Abstract:
Background: Recessive Dystrophic Epidermolysis Bullosa (RDEB) is a genetic skin condition characterized by skin tearing and unremitting blistering upon minimal trauma. Repeated blistering, fibrosis, and scarring lead to aggressive squamous cell carcinoma later in life. RDEB is caused by mutations in the COL7A1 gene encoding collagen type VII (C7), the major component of anchoring fibrils mediating epidermis-dermis adherence. Nonsense mutations in the COL7A1 gene of a subset of RDEB patients leads to premature termination codons (PTC). Similarly, most Junctional Epidermolysis Bullosa (JEB) cases are caused by nonsense mutations in the LAMB3 gene encoding the β3 subunit of laminin 332. Currently, there is an unmet need for the treatment of RDEB and JEB. Zikani Therapeutics has discovered an array of macrocyclic compounds with ring structures similar to macrolide antibiotics that can facilitate readthrough activity of nonsense mutations in the COL7A1 and LAMB3 genes by acting as Ribosome Modulating Agents (RMAs). The medicinal chemistry synthetic advancements of these macrocyclic compounds have allowed targeting the human ribosome while preserving the structural elements responsible for the safety and pharmacokinetic profile of clinically used macrolide antibiotics. Methods: C7 expression was used as a measure of readthrough activity by immunoblot assays in two primary human fibroblasts from RDEB patients (R578X/R578X and R163X/R1683X-COL7A1). Similarly, immunoblot assays in C325X/c.629-12T > A-LAMB3 keratinocytes were used to measure readthrough activity for JEB. The relative readthrough activity of each compound was measured relative to Gentamicin. An imaging-based fibroblast migration assay was used as an assessment of C7 functionality in RDEB-fibroblasts over 16-20 hrs. The incubation period for the above experiments was 48 hrs for RDEB fibroblasts and 72 hours for JEB keratinocytes. Results: 9 RMAs demonstrated increased protein expression in both patient RDEB fibroblasts. The highest readthrough activity at tested concentrations without cytotoxicities increased protein expression up to 179% of Gentamicin (400 µg/ml), with favored readthrough activity in R163X/R1683X-COL7A1 fibroblasts. Concurrent with protein expression, fibroblast hypermotility phenotype observed in RDEB was rescued by reducing motility by ~35% to WT levels (the same level as 690 µM Gentamicin treated cells). Laminin β3 expression was also shown to be increased by 6 RMAs in keratinocytes to 33-83% of (400 µg/ml) Gentamicin. Conclusions: To date, 9 RMAs have been identified that enhance the expression of functional C7 in a mutation-dependent manner in two different RDEB patient fibroblast backgrounds (R578X/R578X and R163X/R1683X-COL7A1). A further 6 RMAs have been identified that enhance the readthrough of C325X-LAMB3 in JEB patient keratinocytes. Based on the clinical trial conducted by us with topical gentamycin in 2017, Zikani’s RMAs achieve clinically significant levels of read-through for the treatment of recessive dystrophic and Junctional Epidermolysis Bullosa. Keywords: epidermolysis bullosa, nonsense mutation, readthrough, ribosome modulation

Procedia PDF Downloads 47412 Effective Practical Proceedings in Breaking the Respiratory Infections Transmission Chain in the Community with the Emphasis on SARS-COV-2 Control
Authors: Fatemeh Aghamohammadzadeh, Mahdi Asghari Ozma
Abstract:
SARS-CoV-2 was transmitted from animals to humans in China and through air transport to almost all world countries, including Iran, creating the first pandemic of the 21st century. The virus was spread through droplets from sneezing, coughing, loud talking, and exhalation of sick and asymptomatic people, even during incubation. It was transmitted from human to human directly by inhalation of viruses in droplets or indirectly through contact with infected surfaces, resulting in the death of a significant number of patients, especially the elderly and those with underlying diseases. The virus is more likely to be transmitted in places with high population densities. The chain of transmission of infection can be broken by observing the following: risk perception, reduced travel, complete quarantine in a particular area, home quarantine, social distancing, use of personal protective equipment (PPE), prevention of gatherings, cleaning and disinfection of public utilities and busy places, identifying, isolating and treating infected people, tracking calls, continuing health education, following health principles by people, especially in poor areas, and washing their hands frequently with soap and water or disinfecting them with 70% ethanol. Keywords: COVID-19, transmission, population density, home quarantine, social distancing

Authors: Soheil Mohtaram, Wu Weidong, Yashar Aryanfar
Abstract:
This research investigates the points with heat recovery potential in a triple-pressure cogeneration combined cycle power plant and determines the amount of waste heat that can be recovered. A modified cycle arrangement is then adopted for accessing thermal potentials. Modeling the energy system is followed by thermodynamic and energetic evaluation, and then the price of the manufactured products is also determined using the Total Revenue Requirement (TRR) method and term economic analysis. The results of optimization are then presented in a Pareto chart diagram by implementing a new model with dual objective functions, which include power cost and produce heat. This model can be utilized to identify the optimal operating point for such power plants based on electricity and heat prices in different regions. Keywords: heat loss, recycling, unused energy, efficient production, optimization, triple-pressure cogeneration

