated with network and distribution networks, is one of these devices. At the same time, by injecting voltage into the system, it can adjust the voltage amplitude and phase in the network. In the form of injections and three-phase syncing, it is used to compensate for the difficulty of energy quality. This article examines the recent use of DVR for power compensation and provides data on the control of each DVR in distribution networks. Keywords: dynamic voltage restorer (DVR), power quality, distribution networks, control systems(PWM)

Abstract:
The research deals with the beginnings of the vernacular Jordanian architecture since the establishment of the Jordanian state in the early nineteenth century until now, where the Jordanian architecture was based on the interactions of the Jordanian society with the surrounding environment, where the local materials available in the construction area were used, and the construction systems inherited from previous civilizations were used. The builders in Jordan relied on exchanging knowledge and transferring it from one generation to another, where they were able to formulate a construction style capable of responding to the requirement of architectural spaces, and each region of Jordan has its own way of building, as there are various geographical areas in Jordan, including agricultural, mountainous and desert areas. Then the research touched on a historical study of the architectural space and identifying the value of the architectural space in the Jordanian social life, which is related to the customs and traditions of a society influenced by the Arab Islamic civilization, and then the construction, the structural structure, its characteristics and the constituent elements of the building were defined in the vernacular l Jordanian architecture. From the structural point of view, and then to identify the structural materials used in the structural structure and the impact of the structural structure on the design from several aspects, leading to the interior space and the factors affecting it. The research aims to explain and clarify the interconnected design and construction solutions in the vernacular Jordanian architecture in a manner that respects the environmental context, taking into account the material cost of the building, where the financial situation of the home owner plays an important role in choosing the building material and construction method. Case studies from heritage buildings from several Jordanian regions will be analyzed to illustrate the idea of the research. Keywords: construction systems, architectural space, environmental context, Jordanian architecture

Authors: Demet Ozgur-Unluakin, Busenur Turkali, Ayse Karacaorenli
Abstract:
Maintenance strategies can be classified into two types, which are reactive and proactive, with respect to the time of the failure and maintenance. If the maintenance activity is done after a breakdown, it is called reactive maintenance. On the other hand, proactive maintenance, which is further divided as preventive and predictive, focuses on maintaining components before a failure occurs to prevent expensive halts. Recently, the number of interacting components in a system has increased rapidly and therefore, the structure of the systems have become more complex. This situation has made it difficult to provide the right maintenance decisions. Herewith, determining effective decisions has played a significant role. In multi-component systems, many methodologies and strategies can be applied when a component or a system has already broken down or when it is desired to identify and avoid proactively defects that could lead to future failure. This study focuses on the comparison of various maintenance strategies on a multi-component dynamic system. Components in the system are hidden, although there exists partial observability to the decision maker and they deteriorate in time. Several predefined policies under corrective, preventive and predictive maintenance strategies are considered to minimize the total maintenance cost in a planning horizon. The policies are simulated via Dynamic Bayesian Networks on a multi-component system with different policy parameters and cost scenarios, and their performances are evaluated. Results show that when the difference between the corrective and proactive maintenance cost is low, none of the proactive maintenance policies is significantly better than the corrective maintenance. However, when the difference is increased, at least one policy parameter for each proactive maintenance strategy gives significantly lower cost than the corrective maintenance. Keywords: decision making, dynamic Bayesian networks, maintenance, multi-component systems, reliability

Procedia PDF Downloads 1027657 An Intensional Conceptualization Model for Ontology-Based Semantic Integration
Authors: Fateh Adhnouss, Husam El-Asfour, Kenneth McIsaac, AbdulMutalib Wahaishi, Idris El-Feghia
Abstract:
Conceptualization is an essential component of semantic ontology-based approaches. There have been several approaches that rely on extensional structure and extensional reduction structure in order to construct conceptualization. In this paper, several limitations are highlighted relating to their applicability to the construction of conceptualizations in dynamic and open environments. These limitations arise from a number of strong assumptions that do not apply to such environments. An intensional structure is strongly argued to be a natural and adequate modeling approach. This paper presents a conceptualization structure based on property relations and propositions theory (PRP) to the model ontology that is suitable for open environments. The model extends the First-Order Logic (FOL) notation and defines the formal representation that enables interoperability between software systems and supports semantic integration for software systems in open, dynamic environments. Keywords: conceptualization, ontology, extensional structure, intensional structure

