2118 Exploring Fluoroquinolone-Resistance Dynamics Using a Distinct in Vitro Fermentation Chicken Caeca Model
Authors: Bello Gonzalez T. D. J., Setten Van M., Essen Van A., Brouwer M., Veldman K. T.
Abstract:
Resistance to fluoroquinolones (FQ) has evolved increasingly over the years, posing a significant challenge for the treatment of human infections, particularly gastrointestinal tract infections caused by zoonotic bacteria transmitted through the food chain and environment. In broiler chickens, a relatively high proportion of FQ resistance has been observed in Escherichia coli indicator, Salmonella and Campylobacter isolates. We hypothesize that flumequine (Flu), used as a secondary choice for the treatment of poultry infections, could potentially be associated with a high proportion of FQ resistance. To evaluate this hypothesis, we used an in vitro fermentation chicken caeca model. Two continuous single-stage fermenters were used to simulate in real time the physiological conditions of the chicken caeca microbial content (temperature, pH, caecal content mixing, and anoxic environment). A pool of chicken caecal content containing FQ-resistant E. coli obtained from chickens at slaughter age was used as inoculum along with a spiked FQ-susceptible Campylobacter jejuni strain isolated from broilers. Flu was added to one of the fermenters (Flu-fermenter) every 24 hours for two days to evaluate the selection and maintenance of FQ resistance over time, while the other served as a control (C-Fermenter). The experiment duration was 5 days. Samples were collected at three different time points: before, during and after Flu administration. Serial dilutions were plated on Butzler culture media with and without Flu (8mg/L) and enrofloxacin (4mg/L) and on MacConkey culture media with and without Flu (4mg/L) and enrofloxacin (1mg/L) to determine the proportion of resistant strains over time. Positive cultures were identified by mass spectrometry and matrix-assisted laser desorption/ionization (MALDI). A subset of the obtained isolates were used for Whole Genome Sequencing analysis. Over time, E. coli exhibited positive growth in both fermenters, while C. jejuni growth was detected up to day 3. The proportion of Flu-resistant E. coli strains recovered remained consistent over time after antibiotic selective pressure, while in the C-fermenter, a decrease was observed at day 5; a similar pattern was observed in the enrofloxacin-resistant E. coli strains. This suggests that Flu might play a role in the selection and persistence of enrofloxacin resistance, compared to C-fermenter, where enrofloxacin-resistant E. coli strains appear at a later time. Furthermore, positive growth was detected from both fermenters only on Butzler plates without antibiotics. A subset of C. jejuni strains from the Flu-fermenter revealed that those strains were susceptible to ciprofloxacin (MIC < 0.12 μg/mL). A selection of E. coli strains from both fermenters revealed the presence of plasmid-mediated quinolone resistance (PMQR) (qnr-B19) in only one strain from the C-fermenter belonging to sequence type (ST) 48, and in all from Flu-fermenter belonged to ST189. Our results showed that Flu selective impact on PMQR-positive E. coli strains, while no effect was observed in C. jejuni. Maintenance of Flu-resistance was correlated with antibiotic selective pressure. Further studies into antibiotic resistance gene transfer among commensal and zoonotic bacteria in the chicken caeca content may help to elucidate the resistance spread mechanisms. Keywords: fluoroquinolone-resistance, escherichia coli, campylobacter jejuni, in vitro model