Abstract:
Since sandwich structures are used in many areas ranging from ships, trains, automobiles, aircrafts, bridge and building, connecting sandwich structures is necessary almost in all industries. So application of metallic joints between sandwich panels is increasing. Various joining methods are available such as mechanically fastened joints (riveting or bolting) or adhesively bonded joints and choosing one of them depends on the application. In this research, sandwich specimens were fabricated with two different types of metallic connections with dissimilar geometries. These specimens included beams and plates and were manufactured using glass-epoxy skins and aluminum honeycomb core. After construction of the specimens, bending and low velocity impact tests were executed on them and the behaviors of specimens were discussed. Numerical models were developed using LS-DYNA software and validated with test results. Finally, parametric studies were performed on the thicknesses and lengths of two connections by employing the numerical models. Keywords: connection, honeycomb, low velocity impact, sandwich panel, static test
Procedia PDF Downloads 569 Self-focused Language and the Reversive Impact of Depression in Negative Mood
Authors: Soheil Behdarvandirad
Abstract:
The relationship between depression and self-focused language has been well documented. The more depressed a person is, the more "I"s, "me"s, and "my"s they will use. The present study attempted to factor in the impact of mood and examine whether negative mood has self-focused impacts similar to those of depression. For this purpose, 160 Iranian native speakers of Farsi were divided into three experimental groups of negative, neutral, and positive groups. After completing the BDI-II inventory and depression measurement, they were presented with pretested mood stimuli (3 separate videos to induce the target moods). Finally, they were asked to write between 10 to 20 minutes about a topic that asked them to freely write about their state of life, how you feel about it and the reasons that had shaped their current life circumstances. While the significant correlation between depression and I-talk was observed, negative mood led to more we-talk in general and seemed to even push the participants away from self-rumination. It seems that it is an emotion-regulatory strategy that participants subconsciously adopt to distract themselves from the disturbing mood. However, negative mood intensified the self-focused language among depressed participants. Such results can be further studied by examining brain areas that are more involved in self-perception and particularly in precuneus. Keywords: self-focused language, depression, mood, precuneus

Low dose, 3. High dose and 4. Treatment, with 6 rats in each group. Full-thickness incisions with a diameter of 2 cm were made on the back of each rat. Rats were topically treated two times a day for 15 days. Group 1-4 were treated with sterile distilled water, 5% and 10% of extract and intrasite gel, respectively. Masson's trichrome and hematoxylin staining techniques are employed for histological analysis revealed strong wound healing potential closer to that of conventional drug intrasite gel. HECP significantly decreased wound area and an increase in hydroxyproline, cellular proliferation, the number of blood vessels and the level of collagen synthesis was observed. Thus, it could be concluded that HECP possesses strong wound healing potential. Keywords: Curcuma purpurascens, wound healing, histopathology, hematoxylin staining
Procedia PDF Downloads 4387 Numerical Simulation of Different Enhanced Oil Recovery (EOR) Scenarios on a Volatile Oil Reservoir
Authors: Soheil Tavakolpour
Abstract:
Enhance Oil Recovery (EOR) can be considered as an undeniable action in reservoirs life period. Different kind of EOR methods are available, but suitable EOR method depends on reservoir properties, like rock and fluid properties. In this paper, we nominated fifth SPE’s Comparative Solution Projects (CSP) for testing different scenarios. We used seven EOR scenarios for this reservoir and we simulated it for 10 years after 2 years production without any injection. The first scenario is waterflooding for whole of the 10 years period. The second scenario is gas injection for ten years. The third scenario is Water-Alternation-Gas (WAG). In the next scenario, water injected for 4 years before starting WAG injection for the next 6 years. In the fifth scenario, water injected after 6 years WAG injection for 4 years. For sixth and last scenarios, all the things are similar to fourth and fifth scenarios, but gas injected instead of water. Results show that fourth scenario was the most efficient method for 10 years EOR, but it resulted very high water production. Fifth scenario was efficient too, with little water production in comparison to the fourth scenario. Gas injection was not economically attractive. In addition to high gas production, it produced less oil in comparison to other scenarios. Keywords: WAG, SPE’s comparative solution projects, numerical simulation, EOR scenarios