Abstract:
The dry-storage systems of nuclear power plants (NPPs) in Taiwan have become one of the major safety concerns. There are two steps considered in this study. The first step is the verification of the TRACE by using VSC-17 experimental data. The results of TRACE were similar to the VSC-17 data. It indicates that TRACE has the respectable accuracy in the simulation and analysis of the dry-storage systems. The next step is the application of TRACE in the dry-storage system of Kuosheng NPP (BWR/6). Kuosheng NPP is the second BWR NPP of Taiwan Power Company. In order to solve the storage of the spent fuels, Taiwan Power Company developed the new dry-storage system for Kuosheng NPP. In this step, the dry-storage system model of Kuosheng NPP was established by TRACE. Then, the steady state simulation of this model was performed and the results of TRACE were compared with the Kuosheng NPP data. Finally, this model was used to perform the safety analysis of Kuosheng NPP dry-storage system. Besides, FRAPTRAN was used tocalculate the transient performance of fuel rods. Keywords: BWR, TRACE, FRAPTRAN, dry-storage

Authors: Degale Desta, Cheng Jian
Abstract:
The most important factor in identifying a person is their face. Even identical twins have their own distinct faces. As a result, identification and face recognition are needed to tell one person from another. A face recognition system is a verification tool used to establish a person's identity using biometrics. Nowadays, face recognition is a common technique used in a variety of applications, including home security systems, criminal identification, and phone unlock systems. This system is more secure because it only requires a facial image instead of other dependencies like a key or card. Face detection and face identification are the two phases that typically make up a human recognition system. The idea behind designing and creating a face recognition system using deep learning with Azure ML Python's OpenCV is explained in this paper. Face recognition is a task that can be accomplished using deep learning, and given the accuracy of this method, it appears to be a suitable approach. To show how accurate the suggested face recognition system is, experimental results are given in 98.46% accuracy using Fast-RCNN Performance of algorithms under different training conditions. Keywords: deep learning, face recognition, identification, fast-RCNN
Procedia PDF Downloads 1407653 An Accurate Computer-Aided Diagnosis: CAD System for Diagnosis of Aortic Enlargement by Using Convolutional Neural Networks
Authors: Mahdi Bazarganigilani
Abstract:
Aortic enlargement, also known as an aortic aneurysm, can occur when the walls of the aorta become weak. This disease can become deadly if overlooked and undiagnosed. In this paper, a computer-aided diagnosis (CAD) system was introduced to accurately diagnose aortic enlargement from chest x-ray images. An enhanced convolutional neural network (CNN) was employed and then trained by transfer learning by using three different main areas from the original images. The areas included the left lung, heart, and right lung. The accuracy of the system was then evaluated on 1001 samples by using 4-fold cross-validation. A promising accuracy of 90% was achieved in terms of the F-measure indicator. The results showed using different areas from the original image in the training phase of CNN could increase the accuracy of predictions. This encouraged the author to evaluate this method on a larger dataset and even on different CAD systems for further enhancement of this methodology. Keywords: computer-aided diagnosis systems, aortic enlargement, chest X-ray, image processing, convolutional neural networks