Authors: Laura Alvarez-Alonso, Francisco Garcia-Carro, Jorge Loredo
Abstract:
Population growth and industrial development imply an increase in the energy demands and the problems caused by emissions of greenhouse effect gases, which has inspired the search for clean sources of energy. Hydrogen (H₂) is expected to play a key role in the world’s energy future by replacing fossil fuels. The properties of H₂ make it a green fuel that does not generate pollutants and supplies sufficient energy for power generation, transportation, and other applications. Supercritical Water Gasification (SCWG) represents an attractive alternative for the recovery of energy from wastes. SCWG allows conversion of a wide range of raw materials into a fuel gas with a high content of hydrogen and light hydrocarbons through their treatment at conditions higher than those that define the critical point of water (temperature of 374°C and pressure of 221 bar). Methane used as a transport fuel is another important gasification product. The number of different uses of gas and energy forms that can be produced depending on the kind of material gasified and type of technology used to process it, shows the flexibility of SCWG. This feature allows it to be integrated with several industrial processes, as well as power generation systems or waste-to-energy production systems. The final aim of this work is to study which conditions and equipment are the most efficient and advantageous to explore the possibilities to obtain streams rich in H₂ from oily wastes, which represent a major problem both for the environment and human health throughout the world. In this paper, the relative complexity of technology needed for feasible gasification process cycles is discussed with particular reference to the different feedstocks that can be used as raw material, different reactors, and energy recovery systems. For this purpose, a review of the current status of SCWG technologies has been carried out, by means of different classifications based on key features as the feed treated or the type of reactor and other apparatus. This analysis allows to improve the technology efficiency through the study of model calculations and its comparison with experimental data, the establishment of kinetics for chemical reactions, the analysis of how the main reaction parameters affect the yield and composition of products, or the determination of the most common problems and risks that can occur. The results of this work show that SCWG is a promising method for the production of both hydrogen and methane. The most significant choices of design are the reactor type and process cycle, which can be conveniently adopted according to waste characteristics. Regarding the future of the technology, the design of SCWG plants is still to be optimized to include energy recovery systems in order to reduce costs of equipment and operation derived from the high temperature and pressure conditions that are necessary to convert water to the SC state, as well as to find solutions to remove corrosion and clogging of components of the reactor. Keywords: hydrogen production, organic wastes, supercritical water gasification, system integration, waste-to-energy
Procedia PDF Downloads 1472116 Operation Parameters of Vacuum Cleaned Filters
Authors: Wilhelm Hoeflinger, Thomas Laminger, Johannes Wolfslehner
Abstract:
For vacuum cleaned dust filters, used e. g. in textile industry, there exist no calculation methods to determine design parameters (e. g. traverse speed of the nozzle, filter area...). In this work a method to calculate the optimum traverse speed of the nozzle of an industrial-size flat dust filter at a given mean pressure drop and filter face velocity was elaborated. Well-known equations for the design of a cleanable multi-chamber bag-house-filter were modified in order to take into account a continuously regeneration of a dust filter by a nozzle. Thereby, the specific filter medium resistance and the specific cake resistance values are needed which can be derived from filter tests under constant operation conditions. A lab-scale filter test rig was used to derive the specific filter media resistance value and the specific cake resistance value for vacuum cleaned filter operation. Three different filter media were tested and the determined parameters were compared to each other. Keywords: design of dust filter, dust removing, filter regeneration, operation parameters

Procedia PDF Downloads 2712113 Improving Lubrication Efficiency at High Sliding Speeds by Plasma Surface Texturing
Authors: Wei Zha, Jingzeng Zhang, Chen Zhao, Ran Cai, Xueyuan Nie
Abstract:
Cathodic plasma electrolysis (CPE) is used to create surface textures on cast iron samples for improving the tribological properties. Micro craters with confined size distribution were successfully formed by CPE process. These craters can generate extra hydrodynamic pressure that separates two sliding surfaces, increase the oil film thickness and accelerate the transition from boundary to mixed lubrication. It was found that the optimal crater size was 1.7 μm, at which the maximum lubrication efficiency was achieved. The Taguchi method was used to optimize the process parameters (voltage and roughness) for CPE surface texturing. The orthogonal array and the signal-to-noise ratio were employed to study the effect of each process parameter on the coefficient of friction. The results showed that with higher voltage and lower roughness, the lower friction coefficient can be obtained, and thus the lubrication can be more efficiently used for friction reduction. Keywords: cathodic plasma electrolysis, friction, lubrication, plasma surface texturing