Procedia PDF Downloads 4346 Annona muricata Leaves Induced Mitochondrial-Mediated Apoptosis in A549 Cells
Authors: Soheil Zorofchian Moghadamtousi, Habsah Abdul Kadir, Mohammadjavad Paydar, Elham Rouhollahi, Hamed Karimian
Abstract:
The present study was designed to evaluate the molecular mechanisms of Annona muricata leaves ethyl acetate extract (AMEAE) against lung cancer A549 cells. Cell viability analysis revealed the selective cytotoxic effect of AMEAE towards A549 cells. Treatment of A549 cells with AMEAE significantly elevated the reactive oxygen species formation, followed by attenuation of mitochondrial membrane potential via upregulation of Bax and downregulation of Bcl-2, accompanied by cytochrome c release to the cytosol. The released cytochrome c triggered the activation of caspase-9 followed by caspase-3. In addition, AMEAE-induced apoptosis was accompanied by cell cycle arrest at G1 phase. Our data showed for the first time that AMEAE inhibited the proliferation of A549 cells, leading to cell cycle arrest and programmed cell death through activation of the mitochondrial-mediated signaling pathway. Keywords: Annona muricata, lung cancer, apoptosis, mitochondria
Procedia PDF Downloads 3235 A Phenomenological-Hermeneutic Account of Design Thinking by Way of an Exposition of Four Species of Negatite: 'Not Being', 'Non-Being', 'Absence', 'Non-Existence'
Authors: Soheil Ashrafi
Abstract:
In this paper, it is attempted to chart and exposit terra incognito of the transcendental intuition of ‘non-being’, a peculiar species of négatité and a form of consciousness which underpins the phenomenal capacity for design thinking, and which serves as the ground of the ‘designing being-relation to the world’. The paper’s contention is that the transcendental intuition of the non-being indwells the agent’s being-relation to the world as a continual tension in that neither does the agent relinquish its ontological leverage and submit altogether to the world’s curbs and dictates, nor is it able to subdue satisfactorily or settle into the world once and for all. By way of phenomenological-hermeneutic analysis, it is endeavoured to argue that design thinking occurs by virtue of a phenomenal transition between the a priori ‘not-being’, the basis of ‘that-which-is’, and the transcendental intuition of non-being through which that-which-is-not-yet announces itself. Along with this, the other two species of négatité as ‘absence’ and ‘non-existence’ are clarified and contrasted with not-being and non-being, which have widely been used in the literature interchangeably as identical terms. In conclusion, it is argued that not only has design thinking in its unadulterated, originary mode historically preceded scientific thinking, but it also has served as the foundation of its emergence. In short, scientific thinking is a derivative, reformed application of design thinking; it indeed supervenes upon it.Keywords: design thinking, designing being-relation to the world, négatité, not-being, non-being

Authors: Marzieh Yassin, Masoud Rashed, Soheil Mansour Sohani, Reza Salehi
Abstract:
Background: Hip arthroplasty is one of the surgical methods to improve symptoms in patients with hip osteoarthritis. The use of electroacupuncture and TENS reduces pain, increases range of motion and improves performance. Methods: In this clinical trial study, 30 patients after hip arthroplasty were randomly divided into two groups: electroacupuncture (n=16) with exercise therapy and TENS with exercise therapy (n=14). Severity of pain, quality of life, range of motion, edema and function were evaluated in two groups before and after the interventions. Interventions of 10 sessions (three sessions per week) were conducted for two groups. The significance level in all tests was below 0.05. Results: The results showed that both groups improved all of the symptoms after the intervention (p≤0.05), although there was no statistically significant difference between the two groups in terms of effectiveness (p≥0.05). Conclusion: The results showed that both methods improve symptoms in patients after surgery. According to this study, electroacupuncture is suggested as a new method effective for the treatment of people with post-Total Hip Arthroplasty. Keywords: electroacupuncture, physical performance, total hip arthroplasty, TENS
Procedia PDF Downloads 823 Indoor Environment Quality and Occupant Resilience Toward Climate Change: A Case Study from Gold Coast, Australia
Authors: Soheil Roumi, Fan Zhang, Rodney Stewart
Abstract:
Indoor environmental quality (IEQ) indexes represented the suitability of a place to study, work, and live. Many indexes have been introduced based on the physical measurement or occupant surveys in commercial buildings. The earlier studies did not elaborate on the relationship between energy consumption and IEQ in office buildings. Such a relationship can provide a comprehensive overview of the building's performance. Also, it would find the potential of already constructed buildings under the upcoming climate change. A commercial building in southeast Queensland, Australia, was evaluated in this study. Physical measurements of IEQ and Energy areconducted, and their relationship will be determined using statistical analysis. The case study building is modelled in TRNSys software, and it will be validatedusingthe actual building's BMS data. Then, the modelled buildingwill be simulated by predicted weather data developed by the commonwealth scientific and industrial research organisation of Australia to investigate the occupant resilience and energy consumption. Finally, recommendations will be presented to consume less energy while providinga proper indoor environment for office occupants. Keywords: IEQ, office buildings, thermal comfort, occupant resilience