Authors: Mona Fawzy Aldaghma
Abstract:
Helical piles are gaining popularity as a viable deep foundation alternative due to their quick installation and multipurpose use in compression and tension. These piles are commonly used as foundations for constructions such as solar panels, wind turbines and offshore platforms. These structures typically transfer various combinations of loads to their helical-pile foundations, including axial and lateral loads. Further research is needed to determine the effects of loading patterns that may act on helical piles as compounds of axial compression and lateral stresses. Multi helical piles are used to increase the efficiency of these piles. In this study, it investigate the behavior of laterally loaded helical piles with multiple helices when subjected to vertical loading conditions in both cohesive and cohesionless soils. Two models of intermediate shaft rigidity are studied with either two or three helices. Additionally, the vertical loading conditions were altered between successive and simultaneous loading. The cohesionless soil is sand with medium density and the cohesive soil is clay with medium cohesion. The study will carried out with numerical analysis using PLAXIS 3D and will be verified by an experimental tests. The numerical simulations reveal that helical piles exhibit different behavior in cohesive soil compared to cohesionless soil. Keywords: helical piles, multi-helix, numerical modeling, PLAXIS 3D, cohesive soil, cohesionless soil, experimental
Procedia PDF Downloads 397651 Water-Sensitive Landscaping in Desert-Located Egyptian Cities through Sheer Reductions of Turfgrass and Efficient Water Use
Authors: Sarah M. Asar, Nabeel M. Elhady
Abstract:
Egypt’s current per capita water share indicates that the country suffers and has been suffering from water poverty. The abundant utilization of turfgrass in Egypt’s new urban settlements, the reliance on freshwater for irrigation, and the inadequate plant selection increase the water demand in such settlements. Decreasing the surface area of turfgrass by using alternative landscape features such as mulching, using ornamental low-maintenance plants, increasing pathways, etc., could significantly decrease the water demand of urban landscapes. The use of Ammochloa palaestina, Cenchrus crientalis (Oriental Fountain Grass), and Cistus parviflorus (with water demands of approximately 0.005m³/m²/day) as alternatives for Cynodon dactylon (0.01m³/m²/day), which is the most commonly used grass species in Egypt’s landscape, could decrease an area’s water demand by approximately 40-50%. Moreover, creating hydro-zones of similar water demanding plants would enable irrigation facilitation rather than the commonly used uniformed irrigation. Such a practice could further reduce water consumption by 15-20%. These results are based on a case-study analysis of one of Egypt’s relatively new urban settlements, Al-Rehab. Such results emphasize the importance of utilizing native, drought-tolerant vegetation in the urban landscapes of Egypt to reduce irrigation demands. Furthermore, proper implementation, monitoring, and maintenance of automated irrigation systems could be an important factor in a space’s efficient water use. As most new urban settlements in Egypt adopt sprinkler and drip irrigation systems, the lack of maintenance leads to the manual operation of such systems, and, thereby, excessive irrigation occurs. Keywords: alternative landscape, native plants, efficient irrigation, low water demand
Procedia PDF Downloads 787650 Study on the Impact of Power Fluctuation, Hydrogen Utilization, and Fuel Cell Stack Orientation on the Performance Sensitivity of PEM Fuel Cell
Authors: Majid Ali, Xinfang Jin, Victor Eniola, Henning Hoene
Abstract:
The performance of proton exchange membrane (PEM) fuel cells is sensitive to several factors, including power fluctuations, hydrogen utilization, and the quality orientation of the fuel cell stack. In this study, we investigate the impact of these factors on the performance of a PEM fuel cell. We start by analyzing the power fluctuations that are typical in renewable energy systems and their effects on the 50 Watt fuel cell's performance. Next, we examine the hydrogen utilization rate (0-1000 mL/min) and its impact on the cell's efficiency and durability. Finally, we investigate the quality orientation (three different positions) of the fuel cell stack, which can significantly affect the cell's lifetime and overall performance. The basis of our analysis is the utilization of experimental results, which have been further validated by comparing them with simulations and manufacturer results. Our results indicate that power fluctuations can cause significant variations in the fuel cell's voltage and current, leading to a reduction in its performance. Moreover, we show that increasing the hydrogen utilization rate beyond a certain threshold can lead to a decrease in the fuel cell's efficiency. Finally, our analysis demonstrates that the orientation of the fuel cell stack can affect its performance and lifetime due to non-uniform distribution of reactants and products. In summary, our study highlights the importance of considering power fluctuations, hydrogen utilization, and quality orientation in designing and optimizing PEM fuel cell systems. The findings of this study can be useful for researchers and engineers working on the development of fuel cell systems for various applications, including transportation, stationary power generation, and portable devices. Keywords: fuel cell, proton exchange membrane, renewable energy, power fluctuation, experimental
Procedia PDF Downloads 1367649 Effect of Constant and Variable Temperature on the Morphology of TiO₂ Nanotubes Prepared by Two-Step Anodization Method
Authors: Tayyaba Ghani, Mazhar Mehmood, Mohammad Mujahid
Abstract:
TiO₂ nanotubes are receiving immense attraction in the field of dye-sensitized solar cells due to their well-defined nanostructures, efficient electron transport and large surface area as compared to other one dimensional structures. In the present work, we have investigated the influence of temperature on the morphology of anodically produced self-organized Titanium oxide nanotubes (TiNTs). TiNTs are synthesized by two-step anodization method in an ethylene glycol based electrolytes containing ammonium fluoride. Experiments are performed at constant anodization voltage for two hours. An investigation by the SEM images reveals that if the temperature is kept constant during the anodizing experiment, variation in the average tube diameter is significantly reduced. However, if the temperature is not controlled then due to the exothermic nature of reactions for the formation of TiNTs, the temperature of electrolyte keep on increasing. This variation in electrolyte bath temperature introduced strong variations in tube diameter (20 nm to 160 nm) along the length of tubes. Current profiles, recorded during the anodization experiment, predict the effect of constant and varying experimental temperatures as well. In both cases, XRD results show the complete anatase crystal structure of nanotube upon annealing at 450 °C. Present work highlights the importance of constant temperature during the anodization experiments in order to develop an ordered array of nanotubes with a uniform tube diameter. Keywords: anodization, ordering, temperature, TiO₂ nanotubes