Authors: Usman D. Hamza, Noor S. Nasri, Mohammed Jibril, Husna M. Zain
Abstract:
Activated carbons (M4P0, M4P2, and M5P2) used in this research were produced from palm shell and polyetherether ketone (PEEK) via carbonization, impregnation, and microwave activation. The adsorption/desorption process was carried out using static volumetric adsorption. Regeneration is important in the overall economy of the process and waste minimization. This work focuses on the thermal regeneration of the CO2 exhausted microwave activated carbons. The regeneration strategy adopted was thermal with nitrogen purge desorption with N2 feed flow rate of 20 ml/min for 1 h at atmospheric pressure followed by drying at 1500C. Seven successive adsorption/regeneration processes were carried out on the material. It was found that after seven adsorption regeneration cycles; the regeneration efficiency (RE) for CO2 activated carbon from palm shell only (M4P0) was more than 90% while that of hybrid palm shell-PEEK (M4P2, M5P2) was above 95%. The cyclic adsorption and regeneration shows the stability of the adsorbent materials. Keywords: activated carbon, palm shell-PEEK, regeneration, thermal
Procedia PDF Downloads 4882111 Study of Biomechanical Model for Smart Sensor Based Prosthetic Socket Design System
Authors: Wei Xu, Abdo S. Haidar, Jianxin Gao
Abstract:
Prosthetic socket is a component that connects the residual limb of an amputee with an artificial prosthesis. It is widely recognized as the most critical component that determines the comfort of a patient when wearing the prosthesis in his/her daily activities. Through the socket, the body weight and its associated dynamic load are distributed and transmitted to the prosthesis during walking, running or climbing. In order to achieve a good-fit socket for an individual amputee, it is essential to obtain the biomechanical properties of the residual limb. In current clinical practices, this is achieved by a touch-and-feel approach which is highly subjective. Although there have been significant advancements in prosthetic technologies such as microprocessor controlled knee and ankle joints in the last decade, the progress in designing a comfortable socket has been rather limited. This means that the current process of socket design is still very time-consuming, and highly dependent on the expertise of the prosthetist. Supported by the state-of-the-art sensor technologies and numerical simulations, a new socket design system is being developed to help prosthetists achieve rapid design of comfortable sockets for above knee amputees. This paper reports the research work related to establishing biomechanical models for socket design. Through numerical simulation using finite element method, comprehensive relationships between pressure on residual limb and socket geometry were established. This allowed local topological adjustment for the socket so as to optimize the pressure distributions across the residual limb. When the full body weight of a patient is exerted on the residual limb, high pressures and shear forces between the residual limb and the socket occur. During numerical simulations, various hyperplastic models, namely Ogden, Yeoh and Mooney-Rivlin, were used, and their effectiveness in representing the biomechanical properties of soft tissues of the residual limb was evaluated. This also involved reverse engineering, which resulted in an optimal representative model under compression test. To validate the simulation results, a range of silicone models were fabricated. They were tested by an indentation device which yielded the force-displacement relationships. Comparisons of results obtained from FEA simulations and experimental tests showed that the Ogden model did not fit well the soft tissue material indentation data, while the Yeoh model gave the best representation of the soft tissue mechanical behavior under indentation. Compared with hyperplastic model, the result showed that elastic model also had significant errors. In addition, normal and shear stress distributions on the surface of the soft tissue model were obtained. The effect of friction in compression testing and the influence of soft tissue stiffness and testing boundary conditions were also analyzed. All these have contributed to the overall goal of designing a good-fit socket for individual above knee amputees. Keywords: above knee amputee, finite element simulation, hyperplastic model, prosthetic socket

Authors: Rami Al Jafar, Paul Elliott, Konstantinos K. Tsilidis, Abbas Dehghan
Abstract:
Although Ramadan fasting is a ritual that is practiced every year by millions of Muslims, studies on Ramadan fasting are still scarce. To the best of our knowledge, none of the previous studies comprehensively explored the effect on the metabolic profile. In London Ramadan Fasting Study, blood samples were collected from 81 participants before and 10-14 days after Ramadan. Blood samples were analysed using nuclear magnetic resonance (NMR) spectroscopy which covers 249 metabolites. Mixed-effects models were used to analyse collected data and assess the effect of Ramadan fasting on the metabolic profile. Our observational study involved 85 individuals with a mean age of 45.2 years, and 53.1% of them were males. After Ramadan, forty metabolites were affected significantly by Ramadan fasting. Most of these metabolites were metabolites ratios (24), and the rest were three Glycolysis, three ketone bodies, nine Lipoprotein subclasses and one inflammation marker. This study suggests that Ramadan fasting is significantly associated with changes in the metabolic profile. However, the changes are assumed to be temporary, and the long-term effect of these changes is unknown. Keywords: metabolic profile, Ramadan fasting, metabolites, intermittent fasting

Authors: Harun Bozna
Abstract:
MOOC (massive open online course) is a groundbreaking education platform and a current buzzword in higher education. Although MOOCs offer many appreciated learning experiences to learners from various universities and institutions, they have considerably higher dropout rates than traditional education. Only about 10% of the learners who enroll in MOOCs actually complete the course. In this case, perceptions of participants and a comprehensive analysis of MOOCs have become an essential part of the research in this area. This study aims to explore the MOOCs in detail for better understanding its content, purpose and primarily drop out issues. The researcher conducted an online questionnaire to get perceptions of graduate students on their learning experiences in MOOCs and arranged a semi- structured oral interview with some participants. The participants are Turkish graduate level students doing their MA and Ph.D. in various programs. The findings show that participants are more likely to drop out courses due to lack of time and lack of pressure. Keywords: distance education, MOOCs, drop out, perception of graduate students