Procedia PDF Downloads 1122 Creep Behaviour of Asphalt Modified by Waste Polystyrene and Its Hybrids with Crumb Rubber and Low-Density Polyethylene
Authors: Soheil Heydari, Ailar Hajimohammadi, Nasser Khalili
Abstract:
Polystyrene, being made from a monomer called styrene, is a rigid and easy-to mould polymer that is widely used for many applications, from foam packaging to disposable containers. Considering that the degradation of waste polystyrene takes up to 500 years, there is an urgent need for a sustainable application for waste polystyrene. This study evaluates the application of waste polystyrene as an asphalt modifier. The inclusion of waste plastics in asphalt is either practised by the dry process or the wet process. In the dry process, plastics are added straight into the asphalt mixture and in the wet process, they are mixed and digested into bitumen. In this article, polystyrene was used as an asphalt modifier in a dry process. However, the mixing process is precisely designed to make sure that the polymer is melted and modified in the binder. It was expected that, due to the rigidity of polystyrene, it will have positive effects on the permanent deformation of the asphalt mixture. Therefore, different mixtures were manufactured with different contents of polystyrene and Marshall specimens were manufactured, and dynamic creep tests were conducted to evaluate the permanent deformation of the modification. This is a commonly repeated loading test conducted at different stress levels and temperatures. Loading cycles are applied to the AC specimen until failure occurs; with the amount of deformation constantly recorded the cumulative, permanent strain is determined and reported as a function of the number of cycles. Also, to our best knowledge, hybrid mixes of polystyrene with crumb rubber and low-density polyethylene were made and compared with a polystyrene-modified mixture. The test results of this study showed that the hybrid mix of polystyrene and low-density polyethylene has the highest resistance against permanent deformation. However, the polystyrene-modified mixture outperformed the hybrid mix of polystyrene and crumb rubber, and both demonstrated way lower permanent deformation than the unmodified specimen. Keywords: permanent deformation, waste plastics, polystyrene, hybrid plastics, hybrid mix, hybrid modification, dry process
Procedia PDF Downloads 1051 The Effects of the Waste Plastic Modification of the Asphalt Mixture on the Permanent Deformation
Authors: Soheil Heydari, Ailar Hajimohammadi, Nasser Khalili
Abstract:
The application of plastic waste for asphalt modification is a sustainable strategy to deal with the enormous plastic waste generated each year and enhance the properties of asphalt. The modification is either practiced by the dry process or the wet process. In the dry process, plastics are added straight into the asphalt mixture, and in the wet process, they are mixed and digested into bitumen. In this article, the effects of plastic inclusion in asphalt mixture, through the dry process, on the permanent deformation of the asphalt are investigated. The main waste plastics that are usually used in asphalt modification are taken into account, which is linear, low-density polyethylene, low-density polyethylene, high-density polyethylene, and polypropylene. Also, to simulate a plastic waste stream, different grades of each virgin plastic are mixed and used. For instance, four different grades of polypropylene are mixed and used as representative of polypropylene. A precisely designed mixing condition is considered to dry-mix the plastics into the mixture such that the polymer was melted and modified by the later introduced binder. In this mixing process, plastics are first added to the hot aggregates and mixed three times in different time intervals, then bitumen is introduced, and the whole mixture is mixed three times in fifteen minutes intervals. Marshall specimens were manufactured, and dynamic creep tests were conducted to evaluate the effects of modification on the permanent deformation of the asphalt mixture. Dynamic creep is a common repeated loading test conducted at different stress levels and temperatures. Loading cycles are applied to the AC specimen until failure occurs; with the amount of deformation constantly recorded, the cumulative, permanent strain is determined and reported as a function of the number of cycles. The results of this study showed that the dry inclusion of the waste plastics is very effective in enhancing the resistance against permanent deformation of the mixture. However, the mixing process must be precisely engineered to melt the plastics, and a homogenous mixture is achieved. Keywords: permanent deformation, waste plastics, low-density polyethene, high-density polyethene, polypropylene, linear low-density polyethene, dry process