Authors: Srijitra Khanpakdee, Teera Butburee, Jung-Ho Yun, Miaoqiang Lyu, Supphasin Thaweesak, Piangjai Peerakiatkhajohn
Abstract:
The synthesis of titanium dioxide (TiO₂) nanorods (NRs) on fluorine-doped tin oxide (FTO) glass via hydrothermal methods was investigated to determine the optimal reaction time for enhanced photocatalytic and optical performance. Reaction times of 4, 6, and 8 hours were studied. Characterization through SEM, UV-vis, XRD, FTIR, Raman spectroscopy and photoelectrochemical (PEC) techniques revealed significant differences in the properties of the TiO₂ NRs based on the reaction duration. XRD and Raman spectroscopy analysis confirmed the formation of the rutile phase of TiO₂. As photoanodes in PEC cells, TiO₂ NRs synthesized for 4 hours exhibited the best photocatalytic activity, with the highest photocurrent density and superior charge transport properties, attributed to their densely packed vertical structure. Longer reaction times resulted in less optimal morphological and photoelectrochemical characteristics. The bandgap of the TiO₂ NRs remained consistent around 3.06 eV, with only slight variations observed. This study highlights the critical role of reaction time in hydrothermal synthesis, identifying 4 hours as the optimal duration for producing TiO₂ NRs with superior photoelectrochemical performance. These findings provide valuable insights for optimizing TiO₂-based materials for solar energy conversion and renewable energy applications. Keywords: titanium dioxide, nanorods, hydrothermal, photocatalytic, photoelectrochemical

Abstract:
In Hassi-Messoud’s oil industry, the systems which are water based (WBM) are generally used for drilling in the first phase. For the rest of the well, the oil mud systems are employed (OBM). In the field of oil exploration, panoply of chemical products is employed in the drilling fluids formulation. These components of different natures and whose toxicity and biodegradability are of ill-defined parameters are; however, thrown into nature. In addition to the hydrocarbon (HC, such as diesel) which is a major constituent of oil based mud, we also can notice spills as well as a variety of other products and additives on the drilling sites. These wastes are usually stored in places called (crud wastes). These may cause major problems to the ecosystem. To treat these wastes, we have considered two methods which are: solidification/ stabilization (chemical) and thermal. So that we can evaluate the techniques of treatment, a series of analyses are performed on dozens of specimens of wastes before treatment. After that, and on the basis of our analyses of wastes, we opted for diagnostic treatments of pollution before and after solidification and stabilization. Finally, we have done some analyses before and after the thermal treatment to check the efficiency of the methods followed in the study. Keywords: wastes treatment, the oil pollution, the norms, wastes drilling