Abstract:
Liquid storage tanks have become widespread during the recent decades due to their extensive usage. Analysis of liquid containing tanks is known to be complex due to hydrodynamic force exerted on tank which makes the analysis a complex one. The objective of this research is to carry out analysis of liquid domain along with structural interaction for various geometries of circular tanks considering seismic effects. An attempt has been made to determine hydrodynamic pressure distribution on the tank wall considering impulsive and convective components of liquid mass. To get a better picture, a comparative study of Draft IS 1893 Part 2, ACI 350.3 and Eurocode 8 for Circular Shaped Tank has been performed. Further, the differences in the magnitude of shear and moment at base as obtained from static (IS 3370 IV) and dynamic (Draft IS 1892 Part 2) analysis of ground supported circular tank highlight the need for us to mature from the old code to a newer code, which is more accurate and reliable. Keywords: liquid filled containers, circular tanks, IS 1893 (part 2), seismic analysis, sloshing

Authors: P. Aarumugam, N. Krishnamoorthy, K. Gunasekaran
Abstract:
The mosquitoes with loss of minimum three legs especially the hind legs have the negative impact on the survival hood of mosquitoes. Three days old unfed adult female laboratory strain was selected in each generation against sublethal dosages (0.004%, 0.005%, 0.007% and 0.01%) of deltamethrin upto 40 generations. Impregnated papers with acetone were used for control. Every fourth generation, survived mosquitoes were observed for functional mortality. Hind legs lost were significantly (P< 0.05) higher in treated than the controls up to generation 24, thereafter no significant lost. In contrary, no significant forelegs lost among exposed mosquitoes. Middle legs lost were also not significant in the exposed mosquitoes except first generation (F1). The field strain (Chennai) did not show any significant loss of legs (fore or mid or hind) compared to the control. The selection pressure on mosquito population influences strong natural selection to develop various adaptive mechanisms. Keywords: Anopheles stephensi, deltamethrin, functional mortality, synthetic pyrethroids

Abstract:
The research leverages the proven HS-FCC technology to directly crack crude oils into petrochemical building blocks. Crude oils were subjected to an optimized hydro-processing process where metal contaminants and sulfur were reduced to an acceptable level for feeding the crudes into the HS-FCC technology. The hydro-processing is achieved through a fixed-bed reactor which is composed of 3 layers of catalysts. The crude oil is passed through a dementalization catalyst followed by a desulfurization catalyst and finally a de-aromatization catalyst. The hydroprocessing was conducted at an optimized liquid hourly space velocity (LHSV), temperature, and pressure for an optimal reduction of metals and sulfur from the crudes. The hydro-processed crudes were then fed into a micro activity testing (MAT) unit to simulate the HS-FCC technology. The catalytic cracking of crude oils was conducted over tailored catalyst formulations under an optimized catalyst/oil ratio and cracking temperature for optimal production of total light olefins. Keywords: petrochemical, catalytic cracking, catalyst synthesis, HS-FCC technology

Abstract:
In this paper, a brief literature review on cavitation jets is presented in order to introduce the cavitation mechanism, strategies to assess when cavitation occurs, and the factors that influence cavitation in cavitating jets. The objectivity of the cavitation number often used to predict cavitation is also discussed. The results show that cavitation cannot be foreseen just using the cavitation number. Therefore, more efforts are needed to innovate and develop a self-resonating jet geometry that would be maintains the flow and the pressure in the cavitation condition just earlier than the flow acts on the target that would be used in such operating conditions. This study focused on a particular aspect related to improving drilling efficiency and the rate of penetration (ROP). In addition, a discussion on the methods used to measure cavitation and the factors that affect cavitation occurrence will be discussed. Two different types of cavitation nozzles were designed and tested. It has been shown that the self-resonating cavitation nozzle presents greater performance than standard non-resonating nozzle. It is thus concluded that a self-resonating cavitation jet present a high potential for improving drilling performance. Keywords: cavitating jet, erosion, cavitation number, rate of penetration (ROP)

Procedia PDF Downloads 3262096 Thermodynamically Predicting the Impact of Temperature on the Performance of Drilling Bits as a Function of Time
Authors: Talal Al-Bazali
Abstract:
Air drilling has recently received increasing acceptance by the oil and gas industry due to its unique advantages. The main advantages of air drilling include the higher rate of penetration, less formation damage, lower risk of loss of circulation. However, these advantages cannot be fully realized if thermal effects in air drilling are not well understood and minimized. Due to its high frictional coefficient, low heat conductivity, and high compressibility, air can impact the temperature distribution of bit and thus affect its bit performances. Based on energy and mass balances, a transient thermal model that predicts bit temperature is presented along with numerical solutions in this paper. In addition, several important parameters that influence bit temperature distribution are analyzed. Simulation results show that the bit temperature increases with increasing weight on bit and rotary speed but decreases as the standpipe pressure and flow rate increase. These results can be used to optimize drilling operations and flow parameters for an improved bit performance as shown in this paper. Keywords: air drilling, rate of penetration, temperature, rotary speed

Three Young’s modulus (EL, ER, ET) and six Poisson ratios (μLR, μLT, μRL, μRT, μTL, μTR) were determined from compression test using bi-axial extensometer at constant moisture content (12 %). Three shear modulus were determined using ultrasound. Six shear wave velocities propagating along the principal axes of anisotropy were measured using EPOCH 650 ultrasonic flaw detector with 1 MHz transverse transducers. The properties of the samples tested were significantly affected by heat treatment by different degree. As a result, softer treatments yielded some amount of increase in Young modulus and shear modulus values, but increase of time and temperature resulted in significant decrease for both values. Poisson ratios seemed insensitive to heat treatment. Keywords: cedar wood, elastic constants, heat treatment, ultrasound

Abstract:
A Memorandum of Understanding (MoU) for the cessation of hostilities was signed by Aceh's longstanding adversaries (the Government of Indonesia and the Free Aceh Movement (GAM) in August 2005. The Government of Indonesia has given the autonomy to Aceh Province of Indonesia, the Law Number 11 of 2006 the authority of the Aceh government to the implementation of the Islamic Sharia. The implementation of Islamic Sharia, Aceh can be a role model of Islam that glorifies women, the implementation of Islamic law in Aceh when enacted and got legality because it supported the socio-cultural and historical community. The value of the value of women's lives is shifted under the pressure of applying Islamic law, with this argument, the importance of justice and equality of policy enforcement in women's lives. Based on interviews conducted in 2016 and 2017 with women's activists, government officials, women non-governmental organizations in Aceh, this paper finds that there is lack of gender balance because of the many problems involving women in the enactment of regional regulations and control policies on women's bodies. The research points to ensure the implementation of Islamic Sharia practitioners have only directed to women and discrimination against women. Keywords: women, policy, Islamic law, social

Abstract:
Supersonic flight promises speed, but the design of the air inlet faces an obstacle: shock waves. They prevent air flow in the mixed compression ports, which reduces engine performance. Our research investigates this using supersonic wind tunnels and schlieren imaging to reveal the complex dance between shock waves and airflow. The findings show clear patterns of shock wave formation influenced by internal/external pressure surfaces. We looked at the boundary layer, the slow-moving air near the inlet walls, and its interaction with shock waves. In addition, the study emphasizes the dependence of the shock wave behaviour on the Mach number, which highlights the need for adaptive models. This knowledge is key to optimizing the combined compression inputs, paving the way for more powerful and efficient supersonic vehicles. Future engineers can use this knowledge to improve existing designs and explore innovative configurations for next-generation ultrasonic applications. Keywords: oblique shock formation, boundary layer interaction, schlieren images, double wedge surface

Abstract:
Graphene is a single-atomic two-dimensional crystal of carbon atoms that has considerable properties due to its unique structure and physics with applications in different fields. Graphene has sensitive electrical properties due to its atomic-thin structure. Along with the substrate materials and their influence on the transport properties in graphene, design and fabrication of graphene-based devices for biomedical and biosensor applications are challenging. In this work, large-area high-quality graphene nanosheets were prepared by low pressure chemical vapor deposition using methane gas as carbon source on copper foil and transferred on the biocompatible substrates. Through deposition of titanium and gold contacts, current-voltage response of the transferred graphene on four biocompatible substrates, including PDMS, SU-8, Nitrocellulose, and Kapton (Fig. 2) were experimentally determined. The considerable effect of the substrate type on the electrical properties of graphene is shown. The sheet resistance of graphene is changed from 0.34 to 14.5 kΩ/sq, depending on the substrate. Keywords: biocompatible substrates, electrical properties, graphene, sheet resistance