Authors: Yongqin Zhang, John Lett
Abstract:
Geospatial technologies have been increasingly used in agriculture for various applications and purposes in recent years. Unmanned aerial vehicles (drones) fit the needs of farmers in farming operations, from field spraying to grow cycles and crop health. In this research, we conducted a practical research project that used drone technology to design and map optimal locations and layouts of irrigation systems for agriculture farms. We flew a DJI Mavic 2 Pro drone to acquire aerial remote sensing images over two agriculture fields in Forest, Mississippi, in 2022. Flight plans were first designed to capture multiple high-resolution images via a 20-megapixel RGB camera mounted on the drone over the agriculture fields. The Drone Deploy web application was then utilized to develop flight plans and subsequent image processing and measurements. The images were orthorectified and processed to estimate the area of the area and measure the locations of the water line and sprinkle heads. Field measurements were conducted to measure the ground targets and validate the aerial measurements. Geospatial analysis and photogrammetric measurements were performed for the study area to determine optimal layout and quantitative estimates for irrigation systems. We created maps and tabular estimates to demonstrate the locations, spacing, amount, and layout of sprinkler heads and water lines to cover the agricultural fields. This research project provides scientific guidance to Mississippi farmers for a precision agricultural irrigation practice. Keywords: drone images, agriculture, irrigation, geospatial analysis, photogrammetric measurements
Procedia PDF Downloads 787642 Forming-Free Resistive Switching Effect in ZnₓTiᵧHfzOᵢ Nanocomposite Thin Films for Neuromorphic Systems Manufacturing
Authors: Vladimir Smirnov, Roman Tominov, Vadim Avilov, Oleg Ageev
Abstract:
The creation of a new generation micro- and nanoelectronics elements opens up unlimited possibilities for electronic devices parameters improving, as well as developing neuromorphic computing systems. Interest in the latter is growing up every year, which is explained by the need to solve problems related to the unstructured classification of data, the construction of self-adaptive systems, and pattern recognition. However, for its technical implementation, it is necessary to fulfill a number of conditions for the basic parameters of electronic memory, such as the presence of non-volatility, the presence of multi-bitness, high integration density, and low power consumption. Several types of memory are presented in the electronics industry (MRAM, FeRAM, PRAM, ReRAM), among which non-volatile resistive memory (ReRAM) is especially distinguished due to the presence of multi-bit property, which is necessary for neuromorphic systems manufacturing. ReRAM is based on the effect of resistive switching – a change in the resistance of the oxide film between low-resistance state (LRS) and high-resistance state (HRS) under an applied electric field. One of the methods for the technical implementation of neuromorphic systems is cross-bar structures, which are ReRAM cells, interconnected by cross data buses. Such a structure imitates the architecture of the biological brain, which contains a low power computing elements - neurons, connected by special channels - synapses. The choice of the ReRAM oxide film material is an important task that determines the characteristics of the future neuromorphic system. An analysis of literature showed that many metal oxides (TiO2, ZnO, NiO, ZrO2, HfO2) have a resistive switching effect. It is worth noting that the manufacture of nanocomposites based on these materials allows highlighting the advantages and hiding the disadvantages of each material. Therefore, as a basis for the neuromorphic structures manufacturing, it was decided to use ZnₓTiᵧHfzOᵢ nanocomposite. It is also worth noting that the ZnₓTiᵧHfzOᵢ nanocomposite does not need an electroforming, which degrades the parameters of the formed ReRAM elements. Currently, this material is not well studied, therefore, the study of the effect of resistive switching in forming-free ZnₓTiᵧHfzOᵢ nanocomposite is an important task and the goal of this work. Forming-free nanocomposite ZnₓTiᵧHfzOᵢ thin film was grown by pulsed laser deposition (Pioneer 180, Neocera Co., USA) on the SiO2/TiN (40 nm) substrate. Electrical measurements were carried out using a semiconductor characterization system (Keithley 4200-SCS, USA) with W probes. During measurements, TiN film was grounded. The analysis of the obtained current-voltage characteristics showed a resistive switching from HRS to LRS resistance states at +1.87±0.12 V, and from LRS to HRS at -2.71±0.28 V. Endurance test shown that HRS was 283.21±32.12 kΩ, LRS was 1.32±0.21 kΩ during 100 measurements. It was shown that HRS/LRS ratio was about 214.55 at reading voltage of 0.6 V. The results can be useful for forming-free nanocomposite ZnₓTiᵧHfzOᵢ films in neuromorphic systems manufacturing. This work was supported by RFBR, according to the research project № 19-29-03041 mk. The results were obtained using the equipment of the Research and Education Center «Nanotechnologies» of Southern Federal University. Keywords: nanotechnology, nanocomposites, neuromorphic systems, RRAM, pulsed laser deposition, resistive switching effect

Iranian city has special relevance problem of assessing the conformity of their planning and development of the concept of sustainable development. Based on theoretical sources, the author concludes that, despite the existence of common indicators for sustainable development of settlements, specialized evaluation criteria city structure planning has not been developed. He is trying to fill this gap and put forward a system of indicators characterizing the level of development of the architectural-planning structure of the city. The proposed system of indicators is designed based on technical and economic urban standard indicators from different countries. Alternative designing systems and requirements of modern rating systems like LEED-ND comprise a criterion for evaluation of urban structures in accordance with principles of "Green" building and New Urbanism. Urban development trends are close in spirit of sustainable development and developed under its influence. The study allowed concluding that a system of indicators to identify the relevant architectural-planning structure of the city, requirements of sustainable development, should be adapted to the conditions of each country, particularly in Iran. The article attempts typology proposed indicators, which are presented in tabular form and are divided into two types: planning and spatial. This article discusses the known indicators of sustainable development and proposed specific system of indicators characterizing the level of development of architectural-planning structure of the city. This article examines indicators for evaluating level of city' planning structure development. The proposed system of indicators is derived from the urban planning standards and rating systems such as LEED-ND, BREEAM Community and CASBEE-UD.Keywords: architectural-planning structure of cities, urban planning indicators, urban space indicators, urban development

Authors: Sanaz Seraj, Shohre Rouhani
Abstract:
Recently, graphene has gained much attention because of its unique optical, mechanical, electrical, and thermal properties. Graphene has been used as a key material in the technological applications in various areas such as sensors, drug delivery, super capacitors, transparent conductor, and solar cell. It has a superior quenching efficiency for various fluorophores. Based on these unique properties, the optical sensors with graphene materials as the energy acceptors have demonstrated great success in recent years. During quenching, the emission of a fluorophore is perturbed by a quencher which can be a substrate or biomolecule, and due to this phenomenon, fluorophore-quencher has been used for selective detection of target molecules. Among fluorescence dyes, 1,8-naphthalimide is well known for its typical intramolecular charge transfer (ICT) and photo-induced charge transfer (PET) fluorophore, strong absorption and emission in the visible region, high photo stability, and large Stokes shift. Derivatives of 1,8-naphthalimides have found applications in some areas, especially fluorescence sensors. Herein, the fluorescence quenching of graphene oxide has been carried out on a naphthalimide dye as a fluorescent probe model. The quenching ability of graphene oxide on naphthalimide dye was studied by UV-VIS and fluorescence spectroscopy. This study showed that graphene is an efficient quencher for fluorescent dyes. Therefore, it can be used as a suitable candidate sensing platform. To the best of our knowledge, studies on the quenching and absorption of naphthalimide dyes by graphene oxide are rare. Keywords: fluorescence, graphene oxide, naphthalimide dye, quenching

Abstract:
A huge revolution has emerged in genome engineering by the discovery of CRISPR (clustered regularly interspaced palindromic repeats) and CRISPR-associated system genes (Cas) in bacteria. The function of type II Streptococcus pyogenes (Sp) CRISPR/Cas9 system has been confirmed in various species. Other S. thermophilus (St) CRISPR-Cas systems, CRISPR1-Cas and CRISPR3-Cas, have been also reported for preventing phage infection. The CRISPR1-Cas system interferes by cleaving foreign dsDNA entering the cell in a length-specific and orientation-dependant manner. The S. thermophilus CRISPR3-Cas system also acts by cleaving phage dsDNA genomes at the same specific position inside the targeted protospacer as observed in the CRISPR1-Cas system. It is worth mentioning, for the effective DNA cleavage activity, RNA-guided Cas9 orthologs require their own specific PAM (protospacer adjacent motif) sequences. Activity levels are based on the sequence of the protospacer and specific combinations of favorable PAM bases. Therefore, based on the specific length and sequence of PAM followed by a constant length of target site for the three orthogonals of Cas9 protein, a well-organized procedure will be required for high-throughput and accurate mining of possible target sites in a large genomic dataset. Consequently, we created a reliable procedure to explore potential gRNA sequences for type I (Streptococcus thermophiles), II (Streptococcus pyogenes), and III (Streptococcus thermophiles) CRISPR/Cas systems. To mine CRISPR target sites, four different searching modes of sgRNA binding to target DNA strand were applied. These searching modes are as follows: i) coding strand searching, ii) anti-coding strand searching, iii) both strand searching, and iv) paired-gRNA searching. The output of such procedure highlights the power of comparative genome mining for different CRISPR/Cas systems. This could yield a repertoire of Cas9 variants with expanded capabilities of gRNA design, and will pave the way for further advance genome and epigenome engineering. Keywords: CRISPR/Cas systems, gRNA mining, Streptococcus pyogenes, Streptococcus thermophiles
